Abstract

PURPOSE: One of components to be determined in a sample is converted into the second substance and the substance is determined using the oxidase corresponding to the second substance wherein the second substance in the sample is previously removed by phosphorylation, then the oxidase in made to act on, thus permitting the precise determination.
CONSTITUTION: In clinical biochemistry or in the field of fermentation, for example, when saccharose or lactose in a sample such as in blood or in a culture medium is determined, the saccharose or the like is converted into glucose (the second substance) and the oxidase corresponding to the second substance is used and the oxygen consumed or hydrogen peroxide formed are measured with an oxygen electrode, hydrogen peroxide electrode or enzyme electrode. At this time, the glucose previously existing in the sample is removed using immobilized kinase and adenosine triphosphate and the saccharose in the sample is converted into the glucose. Thus, the operations are simplified, the amount of the reagent used is lessened and the time for measurement is shortened and accurate results are obtained.
