Abstract
The importance of hand hygiene in health care and community settings is undoubted, with
numerous studies demonstrating the association between proper hand hygiene and reductions in
both rates of nosocomial infections and rates of infectious illnesses in communities. Virus
transfer via human hands is a major route of infection inside and outside medical settings. The
present invention relates to novel broad spectrum sanitizer composition which comprises citric
acid, urea, iodine,combination of essential oils, and isopropyl alcohol. The instant invention has
virucidal activity.

Description
FIELD OF THE INVENTION The present invention relates to novel broad spectrum sanitizer composition and process of manufacture thereof. Specifically, the present invention relates to a novel sanitizer composition comprises of alcohol, citric acid, urea, iodine, clove oil, lavender oil, carbopol and pharmaceutical water which can act on the different types of bacteria, fungi and viruses including envelope and non-envelope virus, includes influenza virus, corona virus and ebola virus. The composition has synergistic effect against different bacteria and viruses. The present application also describes a method of preparation for the same.
BACKGROUND OF THE INVENTION The importance of hand hygiene in health care and community settings is undoubted, with numerous studies demonstrating the association between proper hand hygiene and reductions in both rates of nosocomial infections and rates of infectious illnesses in communities. Hand hygiene is also an infection control procedure that can be rapidly adopted by the public and healthcare workers in the case of emerging and re-emerging infectious diseases to limit the spread of viruses by breaking the transmission.
The present disclosure generally relates to antimicrobial hand sanitizers including alcohols that are effectivein killing microorganisms while providing a moisturizing benefit to the user's skin. More particularly, the alcohol-basedhand sanitizers include a high internal phase emulsion which allows moisturizers or skin protectants such as emollientsand/or silicones to be stably incorporated into the sanitizer.
Virus transfer via human hands is a major route of infection inside and outside medical settings. The efficiency of virus transfer from hands to hands (direct transmission) or via contaminated surfaces (indirect transmission) is closely connected with virus persistence. Viruses of the respiratory tract like the non-enveloped coronavirus can persist on surfaces for several days.
Serious infectious disease outbreaks and hospital-acquired infections are threats to human health. Effective vaccines and drug products are often not available, but antiseptic products for hand washing can help to control the spread of disease.Multiple hand hygiene agents are currently available including plain non-medicated soap, medicated hand washes with various active ingredients, and waterless or alcohol-based hand rubs. It is recognized that some antiseptics and alcohol-based hand rubs are characterized by resistant bacteria and efficacy gaps.
A cleaning composition for hand hygiene therefore would ideally possess broad antiviral spectrum covering non-enveloped in addition to enveloped viruses offering increased protection against persistent, unexpected, or unknown viruses.
Research has been carried out in this area which discloses the formulation possessing alcohol and organic acid. The prior artsare summarized as below.
For example, the patent literatures JP-A3168075 and JP-A6322171 individually disclose ethanol preparations for food preservation comprising ethanol, and an organic acid or a salt thereof, etc.
The patent literatures JP-A115734 and JP-A2002253188 individually describe that microbicide comprising an alcohol such as ethanol and an organic acid can be used for hand wash and hand disinfection.
The patent literature JP-A2004107667 discloses a sanitizing hand cleanser that can be used without water, comprising about 60 to 90 % by weight of an organic alcohol, about 5 to 35% by weight of one or more silicone-based materials, about 5% by weightor less of one or more humectants, about 12% by weight or less of one or more additives for maintaining the skin pH, and one or more thickeners at such a concentration that the viscosity of the cleanser is within the range of 100 to 10,000 cp at about 25°C.
The patent literature JP-A7298862 discloses an alcohol preparation for reducing a population of microorganisms at low temperature, which is used for refrigerated or frozen foods, etc. and which comprises ethyl alcohol and phytic acid as essential ingredients.
The patent literature JP-B3761199 describes a method for disinfection, comprising contact with a 0.05 to 3% by weightaqueous solution of citric acid to inactivate a hepatitis virus B. In this method, lactic acid, malic acid and/or tartaric acidmay be further added to the above solution. The patent literature JP-A6314702 discloses a broad spectrum virucide comprising atleast 70% by weight of ethanol and/or propanol, and 0.5 to 5% by weight of a short chain organic acid. The virucide inquestion has a short chain organic acid especially with 2 to 6 carbon atoms and is effective against a wide range ofviruses, such as polioviruses, vaccinia viruses, SV40-viruses, and adenoviruses. The literature also describes that thisvirucide can be used for hand disinfection as an example of its application.
EP 2135507 which discloses the composition comprising lactic acid, citric acid, zine containing compound and ethanol/isopropyl alcohol. According to inventor, such composition has antiviral effect.
Hand cleaning is important for the prevention of virus transmission in the health care system and environment. The development of broad-spectrumsanitizer composition with activity against enveloped and non-enveloped viruses is limited due to a small number of permissible active ingredients able to inactivate viruses.
The present invention over the preparation which possess virucidal effect.
Accordingly, there remains a need for improved disinfecting compositions that are environmentally sustainable, completely botanical, or natural, biodegradable, non-toxic and capable of providing a broad disinfecting spectrum, and that utilize relatively low concentrations of a limited number of naturally occurring essential plant oils. There is also a need for improved essential oils-based compositions that disperse the essential oils well in a water carrier, that do not require wiping or rinsing off after being applied to inanimate surfaces, and that are stable, have a long shelf life and are affordable and aesthetically appealing to consumers.
Antimicrobial cleaners and hand sanitizers are now in widespread use in the United States, as well as other countries around the world. Many products are effective against bacteria. However, most have limited effectiveness against fungi and virus. Thus, there is a need for effective antimicrobial compositions that are non-toxic and effective against a range of pathogenic agents, including bacterial, viruses and fungi. The present invention provides such compositions.
OBJECT OF THE INVENTION An object of the present invention is to provide a composition that has a virucidal activity and an excellent fast-acting effect.
The main object of the present invention is to provide novel broad spectrum sanitizer composition.
Another object of the present invention is to provide novel broad spectrum sanitizer composition which comprises alcohol, citric acid, urea, iodine, essential oils,and pharmaceutical water.
Another object of the present invention is to provide novel broad spectrum sanitizer composition which comprises citric acid, urea, and iodine useful in the control of the virus spread.
DETAILED DESCRIPTION OF THE INVENTION The present invention relates to novel broad spectrum sanitizer composition which applicable to use for hands and skin, which has a virucidal activity.
The present inventors found that novel broad spectrum sanitizer composition comprising citric acid; urea, iodine, clove oil, lavender oil, carbopoland isopropyl alcohol as highly effective against virusesand excellent fast-acting effect. Moreover, the proposed composition provides moistening effect to skin.
While the invention has been described and illustrated with reference to certain embodiments thereof, those skilled in the art will appreciate that various adaptations, changes, modifications, substitutions, deletions, or additions of procedures and protocols may be made without departing from the spirit and scope of the invention. The present invention can be more readily understood by reading the following detailed description of the invention and study of the included examples. It must be noted that, as used in the specification and appended claims, the singular forms such as "a", "an", and "the" include plural referents unless the content clearly directs otherwise.
The novel broad spectrum sanitizer compositionof the present invention comprises (a) isopropanol, (b) clove oil, (c) citric acid, (d) clove oil, (e) lavender oil. Such novel broad spectrum sanitizer composition of the present invention is highly effective against viruses. Further, the composition of the present invention has an excellent virucidal activity,
The alcohol of the present invention, a microbicidal ingredient of the composition, is isopropyl alcohol. This composition is advantage over the ethanol-based composition which are not effective and known for misuse.
The antiviral surface composition of the present invention comprises specific organic acids, i.e.,citric acid, which produce a synergistic effect with isopropyl alcohol. Therefore, the composition has an enhanced fast-acting and persistent effect.
Essential oils are also widely used in the food industry. For example, clove oil or clove bud oil is used as a compounded oil for the flavoring of pastry, in special sauces and as a condiment. The dried product is used directly in compounded infusions for liqueurs and bitters.
Furthermore, clove oil preparations are commonly accepted in the Ayurvedic system, as a safe and accepted medicine.
Antibacterial activity of clove oil has been reported againstV. cholarae, Gartner'sBacillusand SwineErysipelasbacteria andBrucella andMycobacterium, Trichophyton, AchorionandEpidermophyton.Activityof clove oil volatile fractions is reported by S. Agnihotri and A. D. B. Vaidya againstE. coli, S. aureus, Bacillus subtilis, S. typhi, P. aeruginosa, S. Flexner andP. vulgaris.
Frequent use of disinfectants with high alcohol concentration easily causes skin roughness on hands, which results in less frequent handwashing. In this respect, the novel broad spectrum sanitizer composition of the present invention does not produce such a problem since it possessesmoistening effect. The composition of the present invention may comprise any known moisturizer that is usually added. Examples of such a known moisturizer include, for example, propylene glycol, glycerol and any other of acceptable agent which retains moisture.
These compositions kill or inactivate a broader range of pathogens (e.g., bacteria, viruses, fungi) than do conventional antimicrobial compositions. For example, the compositions described herein are more effective against acid-labile viruses such as adenovirus types 2, 5 and 8. In one embodiment, there is provided an antimicrobial composition comprising:
• (a) An alcohol component that is miscible with water and constitutes about 50% to 90% of the composition; • (b) Water, which constitutes about 10% to 50% of the composition; • (c) An acid component sufficient to maintain the pH below about 5, which constitutes about 0.1-10%, or about 0.1-5%, of the composition; • (d) Essential oils; and • (d) A multivalent cation which constitutes about 0.01% to 5% of the composition. All percentages are by weight, with reference to the weight of the pure component or metal, not including any water (for the non-water components), solvent, or counter-ion (in the case of metals). Each of these components is described in more detail below.
Preparation method The novel broad spectrum sanitizer composition of the present invention can be obtained by mixing all the ingredients, if needed heat-dissolving an ingredient that is in a solid state at ordinary temperature, and usually adjusting the pH of the mixture.

Some illustrative non-limiting examples of the present invention are described below. The examples should not be considered as limiting the scope of this invention in any way, as these examples and other equivalents thereof will become apparent to those versed in the art, in the light of the present disclosure, and the claims.

This novel broad spectrum sanitizer composition possessingisopropyl alcohol, citric acid and urea shown to have virucidal activity. In another embodiment, the present invention uses the nanotechnology for modifying each component of the formulation for better results.
Claims (3)
1. A novel broad spectrum sanitizer composition which applicable to use for hands and skin, which has a virucidal activity said composition comprising isopropanol, clove oil, citric acid, clove oil, and lavender oil.
2. The composition as claimed in claim 1 further comprises any known moisturizer that is usually added like as propylene glycol, glycerol and any other of acceptable agent which retains moisture.
3. The composition as claimed in claim 1 comprising:
(a) An alcohol component that is miscible with water and constitutes about 50% to 90% of the composition;
(b) Water, which constitutes about 10% to 50% of the composition;
(c) An acid component sufficient to maintain the pH below about 5, which constitutes about 0.1-10%, or about 0.1-5%, of the composition;
(d) Essential oils; and
(e) A multivalent cation which constitutes about 0.01% to 5% of the composition.
