Verfahren zur Herstellung von 6, 8-Dithiooctansäureamiden
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 6, 8-Dithiooctansäureamiden der allgemeinen Formel

Durch Untersuchungen, die im Laboratorium und in der Klinik durchgeführt wurden, wurde festgestellt, dass die Verbindungen der allgemeinen Formel I eine Wirkung besitzen, die der der wegen ihrer vitaminähnlichen Wirkung und ihrer Befähigung, die Funktionen der Leber zu fördern und den Körper zu entgiften, bekannten Thioctansäure überlegen ist.
Abgesehen davon sind die Eigenschaften der mit den Verbindungen der allgemeinen Formel I hergestellten pharmazeutischen Präparate günstig. Die Präparate sind stabil, geschmacklos und geruchlos, während die Thioctansäure einige Eigenschaften besitzt, die verbesserungsbedürftig sind.
Es ist bereits bekannt [vgl. A. F. Wagner et al. : J. Am. Chem. Soc. Vol. 78 (1956), S. 5081] Thioctansäure mit kaustischer Soda umzusetzen, das erhaltene Produkt einer Gefriertrocknung zu unterwerfen und das so erhaltene Natriumsalz der Säure mit Oxalylchlorid in trockenem Benzol und dann mit Ammoniak in trockenem Dioxan umzusetzen. Im Hinblick darauf, dass die Säure und auch das Säurechlorid ungewöhnlich instabil sind, und ausserdem die Herstellung des Natriumsalzes auf dem Wege der Gefriertrocknung erfolgen muss, ist das Verfahren sehr umständlich.
Während des kompletten Herstellungsverfahrens muss unter sorgfältigem Ausschluss von Wasser gearbeitet werden ; ausserdem müssen sämtliche Verfahrensbedingungen ständig einer sorgfältigen Kontrolle unterworfen werden, da sonst leicht ungewünschte Nebenreaktionen, wie Aufspaltung des Dithiolan-Ringes und Polymerisation zu verharzten Nebenprodukten eintreten können. Trotz Einhaltung grösster Vorsichtsmassnahmen gelingt es aber nicht, Ausbeuten von über 20% zu erhalten.
Es ist weiterhin bekannt [vgl. L. J. Reed et al. : J. Biol. Chem. Vol. 232 (1958), S. 145] Thioctansäure mit Isobutylchlorformat in Tetrahydrofuran in Anwesenheit von Triäthylamin und das so erhaltene Zwischenprodukt dann mit Ammoniak umzusetzen. Auch dieses Verfahren ist in der Durchführung kompliziert und in der Ausbeute unbefriedigend.-Die bekannten Verfahren sind beide für die Herstellung im technischen Massstab zu unwirtschaftlich.
Die Aufgabenstellung der vorliegenden Erfindung geht dahin, ein neues wirtschaftlich und technisch vorteilhaftes Verfahren zur Herstellung von 6, 8-Dithiooctansäureamiden der allgemeinen Formel I zu finden.
Der Verlauf des Verfahrens gemäss der Erfindung wird durch folgendes Verfahrensschema dargestellt :

Beispiel I : 21, 0 g 6, 8-Dithiooctansäure werden in 100 cm3 Benzol gelöst. 13, 0 g Thionylchlorid werden tropfenweise zu der Lösung unter Kühlen und Rühren hinzugegeben. Nach der Beendigung der Zugabe wird das Gemisch 4 h bei Zimmertemperatur und anschliessend 30 min bei 35-40 C gerührt. Ungefähr 1/3 des Volumens des Benzols wird unter vermindertem Druck entfernt. Die Lösung wird dann in 200 cm3 einer 20%igen wässerigen Lösung von Ammoniak gegossen, wobei mit Eiswasser gekühlt wird.
Das erhaltene Gemisch wird ungefäht 3 h unter Kühlen gerührt. Der erhaltene Niederschlag wird abfiltriert, mit Wasser und Hexan gewaschen und getrocknet. Es werden 19, 7 g (95% d. Th. ) des 6, 8-DiThiooctansäureamids mit dem Fp. 117-122 C erhalten.

CH3OH332m (s144) ou maux 8, 0 g des oben beschriebenen rohen Amids werden in eine Lösung von 1, 6 g NaOH in 20 cm3 Wasser und 80 cm3 Methanol eingetragen. Das Gemisch wird 11/2 h auf einem auf 80 C gehaltenen Wasserbad erhitzt. Dann wird das Methanol unter vermindertem Druck entfernt. Der Rückstand wird mit Chloroform extrahiert. Beispiel 2 : Eine Lösung des rohen 6, 8-Dithiooctansäurechlorids, hergestellt entsprechend Beispiel 1 ausgehend von 20, 6 g 6, 8-Dithiooctansäure, 13, 0 g Thionylchlorid und 100 cm3 Benzol, wird zu minor Lösung von 25, 0 g Diisopropylamin in 200 cm3 Benzol bei Zimmertemperatur zugetropft. Das Gemisch wird 3 h bei Zimmertemperatur gerührt. Das Reaktionsgemisch wird dann nacheinander mit verdünnter Salzsäure, mit einer wässerigen Lösung von Natriumbicarbonat und mit Wasser gewaschen.

Der Chloroformextrakt wird mit Wasser gewaschen und getrocknet.
Das Chloroform wird entfernt. Die so erhaltenen Kristalle des N- (6, 8-Dithiooctanoyl)-DL-methionin werden aus Äthylacetat/Petroläther umkristallisiert. Die nunmehr erhaltenen Kristalle haben den Fp. 102 bis 1030 C.

Die erhaltene Verbindung ergibt keine Depression des Schmelzpunktes, wenn sie mit dem Endprodukt des Beispieles 3 gemischt wird.

Das Chloroform wird entfernt und so Kristalle von N-(6,8-Dithiooctanoyl)-L-Glutaminsäure erhalten. Die Ausbeute beträgt 3, 3 g (46, 1% d. Th). Die zunächst erhaltenen Kristalle werden aus Äthylacetat/Petroläther umkristallisiert, wodurch der Schmelzpunkt von 105-107 C erhalten wird.

The object of the present invention is to find a new, economically and technically advantageous process for the preparation of 6, 8-dithiooctanoic acid amides of the general formula I.

Example I: 21.0 g of 6, 8-dithiooctanoic acid are dissolved in 100 cm3 of benzene. 13.0 g of thionyl chloride are added dropwise to the solution with cooling and stirring.
