Die oben genannten als Ausgangsverbindungen eingesetzten, optisch aktiven Anthracyclinone sind neu, obwohl einige der entsprechenden Racemate von C. M. Wong et al., Canad. J. Chem. 49, 2712 (1971) und Canad. J. Chem. 51, 466 (1973) beschrieben wurden. Die vorgeschlagene Synthese der Racemate erfolgt nach folgendem Reaktionsschema :

der racemischen Form erhalten, während die natürlichen Aglycone optisch aktiv sind und die 7S : 9S-Konfiguration aufweisen (entsprechend der Nomenklatur von Cahn, Ingold und Prelog, Experientia, 1956,12, 81).
Um die natürlichen Anthracyclinantibiotika und/oder deren Analoga, die im Ring D substituiert sind, durch Kondensation derAglyconemit einem geeigneten Daunosaminderivat zu erhalten, wäre es daher vorzuziehen, optisch aktiveAglycone mit der 7S : 9S-Konfiguration zu verwenden, anstatt das Daunosaminderivat mit dem racemischen Aglycon zu kondensieren und anschliessend das (-)-Daunosaminyl- (+)-anthracyelinonvom (-)- Daunosamin- (-)-anthraeyclinon durch ein umständliches und zeitraubendes Verfahren, wie fraktionierte Kristallisation oder Chromatographie, zu trennen.
Es ist bekannt (Eliel, Stereochemistry of Carbon Compounds, Seite 55, McGraw-Hill, 1962), dass die optische Trennung von Alkoholen am besten durch Salzbildung der Emiphthalate mit einer optisch aktiven Base durchgeführt werden kann. Jedoch macht die geringe Löslichkeit der Anthracyclinone (VIII) und ihrer Derivate in den meisten Lösungsmitteln dieses Verfahren praktisch bedeutungslos. Ausserdem sollte vom praktischen und ökonomischen Standpunkt aus die Trennung im frühesten Stadium der Synthese durchgeführt werden, um das Syntheseverfahren am optisch aktiven Zwischenprodukt mit der gewünschten Konfiguration durchzuführen. Das erste Zwischenprodukt mit einem Chiralzentrum ist das Ketolderivat (II), jedoch war vor der Erfindung kein Verfahren zur optischen Trennung derartiger Derivate bekannt.
Zur optischen Trennung eines Ketols der allgemeinen Formel (II) wird (-)-l-Phenyläthylamin mit racemischem 1, 4-Dimethoxy-6-hydroxy-6-acetyltetralin in einem geeigneten Lösungsmittel, wie Acetonitril, zu diastereoisomeren Schiffschen Basen umgesetzt, welche durch Kristallisation getrennt werden und aus welchen die enantiomeren Ketole (II) durch Säurebehandlung gewonnen werden. Die Trennung erfolgt scharf und völlig unerwartet, da keine früheren Fälle der Trennung von Ketonen über Ketimine aus der Literatur bekannt sind (Eliel, loc. eit. Seite 56), wahrscheinlich weil Schiffsche Basen gewöhnlich ziemlich instabil sind und sich leicht während der Kristallisation zersetzen.
Ausserdem wurde gefunden, dass die optisch aktiven Ketole (II) in das racemische Ketol (III) wieder übergeführt werden können, weshalb es möglich ist, das racemische Keton (II) in die gewünschte optisch aktive Form in sehr hoher Ausbeute überzuführen, u. zw. über die Trennung des racemischen Ketols (II) mit (-)-l-PhenyläthyIamin und nachfolgende Rückführung des unerwünschten Isomeren zurück in die racemische Form und Wiederholung der optischen Trennung.
In dem oben erwähnten, von C. M. Wong et al vorgeschlagenen Reaktionsschema werden für mehrere Reaktionsschritte stark saure Bedingungen angewendet, die Racemisierung des Chiralzentrums über ein

Im Hinblick auf die leichte Racemisierung des Ketolchiralzentrums ist die Isolierung der Verbindungen (IV) und (IV") in optisch aktiver Form ausserordentlich überraschend.
Überraschenderweise können die optisch aktiven Verbindungen (IV) und (IV") in die optisch aktiven Formen des Ketals (V) ohne Racemisierung übergeführt werden, indem mit Äthylenglykol in Anwesenheit von p-Toluolsulfonsäure bei erhöhten Temperaturen behandelt wird. Die Behandlung des optisch aktiven Ketals (V) mit N-Bromsuccinimid in Tetrachlorkohlenstoff ergibt ein labiles 7-Bromketal, das bei Behandlung mit Methanol eine Mischung von 7 (S)-und 7 (R)-Methyläthern ergibt.
Diese Methyläther können in Stellung 6 und 11 in Anwesenheit von wasserfreiem Aluminiumchlorid demethyliert werden, wobei ihre optische Aktivität erhalten bleibt, wobei eine Mischung von 7 (S)-und 7 (R)-Methoxyanthracyclinonen erhalten wird, die mit Trifluoressigsäure und anschliessend mit Natriumbicarbonat behandelt wird, wobei die entsprechenden 7 (S)-und 7 (R)-Hydroxyderivate erhalten werden. Diese werden durch Kristallisation oder durch Chromatographie auf Silikagel getrennt, wobei die geeigneten optisch aktiven reinen Verbindungen (VIII') (7S : 9S-

Anwesenheit einer Spur einer starken Säure, wie Toluolsulfonsäure, in einer Vorrichtung behandelt, die mit einem Scheider versehen ist, um das bei der Reaktion gebildete Wasser zu sammeln, wobei das entspre-

chende optisch aktive Ketal (V) erhalten wird, das dann mit einer Wolframlampe am Rückfluss in Tetrachlorkohlenstoff mit N-Bromsuccinimid 5 bis 15 min lang unter Bildung des labilen 7-Bromketals bestrahlt wird. Letztere Verbindung wird nicht isoliert, sondern mit Methanol behandelt, wobei eine Mischung aus 7 (S)-und 7 (R)-Methyläthern (VI) erhalten wird.
Diese Mischung wird ohne Reinigung mit A luminiumchlo- rid in Benzol, Nitrobenzol oder Tetrachloräthan bei einer Temperatur von 5 bis 500C während 1 bis 10 h umgesetzt, wobei eine Mischung aus 7 (S)-und 7 (R)-Methoxyanthraeyclinonen (VII) erhalten wird. Durch Behandlung der Verbindungen (VII) mit Trifluoressigsäure bei 5 bis 300C während 8 bis 20 h und anschlie- ssend mit Natriumbiearbonat während eines kurzen Zeitraumes werden Anthracyclinone der Formel (VIII') zusammen mit ihrem 7 (R)-Epimer erhalten, die durch Kristallisation oder Chromatographie getrennt werden, wobei die reine Verbindung (VIII') (cis-7, 9-Dioldiastereoisomeres) erhalten wird.
Das 7 (R)-Epimere der Formel (VIIIIII) (trans-7,9-Dioldiastereoisomeren) wird isoliert und eventuell mit der Verbindung (VII) zur Trifluoressigsäurebehandlung in den Zyklus rückgeführt. Auf ähnliche Weise kann aus der (+)-Form des Ketols (II) unter Anwendung des obigen Verfahrens die Verbindung (VIII") (7R : 9R-Konfiguration) und deren 7(S)-Epimeres(VIII"") (7R: 9R-Konfiguration) erhalten werden.

Beispiel l : a) Trennung von 1, 4-Dimethoxy-6-hydroxy-6-acetyl-tetralin

Dieser Rückstand wurde in 400 ml Benzol gelöst. 8 g AlCl3 wurden zugesetzt und die Suspension 2 h lang bei 400C gerührt. Die Lösung wurde abgekühlt, 1050 ml 3%ige Oxalsäure wurden zugesetzt und Chloroform zur vollständigen Lösung zugegeben. Die

Eine Mischung von 7 (S)-und 7 (R)-Epimeren von 4-Demethoxy-7-desoxy-7-methoxydaunomycinon (VII, Konfiguration 9S, R 1= = =R = H) (1,5 g), hergestellt wie unter c) beschrieben, wurde in 60 ml Trifluoressigsäure gelöst und über Nacht bei Raumtemperatur stehengelassen.
Die Lösung wurde im Vakuum eingedampft, der Rückstand in 150 ml A ceton aufgenommen und 60 ml5%ige NaHCO wurden zugesetzt. Die Lösung wurde 30 min bei Raumtemperatur stehengelassen, dann mit Wasser verdünnt und wiederholt mit Chloroform extrahiert. Bei Abdampfen des Lösungsmittels verblieb ein Rückstand, der auf Silikagel chromatographiert wurde.
Bei Eluieren mit Chloroform wurden 0, 6 g 4-Demethoxy-daunomycinon (VIII',

In order to obtain the natural anthracycline antibiotics and / or their analogs substituted in ring D by condensation of the aglycones with a suitable daunosamine derivative, it would therefore be preferable to use optically active aglycones with the 7S: 9S configuration rather than the daunosamine derivative with the racemic aglycone to condense and then the (-) - daunosaminyl- (+) - anthracyelinone from (-) - daunosamine- (-) - anthraeyclinone by a cumbersome and time-consuming process such as fractional crystallization or chromatography to separate.
It is known (Eliel, Stereochemistry of Carbon Compounds, page 55, McGraw-Hill, 1962) that the optical separation of alcohols can best be carried out by salt formation of the emiphthalates with an optically active base. However, the low solubility of the anthracyclinones (VIII) and their derivatives in most solvents makes this process practically insignificant. In addition, from the practical and economical point of view, the separation should be carried out at the earliest stage of the synthesis in order to carry out the synthesis process on the optically active intermediate having the desired configuration. The first intermediate product with a chiral center is the ketol derivative (II), but prior to the invention no process for the optical separation of such derivatives was known.
For the optical separation of a ketol of the general formula (II), (-) - l-phenylethylamine is reacted with racemic 1,4-dimethoxy-6-hydroxy-6-acetyltetralin in a suitable solvent, such as acetonitrile, to give diastereoisomeric Schiff bases, which by Crystallization can be separated and from which the enantiomeric ketols (II) are obtained by acid treatment. The separation is sharp and completely unexpected, since no previous cases of separation of ketones via ketimines are known from the literature (Eliel, loc. Eit. P. 56), probably because Schiff bases are usually quite unstable and easily decompose during crystallization.
