La présente invention concerne un procédé de prépara- tion de nouveaux composés hétérocycliques contenant du bore qui ont d'intéressantes propriétés. Ces composés répondent à la for- mule générale:

Les nouveaux composés de formule 1 ont des propriétés remarquables et,entre autres,,un effet marqué sur les micro- organismes. Cet effet peut être mis en évidence tant in vitro que sur l'organisme infecté (après administration des composés par voie parentérale ou orale). Par exemple, la dose curative 50 (c'est-à-dire la dose dont l'administration guérît l'infection chez 50% des animaux d'essaiàen administration par voie orale chez la souris infectée par E. coli,a les valeurs indiquées dans le tableau suivant pour les composés de formule générale I, où Z représente un radical o-phénylène et R1 a les significations précisées dans le tableau.

(Au cours de ces essais, les animaux infectés par E. coli ou B. proteus reçoivent par voie ora- le une dose unique du composé essayé immédiatement après l'infec- tion,mais dans le cas des Infections par S. typhimurium, le traitement est appliqué au moyen d'une même.dose une deuxième fois 24 heures et une troisième fois 48 heures après l'infection).
De nombreux composés de formule 1 sont non seule- ment actifs contre des bactéries à Gram négatif, mais également très efficaces contre les protozoaires et en particulier les trypanozomes et plasmodi es . Par exemple, des groupes de 10 souris ont été infectées au moyen de Plasmodium berghei. Dans un délai de 11 à 30 jours après l'infection, tous les animaux d'un groupe témoin sont emportés par l'infection. Au contraire, 40 jours après l'infection, tous les animaux ayant reçu pendant 5 jours le composé c) survivent, la dose administrée étant de 2,5mg par jour pour une souris de 20 g. Aucune plasmodie ne peut encore ûtre mise en évidence dans des frottis de sang des animaux ainsi traités.

Certains des composés de, formule générale I,et en particulier ceux dans la formule desquels fi représente un radical hétérocyclique éventuellement substitueront en outre des propriétés cytostatiques. Ces propriétés,de même que la présence au bore dans ces composés, les rendent particulièrement intéressants pour le traitement de certaines tumeurs, par exemp- le pour favoriser le traitement des tumeurs par irradiation. Cet effet est particulièrement marqué dans le cas des composés de formule I contenant l'isotope de bore B10.
L'administration des composés de formule I à des animaux d'essai a permis de faire ressortir les différences de toxicité qui sont spécialement marquées lorsque l'administration est poursuivie longtemps. Par exemple, la dose létale 50(ce-est- à-dire la dose dont l'administration entraine la'mort de 50% des animaux d'essai) est la suivante après une .seule administration par voie orale des composés essayés, (les valeurs sont données en mg/kg du poids du corps).

On peut déduire des résultats des essais que les com- posés de formule I sont des produits thérapeutiquement très in- téressants. Par exemple, le composé c) (en raison de sa toxici- té marquée lors d'une administration prolongée) ne convient que pour des traitements locaux ou pour une administration par voie orale de petite durée, mais le composé g) se prête en particulier à l'administration,tant topique qu'orale ou parentérale chez l'hom- me et les animaux,pendant une durée même longue. (Il convient de noter,en outre,que le composé g) a une dose létale 50 de plus de 5000 mg/kg de poids du corps chez la souris et le rat en.administration intrapéritonéale).

Les composés de formule générale I,où Z et/ou R1 portent un ou plusieurs radicaux nitro comme substituants,peuvent être préparés de la manière décrite ci-dessus au moyen de composés de formule II et/ou III portant des radicaux nitro. Toutefois, le composé correspondant exempt de radicaux nitro peut être préparé d'abord puis nitré. Les radicaux nitro dans les composés obtenus de cette manière peuvent être réduits en radicaux amino, par exemple au moyen d'hydrogène naissant ou activé catalytiquement.
On peut préparer les composés de formule générale 1,où Z et/ou fil portent un ou plusieurs radicaux amino,en préparant d'abord les composés correspondants oû Z et/ou fil portent comme substituants un ou plusieurs radicaux acylamino et en éliminant ensuite le ou les radicaux acyle par hydrolyse, de préférence en milieu acide,ou par hydrogénolyse (lorsqu'il s'agit dans le cas du radical acyle, par exemple d'un radical carbobenzoxy d'un radical o-nitrophénoxyacétyle ou d'autres radicaux ana- logues susceptibles d'hydrogénolyse, par exemple au moyen d'hy- drogène activé catalytiquement).
On peut aussi modifier ultérieurement d'autres sub- stituants de Z et/ou R1, par exemple.comme décrit ci--dessus, des radicaux carboxamide, qui se transforment en radicaux amino dans les conditions de la dégradation d'Hofmann, par exemple par traitement au moyen d'un hypochlorite ou hypobromite.
L'invention est davantage illustrée par les exemples suivants, au cours de l' exécution desquels le rendement maximum n'est pas recherché. Les températures indiquées !'le sont pas cor- rigées.
EXEMPLE 1 - On mélange une solution de 3,44 g d'hydrazide diacide benzène-sulfonique dans 20 ml d'éthanol chaud avec une solution

On chauffe le mélange de réaction pendant quelques minutes au reflux. On recueille par filtration les cristaux qui précipi- tent par refroidissement, puis on les recristallise dans l'alcool.
