Description
本発明は、NAD(P)依存性コレステロール脱
水素酵素(Cholesterol dehydrogenase:以下
「NAD(P)−CDH」と略す)に関する。さらに
詳しくは、微生物を好気的条件下で培養し、その
菌体もしくは培養液からNAD(P)−CDHを採取
する方法に関する。
本発明方法により得られたNAD(P)−CDH
は、コレステロールの定量法およびコレステロー
ルの定量用試薬として有用である。ここでいう
NAD(P)−CDHとは、補酵素としてNAD(ニコ
チンアミドアデニン、ジヌクレオチド)、NADP
(ニコチンアミドアデニン、ジヌクレオチドリン
ン酸)を要求し、電子供与体(コレステロール)
から水素をうばい、電子受容体(NAD又は
NADP)に付加する反応を触媒する酵素をいう。
従来好気性微生物が、コレステロールオキシダー
ゼ、コレステロールデヒドラターゼを生産するこ
とは既に知られている。また、ノカルジア・エリ
スロポリスがコレステロールの酸化を触媒する酵
素を生産するとの報告(Ann.Clin.Biochem.,10
巻、79頁、1973年)がある。この酵素の場合は、
NAD(P)への依存性は認められない。また、マ
イコバクテリウム・コレステリカム(J.Biol.
Chem.,206巻、511頁、1953年)、ブレビバクテ
リウム・ステロリカム(特公昭48−1190)につい
ても同様のことがいえる。さらには、絶対嫌気性
微生物である、オイバクテリウムsp.ATCC21408
がNAD(P)−CDHを生産するとの報告(特開昭
53−56090)もあるが、酵素学的性質等の記載は
ほとんどなされていないばかりか、NAD(P)依
存性の記載があるにもかかわらず、NAD(P)の
存在なしにも反応が進行する例が記載されてい
る。したがつてこの酵素は、本発明でいうところ
のNAD(P)依存性脱水素酵素とはいえない。
従来酵素によるコレステロールの定量は、コレ
ステロールオキシダーゼを使用する方法が広く用
いられているが、発色系に導くためにパーオキシ
ダーゼ等が必要であり、操作が繁雑である。しか
も血中のビリルビン、アスコルビン酸等により影
響をうけ、これにより誤差が生じやすいという欠
点を有している。
コレステロールの定量において、NAD(P)の
存在なしには反応が進行しないNAD(P)−CDH
を用い、下記反応式に示される反応により生ずる
NAD(P)Hを直接光度計で測定できれば、操作
が簡単であり、前記のコレステロールオキシダー
ゼを用いる方法の種々の問題も解決される。

本発明者等は、上記反応に適した酵素すなわ
ち、コレステロールに特異性が高く、NAD(P)
依存性である脱水素酵素を広く自然界に求めたと
ころ、意外にも好気的条件下に生育する微生物
が、著量のNAD(P)−CDHを生産することを見
い出した。その中で、特に優れた菌株として、ノ
カルジアsp.No.Ch2−1(Nocardia sp.No.Ch2−
1)、アリカリゲネスsp.No.4(Alcaligenes sp.No.
4)、およびプロテウス・ブルガリス
IAM1025Proteus vulgaris IAM1025が例示され
る。
次にノカルジアsp.No.Ch2−1およびアルカリゲ
ネスsp.No.4の菌学的性質を以下に述べる。
(1) ノカルジアsp.No.Ch2−1(Nocardia sp.No.
Ch2−1)の菌学的性質
(A) 形態的性質
1 細胞の形および大きさ:培養初期菌糸状に
生育し分岐を生じる。その後、不規則な分断
が生じ細胞は桿菌状となる。大きさは0.8〜
1.0μ×1.5〜4.0μ位である。気菌糸を形成せず
胞子のう胞子も形成しない。
2 グラム染色性:陽性
3 抗酸性:陽性
4 運動性:無し
(B) 化学的組成分析
細胞壁中にmeso−ジアノピメリン酸、アラ
ビノース、ガラクトースが含まれL,L−ジア
ミノピメリン酸、グリシンは含まない。
(C) 各培地における生育状態
1 肉汁寒天平板培地:30℃で4日培養後、直
径0.5〜1.0mmの円形コロニーを形成する。周
辺は全縁もしくは波状である。表面は平滑で
半球状であり、中心部が凸状に隆起する場合
もある。色は薄いクリーム色で不透明であ
る。培地中に色素は出さない。
2 シユークロース削酸塩寒天培地
生育中程度で集落の色は白色ないし薄クリ
ーム色である。水溶性色素は出さない。
3 グルコース、アスパラギン寒天培地
生育中程度で集落の色はクリーム色であ
る。水溶性色素は出さない。
4 グリセリン、アスパラギン寒天培地
生育中程度で集落の色は白色ないし薄クリ
ーム色である。水溶性色素は出さない。
5 スターチ無機寒天培地
生育中程度で集落の色は白色ないし薄クリ
ーム色である。水溶性色素は出さない。
6 チロシン寒天培地
生育中程度で集落の色は白色ないし薄クリ
ーム色である。水溶性色素は出さない。
7 栄養寒天培地
生育良好で集落の色はクリーム色である。
水溶性色素は出さない。
8 イースト麦芽寒天培地
生育良好で集落の色はクリーム色である。
水溶性色素は出さない。
9 オートミール寒天培地
生育中程度で集落の色は白色ないし薄クリ

(D) 生理的性質
1 生育温度:15℃〜43℃で生育する。10℃、
45℃で生育しない。最適温度は30〜35℃であ

以上の菌学的性質をBergey′s Manual of
Determinative Bacteriology第8版を参考に検
討した結果、細胞壁中にmeso−ジアミノピメリ
ン酸、アラビノース、ガラクトースを含み、L,
L−ジアミノピメリン酸、グリシンが含まれない
こと、好気性で菌糸状によく生育し、後に分断し
て菌状となること、抗酸性であること、胞子のう
胞子および気菌糸を着生しないこと等から本菌は
Nocardiaに属する菌である。
よつて本菌は、本発明者らがノカルジアsp.No.
Ch2−1(Nocardia sp.No.Ch2−1)と命名し、
工業技術院微生物工業技術研究所に菌寄第6217号
(FERM−PNo.6217)として寄託されている。

照合して検討すると短桿菌で周ベン毛により運動
すること、グラム陰性、好気性であること、栄養
要求性はなく、アンモニウム塩、硝酸塩を利用す
ること、カゼインおよびゼラチンを分解しないこ
と、オキシダーゼ陽性であること等から
Alcaligenes属に分類される。
種については、Alcaligenes eutrophus、A.
paradoxus、A.ruhlandiiおよびA.latusとは主に
独立栄養的生育の点で異なる。またA.faecalisと
は主に42℃における生育、D−フラクトースの資
化性で、A.aquamarinusとは、主にデンプンの
分解性、マルトースの資化性で、A.pacificusと
は、主にスレオニン、ベタインの資化性で、A.
cupidusとは、主にオキシダーゼおよびマンニツ
ト、マンノースの資化性で、A.venustusとは、
主にレブリン酸塩、スレオニン、ベタインの資化
性で、A.aestusとは、主にマンニツト、スレオニ
ン、フエニルアラニンの資化性で異なる。よつて
本菌をアルカリゲネスsp.No.4(Alcaligenes sp.No.
4)と命名し工業技術院微生物工業技術研究所に
菌寄第6216号(FERM−PNo.6216)として寄託
されている。
次にこれらの菌を用いてNAD(P)−CDHを製
造する方法について詳述する。これらの菌はいづ
れも構成的にNAD(P)−CDHを生産する能力を
有し、通常のペプトン、酵母エキス、又は硫酸ア
ンモニウム等のチツソ源及びグルコース、グリセ
ロール等の炭素源、その他無機塩等を含有する培
地でもNAD(P)−CDHを生産するが、コレステ
ロールを培地に添加することによりさらに多重に
NAD(P)−CDHを生産する。この際コレステロ
ールの添加は、培養開始時あるいは途中からのい
ずれでもよい。また、その他培養条件に関して
は、通常行なわれる範囲で実施できる。これらの
菌により生産されたNAD(P)−CDHは、菌体の
みならず、培養液中にも蓄積され、その何れから
でも酵素を回収することができる。これらの培養
濾液又は菌体抽出液を硫酸アンモニウム等による
塩析又は、アセトン、エタノール等の溶剤沈澱し
て得た粗酵素は、そのままコレステロールの定量
に使用するか、あるいは更に精製して使用するこ
ともできる。例えば精製については、イオン交換
クロマトグラフイー、分子篩クロマトグラフイー
等公知の方法により可能である。
ここで得られるNAD(P)−CDHは、コレステ
ロール含有物質中のコレステロール定量等に有利
に使用できる。
本発明に使用するNAD(P)−CDHの活性測定
法を以下に示す。
NAD(P)−CDHの酵素活性は、コレステロー
ルとNAD(P)を基質として反応した場合の
NAD(P)Hの生成量を、340nmにおける吸光度
の増加として、分光光度計で測定し算出する。す
なわち、0.1Mトリス・塩酸緩衝液(PH8.6)2.65
ml、28mMNAD溶液0.1ml、(8%トリトンX−
100溶液0.15ml)、1%コレステロール溶液0.05ml
及びNAD(P)−CDH水溶液0.05mlを混合し、30
℃で反応させ、反応開始後1分間の340nmにおけ
る吸光度の増加を測定する。
対照として上記組成でコレステロールの代りに
水を用い同様の操作を行ない、対照液の340nmに
おける吸光度の増加を試験液のそれから差し引
く。得られた値からNAD(P)Hの生成量を求
め、これより試料中のNAD(P)−CDH活性を算
出する。
酵素活性の表示は、PH8.6、30℃の条件下で1
分間に1μmoleのNAD(P)Hを生成する酵素を
1単位とした。
次に本発明に使用するNAD(P)−CDHの作用
を示す。

応を触媒する。
以下に本発明に使用するNAD(P)CDHの一
般的性質をノカルジアsp.No.Ch2−1(Nocardia

スsp.No.4(Alcaligenes sp.No.4)FERM−P6216
の生産するものについて示す。

次に本発明のNAD(P)−CDHを使用したコレ
ステロールの定量について詳述する。
コレステロールを定量する場合、実際には緩衝
液、NAD(P)、基質(血清、コレステロール等)
及び、NAD(P)−CDHを混合し、一定時間反応
し、生成するNAD(P)Hの増加を吸光度340nm
で測定する。また必要に応じて、生成したNAD
(P)Hの水素をフエナジンメソサルフエート
(PMS)、ジアフオラーゼ等により、ホルマザン
色素等の発色系に導くことも可能である。さらに
は、反応系にコレステロールエステラーゼ、界面
活性剤及び、安定化剤等の添加も可能である。反
応時のPHは6〜10の範囲で実施できるが、優れて
いるPH範囲は7〜9である。
次にNAD(P)−CDHによるコレステロール定
量用試薬の量的組成についての1例を述べれば、
反応系3ml当り、NAD(P)−CDH0.1〜10単位、
NAD(P)10〜100mM、トリトンX−100 1.0%
以下、コレステロールエステラーゼ(ベーリンガ
ー社製)0.1〜10単位が有利である。しかし本発
明はこれらの量的組成に限定されるものではな
い。本発明の測定法における特別な利点は生成す
るNAD(P)Hを直接測定できることであり、ま
た完全に定量できることである。
次に試験例及び実施例につき述べる。
試験例
本発明における菌株3種につき、コレステロー
ル5g/、肉エキス5g/、酵母エキス0.2
g/、少量の消泡剤の組成よりなる培地(PH
7.2)100mlを入れた500ml容坂ロフラスコに植菌
し、30℃で40時間振盪培養した。培養液50mlを遠
心分離(8000rpm10分間)により集菌し、0.1M
リン酸緩衝液PH7.0、50mlで洗浄した。
次に同じ緩衝液50mlに菌体を懸濁し、超音波に
て菌体を破砕した。この破砕液を塩心分離
(10000rpm、10分間)し、清澄液を得た。得られ
た清澄液のNAD(P)−CDH活性を測定し次の結
果を得た。

ーに植菌し、30℃で通気、撹拌(0.5v/v/
min、200rpm)しながら40時間培養した。
培養液を遠心分離し、得られた菌体を0.1Mリ
ン酸緩衝PH70に懸濁し、ガラスビーズにより菌体
を破砕した。この菌体破砕液を遠心分離
(1000rpm、10分間)し、清澄な菌体抽出液を得
た。得られた清澄液に硫酸アンモニウムを35%飽
和になるように加え酵素を沈澱せしめた。
沈澱を遠心分離(8000rpm、10分間)で集め
20mMリン酸緩衝液PH7.0、100mlに溶かし、セロ
フアンチユーブで、20mMリン酸緩衝液PH7.0に
対して24時間透析した。
次に得られた透析液を20mMリン酸緩衝液PH
7.0で平衡化した。DEAE・セルロース200mlを充
填したカラムに通し、酵素を吸着せしめた。同様
の緩衝液でカラムを洗浄後、緩衝液濃度を0.1M
に上げてNAD(P)−CDHを溶出した。NAD
(P)−CDHを含む画分を集め、これを濃縮後
20mMリン酸緩衝液PH7.0に対して透析した。こ
れを同様の緩衝液で平衡化したヘキシルセフアロ
ース20mlを充填したカラムに通し、吸着せしめ
た。このカラムを20mMリン酸緩衝液PH7.0で洗
浄した。次に0.5MNaClを含む同様の緩衝液で
NAD(P)−CDHを溶出し、活性画分を集め、濃
縮し、50単位/mlのNAD(P)−CDH溶液5.0ml
を得た。全体の活性収率は30%であつた。
実施例 2
Alcaligenes sp.No.4FERM−PNo.6216をコレス
テロール10g/、グリセロール2g/、コー
ンスチープリカー5g/、KH2PO45g/、
MgSO47H2O0.2g/、消泡剤0.5g/の組成
よりなる培地に培養し、実施例1と同様の操作を
行ない、40単位/mlのNAD(P)−CDH溶液を
5.0ml得た。全体の活性収率は40%であつた。
実施例 3
Proteus vulgaris IAM1025を用い、実施例1
に準ずる操作を行ない、37単位/mlのNAD(P)
−CDH溶液4mlを得た。全体の活性収率は52%
であつた。
実施例 4
Nocardia sp.No.Ch2−1FERM−PNo.6217をグ
ルコース5g/、肉エキス、酵母エキス0.2ml、
少量の消泡剤の組成よりなる培地(PH7.2)200ml
を入れた500ml容坂ロフラスコに植菌し、30℃で
24時間振盪培養する。この種培養液をコレステロ
ール5g/、グルコース2g/、肉エキス5
g/、KH2PO45g/、MgSO4・7H2O0.2
g/、消泡剤0.5g/の組成よりなる培地
(PH7.2)20を入れた30容ジヤーフアーメンタ
ーに植菌し、30℃で通気、撹拌(0.5v/v/
min、200rpm)しながら、72時間培養した。こ
の培養液を遠心分離(8000rpm、10分間)除菌し
た。得られた培養液に、硫酸アンモニウムを40
%飽和になるように加え酵素を沈澱せしめた。
沈澱を遠心分離(8000rpm、10分間)で集め、
20mMリン酸緩衝液(PH7.0)100mlに溶かし、セ
ロフアンチユーブで、20mMリン酸緩衝液PH7.0
に対して24時間透析した。
次に実施例1〜実施例3に準ずる方法で精製を
行ない、30単位/mlのNAD(P)−CDH溶液を32
ml得た。全体の活性収率は32%であつた。
実施例 5
Alcaligenes sp.No.4FERM−PNo.6216を用い、
実施4に準ずる操作を行ない、25単位/mlの
NAD(P)−CDH溶液を4.2mlを得た。全体の活
性収率は38%であつた。
実施例 6
Proteus vulgaris IAM1025を用い、実施例4
に準ずる操作を行ない、18単位/mlのNAD(P)
−CDH溶液を3.5ml得た。全体の活性収率は49%
