Het vloeibaar en/of oplosbaar maken van zetmeel vindt plaats met het oog op allerhande toepassingen, zoals het fabriceren van maltodextrinen, alsook als eerste stadium van het versuikeringsproces bij het fabriceren van zetmeelstroop, dextrose, levulose e.d. Het is in de meeste gevallen ten zeerste wenselijk dergelijke waterige zetmeeldispersies te maken, waarvan het droge-stofgehalte zo hoog mogelijk is. Het hoogste droge-stofgehalte dat kan worden verkregen volgens de konventionele werkwijze, uitgaande van ongewijzigd zetmeel, ligt echter niet hoger dan ca. 40 gewichts-%. De reden daarvan is
dat zetmeel verstijfselt tijdens het vloeibaar maken volgens
de gebruikelijke werkwijze, waarbij de viskositeit sterk oploopt (z.g. viskositeitspiek) en een brij van verstijfseld,
maar verder ongewijzigd zetmeel met een droge-stofgehalte van

de konventionele installaties. Het is dan ook gebruikelijk het zetmeel vloeibaar te maken bij een droge-stofgehalte van niet meer dan 40% en eventueel daarna het droge-stofgehalte op te voeren tot het gewenste peil door verdamping in een afzonder- . lijk verdampingsstadium. Deze werkwijze biedt echter het nadeel dat ze voor de verdamping een speciale installatie vergt en bovendien dat het energieverbruik vrij hoog is.
Welnu de Aanvraagster heeft een werkwijze gevonden voor het maken van zeer geconcentreerde waterige dispersies van vloeibaar gemaakt en praktisch geheel oplosbaar gemaakt zetmeel, met een droge-stofgehalte van 70 gewichts-% en meer, door middel van een gebruikelijke installatie zonder afzonder- <EMI ID=3.1>
wijze volgens de uitvinding is een waterige dispersie van een zetmeelhydrolysaat met een dextrine-ekwivalent van ca. 1-25, welke waterige dispersie een droge-stofgehalte van ten hoogste 40 gewichts-% heeft. Dit uitgangsmateriaal kan werden gemaakt volgens de een of andere konventionele werkwijze, bijvoorbeeld door hydrolyse in zuur milieu, enzymatische hydrolyse in zuur milieu, enzymatische hydrolyse e.a. Volgens een alternatieve werkwijze is het ook mogelijk uit te gaan van

voorkeur in de vorm van opeenvolgende dosissen, vers korrelvormig zetmeel toegevoegd in aanwezigheid van een bakteriële alfa-amylase, onder omstandigheden waarbij geen noemenswaardige verstijfseling van het zetmeel optreedt, dus zonder de sterke toename van de viskositiet, die typisch is voor het verstijfselen van zetmeel. Bij het begin van het toevoegen van vers zetmeel moet de temperatuur van het mengsel onder het verstijfselingspunt van zetmeel worden gehouden ten einde verstijfseling te voorkomen. Na het toevoegen van het verse zetmeel en het vloeibaar maken ervan door de inwerking van
de alfa-amylase, kan de temperatuur geleidelijk worden opgevoerd tot een punt dat aanmerkelijk boven het "normale" verstijfselingspunt ligt, zonder noemenswaardige verstijfseling noch sterke stijging van de viskositeit. Dank zij het geleidelijk toevoegen en vloeibaar maken van zetmeel aan het mengsel onder omstandigheden zonder gevaar voor verstijfseling, kan het droge-stofgehalte van het mengsel zeer hoog worden opgevoerd, nl. tot 70 gewichts-% en zelfs hoger, in een konventionele installatie zonder enige moeilijkheid met de viskositeit.
Na het toevoegen van de laatste hoeveelheid vers zetmeel en nadat vrijwel al het zetmeel vloeibaar en oplos-

temperatuur desnoods tot 150[deg.]C kan worden verhoogd. Dit opvoeren van de temperatuur dient voor het vloeibaar en grotendeels oplosbaar maken van het resterende zetmeel.
De vereiste hoeveelheid alfa-amylase kan tijdens het toevoegen en vloeibaar maken van het verse zetmeel worden toegevoegd. De meest praktische en doelmatige werkwijze bestaat echter in het toevoegen van een voldoende hoeveelheid alfa-amylase aan het uitganssmengsel met het oog op
een efficiënt vloeibaar maken van de totale hoeveelheid toe te voegen zetmeel. Het voordeel daarvan is dat extra toevoeging van enzyme tijdens het verloop van het proces kan worden voorkomen. Alle faktoren kunnen zo worden geregeld dat het proces vlot verloopt zonder schadelijke inwerking

De temperatuur bij het begin van het toevoegen van het verse zetmeel moet zo worden gekozen dat de werking van

Verkieslijke leveranciers van alfa-amylase voor de werkwijze volgens de uitvinding zijn o.a. sommige mikro-organismen uit het geslacht Bacillus (bakteriën), zoals

Een bijzonder geschikte alfa-amylase wordt geleverd door de bakterie Bacillus licheniformis volgens de voornoemde Oostenrijkse oktrooiaanvrage. Men geeft de voorkeur aan de alfa-amylase, voortgebracht door de Bacillus licheniformis-stam NCIB 8061. Uitstekende alfa-amylasen zijn eveneens afkomstig van de B.

abs. monster.= absorberend vermogen van het monster De gebruikte hoeveelheid alfa-amylase is natuurlijk de vereiste hoeveelheid voor het vloeibaar maken van de gehele hoeveelheid korrelvormig zetmeel en heeft een aktiviteit van ca. 0,5-25 E/g zetmeel (droog). Men kan ook meer gebruiken, maar dit biedt geen praktische voordelen.
Zoals reeds boven werd gezegd, is het verkieslijk de gehele alfa-amylase-dosis toe te voegen bij het begin van het proces, wat natuurlijk niet wegneemt dat het enzyme eventueel ook in deeldosissen tijdens het verloop van het proces kan worden toegevoegd. De pH moet tijdens het vloeibaar maken van het zetmeel natuurlijk zo zijn ingesteld dat de aktiviteit van de alfa-amylase maximaal is, nl. meestal bij 5-7,5
(bij voorkeur bij ca. 6).
De omstandigheden waarbij het toevoegen van het zetmeel plaats vindt, nl. de telkens toe te voegen dosis en de temperatuur, moeten zorgvuldig worden gekozen ten einde waarneembare verstijfseling en te sterk oplopen van

meeldosissen kan plaatsvinden op alle ogenblikken waarop reeds een voldoend deel van het eerder toegevoegde zetmeel vloeibaar gemaakt is, zo dat de toevoeging van vers zetmeel geen al te sterke verhoging van de viskositeit tot gevolg heeft. Bij het gebruik van een traditionele installatie voor de toepassing van de werkwijze volgens de uitvinding is het best alle faktoren van het proces zo te regelen dat de viskositeit niet hoger oploopt dan ca. 3000 cp. Dit neemt niet weg dat de werkwijze volgens de uitvinding ook met goed gevolg kan worden toegepast bij viskositeiten die hoger liggen dan ca. 3000 cp mits een geschikte installatie te gebruiken die zich leent voor het verwerken van mengsels met dergelijke hoge viskositeiten.
Een van de grote voordelen van de werkwijze volgens de uitvinding ligt echter in het lage energieverbruik, te danken aan het feit dat het gebruik van allerhande speciale inrichtingen kan worden vermeden, dat het niet nodig is het mengsel te verdampen of het verdampen althans tot een minimum kan worden beperkt, enz. Het verdient daarom de voorkeur het proces volgens de uitvinding te laten verlopen in een konventionele installa-

Het is een bekend feit dat zetmeel vloeibaar kan worden gemaakt met alfa-amylase zonder verstijfseling. Ook leert de recente ervaring op dit gebied dat het mogelijk
is zetmeel op efficiënte wijze vloeibaar te maken door behandeling met bakteriële alfa-amylase bij temperaturen boven
de normale verstijfselingstemperatuur zonder dat noemenswaardige verstijfseling plaats vindt (zie dienaangaande o.a.
de werkwijze beschreven in de US-oktrooien 3.922.199 op naam van Hebeda e.a., 3.922.200 op naam van Walon e.a., 3.922.198
op naam van Kuske e.a. en 3.922.196 op naam van Leach e.a.).
De toepassing van deze recent ontwikkelde verkwijzen is echter beperkt tot zetmeeldispersies met een zet-

installaties worden gebruikt. Daarbij komt dat, als wordt gewerkt bij temperaturen die aanmerkelijk hoger liggen dan het normale verstijfselingspunt (b.v. 75[deg.]C voor malszetmeel), de behandeling van de waterige zetmeeldispersie met het enzyme eerst moet plaatsvinden bij lagere temperatuur alvorens de temperatuur kan worden opgevoerd tot de vereiste eindtemperatuur, als het erop aankomt verstijfaeling te vermijden. Bij het rechtstreeks toevoegen van zetmeel en alfa-amylase aan heet water met een temperatuur boven het normale verstijfselingspunt van het betrokken zetmeel, begint het zetmeel al dadelijk te verstijfselen. Het is echter een verrassend feit dat, bij toepassing van de werkwijze volgens

de onderhavige uitvinding, het toevoegen van vers korrelvormig zetmeel aan een waterige dispersie van vloeibaar gemaakt zetmeel bij een zo hoge temperatuur als 75-85[deg.]C en zelfs hoger, geen waarneembare verstijfseling, gepaard met sterf oplopen van de viskositeit (viskositeitspiek), optreedt.
Bij de praktijktoepassing van de werkwijze volgens de uitvinding vindt het vloeibaar en oplosbaar maken van zetmeeldispersies met hoog droge-stofgehalte grotendeels plaats tijdens het opvoeren van de temperatuur tot ca.
80-85[deg.]C en is het daarbij verkregen produkt zonder meer geschikt voor verdere verwerking. Het verdient echter aanbeveling de temperatuur uiteindelijk op te voeren tot tenminste

oplosbaar te maken. De aldus verkregen sterk gekoncentreerde dispersie van vloeibaar en oplosbaar gemaakt zetmeel kan
als dusdanig worden gebruikt voor de een of andere gebruikelijke toepassing, zoals bijvoorbeeld voor het maken van maltodextrinen en andere vloeibare zetmeelpreparaten, alsook
als substraat voor verdere versuikering.
Bij het gebruik van gekoncentreerde zetmeel oplossingen, verkregen volgens de werkwijze van de uitvinding, voor verdere versuikering volstaat het de temperatuur en de pH op de vereiste waarde in te stellen, het geschikte versuikeringsenzyme of de geschikte versuikeringsenzymen toe te voegen en het versuikeringsproces op de gebruikelijke wijze te laten verlopen. Als men bijvoorbeeld dextrose of een dextrosehoudend hydrolysaat wenst te maken, moet glyko-amylase worden toegevoegd. Voor het maken van levulosestroop kan glukose-isomerase worden toegevoegd na of tijdens de werking van de gluko-amylase.
Voor het maken van een produkt met hoog maltosegehalte wordt B-amylase met of zonder a-1-6-glukosidase gebruikt. Boven- <EMI ID=24.1>
amylase of B-amylase, met goed gevolg worden toegevoegd bij het begin of tijdens het verder verloop van het vervloeiingsproces te zamen met de bakteriële alfa-amylase, aangezien het proces sneller en doelmatiger verloopt in aanwezigheid van dergelijke enzymen.
De werkwijze volgens de uitvinding leent zich voor het behandelen van allerhande zetmeelsoorten, zoals maïszetmeel, tarwezetmeel, aardappelzetmeel, rijstzetmeel e.a., alsook allerhande hoofdzakelijk uit amylopektine bestaande wasachtige zetmeetsoorten en voorts ook zetmeel met hoog amylosegehalte. Voorts komen ook allerhande fysische en/of chemisch gewijzigde zetmeelsoorten, zoals chemische zetmeelderivaten e.d., in aanmerking voor behandeling volgens de werkwijze van de uitvinding. Dit geldt eveneens voor zetmeelhoudende produkten, zoals bloem, meel e.d.
De onderstaande voorbeelden dienen tot nadere toelichting van het gebruik van de werkwijze volgens de uitvin-
