Verfahren zur Herstellung neuer Pyridazon- (3)-Verbindungen, deren Salzen bzw. quartären Trialkylammoniumsalzen
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen mit analeptischer, kardiovaskulärer, gefässerweiternder und blutdrucksteigernder Wirkung, die bei der menschlichen Therapeutik Anwendung finden.
Das erfindungsgemässe Verfahren zur Herstellung neuer Pyridazon- (3)-Verbindungen der allgemeinen Formel

worin R, Ri und R2 wie oben definiert sind bzw. deren Alkylester kondensiert, in einer zweiten Stufe das so erhaltene, in Stellung 4 hydroxylierte Pyridazinon in einem sauren Medium dehydratisiert und gegebenenfalls, wenn Rg ein Wasserstoffatom bedeutet, in das Reaktionsprodukt eine Gruppe- (CHX) n-Z, in welcher X, Z und n die obgenannte Bedeutung haben, einführt und erwünschtenfalls die erhaltenen Verbindungen in ihre Salze bzw. quartäre Trialkylammoniumverbindungen überführt.
Das zu kondensierende Hydrazin kann in Stellung 2 an das Stickstoffatom mit einer entsprechenden basischen Aminoalkylgruppe substituiert sein. Sollte jedoch das zu kondensierende Hydrazin nicht substituiert sein, dann wird das Reaktionsprodukt der ersten Stufe zweckmässig mit einem Aminoalkylhalogenid in Gegenwart von Natriumalkoholat zwecks Substituierung unter Erhitzen versetzt.
Die Kondensation wird in einem Lösungsmittel wie Butanol, das ein azeotropes Gemisch mit dem während der Reaktion ausgeschiedenen Wasser bildet, durchgeführt.
Die Dehydratisierung des in der ersten Stufe erhaltenen Pyridazinons erfolgt durch mässiges Erhitzen in einem sauren Medium, wobei anschliessend das entsprechende Pyridazon mittels Destillation erhalten wird.

Die nach dem erfmdung < gemässen Verfahren hergestellten Verbindungen sind besonders zur Behand- lung von kardiovaskulären Kollaps und von Atmungskollaps, von Arterienentzündungen und zur Be- kämpfung aller Beschwerden, für die eine periphere gefässerweiternde Wirkung erforderlich ist, geeignet.
Das erfindungsgemässe Verfahren wird in zwei Stufen durchgeführt.
1. Stufe :
Ein Hydrazin oder ein durch eine basische Aminoalkylgruppe an das Stickstoffatom in Stellung 2 substituiertes Hydrazin wird unter Cyclisierung mit einer oc-Hydroxyl-y-carbonyl-carbonsäure, d. h. mit einer y-Aldehydsäure oder einer y-Ketonsäure, versetzt. An Stelle vorgenannter Säuren können auch die Ester derselben zur Anwendung kommen. Im Falle eines in Stellung 2 nichtsubstituierten Hydrazins, wird die Substitution des aktiven Wasserstoffatoms mittels eines Aminoalkylhalogenids, in Gegenwart eines Natriumalkoholates (Methylat oder Äthylat), unter Erhitzen durchgeführt.
2. Stufe :
Das in der ersten Stufe erhaltene, in Stellung 4 hydroxylierte Pyridazinon wird in einer sauren Lösung unter mässigem Erhitzen dehydratisiert ; anschliessend wird das so gewonnene Pyridazon mittels Destilla- tion von Reaktionsgemisch getrennt.
Es ist hervorzuheben, dass die zur Anwendung kommenden Säuren mittels Kondensation bei Zimmer- temperatur von Alkylketonen mit dem Alkalisalz einer < x-Ketonsäure in einer alkoholischen Kaliumcarbonat- lösung hergestellt werden können. Anschliessend wird das Reaktionsgemisch neutralisiert und die so erhaltene Säure beispielsweise mit angesäuertem Äther entzogen. Ebenfalls ist es möglich, die Ester vor- genannter Säuren mittels Kondensation bei Zimmertemperatur von aliphatischen Aldehyden mit Äthyl- pyruvat, in Gegenwart eines Kondensationsmittels wie Diäthylamin, herzustellen. nichtsubstituierten oder entsprechend substituierten Hydrazin so durchgeführt, dass ein in Stellung 4 hydroxyliertes Pyridazinon erhalten wird.
Sollte das Hydrazin entsprechend substituiert sein, so findet die Kondensation in einem Lösungsmittel statt, beispielsweise in Butanol. Auf diese Weise ist es möglich, ein azeotropes Gemisch zu erhalten, so dass die zwei Wassermoleküle, die sich infolge der Kondensation bilden, leicht zu entfernen sind.
Die zweite Stufe des erfindungsgemässen Verfahrens, d. h. die Dehydratisierung, kann ohne Abscheidung des in der ersten Stufe erhaltenen Pyridazinons durchgeführt werden, wobei eine sehr hohe Ausbeute erreicht wird. Sollte jedoch ein freies Hydrazin zur Anwendung kommen, so wird das Wasserstoffatom des Stickstoffes des Pyridazons in Stellung 2 durch Reagieren mit einem Aminoalkylhalogenid in Gegenwart von Natriumalkoholat unter Erhitzen aminoalkyliert. Vorgenannte Aminoalkylation wird erst nach der Kondensation durchgeführt. Um eine bessere Verwendung der nach dem erfindungsgemässen Verfahren hergestellten Verbindung zu erreichen, kann ebenfalls das basische Pyridazon in ein Salz, beispielsweise in Hydrochlorid oder in ein Salz einer organischen Säure oder in ein Salz einer quartären Ammoniumverbindung, umgewandelt werden.
Zur näheren Erläuterung der Erfindung dienen folgende Beispiele. Die Erfindung ist jedoch nicht auf die dargestellten Beispiele beschränkt : Beispiel l : Herstellung von 2-Dimethylaminoäthyl-4-methyl-5-äthyl-3-pyridazon.

gebracht. Dabei wurden etwas mehr als 10 cm3 Wasser aus dem Reaktionsgemisch entfernt. Anschliessend wurde das Benzol unter vermindertem Druck abgedampft. Dem Reaktionsgemisch wurden 105 cm3 kristalli- sierte Essigsäure und 3 cm3 konz. Salzsäure zugefügt und das so erhaltene Gemisch 2 h lang im Trocken- schrankbei 1000 C gehalten. Es erfolgte anschliessend die VerdunstUng der Säuren unter vermindertem Druck.
Dabei wurde eine kristalline bräunliche Masse, die das 4-Methyl-5-äthyl-3-pyridazon darstellte, erhalten.
Vorgenannte Masse wurde unmittelbar in etwa 30 cm3 Äthanol aufgelöst, anschliessend gefiltert und das Filtrat zum Auskristallisieren gelassen.
Ausbeute : 71% (19, 7 g).
Das so gewonnene Produkt wurde zum zweiten Mal aus 25 cm3 Äthanol (Ausbeute 64% bzw. 17, 87 g) und darauf zum dritten Mal aus 20 cm3 Äthanol (Ausbeute 59% bzw. 16, 39 g) umkristallisiert. Das so

In einem Erlen-Meyer-Kolben wurden 0, 058 Mol (8 g) 4-Methyl-5-äthyl-3-pyridazon eingeführt, das in 43, 5 cm3 absoluten Alkohol aufgelöst wurde. Es erfolgte der Zusatz einer Natriumäthylatlösung, die aus 0, 061 Grammatomen (1, 4 g) metallischem Natrium und 43, 5 cm3 absolutem Alkohol hergestellt wurde. Anderseits wurden 0, 061 Mol (8, 7 g) Dimethylamino-chloräthan-hydrochlorid in etwa 43, 5 cm3 absolutem Alkohol aufgelöst.
Der letztgenannten Lösung wurde eine Natriumäthylatlösung (0, 061 Grammatome bzw. 1, 4 g metallisches Natrium in 43, 5 cm3 absolutem Alkohol) unter Abkühlung zugefügt. Dabei bildete sich sofort ein Niederschlag von Natriumchlorid und basischem Dimethylamino-chloräthan.
Das sich in Suspension befindende Gemisch von Dimethylamino-chloräthan und Natriumchlorid wurde unfiltriert der Lösung von 4-Methyl-5-äthyl-3-pyridazon und Natriumäthylat beigegeben. Das so erhaltene Reaktionsgemisch wurde bei Zimmertemperatur 1 h lang gerührt, darauf 8 h unter Rückfluss erhitzt und hernach stehengelassen, anschliessend auf einen Buchnerfilter filtriert und das im Filtrat enthaltene Lösungsmittel unter vermindertem Druck abgedampft. Der braune Rückstand wurde anschliessend bei einer Temperatur von 106 bis 120 C/0, 2 mm Hg destilliert.
Es erfolgte die Auflösung des so erhaltenen Produktes in Aceton, wobei in die Lösung gasförmige Salzsäure eingeblasen wurde. Es bildeten sich weisse Kristalle von 2-Dimethylaminoäthyl-4-methyl-5- äthyl-3-pyridazon-hydrochlorid, welche filtriert und mit Äther gewaschen wurden. Das vorgenannte rohe Reaktionsprodukt wurde in der kleinstmöglichen Menge absoluten Alkohols aufgelöst und aus Äther umkristallisiert. Auf diese Weise wurde das Hydrochlorid in Form perlmutterartiger, weisser Kristalle mit einem Schmelzpunkt von 160 C erhalten.
Andere Verbindungen, die nach dem erfindungsgemässen Verfahren hergestellt werden können, werden in nachstehender Tabelle genannt, wobei die Bedeutung der substituierten Radikale in der allgemeinen Formel und die Schmelzpunkte der Basen oder die Salze ebenfalls angegeben werden :

Beispiel 1 hergestellte Verbindung), die Verbindung Nr. 42-Morpholinoäthyl-4-methyl-5-äthyl-3-pyridazon hydrochlorid, die Verbindung Nr. 5 ss-2-Trimethyläthylammonium-4-methyl-5-äthyl-3-pyridazon- hydrojodid.
Die Verbindung Nr. 5 wurde leicht aus der nach Beispiel 1 erhaltenen Base wie folgt hergestellt :
In einem 100 cm3 Erlen-Meyer-Kolben wurden 0, 02 Mol (4, 2 g) der nach Beispiel 1 hergestellten Base in 42 cm3 Aceton eingeführt. Anschliessend erfolgte der Zusatz von 0, 021 Mol (2, 9 g bzw. 1, 31 cm3) Methyljodid. Darauf wurde filtriert und das Reaktionsprodukt aus 60 cm3 absolutem Alkohol umkristallisiert. Auf diese Weise wurde vorgenanntes Hydrojodid in Form von weissen Kristallen, die eine sehr gute Wasserlöslichkeit aufweisen und auch in heissem Alkohol löslich waren, erhalten. Die so hergestellte Verbindung besass einen Schmelzpunkt von 198 C.
Wie schon erwähnt, besitzen die nach dem erfindungsgemässen Verfahren hergestellten Verbindungen eine aussergewöhnlich analeptische, kardiovaskuläre, gefässerweiternde und blutdrucksteigernde Wirkung, insbesondere das nach Beispiel 1 hergestellte 2-Dimethylaminoäthyl-4-methyl-5-äthyl-3-pyridazon.
Dabei kann vorgenannte Verbindung entweder als 5-10%igue Lösung in 5-10 cm3 injizierbaren Ampullen oder als Tabletten mit einem Wirkstoffgehalt von 100 bis 500 mg zur Anwendung kommen.

The process according to the invention is carried out in two stages.

In a 100 cm3 Erlen-Meyer flask, 0.02 mol (4.2 g) of the base prepared according to Example 1 in 42 cm3 of acetone were introduced. This was followed by the addition of 0.021 mol (2.9 g or 1.31 cm3) of methyl iodide. It was then filtered and the reaction product was recrystallized from 60 cm3 of absolute alcohol. In this way, the aforementioned hydroiodide was obtained in the form of white crystals which have very good solubility in water and which were also soluble in hot alcohol. The compound so produced had a melting point of 198 C.
As already mentioned, the compounds produced by the process according to the invention have an extraordinarily analeptic, cardiovascular, vasodilating and blood pressure-increasing effect, in particular the 2-dimethylaminoethyl-4-methyl-5-ethyl-3-pyridazone produced according to Example 1.
The aforementioned compound can be used either as a 5-10% solution in 5-10 cm3 injectable ampoules or as tablets with an active ingredient content of 100 to 500 mg.
