Abstract

En el presente descripción se describen métodos para tratar la distrofia muscular que comprenden administrar un vector de AAV recombinante (rAAV) scAAVrh74.MHCK7.hSGCG, métodos para expresar el gen de sarcoglucano g en un paciente, composiciones farmacéuticas que comprenden el rAAV, y métodos para generar el rAAV. Reivindicación 1: Un método para tratar la distrofia muscular en un sujeto que lo necesita que comprende la etapa de administrar un virus adenoasociado recombinante (rAAV) scAAVrh74.MHCK7.hSGCG al sujeto, en donde el rAAV se administra mediante el uso de una vía de administración sistémica y a una dosis de aproximadamente 2 ´ 10¹² vg/kg a aproximadamente 5,0 ´ 10¹⁴ vg/kg en función de un plásmido linealizado como el estándar de cuantificación; en donde el nivel de creatina cinasa (CK) sérica en el sujeto disminuye después de la administración del rAAV en comparación con el nivel de CK sérica antes de la administración del rAAV. Reivindicación 42: Un método para aumentar la expresión de sarcoglucanos en el tejido muscular de un sujeto que comprenden administrar al sujeto un constructo que comprende una secuencia de nucleótidos que codifica un primer sarcoglucano, y detectar un aumento de la expresión de al menos un segundo sarcoglucano en la membrana celular de la célula que expresa dicho primer sarcoglucano. Reivindicación 50: Una composición, que comprende un vector rAAV scAAVrh74.MHCK7.hSGCG, un agente amortiguador, un agente de resistencia iónica, y un tensioactivo. Reivindicación 65: Una composición farmacéutica que comprende un AAV recombinante (rAAV) scAAVrh74.MHCK7.hSGCG, en donde el scAAVrh74.MHCK7.hSGCG comprende una secuencia de nucleótidos que es al menos 95% o 99% idéntica a la SEQ ID Nº 7.
