Abstract
Se proporciona un método cromatográfico de afinidad para preparar a partir de un material de partida que contiene IgG una formulación de IgG que incluye una o más de IgG1, IgG2, IgG3 e IgG4. Un método de la invención incluye poner en contacto el material de partida que contiene IgG con un primer medio cromatográfico que incluye un primer soporte sólido que tiene proteína A unida al mismo, formando una fracción unida a proteína A de una subclase de IgG con afinidad de unión a proteína A y formando un primer flujo continuo incluyendo una subclase de IgG que no se une a la proteína A; y poner en contacto el primer flujo continuo con un primer medio cromatográfico de ligando de afinidad que incluye un segundo soporte sólido que tiene un primer ligando de afinidad con afinidad de unión a IgG unido al mismo, formando una primera subclase de IgG unida a ligando de afinidad y un segundo flujo continuo que incluye una subclase de IgG no unida a la primera subclase de IgG unida a ligando de afinidad. Reivindicación 43: Una solución de IgG acuosa que comprende al menos aproximadamente 99,5% de IgG y menos de aproximadamente 0,5% de impurezas donde la solución de IgG acuosa comprende Factor XI como una impureza en una actividad de menos de aproximadamente 0,0025 E/mg de IgG total, preferentemente menos de aproximadamente 0,0020 E/mg de IgG total, más preferentemente menos de aproximadamente 0,0015 E/mg de IgG total y con la máxima preferencia menos de aproximadamente 0,0012 E/mg de IgG total.
