Process for the production of new 1-oxo-isoindolines and their salts
The aim of the present invention is to produce new 3-substituted l-oxo-isoindolines and 2,3-substituted l-oxo-isoindolines of the general formula
EMI1.1
In this, X denotes H, Cl, O-alkyl, CH3 or OH, Y H, branched or unbranched alkyl (Ci-C, arylalkyl, alkylcycloalkyl or an alkylheterocyclic group, and R
EMI1.2
where n is 2-6, Ri is H, branched or unbranched alkyl, R, Rg either independently of one another each branched or unbranched alkyl or both together, together with the N atom, denote a heterocyclic ring, and Z denotes O or S.
These products are, under suitable reaction conditions, by condensation of the substituted or unsubstituted 3-hydroxyphthalimidine with amines of the formula
EMI1.3
wherein Z, n, Ri, R2 and Rs have the meanings given above.
By alkylation in position 2, the products in which Y has the meaning given above other than hydrogen are obtained.
If Ri is H, products of the following general formula are formed in the alkylation reaction
EMI1.4
(Y 'has the same meaning as Y, with the exception of hydrogen.)
After reduction, this results in products of the general formula I.
<Desc / Clms Page number 2>
If Y is not H but has one of the other meanings given, the products of the general formula I are more favorable by reaction of compounds of the general formula
EMI2.1
wherein X and Y 'have the meanings given above and Hal is halogen, obtained with amines of the formula II.
By reacting these products with organic or inorganic acids (e.g. hydrochloric, sulfuric, citric, tartaric acid or the like), the corresponding water-soluble salts are obtained.
Many of these products have interesting pharmacological properties and are therefore useful as antispasmodics, analgesics and antitussives in therapy.
The following examples are intended to illustrate the present invention without restricting it.
Example 1: 1-Oxo-3- (aminophenyl-p-ethoxy-pipoeridino) -isoindoline.

The precipitate obtained in this way is crystallized from ethanol (99.9%) and gives 1-oxo-3- (aminophenyl-p-ethoxy-pipoeridino) -isoindoline (melting point 180-181 ° C.).

The precipitate obtained in this way is crystallized from ethanol (99.9%) and gives 1-oxo-3- (aminophenyl-p-ethoxy-morpholino) isoindoline (melting point 130-131 ° C.).
Example 3: 1-Oxo-3- (aminophenyl-p-ethoxy-piperidino) -6-chloro-isoindoline.
1.3 g (0.078 mol) of 6-chloro-3-hydroxyphthalimidine are dissolved in 50 ml of N, N-dimethylformamide.
3.11 g (0.0156 mol) of p-aminophenyl-ethoxy-piperidine are added to the solution obtained in this way and the mixture is refluxed for 5 hours; then it is cooled and ice is added. The precipitate obtained in this way is crystallized from ethanol (50%) and gives 1-oxo-3- (aminophenyl-p-ethoxy-piperidino) -6-chloroisoindoline (melting point 160-161 ° C.).
