"Vaccine against trachoma and method for preparing it" @ The invention relates to a method for preparing a viral vaccine against trachoma as well as the vaccine prepared by this method.
More particularly, the invention aims to separate the trachoma virus from the undesirable products which it contains, so that the purified vaccine thus obtained can be used successfully both in the prevention and in the treatment of trachoma. -trachoma was first isolated by T'ang et al (Chainese Med. J. 75.1957, page 429) in 1957 and the following year LH
Collier and others (J. Lancet 1958, I, page 993) isolated- this virus-by the method, of T'ang and confirmed its identity -in infecting the eyes of a patient with trachoma. unintentionally, Subsequently, in different laboratories, the trachoma virus was isolated from patients with trachoma *

In the yolk sac Boeufs incubated at +39 C for
6 days, the viral strain diluted with a buffer solution of sucrose, potassium phosphate and glutamate at pH 7 is injected (PGS buffer described in J. Bact. 59, 1950, page 509).
When 50% egg mortality is reached, the eggs still alive are opened and the membranes (maintained at -70 ° C) are used to inoculate other eggs. The infected membranes are suspended 50% in a solution of sucrose, potassium phosphate and potassium glutamate, and kept at a low temperature until they are used to prepare the vaccine.
The suspension is homogenized by stirring and a part of homogenized suspension is added 3 parts of trypsin suspension in a phosphate buffer solution (100 mg of trypsin / 100 cm3 of buffer solution)
at pH 7.5-8, at a temperature. from 34-40 C. The suspension obtained is homogenized by stirring it for 15 minutes at a temperature of
35-40 C during this phase, the virus separates from the protein part which is then removed by centrifugation at
125-650 G; then- active part of the upper portion which floats by centrifuging at 2000-6500 G.
The active part obtained is brought back to the initial volume by means of a phosphate buffer solution at pH 7.5-8; then we add diatomaceous earth (at a rate of 2 g per 100 cm3 'of suspension) to eliminate the other foreign substances which are finally separated by centrifugation at 650 G.
We receive @ ille the floating part. And we. the. centrifuged at 2000-6500 G .. to separate the active part again .. This is suspended in a phosphate buffer solution, in a volume .. equal to that of the 50% suspension. of the infected mombranes. , and we make it react. all for a few minutes with an equal slide of ethyl ether to extract the fats and foreign substances soluble in the ether.
