L'invention a encore pour objet un procédé permet- tant de transformer des #4-3-céto-prégnènes directement et / avec un rendement élevé; en #1,4-3-céto-prégnadiènes.
L'invention a aussi pour objet la production de #1,4-3-céto-stéroïdes de la série du prégnane directement et avec un rendement élevé à partir des 3-céto-prégnanes correspondants.
On a découvert à présent que la déshydrogénation sélective de composés stéroïdes peut être exécutée par un nouveau processus de déshydrogénation bactériologique, consistant à mettre un composé stéroïde, en particulier un composé stéroïde 3-oxygéné C-5 non saturé, avec l'activité déshydrogènante de microorganismes du bonne Mycobacterium, de préférence des microorganismes appartenant à l'espèce Mycobacterium smegmatis, Mycobacterium phlei, Mycobacterium lacticola, et Mycobacterium tuberculosis. Les microorganismes du genre Mycobacterium peuvent être obtenus à partir de sources connues, telles que celles de la American type culture collection, Washington,D.C. ou peuvent être isolés de sources naturelles, telles que sol ou maladies, par des procédés connus .
La déshydrogénation peut s'effectuer en mettant le composé stéroide en contact avec les microorganismes du genre Mycobacterium eux-mêmes ou, si on le préfère, avec des systèmes enzymatiques des microorganismes du genre Mycobactérium, de façon que l'hydrogène attaché aux atomes de carbone C-1 et C-2 soit sélectivement éliminé, de manié-, re à produire le #1-stéroïde désiré sensiblement non contaminé par des sous-produits non désirés.

La déshydrogénation bactériologique suivant la présente invention s'opère en mettant le composés stéroïde 3-oxygéné en contact avec l'activité @ @hydrogénante de mi- croorganismes du genre Mycobacterium. (;-,.ci peut se faire en ajoutant le composé stéroïde, sous forme de solide ou sous forme d'une solution dans un solvant, tel que, par exemple, une dialcoyl-cétone comme l'acétone, une dialcoylformamide comme la diméthylformamide, et analogues, dans des condi- tions stériles, à une culture de microorganisme; dans un milieu nutritif et en agitant le mélange résultant de façon à assurer la croissance du microorganisme et la déshydrogé- nation du composé stéroïde.
Le stéroïde peut être ajouté au moment où le milieu nutritif est inoculé à l'aiae de la cultt . re de microorganismes du genre Mycobacterium, ou bien il peut être ajouté à une culture établie. Au lieu d'ajouter le com- posé stéroïde à la culture établie dans le milieu nutritif, les cellules de cette culture établie peuvent être séparées par filtration du @ouillon, lavées à l'eau distillée, puis mises en suspension dans une solution aqueuse tamponnée con- tenant le composé stéroïde 3-oxygéné, le mélange résultant étant agité, de manière à déshydrogéner le composé sté- roïde, de façon à former le /\ -stéroïde 3-oxygéné.
Ce der- nier est plus facilement récupéré de ce milieu que du mélan- ge obtenu, lorsque le stéroïde est incubé avec le microorga- nisme dans .le milieu nutritif original. Ou bien .e composé

stéroïde 3-oxygéné peut être mis en contact avec des prépa- rations enzymatiques déshydrogénantes provenant de la crois- sance de microorganismes du genre Mycobacterium, de manière à former le composé stéroïde #1-3-oxygéné correspondant.
Les milieux nutritifs utilisés pour exécuter cette déshydrogénation bactériologique sont ceux ordinairement uti- lisés pour la propagation des mioroorganismes du genre Myco- bacterium. Les milieux nutritifs usuels contiennent une source d'azote et de carbone, des sels inorganiques et,au besoin, des facteurs de croissance. Le carbone peut être fourni par des composés, tels qu'acétates, lactates et analo- gues. L'azote peut être fourni;par un sel ammonique, des a- mino-acides ou des protéines,: telles que graines de soja, avoines, levure, extraits de levure, produit de digestion, truptique de caséine, extrait de viande, farine de sang, vi- ande protéinique et farine dbs, farine de saumon, farines de poisson, solubles de poisson, solubles de distillateurs et analogues.
Si on le désire, les microorganismes du genre
Mycobacterium peuvent être propagées en utilisant des pro- téïnes (ou des amino-acides) sans qu'un hydrate de.carbone quelconque soit présent dans le milieu, auquel cas les pro- téines ou amino-acides servent de source de carbone et d'azo- te requise par les microorganismes.
Bien que, comme on l'a signalé plus haut, la déshy- drogénation du stéroïde 3-oxygéné puisse être exécutée en utilisant des préparations enzymatiques déshydrogénantes pro- venant de la croissance de microorganisme du type Mycobacte- rium ou en mettant le composé stéroïde en contact avec une suspension d'une culture établie dans de l'eau distillée,on préfère ordinairement ajouter le stéroide 3-oxygéné à un milieu nutritif contenant une \ culture de 24 heures de micro- organismes du genre Mycobacterium. stéroïde, qui peut être ajoutée au milieu varie selon le substrat particulier à déshydrogéner, mais on' préfère or- dinairement employer une concentration d'environ 0,
005% à' 0,2% de stéroïde 3-oxygéné, bien que des concentrations plus élevées ou moins élevées puissent être utilisées, si on le désire. La culture contenant le stéroïde 3-oxygéné ajou- té est alors incubée, de préférence en agitant et en aérant ! pendant une période additionnelle, qui varie ordinairement entre environ 10 heures et 50 heures, bien que des durées de fermentation plus courtes ou plus longues puissent être avantageuses pour la déshydrogénation de 3-céto-stéroïdes particuliers.
En raison du fait que des fermentations pro- longées peuvent- donner lieu à la destruction d'une partie du #1-3-céto-stéroïde, on préfère ordinairement appliquer une durée de fermentation d'environ 10 heures à 24 heures, qui, selon le substrat stéroïde, a donné les rendements max- ima en produit stéroïde #1-déshydrogéné.
Lorsque le processus de déshydrogénation est ter- miné, le produit est avantageusement .récupéré du bouillon fermenté par extraction à l'aide d'un solvant non miscible à l'eau, tel qu'un hydrocarbure chloré comme le chloroforme, le une cétone, tel que la méthylisobutylvétone, un alcan ate d'alcoyle, tel que l'acétate d'éthyle, et analogues. L'ex- trait de stéroïde /\ -déshydrogéné et de maniètre de départ n'ayant pas réagi qui peut être présente est avantageusement purifié par chromatographie à l'aide de. gel de silice, dta- lumine activée et analogues ou si on le désire, à l'aide de chromatogrammes sur papier descendants.
Après séparation du produit déshydrogéné de la matière de départ n'ayant pas réagi, le produit peut être purifié davantage, si on le désira par-. recristallisation dans en solvant, tel que l'acé- tate d'éthyle, les mélanges acétate d'éthyle et d'éther de pétrole, et analogues.
