L'amine réagit avec le 1-(oméga-halogénoalcoxy)-benzène substitué en position 2- en quantité équimoléculaire. Toutefois, l'utilisation d'un excès d'amine accélère la réaction. Normalement, on utilise de 1 à 100 moles et de préférence de 2 à 40 moles de <EMI ID=61.1>
position 2-. Lorsque l'excès est important, il sert de solvant. La réaction peut être effectuée en l'absence de solvant étranger mais l'utilisation d'un solvant inerte dans la réaction permet d'opérer en milieu homogène. Parmi les solvants qui conviennent, on peut citer l'eau, le dioxanne, le tétrahydrofuranne, le diméthylsulfoxy.de, les alcools aliphatiques inférieurs et leurs mélanges. Les solvants préférés sont les mélanges alcool aliphatique inférieur-eau.
La température de réaction ne constitue pas un facteur critique mais se situe normalement entre la température ambiante et 150[deg.]C, de préférence entre la température ambiante et 100[deg.]C. La durée de réaction varie très largement avec la température et la réactivité des composés de départ mais normalement elle se situe entre 10 mn et 40 heures.
Lorsqu'on opère en présence d'une base capable ce neutraliser un halogénure d'hydrogène formé.dans le cours de la réaction, celle-ci est accélérée. Parmi les bases qui conviennent, on citera des bases minérales telles que l'hydroxyde de potassium, l'hydroxyde de sodium, le carbonate de potassium, le carbonate de sodium et les bases analogues, et les amines tertiaires telles que la pyridine, la triéthylamine et les amines tertiaires analogues. Normalement, on utilise la base en quantités de 1 à 5.moles par mole du 1-(oméga-halogénoalcoxy)-benzène substitué en position 2-.
Si l'on opère en l'absence d'une base, le 1-(omégaaminoalcpxy)-benzène substitué en position 2- réagit avec l'halogénure d'hydrogène formé dans le cours de la réaction et est converti-en sel de cet halogénure d'hydrogène.
On peut par ailleurs-préparer facilement les sels des 1-(oméga-aminoalcoxy)-benzènes substitués en position 2- en les mettant en contact avec un acide approprié.
Les 1-(oméga-aminoalcoxy)-benzènes substitués en position 2- et'leurs sels formés par addition avec des acides peuvent être purifiés par recristallisation dans un solvant approprié, par exemple un mélange alcool-éther.
Des essais pharmacologiques effectués sur les 1-(oméga-aminoalcoxy)-benzènes substitués en position 2- ont
montré que ces composés pouvaient être utilisés comme agents anti- <EMI ID=62.1>
par l'effet antagoniste sur l'hypothermie provoquée par administration de réserpine à des souris.
On a procédé sur des souris à des essais visant à mettre en évidence une activité anti-dépressive, sédative, anti-convulsive,

Les résultats rapportés dans les tableaux I et II montrent que les 1-(oméga-aminoalcoxy)-benzènes substitués en position 2- manifestent une activité anti-reserpine comparable
à celle de l'Amitriptyline, avec toutefois une faible toxicité,
une faible activité dépressive CNS et une faible action anticholinergique.
Les composés selon l'invention peuvent être administrés par un mode quelconque permettant de pallier aux états dépressifs chez les êtres vivants à sang chaud. Ainsi par exemple, on peut les administrer par voie parentérale, sous-cutanée, intra-veineuse, intramusculaire ou intrapéritonéale. On peut encore les administrer, uniquement ou concurremment à un. autre mode d'administration, par voie orale. La posologie dépend de l'âge, de l'état de santé et du poids du patient, de l'état de dépression, des types de Traitements conjoints éventuels, de la fréquence du traitement et de la nature de l'effet recherché. En général, la posologie quotidienne est d'environ 0,5 à 50 mg de composant actif par kg de poids corporel. Normalement, une posologie quotidienne de 1 à 30 mg par kg en une ou plusieurs doses permet de parvenir aux résultats recherchés.
Les composés de formule I peuvent être administrés sous forme d'unités de dosage tels que des comprimés, des capsules,
des sachets de poudre ou des solutions, suspensions ou élixirs liquides pour l'administration orale, ou sous forme de compositions <EMI ID=77.1>
