Abstract

懸濁液への電界の印加を適正に実施できるエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置を提供する。電極対の間を生体由来物と生体活性物質とを含有する懸濁液と、シース液とを、シース液で懸濁液を挟むようにして主流路に連続的に送液し、電極対によって懸濁液に電界を印加することで、生体由来物に生体活性物質を導入する、エレクトロポレーション方法であって、電極対の間において懸濁液及びシース液の流れが安定した後に、懸濁液に電界を印加する印加工程を有する、エレクトロポレーション方法である。

Description

エレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置
 本発明は、シース液で、生体由来物と生体活性物質とを含有する懸濁液を挟むようにして、電極対の間に連続的に送液するエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置に関し、特に、電極対の間において懸濁液及びシース液の流れが安定した後に、懸濁液に電界を印加するエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置に関する。
 電気穿孔法は、電気パルスにより細胞膜に孔をあけることにより細胞内に物質を導入する手法であり、エレクトロポレーションとも呼ばれる。例えば、細胞懸濁液に電極対によって電界を印加することで細胞膜に微小な孔を開け、DNA(Deoxyribonucleic acid)を細胞内部に導入することで、細胞の形質転換を行うことが可能である。
 なお、以下、エレクトロポレーションのことを、EPと略すことがある。
 電気穿孔法を用いて、細胞、細胞誘導体、細胞小器官、細胞内顆粒及び小胞等の生体由来物に、DNA、RNA(Ribonucleic acid)及びタンパク質等の生体活性物質を導入するEPとして、バッチ式が広く用いられている。
 バッチ式においては、例えば、内面に電極対が設置された容器に、生体由来物及び生体活性物質を含む懸濁液が収容され、電極対によって電界が印加される。これにより生体由来物の表面を覆う膜に微小な孔が開き、膜の透過性が高くなる。また、ここで、生体活性物質が拡散や電気泳動により透過性が高まった膜を通過することで、生体活性物質が生体由来物へ導入される。
 一方、例えば、特許文献1及び2に示されるように、電極対が設置された流路中に、生体由来物及び生体活性物質を含む懸濁液を流通させるフロー式のエレクトロポレーションも開発されている。

 特許文献1及び2では、懸濁液にEPを実施する際に、対向する2つの電極に、所定の電界強度となるように電気パルスが加えられる。懸濁液を送液しながらEPを実施するフロー式のエレクトロポレーションでは、懸濁液の流れが安定な状態ではないと導入効率が低下する場合があり、生産性が低い。
 本発明の目的は、生産性が高いフロー式のEPを行うことができるEP方法、有用物質の製造方法及びEP装置を提供することにある。
 以下の構成により、上述の目的を達成することができる。
 発明[1]は、電極対の間を生体由来物と生体活性物質とを含有する懸濁液と、シース液とを、シース液で懸濁液を挟むようにして主流路に連続的に送液し、電極対によって懸濁液に電界を印加することで、生体由来物に生体活性物質を導入する、EP方法であって、電極対の間において懸濁液及びシース液の流れが安定した後に、懸濁液に電界を印加する印加工程を有する、EP方法である。
 発明[2]は、懸濁液を挟むようにシース液を主流路に送液する際に、懸濁液の供給に第1チューブが用いられ、シース液の供給に第2チューブが用いられ、第1チューブにおいて懸濁液の先頭の待機位置が予め定められ、第2チューブにおいてシース液の先頭の待機位置が予め定められており、懸濁液及びシース液の全ての先頭が、待機位置に到達した後に、懸濁液及びシース液の送液を開始する、発明[1]に記載のEP方法である。
 発明[3]は、印加工程の前に、電極対の間において懸濁液及びシース液の流れの安定を判定する判定工程を有し、判定工程は、電極対の間における懸濁液及びシース液の流れの電気抵抗を測定して、電気抵抗に基づいて、流れの安定を判定するか、又は、判定工程は、第1チューブの懸濁液の流量及び第2チューブのシース液の流量の安定性を検出して流れの安定を判定する、発明[1]又は[2]に記載のEP方法である。
 発明[4]は、判定工程は、電極対の主流路における上流側又は下流側において、電極対の間における懸濁液及びシース液の流れの電気抵抗を測定して、電気抵抗に基づいて、流れの安定を判定する、発明[3]に記載のEP方法である。
 発明[5]は、懸濁液及びシース液を送液するタイミングは、懸濁液とシース液とで同時である、発明[1]~[4]のいずれか1つに記載のEP方法である。
 発明[6]は、懸濁液及びシース液を送液するタイミングは、懸濁液よりもシース液の方が先である、発明[1]~[4]のいずれか1つに記載のEP方法である。
 発明[7]は、電極対の間を生体由来物と生体活性物質とを含有する懸濁液と、シース液とを、シース液で懸濁液を挟むようにして主流路に連続的に送液し、電極対によって懸濁液に電界を印加することで、生体由来物に生体活性物質を導入する、EP方法であって、懸濁液を挟むようにシース液を主流路に送液する際に、懸濁液の供給に第1チューブが用いられ、シース液の供給に第2チューブが用いられ、第1チューブにおいて懸濁液の先頭の待機位置が予め定められ、第2チューブにおいてシース液の先頭の待機位置が予め定められており、懸濁液及びシース液の全ての先頭が、待機位置に到達したことを検知した後に、懸濁液及びシース液の送液を開始し、懸濁液及びシース液の送液後、電極対の間において懸濁液及びシース液の流れが安定した後に、懸濁液に電界を印加する、EP方法である。
 発明[8]は、印加工程では、懸濁液及びシース液の送液を開始後、予め設定した時間経過後に、懸濁液に電界を印加する、発明[1]~[7]のいずれか1つに記載のEP方法である。
 発明[9]は、第1チューブにおける懸濁液の先頭の位置は、超音波センサで検知される、発明[7]に記載のEP方法である。
 発明[10]は、発明[1]~[9]のいずれか1つに記載のEP方法を含む、有用物質の製造方法である。
 発明[11]は、生体由来物と生体活性物質とを含有する懸濁液を連続的に送液して電界を印加することで、生体由来物に生体活性物質を導入する、EP装置であって、主流路と、対向して配置された電極を備え、主流路を流れる懸濁液に電界を印加する電極対と、主流路に懸濁液を供給する懸濁液供給口と、主流路内の懸濁液供給口の下流側に設けられ、主流路にシース液を供給するシース液供給口と、懸濁液及びシース液を送液するポンプと、電極対の間において懸濁液及びシース液の流れが安定した後に、懸濁液に電界を印加する電源部とを有する、EP装置である。
 発明[12]は、シース液供給口と連通され、主流路において懸濁液にシース流を合流させるシース液流路と、懸濁液供給口に接続され、懸濁液を主流路に供給するための第1チューブと、シース液供給口に接続され、シース液を主流路内に供給するための第2チューブとを有し、第1チューブにおいて懸濁液の先頭の待機位置が予め定められ、第2チューブにおいてシース液の先頭の待機位置が予め定められており、ポンプにより、懸濁液及びシース液の全ての先頭が、待機位置に到達した後に、懸濁液及びシース液の送液を開始させる制御部を更に有する、発明[11]に記載のEP装置である。
 発明[13]は、電極対の間において懸濁液及びシース液の流れの安定を判定する判定部を有し、判定部は、電極対の間における懸濁液及びシース液の流れの電気抵抗を測定して、電気抵抗に基づいて、流れの安定を判定するか、又は、判定部は、第1チューブの懸濁液の流量及び第2チューブのシース液の流量の安定性を検出して流れの安定を判定する、発明[12]に記載のEP装置である。
 発明[14]は、電極対の主流路における上流側又は下流側に設けられた補助電極を有し、判定部は、補助電極を用いて懸濁液及びシース液の流れの厚み方向の電気抵抗を測定して、電気抵抗に基づいて、流れの安定を判定する、発明[13]に記載のEP装置である。
 発明[15]は、第1チューブにおける懸濁液の先頭の位置、及び第2チューブにおけるシース液の先頭の位置を検知するセンサを有し、制御部は、センサにより検知された、第1チューブにおける懸濁液の先頭の位置、及び第2チューブにおけるシース液の先頭の位置に基づいて、ポンプによる懸濁液及びシース液の送液を制御する、発明[12]~[14]のいずれか1つに記載のEP装置である。
 発明[16]は、生体由来物と生体活性物質とを含有する懸濁液を連続的に送液して電界を印加することで、生体由来物に生体活性物質を導入する、EP装置であって、主流路と、対向して配置された電極を備え、主流路を流れる懸濁液に電界を印加する電極対と、懸濁液に電界を印加する電源部と、主流路に懸濁液を供給する懸濁液供給口と、主流路内の懸濁液供給口の下流側に設けられ、主流路にシース液を供給するシース液供給口と、懸濁液及びシース液を送液するポンプと、懸濁液供給口に接続され、懸濁液を主流路に供給するための第1チューブと、シース液供給口に接続され、シース液を主流路内に供給するための第2チューブと、第1チューブにおける懸濁液の先頭の位置、及び第2チューブにおけるシース液の先頭の位置を検知するセンサと、ポンプによる懸濁液及びシース液の送液を制御する制御部とを有し、第1チューブにおいて懸濁液の先頭の待機位置が予め定められ、第2チューブにおいてシース液の先頭の待機位置が予め定められており、制御部は、センサにより懸濁液及びシース液の全ての先頭が、待機位置に到達したことが検知された後に、ポンプにより懸濁液及びシース液の送液を開始させ、懸濁液及びシース液の送液後、電極対の間において懸濁液及びシース液の流れが安定した後に、電源部により懸濁液に電界を印加する、EP装置である。
 発明[17]は、制御部は、ポンプにより懸濁液及びシース液の送液を開始し、予め設定した時間経過後に、電源部により懸濁液に電界を印加する、発明[12]~[16]のいずれか1つに記載のEP装置である。
 発明[18]は、第1チューブにおける懸濁液の先頭の位置を検知するセンサは、超音波センサである、発明[15]又は[16]に記載のEP装置である。
 本発明によれば、生産性が高いフロー式のエレクトロポレーションを行うことができるエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置を提供できる。

 以下に、添付の図面に示す好適実施形態に基づいて、本発明のエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置を詳細に説明する。
 なお、以下に説明する図は、本発明を説明するための例示的なものであり、本発明を説明するために簡略化等をしており、以下に示す図に本発明が限定されるものではない。
 以下において数値範囲を示す「~」とは両側に記載された数値を含む。例えば、εが数値εα~数値εβとは、εの範囲は数値εαと数値εβを含む範囲であり、数学記号で示せばεα≦ε≦εβである。
 また、特定の角度、平行及び直交並びに鉛直等について、特に記載がなければ、該当する技術分野で一般的に許容される誤差範囲を含む。更に、長さ、幅、及び厚さ等に関し、特に記載がなければ、該当する技術分野で一般的に許容される誤差範囲を含む。また、温度、流量、電気伝導度、導電率及び体積分率等に関し、特に記載がなければ、該当する技術分野で一般的に許容される誤差範囲を含む。
(エレクトロポレーション装置の第1の例)
 図1は本発明の実施形態のエレクトロポレーション装置(EP装置)を有するエレクトロポレーションシステムの一例を示す模式図である。図2は本発明の実施形態のエレクトロポレーション装置の流路デバイスの第1の例を示す模式的断面図である。図3は本発明の実施形態のエレクトロポレーション装置の第1の例の流路デバイスの作用を説明するための模式的断面図である。図2は図1のA-A線による模式的断面図である。
 図1及び図2に示すEP装置10は、対向して配置された電極を備える電極対34の間を生体由来物と生体活性物質とを含有する懸濁液Q(図3参照)と、シース液s(図3参照)とを、シース液sで懸濁液Qを挟むようにして主流路30に連続的に送液し、電極対34によって懸濁液Qに電界を印加することで、生体由来物に生体活性物質を導入するエレクトロポレーションを実施する装置である。
 エレクトロポレーション装置10は、図3に示すように懸濁液Qとシース液sを用いるフロー式のEP装置であり、シース液sで懸濁液Qを挟むようにして電極対34の間に連続的に送液してEPを実施する。
 電極対34の間に、懸濁液Q及びシース液sを、シース液sで懸濁液Qを挟んで連続的に送液してEPを実施することを連続エレクトロポレーションともいう。シース液sで懸濁液Qを挟むようにして送液しており、2つのシース液sの流れ、すなわち、2つのシース液流を有する。シース液流、懸濁液流及びシース液流の3層の積層流れとなる。
 EP装置10は、図1、2に示すように流路デバイス11と、流路デバイス11に懸濁液Qを供給するポンプ12a、12bと、EPを実施する電源部14と、制御部16と、判定部18とを有する。更に、流路デバイス11にシース液sを供給するポンプ20a、20bと、シース液sを貯留するタンク22とを有する。
 また、EP装置10のポンプ12aは、例えば、懸濁液の供給部である培養装置24に、配管15により接続されている。混合器25と培養装置24との間にポンプ12aが設けられている。培養装置24は、EP装置10を構成するものではない。
 また、生体活性物質を貯留するタンク23及び混合器25もEP装置10を構成するものではない。懸濁液の供給部としては、図1では培養装置24を示しているが、培養装置24から取り出された懸濁液を貯留するタンクでもよい。
 また、流路デバイス11は、流路デバイス11から外部に排出された懸濁液Q及びシース液s等の液体を貯留する廃棄槽26及び容器27と第3チューブ28により接続されている。第3チューブ28にバルブ29が設けられている。第3チューブ28は、流路デバイス11から外部に排出された懸濁液Q及びシース液s等の液体の流路を構成する。バルブ29は送液先を廃棄槽26又は容器27に切り替える。流路デバイス11から外部に排出された液体は、第3チューブ28を通過し、バルブ29により廃棄槽26又は容器27に送液される。
 なお、廃棄槽26、容器27及びバルブ29は、EP装置10を構成するものではない。
 EP装置10に、培養装置24、後述のタンク23及び混合器25、並びに廃棄槽26、容器27及びバルブ29を含めてエレクトロポレーションシステム40ともいう。
 流路デバイス11は、図1及び図2に示すように主流路30を有する。主流路30は直線状の流路である。また、主流路30は、延在方向Dと直交方向における断面の形状が、例えば、内角が全て90°の四角形である。主流路30の上述の断面の形状は、内角が全て90°の四角形に限定されるものではない。
 図2に示すように主流路30の一方の端30aに、懸濁液Qを主流路30に供給するための流入口31が、例えば、流路デバイス11の表面11aに設けられている。主流路30の他方の端30bに、懸濁液Q及びシース液s等の液体を外部に排出するための排出口32が、例えば、流路デバイス11の裏面11bに設けられている。主流路30において、流入口31側が上流側であり、排出口32側が下流側である。
 さらに、図2に示すように、主流路30の一方の端30aに、シース液sを主流路30に供給するための第1流入口31aが、例えば、流路デバイス11の表面11aに設けられている。シース液sを主流路30に供給するための第2流入口31bが、例えば、流路デバイス11の裏面11bに設けられている。例えば、第1流入口31aと第2流入口31bとは延在方向Dにおける位置が同じである。
 流入口31に第1チューブ13が接続されている。流入口31に接続された第1チューブ13及び配管15を介して、流入口31にポンプ12aが接続されている。第1チューブ13は、懸濁液Qを挟むようにシース液sを主流路30に送液する際に、懸濁液Qの供給に用いられるものである。
 第1チューブ13及びポンプ12aの構成は、特に限定されるものではなく、懸濁液Qの粘度、懸濁液Qの送液量等により、当該技術分野において公知なもの等を適宜利用可能である。第1チューブ13は、例えば、シリコンチューブのような可撓性を有するものでもよい。
 第1流入口31aに第2チューブ21aが接続されている。第1流入口31aに接続された第2チューブ21aを介して、第1流入口31aにポンプ20aが接続されている。ポンプ20aに配管21cを介してタンク22が接続されている。
 第2流入口31bに第2チューブ21bが接続されている。第2流入口31bに接続された第2チューブ21bを介して、第2流入口31bにポンプ20bが接続されている。ポンプ20bに配管21dを介してタンク22が接続されている。
 シース液sを供給する第1流入口31aと第2流入口31bがあるため、第2チューブ21a、21bが2つある。2つの第2チューブ21a、21bは、それぞれ懸濁液Qを挟むようにシース液sを主流路30に送液する際に、シース液sの供給に用いられるものである。
 第2チューブ21a、21b及びポンプ20a、33bの構成は、特に限定されるものではなく、シース液sの粘度、シース液sの送液量等により、当該技術分野において公知なもの等を適宜利用可能である。第2チューブ21a、21bは、例えば、シリコンチューブのような可撓性を有するものでもよい。
 ポンプ20aとポンプ20bを設ける構成としたが、これに限定されるものではなく、第1流入口31a及び第2流入口31bに対して、ポンプを1つ設ける構成でもよい。すなわち、シース液sを1つのポンプで流路デバイス11に供給するようにしてもよい。
 タンク22は、シース液sを貯留するものである。タンク22はシース液sを貯留することができれば、その構成は、特に限定されるものではない。
 上述のように流入口31を流路デバイス11の表面11aに設け、排出口32を流路デバイス11の裏面11bに設ける構成としたが、これに限定されるものではない。例えば、流入口31及び排出口32を、いずれか一方を流路デバイス11の表面11aに設け、他方を裏面11bに設ける構成としてもよい。
 図2に示す流路デバイス11は、水平面Hpに対して、例えば、直線状の主流路30の延在方向Dが直交するように配置されており、主流路30の延在方向Dは鉛直方向zと平行である。主流路30の幅方向は、鉛直方向zと直交する方向xと平行な方向であり、主流路30の延在方向Dと直交する方向でもある。
 上述の水平面Hpに対して、例えば、直線状の主流路30の延在方向Dが直交するとは、水平面Hpに対する主流路30の延在方向Dの角度が90°であるが、90°に対して±10°許容する。
 このため、水平面Hpに対する主流路30の延在方向Dの角度が90°の場合、主流路30の延在方向Dは鉛直方向zと平行であるが、主流路30の延在方向Dは鉛直方向zに対して、±10°の傾きを許容する。
 主流路30の延在方向Dが、主流路30内を懸濁液Qが流れる方向Dfである。エレクトロポレーション装置10において、電極対の間における懸濁液Qの送液方向Ddは、鉛直方向の下方から上方に向かう方向である。主流路30内を懸濁液Qが、鉛直方向の下方から上方に向かう方向に流れる。
 懸濁液Qは、流入口31から供給されて、主流路30を図2に示す方向Dfに沿って流れて排出口32から排出される。
 シース液sは、第1流入口31aと第2流入口31bから供給されて、主流路30を図2に示す方向Dfに沿って流れて排出口32から排出される。
 図1及び図2に示す方向Dfは、主流路30の上流から下流に向かう方向でもある。
 培養装置24は、生体由来物と培地を含む培養液(図示せず)を得るものである。培養装置24は、例えば、細胞培養により、培地を含む培養液を得る。
 混合器25にタンク23が接続されている。混合器25とタンク23との間にポンプ12bが設けられている。
 タンク23に生体活性物質が、例えば、液体に分散した状態で貯留されている。タンク23は生体活性物質を貯留することができれば、その構成は、特に限定されるものではない。
 混合器25は、培養液と生体活性物質とを混合するものである。培養液と生体活性物質とが混合されたものを懸濁液Qといい、懸濁液Qは培地を含む。
 ポンプ12aを用いて培養装置24内の培養液を吸引し、ポンプ12bを用いてタンク23内の生体活性物質を吸引することにより、培養液の培地をエレクトロポレーション(EP)用バッファーと交換することなく、混合器25で培養装置24の培養液と、タンク23の生体活性物質とが混合され、懸濁液Qとなる。
 混合器25は、培養液と生体活性物質とを混合することができれば、その構成は、特に限定されるものではなく、公知のものを利用可能である。
 懸濁液Qがポンプ12a、12bにより、第1チューブ13及び流入口31を通り、主流路30に供給される。このように培養液の培地を交換せずに、培地を含む懸濁液Qが主流路30に供給される。懸濁液Qは、電極対34の間を鉛直方向の下方から上方に向かう方向に送液される。
 なお、培養装置24は必ずしも必要ではない。培養装置24に換えて、例えば、懸濁液Qが貯留されたタンク(図示せず)としてもよい。この場合、タンクがポンプ12aに接続される。
 上述のポンプ12a、12bは、培養装置24等の懸濁液の供給部から懸濁液を送液するポンプである。このようなポンプ12a、12bは特に限定されるものではなく、公知のものを適宜利用可能であるが、シリンジポンプ等が挙げられる。
 排出口32に第3チューブ28が接続されている。第3チューブ28は、例えば、シリコンチューブのような可撓性を有するものでもよい。
 廃棄槽26は、流路デバイス11から外部に排出された懸濁液Q及びシース液s等の液体のうち、後培養に適さないもの等を貯留する。
 容器27は、流路デバイス11から外部に排出された懸濁液Q及びシース液s等の液体のうち、後培養に利用するものを貯留する。
 廃棄槽26及び容器27は、いずれも流路デバイス11から外部に排出された懸濁液Q及びシース液s等の液体を貯留することができれば、その構成は、特に限定されるものではない。
 廃棄槽26には、例えば、懸濁液Q及びシース液sの送液開始から電界印加迄に、排出口32から排出された懸濁液Q及びシース液sが送液されて貯留される。
 容器27には、例えば、懸濁液Qへの電界印加後、予め設定された時間経過後、エレクトロポレーションされた懸濁液Qが送液されて貯留される。
 バルブ29は、送液先を廃棄槽26又は容器27に切り替えることができれば、その構成は特に限定されるものではなく、公知のバルブを適宜利用可能である。また、バルブ29は制御部16により制御可能なものでもよく、この場合、制御部16により送液先が廃棄槽26又は容器27に切り替えられる。
 主流路30において、図3に示すように懸濁液流とシース液流とが合流する合流部59の下流側に、電極対34が設けられている。電極対34は、対向して配置された電極を備え、主流路30を流れる懸濁液Qに電界を印加する。
 懸濁液Qに電界を印加する電極対34は、対向して配置された第1電極35と、第2電極36とを有する。第1電極35は電極面35aを有し、電極面35aは平面である。第2電極36は電極面36aを有し、電極面36aは平面である。第1電極35の電極面35aと、第2電極36の電極面36aとは対向し、かつ平行に配置されている。
 第1電極35の電極面35aと、第2電極36の電極面36aとは、それぞれ主流路30の内面30cに面して配置されている。第1電極35の電極面35aと、第2電極36の電極面36aとは、それぞれ主流路30の内面30cを構成しており、例えば、エレクトロポレーションを実施する際に、主流路30に懸濁液Qとシース液sとが供給されると、シース液sと接する。
 電極対34を構成する第1電極35の電極面35aと、第2電極36の電極面36aとの対向している間の空間37が、電極対34の間である。
 電極対34の第1電極35と第2電極36とは、図2に示すように電源部14に電気的に接続されている。電源部14により電極対34に電圧が印加される。すなわち、電源部14により、対向する第1電極35と第2電極36とに電圧が印加される。
 電源部14は、例えば、電極対34に、パルス電圧を印加し、パルス幅及びパルス周期を調整できるものであれば、その構成は、特に限定されるものではない。電源部14には、例えば、パルス電源が用いられる。
 また、判定部18は、電極対34の間において懸濁液Q及びシース液sの流れの安定を判定する。判定部18は、流れが安定した情報を含む判定信号を制御部16に出力し、制御部16は判定信号を受信する。制御部16は、受信した判定信号に基づいて、電源部14により懸濁液Qに電界を印加する等の各部を制御する。電極対34の間における懸濁液及びシース液の流れの安定の判定について後に説明する。
 例えば、第1チューブ13の流入口31の近傍にセンサ38が設けられている。また、例えば、第2チューブ21aの第1流入口31aの近傍にセンサ39aが設けられており、第2チューブ21bの第2流入口31bの近傍にセンサ39bが設けられている。
 流入口31の近傍とは、流入口31から10cm以内の範囲であり、好ましくは5cm以内の範囲である。
 第1流入口31aの近傍とは、第1流入口31aから10cm以内の範囲であり、好ましくは5cm以内の範囲である。
 第2流入口31bの近傍とは、第2流入口31bから10cm以内の範囲であり、好ましくは5cm以内の範囲である。
 懸濁液Qの先頭の位置とは、第1チューブ13において懸濁液Qの流路デバイス11に最も近い位置の液面のことである。
 シース液sの先頭の位置とは、第2チューブ21aにおいてシース液sの流路デバイス11に最も近い位置の液面のことであり、第2チューブ21bにおいてシース液sの流路デバイス11に最も近い位置の液面のことである。
 センサ38は第1チューブ13における懸濁液Qの先頭の位置を検知するものである。センサ38は、例えば、超音波センサが用いられる。
 超音波センサでは、超音波は気体よりも液体及び固体中を伝搬しやすい性質があるため、生体由来物を用いる場合には誤検知されることが少ない。また、上述のように超音波は気体よりも液体及び固体中を伝搬しやすい性質があるため、懸濁液Qの生体由来物の濃度が高い場合であっても、検知精度の低下が抑制される。このようなことから、懸濁液Qを検知に用いるセンサ38は、超音波センサであることが好ましい。
 センサ39a、39bは、第2チューブ21a、21bにおけるシース液sの先頭の位置を検知するものである。センサ39a、39bは、第2チューブ21a、21bにおけるシース液sの先頭の位置を検知できれば、その構成は、特に限定されるものではない。
 また、センサ38は、第1チューブ13において懸濁液Qの先頭を検知した場合、制御部16に検知信号を送信することが好ましい。センサ39a、39bは、第2チューブ21a、21bにおいてシース液sの先頭を検知した場合、制御部16に検知信号を送信することが好ましい。制御部16は、上述のセンサ38、センサ39a及びセンサ39bからの検知信号を受信する。
 エレクトロポレーション装置10では、第1チューブ13において懸濁液Qの先頭の待機位置が予め定められており、第2チューブ21a、21bおいてシース液sの先頭の待機位置が予め定められている。このため、センサ38を、第1チューブ13における懸濁液Qの先頭の予め定められた待機位置に配置することが好ましい。上述の第1チューブ13の待機位置は、例えば、第1チューブ13の流入口31の近傍である。
 また、センサ39a、39bを、第2チューブ21a、21bおけるシース液sの先頭の予め定められた待機位置に配置することが好ましい。上述の第2チューブ21aの待機位置は、例えば、第2チューブ21aの第1流入口31aの近傍である。上述の第2チューブ21bの待機位置は、例えば、第2チューブ21bの第2流入口31bの近傍である。
 EP装置10の制御部16は、ポンプ12と電源部14と、判定部18と、ポンプ20a、20bと、センサ38、39a、39bとに接続されている。制御部16によりポンプ12a、12b及びポンプ20a、20bの動作が制御され、懸濁液Qとシース液sの送液の開始のタイミングと、懸濁液Qとシース液sの送液の停止のタイミングが制御される。
 また、制御部16により、電源部14が電極対34に印加する電圧の大きさ、パルス幅及びパルス周期が調整される。また、制御部16は、ポンプ12a、12b及びポンプ20a、20bの動作のタイミング、すなわち、懸濁液Qとシース液sの送液のタイミングと、電源部14の電極対34への電圧の印加のタイミングを調整する。
 制御部16は、判定部18による電極対34の間における懸濁液及びシース液の流れの安定の判定に基づいて、ポンプ12a、12b及びポンプ20a、20bの動作のタイミング、すなわち、懸濁液Qとシース液sの送液のタイミングと、電源部14の電極対34への電圧の印加のタイミングを調整することもできる。
 また、制御部16及び判定部18は、時間を計測する機能を有し、例えば、時間を計測するクロック(図示せず)を有する。制御部16は、クロックにより、ポンプ12a、12b、20a、20bの動作のタイミングを調整でき、決まった時間差で、ポンプ12a、12b、20a、20bを動作させることができる。これにより、懸濁液Q及びシース液sの送液のタイミングを制御できる。
 また、判定部18は、クロックにより、電極対の間における懸濁液Q及びシース液sの流れが安定した状態を判定できる。
 なお、制御部16及び判定部18は、例えば、プログラムが実行されることで機能するコンピュータによって構成されてもよいし、専用回路で構成された専用装置であってもよい。制御部16及び判定部18は、流路デバイス11とは別に、例えば、クラウド上に設けられた仮想的な構成とすることもでき、例えば、クラウド上で実行されるようにサーバーで構成してもよい。
 制御部16及び判定部18を上述のように仮想的な構成とする場合、制御部16はポンプ12a、12b及びポンプ20a、20b並びに電源部14と、インターネット又はLAN(local area network)等を介して接続される。
 上述のコンピュータ、専用装置及びサーバーは、例えば、プロセッサを有する。プロセッサは、1又は複数のハードウェアによって構成してもよく、ハードウェアの種類は限定されない。例えば、プロセッサは、CPU(Central Processing Unit)、MPU(Micro Processing Unit)、FPGA(Field Programmable Gate Array)等のプログラマブルロジックデバイス、ASIC(Application Specific Integrated Circuit)等の特定の処理を実行させるための専用回路、GPU(Graphic Processing Unit)、又はNPU( Neural Processing Unit)等のハードウェアによって構成され得る。また、プロセッサは、本実施形態における各種の処理を実行する各部(Unit)、又は各手段(Means)を有している。また、ハードウェアの種類は、異なる種類のハードウェアを組み合わせたものであってもよい。複数のハードウェアが、あるプロセッサの1または複数の処理を実行するように構成される場合、当該複数のハードウェアは互いに物理的に離れた装置内に存在していてもよく、同じ装置内に存在していてもよい。また、いずれの実施形態においても、プロセッサによる各処理の順序は上記した順序に限定されるものではなく、適宜変更してもよい。なおハードウェアは半導体素子等の回路素子を組み合わせた電気回路(circuitry)等により構成される。
 EP装置10において、流路デバイス11は、例えば、図2に示すように、第1基板52と、第2基板54と、第1導入部56と、第2導入部58と、第1基板52及び第2基板54の間に設けられる流路基板55とを有して構成される。第1導入部56、第1基板52、流路基板55、第2基板54、及び第2導入部58の順で積層されている。
 第1基板52に第1電極35が、第2基板54に第2電極36が、それぞれ組み込まれ、電極対34を構成する。上述のように第1電極35の電極面35aと、第2電極36の電極面36aとは、それぞれ主流路30の内面30cを構成している。
 上述のように、流路デバイス11は、懸濁液Qを供給する流入口31、主流路30、フロー式のEPを行った処理済懸濁液を排出する排出口32、シース液sを流入する第1流入口31a及び第2流入口31bを有する。主流路30は、第1基板52と第2基板54との間に略直線状に形成される。
 第1導入部56、第1基板52、流路基板55、第2基板54及び第2導入部58の積層方向Dsに延在する貫通孔56bが、第1導入部56に設けられている。また、第1基板52に、第1導入部56の貫通孔56bと連通し、積層方向Dsに延在する貫通孔52bが設けられている。貫通孔52bは主流路30の最上流部と連通する。第1導入部56の貫通孔56bの第1基板52の反対側の開口が流入口31である。
 流入口31は、第1導入部56の貫通孔56b及び第1基板52の貫通孔52bにより主流路30の最上流部と連通する。
 主流路30(流路基板55)の内面30cに、電極対34における電極の対向方向、すなわち、第1電極35と第2電極36との対向方向に突出した凸部55a及び凸部55bが、最上流部から下流に向かって設けられている。
 なお、積層方向Dsは、鉛直方向zと直交する方向yと平行な方向である。また、電極対34における電極の対向方向は、鉛直方向zと直交する方向yと平行な方向である。
 また、第1基板52に、主流路30と連通する貫通孔52aが設けられている。第2基板54に、主流路30と連通する貫通孔54aが設けられている。貫通孔52a及び貫通孔54aは、主流路30に対して傾斜して設けられている。
 第1導入部56に、第1基板52の貫通孔52aと連通する貫通孔56aが設けられている。
 また、第2導入部58に、第2基板54の貫通孔54aと連通する貫通孔58aが設けられている。貫通孔56aは、第1流入口31aと第1基板52の貫通孔52aと連通するものであり、第1流入口31aから積層方向Dsに延びており、第1基板52との境界で主流路30の延在方向Dに屈曲している。このような形状の貫通孔56aを便宜的に略L字状の貫通孔56aという。
 貫通孔58aは、第2流入口31bと第2基板54の貫通孔54aと連通するものであり、第2流入口31bから積層方向Dsに延びており、第2基板54との境界で主流路30の延在方向Dに屈曲している。このような形状の貫通孔58aを便宜的に略L字状の貫通孔58aという。
 第1導入部56の貫通孔56a及び第1基板52の貫通孔52aと、第2導入部58の貫通孔58a及び第2基板54の貫通孔54aとは、鉛直方向zに延在する主流路30に対して対称に配置されている。
 第1流入口31aは、第1導入部56に設けられた略L字状の貫通孔56aを経て、第1基板52に設けられた貫通孔52aを経て主流路30と連通する。ここで、貫通孔52aは、主流路30の延在方向Dに対して、下流方向に向かって傾斜しており、主流路30の凸部55aの直下流に開口する。
 なお、貫通孔56a(シース液流路の一部)は、シース液流の幅方向の流速が均一になるように、例えば、略L字状の流路が曲がった位置から下流に向けて、流路を幅方向に広げてもよい。この点に関しては後述する貫通孔58aも同様である。
 また、貫通孔56aは第1導入部56及び第1基板52に跨って設けられていてもよい。
 他方、第2流入口31bは、第2導入部58に設けられた略L字状の貫通孔58aを経て、第2基板54に設けられた貫通孔54aを経て主流路30と連通する。ここで、貫通孔54aは、主流路30の延在方向Dに対して、下流方向に向かって傾斜しており、主流路30の凸部55bの直下流に開口する。なお、貫通孔58aは、第2導入部58及び第2基板54に跨って設けられていてもよい。凸部55a及び凸部55bの下流側端部55cが、流路デバイス11において、主流路30に懸濁液Qを供給する懸濁液供給口55dとなる。この下流側端部55c、すなわち、懸濁液供給口55dに貫通孔52b、貫通孔56b及び流入口31を経て第1チューブ13が接続されている。
 また、貫通孔52aの主流路30への開口部、及び貫通孔54aの主流路30への開口部が、流路デバイス11における主流路30へのシース液sのシース液供給口52c、54cとなる。なお、懸濁液Qの懸濁液供給口とシース液sのシース液供給口52c、54cとの幅は、ほぼ同一であることが好ましい。
 また、流路デバイス11において、下流側端部55cが懸濁液流とシース液流とが合流する合流部59となる。
 主流路30に電極対34の電極に接触するシース液流を形成するシース液供給口52cが主流路30内の懸濁液供給口55dの下流側に設けられている。
 貫通孔52aと貫通孔56aとにより、主流路30において合流部59で懸濁液Qにシース液流を合流させるシース液流路64が形成される。シース液流路64に第2チューブ21aが接続されている。第2チューブ21aは第1流入口31a、貫通孔56a及び貫通孔52aを経てシース液供給口52cに接続されている。
 主流路30に電極対34の電極に接触するシース液流を形成するシース液供給口54cが主流路30内の懸濁液供給口55dの下流側に設けられている。
 貫通孔54aと貫通孔58aとにより、主流路30において合流部59で懸濁液Qにシース液流を合流させるシース液流路65が形成される。シース液流路65に第2チューブ21bが接続されている。第2チューブ21bは第2流入口31b、貫通孔58a及び貫通孔54aを経てシース液供給口54cに接続されている。
 シース液流路64、65は、主流路30における液体の流れ方向に対して、下流方向に向かって傾斜するように設けられ、懸濁液Qの供給口の下流であって電極対34の上流にシース液sが流入するように開口する。主流路30の上流側であって、懸濁液Qのみが流れる領域を懸濁液流路ということがあり、懸濁液流路の下流側であり、凸部55a及び凸部55bに挟まれた領域が規制領域となる。シース液流路64、65は、シース液のみが流れる。
 シース液流路64、65は、下流に向けて流路が幅方向に広がる領域を有することが好ましい。この結果、主流路30の幅が広くても、下流での流速分布が均一になり、懸濁液Qに対して均一にEPできる。このとき、懸濁液流路にも下流に向けて流路が幅方向に広がる領域を有することが好ましく、懸濁液流路でも流速分布が均一になる。
 なお、流路デバイス11において、流路基板55は、公知の各種の方法で形成すればよい。また、複数枚の流路板を用いて、流路基板55を形成してもよい。一例として、凸部55aに対応する部分(非開口部)を有し、主流路30に対応する開口を有する流路板、主流路30全域に対応する開口を有する流路板、及び、凸部55bに対応する部分(非開口部)を有し、主流路30に対応する開口を有する流路板の、3枚の流路板を用いて流路基板55を形成してもよい。
 流路デバイス11において、懸濁液Qは、第1チューブ13から流入口31に供給され、最上流部から主流路30に流入して、主流路30から排出口32に至る。
 シース液sは、第2チューブ21aから第1流入口31aに供給され、第2チューブ21bから第2流入口31bに供給される。
 第1流入口31aに流入されたシース液sは、第1導入部56に設けられた貫通孔56a及び第1基板52に設けられた貫通孔52aを経て、すなわち、シース液流路64を経て主流路30に供給され、主流路30から排出口32に至る。また、第2流入口31bに流入されたシース液sは、第2導入部58に設けられた貫通孔58a及び第2基板54に設けられた貫通孔54aを経て、すなわち、シース液流路65を経て主流路30に供給され、主流路30から排出口32に至る。
 ここで、上述のように、主流路30内(流路基板55)には、電極対34における第1電極35と第2電極36との対向方向に突出して、最上流部から下流に向かって、凸部55a及び凸部55bが設けられている。凸部55a及び凸部55bを有する領域では、懸濁液Qの厚さDmは凸部55aと凸部55bとの間隔dである。
 また、上述のように、貫通孔52aは、主流路30における液体の流れ方向に対して、下流方向に向かって傾斜するように設けられ、主流路30の凸部55a(凸部55aと凸部55bとで挟まれた懸濁液流路の領域を規制領域ということがある)の直下流に開口する。上述のように、凸部55a及び凸部55bで挟まれた領域が規制領域である。他方、貫通孔54aは、主流路30における液体の流れ方向に対して、下流方向に向かって傾斜するように設けられ、主流路30の凸部55bの直下流に開口する。
 なお、図2に示す態様では、シース液流路が貫通孔56a(58a)及び貫通孔52a(54a)から構成されているが、シース流路としては貫通孔52a(54a)のみがあればよく、流路構成はデバイス構成に応じて適宜設計される。
 貫通孔56a及び貫通孔52aで構成されているシース液流路64は、シース液流路64としては貫通孔52aのみがあればよく、シース液流路の構成は、流路デバイスの構成に応じて適宜決定される。
 また、貫通孔54aと貫通孔58aで構成されているシース液流路65は、シース液流路65としては貫通孔54aのみがあればよく、シース液流路の構成は、流路デバイスの構成に応じて適宜決定される。
 従って、電極対34の上流では、図3に概念的に示すように、懸濁液Qによる懸濁液流の対向方向の両側に、シース液sからなるシース液流を形成して、シース液流/懸濁液流/シース液流の3層の積層流が形成され、電極対34の間には3層の積層流が流入する。
 EP装置10では、懸濁液Qとシース液sとが、シース液流/懸濁液流/シース液流の積層流を形成した状態で電極対34の間、すなわち、上述の空間37に連続的に送液されて、第1電極35及び第2電極36によって、3層の積層流の状態で懸濁液Qに、例えば、パルス電界を印加することにより、上述のようにシース液流を用いたフロー式のEPによる処理を行うことができる。この場合、懸濁液流の厚さは規制領域の厚さと略等しくなり、少なくとも電極対34の間、かつ電極対34の上流端部で懸濁液流の厚さDmが1~10mmに維持される。
 なお、図3では、図2に示す第1チューブ13、第2チューブ21a、21b、ポンプ12a、12b、20a、20b、タンク22、培養装置24及びタンク23、電源部14、制御部16、判定部18及び混合器25の図示を省略している。図3では、センサ38、39a、39bの図示も省略している。
 上述のように第1流入口31aから供給されたシース液sは、下流方向に向かって傾斜するように主流路30の凸部55aの直下流に流入する。他方、第2流入口31bから供給されたシース液sは、下流方向に向かって傾斜するように主流路30の凸部55bの直下流に流入する。
 また、シース液sの流入部の上流では、主流路30に凸部55a及び凸部55bが形成されており、シース液sの流入位置よりも下流に比して、主流路30の対向方向の厚さが薄くなっている。また、この領域における主流路30の対向方向の厚さ、すなわち、懸濁液流の厚さDmは、凸部55a及び凸部55bの間隔で決定する。
 このように凸部55a及び凸部55bはシース液流を形成し、かつ懸濁液流の厚さDmを規制する機能を有する。流路デバイス11では、主流路30における凸部55a及び凸部55bが形成された領域が、シース液流との合流前の懸濁液流の厚さを規制する規制領域となる。シース液流との合流前の懸濁液流の厚さを規制する規制領域を懸濁液流路ということがある。
 従って、流路デバイス11においては、凸部55aと凸部55bとの間隔dを1~10mmとすることにより、規制領域を流れる懸濁液流の厚さDmを1~10mmとする。
 これにより、流路デバイス11においては、上述のフロー式のEPにおけるシース液流/懸濁液流/シース液流の3層の積層流れにおいて、懸濁液流の厚さDm(図3参照)を1~10mmに制御できる。なお、懸濁液流路は、その一部が規制領域となっていればよく、下流側の一部が規制領域となり間隔dが1~10mmになっていればよい。
 この規制領域における懸濁液流の厚さDm、すなわち、間隔d(図2参照)は、1~8mmが好ましく、2~5mmがより好ましく、2~3mmが更に好ましい。規制領域の厚さの下限は、2mmが好ましく、3mmがより好ましい。間隔d(懸濁液流の厚さDm)は、2~10mmでもよく、3~10mmでもよい。
 また、凸部55a及び凸部55bの厚さの合計、すなわち、電極の対向方向の高さの合計を、凸部55aと凸部55bとの間隔d以下とすることにより、シース液流の厚さの合計を、懸濁液流の厚さ以下として、フロー式のEPを行った後の懸濁液の希釈率を2倍以下にできる。
 また、シース液sによるシース液流の厚さds(図3参照)に制限はないが、懸濁液流との合流時におけるシース液流の厚さは、0.1mm以上が好ましく、0.2mm以上がより好ましく、0.3mm以上が更に好ましく、0.5mm以上が特に好ましい。2つのシース液の厚さの合計を、懸濁液流の厚さDm以下とすることにより、シース液流の厚さの合計を、懸濁液流の厚さDm以下として、フロー式のEPを行った後の懸濁液の希釈率を2倍以下にできる。このため、2つのシース液流の厚さds(図3参照)の合計を、懸濁液流の厚さDm以下とすることが好ましい。すなわち、懸濁液流との合流時におけるシース液流の厚さの合計は、規制領域における懸濁液流の厚さの1倍以下であることが好ましい。
 懸濁液流の厚さDmは、上述のように凸部55aと凸部55bとの間隔d(図2参照)によって決定される。凸部55aと凸部55bは、懸濁液流の厚さDmの規制部材として機能する。このため、凸部55aと凸部55bの対向方向の厚さを変えることにより、懸濁液流の厚さDmを調整できる。この場合、シース液流の厚さds(図3参照)も調整される。
 このような流路デバイス11において、懸濁液流とシース液流とを合流させる際における懸濁液流とシース液流とが成す角度、すなわち、主流路30と、貫通孔52a及び貫通孔54aとが成す角度には、制限はないが、小さい方が好ましい。
 具体的には、主流路30の延在方向Dと、貫通孔52aの延在方向及び貫通孔54aの延在方向とが成す角度は、90°以下が好ましく、60°以下がより好ましく、45°以下が更に好ましく、30°以下が更に一層好ましい。
 また、懸濁液流とシース液流とを合流させる際における両者は流速差が小さいことが好ましい。具体的には、懸濁液流とシース液流とを合流させる際におけるシース液流の流速は、懸濁液流の流速の±10%以下であることが好ましく、±5%以下であることがより好ましく、±3%以下であることが更に好ましく、±0%、すなわち、懸濁液流とシース液流とが等速であることが最も好ましい。
 図2の流路デバイス11において、第1導入部56と第1基板52との間、第1基板52と流路基板55との間、流路基板55と第2基板54との間、第2基板54と第2導入部58との間には、懸濁液Qの液漏れ防止用のシール部材(図示せず)としてOリングがあることが好ましい。
 EP装置10では、ポンプ12a、ポンプ12b、ポンプ20a、ポンプ20bは、電極対34の間、すなわち、上述の空間37において、懸濁液Q及びシース液sの送液方向Ddが鉛直方向zの下方から上方に向かう方向にとなるように、懸濁液Q及びシース液sを送液する。電源部14を用いて懸濁液Qに電界を印加してEPを実施し、対向する第1電極35と第2電極36との間に気泡が発生した場合、発生した気泡は、懸濁液Q及びシース液sに比して比重が小さいため、鉛直方向zに懸濁液Q及びシース液sよりも移動しやすい。このため、電極対34の間に発生した気泡は、懸濁液Q及びシース液sの送液により電極対34の間に滞留することなく、鉛直方向zに移動することで電極対34の間から除去され、気泡による懸濁液Qへの電界の印加の阻害を抑制できる。これにより、EP装置10では、懸濁液Qへの電界の印加を適正に実施でき、その結果、導入効率及び生体活性物質の利用効率を適正なものにできる。
 EP装置10では、図3に示すように、懸濁液流と共に、懸濁液流と第1電極35及び第2電極36との間に、懸濁液とは異なるシース液sからなるシース液流を形成して、フロー式のEPを行う。
 シース液sを用いることにより、懸濁液Qにおける生体由来物の濃度が高い場合でも、主流路30の壁面近傍に生体由来物の流れが滞留することを防止して、高効率のフロー式のEPを行うことができる。
 本発明者等の検討によれば、懸濁液Qにおける生体由来物の濃度が高いがシース液sがない場合、主流路30の壁面の極近傍で生体由来物の流れが滞留してしまう。生体由来物の流れが滞留する滞留層では、生体由来物が流動している領域に比して生体由来物が密着しているため、電気抵抗が高い。そのため、この滞留層によって電極対34が印加する電界を大量に消費してしまう。その結果、本来、電界を印加すべきである流動部に印加する電界が低くなって十分な電界を印加できず、EPの導入効率が低下してしまう。
 また、シース液sがない場合、生体由来物が電極付近で滞留層として滞留するため、生体由来物は、繰り返し電界を印加される。その結果、電極の近傍で生体由来物が焦げ付き、焦げ付いた生体由来物が電極の表面に付着してしまう。このような異物が電極表面に付着すると、電極が印加する電界を遮蔽してしまうので、EPの導入効率が低下してしまう。このことから、シース液sがあることが好ましい。
 さらに、本発明者等は、検討の結果、このようなシース液sを用いるフロー式のEPにおいて、電極対を構成する電極の対向方向における懸濁液流の厚さDmを厚くすることにより、高効率のフロー式のEPが行えることも見出した。
 具体的には、フロー式のEPにおいて、電極対を構成する電極の対向方向における懸濁液流の厚さDmを、1~10mmとすることで、高効率のフロー式のEPを行うことができる。なお、電極対の間の一部で懸濁液流の厚さDmが1~10mmとなっていればよく、電極対の間かつ電極上流端部で懸濁液流の厚さDmが1~10mmとなっていればよい。
 EP方法は、このように懸濁液流の厚さDmを厚くしてフロー式のEPを行うことにより、より高効率のフロー式のEPを行うことを可能にしている。具体的には、スループット、導入効率、及び生体活性物質の利用効率が高い、より高効率のフロー式のEPを行うことができる。なお、スループットとは、単位時間当たりの細胞処理量である。
 また、EP方法は、懸濁液流の厚さDmを厚くしてフロー式のEPを行うことにより、処理量を増大させつつ、高濃度の懸濁液に対して、効率的に大量のフロー式のEPを行うことができ、高い生産性を達成することができる。
 ここで、従来のフロー式のEPでは、懸濁液流の厚さを、できるだけ薄くして処理を行っている。
 例えば、特許文献2では、好ましい態様として、懸濁液流の厚さ、すなわち、シース液流間の間隔を50~100μmとして、フロー式のEPを行うことを例示している。特許文献2では、これにより、電極対の間を通過する生体由来物(細胞)が電極の対向方向に重ならないように流路内を流す単層流とすることができる、としている。
 従来のフロー式のEPでは、このように懸濁液流の厚さを薄くすることにより、電極対の間における生体由来物の流れを均一にし、かつ、生体由来物に印加する電界を均一にすることで、効率の高い処理を可能にしていると考えられる。
 しかしながら、本発明者等は、検討の結果、逆に懸濁液流の厚さを大幅に厚くすることで、より効率が良いフロー式のEPを行えることを見出した。
 フロー式のEPでは、懸濁液に含まれる生体由来物及び生体活性物質の拡散によって、シース液流と懸濁液流との界面に、生体由来物及び生体活性物質が拡散された拡散領域が形成される。
 特許文献2のように、懸濁液流の厚さを50~100μmとした場合、懸濁液流の厚さに対して拡散領域が厚くなる。その結果、フロー式のEPに与える生体由来物及び生体活性物質の拡散の影響が大きく、導入効率及び生体活性物質の利用効率が落ちてしまう。
 一方、懸濁液流の厚さを500μm程度にすると、懸濁液流の厚さに対する拡散領域の厚さは小さくなり、拡散の影響は少なくなるが、懸濁液流における電界の均一性が悪くなってしまう。その結果、特許文献2のように、懸濁液流の厚さを50~100μmとした場合よりも、導入効率及び生体活性物質の利用効率が落ちてしまう。
 これに対して、懸濁液流の厚さDmを大幅に厚く1mm以上とすることにより、生体由来物等の拡散による影響を抑制できる。
 すなわち、拡散領域は拡散によって形成されるので、拡散領域の厚さは懸濁液流の厚さDmには影響されない。そのため、懸濁液流の厚さDmを1mm以上とすることにより、拡散領域の厚さに対して拡散の影響がない均一な懸濁液Qの厚さを十分に厚くして、多くの懸濁液Qを処理することが可能になる。
 加えて、懸濁液流の厚さDmを1mm以上とすることにより、電界均一性も良好にできる。すなわち、EPにおいて、生体由来物に印加される電界の強さは、電極対の間方向に存在する生体由来物の数によって変動する。例えば、特許文献2のように、懸濁液流の厚さが50μm程度と十分に狭い場合には、電極対の間方向で生体由来物が重なる確率は低く、各生体由来物に印加される電界は、ほぼ一定で電界均一性は良好である。これに対して、懸濁液流の厚さが500μm程度である場合には、電極対の間方向に存在する生体由来物の数は流れ方向の位置によって異るため、各生体由来物に印加される電界に差が生じてしまい、電界均一性が悪くなる。これに対して、懸濁液流の厚さDmを1mm以上と十分に厚くすることにより、流れ方向の位置における電極対の間方向に存在する生体由来物の数を均一化することができ、その結果、各生体由来物に印加される電界を、ほぼ一定にでき、電界均一性を良好にできる。
 その結果、EP方法によれば、懸濁液流を薄くした従来のフロー式のEPに比して高効率のフロー式のEPを行うことが可能になる。
<電極対の間における懸濁液及びシース液の流れの安定>
 EP装置10及び後述のEP方法では、懸濁液Q及びシース液sの送液後、電極対の間において懸濁液Q及びシース液sの流れが安定した後に、電源部14により懸濁液Qに電界を印加する。以下、電極対の間における懸濁液及びシース液の流れの安定について説明する。
 ここで、図4は本発明の実施形態のエレクトロポレーション装置の第1の例の流路デバイスの電極対の間における懸濁液及びシース液の流れの安定を示す模式的断面図である。図5は本発明の実施形態のエレクトロポレーション装置の第1の例の流路デバイスの電極対の間における懸濁液及びシース液の流れの不安定の一例を示す模式的断面図である。図6は本発明の実施形態のエレクトロポレーション装置の第1の例の流路デバイスの電極対の間における懸濁液及びシース液の流れの不安定の他の例を示す模式的断面図である。
 なお、図4~6において、図1及び図2に示すEP装置10と同一構成物には同一符号を付して、その詳細な説明は省略する。
 電極対34の間における懸濁液Q及びシース液sの流れが安定した状態とは、シース流でない場合に流路の壁面近傍で生体由来物の流れが滞留する滞留層が生じずに、懸濁液流が円滑に流れる状態をいう。好ましくは、懸濁液流の流速分布がほぼ均一な状態をいう。さらに好ましくは、図4に示すように懸濁液流の厚さDmと、2つのシース液流の厚さdsとが予め設定された範囲にあり、懸濁液流とシース液流との界面Bsが波打つことなく平面である。
 EP装置10では、電極対34の間における懸濁液Q及びシース液sの流れが安定した状態で、EPを実施する。これにより、懸濁液Qへの電界の印加を適正に実施でき、その結果、導入効率及び生体活性物質の利用効率が高くなり、懸濁液Qの廃棄も少なくなり、生産性が高いEPを実施できる。
 図5に示すように電極対34の間において、予め設定された範囲に対して2つのシース液流の厚さdsが厚く、懸濁液流の厚さDmが薄い場合、懸濁液流とシース液流との界面Bsが波打つことなく平面であっても、電極対の間における懸濁液Q及びシース液sの流れが安定した状態ではない。電極対34の間における懸濁液Q及びシース液sの流れが図5に示す状態の場合、シース液流が厚く、懸濁液Qへの電界の印加を適正に実施できず、その結果、導入効率及び生体活性物質の利用効率が低くなり、生産性が低い。
 また、図6に示すように電極対34の間において、懸濁液流とシース液流との界面Bsが、主流路30の延在方向Dに沿って第1電極35と第2電極36との対向方向に、例えば、波打つように変動する場合、電極対34の間における懸濁液Q及びシース液sの流れが安定した状態ではない。上述の界面Bsの波打つような変動は、懸濁液Q又はシース液sの送液の際の圧力変動、いわゆる脈動により生じる。図6に示すように懸濁液流とシース液流との界面Bsが波打つように変動する場合、電極対34の間においてシース液流の厚みが不均一であり、懸濁液Qへの電界の印加を適正に実施できず、その結果、導入効率及び生体活性物質の利用効率が低くなり、生産性が低い。
 こられのことから図5及び図6に示すような電極対34の間における懸濁液Q及びシース液sの流れが不安定の状態を避けて、EPを実施する必要がある。
 また、図4、図5及び図6では、電極対34の間において、シース液の厚さdsと、懸濁液Qの厚さDmとが異なっていることから、電極対34の間の電気抵抗が異なる。電極対34の間の電気抵抗は、シース液の厚さdsと、懸濁液Qの厚さDmとに応じた値を示し、シース液流/懸濁液流/シース液流の3層の積層流の厚み方向の電気抵抗を反映する。このことを利用して、電極対34の間の電気抵抗を、後述の測定部19(図1、2等参照)で測定して、電極対34の間における懸濁液Q及びシース液sの流れの電気抵抗を得て、この流れの電気抵抗に基づいて、判定部18が流れの安定を判定し、判定信号を制御部16に出力する。
 例えば、電気抵抗として、インピーダンスを測定して、このインピーダンスに基づいて、電極対34の間における懸濁液Q及びシース液sの流れが安定しているか否かを判定できる。
 このため、例えば、図4に示すように懸濁液Q及びシース液sの流れが安定した状態における電極対34の間の電気抵抗又はインピーダンスを予め測定し、流れが安定であることを示す電極対34の間の電気抵抗又はインピーダンスの範囲を予め設定しておき、判定部18に記憶する。電極対34の間の電気抵抗又はインピーダンスの測定値が予め設定された範囲内にある場合、判定部18は、流れが安定していると判定し、判定信号を制御部16に出力する。
 図7は本発明の実施形態のエレクトロポレーション装置の第1の例の流路デバイスの電極対の間における懸濁液及びシース液の流れの安定を説明するグラフである。
 図7のグラフでは縦軸を流量とし、横軸を時間としている。図7において、符号70の線は、2つのシース液のうち、一方のシース液sの流量変化を示す。符号71の線は、懸濁液Qの流量変化を示す。符号72の線は、2つのシース液のうち、他方のシース液sの流量変化を示す。
 例えば、図7に示すように、懸濁液Q及びシース液sの送液を開始後、所定の時間経過すると、懸濁液Qの流量及びシース液sの流量が、それぞれ設定された流量となり、かつ流量が安定する。この場合、電極対の間における懸濁液Q及びシース液sの流れが安定した状態になる。経過時間による流量の安定を利用して、電極対の間における懸濁液Q及びシース液sの流れが安定した状態を時間制御により得ることができる。
 図7では、懸濁液Q及びシース液sの送液の開始時刻をtとし、電極対の間における懸濁液Q及びシース液sの流れが安定した状態になった時刻をtsとするとき、開始時刻tから時刻tsまでの時間により、電極対の間における懸濁液Q及びシース液sの流れの安定を制御する。
 本発明では高濃度の懸濁液Qを使用するため、懸濁液Q及びシース液sの流れを安定させるためには、開始時刻tから時刻tsまでの時間は、5秒以上が好ましく、10秒以上がより好ましく、15秒以上が更に好ましい。
 懸濁液Q及びシース液sの流れが安定した状態になるまでの時間が長いと、すなわち、開始時刻tから時刻tsまでの時間が長いと、懸濁液Q及びシース液sの廃棄が増える。このため、開始時刻tから時刻tsまでの時間の上限としては、60秒である。
 また、開始時刻tから時刻tsまでの時間は、短すぎると懸濁液Q及びシース液sの流れが安定した状態にならず、長すぎると懸濁液Q及びシース液sの廃棄が増える。そのため、開始時刻tから時刻tsまでの時間は5~60秒が好ましく、10~45秒がより好ましく、15~30秒が更に好ましい。
 図7に示すように、懸濁液Q及びシース液sの送液が安定した後、すなわち、時刻tsの後、時刻tから、電極対による懸濁液Qへの電界の印加を開始し、フロー式のEPを実施する。時刻tの後、時刻tから、容器27(図1参照)に送液する。
 なお、開始時刻tから時刻tまで、懸濁液Q及びシース液sを廃棄槽26(図1参照)に送液する。廃棄槽26(図1参照)と容器27(図1参照)の送液の切り替えは、バルブ29により実施される。
 時刻tsから、予め設定された時間を経過した時刻tで、電極対による懸濁液Qへの電界の印加を停止する。また、懸濁液Q及びシース液sの送液も停止する。
 図7では時刻tsから時刻tまでが、電極対の間における懸濁液Q及びシース液sの流れが安定した状態に該当する。
 EP装置10では、第1チューブ13において懸濁液Qの先頭の待機位置が予め定められており、第2チューブ21a、21bおいてシース液sの先頭の待機位置が予め定められている。制御部16は、ポンプ12a、12b、20a、20bにより、懸濁液Q及びシース液sの全ての先頭が、待機位置に到達した後に、懸濁液Q及びシース液sの送液を開始させる。
 センサ38を、第1チューブ13における懸濁液Qの先頭の待機位置に配置することにより、懸濁液Qの先頭の待機位置を検知できる。第1チューブ13において懸濁液Qの先頭が、センサ38により検知された場合、センサ38から制御部16に検知信号が送信され、制御部16は検知信号を受信する。
 センサ39a、39bを、第2チューブ21a、21bおけるシース液sの先頭の待機位置に配置することにより、シース液sの先頭の待機位置を検知できる。第2チューブ21a、21bにおいてシース液sの先頭がセンサ39a、39bにより検知された場合、センサ39a、39bから制御部16に検知信号が送信され、制御部16は検知信号を受信する。このように、センサ38からの検知信号と、センサ39a、39bからの検知信号とにより、制御部16では、ポンプ12a、12bによる懸濁液Qの送液と、ポンプ20a、20bによるシース液sの送液とを制御できる。
 例えば、制御部16では、懸濁液Q及びシース液sの全ての先頭が、待機位置に到達した場合、ポンプ12a、12b、20a、20bを停止して懸濁液Q及びシース液sの送液を停止する。この状態で、次に、ポンプ12a、12b、20a、20bによる懸濁液Q及びシース液sの送液を開始する。制御部16は、電極対の間において懸濁液Q及びシース液sの流れが安定した後に、電源部14により懸濁液Qに電界を印加させる。
 懸濁液Q及びシース液sを送液するタイミングは、懸濁液Qとシース液sとで同時であってもよく、懸濁液Qよりもシース液sの方が先であってもよい。
 懸濁液Q及びシース液sを送液するタイミングが同時とは、懸濁液Qを送液するポンプ12a、12bと、シース液sを送液するポンプ20a、20bとの駆動タイミングが同じであることである。すなわち、図7に示す開始時刻tでポンプ12a、12bと、ポンプ20a、20bとを駆動して、懸濁液Qとシース液sとを送液することである。
 懸濁液Q及びシース液sを送液するタイミングが、懸濁液Qよりもシース液sの方が先とは、シース液sを送液するポンプ20a、20bの駆動タイミングが、懸濁液Qを送液するポンプ12a、12bの駆動タイミングよりも先であることである。すなわち、図7に示す開始時刻tでポンプ20a、20bを駆動してシース液sを送液した後、ポンプ12を駆動して懸濁液Qを送液することである。
 なお、送液するタイミングが懸濁液Qよりもシース液sの方が先とは、第1電極35の電極面35aと、第2電極36の電極面36aとの間の流路をシース液sが満たすことである。
 懸濁液Qよりもシース液sを先に送液した場合、シース液sにより第1電極35の電極面35aと、第2電極36の電極面36aとが覆われるため、第1電極35の電極面35aと、第2電極36の電極面36aとに懸濁液Qが接触することが抑制される。このため、第1電極35の電極面35aと第2電極36の電極面36aとが汚れることが防止される。
 判定部18は、電極対の間において懸濁液Q及びシース液sの流れの安定を判定するものである。
 判定部18は、例えば、電極対34の間における懸濁液Q及びシース液sの流れの電気抵抗を測定して、電気抵抗に基づいて、流れの安定を判定する。この場合、判定部18に、測定部19(図1、2等参照)を設ける。上述の流れの安定の判定に電気抵抗を用いる場合、測定部19は、電極対34の第1電極35と、配線19aにより電気的に接続され、第2電極36と、配線19bにより電気的に接続される。測定部19は、電極対34の間の電気抵抗を測定して、電極対34の間における懸濁液Q及びシース液sの流れの電気抵抗を測定する。測定部19は、上述の電極対34の間の電気抵抗を測定できれば、その構成は、特に限定されるものではない。
 また、判定部18に測定部19がある場合、電源部14及び判定部18の動作時に、互いに影響を与えないように、電源部14と判定部18とを電気的に絶縁状態にできる構成にする。例えば、電源部14及び判定部18に、それぞれ開閉器等の遮断器(図示せず)が設けられている。遮断器により、電源部14と、電極対34とを導通状態又は電気的に絶縁状態にできる。また、遮断器により、判定部18と、電極対34とを導通状態又は電気的に絶縁状態にできる。
 電源部14と電極対34とが導通状態のときには、遮断器により、判定部18と電極対34とを電気的に絶縁状態にして、電源部14と判定部18とを電気的に絶縁状態にする。一方、判定部18と電極対34とが導通状態のときには、遮断器により、電源部14と電極対34とを電気的に絶縁状態にして、電源部14と判定部18とを電気的に絶縁状態にする。
 これにより、電源部14の動作時に判定部18に電圧が印加されない。また、判定部18の測定部19による電気抵抗の測定時に、電源部14を含めた電気抵抗を測定することがない。
 なお、上述の流れの安定の判定に電気抵抗を用いない場合、判定部18に測定部19を設ける必要がない。
 判定部18を用いて、電極対34の間における懸濁液Q及びシース液sの流れの電気抵抗として、例えば、インピーダンスを測定する。図4に示す状態の電気抵抗が予め測定されており、流れが安定であることを示す電気抵抗の範囲が予め設定されており、判定部18に記憶されている。判定部18は、測定部19を用いて測定された、電極対34の間における電気抵抗が予め設定された範囲内にある場合、流れが安定していると判定する。この場合、判定部18は判定信号を制御部16に出力する。
 また、判定部18は、第1チューブ13の懸濁液Qの流量及び第2チューブ21a、21bのシース液sの流量の安定性を検出して、懸濁液Q及びシース液sの流れの安定を判定することもできる。この場合、第1チューブ13のポンプ12aの下流側、かつ流入口31迄の領域に流量計(図示せず)を設ける。また、第2チューブ21aのポンプ20aの下流側、かつ第1流入口31a迄の領域に流量計(図示せず)を設ける。また、第2チューブ21bのポンプ20bの下流側、かつ第2流入口31b迄の領域に流量計(図示せず)を設ける。
 各流量計による流量のデータを判定部18に出力させる。判定部18が、各流量計により計測された流量の変動より流れの安定を判定する。判定部18では、例えば、設定流量と、計測された流量とを比較し、例えば、図7に示すように、設定流量に達した後、設定流量が予め設定した時間維持された場合に、懸濁液Q及びシース液sの流れが安定していると判定する。このようにして、第1チューブ13の懸濁液Qの流量及び第2チューブ21a、21bのシース液sの流量の安定性を検出して、懸濁液Q及びシース液sの流れの安定を判定できる。なお、上述の予め設定した時間は、判定部18に記憶されている。
 流量の安定性の判定において、懸濁液Qの流量を設定流量と比較するが、シース液流は2つある。例えば、2つのシース液流の流量を、それぞれ設定流量と比較して、2つとも設定流量に達した後、設定流量が予め設定した時間維持された場合に、懸濁液Q及びシース液sの流れが安定していると判定する。
 流量計は、懸濁液Q及びシース液sの流量を測定できれば、特に限定されるものではない。流量計には、例えば、超音波式、コリオリ式、又は熱MEMS(Micro Electro Mechanical Systems)式のものが用いられる。
 さらには、判定部18は、上述の時間制御により、電極対の間において懸濁液Q及びシース液sの流れの安定を判定してもよい。この場合、懸濁液Qを送液するポンプ12aと、シース液sを送液するポンプ20a、20bとの駆動開始時刻(開始時刻t)から時間を、例えば、判定部18で計測する。懸濁液Q及びシース液sの送液を開始後、予め設定した時間経過した場合、判定部18は、流れが安定していると判定し、判定信号を制御部16に出力する。制御部16は、受信した判定信号に基づいて、電源部14により懸濁液Qに電界を印加する等の各部を制御する。
(エレクトロポレーション方法の第1の例)
 エレクトロポレーション方法(EP方法)の第1の例は、電極対34の間、すなわち、上述の空間37に生体由来物と生体活性物質とを含有する懸濁液Qと、シース液sとを、シース液sで懸濁液Qを挟むようにして主流路30に連続的に送液して、電極対34によって懸濁液Qに電界を印加することで、生体由来物に生体活性物質を導入する方法である。EP方法の第1の例には、例えば、図1及び図2に示すEP装置10が用いられる。
 EP方法の第1の例では、EP装置10のポンプ12aにより培養装置24内の培養液を吸引し、ポンプ12bによりタンク23内の生体活性物質を吸引して、培養液の培地をEP用バッファーに交換することなく、混合器25で培養装置24の培養液と、タンク23の生体活性物質とが混合された懸濁液Qとなる。ポンプ12aにより懸濁液Qを第1チューブ13及び流入口31を通して主流路30に供給する。また、ポンプ20aによりタンク22内のシース液sを、第2チューブ21a、第1流入口31a、第1導入部56に設けられた貫通孔56a及び第1基板52に設けられた貫通孔52aを通して主流路30に供給する。また、ポンプ20bによりタンク22内のシース液sを、第2チューブ21b、第2流入口31b、第2導入部58に設けられた貫通孔58a及び第2基板54に設けられた貫通孔54aを通して主流路30に供給する。これにより、電極対34の間でシース液流/懸濁液流/シース液流の3層の積層流が形成され、電極対34の間を鉛直方向の下方から上方に向かう方向に懸濁液Q及びシース液sを連続的に送液する。このとき、電源部14により電極対34に、例えば、パルス電圧を印加してEPを実施する。
 EP方法の第1の例では、懸濁液Q及びシース液sの送液方向Ddが鉛直方向zの下方から上方に向かう方向である。電源部14を用いて懸濁液Qに電界を印加してEPを実施し、対向する第1電極35と第2電極36との間に気泡が発生した場合、発生した気泡は、懸濁液Qに比して比重が小さいため、鉛直方向zに懸濁液Qよりも移動しやすい。このため、電極対34の間に発生した気泡は、懸濁液Q及びシース液sの送液により電極対34の間に滞留することなく、鉛直方向zに移動することで電極対34の間から除去され、気泡による懸濁液Qへの電界の印加の阻害を抑制できる。これにより、EP方法の第1の例では、懸濁液Qへの電界の印加を適正に実施でき、その結果、導入効率及び生体活性物質の利用効率を適正なものにできる。
 また、懸濁液Qの組成が、懸濁液Qに電界を印加した際に気泡が発生しやすいものである場合にも、上述のように電極対34の間で気泡が発生した場合、発生した気泡は、懸濁液Qに比して比重が小さいため、鉛直方向zに懸濁液Qよりも移動しやすく、懸濁液Q及びシース液sの送液により電極対34の間に滞留することなく、鉛直方向zに移動することで電極対34の間から除去され、気泡による懸濁液Qへの電界の印加の阻害を抑制できる。これにより、EP方法の第1の例では、懸濁液Qへの電界の印加を適正に実施でき、その結果、導入効率及び生体活性物質の利用効率を適正なものにできる。
 EP方法では、例えば、EPを実施する前に、細胞培養により、生体由来物と培地を含む培養液を得る培養工程と、培養液と生体活性物質とを混合して懸濁液とする混合工程とを有してもよい。通常、EPを実施する前には、培養液から生体由来物(例えば、細胞)を取り出して、EP用バッファーに懸濁させて懸濁液を得る。この培地をEP用バッファーに交換することを、単に培地交換又はバッファー交換という。
 EP方法の第1の例では、培養工程で得られた、培養液の培地をEP用バッファーに交換せずに、すなわち、EP用バッファーを添加せずに、生体活性物質と混合して、懸濁液を電極対34の間に連続的に送液する。
 EP方法の第1の例では、培養液の培地をEP用バッファーに交換しないことから、通常EPされる懸濁液よりも培地を多く含み、電気伝導度が高い。そのため、電界を印加すると気泡が大量に発生する。このような懸濁液に対しても、懸濁液は電極対の間を鉛直方向の下方から上方に向かって移動しながらEPされるため、電極対の間に発生した気泡は、鉛直方向に移動することで電極対の間から除去され、気泡による懸濁液への電界の印加の阻害を抑制できる。
 培養工程は、上述のように培養装置24で実施される。混合工程は、上述のようにポンプ12a、12bを用いて培養装置24の培養液と、タンク23の生体活性物質とを吸引することにより混合器25で実施される。
 以下、EP方法の第1の例について、より具体的に説明する。
 図8は本発明の実施形態のエレクトロポレーション方法の第1の例を示すフローチャートである。
 上述のように第1チューブ13において懸濁液Qの先頭の待機位置が予め定められ、第2チューブ21a、21bにおいて、それぞれシース液sの先頭の待機位置が予め定められている。
 まず、懸濁液Qの先頭の待機位置に配置されたセンサ38(図1及び図2参照)により、第1チューブ13における懸濁液Qの先頭の待機位置の検知を開始する。シース液sの先頭の待機位置に配置されたセンサ39a、39b(図1及び図2参照)により、第2チューブ21a、21bにおけるシース液sの先頭の待機位置の検知を開始する。
 ポンプ12a、12bにより、懸濁液Qを第1チューブ13に送液する。また、ポンプ20aによりタンク22内のシース液sを第2チューブ21aに送液する。ポンプ20bによりタンク22内のシース液sを第2チューブ21bに送液する(ステップS10)。
 このとき、上述のように、懸濁液Q及びシース液sの先頭の待機位置の検知はセンサ38(図1及び図2参照)、及びセンサ39a、39b(図1及び図2参照)により開始されている。
 次に、懸濁液Qの先頭及びシース液sの先頭が、待機位置に到達したか否かを判定する(ステップS12)。ステップS12では、懸濁液Qの先頭の待機位置に配置されたセンサ38(図1及び図2参照)、及びシース液sの先頭の待機位置に配置されたセンサ39a、39b(図1及び図2参照)から検知信号を制御部16で受信した場合、制御部16が懸濁液Qの先頭及びシース液sの先頭が、待機位置に到達したと判定する。
 ステップS12において、懸濁液Q及びシース液sの全ての先頭が、待機位置に到達したと判定された場合、一旦、ポンプ12a、12b、20a、20bの駆動を停止し、懸濁液Q及びシース液sの送液を停止する。その後、懸濁液Q及びシース液sの全ての先頭が、待機位置にある状態で、ポンプ12a、12b、20a、20bの駆動を開始して、懸濁液Q及びシース液sの送液を開始する(ステップS14)。
 一方、制御部16が上述の検知信号を受信していない場合、制御部16が懸濁液Qの先頭及びシース液sの先頭は待機位置に到達していないと判定する。この場合、懸濁液Qの先頭及びシース液sの送液を継続する(ステップS26)。再度、懸濁液Qの先頭及びシース液sの先頭が、待機位置に到達したか否かを判定する(ステップS12)。制御部16により懸濁液Q及びシース液sの全ての先頭が待機位置に到達したと判定されるまで、ステップS12及びステップS26を繰り返す。
 ステップS14で懸濁液Q及びシース液sの送液を再開した後、電極対34の間において懸濁液及びシース液の流れの安定を判定する(ステップS16)。ステップS16が判定工程である。
 ステップS16(判定工程)では、上述のように、例えば、判定部18の測定部19により電極対34の間における懸濁液Q及びシース液sの流れの電気抵抗を測定して、電極対34の間の電気抵抗に基づいて、流れの安定を判定する。例えば、上述のように流れが安定であることを示す電極対34の間の電気抵抗の範囲が予め設定されており、測定された電極対34の間の電気抵抗が予め設定された範囲内にある場合、判定部18により、流れが安定していると判定され、判定部18は判定信号を制御部16に出力する。
 また、ステップS16(判定工程)では、流量の安定性を検出して、上述の流れの安定を判定できる。この場合、上述のように判定部18では、例えば、設定流量と、計測された流量とを比較し、例えば、図7に示すように、設定流量に達した後、設定流量が予め設定した時間維持された場合に、流れが安定していると判定し、判定信号を制御部16に出力する。判定工程は後述の印加工程の前に実施される。
 また、ステップS16では、上述の時間制御により、電極対34の間において懸濁液Q及びシース液sの流れの安定を判定することもできる。
 ステップS16では、上述のいずれかの方法により、上述の流れの安定を判定すればよい。
 ステップS16において、流れが安定していると判定された場合、制御部16が電源部14により懸濁液Qに電界を印加させる(ステップS18)。ステップS18により、EPが実施される。ステップS18が印加工程である。連続的なEPであるため、電界は連続的に継続して、所定の期間印加される。
 なお、ステップS16で流れの安定の判定に電気抵抗を用いた場合、ステップS16では、上述のように遮断器により、判定部18と電極対34とを導通状態にし、電源部14と電極対34とを電気的に絶縁状態にして電源部14と判定部18とを電気的に絶縁状態にした後に、測定部19により、電極対34の間における電気抵抗を測定する。
 ステップS16で流れの安定の判定に電気抵抗を用いた場合、ステップS18では、上述のように遮断器により、電源部14と電極対34とを導通状態にし、判定部18と電極対34とを電気的に絶縁状態にして電源部14と判定部18とを電気的に絶縁状態にした後に、電源部14により懸濁液Qに電界を印加する。
 一方、ステップS16において、流れが安定していると判定されない場合、懸濁液Q及びシース液sの送液を継続する(ステップS28)。再度、懸濁液Q及びシース液sの流れが安定しているか否かを判定する(ステップS16)。懸濁液Q及びシース液sの流れが安定している判定されるまで、ステップS16及びステップS28を繰り返す。
 次に、ステップS18(印加工程)の電界の印加から所定の時間経過後、排出先を変更する(ステップS20)。ステップS20において、ステップS18の電界の印加からの経過時間を、例えば、制御部16で計測する。
 ここで、懸濁液Q及びシース液sの送液開始(ステップS10)から電界印加(ステップS18)迄は、排出口32から排出された懸濁液Q及びシース液sを廃棄槽26(図1参照)に送液している。電界印加(ステップS18)後、予め設定された時間経過後、例えば、図7に示す時刻tから、バルブ29(図1参照)により送液先を切り替えて、排出口32から排出された懸濁液Q及びシース液sの送液先を容器27(図1参照)に変更する。
 ステップS16で流れが安定したと判定された後、予め設定された時間が経過したか否かを判定する(ステップS22)。上述のステップS16の流れが安定したと判定されてからの経過時間は、例えば、制御部16で計測し、予め設定された時間が経過したか否かを制御部16が判定する。
 ステップS22において、ステップS16の流れが安定したと判定されてからの経過時間が、予め設定された時間を経過したと判定された場合、制御部16により、電源部14を制御して懸濁液Qへの電界の印加を停止し、更には、ポンプ12a、12b及びポンプ20a、20bを停止して懸濁液Q及びシース液sの送液を停止する(ステップS24)。
 一方、ステップS22において、ステップS16の流れが安定したと判定されてからの経過時間が、予め設定された時間を経過していないと判定された場合、ステップS16の流れが安定したと判定されてからの経過時間が、予め設定された時間を経過したと判定される迄、懸濁液Q及びシース液sの送液及び電界の印加が継続される。ステップS22において、予め設定された時間を経過したと判定された場合、制御部16により、電源部14を制御して懸濁液Qへの電界の印加を停止し、更には、ポンプ12a、12b及びポンプ20a、20bを停止して懸濁液Q及びシース液sの送液を停止する(ステップS24)。
(エレクトロポレーション装置の第2の例)
 図9は本発明の実施形態のエレクトロポレーション装置(EP装置)の流路デバイスの第2の例を示す模式的断面図である。
 なお、図9において、図1及び図2に示すEP装置10と同一構成物には同一符号を付して、その詳細な説明は省略する。
 図9に示すEP装置10aは、図1及び図2に示すEP装置10に比して、補助電極60を有し、補助電極60が判定部18の測定部19に電気的に接続されている点が異なり、それ以外の構成は、図1及び図2に示すEP装置10と同様の構成である。
 EP装置10aは、補助電極60が電極対34の主流路30の下流側に設けられている。補助電極60は、懸濁液Q及びシース液sの流れの厚み方向の電気抵抗の測定に用いられる。判定部18が補助電極60を用いて懸濁液Q及びシース液sの流れの厚み方向の電気抵抗を測定する。補助電極60を用いて測定する厚み方向の電気抵抗には、インピーダンスが含まれる。
 補助電極60は、対向して配置された第1電極61と、第2電極62とを有する。第1電極61は電極面61aを有し、電極面61aは平面である。第2電極62は電極面62aを有し、電極面62aは平面である。第1電極61の電極面61aと、第2電極62の電極面62aとは対向し、かつ平行に配置されている。補助電極60は、第1電極61と第2電極62とを有する対電極である。
 補助電極60を用いて懸濁液Q及びシース液sの流れの厚み方向の電気抵抗は、懸濁液Q及びシース液sの流れの補助電極60を構成する第1電極61と第2電極62の間の電気抵抗である。
 第1電極61の電極面61aと、第2電極36の電極面36aとは、それぞれ主流路30の内面30cに面して配置されている。第1電極61の電極面61aと、第2電極62の電極面61aとは、それぞれ主流路30の内面30cを構成しており、例えば、EPを実施する際に、主流路30に懸濁液Qとシース液sとが供給されると、シース液sと接する。
 第1電極61は、配線19aにより測定部19と電気的に接続されている。第2電極62は、配線19bにより測定部19と電気的に接続されている。
 EP装置10aでは、判定部18の測定部19と補助電極60を用いて、懸濁液Q及びシース液sの流れの厚み方向の電気抵抗として、例えば、インピーダンスを測定する。補助電極60の場合でも、図4に示すように懸濁液Q及びシース液sの流れが安定した状態の厚み方向の電気抵抗又はインピーダンスが予め測定されており、流れが安定であることを示す厚み方向の電気抵抗又はインピーダンスの範囲を予め設定する。予め設定された厚み方向の電気抵抗又はインピーダンスの範囲は、判定部18に記憶されている。
 判定部18は、補助電極60を用いて測定された厚み方向の電気抵抗又はインピーダンスの測定値が予め設定された範囲内にある場合、流れが安定していると判定する。判定部18の判定結果を示す情報を制御部16に出力する。制御部16は、受信した判定結果に基づいて電源部14により懸濁液Qに電界を印加する等の各部を制御する。
 補助電極60を電極対34の主流路30の下流側に設けることにより、電極対34の下流側にある補助電極60で、懸濁液Q及びシース液sの流れが安定していれば、補助電極60の上流側の電極対34の間でも懸濁液Q及びシース液sの流れが安定しているとしている。
 EP装置10aは、補助電極60を電極対34の主流路30の下流側に設ける構成としたが、これに限定されるものではなく、例えば、補助電極60を電極対34の主流路30の上流側に設けてもよい。より具体的には、補助電極60を、主流路30において合流部59と電極対34との間に相当する領域に設けてもよい。
 補助電極60を電極対34の主流路30の上流側に設けた場合、補助電極60で、懸濁液Q及びシース液sの流れが安定していれば、補助電極60の下流側の電極対34の間でも懸濁液Q及びシース液sの流れが安定しているとしている。
 EP装置10aは、上述のEP装置10と同様の効果を得ることができる。EP装置10aでも、懸濁液Qへの電界の印加を適正に実施でき、その結果、導入効率及び生体活性物質の利用効率が高くなり、懸濁液Qの廃棄も少なくなり、生産性が高いEPを実施できる。
(エレクトロポレーション方法の第2の例)
 エレクトロポレーション方法(EP方法)の第2の例には、例えば、図9に示すEP装置10aが用いられる。
 EP方法の第2の例は、EP方法の第1の例に比して、電極対34の間において懸濁液及びシース液の流れの安定を判定する判定工程(ステップS16)が、電極対34の間における電気抵抗ではなく、補助電極60を用いて測定した、懸濁液Q及びシース液sの流れの厚み方向の電気抵抗である点が異なる。それ以外の工程は、EP方法の第1の例と同様の工程であるため、その詳細な説明は省略する。
 ステップS16において、例えば、補助電極60を用いて、判定部18の測定部19により懸濁液Q及びシース液sの流れの厚み方向の電気抵抗を測定して、補助電極60を用いた厚み方向の電気抵抗に基づいて、流れの安定を判定する。例えば、上述のように流れが安定であることを示す懸濁液Q及びシース液sの流れの厚み方向の電気抵抗の範囲が予め設定されており、測定された、懸濁液Q及びシース液sの流れの厚み方向の電気抵抗が予め設定された範囲内にある場合、判定部18は、流れが安定していると判定する。
 なお、例えば、補助電極60を電極対34の主流路30の上流側に設けた場合、ステップS16(判定工程)では、電極対34の流路における上流側において、補助電極60により懸濁液Q及びシース液sの流れの厚み方向の電気抵抗を測定して、判定部18が厚み方向の電気抵抗に基づいて、懸濁液Q及びシース液sの流れの安定を判定する。判定部18は、判定結果を示す情報を制御部16に出力する。
 EP方法の第2の例は、EP方法の第1の例と同様の効果を得ることができる。
 上述のEP装置及びEP方法は、いずれも目的のタンパク質又はウイルス等の有用物質の製造に用いることができる。すなわち、有用物質の製造方法は、上述のEP方法を含む。
 以下、EP装置の各部について説明する。
〔電極対の間の距離D〕
 電極対34の間の距離D(図2、9参照)は、第1電極35の電極面35aと第2電極36の電極面36aとの対向方向における距離であり、1~10mmであることが好ましい。電極対の間の距離Dの下限値は、2mmがより好ましく、3mmが更に好ましい。電極対の間の距離Dの上限値は、6mmがより好ましく、5mmが更に好ましい。
 電極対の間の距離Dが大きくなり過ぎると、必要以上の電界が発生し、発熱、電極端部への電界集中が生じ、沸騰又は放電が発生して、生存率又は導入効率が低下する場合がある。発熱を防ぐ点からは、電極対の間の距離Dは、1~6mmであることが好ましい。
 また、電極対の間の距離Dは1~3mmであると、発熱及び放電の発生が抑制され、発熱又は放電による細胞等の生体由来物の生存率又は導入効率の低下も抑制されることから、更に好ましい。
 なお、電極対の間の距離Dは、流路基板55の厚さに相当する。電極対の間の距離Dを、電極間ギャップ、又はギャップともいう。
 上述電極対の間の距離Dは、該当する部分の長さを、ノギス又はマイクロメーターを用いて測定できる。
<電極対>
 電極対34を構成する第1電極35及び第2電極36は、例えば、金属材料又は炭素材料で構成される。具体的には、国際公開第2023/157673号に記載の電極と同様の構成が採用できる。
(厚さ)
 第1電極35の厚さ及び第2電極36の厚さは特に制限されないが、0.5~10mmであることが好ましく、1~5mmであることがより好ましい。
 上述の第1電極35の厚さ及び第2電極36の厚さは、いずれも該当する部分の厚さを、ノギス又はマイクロメーターを用いて測定できる。
<補助電極>
 補助電極60を構成する第1電極61及び第2電極62は、電極対34を構成する第1電極35及び第2電極36と同様の構成であり、例えば、上述の金属材料で構成される。
 第1電極61の厚さ及び第2電極62の厚さについても、特に制限されないが、第1電極35及び第2電極36と同様に0.5~10mmであることが好ましく、1~5mmであることがより好ましい。
 上述の第1電極61の厚さ及び第2電極62の厚さは、いずれも該当する部分の厚さを、ノギス又はマイクロメーターを用いて測定できる。
 電極対の間の距離Dについても、特に制限されないが、第1電極35及び第2電極36と同様であり、1~10mmであることが好ましく、2~5mmであることがより好ましく、2~3mmが更に好ましい。
 補助電極60は、主流路30の延在方向Dにおける長さが電極対34よりも短いことが好ましく、2~10mmであることがより好ましい。
<流路デバイスの構成>
 図2の流路デバイス11の第1導入部56と第1基板52と流路基板55と第2基板54と第2導入部58とを構成する材料は、特に限定されるものではない。
 これらを構成する材料は、例えば、懸濁液、又は懸濁液及びシース液に対して十分な耐性を有し、かつ、必要な剛性を確保できるものであれば、各種の材料が利用可能である。
<懸濁液>
 エレクトロポレーション(EP)の対象となる懸濁液は、生体由来物と生体活性物質とを含有する。また、懸濁液は、培地がEP用バッファーに交換されたものではなく、EP用バッファーが添加されたものではない。
 懸濁液は、電気伝導度が2mS/cm以上であることが好ましく、3mS/cm以上であることがより好ましく、4mS/cm以上であることが更に好ましい。通常、懸濁液は、EP前に、培養液の培地をEP用バッファーに交換されて、懸濁液中の培地はEP用バッファーに置き換わり、懸濁液の電気伝導度は小さくなり、電界印加に伴う気泡の発生は少ない。上記の電気伝導度の懸濁液は、通常はEPに使用できないが本発明の方法では使用できる。
 電気伝導度(mS/cm)は、電気伝導率計を用い、温度25℃の懸濁液を測定した値である。電気伝導度(mS/cm)は、導電率と同義であり、導電率は電気伝導度と単位も同じである。
 電気伝導度を調整する方法としては、例えば、懸濁液に含まれる生体由来物の体積分率を調整する方法、及び培地の種類を変える方法が挙げられる。
 通常、EP対象の懸濁液の培地(生体由来物以外の液体成分)は、培養液の培地をEP用バッファーに交換するため、ポロキサマー含有量が0.1g/L未満である。また塩化ナトリウムの濃度は0.01g/L未満である。
 他方、本発明では、培養液の培地をEP用バッファーに交換しないため、培地のポロキサマー含有量が0.1g/L以上である。ポロキサマー含有量は1g/L以上でもよく、3g/L以上でもよく、5g/L以上でもよい。本発明では、ポロキサマー含有量が多くても、気泡による懸濁液Qへの電界の印加の阻害を抑制できる。
 EP対象の懸濁液の培地は、塩化ナトリウムを含有し、塩化ナトリウムの含有量が0.001g/L以上であり、より好ましくは0.01g/L以上であり、0.1g/L以上であり、1g/L以上である。
 上記含有量は、懸濁液を300Gで15分間遠心分離し、遠心上清液を採取し、高速液体クロマトグラフィーにて測定することで求めることができる。
 懸濁液の流量は、1mL/分以上であることが好ましく、2mL/分以上であることがより好ましく、3mL/分以上であることが更に好ましい。懸濁液の流量の上限は1000mL/分であることが好ましい。
 懸濁液の流量を1mL/分以上とすることにより、EPの処理量を多くできる点で好ましい。
 上記流量は、送液流路中に各種流量センサ(超音波式、コリオリ式、熱MEMS式)を設置して測定することができる。あるいは、上記流量は、回収容器の重量変化と送液時間から算出してもよい。
(生体由来物)
 生体由来物は特に制限されないが、具体例としては、細胞、細胞小器官、細胞内顆粒及び小胞等、並びに菌等が挙げられる。なかでも、本発明の効果等がより優れる理由から、細胞であることが好ましく、動物細胞であることがより好ましく、哺乳類細胞であることが更に好ましく、ヒト又はチャイニーズハムスター由来細胞であることが最も好ましい。細胞の具体例としては、ヒトT細胞、HEK(Human Embryonic Kidny)293、A549、SF9、EB66、Daudi、Hela、Vero、MDCK、BHK(Baby Hamster Kidney)、CHO(Chinese Hamster Ovary)、NS0、SP2/0、ハイブリドーマ等が挙げられる。医薬品製造の点では、HEK293又はCHOでの遺伝子導入が最も使われる。
(生体活性物質)
 生体活性物質は、核酸(例えば、DNA、RNA)及びタンパク質等の、生体由来物に導入されることにより、生体由来物に何らかの作用をもたらす物質である。核酸としては、例えば、プラスミド、線形DNA、mRNA(messenger RNA)があり、プラスミドが特に好ましい。
 懸濁液における生体活性物質の濃度は、1~1000μg/mLが好ましく、2.5~500μg/mLがより好ましく、10~200μg/mLが更に好ましい。
 懸濁液中の培地に対する、懸濁液中の生体活性物質の濃度は、10~500μg/mLが好ましい。
 懸濁液における生体由来物あたりの生体活性物質濃度は、20pg/個以下が好ましく、5pg/個以下がより好ましく、1pg/個以下が更に好ましく、0.5pg/個以下が特に好ましい。懸濁液における生体由来物あたりの生体活性物質濃度が低いほど、生体由来物あたりの生体活性物質の使用量を低減できるため、コストを低減することができる。
 懸濁液Qにおける生体由来物の濃度には制限はないが、懸濁液Qは生体由来物の体積分率が20%以上であることが好ましい。
 EPにおいては、懸濁液Qにおける生体由来物の濃度を高くすることで、同量の低濃度の懸濁液を導入した場合に比して生体由来物の処理量を多くできる。例えば、濃度が2倍であれば、2倍の処理を行うことが可能になり、EPのスループット及び生体由来物の利用効率を向上することができる。
 加えて、EPでは、上述のように電気泳動によって生体由来物の中に生体活性物質が入る。そのため生体由来物の濃度を高くしても、それに応じて生体活性物質の量を増やす必要が無いので、生体活性物質の利用効率も向上できる。
 懸濁液Qは生体由来物の体積分率が20%以上であることにより、上述した効果を好適に得て、効率の良い処理を行うことができる。
 また、懸濁液Qの濃度が高過ぎると、細胞懸濁液の粘度が急激に増加し、流路が詰まってしまう可能性が生じる。しかしながら、懸濁液Qは生体由来物の体積分率を70%以下とすることにより、懸濁液Qの濃度が高過ぎることに起因する流路の詰まりを好適に防止できる。
 懸濁液Qにおける生体由来物の体積分率は、30%以上がより好ましく、40%以上がより好ましく、50%以上が更に好ましい。懸濁液Qの生体由来物の体積分率の上限は、65%がより好ましく、60%が更に好ましい。
 上記懸濁液Qにおける生体由来物の体積分率は、生体由来物の平均直径及び濃度(単位体積あたりの個数)を画像解析(例えば、Vi-CELL XR(ベックマン・コールター)等)により測定し、体積分率(=4÷3×(平均直径÷2)3×π×濃度×100(%))を算出することができる。なお、体積分率は、生体由来物が球であるとして算出している。より具体的には、例えば、懸濁液Qにおける生体由来物の平均直径が20μm、濃度が40×10個/mL(ミリリットル)である場合、懸濁液Qにおける生体由来物の体積分率は17%と算出できる。
 培地としては、通常の動物細胞の培養で使用されている液状培地を用いることができる。培地としては、PBS(リン酸緩衝食塩水)をベースとしたHEK293等の細胞の培養に最適化した培地が各社から販売されている。培地は、アミノ酸、塩、糖(グルコース等)類、ビタミン、ホルモン、増殖因子、脂質、微量元素等を含むが、特に限定されない。培地のpH(水素イオン指数)は6~8であり、好ましくは6.8~7.6であり、より好ましくは7.2~7.6である。
 培地には、更に消泡剤を添加してもよい。消泡剤としては、シリコーン系が好ましく、ジメチコンが特に好ましい。消泡剤としては、ポリジメチルシロキサンを含むものが好ましく、より好ましくはポリジメチルシロキサンに微粉末シリカを含むもの(シメチコン)である。培養液量に対するシメチコンの添加速度は特に限定されないが、好ましくは5mg/hr/L以下であり、より好ましくは2mg/hr/L以下である。
 培地には、ポリオキシプロピレンとポリオキシエチレンのブロック共重合体を添加してもよい。ポリオキシプロピレンとポリオキシエチレンの共重合体としては、ポロキサマーが好ましく、より好ましくは、ポロキサマー-188である。
 本発明においては、電極対の間の一部で懸濁液流の厚さDmが1~10mmとなっていればよく、電極対の間かつ電極上流端部で懸濁液流の厚さDmが1~10mmとなっていればよい。
 上述電極対の間における懸濁液流の少なくとも一部の厚さは、細胞濃度が80%以上の懸濁液流を特定し、特定した懸濁液流の厚さを測定する。
<シース液>
 シース液流に使用されるシース液は、懸濁液以外の液体であれば、各種の液体が利用可能である。中でも、シース液としては、懸濁液に用いられる培地が好ましく例示される。この培地は、細胞等の生体由来物の増殖に用いた培地であることが好ましい。
 上述のようにシース液流は2つある。2つのシース液流は、それぞれのシース液の流量が、0.1mL/分以上であることが好ましい。シース液の流量の上限は、懸濁液の流量以下であることが好ましい。
 シース液の流量を0.1mL/分以上とすることにより、懸濁液が電極に接触することを好適に防止できる点で好ましい。また、シース液の流量を、懸濁液の流量以下とすることにより、シース液の量を抑制できる、懸濁液の希釈を抑制できる等の点で好ましい。
 ポンプは、懸濁液を1mL/分以上の流量で送液することが好ましく、2mL/分以上の流量で送液することがより好ましく、3mL/分以上の流量で送液することが更に好ましい。ポンプによる懸濁液の流量の上限は10mL/分であることが好ましい。
 ポンプは、2つのシース液流のうち、それぞれのシース液流を形成するシース液を0.1mL/分以上の流量で送液することが好ましい。2つのシース液流のうち、それぞれのシース液流を形成するシース液のポンプによる流量の上限は、懸濁液の流量以下であることが好ましい。
 ここで、EPでは、電界が印加されると、その時点で、生体由来物の膜(又は殻)に孔が開き、電気泳動によって生体由来物内に生体活性物質が入る。
 生体由来物の膜に形成された孔は、継時と共に徐々に閉じるが、孔が閉じる前に、例えば、懸濁液の希釈等の操作により生体由来物の外部の液成分(浸透圧、イオン濃度等)が急変すると、孔を介したイオン及び媒質の移動等により、生体由来物内における成分比率の急変、並びに、変形及び膨張等のダメージを受けてしまう。
 その結果、例えば、細胞への物質導入の場合には生体由来物の生存率が低下して、EPにおける導入効率及び生体活性物質の利用効率が低くなってしまう可能性がある。
 シース液流の厚さdsが懸濁液流の厚さDmよりも大きく、懸濁液Qの希釈率が高いと、例えば、細胞への物質導入の場合であれば、生体由来物の生存率が低下し(ダメージを受け)、EP効率が低くなる場合がある。発明者らが検討した結果、例えば、細胞懸濁液を培地シース液流で希釈する場合、希釈率が2倍以上で、細胞生存率の顕著な低下が見られる場合が多く確認された。
 このため、上述のように、懸濁液流との合流時におけるシース液流の厚さの合計を、規制領域における懸濁液流の厚さ以下、すなわち、懸濁液Qの希釈率を2倍以下とすることにより、生体由来物に大量のシース液sが混入することに起因する不都合、例えば、細胞への物質導入であれば生体由来物の生存率低下を抑制して、効率の良い処理を行うことが可能になる。
 この効果を、より好適に得られる点で、懸濁液流との合流時におけるシース液流の厚さの合計は、上述のように規制領域における懸濁液流の厚さの1倍以下であることが好ましく、0.8倍以下がより好ましく、0.5倍以下が更に好ましい。すなわち、電極対の下流における懸濁液Qの希釈率が、上述のように2倍以下であることが好ましく、1.8倍以下であることがより好ましく、1.5倍以下であることが更に好ましい。
<エレクトロポレーションの電圧>
 エレクトロポレーションの電圧は、電極対の間の懸濁液に印加される電界が、所望の電界強度になるように、懸濁液流の厚さ、懸濁液流の電気伝導度(導電率)、シース液流の厚さ、及びシース液流の電気伝導度(導電率)等をふまえて、懸濁液に印加される電界が、所望の電界強度となるように設定する。
 なお、EPにおける導入効率及び生体活性物質の利用効率の低下を引き起こす可能性が有るので、電極の電気抵抗は懸濁液及びシース液の抵抗よりも十分に低いことが好ましい。
 懸濁液に印加する電界の最適値は、生体由来物の種類及びサイズ等によって変わるが、通常、100~2000V/cm程度である。
 電圧はパルス電圧であることが好ましい。また、電極反応(電気分解によるガスの発生、電極劣化等)を均一化するためにバイポーラパルス(プラスとマイナスを交互)としてもよい。
<パルス幅>
 パルス幅の最適値は、生体由来物の種類等によって変わるが、通常0.1~100ms(ミリ秒)、好ましくは1~10ms程度である。
<パルス周期(パルス間隔)>
 パルス周期は、生体由来物が電極(電極長L(図2、9参照))を通過する時間に同期させる(整数倍にする)ことが好ましい。例えば、生体由来物が電極(電極長L)を通過する間に、平均して1~5回、好ましくは1回のパルス電圧を印加する。
 なお、生体由来物が電極(電極長L)を通過する時間は、流量と流路断面積とによって決まる。流路の断面の形状が、全ての内角が90°の四角形の場合、流路断面積は、(電極幅W)×(電極対の間の距離D)である。なお、電極幅Wは図1に示し、電極対の間の距離Dは図2、9に示している。
 本明細書には、国際出願番号PCT/JP2024/012393の(1)細胞の前培養、(2)細胞濃度の調整(濃縮等)、(3)核酸の添加及び混合、(4)エレクトロポレーション(遺伝子導入)、(5)ウイルス産生のための培養(本培養)、並びに(6)ウイルスの回収及び精製の内容が援用され、これらは参照によって本明細書に組み込まれる。
<有用物質の製造方法>
 上述のように有用物質の製造方法は、本発明のEP方法を含む。
 有用物質の製造方法において、好ましくは、生体由来物は細胞であり、生体活性物質は核酸である。本発明のEP方法によって、細胞に核酸を導入し、核酸が導入された細胞を培養することにより、細胞に有用物質を生産させる。
 有用物質の種類は特に限定されないが、好ましくはタンパク質又はウイルスである。
 有用物質としては、例えば、組み換えポリペプチド鎖、組み換え分泌ポリペプチド鎖、抗原結合タンパク質、ヒト抗体、ヒト化抗体、キメラ抗体、マウス抗体、バイスペシフィック抗体、Fc融合タンパク質、断片化免疫イムノグロブリン、一本鎖抗体(scFv)、及び非エンベロープウイルスが挙げられる。
 非エンベロープウイルスは、より具体的には、アデノ随伴ウイルス、アデノウィルス、レンチウイルス、バキュロウイルス、及びレトロウイルス等が挙げられる。非エンベロープウイルスは、当技術分野で公知であり、国際公開第2015/005430号に記載されており、これらは参照によって本明細書に組み込まれる。
 有用物質は、好ましくはヒト抗体、ヒト化抗体、キメラ抗体、マウス抗体、及びアデノ随伴ウイルスである。
 有用物質の製造方法は、回収及び精製工程を有してもよい。
 本発明のエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置は、遺伝子治療薬の製造等に好適に利用可能である。
 本発明は、基本的に以上のように構成されるものである。以上、本発明のエレクトロポレーション方法、有用物質の製造方法及びエレクトロポレーション装置について詳細に説明したが、本発明は上述の実施形態に限定されず、本発明の主旨を逸脱しない範囲において、種々の改良又は変更をしてもよいのはもちろんである。

Claims (18)

  1.  電極対の間を生体由来物と生体活性物質とを含有する懸濁液と、シース液とを、前記シース液で前記懸濁液を挟むようにして主流路に連続的に送液し、前記電極対によって前記懸濁液に電界を印加することで、生体由来物に生体活性物質を導入する、エレクトロポレーション方法であって、
     前記電極対の間において前記懸濁液及び前記シース液の流れが安定した後に、前記懸濁液に電界を印加する印加工程を有する、エレクトロポレーション方法。
  2.  前記懸濁液を挟むように前記シース液を前記主流路に送液する際に、前記懸濁液の供給に第1チューブが用いられ、前記シース液の供給に第2チューブが用いられ、
     前記第1チューブにおいて前記懸濁液の先頭の待機位置が予め定められ、前記第2チューブにおいて前記シース液の先頭の待機位置が予め定められており、
     前記懸濁液及び前記シース液の全ての先頭が、前記待機位置に到達した後に、前記懸濁液及び前記シース液の送液を開始する、請求項1に記載のエレクトロポレーション方法。
  3.  前記印加工程の前に、前記電極対の間において前記懸濁液及び前記シース液の前記流れの安定を判定する判定工程を有し、
     前記判定工程は、前記電極対の間における前記懸濁液及び前記シース液の前記流れの電気抵抗を測定して、前記電気抵抗に基づいて、前記流れの安定を判定するか、又は、
     前記判定工程は、前記第1チューブの前記懸濁液の流量及び前記第2チューブの前記シース液の流量の安定性を検出して前記流れの安定を判定する、請求項1又は2に記載のエレクトロポレーション方法。
  4.  前記判定工程は、前記電極対の前記主流路における上流側又は下流側において、前記電極対の間における前記懸濁液及び前記シース液の前記流れの前記電気抵抗を測定して、前記電気抵抗に基づいて、前記流れの安定を判定する、請求項3に記載のエレクトロポレーション方法。
  5.  前記懸濁液及び前記シース液を送液するタイミングは、前記懸濁液と前記シース液とで同時である、請求項1又は2に記載のエレクトロポレーション方法。
  6.  前記懸濁液及び前記シース液を送液するタイミングは、前記懸濁液よりも前記シース液の方が先である、請求項1又は2に記載のエレクトロポレーション方法。
  7.  電極対の間を生体由来物と生体活性物質とを含有する懸濁液と、シース液とを、前記シース液で前記懸濁液を挟むようにして主流路に連続的に送液し、前記電極対によって前記懸濁液に電界を印加することで、生体由来物に生体活性物質を導入する、エレクトロポレーション方法であって、
     前記懸濁液を挟むように前記シース液を前記主流路に送液する際に、前記懸濁液の供給に第1チューブが用いられ、前記シース液の供給に第2チューブが用いられ、
     前記第1チューブにおいて前記懸濁液の先頭の待機位置が予め定められ、前記第2チューブにおいて前記シース液の先頭の待機位置が予め定められており、
     前記懸濁液及び前記シース液の全ての先頭が、前記待機位置に到達したことを検知した後に、前記懸濁液及び前記シース液の送液を開始し、
     前記懸濁液及び前記シース液の前記送液後、前記電極対の間において前記懸濁液及び前記シース液の流れが安定した後に、前記懸濁液に電界を印加する、エレクトロポレーション方法。
  8.  前記印加工程では、前記懸濁液及び前記シース液の送液を開始後、予め設定した時間経過後に、前記懸濁液に前記電界を印加する、請求項1又は7に記載のエレクトロポレーション方法。
  9.  前記第1チューブにおける前記懸濁液の前記先頭の前記位置は、超音波センサで検知される、請求項7に記載のエレクトロポレーション方法。
  10.  請求項1~9のいずれか1項に記載のエレクトロポレーション方法を含む、有用物質の製造方法。
  11.  生体由来物と生体活性物質とを含有する懸濁液を連続的に送液して電界を印加することで、生体由来物に生体活性物質を導入する、エレクトロポレーション装置であって、
     主流路と、
     対向して配置された電極を備え、前記主流路を流れる前記懸濁液に前記電界を印加する電極対と、
     前記主流路に前記懸濁液を供給する懸濁液供給口と、
     前記主流路内の前記懸濁液供給口の下流側に設けられ、前記主流路にシース液を供給するシース液供給口と、
     前記懸濁液及び前記シース液を送液するポンプと、
     前記電極対の間において前記懸濁液及び前記シース液の流れが安定した後に、前記懸濁液に電界を印加する電源部とを有する、エレクトロポレーション装置。
  12.  前記シース液供給口と連通され、前記主流路において前記懸濁液に前記シース流を合流させるシース液流路と、
     前記懸濁液供給口に接続され、前記懸濁液を前記主流路に供給するための第1チューブと、前記シース液供給口に接続され、前記シース液を前記主流路内に供給するための第2チューブとを有し、
     前記第1チューブにおいて前記懸濁液の先頭の待機位置が予め定められ、前記第2チューブにおいて前記シース液の先頭の待機位置が予め定められており、
     前記ポンプにより、前記懸濁液及び前記シース液の全ての先頭が、前記待機位置に到達した後に、前記懸濁液及び前記シース液の送液を開始させる制御部を更に有する、請求項11に記載のエレクトロポレーション装置。
  13.  前記電極対の間において前記懸濁液及び前記シース液の前記流れの安定を判定する判定部を有し、
     前記判定部は、前記電極対の間における前記懸濁液及び前記シース液の前記流れの電気抵抗を測定して、前記電気抵抗に基づいて、前記流れの安定を判定するか、又は、
     前記判定部は、前記第1チューブの前記懸濁液の流量及び前記第2チューブの前記シース液の流量の安定性を検出して前記流れの安定を判定する、請求項12に記載のエレクトロポレーション装置。
  14.  前記電極対の前記主流路における上流側又は下流側に設けられた補助電極を有し、
     前記判定部は、前記補助電極を用いて前記懸濁液及び前記シース液の前記流れの厚み方向の前記電気抵抗を測定して、前記電気抵抗に基づいて、前記流れの安定を判定する、請求項13に記載のエレクトロポレーション装置。
  15.  前記第1チューブにおける前記懸濁液の先頭の位置、及び前記第2チューブにおける前記シース液の先頭の位置を検知するセンサを有し、
     前記制御部は、前記センサにより検知された、前記第1チューブにおける前記懸濁液の前記先頭の位置、及び前記第2チューブにおける前記シース液の前記先頭の位置に基づいて、前記ポンプによる前記懸濁液及び前記シース液の送液を制御する、請求項12に記載のエレクトロポレーション装置。
  16.  生体由来物と生体活性物質とを含有する懸濁液を連続的に送液して電界を印加することで、生体由来物に生体活性物質を導入する、エレクトロポレーション装置であって、
     主流路と、
     対向して配置された電極を備え、前記主流路を流れる前記懸濁液に前記電界を印加する電極対と、
     前記懸濁液に前記電界を印加する電源部と、
     前記主流路に前記懸濁液を供給する懸濁液供給口と、
     前記主流路内の前記懸濁液供給口の下流側に設けられ、前記主流路に前記シース液を供給するシース液供給口と、
     前記懸濁液及び前記シース液を送液するポンプと、
     前記懸濁液供給口に接続され、前記懸濁液を前記主流路に供給するための第1チューブと、
     前記シース液供給口に接続され、前記シース液を前記主流路内に供給するための第2チューブと、
     前記第1チューブにおける前記懸濁液の先頭の位置、及び前記第2チューブにおける前記シース液の先頭の位置を検知するセンサと、
     前記ポンプによる前記懸濁液及び前記シース液の送液を制御する制御部とを有し、
     前記第1チューブにおいて前記懸濁液の先頭の待機位置が予め定められ、前記第2チューブにおいて前記シース液の先頭の待機位置が予め定められており、
     前記制御部は、前記センサにより前記懸濁液及び前記シース液の全ての先頭が、前記待機位置に到達したことが検知された後に、前記ポンプにより前記懸濁液及び前記シース液の前記送液を開始させ、
     前記懸濁液及び前記シース液の前記送液後、前記電極対の間において前記懸濁液及び前記シース液の流れが安定した後に、前記電源部により前記懸濁液に前記電界を印加する、エレクトロポレーション装置。
  17.  前記制御部は、前記ポンプにより前記懸濁液及び前記シース液の送液を開始し、予め設定した時間経過後に、前記電源部により前記懸濁液に前記電界を印加する、請求項12又は15に記載のエレクトロポレーション装置。
  18.  前記第1チューブにおける前記懸濁液の前記先頭の前記位置を検知する前記センサは、超音波センサである、請求項15又は16に記載のエレクトロポレーション装置。
