Abstract
Reivindicación 1: Procedimiento de inmovilización de ligandos nucleicos que comprenden, al menos, una función amina primaria, por injerto sobre un soporte sólido que presenta grupos ácidos carboxílicos activados, que comprenden una etapa de acoplamiento covalente de dichos ácidos nucleicos sobre dicho soporte sólido a un pH inferior a 6.Reivindicación 13: Método de preparación de un soporte de afinidad que comprende la implementación del procedimiento de inmovilización tal como se ha definido en una cualquiera de las reivindicaciones 1 a 12. Reivindicación 19: Complejo resultante de la interacción de un ligando nucleico y una molécula diana, donde dicho complejo está formado en la superficie de un soporte sólido tal como se ha definido en una cualquiera de las reivindicaciones 14 a 18. Reivindicación 20: Utilización de un soporte de afinidad tal como se ha definido en una cualquiera de las reivindicaciones 14 a 18 para la purificación de una molécula diana. Reivindicación 22: Procedimiento de purificación de una molécula diana con un soporte de afinidad tal como se ha definido de acuerdo con una cualquiera de las reivindicaciones 14 a 18, que comprende las etapas siguientes: a) poner en contacto una composición por purificar que comprende la molécula diana de interés con un soporte sólido de afinidad tal como se ha definido en una de las reivindicaciones 14 a 18, con el objeto de formar un complejo entre (i) ligandos nucleicos injertados sobre dicho soporte y (ii) dicha molécula diana, y b) liberar dicha molécula diana a partir del complejo formado en la etapa a), y recuperar dicha molécula diana purificada.Claim 1: Method of immobilization of nucleic ligands comprising at least one primary amine function, by grafting on a solid support having activated carboxylic acid groups, comprising a step of covalent coupling of said nucleic acids on said solid support to a solid support. pH lower than 6. Claim 13: Method of preparation of an affinity support comprising the implementation of the immobilization process as defined in any one of claims 1 to 12. Claim 19: Complex resulting from the interaction of a ligand nucleic and a target molecule, wherein said complex is formed on the surface of a solid support as defined in any one of claims 14 to 18. Claim 20: Use of an affinity support as defined in any one of claims 14 to 18 for the purification of a target molecule. Claim 22: Purification process of a target molecule with an affinity support as defined according to any one of claims 14 to 18, comprising the following steps: a) contacting a composition to be purified comprising the target molecule of interest with a solid affinity support as defined in one of claims 14 to 18, in order to form a complex between (i) nucleic ligands grafted onto said support and (ii) said target molecule, and b ) releasing said target molecule from the complex formed in step a), and recovering said purified target molecule.
