Abstract

Los copolímeros injertados de polietilenglicol y poli-L-lisina (PEG-g-PLL) resultanparticularmente efectivos y ejecutan picos de expresiones genéticas dentro de las 24 horas, con persistencia de por lo menos 96 horas.A non-toxic polymeric gene transporter used to introduce a selected nucleic acid into a host cell comprising a grafted copolymer composed of straight-chain amphiphilic side polymers, grafted to a polycationic main polymer. The primary polycationic polymer forms a complex with nucleic acids by electrostatic interaction. Amphiphilic side polymers reduce the cytotoxicity of polycation and increase solubility. The molar ratio of the side polymers can be adjusted to optimize transfection efficiency. Similarly, the ratio of the nucleic acid to the copolymer can be adjusted to optimize efficiency. The composition can be used in a method for transforming the host cell with a selected nucleic acid. Grafted copolymers of polyethylene glycol and poly-L-lysine (PEG-g-PLL) are particularly effective and perform gene expression peaks within 24 hours, with persistence of at least 96 hours.
