A ligand (L) is determined by mixing a test sample with (a) a fluorescer (F) which does not react (chemically or immunologically) with L and (b) a reagent system (R) which does not react with F and which, in presence of L, changes the transmission properties of the soln. within a wavelength which overlaps the excitation and/or emission wavelength bands of F. The soln. is then irradiated to excite F and the intensity of fluorescence determined to provide a measure of L concn. R is a turbidimetric or esp. chromogenic reagent.
