Verfahren zur Herstellung von neuen 5H-Dibenz [ b, f ] azepinen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen N-heterocyclischen Verbindungen mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Derivate des 5H-Dibenz Eb, f lazepins und des 10, 11-Di- hydro-5H-dibenz f]azepins der allgemeinen Formel I :

worin R einen Alkyl-, Alkenyl-, Aryl- oder Aralkylrest, worin Benzolkerne durch Halogenatome substituiert sein können, R ein Wasserstoffatom oder einen Alkyl- oder Alkenylrest, Rs einen niederen Alkyl-oder Alkenylrest, X undX ein Wasserstoffatom oder zusammen eine zusätzliche Bindung und Z einen geradkettigen oder verzweigten Alkylenrest mit 2 - 6 Kohlenstoffatomen bedeuten, und ihre Salze mit anorganischen oder organischen Säuren wertvolle pharmakologische Eigenschaften, insbesondere antiallergische, spasmolytische,
Serotonin-antagonistische und Reserpin-antagonistische Wirksamkeit besitzen, sowie auch die Wirkung anderer Arzneistoffe, insbesondere von Narkotica, potenzieren.
In den Verbindungen der allgemeinenFprmel I ist Rl beispielsweise ein Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, Allyl-, Crotyl-, Methallyl-, Phenyl-, Benzyl-, ss-Phenyläthyl- oder y-Phenylpropylrest und R2 ein Wasserstoffatom oder einen der oben genannten Alkyl- und Alkenylreste.
R ist beispielsweise durch den Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek. Butyl-, n-Amyl-, Isoamyl-, Diäthylmethyl-, Allyl-, Methallyl- oder Crotylrest und Z z. B. durch den Äthylen-, Propylen-, Trimethylen-, 1-Methyl-trimethylen-, 2-Methyl-trimethylen-, 1, 3 - Dimethyl-trimethylen -, 2, 2-Dimethyl-trimethylen-, Tetramethylen-, Pentamethylen- oder Hexamethylenrest verkörpert.
Zur Herstellung der neuen Verbindungen der allgemeinen Formel I wird ein reaktionsfähiger Ester einer Verbindung der allgemeinen Formel II :

Je nach dem Siedepunkt des verwendeten Amins und des Lösungsmittels sowie der notwendigen Reaktionstemperatur ist die Umsetzung gegebenenfalls in geschlossenem Gefäss durchzuführen.
Insbesondere bei Umsetzungen mit primären Aminen, deren Siedepunkt im Bereich der Reaktionstemperatur liegt, kann auch ein Überschuss derselben als alleiniges Verdünnungsmittel dienen. Zu reaktionsfähigen Estern von Hydroxyverbindungen der allgemeinen Formel II gelangt man beispielsweise durch Umsetzung von Alkalimetallverbindungen von Verbindungen der allgemeinen Formel IV :

Umsetzung der oben genannten Alkalimetallverbindungen mit Alkylenoxyden und Behandlung der entstandenen Hydroxyalkylverbindungen mit anorganischen Säurehalogeniden, Arylsulfonsäurechloriden oder Methansulfochlorid, wobei man Halogenide, Arylsulfonsäureester bzw. Methansulfonsäureester von Verbindungen der allgemeinen Formel II erhält.
Als Beispiele von primären Aminen der allgemeinen Formel III seien Methylamin, Äthylamin, n-Propylamin, Isopropylamin, n-Butylamin, Isobutylamin, sek. Butylamin, tert. Butylamin, n-Amylamin, Isoamylamin, 3-Amino-pentan, tert. Amylamin, n-Hexylamin, n-Heptylamin, -Octylamin, Allylamin, Crotylamin und Methallylamin genannt.
Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, ss-Hydroxy-äthansulfonsäure, Äthandisulfonsäure, Essigsäure, Milchsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure und Mandelsäure bilden die Verbindungen der allgemeinen Formel I Salze, die zum Teil wasserlöslich sind.
Das nachfolgende Beispiel erläutert die Herstellung der neuen Verbindungen näher. Teile bedeuten darin Gew.-Teile, diese verhalten sich zu Vol. -Teilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel : a) 21,0 Teile 10-Methyl-5H-dibenz [b, f] azepin werden in 200 Vol. -Teilen abs. Benzol gelöst und mit 22 Teilen 1 - Chlor - 3 - brom - propan versetzt. Bei 60 - 620 wird eine Suspension von 5,6 Teilen Natriumamid in Toluol zugetropft und das Gemisch wird während 18 h bei 650 weiter gerührt. Man kühlt ab und zersetzt mit Wasser. Die benzolische Lösung wird abgetrennt, getrocknet und
