Abstract
The invention relates to a method for the in vitro selection of binders capable of binding to a target molecule, from a molecule library. According to the inventive method, immobilised target molecules are brought into contact with a plurality of molecules bound to a reference complex, so that target molecules can react with possible binders and form a linkage from the reference complex, binding agent and target molecule. Non-bound constituents can then be separated and a force is applied to the linkages such that a link in the linkage is dissolved, and the binders bound to the target molecules and or complementary binding agents thereof are identified and/or amplified. Said reference complex consists of two constituents, the binding force thereof being selected in such a way that, when a tractional force is applied, the binding force of the two reference constituents is smaller than the binding force of a specifically bound molecule.
