In het bijzonder werd daar ook beschreven hoe men zoodanige derivaten kan bereiden, die in het ringskelet minstens één vrije hydroxylgroep aan het koolstofatoom C3 bevatten en in de zijketen aan het koolstofatoom C21 daarentegen een door aoyleering of veraethering, eventueel met een gemakkelijk afsplitsbare alkylrest, besoherm- de hydroxylgroep of een halogeenatoom dragen ; dealgemeene formule van dergelijke verbindingen is dus

Er werd nu gevonden, dat men deze verbindingen door voorzichtige oxydatie, bijv. met chroomzuur, kan omzetten in de overeenkomstige, aan het koolstofatoom C21 gesubstitueerde, verzadigde of onverzadigde derivaten van pregnaan-dion-3,20, doordat de vrije hydroxylgroepen, speciaal die aan het koolstof atoom C3, tot ketogroepen worden geoxydeerd. Zulke oxyda-' ties zijn voor derivaten, die aan het koolstofatoom C21 niet gesubstitueerd zijn, herhaaldelijk beschreven. Men kon echter niet van te voren weten, dat deze ook zouden kunnen worde,n toegepast op de derivaten, die aan het koolstofatoom C21 als beschreven zijn gesubstitueerd, omdat deze verbindingen zeer

gevoelig zijn voor velé oxydatiemiddelen en bijv. door ammoni- akale zilveroplossing reeds in de koude snel in de zijketen worden geoxydeerd.
Uiterste voorzichtigheid is vooral noodig als het ringskelet een dubbele binding bevat, speciaal aan het koolstofatoom C5, daar de oxydatie dan met nevenreaoties verloopt.
In dit geval kan het aanbeveling verdienen de dubbele bindingen volgens op zichzelf bekende methoden door additie van halogeen of halogeenwaterstof tijdelijk te beschermen en na de oxydatie de dubbele binding te regenereeren, bijv. door behandeling met reduotiemiddelen; eventueel kan daarbij de dubbele binding verschuiven. Deze werkwijze is echter alleen toe te passen bij die derivaten, die aan het koolstofatoom C21 een beschermde hydroxylgroep dragen. Bevindt zich daar een halogeenatoom, dan kan men niet zoo te werk gaan omdat bij het verwijderen van de halogeenatomen uit het ringskelet het halogeenatoom in de zijketen aanleiding geeft tot nevenreaoties.
In het geval, dat men te maken heeft met derivaten van het éénmaal onverzadigde pregneen-5-ol-3-on-20 kan de gewenschte omzetting worden bereikt, onder gelijktijdige vervanging van het halogeenatoom aan het koolstofatoom C21, door een aoyloxyl- of een hydroxylgroep op de volgende wijze : Men addeert halogeen, oxydeert en verwarmt daarna met zouten van organische of zwakke anorganische zuren,tot het halogeen in de zijketen door acyloxyl of hydroxyl is vervangen. Gelijktijdig wordt in het ringskelet een mol. halogeenwaterstof afgesplitst onder vorming van een gehalogeneerd onverzadigd keton. Het halogeenatoom aan het koolstofatoom C6 kan daarna door reductie worden verwijderd.
Deze werkwijze heeft echter geen voordeelen als men tot polyketonen wil komen, die aan hetkoolstofatoom C21 een zuurstofrest dragen ; dat geval kan men beter direct van de derivaten met een beschermde hydroxylgroep uitgaan.

Bij de verzadigde derivaten daarentegen levert de vervanging van het 21-halogeen-atoom door een hydroxyl- of een beschermde hydroxylgroep geen moeilijkheden op. De polyketonen met een vrije hydroxylgroep aan het koolstofatoom C21 krijgt men door verzeeping van de overeenkomstige verbindingen met een door acyleering of veraethering beschermde hydroxylgroep, dus door hydrolyse of alcoholyse, waarbij sterk alkalische middelen dienen te worden vermeden. Sterke zuren kunnen meestal wel worden gebruikt. De hydroxy-21-ketonen kunnen ook uit de halogeen-81-ketenen worden bereid door inwerking van zouten van organische of zwakke anorganische zuren bij aanwezigheid van water of alcoholen. Om de boven besproken redenen komt dit echter alleen voor de verzadigde derivaten in aanmerking.
De vrije hydroxylgroep van het koolstof atoom C21 kan naderhand weer veresterd of veraetherd worden.
Alle genoemde reacties kunnen met voordeel worden uitgevoerd in aanwezigheid van oplos- of verdunningsmiddelen. Bovendien is het niet noodig de als tusschenproducten optredende verbindingen te isoleeren.
Ter verduidelijking zal de werkwijze voor een eenvoudig geval door formules worden geïllustreerd en wel voor de bereiding van aan het koolstofatoom C21 gesubstitueerde pregneen- 4-dion-3.20-derivaten. Een voorbeeld voor de bereiding der genoemde verbindingen volgt daarna.

Men gaat bijv. uit van aceetoxy-21-pregnean-5-ol-3-on-20 ( formule II, R = aoetyl ), addeert broom en behandelt het gevormde dibromide ( III ) met oxydatiemiddelen, bijv. chroom- zuur, waarbij het dibroomketon ( IV ) ontstaat. Dit levert met reductiemiddelen het onverzadigde keton ( V ), dat bijv. door inwerking van zuur onder verplaatsing van de dubbele binding in het isomere onverzadigde keton ( VI ) overgaat. Door

verzeeping krijgt men daaruit het vrije hydroxy-di keton (VII).
De voor de onverzadigde halogeen-21-ketenen mogelijke omweg wordt door de formules ( VIII ) en volgende aangegeven.
Na verzadiging van de dubbele binding met broom ( IX ) wordt geoxydeerd tot ( X ) en dit levert door behandeling met natriumacetaat enz. de ester van een onverzadigd broomketon met de waarschijnlijke formule ( XI ). Hieruit kan dan met reductiemiddelen het broomatoom worden afgesplitst, waarbij het diketon ( VI, R = acyl ) ontstaat.
Op analoge wijze kunnen pregnaanderivaten met meer dan één vrije hydroxylgroep in het ringskelet en halogeen of een beschermde hydroxylgroep aan het koolstofatoom C21 worden omgezet in de overeenkomstige polyketonen.
De verkregen verbindingen kunnen worden gebruikt voor de bereiding van geneesmiddelen.

1 g acestoxy-21-pregneen-5-o1-3-on-0 ( kristallen met smeltpunt 180 - 182 C (geoorr.) ) wordt opgelost in 5 cm3 chloroform, waarna bij 0 C een oplossing van 0,45 g broom in chloroform wordt toegevoegd,'waarvan eerst de laatste druppels een blijvende geelkleuring veroorzaken. Spoedig daarna wordt het mengsel in vacuo droog gedampt en het residu in weinig ijsazijn opgelost ; deze oplossing wordt een oplossing van 0,75 g chroomtrioxyde in 30 cm3 ijsazijn toegevoegd, waarna men het geheel 12 uur bij kamertemperatuur laat staan. Hierna giet men het mengsel in water uit en schudt het daarbij neerslaande broomketon met aether uit. De aether-oplossing wordt met wat water gewasschen en korten tijd gedroogd.
Daarna voegt men 2 g zinkstof en 1 g watervrij natriumacetaat toe; de aether wordt nu bij 60 badtemperatuur onder schudden af gedestilleerd en het residu wordt onder herhaald schudden, nog gedurende ongeveer 30 minuten op dezelfde temperatuur verwarmd, maar in

elk geval zoodanig, dat/een proefje in water uitgegoten en met aether uitgeschud, broomvrij is geworden. Dan wordt het mengsel in aether opgenomen, de oplossing wordt gefiltreerd, met water gewassohen, gedroogd en ingedampt. Het gekristalliseerde residu wordt gedurende 10 minuten met ijsazijn tot zacht koken verwarmd, daarna in vacuo droog gedampt.
De kristallen kunnen worden gezuiverd door sublimeering in hoogvacuum ( een badtemperatuur van 190 geeft bij een druk van 0,02 mm en voldoende groot verwarmingsoppervlak al een voldoende destillatiesnelheid, voor kleine hoeveelheden), daarna volgt omkristalliseeren uit weinig aoeton onder toevoeging van petroleumaether.
Men verkrijgt zoo ongeveer 0,7 g zuiver aceetoxy-21-pregneen- 4-dion-3.20 als kleurlooze naalden, die bij verwarming bij ongeveer 60 C ondoorschijnend worden en bij 158 - 1600 C ( geoorr.) smelten. Na goed drogen is hun samenstelling C23H32O4.
De oplossing in aethanol of methanol reduceert bij kamertemperatuur snel ammoniakale' zilveroplossing en vertoont in het ultraviolet-,spectrum de band bij ongeveer 240 m , die voor de [alpha]- ss-onverzadigde ketonen karakteristiek is. De verbinding 'is goed oplosbaar in ijsazijn, aethanol, methanol, aoeton, dioxaan en benzeen; vrij goed in aether en moeilijk in petroleumaether en water.
Bij de reductie met zink kan men ook vanaf het begin sterker verhitten, waardoor het achteraf verhitten met ijsazijn voor het verschuiven van de dubbele binding komt te vervallen.
Natuurlijk kan men ook andere reduotiemiddelen gebruiken. Zoo gelukt de debromeering ook heel goed door een uur met overmaat natriumjodide in ijsazijn te verwarmen, waarbij dezelfde verbinding ontstaat. Indien men aethanol als oplosmiddel gebruikt dan wordt gemakkelijk een deel van het acetyl aan het koolstofatoom C21 verzeept onder vorming van een vrije hydroxylgroep.

VOORBEELD 2 :
Aan een oplossing van 0,2 g van het bovenbeschreven aceet-

oxy-21-pregneen-4-dion-3.20 in 10 oma aethanol worden 10 em.3 water en 1 cm3 geconcentreerd zoutzuur toegevoegd. Het mengsel wordt 40 minuten onder terugvloeiing gekookt. Na afkoeling wordt bij kamertemperatuur in vaeuo ingedampt tot een troebeling optreedt, waarna men laat kristalliseeren. De goed ontwikkelde kleurlooze kristallen worden afgezogen, eerst met sterk verdunde aethanol, dan met water gewassohen en aan de lucht gedroogd. Door indampen wordt uit de moederloog de rest verkregen. Bij verwarmen worden de kristallen van het pregneen- 4-ol-21-dion-3.20 even boven 100 C ondoorsohijnend en smelten bij 137 - 139 C. Na zorgvuldig drogen is hun samenstelling C21H30O3.
De oplossing reduceert ammoniakale zilveroplossing in de koude en vertoont in het ultraviolet-absorptie-spectrum de " cholestenon "- band. De oplosbaarheid is analoog aan die van-het acetaat, echter in oplosmiddelen zonder hydroxylgroepen meestal iets geringer. Uit weinig aceton, onder toevoeging van petroleum-aether worden goed gevormde karakteristieke lange naalden verkregen; soms ook driehoeken of afgeknotte driehoeken. In een goed vacuüm kan men kleine hoeveelheden zonder ontleding sublimeeren, waarbij 190 en 0,02 mm druk reeds voldoende zijn als het verwarmingsoppervlak groot genoeg is.
Op geheel analoge wijze kan dezelfde verbinding uit andere esters worden bereid en ook door aoyleering weer in esters worden omgezet, terwijl met alkyleeringsmiddelen aethers ontstaan, waarbij sterk alkalische reacties moeten worden vermeden.

met 0,08 g broom in chloroform behandeld. Na indampen in vacuo wordt het residu zonder verwarmen opgelost in de noodige hoeveelheid ijsazijn, waarna een oplossing van 0,15 g ohroomtrioxyde in 7 cm3 ijsazijn wordt toegevoegd. Na 8 uur staan bij kamertemperatuur giet men het mengsel in water, schudt met aether uit. De aether-oplossing wordt met water gewasschen, boven natriumsulfaat gedroogd en na toevoeging van 0,5 g zinkstof en wat natriumacetaat op een waterbad ingedampt.
Het residu wordt na toevoeging van een beetje aethanol nog een uur op een kokend waterbad verwarmd, waarbij behalve debromeering ook verschuiving van de dubbele binding en afsplitsing van de triphenylmethylrest plaats vindt. Men verwijdert de aethanol in vaouo, voegt aether toe, filtreert, wascht de oplossing met water en soda-oplossing, droogt en dampt in. Het residu kan, om de afsplitsing van de triphenylmethylrest volledig te maken, nog eens met waterige alcohol en zoutzuur worden verwarmd en daarna in vacuo worden droog gedampt. Het ontstane triphenyloarbinol kan dan door verdeeling van het reaotiemengsel tus- sahen waterige aethanol of 50 %-ige methanol en petroleumaether worden verwijderd.
Uit de waterige alcoholische oplossing wordt het pregneen-4-ol-21-dion-3.20 als in voorbeeld 2 afgescheiden.

0,22 g ohloor-81-pregneen-5-ol-3-on-30 ( kleurlooze naal- den met smeltpunt 162 - 164 C (geoorr.) ) worden met0,108 g broom in chloroform gebromeerd en daarna als boven met 0,2 g ohroomtrioxyde in ijsazijn gedurende 14 uur bij kamertemperatuur behandeld. Het mengsel wordt in water gegoten en met aether uitgeschud, waarna de aether-oplossing met water wordt gewasschen, gedroogd en bij 50 C ingedampt. Het residu wordt verscheidene uren met 3 cm3 ijsazijn en 0,8 g watervrij natri- umacetaat op een waterbad verwarmd. Daarna voegt men 0,5 g

zinkstof toe en verwarmt nog ongeveer een half uur, afhankelijk van de kwaliteit van het zink, maar in elk geval zoo lang, dat een proefje, dat in water wordt gegoten en met aether uitgeschud, nagenoeg halogeenvrij is.
Dan verdunt men met veel aether, filtreert, wascht met water en soda-oplossing en dampt droog. Het residu wordt bij voorkeur eerst in hoogvacuum gedestilleerd, waarna een scheiding volgt door adsorptie, bijv. aan actief aluminiumoxyde uit een mengsel van petroleum-aether en benzol en elutie met aether en aceton. Daarna wordt omgekristalliseerd zooals in voorbeeld 1. De verkregen kristallen zijn identiek met de daar beschrevene. Voor de omzetting van verzadigde halogeen-21-ketonen met natriumacetaat in ijsazijn, enz. heeft het voordeel nog iets sterker te verhitten, bijv.
3 uur op 130 C. Men krijgt dan direct de aceetoxy-21-ketonen in zuiveren toestand. Bij de dibroomderivaten echter kan men in het algemeen beter niet te sterk verhitten.

1 g of acestoxy-21-pregnene-5-o1-3-one-O (crystals with m.p. chloroform is added, the last drops of which first cause a permanent yellow coloration. Soon after, the mixture is evaporated to dryness in vacuo and the residue dissolved in a little glacial acetic acid; to this solution, a solution of 0.75 g of chromium trioxide in 30 ml of glacial acetic acid is added and the whole is allowed to stand at room temperature for 12 hours. The mixture is then poured into water and the bromocetone which precipitates in the process is shaken out with ether. The ether solution is washed with some water and dried for a short time.
Then 2 g of zinc dust and 1 g of anhydrous sodium acetate are added; the ether is now distilled off at 60 bath temperature with shaking and the residue is heated at the same temperature for about 30 minutes with repeated shaking, but in

After careful drying, their composition is C21H30O3.
The solution reduces ammoniacal silver solution in the cold and shows the "cholestenone" band in the ultraviolet absorption spectrum. The solubility is analogous to that of the acetate, but usually slightly lower in solvents without hydroxyl groups. treated with 0.08 g of bromine in chloroform. After evaporation in vacuo, the residue is dissolved without heating in the necessary amount of glacial acetic acid, after which a solution of 0.15 g of chromium trioxide in 7 ml of glacial acetic acid is added. After standing at room temperature for 8 hours, the mixture is poured into water and shaken with ether. The ether solution is washed with water, dried over sodium sulfate and, after adding 0.5 g of zinc dust and some sodium acetate, evaporated on a water bath.
The residue is heated for another hour on a boiling water bath after adding a little ethanol, during which time, in addition to debromination, the double bond is shifted and the triphenylmethyl residue is split off. The ethanol is removed in vacuo, ether is added, filtered, the solution is washed with water and soda solution, dried and evaporated. To complete the cleavage of the triphenylmethyl residue, the residue can be heated again with aqueous alcohol and hydrochloric acid and then evaporated to dryness in vacuo. The triphenyloarbinol formed can then be removed by partitioning the reaction mixture between aqueous ethanol or 50% methanol and petroleum ether.
The pregnene-4-ol-21-dione-3.20 is separated from the aqueous alcoholic solution as in Example 2.
