Abstract
Description
本願は、米国仮特許出願第61/785,030号(2013年3月14日出願)に対する優先権を主張し、上記出願の内容は、参照により本明細書に引用される。
心筋梗塞につながるアテローム性動脈硬化症およびプラーク破裂は、世界中で主要な死因のままである[1]。炎症ならびに基礎的な細胞および分子機構[2−4]は、発生から進行を通したアテローム発生、プラーク破裂、および最終的には血栓症の一因となる。近年、Virmani[5]によって「薄い被膜の線維性アテローム」として定義されている、脆弱なプラークは、炎症に起因し、典型的には、厚さが65μm未満の薄い線維性被膜、減少した平滑筋細胞を伴うマクロファージの浸潤の増加、および安定したプラークと比較して増加した脂質コアのサイズを有するものとして特徴付けられる[6−8]。

カテーテルベースの強度OCT−TPLシステム(図5Aおよび5Bに示される)の実施例は、同調可能なフェムト秒レーザ励起(例えば、760nm〜1040nm、6nJ〜5μJ、100fs〜1ps、500kHz〜80MHz)を使用するTPLと組み合わせられた、1310nmで動作するスペクトル領域OCTシステムを組み込むことができる。フェムト秒レーザ光の群分散遅延を事前に補償して、管腔表面に変換制限パルスを提供するために、パルス圧縮機を利用することができる。撮像カテーテルは、OCT光およびTPL励起光の両方の単一モード伝搬、ならびにTPL放射光の透過(例えば、OCT光およびTPL励起光が同時に透過させられる)を可能にすることができる、OCT−TPL撮像システムのフォトニック結晶ファイバ(PCF)(例えば、LMA−20、NKT Photonics)に接続される。ある実施形態では、PCFは、15μmコア(例えば、LMA−15)を備え得る。
カテーテルベースの高偏光感度OCT−TPLシステム(図6に示される)の一実施例は、同調可能なフェムト秒レーザ励起(例えば、760nm〜1040nm、6nJ〜5μJ、100fs〜1ps、500kHz〜80MHz)を使用するTPLと組み合わせられた、1310nmで動作するスペクトル領域高偏光感度OCT(PSOCT)システムを組み込むことができる。図6のシステムは、インラインファイバ偏光計の役割を果たす偏光維持ファイバ区画(PM1およびPM2)と、偏光ビームスプリッタ(PBS)と、帯域通過フィルタ(BP)と、ショートパスフィルタ(SP)と、光子増倍管(PMT)と、フォトニック結晶ファイバ(PCF)とを備えている。
カテーテルベースの高位相感度OCT−TPLシステム(図7に示される)の一実施例は、同調可能なフェムト秒レーザ励起(例えば、760nm〜1040nm、6nJ〜5μJ、100fs〜1ps、500kHz〜80MHz)を使用するTPLと組み合わせられた、1310nmで動作するスペクトル領域高位相感度OCT(PhSOCT)システムを組み込むことができる。図7のシステムは、偏光ビームスプリッタ(BS)と、帯域通過フィルタ(BP)と、ショートパスフィルタ(SP)と、光子増倍管(PMT)と、フォトニック結晶ファイバ(PCF)とを備えている。
本実施例では、OCT−TPLカテーテルが、TPL励起および放射を組み込むように、現在のOCTカテーテルを修正するであろう。以前、ナノ粒子を装填されたマクロファージの検出は、特注の多重光子顕微鏡を使用して行われた[30]。したがって、提案されたカテーテルベースのOCT−TPL撮像システムのTPL励起効率を、多重光子顕微鏡と比較することが望ましい。表1は、両方の撮像システムからのレーザ励起の特性評価を示す。
金ナノロッドは、マクロファージ(アテローム性動脈硬化症および癌に関与する重要な初期細胞マーカー)によって取り込まれ、マクロファージ標的化のための種々の撮像技法用の造影剤として使用されることができる。この研究の目的は、それぞれ、700、756、844、および1060nmでの表面プラズモン共鳴をともなう4つのサイズの金ナノロッドの二光子発光(TPL)性質を比較することである。レーザ走査TPL顕微鏡を使用して、単一ナノロッドおよびローダミン6G粒子からのTPLが測定された。ナノロッドからのTPL放射スペクトルが、光子増倍CCDを伴う分光計によって記録された。4つ全てのサイズのナノロッドは、励起波長への依存性を伴う強いTPL強度を生成し、二光子作用断面(TPACS)がプラズモン増進されることを示した。(TPLプロセスを確認する)励起出力への発光強度の二次の依存性が、低出力レベルで観察され、その後、光退色効果による高出力レベルでの強度飽和または減少が続いた。単一のナノロッドの最大TPACSは、760nm励起での単一のローダミン6G粒子の25GMと比較して、12271GMと測定された。ナノロッドTPL放射スペクトルの特性は、ブリユアン領域中のXおよびY対称点付近の穴とのフェルミレベル付近の電子の再結合によって説明することができる。選択された撮像用途のためのもっとも明るい造影剤の選択を誘導するために、ナノロッドのTPL輝度、TPACS、および放射スペクトルの比較結果を使用することができる。
(サンプル調製)
以前に説明されたような種結晶成長方法を使用して、金ナノロッドが溶液中で合成された[68]。それぞれ、700、756、844、および1060nmで表面プラズモン共鳴を伴う4つのサイズのナノロッドが、Nanopartzから購入され、簡潔に超音波分解され、使用前に原液濃度から10回希釈された。ナノロッドサンプルが、スライドガラス上に5μl希釈を分散することによって調製され、カバースリップによって覆われ、厚さ5μmのナノロッド溶液を形成した。スライドガラス上の4つのサイズのナノロッドの最終濃度は、それぞれ、5.7×1010、4×1010、7.2×1010、および2.8×1010ナノ粒子/mlである。透過電子顕微鏡法(TEM)は、ナノロッドの形態を明らかにし、TPL画像は、回折限界での単一のナノロッドの形状を示した(図10a−d)。図10は、研究で使用された金ナノロッドのTEM画像、(a)Au700、(b)Au756、(c)Au844、(d)Au1060を提供する。(a、b、c、d)中の差し込み図は、400nm〜700nmのスペクトル範囲内の840nm励起での単一のナノロッドのTPL画像である。TEMおよびTPL画像中のスケールバーは、それぞれ、20nmおよび1μmを表す。(e)レーザ走査TPL顕微鏡の概略図。EOM:電気光学変調器、PMT:光電子増倍管。
レーザ走査TPL顕微鏡(図10b、Prairie Technologies,Middleton,WI)を使用して、ナノロッドからのTPLが測定された。760nm〜1040nm(80MHz、100fs)で放射するフェムト秒Ti:サファイアレーザ(Mai Tai HP,Newport,Irvine,CA)が、励起光源として使用された。顕微鏡に進入するレーザビームの強度が、電気光学変調器(350−80,ConOptics,Danbury,CT)によって変調され、サンプルに送達される電力を測定するための電力計を伴うピックミラー(反射率1%)によって監視された。2D画像を生成するために、一対のガルバノメトリック走査鏡を使用して、x−y面内のサンプルにわたって対物レンズ(40倍、NA=0.8、水出現、Olympus,Center Valley,PA)の焦点量が走査された。サンプルからのTPL放射が、同一の対物レンズを通して収集され、720nmロングパスダイクロイックミラーによって励起レーザ線から分離され、4つのチャネルの中へ向かわせられ、それぞれ、640nm〜680nm、570nm〜620nm、490nm〜560nm、および435nm〜485nmのスペクトル範囲内で4つの光電子増倍管(PMT1、2:H7422P−40、PMT3、4:R3896、Hamamatsu,Bridgewater,NJ)によって検出された。励起レーザ線からの光子カウントを最小化するために、ショートパスフィルタ(et720sp,Chroma Technology,Bellows
Falls,VT)がダイクロイックミラーの後に配置された。この研究では、検出光路中のダイクロイックミラーおよび帯域通過フィルタなしでTPL放射信号(720nm未満)を収集するために、PMT1のみが使用された。TPLはまた、PMT1を光子増倍CCD(Shamrock 303i,Andor Technology,Belfast,Ireland)を伴うファイバ連結分光計と交換することによっても測定された。
ナノロッドのTPACSが、基準ローダミン6GサンプルからのTPL放射の比較方法によって決定された。サンプルからのTPL放射は、関連パラメータを用いた方程式(1)で表すことができる[69]。
(ナノロッド輝度の電力依存性)
図11aは、4×1011ナノ粒子/mlの濃度で測定された4つのサイズのナノロッドの単一光子吸収スペクトルを示す。図11bは、760、840、および1040nmの波長でのナノロッドの励起レーザ出力(132μW〜4.8mW)へのMPL強度依存性を示す。図11cは、(b)の低電力レベルでの励起レーザへのナノロッドの発光強度の二次の依存性を示す。(異なる励起波長でのナノロッドの各サイズに対する)1.7〜2.2の傾きが、TPLプロセスを確認する。
ナノロッドのTPACSが決定されることができる前に、ローダミン6GのTPACSが測定される必要がある。690nm〜960nmの励起波長範囲を伴うローダミン6G溶液のTPACSが、Albotaら[71]によって報告されているが、このデータは、960nm〜1040nmの波長範囲を含まない。この研究では、我々は、Albotaらのデータを80nmだけ拡張する760nm〜1040nmの励起波長範囲で方程式(1)を使用して、ローダミン6G溶液および単一粒子の両方の正規化TPACSを測定して計算した。ローダミン6GのTPLプロセスが、全ての励起波長およびを適用された電力範囲(データ図示せず)において観察され、ローダミン6G溶液の測定は、820nmで重複した主要吸収ピークを伴う760nm〜960nm範囲内の報告値に合理的に合致する。単一のローダミン粒子の吸収ピークは、800nmへの青色偏移を有し、1000nmでの第2のピークは、ローダミン6G溶液と比較して著しく減衰させられる。次いで、単一のローダミン6G粒子のTPACSが、方程式(2)に従ってナノロッドと比較するために輝度基準として使用された。
ナノロッドのTPL放射をより良く特性評価するために、TPL放射スペクトルが、複数の励起波長(すなわち、760、800、840、および1040nm)において350nm〜700nmの範囲内でナノロッド溶液(80×80μm2視野)から収集された。全てのナノロッドに対する平均励起出力は、TPLプロセスを満たすことができるように、1mW未満に保たれた。次いで、TPL放射が、入射光子の数およびナノロッドの濃度によって正規化され、図14に示された。
ナノロッド輝度が、マクロファージ標的および検出における非常に重要なパラメータであり、それは、撮像システムの感度も決定するので、最も強いTPL信号を生じるナノロッドのサイズを選択することは、大いに臨床的関心であり、重要である。この研究では、4つのサイズのナノロッドが比較され、Au756が、同一の励起出力で、および対応する縦SPRの励起波長で最も強く放射することが分かった。実際、金ナノロッドからの単一光子励起による蛍光放射は、EustisおよびEl−Sayedによって実験およびシミュレーション研究[73]で実証されるように、3つの要因、すなわち、(1)増加するナノロッドアスペクト比とともに増加するはずである、縦SPR波長での単一光子吸収度の強度(図11a)、(2)d帯と伝導帯との間の電子遷移に起因し、約1.8eV(689nm)の閾値エネルギーで開始された、SPR吸収帯とバンド間遷移との間の重複[74、75]、(3)約525nmでピークになり、その後減少し、約750nmを超えると減退する、バルク金のSPR吸収と蛍光帯との間の重複[47]によって決定される。第1の要因は、他の2つの要因に対する競合構成要素であり、その正味の効果は、TPL放射の増進を決定する。この研究では、ナノロッドのアスペクト比が2.9から6.7まで増加すると、縦SPRの増進が増加する一方で、SPR吸収帯とバンド間遷移との間の重複、または金のバルク蛍光が両方とも減少する。したがって、観察された最も強い増進は、3.5のアスペクト比を伴うAu756をもたらし、それは、Au1060より約12倍高い。この観察は、単一光子励起を伴うナノロッドについて報告されたものに非常に類似し、ナノロッドの量子収量は、3.4を下回るアスペクト比に対して二次的に増加し、その後減少し[76]、蛍光放射は、アスペクト比が3.25を超えて増加すると、6のアスペクト比で約1桁弱くなるまで減退し始める[73]。
TPLMを利用することによって、金ナノロッドのTPL性質が調査され、特性評価された。700、756、844、および1060nmの縦SPR波長を伴う4つのサイズのナノロッドが、複数の励起波長(すなわち、760、840、1040nm)で励起させられた。Au756は、全てのナノロッドの間で、同一の励起出力を用いて760nm励起で最も強いTPL信号を放出することが観察された。励起出力へのTPL強度の二次の依存性が、低電力レベル(例えば、<1.6mW)で満たされた一方で、光退色効果は、特により大きいサイズのナノロッドについて、高電力レベル(例えば、>1.6mW)で明白であった。3つの励起波長でのナノロッドのTPACSが、単一のローダミン6G粒子の正規化TPACSスペクトルの測定に基づいて計算された。ナノロッドのTPL放射スペクトルは、金の電子バンド計算に合致し、TPL輝度測定と一致する。結果は、金ナノロッドがTPLMのための有望な造影剤であり、ナノロッドのTPL輝度、TPACS、および放射スペクトルの比較によって、最も明るいナノロッドを決定できることを示唆する。
