 本発明は、ハイポブラスト幹細胞の製造方法、ハイポブラスト幹細胞の誘導用培地、ハイポブラスト幹細胞、卵黄嚢オルガノイドの製造方法、及び卵黄嚢オルガノイドに関する。
 本願は、2024年5月28日に、日本に出願された特願2024-086184号に基づき優先権を主張し、その内容をここに援用する。
 ヒトの着床前胚は、将来、胎児に寄与するエピブラスト、胎盤に寄与する栄養外胚葉、及び卵黄嚢に寄与するハイポブラストの3種類の細胞で構成されている。エピブラストからは胚性幹細胞(Embryonic stem cell:ES細胞)が樹立されている。栄養外胚葉からは栄養膜幹細胞(Trophoblast stem cell:TS細胞)が樹立されている(特許文献1)。これらの細胞は、基礎から応用まで幅広い分野で活用が進んでいる。
 一方、マウスにおいては、卵黄嚢への分化能を有する細胞として、原始内胚葉幹細胞(Primitive endoderm stem cell:PrES細胞)が報告されている(非特許文献1)。また、ヒト多能性幹細胞から、ハイポブラスト様細胞が誘導されたことが報告されている(非特許文献2)。

 ES細胞及びTS細胞に加えて、卵黄嚢への分化能を有するハイポブラスト幹細胞を樹立することができれば、哺乳動物の初期発生の基礎研究、妊娠初期の胎子に対する薬物影響試験、及び生殖補助医療等に有用であると期待される。
 非特許文献1では、マウス胚盤胞を、線維芽細胞増殖因子4(Fibroblast Growth Factor 4:FGF4)、血小板由来成長因子(Platelet-Derived Growth Factor:PDGF)、及びGSK3β阻害剤の存在下で培養することにより、マウスPrES細胞を誘導している。しかしながら、非特許文献1に記載の条件では、ヒトのハイポブラスト幹細胞を誘導することはできない。
 非特許文献2では、ヒト多能性幹細胞を、FGF4、骨形成因子4(Bone morphogenetic Protein:BMP4)、インターロイキン6(Interleukin 6:IL-6)、PDGF、Wnt阻害剤、及びTGFβ/SMAD阻害剤の存在下で培養することにより、ハイポブラスト様細胞を誘導している。しかしながら、非特許文献2に記載のハイポブラスト様細胞は、長期間(例えば、6カ月以上)培養して維持することができない。
 本開示は、ヒトにも適用可能であり、長期培養により維持可能なハイポブラスト幹細胞を製造可能な、ハイポブラスト幹細胞の製造方法、前記製造方法に使用可能なハイポブラスト幹細胞の誘導用培地、前記製造方法により製造されるハイポブラスト幹細胞、前記ハイポブラスト幹細胞を用いた卵黄嚢オルガノイドの製造方法、及び前記ハイポブラスト幹細胞を含む卵黄嚢オルガノイドを提供することを課題とする。
 本開示は、以下の態様を含む。
(1)哺乳動物の卵黄嚢から単離された細胞を、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で培養する工程を含む、ハイポブラスト幹細胞の製造方法。
(2)前記培地が、TGFβ/SMAD阻害剤をさらに含有する、(1)に記載のハイポブラスト幹細胞の製造方法。
(3)前記培地が、HDAC阻害剤及びp38阻害剤からなる群より選択される少なくとも1種をさらに含有する、(1)又は(2)に記載のハイポブラスト幹細胞の製造方法。
(4)上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する、ハイポブラスト幹細胞の誘導用培地。
(5)TGFβ/SMAD阻害剤をさらに含有する、(4)に記載のハイポブラスト幹細胞の誘導用培地。

 本開示によれば、ヒトにも適用可能であり、長期培養により維持可能なハイポブラスト幹細胞を製造可能な、ハイポブラスト幹細胞の製造方法、前記製造方法に使用可能なハイポブラスト幹細胞の誘導用培地、前記製造方法により製造されるハイポブラスト幹細胞、前記ハイポブラスト幹細胞を用いた卵黄嚢オルガノイドの製造方法、及び前記ハイポブラスト幹細胞を含む卵黄嚢オルガノイドを提供することができる。

[定義]
 「を含む」(comprise)という用語は、対象となる構成要素以外の構成要素を含んでいてもよいことを意味する。「からなる」(consist of)という用語は、対象となる構成要素以外の構成要素を含まないことを意味する。「から本質的になる」(consist essentially of)という用語は、対象となる構成要素以外の構成要素を特別な機能を発揮する態様(発明の効果を完全に喪失させる態様など)では含まないことを意味する。本明細書において、「を含む」(comprise)と記載する場合、「からなる」(consist of)態様、及び「から本質的になる」(consist essentially of)態様を包含する。
 「~」を用いて表される数値範囲は、「~」の前後に記載される数値を下限値及び上限値として含む範囲を意味する。特定のパラメータについて、複数の上限値及び下限値が記載されている場合、これらの上限値及び下限値の内、任意の上限値と下限値とを組合せて好適な数値範囲とすることができる。
 タンパク質、細胞、及びオルガノイドは、単離されたものであり得る。「単離された」とは、他の成分から分離された状態を意味する。「単離された」成分は、天然状態から分離された状態であってもよい。「単離された」ものは、他の成分を実質的に含まないものであり得る。「他の成分を実質的に含まない」とは、単離された成分に含まれる他の成分の含有量が無視できる程度であることを意味する。単離された成分に含まれる他の成分の含有量は、例えば、10質量%以下、5質量%以下、4質量%以下、3質量%以下、2質量%以下、1質量%以下、0.5質量%以下、又は0.1質量%以下であり得る。本明細書に記載されるタンパク質、細胞、及びオルガノイドは、それぞれ、単離されたタンパク質、単離された細胞、及び単離されたオルガノイドであり得る。
[ハイポブラスト幹細胞の製造方法]
 本開示の第1の態様は、ハイポブラスト幹細胞の製造方法である。前記製造方法は、哺乳動物の卵黄嚢から単離された細胞を、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で培養する工程(以下、「培養工程」ともいう)を含む。
(ハイポブラスト幹細胞)
 「ハイポブラスト幹細胞」(以下、「HS細胞」ともいう)とは、下記の(a)~(e)の特徴を有する細胞を指す。HS細胞は、下記(b)~(e)の特徴を有する細胞であってもよい。
 (a)哺乳動物の卵黄嚢から単離された細胞から誘導された細胞である;
 (b)前記哺乳動物の卵黄嚢から単離された細胞と比較して、α-フェトプロテインの発現量が低い;
 (c)α-フェトプロテイン及びアルブミンからなる群より選択される少なくとも1種を発現する細胞を含む卵黄嚢オルガノイドへの分化能を有する;
 (d)増殖能を有する;及び
 (e)細胞塊を形成する。
(a)について:
 HS細胞は、卵黄嚢から単離された細胞(以下、「卵黄嚢細胞」ともいう)から誘導された細胞である。HS細胞は、卵黄嚢から単離された細胞を、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で培養することにより、誘導することができる。培養方法の具体例については後述する。
(b)について:
 HS細胞は、卵黄嚢細胞と比較して、α-フェトプロテイン(AFP)の発現量が低い。比較対象となる卵黄嚢細胞は、HS細胞の誘導元である卵黄嚢細胞である。AFPは、卵黄嚢において高発現している卵黄嚢マーカーである。卵黄嚢細胞からHS細胞を誘導すると、AFPの発現量が低下する。HS細胞におけるAFPの発現量の低下は、HS細胞が、卵黄嚢細胞よりも未分化な状態に移行したことを示すと考えられる。
 卵黄嚢細胞及びHS細胞におけるAFPの発現量は、公知の方法により、測定することができる。そのような方法としては、例えば、RNA-seq解析、定量的逆転写PCR(RT-qPCR)、ノーザンブロッティング解析、蛍光免疫染色、ウェスタンブロッティング解析、ELISA等が挙げられる。AFPの発現量の測定に使用可能なAFPの配列情報は、GeneBank等の公知の配列データベースから入手することができる。ヒトAFPの配列情報は、例えば、GeneBankに、アクセッション番号NM_001134.3、及びNM_001354717.2で登録されているもの等を利用することができる。AFPの発現量の測定に使用可能な抗AFP抗体は、市販のものを用いることができる。
(c)について:
 HS細胞は、卵黄嚢オルガノイドへの分化能を有する。「卵黄嚢オルガノイド」は、AFP及びアルブミンからなる群より選択される少なくとも1種を発現する細胞(以下、「AFP/Alb発現細胞」ともいう)を含むオルガノイドである。AFP/Alb発現細胞は、AFPのみを発現する細胞でもよく、アルブミンのみを発現する細胞でもよく、AFP及びアルブミンの両方を発現する細胞でもよい。卵黄嚢オルガノイドは、AFP/Alb発現細胞から主に構成されていることが好ましい。卵黄嚢オルガノイドは、例えば、全細胞の50%以上、60%以上、70%以上、80%以上、90%以上、又は95%以上がAFP/Alb発現細胞により構成されていることが好ましい。
 卵黄嚢オルガノイドにおけるAFP及びアルブミンの発現は、上記のような公知の方法により測定することができる。AFP及びアルブミンの発現量の測定に使用可能なAFPの配列情報は、GeneBank等の公知の配列データベースから入手することができる。ヒトアルブミンの配列情報は、例えば、GeneBankに、アクセッション番号NM_000477.7で登録されているもの等を利用することができる。アルブミンの発現量の測定に使用可能な抗アルブミン抗体は、市販のものを用いることができる。
 卵黄嚢オルガノイドにおけるAFP及びアルブミンの発現量は、HS細胞の誘導元である卵黄嚢と同程度であることが好ましい。例えば、卵黄嚢におけるAFPの発現量を1とした場合、卵黄嚢オルガノイドにおけるAFPの相対発現量は、0.1以上、0.2以上、0.5以上、0.6以上、又は0.7以上であることが好ましい。卵黄嚢におけるAFPの発現量を1とした場合の、卵黄嚢オルガノイドにおけるAFPの相対発現量の上限値は、例えば、10.0以下、9.0以下、8.0以下、7.0以下、6.0以下、5.0以下、4.0以下、3.0以下、又は2.0以下である。例えば、卵黄嚢におけるアルブミンの発現量を1とした場合、卵黄嚢オルガノイドにおけるアルブミンの相対発現量は、0.1以上、0.2以上、0.5以上、0.6以上、又は0.7以上であることが好ましい。卵黄嚢におけるアルブミンの発現量を1とした場合の、卵黄嚢オルガノイドにおけるアルブミンの相対発現量の上限値は、例えば、10.0以下、9.0以下、8.0以下、7.0以下、6.0以下、5.0以下、4.0以下、3.0以下、又は2.0以下である。
 卵黄嚢オルガノイドは、卵黄嚢と遺伝子発現パターンが類似した細胞から構成されることが好ましい。例えば、RNA-seq解析又はシングルRNA-seq解析により得られた遺伝子発現データのUMAP解析を行った場合に、卵黄嚢と同じクラスターに位置することが好ましい。
 細胞が、卵黄嚢オルガノイドへの分化能を有するか否かは、評価対象の細胞をハイドロゲル内で培養することにより、確認することができる。評価対象の細胞がオルガノイドへの分化能を有する場合、ハイドロゲル内で5~20日程度培養することにより、オルガノイドが形成される。形成されたオルガノイドが、AFP/Alb細胞を含む場合、当該オルガノイドは、卵黄嚢オルガノイドであると判定することができる。形成されたオルガノイドが卵黄嚢オルガノイドである場合、当該細胞は卵黄嚢オルガノイドへの分化能を有する、と判定することができる。HS細胞からの卵黄嚢オルガノイドの形成方法の詳細については、後述する。
(d)について:
 HS細胞は、増殖能を有する。HS細胞が増殖可能な培地としては、例えば、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地が挙げられる。そのような培地の具体例としては、後述のハイポブラスト幹細胞の誘導用培地が挙げられる。
 HS細胞の培養条件としては、動物細胞の培養に通常用いられる条件を用いることができる。培養温度としては、32~40℃(好ましくは35~38℃、典型的には37℃)が挙げられる。CO濃度としては、2~5%(好ましくは5%)が挙げられる。HS細胞の培養は、ラミニン等の培養基質を用いた接着培養とすることができる。「培養基質」とは、細胞の培養において、細胞が接着するための足場となる物質を指す。培養基質としては、細胞外マトリクスが挙げられる。
 HS細胞は、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地を用いて、培養基質の存在下、37℃、5%CO条件下で培養した場合に、増殖可能な細胞であるともいえる。HS細胞が増殖すると、後述の(e)に記載される細胞塊のサイズが大きくなっていく。そのため、細胞塊のサイズの増大により、HS細胞の増殖を確認することができる。
(e)について:
 HS細胞は、細胞塊を形成する。より具体的には、HS細胞は、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で培養することにより、図1に示すような球状の細胞塊を形成する。細胞塊のサイズは、例えば、最小径20~200μm程度である。細胞塊の最大径は、例えば、20~200μm程度である。HS細胞により形成される細胞塊は、位相差顕微鏡を用いて、倍率20倍で観察した場合、通常、細胞間の境界を明瞭に観察することができない。これは、細胞が密に集まった細胞密度の高い細胞塊であるためと考えられる。
 HS細胞は、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地を用いて、培養基質の存在下、37℃、5%CO条件下で培養した場合に、細胞塊を形成可能な細胞であるともいえる。
 その他の特徴:
 HS細胞は、上記(a)~(e)の特徴に加えて、下記(f)の特徴を有することが好ましい。
(f)上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で継代培養を行うことにより、6カ月以上の維持培養が可能である。
 上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地としては、後述のハイポブラスト幹細胞の誘導用培地が挙げられる。継代培養の培養条件としては、動物細胞の培養に通常用いられる条件を用いることができる。培養温度としては、上記(d)で挙げた培養条件と同様の培養条件が挙げられる。
 継代は、例えば、3~10日毎に行ってもよく、培養容器がコンフルエントな状態となったときに行ってもよい。HS細胞の継代は、動物細胞の継代に通常用いられる方法により、行うことができる。例えば、タンパク質分解酵素(例えば、トリプシン、トリプシン代替品等)を添加し、ピペッティング等により細胞を培養容器から剥離する。この際、細胞塊を、遊離細胞に分散させてもよい。剥離した細胞を遠心分離等により回収する。回収した細胞を新たな培地に播種することにより、継代を行うことができる。
 HS細胞は、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地を用いて、培養基質の存在下、37℃、5%CO条件下で継代培養を行うことにより、6カ月以上維持培養が可能な細胞であることが好ましい。
 「維持培養」とは、細胞の性質が維持されたまま、細胞を培養することをいう。HS細胞においては、維持培養中、上記(a)~(e)の特徴が維持される。
 HS細胞のサイズは、誘導元の卵黄嚢細胞が由来する生物により異なる。例えば、ヒト卵黄嚢細胞から誘導されたHS細胞のサイズは、細胞径10~50μm程度の大きさである。
(培養工程)
 培養工程では、哺乳動物の卵黄嚢から単離された細胞(卵黄嚢細胞)を、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で培養する。
<卵黄嚢細胞>
 卵黄嚢細胞が由来する哺乳動物は、妊娠時に卵黄嚢を有する哺乳動物である。哺乳動物としては、例えば、ヒト、及びヒト以外の哺乳動物が挙げられる。ヒト以外の哺乳動物としては、例えば、ヒト以外の霊長類(サル、チンパンジー、ゴリラ、マーモセットなど)、齧歯類(マウス、ハムスター、ラット、モルモットなど)、ウサギ、イヌ、ネコ、ウシ、ブタ、ヤギ、ヒツジ、ウマ等が挙げられるが、これらに限定されない。卵黄嚢細胞が由来する哺乳動物としては、霊長類が好ましく、ヒトがより好ましい。
 卵黄嚢細胞は、卵黄嚢から単離された細胞である。卵黄嚢細胞は、卵黄嚢から単離された細胞の培養株を用いてもよい。卵黄嚢は、例えば、妊娠初期の哺乳動物から摘出してもよい。哺乳動物がヒトである場合、妊娠中絶手術により摘出されたものを用いてもよい。
 卵黄嚢からの細胞の単離方法は、公知の方法を用いることができる。卵黄嚢細胞は、例えば、卵黄嚢に機械的及び/又は酵素的処理を適宜使用して、細胞を分離することにより、単離することができる。機械的処理としては、ハサミ又はメスによる切断等が挙げられる。酵素処理としては、タンパク質分解酵素(トリプシン等)、細胞外マトリクス分解活性を有するプロテアーゼ(例えば、ディスパーゼ、コラゲナーゼ等)による処理が挙げられる。子宮内膜上皮組織の機械的及び/又は酵素的処理の後、遠心分離、ストレーナー又はフィルター等を用いて卵黄嚢細胞を回収することができる。回収した卵黄嚢細胞は、適宜、培地(TS培地等)、緩衝液(リン酸緩衝生理食塩水(PBS等))等で洗浄してもよい。
<培地>
 培地としては、例えば、基礎培地に、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤等を添加した培地が挙げられる。
≪基礎培地≫
 基礎培地としては、動物細胞の培養に一般的に用いられる培地が挙げられる。基礎培地としては、例えば、Doulbecco’s modified Eagle’s Medium(DMEM)培地、DMEM/F12培地、Advanced DMEM/F12培地、IMDM培地、Medium199培地、Eagle’sMinimum Essential Medium(EMEM)培地、αMEM培地、Ham’s F12培地、RPMI1640培地、Fischer’s培地、及びこれらの2種以上の混合培地等が挙げられる。好ましい基礎培地としては、例えば、DMEM/F12が挙げられる。
 基礎培地は、必要に応じて、血清(牛胎児血清(FBS)など)、及び/又は血清代替物を含んでいてもよい。血清代替物としては、例えば、アルブミン、トランスフェリン、亜セレン酸ナトリウム、ITS-X(Invitrogen)、ノックアウト血清代替物(Knockout Serum Replacement(KSR)、N2サプリメント(Invitrogen)、B27サプリメント(Invitrogen)、脂肪酸、インスリン、コラーゲン前駆体、微量元素、2-メルカプトエタノール、3’-チオールグリセロール等が挙げられる。基礎培地は、必要に応じて、脂質、アミノ酸、L-グルタミン、Glutamax、非必須アミノ酸、ビタミン、増殖因子、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類等の成分を含んでいてもよい。これらは、適宜組み合わせて用いることができる。
 基礎培地としては、例えば、上記のような基礎培地(例えば、DMEM/F12)に、ウシ血清アルブミン(BSA)、ITS-X、血清代替物(KSR等)、及び抗生物質(ペニシリン、ストレプトマイシン等)を添加した培地が挙げられる。基礎培地の具体例としては、後述の実施例で用いられたTS basal培地が挙げられる。
≪上皮成長因子≫
 上皮成長因子(epidermal growth factor:EGF)は、細胞表面に存在するEGF受容体(EGFR)に結合してEGFシグナル伝達を誘発し、分裂促進因子として作用する。EGFが由来する生物は、特に限定されないが、卵黄嚢細胞が由来する生物と同じことが好ましい。ヒト卵黄嚢細胞を用いる場合、ヒトEGFが好ましい。ヒトEGFは、非ヒト細胞により生産された組換え体であってもよい。EGFは、市販のものを用いることができる。培地中のEGFの濃度は、特に限定されないが、例えば、5~200ng/mLとすることができる。培地中のEGFの濃度は、10~150ng/mLが好ましく、10~100ng/mLがより好ましく、10~80ng/mLがさらに好ましく、10~60ng/mLが特に好ましい。
≪GSK3β阻害剤≫
 GSK(Glycogen Synthase Kinase)3β阻害剤は、GSK3βの機能、例えば、キナーゼ活性(例えば、βカテニンに対するリン酸化能)を阻害する物質である。GSK3β阻害剤としては、例えば、6-[[2-[[4-(2,4-ジクロロフェニル)-5-(4-メチル-1H-イミダゾール-2-イル)-2-ピリミジニル]アミノ]エチル]アミノ]ニコチノニトリル、ケンパウロン(Kenpaullone)、1-アザケンパウロン(Azakenpaullone)、CHIR98014、AR-A014418、CT99021、CT20026、SB216763、AR-A014418、リチウム、SB415286、TDZD-8、BIO、BIO-アセトキシム、(5-メチル-1H-ピラゾール-3-イル)-(2-フェニルキナゾリン-4-イル)アミン、ピリドカルバゾール-シクロペンタジエニルルテニウム(cyclopenadienylruthenium)複合体、TDZD-8 4-ベンジル-2-メチル-1,2,4-チアジアゾリジン-3,5-ジオン、2-チオ(3-ヨードベンジル)-5-(1-ピリジル)-[1,3,4]-オキサジアゾール、OTDZT、アルファ-4-ジブロモアセトフェノン、AR-AO 144-18、3-(1-(3-ヒドロキシプロピル)-1H-ピロロ[2,3-b]ピリジン-3-イル]-4-ピラジン-2-イル-ピロール-2,5-ジオン;TWSl 19ピロロピリミジン化合物、L803 H-KEAPPAPPQSpP-NH2又はそのミリストイル化形態;2-クロロ-1-(4,5-ジブロモ-チオフェン-2-イル)-エタノン、SB216763、及びSB415286等の低分子阻害剤が挙げられる。GSK3β阻害剤は、GSK3βに対するアンチセンス核酸、siRNA、ドミナントネガティブ変異体、及びそれらの発現ベクター等であってもよい。6-[[2-[[4-(2,4-ジクロロフェニル)-5-(4-メチル-1H-イミダゾール-2-イル)-2-ピリミジニル]アミノ]エチル]アミノ]ニコチノニトリルの市販品としては、CHIR99021等が挙げられる。GSK3β阻害剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
 GSK3β阻害剤は、CHIR99021を用いることが好ましい。
 培地中のGSK3β阻害剤の濃度は、特に限定されないが、例えば、0.1~50μMとすることができ、0.2~30μMが好ましく、0.5~20μMがより好ましく、1~10μMがさらに好ましい。
≪ROCK阻害剤≫
 ROCK(Rho associated coiled-coil containing protein kinase:Rho結合キナーゼ)阻害剤は、Rho結合キナーゼの機能を阻害する物質である。ROCK阻害剤としては、例えば、trans-N-(4-ピリジル)-4-(1-アミノエチル)-シクロヘキサンカルボキサミド、1-(5-イソキノリニルスルホニル)ホモピペラジン(1-(5-Isoquinolinylsulfonyl)homopiperazine)、またはこれらの塩等が挙げられる。ROCK阻害剤としては、例えば、Fasudil/HA1077、H-1152、およびWf-536等の低分子阻害剤、並びにそれらの誘導体等も挙げられる。ROCK阻害剤は、ROCKに対するアンチセンス核酸、siRNA、ドミナントネガティブ変異体、及びそれらの発現ベクター等であってもよい。
 trans-N-(4-ピリジル)-4-(1-アミノエチル)-シクロヘキサンカルボキサミド又はその塩の市販品としては、Y27632((R)-(+)-trans-N-(4-pyridyl)-4-(1-aminoethyl)-cyclohexanecarboxamide・2HCl・HO)等が挙げられる。ROCK阻害剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
 ROCK阻害剤は、Y27632を用いることが好ましい。
 培地中のROCK阻害剤の濃度は、特に限定されないが、例えば、0.1~50μMとすることができ、0.5~20μMが好ましく、1~15μMがより好ましく、1~10μMがさらに好ましい。
≪他の因子≫
 培地は、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤に加えて、他の因子を含有してもよい。他の因子としては、例えば、TGFβ/SMAD阻害剤、HDAC阻害剤、p38阻害剤、ニコチンアミド等が挙げられる。
・TGFβ/SMAD阻害剤
 TGFβ(Transforming Growth Factor β)/SMAD阻害剤は、TGFβのALK(Activin receptor-Like Kinase)ファミリーへの結合を阻害する物質、又はALKファミリーによるSMADのリン酸化を阻害する物質である。TGFβ/SMAD阻害剤としては、例えば、ALK4、ALK5又はALK7等のキナーゼ活性を阻害する物質等が挙げられる。TGFβ/SMAD阻害剤としては、例えば、Lefty-1(NCBIアクセッション番号として、マウス:NP_034224.1、ヒト:NP_066277.1が例示される)、SB431542、SB202190(以上、R.K.Lindemann et al., Mol. Cancer, 2003, 2:20)、SB505124 (GlaxoSmithKline)、NPC30345、SD093、SD908、SD208(Scios)、LY2109761、LY364947、LY580276(Lilly Research Laboratories)、A83-01(3-(6-メチル-2-ピリジニル)-N-フェニル-4-(4-キノリニル)-1H-ピラゾール-1-カルボチオアミド、WO2009146408)、ALK5阻害剤II(2-[3-[6-メチルピリジン-2-イル]-1H-ピラゾル-4-イル]-1,5-ナフチリジン)、TGFβRIキナーゼ阻害剤VIII(6-[2-tert-ブチル-5-[6-メチル-ピリジン-2-イル]-1H-イミダゾル-4-イル]-キノキサリン)並びにこれらの誘導体等が挙げられるが、これらに限定されない。TGFβ/SMAD阻害剤は、ALKファミリーに対するアンチセンス核酸、RNA干渉誘導性核酸(例、siRNA)、ドミナントネガティブ変異体、及びそれらの発現ベクター等であってもよい。TGFβ/SMAD阻害剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
 TGFβ/SMAD阻害剤は、A83-01を用いることが好ましい。
 培地中のTGFβ/SMAD阻害剤の濃度は、特に限定されないが、例えば、0.1~50μMとすることができ、0.5~20μMが好ましく、1~15μMがより好ましく、1~10μMがさらに好ましい。
・HDAC阻害剤
 HDAC(Histone Deacetylase)阻害剤は、ヒストンデアセチラーゼの機能を阻害する物質である。HDAC阻害剤としては、HDACの機能を抑制できるものであれば、特に限定されない。HDAC阻害剤としては、例えば、バルプロ酸(VPA)、トリコスタチンA、酪酸ナトリウム、MC1293、M344等の低分子阻害剤が挙げられる。HDAC阻害剤は、HDACに対するアンチセンス核酸、siRNA、ドミナントネガティブ変異体、およびそれらの発現ベクター等であってもよい。HDAC阻害剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
 HDAC阻害剤は、VPAを用いることが好ましい。
 培地中のHDAC阻害剤の濃度は、特に限定されないが、例えば、0.01~10μMとすることができ、0.05~5μMが好ましく、0.1~2μMがより好ましく、0.5~1μMがさらに好ましい。
・p38阻害剤
 p38阻害剤は、p38 MAPK(P38 mitogen-activated protein kinase)の機能を阻害する物質である。p38阻害剤としては、例えば、SB202190(4-(4-フルオロフェニル)-2-(4-ヒドロキシフェニル)-5-(4-ピリジル)-1H-イミダゾール)、SB203580(4-[4-(4-フルオロフェニル)-2-[4-(メチルスルフィニル)フェニル]-1H-イミダゾール-5-イル]ピリジン)、VX702(6-(N-カルバモイル-2,6-ジフルオロアニリノ)-2-(2,4-ジフルオロフェニル)ピリジン-3-カルボキシアミド)、VX745(5-(2,6-ジクロロフェニル)-2-[2,4-ジフルオロフェニル)チオ]-6H-ピリミド[1,6-b]ピリダジン-6-ワン)、PD169316(4-(4-フルオロフェニル)-2-(4-ニトロフェニル)-5-(4-ピリジル)-1H-イミダゾール)、RO4402257(6-(2,4-ジフルオロフェノキシ)-2-{[3-ヒドロキシ-1-(2-ヒドロキシエチル)プロピル]アミノ}-8-メチルピリド[2,3-D]ピリミジン-7(8h)-ワン)、BIRB796(1-[5-tert-ブチル-2-(4-メチルフェニル)ピラゾール-3-イル]-3-[4-(2-モルフォリン-4-イレトキシ)ナフタレン-1-イル]ウレア)等が挙げられる。p38阻害剤は、p38 MAPKに対するアンチセンス核酸、siRNA、ドミナントネガティブ変異体、及びそれらの発現ベクター等であってもよい。p38阻害剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
 p38阻害剤は、SB202190を用いることが好ましい。
 培地中のp38阻害剤の濃度は、特に限定されないが、例えば、0.01~50μMとすることができ、0.1~20μMが好ましく、0.5~10μMがより好ましく、0.5~5μMがさらに好ましい。
・ニコチンアミド
 培地は、上記成分に加えてニコチンアミドを含有してもよい。
 培地中のニコチンアミドの濃度は、特に限定されないが、例えば、0.01~500nMとすることができ、0.1~100nMが好ましく、0.5~50nMがより好ましく、1~30nMがさらに好ましい。
 培地としては、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、及びTGFβ/SMAD阻害剤を含有する培地;上皮成長因子、GSK3β阻害剤、及びROCK阻害剤に加えて、HDAC阻害剤及びp38阻害剤からなる群より選択される少なくとも1種を含有する培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、及びTGFβ/SMAD阻害剤に加えて、HDAC阻害剤及びp38阻害剤からなる群より選択される少なくとも1種を添加した培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、及びHDAC阻害剤を含有する培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、及びp38阻害剤を含有する培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、TGFβ/SMAD阻害剤、及びHDAC阻害剤を含有する培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、TGFβ/SMAD阻害剤、及びp38阻害剤を含有する培地;上皮成長因子、GSK3β阻害剤、ROCK阻害剤、TGFβ/SMAD阻害剤、HDAC阻害剤、及びp38阻害剤を含有する培地等が挙げられる。培地は、上記各培地がさらにニコチンアミドを含有するものであってもよい。培地は、基礎培地(例えば、TS basal培地)に、上記各因子を含有させたものであってもよい。培地の具体例としては、例えば、後述の実施例で用いられたYS-2D培地、YS培地、YS-2D-3培地、YS-2D-5培地等が挙げられる。培地は、液体培地が好ましい。
<培養条件>
 培養条件は、動物細胞の培養に一般的に用いられる条件とすることができる。例えば、培養温度32~40℃(好ましくは35~38℃、典型的には37℃)、CO濃度2~5%(好ましくは5%)とすることができる。
 培養開始時の細胞播種密度としては、例えば、10~10個/ml、又は10~10個/mlが挙げられる。
 培養は、例えば、培養基質を用いた接着培養とすることができる。培養基質としては、例えば、細胞外マトリクスを用いることができる。細胞外マトリクスとしては、例えば、ラミニン、IV型コラーゲン、ヘパラン硫酸プロテオグリカン、エンタクチン等の基底膜系細胞外マトリクス;ヒアルロン酸、II型コラーゲン、リンクタンパク質等の軟骨系細胞外マトリクス等;I型コラーゲン、プロテオグリカン(バーシカン、デコリン等)、フィブロネクチン等の間質系細胞外マトリクス等が挙げられる。培養基質は、Matrigel(CORNING)、iMatrix(登録商標)(FUJIFILM Wako)等の市販の製品を用いてもよい。培養基質としては、例えば、iMatrix511等が好ましい。
 培養容器は、動物細胞の培養に一般的に用いる培養容器を用いることができる。培養容器としては、例えば、ウェルプレート、培養ディッシュ、培養フラスコ等が挙げられる。
 培養期間は、例えば、2~60日程度、4~50日程度、又は5~30日程度とすることができる。培養は、細胞におけるAFP、アルブミン等の卵黄嚢マーカーの発現量が低下するまで行ってもよい。例えば、卵黄嚢細胞と比較して、AFP、アルブミン等の卵黄嚢マーカーの発現量が、1/100以下、1/300以下、1/500以下、1/800以下、又は1/1000以下となるまで、培養を行ってもよい。
 培養は、例えば、細胞が、細胞塊(例えば、図1のような細胞塊)を形成するまで行ってもよい。
 培養は、適度な間隔で、継代培養を行ってもよい。継代は、例えば、コンフルエントな状態となったときに行うことができる。継代は、例えば、2~10日毎、2~8日毎、2~5日毎、又は2~4日毎に行うことができる。継代培養の際には、ピペッティング等により、細胞を剥離してもよい。細胞の剥離の際には、トリプシン等のタンパク質酵素を用いてもよい。
 継代培養に用いる培地としては、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地が挙げられる。継代培養に用いる培地は、上記<培地>の項で説明した培地と同様のものを用いることができる。
 卵黄嚢細胞を上記のように培養することにより、HS細胞を得ることができる。
(他の工程)
 本実施形態の製造方法は、上記培養工程に加えて、他の工程を含んでもよい。他の工程としては、例えば、培養工程後にHS細胞を回収する工程、HS細胞における卵黄嚢マーカー発現量を測定する工程、HS細胞を位相差顕微鏡等で観察する工程等が挙げられる。
 HS細胞の回収は、ピペッティング等により細胞を剥離した後、細胞懸濁液を遠心分離することにより、行うことができる。回収した細胞は、新たな培地に継代してもよく、細胞凍結保存液等を用いて凍結保存してもよい。細胞凍結保存液としては、CELLBANKER(登録商標)1(ゼノジェンファーマ)等の市販の凍結保存液を用いることができる。あるいは、上記のような方法で、継代培養により維持してもよい。
 HS細胞は、AFP、アルブミン等の卵黄嚢マーカーの発現量を測定してもよい。これらの卵黄嚢マーカーの発現量が、卵黄嚢細胞と比較して低下している場合、HS細胞が誘導されたと判断することができる。卵黄嚢マーカーの発現量の測定は、上記のような公知の方法により行うことができる。
 HS細胞は、位相差顕微鏡等により、細胞塊の形成を観察してもよい。細胞が細胞塊を形成している場合、HS細胞が誘導されたと判断することができる。
 本実施形態の製造方法により得られたHS細胞は、卵黄嚢への分化能を有すると考えられる。そのため、HS細胞は、後述の卵黄嚢オルガノイド(以下、「HSオルガノイド」ともいう)の形成に加えて、妊娠初期における薬物評価試験のモデル細胞、再生医療、生殖補助医療等への応用が期待される。
[ハイポブラスト幹細胞の誘導用培地]
 本開示の第2の態様は、ハイポブラスト幹細胞の誘導用培地(以下、「HS細胞誘導用培地」ともいう)である。HS細胞誘導用培地は、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する。
 HS細胞誘導用培地は、上記[ハイポブラスト幹細胞の製造方法]における(培養工程)の<培地>の項で説明した培地と同じである。
 HS細胞誘導用培地は、卵黄嚢細胞から、HS細胞を誘導するために用いることができる。
[ハイポブラスト幹細胞]
 本発明の第3の態様は、ハイポブラスト幹細胞(HS細胞)である。HS細胞は、上記[ハイポブラスト幹細胞の製造方法]における(ハイポブラスト幹細胞)の項で説明したHS細胞と同じである。一実施形態において、HS細胞は、ヒトの卵黄嚢から単離された細胞から誘導された細胞である。
[卵黄嚢オルガノイドの製造方法]
 本発明の第4の態様は、卵黄嚢オルガノイドの製造方法である。前記製造方法は、上記第3の態様に係るハイポブラスト幹細胞を、ハイドロゲル内で培養する工程(以下、「ハイドロゲル内培養工程」ともいう)を含む。
(ハイドロゲル内培養工程)
 ハイドロゲル内培養工程では、HS細胞を、ハイロドゲル内で培養する。
 ハイドロゲルは、水を内部に含んでゲル化した高分子材料である。ハイドロゲルに用いられる高分子材料としては、例えば、コラーゲン(I型、II型、III型、V型、XI型等)、ゼラチン、エラスチン、プロテオグリカン、グリコサミノグリカン、フィブロネクチン、ビトロネクチン、ラミニン、ペクチン、ヒアルロン酸、キチン、及びキトサン等の細胞外マトリクス;アルギン酸、及びデンプン等の多糖;ポリリジン、及びポリアルギニン等のアミノ酸ポリマー;フィブリン等の繊維状タンパク質;並びに、ポリエチレングリコール等の合成高分子ポリマー等が挙げられるが、これらに限定されない。ハイドロゲルは、Matrigel(登録商標)(CORNING)、iMatrix(登録商標)(FUJIFILM Wako)等の市販品を用いてもよい。
 培養は、HS細胞をハイドロゲル内に埋め込むことにより、開始することができる。HS細胞は、上記のように継代培養した細胞を用いてもよく、凍結保存した細胞を用いてもよい。凍結保存していた場合、ハイドロゲル内での培養の前に、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で、HS細胞を培養することが好ましい。この際の培地としては、上記[ハイポブラスト幹細胞の製造方法]における(培養工程)の<培地>の項で説明した培地と同様のものが挙げられる。
 HS細胞は、ハイドロゲル内に埋め込む前に、ピペッティング等により培養容器から剥離してもよい。HS細胞が、細胞塊を形成している場合、ピペッティング等により、細胞塊を分離させて、遊離の細胞としてもよい。細胞の剥離及び/又は細胞塊の分離の際には、トリプシン等のタンパク質酵素を用いてもよい。
 ハイドロゲル内に埋め込むHS細胞の細胞数は、例えば、10~1010個とすることができる。
 ハイドロゲルにおいて、HS細胞を埋め込む位置としては、例えば、ハイドロゲルの表面から深さ0.1~5cm程度、又は0.5~3cm程度が挙げられる。
 HS細胞を埋め込んだハイドロゲルを、適切な液体培地に浸漬して培養することができる。前記培地としては、例えば、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地が挙げられる。前記培地としては、上記[ハイポブラスト幹細胞の製造方法]における(培養工程)の<培地>の項で説明した培地と同様の培地が挙げられる。
 培養条件は、動物細胞の培養に一般的に用いられる条件とすることができる。例えば、培養温度32~40℃(好ましくは35~38℃、典型的には37℃)、CO濃度2~5%(好ましくは5%)とすることができる。
 培養期間は、卵黄嚢オルガノイドが形成されるのに十分な期間であればよい。卵黄嚢オルガノイドの形成は、位相差顕微鏡による観察等により、確認することができる。培養期間としては、例えば、3日以上、5日以上、7日以上、又は10日以上が挙げられる。培養期間の範囲としては、例えば、3~50日程度、5~50日程度、7~50日程度、又は10~50日程度が挙げられる。
 本実施形態の製造方法により得られる卵黄嚢オルガノイドは、上記[ハイポブラスト幹細胞の製造方法]における(ハイポブラスト幹細胞)の項の「(c)について」で説明した卵黄嚢オルガノイドと同じである。
 本実施形態の製造方法により得られる卵黄嚢オルガノイドは、卵黄嚢と同様の遺伝子発現パターンを有し、卵黄嚢と類似の機能を有すると推測される。そのため、前記卵黄嚢オルガノイドは、妊娠初期における薬物評価試験用モデル、再生医療、生殖補助医療等への応用が期待される。
 別の態様において、本開示は、上記第4の態様に係る製造方法により得られる卵黄嚢オルガノイドもまた提供する。前記卵黄嚢オルガノイドは、上記[ハイポブラスト幹細胞の製造方法]における(ハイポブラスト幹細胞)の項で説明したHS細胞を含む。
 以下、実施例により本発明をさらに詳細に説明するが、本発明はこれらの例によって限定されるものではない。
[試薬類、培地]
 実験に使用した試薬類を表1に示す。
 各阻害剤として、表2に示す試薬を用いた。
 培地の調製のために、表3に示す試薬溶液を調製した。
 TS basal培地の組成を表4に示す。TS basal培地は、4℃で保存し、調製後、1カ月以内に使用した。
 YS-2D培地の組成を表5に示す。YS-2D培地は、4℃で保存し、調製後、2週間以内に使用した。
 YS培地の組成を表6に示す。YS培地は、4℃で保存し、調製後、2週間以内に使用した。
[実験例1]
(ハイポブラスト幹細胞の誘導)
<ヒト卵黄嚢からの細胞の単離>
 ヒト卵黄嚢組織をハサミで細断し、TrypLE溶液中で、37℃で10分間処理を行った後、遠心分離によって細胞(卵黄嚢細胞)を回収した。
<ハイポブラスト幹細胞の誘導>
 以下の手順に従い、ヒト卵黄嚢細胞から、ハイポブラスト幹細胞(以下、「HS細胞」ともいう)を誘導した。
(1)12ウェルディッシュの各ウェルに、1mlのYS-2D培地及び1μlのiMatrix511を添加した。
(2)37℃で、少なくとも10分間、12ウェルディッシュを維持した。
(3)各ウェルのYS-2D培地に、約0.5×10個/ウェルで細胞を懸濁した。
(4)37℃、5%CO条件下で培養し、3日毎に、YS-2D培地を用いて、継代した。
 上記のように調製した細胞は、CELLBANKER(登録商標)1(ゼノジェンファーマ)を用いて凍結保存することができた。
 細胞の継代は、以下の手順に従って行った。
(1)ウェル内のYS-2D培地を吸引した。
(2)各ウェルに、0.25mlのPBS(phosphate-buffered saline)及び0.25mlのTrypLEを添加した。
(3)37℃で、約10分間、12ウェルディッシュを維持した。
(4)ピペッティングにより、細胞を剥離した。
(5)0.5mlのTS basal培地を含む新しいチューブに、剥離した細胞を移した。
(6)約380gで、2分間、前記チューブを遠心分離した。
(7)新しい12ウェルディッシュのウェルに、YS-2D培地及びiMatrix511を入れ、37℃に温めた。このウェルに、遠心分離で得られた細胞ペレットを播種した(split ratio=1:6~1:8)。
 図1に、上記の方法により誘導されたHS細胞の位相差顕微鏡画像を示す。図1のHS細胞は、YS-2D培地において、卵黄嚢細胞を約1カ月間継代培養することにより得られたものである。最後の継代から6日目のHS細胞を用いた。HS細胞は、細胞塊を形成しており、卵黄嚢細胞とは、外観が異なっていた。HS細胞の細胞塊において、細胞間の境界は明瞭には観察されなかった。このことから、細胞密度の高い細胞塊であると推測された。HS細胞の外観は、以前に報告されたマウスPrES細胞(Ohinata et al. Science. 2022 Feb 4;375(6580):574-578.)及びハイポブラスト様細胞(Okubo et al. Nature. 2024 Feb;626(7998):357-366)とも異なっていた。
[実験例2]
(卵黄嚢オルガノイドの形成)
 実験例1で得られたHS細胞を用いて、以下の手順により、卵黄嚢オルガノイドを形成した。
(1)ウェル内のYS-2D培地を吸引した。
(2)各ウェルに、0.25mlのPBS(phosphate-buffered saline)及び0.25mlのTrypLEを添加した。
(3)37℃で、約10分間、12ウェルディッシュを維持した。
(4)ピペッティングにより、細胞を剥離した。
(5)0.5mlのTS basal培地を含む新しいチューブに、剥離した細胞を移した。
(6)約380gで、2分間、前記チューブを遠心分離した。
(7)Matrigelに、遠心分離で得られた細胞ペレットを埋め込み、新しい12ウェルディッシュのウェルに入れた。
(8)(7)のMatrigelを、YS培地中で、37℃、5%CO条件下で培養した。
 卵黄嚢オルガノイドの継代は、以下の手順に従って行った。
(1)Matrigelの周囲のYS培地を吸引した。
(2)PBS及びTrypLE(PBS:TrypLE=1:1;1.5ml/35cmディッシュ)を35cmディッシュに添加した。
(3)ピペットマンP1000(ギルソン)を用いて、ディッシュ内の液体を、約5回ピペッティングした。
(4)37℃で、約15分間、ディッシュを維持した。
(5)ピペットマンP200(ギルソン)を用いて、ディッシュ内の液体を、約10回ピペッティングした。
(6)5mlのTS basal培地を含む新しい15mlチューブに、細胞塊を移した。
(7)約800gで、2分間、前記チューブを遠心分離した。
(8)Matrigelに、遠心分離で得られた細胞ペレットを埋め込み(split ratio=1:4~1:8)、新しい12ウェルディッシュのウェルに入れた。これを、YS培地中で、37℃、5%CO条件下で培養した。
 図2に、上記の方法で得られたオルガノイドの位相差顕微鏡画像(左図)と免疫蛍光染色画像(右図)を示す。図2のオルガノイドは、Matrigel内で、HS細胞を約1カ月間継代培養することにより得られたものである。最後の継代から11日目のオルガノイドを用いた。免疫蛍光染色は、抗AFP(α-フェトプロテイン)抗体を用いて行った。AFPは、卵黄嚢で高発現する卵黄嚢マーカーである。HS細胞から誘導された卵黄嚢オルガノイド(以下、「HSオルガノイド」ともいう)は、AFPを高発現していた。
[実験例3]
(RNA-seq解析)
 実験例1で得られたHS細胞及び実験例2で得られたHSオルガノイドについて、RNA-seq解析により、遺伝子発現を調べた。
 図3Aに、RNA-seq解析により得られたAFPの発現量を示す。HS細胞では、AFPはほとんど発現していなかった。一方、HSオルガノイドでは、AFPが発現していた。
 図3Bに、RNA-seq解析により得られたアルブミンの発現量を示す。HS細胞では、アルブミンはほとんど発現していなかった。一方、HSオルガノイドでは、アルブミンが発現していた。
 図4に、RNA-seq解析により得られた遺伝子発現データを、UMAP(Uniform Manifold Approximation and Projection)解析した結果を示す。卵黄嚢の遺伝子発現データとして、Issac Goh et al.(Science. 2023 Aug 18;381(6659))で報告されたデータを利用した。HS細胞及びHSオルガノイドの遺伝子発現パターンは、卵黄嚢内胚葉の遺伝子発現パターンを類似していた。
[実験例4]
(HS細胞の誘導条件の検討)
 YS-2D培地(表5参照)のY27632、EGF、A83-0、CHIR99021、SB02190、及びVPAの組合せに替えて、表7に示す因子の組み合わせを用いたこと以外は、YS-2D培地と同様の組成で各培地を調製した。
 表7に示す因子の組み合わせを含む培地をそれぞれ用いたこと以外は、実験例1と同様の方法により、卵黄嚢細胞からHS細胞の誘導を行った。表7の各培地で5日間培養後、自動セルカウンター(Olympus Cell Counter model R1)を用いて細胞数を計測し、細胞増殖を評価した。その結果を表7に併せて示す。「+」は細胞増殖が確認されたことを示し、「+」の数が多いほど、細胞増殖量が多かったことを示す。「-」は、細胞増殖が確認されなかったことを示す。
 細胞増殖が確認された培地では、図1と同様の形状のHS細胞の細胞塊が観察された。細胞増殖が確認されなかった培地では、HS細胞の細胞塊は観察されなかった。
 6種全ての因子を含むYS-2D培地での細胞増殖が最も良好であった。次いで、YS培地(VPA非含有)及びYS-2D-5培地(SB202190非含有)での細胞増殖が良好であった。YS-2D-3培地(A83-01非含有)では、HS細胞の細胞塊は観察されたが、細胞増殖は低かった。YS-2D-1培地(EGF非含有)、YS-2D-2培地(CHIR99021非含有)及びYS-2D-4培地(Y27632非含有)では、細胞増殖が観察されず、HS細胞の細胞塊も観察されなかった。
 以上の結果から、EGF、GSK3β阻害剤及びROCK阻害剤は、卵黄嚢細胞からHS細胞への誘導に、必須の因子であることが確認された。TGFβ/SMAD阻害剤は、上記3種の因子と組み合わせて用いることにより、HS細胞の誘導を効率的に行うことができると考えられた。さらに、HDAC阻害剤及びp38阻害剤を培地に添加することにより、HS細胞の誘導をより効率的に行うことができると考えられた。
[実験例5]
(HS細胞の長期培養)
 実験例1で得られたHS細胞について、YS-2D培地又はYS培地を用いて、37℃で、6カ月以上の長期培養を行った。HS細胞は、3日毎に、実験例1で記載した方法により継代した。
 6カ月以上の培養期間中、HS細胞は、継代後の培養により、図1と同様の細胞塊を形成した。6カ月以上の培養後のHS細胞を用いて、実験例2と同様の方法により、HSオルガノイドを形成した。その結果、図2と同様のHSオルガノイドを形成することが確認された。
 上記の結果から、HS細胞は、6カ月以上の長期培養による維持が可能であることが確認された。
[実験例6]
(シングルセルRNA-seq解析)
 実験例2と同様の方法によりHSオルガノイドを得た。HS細胞及びHSオルガノイドについて、シングルセルRNA-seq解析を行った。
 図5に、シングルセルRNA-seq解析により得られた遺伝子発現データを、UMAP(Uniform Manifold Approximation and Projection)解析した結果を示す。卵黄嚢の遺伝子発現データとして、Issac Goh et al.(Science. 2023 Aug 18;381(6659))で報告されたデータを利用した。実験例3における結果と同様に、HSオルガノイドの遺伝子発現パターンは、卵黄嚢内胚葉の遺伝子発現パターンを類似していた。
 図6に、シングルセルRNA-seq解析により得られた遺伝子発現データに基づいて、HS細胞とHSオルガノイドとの遺伝子発現を比較した結果を示す。図6における上段(A)は、DNA複製及び細胞分裂等の細胞増殖に関わる遺伝子の発現を示す。図6における下段(B)は、生体分子(脂質、糖質、タンパク質)の取込、代謝、輸送及び硫酸抱合;並びにMAPKシグナル活性等に関わる遺伝子の発現を示す。HSオルガノイドでは、HS細胞と比較して、細胞増殖に関わる遺伝子の発現が減少し、卵黄嚢の機能に関する遺伝子の発現が亢進した。
 本開示によれば、ヒトにも適用可能であり、長期培養により維持可能なハイポブラスト幹細胞を製造可能な、ハイポブラスト幹細胞の製造方法、前記製造方法に使用可能なハイポブラスト幹細胞の誘導用培地、前記製造方法により製造されるハイポブラスト幹細胞、前記ハイポブラスト幹細胞を用いた卵黄嚢オルガノイドの製造方法、及び前記ハイポブラスト幹細胞を含む卵黄嚢オルガノイドを提供することができる。前記ハイポブラスト幹細胞及び卵黄嚢オルガノイドは、妊娠初期を対象とする薬物評価試験、再生医療、生殖補助医療等への応用が期待される。
 以上、本発明の好ましい実施例を説明したが、本発明はこれら実施例に限定されることはない。本発明の趣旨を逸脱しない範囲で、構成の付加、省略、置換、およびその他の変更が可能である。本発明は前述した説明によって限定されることはなく、添付のクレームの範囲によってのみ限定される。

Claims (12)

  1.  哺乳動物の卵黄嚢から単離された細胞を、上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する培地で培養する工程を含む、ハイポブラスト幹細胞の製造方法。
  2.  前記培地が、TGFβ/SMAD阻害剤をさらに含有する、請求項1に記載のハイポブラスト幹細胞の製造方法。
  3.  前記培地が、HDAC阻害剤及びp38阻害剤からなる群より選択される少なくとも1種をさらに含有する、請求項1又は2に記載のハイポブラスト幹細胞の製造方法。
  4.  上皮成長因子、GSK3β阻害剤、及びROCK阻害剤を含有する、
     ハイポブラスト幹細胞の誘導用培地。
  5.  TGFβ/SMAD阻害剤をさらに含有する、請求項4に記載のハイポブラスト幹細胞の誘導用培地。
  6.  HDAC阻害剤及びp38阻害剤からなる群より選択される少なくとも1種をさらに含有する、請求項4に記載のハイポブラスト幹細胞の誘導用培地。
  7.  哺乳動物の卵黄嚢から単離された細胞から、ハイポブラスト幹細胞を誘導するために用いられる、請求項4~6のいずれか一項に記載のハイポブラスト幹細胞の誘導用培地。
  8.  下記(a)~(e)の特徴を有する、ハイポブラスト幹細胞:
     (a)哺乳動物の卵黄嚢から単離された細胞から誘導された細胞である;
     (b)前記哺乳動物の卵黄嚢から単離された細胞と比較して、α-フェトプロテインの発現量が低い;
     (c)α-フェトプロテイン及びアルブミンからなる群より選択される少なくとも1種を発現する細胞を含む卵黄嚢オルガノイドへの分化能を有する;
     (d)増殖能を有する;及び
     (e)細胞塊を形成する。
  9.  請求項4~6のいずれか一項に記載のハイポブラスト幹細胞の誘導用培地で継代培養を行うことにより、6カ月以上の維持培養が可能な、請求項8に記載のハイポブラスト幹細胞。
  10.  ヒトの卵黄嚢から単離された細胞から誘導された細胞である、請求項8に記載のハイポブラスト幹細胞。
  11.  請求項8に記載のハイポブラスト幹細胞を、ハイドロゲル内で培養する工程を含む、卵黄嚢オルガノイドの製造方法。
  12.  請求項8に記載のハイポブラスト幹細胞を含む、卵黄嚢オルガノイド。
