Die Erfindung betrifft ein Verfahren zur Herstellung von neuem 3-Fluor-D-alanin oder seinen Deuteroanalogen, wie z. B. 2-Deutero-3-fluor-D-alanin und 2, 3, 3-Trideutero-3--fluor-D-alanin. Das 3-Fluor-D- - alanin und seine Deutero-Analogen sind starke antibakterielle Mittel und eignen sich zum Hemmen des Wachstums von grampositive und gramnegativen pathogenen Bakterien.

Wenn 3-Fluor-D-alaninverbindungen durch Zerlegung von Salzen einer optisch aktiven Säure mit einem Derivat der Carboxylgruppe des 3-Fluor-DL-alanins hergestellt werden, erhält man schliesslich die entsprechenden Derivate des 3-Fluor-D-alanins, wie (nach Zerlegung unter Verwendung von z. B. Weinsäure, Campher-10-sulfonsäure und Dibenzoyl-d-weinsäure als Zerlegungsmittel) den Benzylester von 3-Fluor- - D-alanin, D-2-Amino-3-fluorpropionitril, oder D-2-Amino-3-fluorpropionsäureamid.
Beispeil 1: N-1-Menthoxyacetyl-3-fluor-D-alanin wird mit wässeriger 6n Salzsäure, die zu Ver- flüchtigungszwecken etwas Äthanol enthält, auf Rückflusstemperatur erhitzt. Nach 18stündigem Kochen am Rückflusskühler wird die Lösung teilweise eingedampft und zweimal mit Äther extrahiert. Dann wird die wässerige Lösung zur Trockne eingedampft, wobei die überschüssige Salzsäure abgetrieben wird. Das als Rückstand hinterbleibende 3-Fluor-D-alanin-hydrochlorid wird in der geringstmöglichen Menge Wasser gelöst und die Lösung mit konzentrierter wässeriger Ammoniumhydroxydlösung auf einen pH-Wert von 4, 8 eingestellt.
Das Gemisch wird auf 00C gekühlt, worauf man durch Abfiltrieren optisch reines 3-Fluor-D-

Nach einer andern Verfahrensweise wird das 3-Fluor-D-alanin-hydrochlorid in Wasser gelöst und die Lösung durch ein stark saures Kationenaustauscherharz (in der Wasserstoffionenform) geleitet. Die Chlorionen werden aus der Ionenaustauschersäule mit Wasser ausgewaschen, dann wird die Säule mit verdünnter

wässeriger Ammoniumhydroxydlösung eluiert und das Eluat eingedampft. Das dabei ausfallende 3-Fluor-D- - alanin wird abfiltriert und getrocknet.
Das Ausgangsmaterial erhält man wie folgt : Zu einer Lösung von 20, 4 g 3-Fluor-DL-alanin in 200 ml
Wasser und 56ml 2, 5n-wässeriger Natronlauge werden unter schnellem Rühren 28, 0 g linksdrehendes Menth- oxyacetylchlorid zugetropft. Man setzt weitere wässerige 2, 5n-Natronlauge zu, um den PH-Wert zwischen
10 und 11 zu halten. Danngiesstman die Reaktionslösung langsamineishaltigeverdünnte wässerige Salzsäure.
Der Niederschlag wird abfiltriert und die gemischten Diastereoisomeren werden durch fraktionierte Kri- stallisation aus wässerigem Äthanol voneinander getrennt.
Beispiel 2 : Das wie nachstehend angegeben hergestellte Hydrochlorid des Benzylesters von 3-Fluor- -D-alanin wird hydrolysiert, indem man die wässerige saure Lösung 5 h zum Sieden erhitzt. Die Reaktion- lösung wird gekühlt und mit Methylenchlorid extrahiert. Dann wird die wässerige Lösung zur Trockne ein- gedampft, wobei die überschüssige Salzsäure abgetrieben wird. Das hinterbleibende 3-Fluor-D-alanin-hy- drochlorid wird in der geringstmöglichen Menge Wasser gelöst, der pH-Wert mit konzentriertem wässeri- gem Ammoniumhydroxyd auf 4, 8 eingestellt und der Niederschlag von 3-Fluor-D-alanin durch Abfiltrieren gewonnen. Fp. 166 bis 168 C (Zers.).
Das Ausgangsmaterial erhält man wie folgt : Eine Suspension von 20, 7 g 3-Fluor-DL-alanin in 200 ml
Benzylalkohol wird unter Hindurchleiten von trockenem Chlorwasserstoff auf 1000C erhitzt. Das Reaktion- gemisch wird auf 1000C gehalten, bis sich eine klare Lösung gebildet hat. Dann dampft man den Benzylalko- hol unter vermindertem Druck ab, löst das hinterbleibende Öl in 50 ml Wasser und setzt 200 ml Methylen- chlorid zu. Hierauf setzt man so lange festes Natriumcarbonat unter Rühren zu, bis das Aufbrausen aufhört.
Die Methylenohloridschicht wird abgetrennt und mit Wasser gewaschen. Dann wird die Methylenchlorid- schicht im Vakuum eingedampft, wobei der Benzylester von 3-Fluor-DL-alanin als zähflüssiges Öl hinter- bleibt.
10 g des so hergestellten Benzylesters von 3-Fluor-DL-alanin und 9, 0 g Dibenzoyl-d-weinsäure werden in 100 ml Äthanol miteinander vermischt und das Gemisch wird gelinde erwärmt, bis Lösung eingetreten ist.
Die Lösung wird zur Trockne eingedampft. Der Rückstand wird in der geringstmöglichen Menge heissen Äthanols gelöst und das Gemisch der Diastereoisomeren durch fraktionierte Kristallisation getrennt.
Das so erhaltene Dibenzoyl-d-weinsäuresalz des Benzylesters von 3-Fluor-D-alanin wird zu verdünnter wässeriger Salzsäure zugesetzt und das Gemisch fünfmal mit je 25 ml Methylenchlorid extrahiert, wodurch die Dibenzoyl-d-weinsäure aus der das Hydrochlorid des Benzylesters von 3-Fluor-D-alanin enthaltenden sauren wässerigen Lösung extrahiert wird.
Beispiel 3 : N-Benzoyl-3-fluor-D-alanin wird nach Zusatz von konzentrierter wässeriger Salzsäure 8 h auf Rückflusstemperatur erhitzt, wodurch die Benzylgruppe abgespalten wird.
Die Lösung wird zur Trockne eingedampft und der Rückstand in der geringstmöglichen Menge Wasser gelöst. Der pH-Wert der wässerigen Lösung wird mit konzentriertem wässerigem Ammoniumhydroxyd auf 4, 8 eingestellt, die Lösung gekühlt und der Niederschlag abfiltriert. Das rohe 3-Fluor-D-alanin wird 24 h bei 250C mit dem Fünffachen seines Volumens an Wasser verrührt. Durch Abfiltrieren erhält man praktisch reines 3-Fluor-D-alanin. Fp. 166 bis 1680C (Zers.).
Das Ausgangsmaterial erhält man wie folgt : 0, 1 mMol asymmetrischer Diphosphinrhodium-Katalysator (hergestellt nach "Chem. Comm. ", [1971], S. 481) wird in 50 ml eines Gemisches aus 1 Teil Benzol und 2 Teilen Äthanol gelöst und die Lösung mit 11, 0 o ;-Benzamido-ss-fluoracrylsäure versetzt. Das Gemisch wird bei 7 kg/cm2 und 2500 mit Wasserstoff umgesetzt. Die Reaktionslösung wird zur Trockne eingedampft und der Rückstand in Chloroform gelöst. Der Chloroformextrakt wird mit Wasser gewaschen, mit Holzkohle behandelt, filtriert und zur Trockne eingedampft.
B e i s p i e 1 4 : N-Benzoyl-2, 3-dideutero-3-fluor-D-alanin wird nach Zusatz von konzentrierter wässeriger Salzsäure 8 h auf Rückflusstemperatur erhitzt, wobei die Benzylgruppe abgespalten wird. Die Lösung wird dann zur Trockne eingedampft und der Rückstand in der geringstmöglichen Menge Wasser gelöst. Der pH-Wert derwässerigen Lösungwird mit konzentriertem wässerigem Ammoniumhydroxyd auf 4, 8 eingestellt, die Lösung gekühlt und der Niederschlag abfiltriert. Das als Filterrückstand erhaltene rohe 2, 3-Dideutero- - 3-fluor-D-alanin wird 24 h bei 25 C mit dem Fünffachen seines Volumens an Wasser gerührt. Durch Abfiltrieren erhält man praktisch reines 2, 3-Dideutero-3-fluor-D-alanin.
Das Ausgangsmaterial erhält man wie folgt : 0, 1 mMol asymmetrischer Diphosphinrhodium-Katalysator (hergestellt gemäss "Chem. Comm. Der Chloroformextraktwird mit Wasser gewaschen, mit Adsorptionskohle behandelt, filtriert und das Filtrat zur Trockne eingedampft. Man erhält N-Benzoyl-2, 3-dideutero-3- - fluor-D-alanin.
Beispiel 5 : 2-Isooyanato-3-fluor-D-propionsäuremethylester wird mit 100 ml Wasser und 50 ml

konzentrierter wässeriger Salzsäure versetzt und das Gemisch 2 h auf Rückflusstemperatur erhitzt. Unter diesen Bedingungen wird sowohl die Isocyanatgruppe als auch die Methylestergruppe durch Hydrolyse gespalten. Durch Eindampfen der wässerigen Reaktionslösung im Vakuum erhält man rohes 3-Fluor-D-alanin- - hydrochlorid (S-3-Fluoralanin-hydrochlorid).
Das Hydrochlorid des 3-Fluor-D-alanins wird in Wasser gelöst und die Lösung durch ein starkes Kationenaustauscherharz (inder Wasserstoffionenform) hindurchgeleitet. Die Ionenaustauschersäule wird zwecks Entfernung der Chlorionen mit Wasser gewaschen und dann mit verdünnter wässeriger Ammoniumhydroxyd-

Den als Ausgangsmaterial eingesetzten N-Carbonyl-3-fluor-D-alaninmethylester erhält man wie folgt : 50gDimethylmethylenmalonat (d. h. 2-Methoxycarbonylacrylsäuremethylester) werden bei einer Temperatur unter -200C zu 200 ml wasserfreiem Fluorwasserstoff zugesetzt. Nach dem Abdampfen des Fluorwasserstoffs setzt man 500 ml Äther zu. Die Ätherlösung wird mit Eiswasser gewaschen, getrocknet und filtriert. Das
Filtrat wird zur Trockne eingedampft und das als Rückstand hinterbleibende Öl im Vakuum destilliert. Man erhält praktisch reinen 3-Fluormethylmalonsäuredimethylester (d. h. 2-Methoxycarbonyl-3-fluorpropion- säuremethylester).
16, 4 g 2-Methoxycarbonyl- 3-fluorpropionsäuremethylester werden in 100 ml Methanol gelöst, und die Losungwird unter Innehaltung einer Temperatur von 00C mit 10 ml wässeriger 1n Natronlauge versetzt. Hie- durch erhält man eine wässerige-methanolische Lösung, die das Natriumsalz von 2-Carboxy-3-fluor-pro- pionsäuremethylester enthält. Diese Lösung wird mit verdünnter wässeriger Salzsäure neutralisiert, mit Chloroform extrahiertund die Chloroformlosung getrocknet und im Vakuum eingedampft. Man erhält 2-Carb- oxy-3-fluorpropionsäuremethylester.
15 g dieses Esters werden in einem Gemisch aus Methanol und Benzol gelöst, worauf man die stöchio- metrisch äquivalente Menge L-a-Methylbenzylamin zusetzt und die diastereoisomerenSalzedes 2-Carboxy- - 3-fluorpropionsäuremethylesters durch fraktionierte Kristallisation voneinander trennt. Das so erhaltene L-o'-Methylbenzylaminsalz von S-2-Carboxy-3-fluorpropionsäuremethylester wird in wässeriger Lösung mit der stöchiometrisch äquivalenten Menge wässeriger Salzsäure umgesetzt, die wässerige Lösung mit Chloroform extrahiertund der Chloroformextraktgetrocknet und im Vakuum eingedampft. Man erhält S-2-Carboxy- - 3-fluorpropionsäuremethylester.
3, 0 g S-2-Carboxy-3-fluorpropionsäuremethylester werden in 25 ml Aceton gelöst, und die Lösung wird bei 00C zunächst mit 2, 02 g Triäthylamin und dann mit überschüssigem Chlorameisensäureäthylester versetzt. Zu dem Gemisch setzt man 10 ml Wasser zu und rührt die Lösung 30 min bei 0 C, wobei man eine wässerige Acetonlösung erhält, die das entsprechende Carboäthoxyester-Mischanhydrid von S-2-Carboxy- - 3-fluor-propionsäuremethylester enthält. Diese Lösung wird mit 1, 3 g Natriumazid versetzt und das Gemisch weitere zwei Stunden bei-5 bis 00C gerührt. Das Reaktionsgemisch wird in eiskalte gesättigte wässerige Kochsalzlösung gegossen und das wässerige Gemisch mehrmals mit Äther extrahiert.
Die den S-2- - Azidocarbonyl-3-fluorpropionsäuremethylester enthaltende Lösung wird über wasserfreiem Natriumsulfat getrocknet, filtriert, mit 100 ml Dioxan versetzt, der Äther dann abgedampft und die Dioxanlösung vorsichtig (hinter einem Schutzfenster) zum Sieden erhitzt, wobei sich das Azid zersetzt und sich zu S-2-Isocyanato- - 3-fluorpropionsäuremethylester, d. h. N-Carbonyl-3-fluor-D-alaninmethylester, umlagert.
Wennman den S-2-Azidocarbonyl-3-fluorpropionsäuremethylesterin Äthanollösung zersetzt, bildet sich das entsprechende Äthylurethan des 3-Fluor-D-alaninmethylesters (Äthylurethan von S-3-Fluoralaninmethylester). Das letztere wird dann mit wässeriger Salzsäure zu 3-Fluor-D-alanin-hydrochlorid umgesetzt, welches, wie oben beschrieben, in 3-Fluor-D-alanin übergeführt wird.
Die optisch aktiven Verbindungen dieses Beispiels werden mitunter mit"R" bzw."S" gemäss den"IUPAC Tentative Rules for the Nomenclature of Organic Chemistry ; Section E Fundamental Stereochemistry", Journal of Organic Chemistry, Bd. 35 [1970], S. 2863 bis 2867, bezeichnet.
In ähnlicher Weise wird das Carboäthoxyester-Mischanhydrid von S-2-Carboxy-3-fluorpropionsäuremethylester (statt mit Natriumazid) mit Ammoniak zu S-2-Aminocarbonyl-3-fluorpropionsäuremethylester umgesetzt, der dann mit Natriumhypobromitzur Reaktion gebracht wird, wobei die Amidogruppe sich umlagert und, wenn man Dioxan als Lösungsmittel verwendet, S-2-Isocyanato-3-fluorpropionsäuremethylester, und wenn man Äthanol als Lösungsmittel verwendet, das entsprechende Äthylurethan von 3-Fluor-D-alanin, nämlich N- Äthoxycarbonyl-3-fluor-D-alanin, entsteht.

Hiebei werden die Isocyanatgruppe und die Nitrilgruppe durch Hydrolyse gespalten, und es entsteht 3-Fluor-D-alanin-hydrochlorid.
Nach einer ändern Methode erhält man D-2-Amino-3-fluorpropionitrilverbindungendurch Zerlegung der entsprechenden DL-2-Amino-3-fluorpropionitrilverbindungen. DL-2-Amino-3-fluorpropionitril selbst wird z. B. unter Verwendung eines optisch aktiven Zerlegungsmittels, wie Weinsäure oder Campher-10-sulfon- säure zerlegt, wobei man das optisch aktive Salz des D-2-Amino-3-fluorpropionitrils erhält, welches durch Behandeln mit wässerigem Ammoniak in das freie D-2-Amino-3-fluorpropionitril übergeführt wird. Das letztere wird aus der wässerigen Lösung mit Äther extrahiert.
Das D-2-Amino-3-fluorpropionitril wird dann bei Temperaturen unter 00C mit an Chlorwasserstoff angereicherter wässeriger Salzsäure (50% HC1) zu D-2-Amino-3-fluorpropionsäureamid umgesetzt udn das letztere mit konzentrierter wässeriger Salzsäure auf Rückflusstemperatur erhitzt, wobei man das Hydrochlorid des 3-Fluor-D-alanins erhält.

The starting material is obtained as follows: To a solution of 20.4 g of 3-fluoro-DL-alanine in 200 ml
Water and 56 ml of 2.5N aqueous sodium hydroxide solution are added dropwise with rapid stirring 28.0 g of levorotatory mentholoxyacetyl chloride. Another aqueous 2, 5N sodium hydroxide solution is added to the pH value between
10 and 11 to keep. Then, dilute aqueous hydrochloric acid containing ice is slowly poured into the reaction solution.
The precipitate is filtered off and the mixed diastereoisomers are separated from one another by fractional crystallization from aqueous ethanol.
EXAMPLE 2 The hydrochloride of the benzyl ester of 3-fluoro-D-alanine prepared as indicated below is hydrolyzed by heating the aqueous acidic solution to the boil for 5 h. The reaction solution is cooled and extracted with methylene chloride. The aqueous solution is then evaporated to dryness, the excess hydrochloric acid being driven off. The remaining 3-fluoro-D-alanine hydrochloride is dissolved in the smallest possible amount of water, the pH value is adjusted to 4.8 with concentrated aqueous ammonium hydroxide and the 3-fluoro-D-alanine precipitate is collected by filtration . The starting material is obtained as follows: A suspension of 20.7 g of 3-fluoro-DL-alanine in 200 ml
Benzyl alcohol is heated to 1000 ° C. while dry hydrogen chloride is passed through. The reaction mixture is kept at 1000C until a clear solution has formed. The benzyl alcohol is then evaporated off under reduced pressure, the oil which remains is dissolved in 50 ml of water and 200 ml of methylene chloride are added. Solid sodium carbonate is then added with stirring until the effervescence stops.
The methylene chloride layer is separated and washed with water. The methylene chloride layer is then evaporated in vacuo, leaving the benzyl ester of 3-fluoro-DL-alanine as a viscous oil.
10 g of the benzyl ester of 3-fluoro-DL-alanine thus prepared and 9.0 g of dibenzoyl-d-tartaric acid are mixed with one another in 100 ml of ethanol and the mixture is gently warmed until solution has occurred.
The solution is evaporated to dryness. The residue is dissolved in the smallest possible amount of hot ethanol and the mixture of diastereoisomers is separated by fractional crystallization.
The dibenzoyl-d-tartaric acid salt of the benzyl ester of 3-fluoro-D-alanine thus obtained is added to dilute aqueous hydrochloric acid and the mixture is extracted five times with 25 ml of methylene chloride each time, whereby the dibenzoyl-d-tartaric acid from the hydrochloride of the benzyl ester of 3 -Fluoro-D-alanine containing acidic aqueous solution is extracted.
Example 3: N-Benzoyl-3-fluoro-D-alanine is heated to reflux temperature for 8 h after the addition of concentrated aqueous hydrochloric acid, whereby the benzyl group is split off.
The solution is evaporated to dryness and the residue is dissolved in the smallest possible amount of water. The pH of the aqueous solution is adjusted to 4.8 with concentrated aqueous ammonium hydroxide, the solution is cooled and the precipitate is filtered off. The crude 3-fluoro-D-alanine is stirred for 24 hours at 250C with five times its volume of water. By filtering off, practically pure 3-fluoro-D-alanine is obtained. M.p. 166 to 1680C (dec.).

Evaporation of the aqueous reaction solution in vacuo gives crude 3-fluoro-D-alanine hydrochloride (S-3-fluoroalanine hydrochloride).

The resulting L-o'-methylbenzylamine salt of S-2-carboxy-3-fluoropropionic acid methyl ester is reacted in aqueous solution with the stoichiometrically equivalent amount of aqueous hydrochloric acid, the aqueous solution is extracted with chloroform and the chloroform extract is dried and evaporated in vacuo. S-2-carboxy- 3-fluoropropionic acid methyl ester is obtained.

H. N-carbonyl-3-fluoro-D-alanine methyl ester, rearranged.
If the S-2-azidocarbonyl-3-fluoropropionic acid methyl ester is decomposed in ethanol solution, the corresponding ethyl urethane of 3-fluoro-D-alanine methyl ester (ethyl urethane of S-3-fluoroalanine methyl ester) is formed. The latter is then reacted with aqueous hydrochloric acid to form 3-fluoro-D-alanine hydrochloride, which, as described above, is converted into 3-fluoro-D-alanine.

Another method is used to obtain D-2-amino-3-fluoropropionitrile compounds by decomposing the corresponding DL-2-amino-3-fluoropropionitrile compounds. DL-2-amino-3-fluoropropionitrile itself is z. B. using an optically active decomposition agent such as tartaric acid or camphor-10-sulfonic acid decomposed, the optically active salt of D-2-amino-3-fluoropropionitrile being obtained, which by treatment with aqueous ammonia in the free D- 2-Amino-3-fluoropropionitrile is converted. The latter is extracted from the aqueous solution with ether.
The D-2-amino-3-fluoropropionitrile is then reacted at temperatures below 00C with aqueous hydrochloric acid (50% HC1) enriched in hydrogen chloride to give D-2-amino-3-fluoropropionic acid amide udn the latter is heated to reflux temperature with concentrated aqueous hydrochloric acid, with the hydrochloride of 3-fluoro-D-alanine is obtained.
