Les procédés actuellement connus pour la préparation de composés de la formule générale I emploient comme produit de départ la 7-chloro-1,3-dihydro-3amino-5-phényl-2H-1,4-benzodiazépine-2-one avec formation du sel de diazonium comme composé intermédiaire, alors que le procédé suivant l'invention utilise comme matière de départ la 7-chloro-1,3-dihydro-3-hydrazino-5-phényl2H-1,4-benzodiazépine-2-one, le produit intermédiaire étant le dérivé 2-azido.
Le procédé suivant l'invention est plus aisé à réaliser, parce qu'il élimine la réduction compliquée
et dispendieuse du groupe N-oxyde, qui se forme dans le procédé connu de préparation du 4-oxyde de la 7-chloro-

one par cyclisation de l'oxime de la benzophénone à l'aide de l'acide trifluor-acétique.
Le procédé suivant l'invention est réalisé en ajoutant une solution aqueuse du nitrite de sodium sous agitation à la solution du composé de départ dans un acide minéral aqueux, maintenue par refroidissement entre
-5 et O[deg.]C. Après neutralisation du mélange réactionnel, on sépare par filtration le produit réactionnel formé et on le lave à l'eau. Le produit brut obtenu peut être purifié par une technique usuelle comme la recristallisation dans un
alcool aliphatique inférieur ou une chromatographie en couche mince ou sur colonne de gel.
L'acide minéral utilisé pour acidifier le milieu réactionnel est avantageusement l'acide chlorhydrique ou l'acide sulfurique.
Les composés de départ de la formule II, c'est-à-
dire les 7-chloro-1,3-dihydro-3-hydrazino-5-phényl2H-1,4-benzodiazépine-2-ones, constituent eux aussi des composés nouveaux et l'un des objets de l'invention.
Le procédé pour leur préparation consiste à faire réagir un composé de la formule

organique aprotique, inerte dans les conditions réactionnelles vis-à-vis des réactifs et du produit formé, en présence d'un excès d'hydrazine, le produit réactionnel étant séparé par filtration et lavé, d'abord avec un solvant organique inerte, puis à l'eau.
Les composés de la formule IV et l'hydrazine sont
des produits disponibles sur le marché.
Les composés de la formule générale I agissent sur
le système nerveux central et sont employés comme médicaments tranquillisants. Ils possèdent des effets anxiolytiques, sédatifs, anti-convulsifs et myo-résolutifs prononcés. Ils ont de plus l'avantage d'être nettement moins toxiques que d'autres dérivés benzodiazépiniques analogues.
L'invention est illustrée ci-après par quelques exemples non limitatifs de modes de réalisation. Préparation des composés de la formule II a) A la solution de 5,0 g (0,17 mole) d'hydrazine anhydre dans 50 ml de nitrile acétique anhydre,refroidie à 0[deg.]C, on ajoute en l'espace de 30 mn et en agitant la solution de 11,9 g (35 millimoles) de chlorhydrate de 3,7-dichloro-5-phényl-2H-1,3-dihydro1,4-benzodiazépine-2-one dans 380 ml de nitrile acétique anhydre. Après une agitation de 30 mn supplémentaires entre 0 et 5[deg.]C, on sépare le produit réactionnel pré- cipité par filtration, on le lave avec 50 ml de nitrile acétique et le résidu mis en suspension dans 250 ml
d'eau est agité pendant 30 mn. Par filtration et séchage,

b) A la solution de 0,8 g (27 millimoles) d'hydrazine anhydre dans 6 ml de N,N-diméthyl-formamide anhydre, refroidie à 0[deg.]C, on ajoute en l'espace de 30 mn et en agitant la solution de 1,18 g (3,lmmoles) de chlorhydrate

Après 30 mn d'agitation supplémentaires entre 0 et 5[deg.]C, le mélange réactionnel est versé dans 100 ml d'eau refroidie à la glace et une agitation de 15 mn conduit à la précipitation du produit brut, qui est séparé par filtration et lavé avec 20 ml d'eau distillée.

On prépare une solution de 3,0 g (1 millimole) de 7-chloro-3-hydrazino-5-phényl-2H-1,3-dihydro-1,4-benzodiazépine-2-one dans 100 ml d'une solution aqueuse à

froidie à -5[deg.]C, on ajoute en agitant la solution aqueuse de 0,76 g (11 millimoles) de nitrite de sodium (donnant une réaction positive avec l'indicateur potassium-amidon) ,

a En refroidissant vers -5[deg.]C et en agitant, on prépare une solution de 0,2 g (6 millimoles) de 7-chloro-3- hydrazino-5-(2-chlorophényl)-2H-1,3-dihydro-1,4-benzodia-

En maintenant la solution dans cette gamme de températures, on y ajoute lentement et en agitant une solution aqueuse de 50 mg (0,07=mole) de nitrite de sodium
(donnant una réaction positive avec l'indicateur au potassium-amidon), puis on poursuit l'agitation pendant
10 mn dans la même gamme de températures, avant de neutraliser le mélange réactionnel à l'aide d'une solution aqueuse saturée d'hydrogénocarbonate de sodium.
Le produit précipité est séparé par filtration et lavé avec 10 ml d'eau, puis séché à l'air.
Par purification du produit brut obtenu par une chromatographie en couche mince, on obtient 0,06 mg,

The processes currently known for the preparation of compounds of general formula I use as starting material 7-chloro-1,3-dihydro-3amino-5-phenyl-2H-1,4-benzodiazepine-2-one with formation of the diazonium salt as intermediate compound, whereas the process according to the invention uses as starting material 7-chloro-1,3-dihydro-3-hydrazino-5-phenyl2H-1,4-benzodiazepine-2-one, the intermediate product being the 2-azido derivative.
The process according to the invention is easier to carry out, because it eliminates the complicated reduction
and expensive N-oxide group, which is formed in the known process for the preparation of 4-oxide of 7-chloro-

one by cyclization of the benzophenone oxime using trifluoracetic acid.
The process according to the invention is carried out by adding an aqueous solution of sodium nitrite with stirring to the solution of the starting compound in an aqueous mineral acid, maintained by cooling between
-5 and O [deg.] C. After neutralization of the reaction mixture, the reaction product formed is filtered off and washed with water. The crude product obtained can be purified by a usual technique such as recrystallization in a
lower aliphatic alcohol or thin layer or gel column chromatography.
The mineral acid used to acidify the reaction medium is advantageously hydrochloric acid or sulfuric acid.
The starting compounds of formula II, i.e.
