Die Erfindung betrifft ein Verfahren zur Herstellung neuer Cephalosporanderivate der Formel

Erfindungsgemäss gelangt man zu den Verbindungen der Formel (I), indem man eine Verbindung der Formel

worin Rund Rl obige Bedeutungen besitzen und R2 für die Methyl- oder Aminogruppe steht, mit einem Jodierungsmittel umsetzt.
Das erfindungsgemässe Verfahren kann nach bekannten Jodierungsreaktionen durchgeführt werden, beispielsweise mi einem Trialkyljodsilan.
Die erfindungsgemässe Umsetzung der Verbindungen der Formel (II) kann beispielsweise so vorgenommen werden, dass eine Verbindung der Formel (II) in einem unter den Reaktionsbedingungen inerten Lösungsmittel gelöst wird, vorzugsweise in Dichlormethan, 1, 1 bis 1, 5 Äquivalente Trialkyljodsilan, z. B. Trimethyljodsilan, zugegeben werden und die Reaktion bei Temperaturen von - 20 bis +50 C, vorzugsweise bei 0 bis 45 C, insbesondere jedoch zwischen 15 und 30 C, durchgeführt wird. Es ist überraschend, dass eine Verbindung der Struktur von 3-Jodmethyl-N, 0-bistrialkyl- silyl-7-aminocephalosporansäure stabil ist und in reiner Form isoliert werden kann.
Bei einigen ähnlichen Strukturen, die in der Literatur beschrieben werden, erfolgt exzessive Selbstalkylierung im Stickstoff, und die Verbindungen können nicht in reiner Form erhalten werden (DE-OS 3212900). Die Tatsache, dass die erfindungsgemäss hergestellten N, 0-Bis-trialkylsilyl-3-jodmethyl-7-aminocepha- losporansäuren in Lösung und in fester Form stabil sind, kann der sterischen Hinderung der Trialkylsilylgruppe am Stickstoff zugeschrieben werden. Es ist deshalb essentiell, reine Bissilylverbindungen vorliegen zu haben, da das Vorliegen von Monosilylverbindungen zu exzessiver Zersetzung und Polymerisation führt.
Bistrimethylsilyl-3-jodmethyl-7-aminocephem (3)-4-carbonsäuren können durch Entfernen des überschüssigen Lösungsmittels und Trialkyljodsilans im Vakuum in reiner Form isoliert und mittels des NMR-Spektrums charakterisiert werden.
Die als Ausgangsprodukte benötigten Verbindungen der Formel (II) können erhalten werden, indem man Verbindungen der Formel

Dieses Verfahren kann beispielweise folgendermassen ausgeführt werden :
Eine 7-Aminocephem (3)-4-carbonsäure der Formel (III), vorzugsweise 7-ACA, wird in einem unter den Reaktionsbedingungen inerten Lösungsmittel suspendiert und in Gegenwart eines Überschus- ses an Silylierungsmittel rückflusserhitzt. Als Lösungsmittel für diese Silylierungsreaktion kommen halogenierte Kohlenwasserstoffe in Frage, wie beispielsweise Dichlormethan, Chloroform, 1, 2-Dichlor- äthan, 1, 1, 2-Trichloräthan und Tetrachloräthylen, organische Nitrile, wie Acetonitril, Propionitril,
Nitroalkane, wie Nitromethan, oder Sulfone, wie Sulfolan.
Die Silylierungsreaktion kann entweder bei hochsiedenden Lösungsmitteln durch exzessives Erhitzen auf die entsprechenden Temperaturen oder bei niedriger siedenden Lösungsmitteln durch eine spezielle Katalyse einer organischen Sauer- stoffsäure, z. B. Quadratsäure, Trifluoressigsäure und deren Gemische, durchgeführt werden. Als
Silylierungsmittel bei dieser Reaktion können Hexamethyldisilazan allein oder im Gemisch mit Tri- methylchlorsilan, Bistrimethylsilylharnstoff, N-Trimethylsilyldialkylamine oder monosilylierte Acetami- de bzw. Gemische dieser Reagenzien, verwendet werden. Zur Charakterisierung der silylierten Ver- bindungen wird üblicherweise das überschüssige Silylierungsmittel im Vakuum abgezogen, der kri- stalline Rückstand in einem deuterierten Lösungsmittel gelöst und NMR-spektroskopisch charakteri- siert.
Es ist essentiell, dass die Bissilylierung zu 100% verläuft, da geringe Mengen an Monosilyl- derivat die weitere Umsetzung erheblich stören.
Die Cephalosporanderivate der Formel (I) können leicht in 3-substituierte 7-Aminocephem (3) - - 4-carbonsäuren überführt werden. 3-substituierte 7-Aminocephem (3)-4-carbonsäuren sind wertvolle
Zwischenverbindungen zur Herstellung von Cephalosporan-Wirkstoffen. Diese Verbindungen können in 3-Stellung entweder durch einen Mercapto-Heterocyclus substituiert sein, oder sie können als quartäre Ammoniumsalze vorliegen, wobei in diesem Fall die Carbonsäuren als Betaine vorliegen.
In Spezialfällen können derartige Zwischenverbindungen auch in 3-Stellung an einem Stickstoff alkylierte Heterocyclen tragen, wobei in diesem Fall keine Betainform vorliegt.
Der Stand der Technik zur Herstellung von derartigen Verbindungen weist im wesentlichen drei Verfahren auf, die jedoch alle mit gewissen Nachteilen behaftet sind. So beschreibt die DE-OS 2804896 die Umsetzung von 7-Aminocephem (3) -4-carbonsäuren mit Lewis-Säuren bzw. mit Protonensäuren im Überschuss, wobei vorzugsweise Bortrifluorid verwendet wird. Es ist bekannt, dass derartige Reagenzien äusserst toxisch sind, Umweltprobleme verursachen und ausserdem ss-Lactame unter stark sauren Bedingungen im protischen Medium instabil sind.
Ein zweites Verfahren beschreibt die Austauschreaktion in 3-Stellung in wässerigen Puffersystemen (US-PS Nr. 3, 516, 997). Dieses Verfahren gibt geringe Ausbeuten, da unter den verwendeten Bedingungen (7-ACA + heterocyclisches Thiol) Cephalosporine in Pufferlösungen zwischen PH 5 und 9 zu einem erheblichen Teil zersetzt und daher die Titelverbindungen in ungenügender Reinheit erhalten werden.
Ein drittes Verfahren beschreibt die Acylgruppen-Abspaltung der Seitenkette in 7-Position bei einem Cephalosporin, das schon den richtigen Substituenten in 3-Stellung trägt (z. B. US-PS Nr. 4, 369, 313). Zur Herstellung der in diesem Patent verwendeten Ausgangsmaterialien muss jedoch 7-ACA zunächst in 7-Stellung acyliert, dann der Austausch in 3-Stellung vorgenommen und zum Schluss die Seitenkette in Position 7 abgespalten werden. Gesamthaft gesehen ist diese Reaktionsfolge von der Ausbeute ab 7-ACA als unwirtschaftlich zu betrachten.
Alle diese Verfahren werden von dem unter Verwendung der Verbindungen der Formel (I) durchgeführten Verfahren bei weitem übertroffen. Dieses Verfahren geht von kommerziell leicht erhältlichen Verbindungen der Formel (III) aus und führt dieses Ausgangsmaterial durch eine

Ein weiterer besonderer Vorteil dieses Verfahrens ist es, dass einige nach a) umgesetzte 7-Ami- noceph (3) -4-carbonsäurederivate schwer lösliche Hydrojodide bilden, die aus Wasser in sehr reiner Form anfallen und kristallin isoliert werden können. Ein bevorzugtes Beispiel hiefür ist (6R, 7R)-7- - Amino-3- (1-pyridiniummethyl)-cephem (3)-4-carboxylat. Hydrojodid.
Im nachfolgenden Beispiel, das die Erfindung erläutern soll, ohne jedoch ihren Umfang einzuschränken, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel : 7-Trimethylsilylamino-3-jodmethylcephem (3)-4-carbonsäuretrimethylsilylester :
Die N. O-bissilylierte 7-ACA beispielsweise hergestellt wie unter A) oder B) beschrieben wird in 125 ml trockenem Dichlormethan gelöst, die Lösung auf 0'gekühlt und 1, 87 ml 3-Trimethyljodsilan zugetropft. Man lässt die Lösung auf Raumtemperatur kommen und rührt 2 h nach. Nach

In special cases, such intermediate compounds can also carry heterocycles alkylated in the 3-position on a nitrogen, in which case no betaine form is present.
The prior art for the production of such connections essentially has three methods, but all of them have certain disadvantages. For example, DE-OS 2804896 describes the reaction of 7-aminocephem (3) -4-carboxylic acids with Lewis acids or with excess protonic acids, preferably boron trifluoride. It is known that such reagents are extremely toxic, cause environmental problems and, in addition, ss-lactams are unstable in the protic medium under strongly acidic conditions.
A second method describes the exchange reaction in the 3-position in aqueous buffer systems (US Pat. No. 3,516,997). This process gives low yields since under the conditions used (7-ACA + heterocyclic thiol) cephalosporins in buffer solutions between PH 5 and 9 decompose to a considerable extent and therefore the title compounds are obtained in insufficient purity.
A third method describes the acyl group cleavage of the side chain in the 7-position in the case of a cephalosporin which already bears the correct substituent in the 3-position (e.g. US Pat. No. 4,369,313). To produce the starting materials used in this patent, however, 7-ACA must first be acylated in the 7-position, then exchanged in the 3-position and finally the side chain in position 7 must be split off. Viewed overall, this reaction sequence can be regarded as uneconomical from the yield of 7-ACA.
