La/présente invention est relative à. un nouveau pro- cédé pour la préparation de nouveaux produits thérapeuti- ques pour le traitement d'affections allergiques ; elle con- cerne plus particulièrement un nouveau procédé pour la culture de bactéries sélectées destinées à la préparation d'un concen- tré complexe en tant que produit du métabolisme bactérien, concentré qui, comme il résulte,des preuves fournies par la pratique, permet de traiter avec succès toutes espèces d'af- fections allergiques, ainsi que l'asthme et même le rhumatisme.
Le présente invention a principalement pour objet d'établir un procédé nouveau pour la préparation d'un concentré

complexe en tant que produit du métabolisme bactérien, ce concentré étant capable de traiter avec succès des affec- tions allergiques, l'asthme et le rhumatisme et étant ca- ractérisé en ce qu'il comporte les opérations qui consistent: à préparer un milieu de culture à base de bouillon, de viande, de sang ou de plasma humains et de glucose;à ensemencer ce milieu de culture avec un mélange de bactéries sélectéesdu groupe comprenant les staphylocoques, les streptocoques, les pneu- mocoques, les microcoques du catarrhe, et analogues; et à laisser le métabolisme bactérien s'opérer jusqu'à la consom- mation pratiquement totale des substances nutritives du milieu de culture.
Plus particulièrement, l'objet de la présente inven- tion consiste à établir un nouveau procédé pour la prépara- tion d'un concentré complexe en tant que produit de métabo- lisme bactérien, ce procédé comprenant la combinaison des trois opérations, à savoir : la préparation d'un milieu de culture spécifique à base de bouillon de viande, de sang humain et de glucose; l'ensemencement de ce milieu de cul- ture avec les bactéries sélectées mentionnées plus haut et la poursuite du métabolisme bactérien dans le milieu de cul- ture précité ; finalement, la séparation et l'élaboration du concentré complexe produit par ce métabolisme bactérien.
Selon une caractéristique de l'invention, le procédé dont il est question plus haut comprend les opérations qui consistent à mélanger un bouillon de viande avec le-chlorure de sodium et la peptone bactériologique; à neutraliser le mélange avec une solution de carbonate de sodium; et à sté- riliser à une température voisine mais non supérieure à 125 C, en effectuant simultanément la précipitation des constituants' coagulables, de protéines étrangères et de sels insolubles

fermés par la noutralisation à la température appliquée.
Selon une autre caractéristique de l'invention, le procède indiqué plus haut comprend les opérations consis- tant à ajouter le glucose après la première stérilisation .mentionnée plus haut; à laisser la masse se refroidir; ajuster ensuite le pH .de la masse à environ 6,8 à 7,2;et . stériliser ensuite nouveau à une température quelque peu inférieure, soit une. température d'environ 110-115 C.
Selon une autre caractéristique de la présente in- vention, le procédé indiqué plus haut défini ci-après com- prend l'incorporation, au milieu de culture, de sang humain hémolyse, dans l'eau distillée stérilisée, après la seconde stérilisation susdite et après refroidissement du milieu de culture jusqu'à la température' normale du'sang.
Selon une autre particularité'de l'invention, le pro- cédé dont il est question plus haut et qui a été défini dans le paragraphe précédent, comprend l'incorporation, au milieu de culture, de substances telles que des vitamines et des ,sels inorganiques,.capables de stimuler la croissance de bactéries.
Selon une caractéristique subsidiaire de la présente invention, le procédé dont il est question plus haut et qui a été défini dans les. paragraphes précédents, comporte l'en- semencement du milieu de culture avec des bactéries du groupe spécifié ci-dessus et obtenu à partir de souches.développées dans la région géographique dans laquelle le produit appelé à Stre préparé par le procédé selon l'invention doit être employé.
Selon une autre caractéristique de l'invention, le procédé dont il est question plus haut et qui a été défini dans les paragraphes précédents, est caractérisé par le fait

qu'on laissera métabolisme bactérien se poursuivre dans le milieu de culture en maintenant la température de ce dernier à environ 38 à 38,5 C pendant environ 3-4 jours, de préfé- rence avec agitation périodique et,éventuellement, en répé- tant l'ensemencement de manière que le métabolisme bactérien se poursuive jusqu'à la consommation pratiquement totale des substances-nutritives du milieu de culture, assurant ainsi la formation d'un concentré complexe produit par le métabo- lisme bactérien susdit.
Selon une autre caractéristique de l'invention, le pro- cédé dont il est question plus haut et qui $ été indiqué dans les paragraphes précédents comprend, à titre d'opérations fi- nales visant à séparer et à élaborer le concentré complexe précité, la stérilisation du'milieu de culture traité, à l'aide d'une solution dàcide phénique dans le camphre ; décantation du milieu de culture ainsi .stérilisé; la purifi- cation du liquide décanté; la stérilisation finale dû-liquide .purifié; et un contrôle de stérilité pendant plusieurs jours, après quoi le produit peut être introduit dans des ampoules, ou si on le désire, déshydraté par lyophilisation.
Le produit¯ nouveau ainsi obtenu est un composé concen- tré complexe d'un pH d'environ 5,5, produit par métabolisme bactérien et dont la composition chimique n'a pas encore pu être déterminée. Ce produit, applique.dans :plus de 1. 000 cas par injection sous-cutanée, s'est montré très efficace en permettant de traiter avec succès toutes espèces d'allergies, l'asthme et même le rhumatisme. L'effet anti-allergique de ce produit (contrairement au phénomène dit de Burky) consiste à insensibiliser les personnes allergiques vis-à-vis de la plupart des substances allergènes, et cela en dépit de l'ab- sence d'une quelconque de ces substances dans le nouveau produit selon la présente invention.

Un bouillon de viande animale a été préparé de la manière habituelle et additionne de chlorure de sodium et de peptone bactériologique, ce bouillon étant ensuite neu- tralisé avec du carbonate de sodium en solution normale.
Le mélange a été ensuite stérilisé pendant 15 minutes en- viron dans des récipients d'une capacité ne dépaaseant pas 1 litre, à une température voisine maisnon supérieur à 125 C, de manière à provoquer la précipitation simultanée des constituants coagulables, des protéines étrangères et des sels insolubles formés par la neutralisation à la tem- pérature appliquée.
On laisse refroidir le bouillon de viande ainsi pré- paré jusqu'au voisinage de la température ambiante et l'on ajoute, à 100 parties en volume de ce bouillon, 2 parties en volume de glucose. A ce mélange on ajoute de préférence une ou plusieurs vitamines et des sels organiques capables d'agir comme agent stimulant la croissance des bactéries, par exemple :

Le pH de ce milieu de culture est ajusté d'une manière connue en soi à 7,0-7,2. Ce milieu de culture est ensuite stérilisé à 115 0 et, après avoir été refroidi à environ 38-38,5 C, est additionné de deux parties en volume de sang humain hémolyse dans l'eau distillée stérilisée.
Le mili. de culture ainsi préparé et qui possède la température indiquée plus haut, est ensemencé avec un mélange de germes sélectés comportant deux ou plusieurs membres du groupe composé de staphylocoques, de streptocoques, de pneu- mocoques, de microcoques du catarrhe, le métabolisme bacté- rien du milieu de culture ensemencé étant effectué dans des récipients qui exposent une superficie relativement impor- tante du milieu de culture, en maintenant la température à 38-38,5 C pendant 4 jours environ, avec agitation périodique, jusqu'', la consommation pratiquement totale des substances nutritives du milieu de culture, après quoi la stérilisation est opérée à l'aide d'une solution à.65% d'acide phénique dans le camphre,

Ensuite, le concentré complexe produit par le métabolisme bactérien est séparé par décantation et est filtré ou purifié d'une autre manière connue en soi. La stérilité du produit purifié est contrôlée pendant plusieurs jours, ce' produit étant ensuite introduit dans des ampoules, en contrôlant strictement la stérilité..
Afin de préserver le produit ainsi obtenu pendant un laps de temps indéfini, il est avantageux de le soumettre à la lyophilisation.

complex as a product of bacterial metabolism, this concentrate being capable of successfully treating allergic conditions, asthma and rheumatism and being characterized in that it comprises the operations which consist of: preparing a culture based on broth, human meat, blood or human plasma and glucose; inoculating this culture medium with a mixture of bacteria selected from the group comprising staphylococci, streptococci, pneumococci, catarrh micrococci, and analogues; and allowing bacterial metabolism to proceed until substantially all of the nutrients from the culture medium are consumed.
More particularly, the object of the present invention consists in establishing a new process for the preparation of a complex concentrate as a product of bacterial metabolism, this process comprising the combination of the three operations, namely: the preparation of a specific culture medium based on meat broth, human blood and glucose; the inoculation of this culture medium with the selected bacteria mentioned above and the pursuit of bacterial metabolism in the above culture medium; finally, the separation and elaboration of the complex concentrate produced by this bacterial metabolism.

that bacterial metabolism should be allowed to continue in the culture medium while maintaining the temperature of the latter at about 38 to 38.5 C for about 3-4 days, preferably with periodic agitation and, optionally, repeating seeding so that the bacterial metabolism continues until the practically total consumption of the nutrients of the culture medium, thus ensuring the formation of a complex concentrate produced by the aforesaid bacterial metabolism.
According to another characteristic of the invention, the process referred to above and which has been indicated in the preceding paragraphs comprises, as final operations aimed at separating and preparing the aforementioned complex concentrate, the sterilization of the treated culture medium, using a carbolic acid solution in camphor; decantation of the culture medium thus sterilized; purification of the decanted liquid; the final sterilization of the purified liquid; and a sterility check for several days, after which the product can be introduced into ampoules, or if desired, dehydrated by lyophilization.
The new product thus obtained is a complex concentrated compound with a pH of about 5.5, produced by bacterial metabolism and the chemical composition of which has not yet been determined. This product, applied in: more than 1,000 cases by subcutaneous injection, has been shown to be very effective in successfully treating all kinds of allergies, asthma and even rheumatism. The anti-allergic effect of this product (unlike the so-called Burky phenomenon) consists in making people who are allergic to most allergenic substances insensitive, despite the absence of any of the these substances in the new product according to the present invention.
