Verfahren zur Herstellung von zur Injektion geejgneten Cholesterinpräparaten.
Wie aus der neueren Literatur hervorgeht, liegt in der therapeutischen Praxis das Bedürfnis vor, Cholesterin in zu Injektionen geeignete Lösungen zu bringen (vergl. Grimm, Deutsche med. Wochenschrift" 1910, Seite 176). Das einzige, bisher in Betracht kommende Lösungsmittel für Cholesterin ist Olivenöl, dessen Lösungsvermögen jedoch so gering ist, dass man nicht mehr als etwa 3"/oige Losungen damit erhalten kann (vergl. Morgenroth und Reicher, Charité-Annalen 33, Separat Seite 2).
Es wurde nun gefunden, dass die Ester höherer Fettsäuren, das Olivenöl inbegriffen, schon in sehr geringen Mengen das Vermögen besitzen, den Schmelzpunkt der physiologisch wichtigen Cholesterinester (vergl. Hoppe-Seylers Zeitschrift für physiologische Chemie", Bd. 21, Seite 331 u. ff.) so stark herabzndrücken, dass derartige Gemische unter Körpertemperatur zu vollkommen klaren, homogenen Flüssigkeiten schmelzen und, einmal ge- schmolzen, längere Zeit bei Zimmertemperatur flüssig bleiben. Auf diese Weise lassen sich Präparate erhalten, mittels deren dem Körper grössere Mengen Cholesterin subkutan oder per os zugeführt worden können, ohne dass er durch indifferente Stoffe unnötig belastet wird.
Eine innige Mischung von 2 Teilen Cholesterinölsäureester mit 1 Teil Ölsäureäthylester beginnt bei 290 C zu schmelzen, eine solche mit 1 Teil Palmitinsäuremethylester bei 310, während der reine Cholesterinölsäureester bei 40 bis 45 schmilzt. Als brauchbar für den vorgenannten Zweck erweisen sich auch Mischungen von Cholesterinölsäureester mit Olivenöl oder Phenylpalmitat.

As can be seen from the recent literature, there is a need in therapeutic practice to bring cholesterol into solutions suitable for injections (cf. Grimm, Deutsche med. Wochenschrift "1910, page 176). The only solvent for cholesterol that has been considered so far is olive oil, but its dissolving power is so low that you cannot get more than about 3% solutions with it (cf. Morgenroth and Reicher, Charité-Annalen 33, separate page 2).
It has now been found that the esters of higher fatty acids, including olive oil, have the ability, even in very small quantities, to reduce the melting point of the physiologically important cholesterol esters (cf. Hoppe-Seylers Zeitschrift für Physiologische Chemie ", Vol. 21, Page 331 u. ff.) so much that such mixtures melt into completely clear, homogeneous liquids below body temperature and, once melted, remain liquid for a long time at room temperature. In this way, preparations can be obtained which give the body larger amounts of cholesterol subcutaneously or can be supplied per os without being unnecessarily burdened by indifferent substances.
An intimate mixture of 2 parts of cholesterol oleic acid ester with 1 part of oleic acid ethyl ester begins to melt at 290 ° C., one with 1 part of palmitic acid methyl ester at 310 ° C., while the pure cholesterol oleic acid ester melts at 40 to 45 ° C. Mixtures of cholesterol oleic acid esters with olive oil or phenyl palmitate also prove to be useful for the aforementioned purpose.
