Abstract
Se revela un fragmento aislado de ácidos nucleicos que codifica una proteina reguladora del ciclo celular que se selecciona entre una proteina CDC2 y unaprotein quinasa de PITSLRE. Se construye un gen quimérico que codifica toda o una porcion de dicha proteína, en orientacion sentido o antisentido, en donde laexpresion de dicho gen quimérico resulta en la produccion de niveles alterados de dicha proteína regulatoria del ciclocelular en una célula huéspedtransformada. Se revelan también métodos para obtener dicho fragmento de ácidos nucleicos y un método para evaluar por lo menos un compuesto respecto de sucapacidad para inhibir la actividad de una proteína de regulacion del ciclo celular. La disponibilidad de secuencias de ácidos nucleicosque codifiquen toda ouna porcion de una proteína reguladora del ciclo celular facilita los estudios para comprender mejor la regulacion del ciclo celular en plantas, proporcionaherramientas genéticas para mejorar el crecimiento celular en cultivos detejidos, incrementa la eficacia de la transferencia de genes y ayuda a obtenertransformaciones más estables.An isolated nucleic acid fragment encoding a cell cycle regulatory protein that is selected from a CDC2 protein and a PITSLRE protein kinase is revealed. A chimeric gene is constructed that encodes all or a portion of said protein, in a sense or antisense orientation, where the expression of said chimeric gene results in the production of altered levels of said regulatory cell protein in a transformed host cell. Methods for obtaining said nucleic acid fragment and a method for evaluating at least one compound for its ability to inhibit the activity of a cell cycle regulating protein are also disclosed. The availability of nucleic acid sequences that encode any portion of a cell cycle regulatory protein facilitates studies to better understand cell cycle regulation in plants, provides genetic tools to improve cell growth in detected cultures, increases the efficiency of cell transfer. genes and helps to obtain more stable transformations.
