Abstract
本発明は、薬物と、ホスホコリン基を有する薬物送達用ポリマーと、を含み、STARD7に対する解離定数(KD)が1.0×10-6M以下である、薬物-ポリマー複合体を提供する。
Description
本発明は、薬物-ポリマー複合体、医薬組成物、薬物送達用ポリマー、および薬物送達方法に関する。
一般に、経口や静脈内注射によって薬物を全身投与すると、投薬対象の患部だけでなく、正常組織にも薬物が集積する。その結果、薬物投与による副作用が認められ、治療方法の変更や中断が必要になる場合がある。これに対し、副作用の低減を目的として、患部に薬物を選択的に輸送する薬物送達システム(DDS)が開発されている。
薬物の患部への集積量の増大に向けて、患部を指向する標的指向型のDDSの開発が進められている。従来は、細胞取り込み量を増大するための研究が中心であったが、近年、がん、心臓病、糖尿病等の難治性疾患の多くがミトコンドリアと密接な関係があることが明らかになってきていることから、細胞内のミトコンドリアへの薬物送達にも関心が向けられている。
ミトコンドリアへの薬物送達に関しては、これまでに、正電荷を有し、かつ、疎水的な構造体または部位を利用した技術が提案されている。例えば、脂質二重層を有するリポソームによって構成され、表面にオクタアルギニンを含むミトポーターの使用(非特許文献1)、および、トリフェニルホスホニウム(TPP)基またはミトコンドリア膜上の輸送タンパク質によって認識されるシグナルを含むタンパク質またはペプチドの利用が提案されている(例えば、非特許文献2、非特許文献3)。また、比較的極性が高く、双性イオンであるホスホコリン(PC)基の利用も提案されている(特許文献1)。

正電荷を有し、かつ、疎水的な構造体または部位を利用したミトコンドリアターゲティングは、in vivoでの適用において、(i)血液中で負に帯電した血漿タンパク質との凝集体を生じさせる場合があり、当該凝集体は細網内皮系によって体内から排除される、または、毛細血管のつまりを引き起こす可能性がある;(ii)脂質二重層と非特異的に相互作用する結果、種々の細胞の細胞膜に吸着してオフターゲティングが生じる場合がある;(iii)細胞に取り込まれたとしても、mRNA等の種々の負に帯電した細胞内物質と相互作用する結果、細胞内でオフターゲティングが生じる場合がある;(iv)ミトコンドリアに送達されたとしても、界面活性剤のような構造が脂質二重層と混ざり合う結果、ミトコンドリア膜を破壊する場合がある;等の問題があり、実用化が困難である。また、特許文献1に記載のPC基を用いたミトコンドリアターゲティングに関しては、in vivoでの適用に関して十分な検討がなされていない。
本発明は、上記課題を解決するためになされたものであり、その主たる目的は、in vivoでミトコンドリアへ薬物を送達可能な技術、なかでも、がん細胞のミトコンドリアへ選択的に薬物を送達可能な技術を提供することにある。
[1]本発明の1つの局面によれば、薬物と、ホスホコリン基を有する薬物送達用ポリマーと、を含み、STARD7に対する解離定数(KD)が1.0×10-6M以下である、薬物-ポリマー複合体が提供される。
[2]上記[1]に記載の薬物-ポリマー複合体は、ミトコンドリアに上記薬物を送達するためのものであってよい。
[3]上記[1]または[2]に記載の薬物-ポリマー複合体は、平均粒子径が、5.2nmを超えてもよい。

[16]上記[12]から[15]のいずれかに記載の薬物送達用ポリマーは、ポリ(エチレングリコール)、ポリ(サッカライド)、ポリ(ビニルピロリドン)、ポリ(ビニルアルコール)、ポリ(アクリルアミド)、ポリ(アクリル酸)、ポリ(メタクリルアミド)、ポリ(メタクリル酸)、ポリ(メタクリル酸エステル)、ポリ(アクリル酸エステル)、ポリ(アミノ酸)、ポリ(リンゴ酸)、またはこれらの誘導体から構成される親水性ポリマーを含んでよい。
本発明によれば、ホスファチジルコリンをミトコンドリアへ輸送する活性を有するリン脂質輸送タンパク質「STARD7(StAR-related lipid transfer domain protein 7)」と一定以上の結合親和性を有する薬物-ポリマー複合体または薬物送達用ポリマー(DDSポリマー)を用いることにより、ミトコンドリアターゲティング、好ましくはin vivoでのミトコンドリアターゲティングを好適に実施することができる。
以下、本発明の好ましい実施形態(例えば、in vivoでのミトコンドリアターゲティングに好適に適用され得る薬物-ポリマー複合体およびDDSポリマー等)について説明するが、本発明はこれらの実施形態には限定されない。また、文脈上不適切である場合を除き、各実施形態は適宜組み合わせることができる。さらにまた、本明細書中で、数値範囲を表す「~」は、その上限および下限の数値を含む。
A.薬物-ポリマー複合体
本発明の実施形態による薬物-ポリマー複合体は、薬物と、ホスホコリン(PC)基を有する薬物送達用ポリマー(PC基含有DDSポリマー)と、を含む。本発明の実施形態による薬物-ポリマー複合体の平均粒子径は、代表的には5nmを超え、例えば5.2nm超、5.5nm超、または6nm超である。PC基は、下記式(I)で表される双性イオン性基である。
本発明の実施形態による薬物-ポリマー複合体は、薬物と、ホスホコリン(PC)基を有する薬物送達用ポリマー(PC基含有DDSポリマー)と、を含む。本発明の実施形態による薬物-ポリマー複合体の平均粒子径は、代表的には5nmを超え、例えば5.2nm超、5.5nm超、または6nm超である。PC基は、下記式(I)で表される双性イオン性基である。
本発明の実施形態による薬物-ポリマー複合体のSTARD7に対する解離定数(KD)は、例えば1.0×10-6M以下、好ましくは5.0×10-7M以下、より好ましくは3.0×10-7M以下、さらに好ましくは2.0×10-7M以下、さらにより好ましくは5.0×10-8M以下であり、例えば1.0×10-9M以上または1.0×10-8M以上であってよい。解離定数が上記範囲内であれば、STARD7を介した薬物-ポリマー複合体のミトコンドリア内への取り込みが好適に行われ得る。このような効果が得られる理由としては、本発明を何ら制限するものではないが、上記解離定数を有する薬物-ポリマー複合体は、PC基がSTARD7に好適に認識され得る結果、ミトコンドリア内に効率的に取り込まれると推測される。上記解離定数は、表面プラズモン共鳴(SPR)法を用いて測定することができる。STARD7は、代表的には哺乳動物のSTARD7、好ましくはヒトのSTARD7である。
以下、本発明の実施形態による薬物-ポリマー複合体に関して、薬物-ポリマー複合体に含まれる薬物送達用ポリマー(DDSポリマー)の数が1である実施形態(第1の実施形態)と薬物-ポリマー複合体に含まれるDDSポリマーの数が2以上である実施形態(第2の実施形態)とに分けて説明する。なお、上記薬物-ポリマー複合体は、共有結合および/または非共有結合性の相互作用を利用して形成され得、例えば、結合体(コンジュゲート)、会合体、または凝集体であり得る。
A-1.第1の実施形態
第1の実施形態における薬物-ポリマー複合体は、1つのPC基含有DDSポリマーと1つ以上の薬物とを含む。PC基含有DDSポリマーと薬物とは、直接またはリンカーを介して共有結合(配位結合を含む)していてもよく、非共有結合性の相互作用によって複合体化していてもよい。好ましくは、PC基含有DDSポリマーと薬物とは、共有結合している。リンカーとしては、DDS用途で用いられる任意のリンカーを用いることができる。リンカーの主鎖の原子数は、例えば1~20、好ましくは1~15、より好ましくは1~10とすることができる。
第1の実施形態における薬物-ポリマー複合体は、1つのPC基含有DDSポリマーと1つ以上の薬物とを含む。PC基含有DDSポリマーと薬物とは、直接またはリンカーを介して共有結合(配位結合を含む)していてもよく、非共有結合性の相互作用によって複合体化していてもよい。好ましくは、PC基含有DDSポリマーと薬物とは、共有結合している。リンカーとしては、DDS用途で用いられる任意のリンカーを用いることができる。リンカーの主鎖の原子数は、例えば1~20、好ましくは1~15、より好ましくは1~10とすることができる。
1つのPC基含有DDSポリマーと1つ以上の薬物とが結合した構成を有する薬物-ポリマー複合体において、PC基含有DDSポリマーは、2つ以上のポリマー鎖を有する分岐型ポリマーであり得る。具体的には、上記薬物-ポリマー複合体は、2つ以上のポリマー鎖を有する分岐型のPC基含有DDSポリマーと、当該2つ以上のポリマー鎖の少なくとも1つに結合された薬物と、を含み得る。
PC基含有DDSポリマーを構成するポリマーとしては、生体適合性が高い親水性ポリマーが好ましい。例えば、ポリ(エチレングリコール)、ポリ(サッカライド)、ポリ(ビニルピロリドン)、ポリ(ビニルアルコール)、ポリ(アクリルアミド)、ポリ(アクリル酸)、ポリ(メタクリルアミド)、ポリ(メタクリル酸)、ポリ(メタクリル酸エステル)、ポリ(アクリル酸エステル)、ポリ(アミノ酸)、ポリ(リンゴ酸)、またはこれらの誘導体が例示でき、なかでも、ポリ(エチレングリコール)が好ましい。1つの実施形態において、PC基含有DDSポリマーは、生理的pH(pH7.4)において、電荷を有さないポリマーによって構成されている。
分岐型のPC基含有DDSポリマーが有するポリマー鎖の数は、例えば3以上、好ましくは4以上、より好ましくは5以上、さらに好ましくは6~32であり、例えば7以上または8以上であってよく、また例えば24以下、20以下、または16以下であってよい。ポリマー鎖の数が上記範囲内であると、立体障害の抑制とPC基の導入量の確保とを好適に両立することができる。
PC基含有DDSポリマーは、少なくとも1つのPC基を有する。PC基含有DDSポリマーが有するPC基の数(ポリマー1分子当たりのPC基の数)は、薬物-ポリマー複合体が上記STARD7に対する解離定数を満たす限りにおいて制限されず、好ましくは3以上、より好ましくは4以上、さらに好ましくは5以上、さらにより好ましくは6~32であり、例えば7以上または8以上であってよく、また例えば24以下、20以下、または16以下であってよい。1つの実施形態において、PC基含有DDSポリマー自身(薬物と結合される前のPC基含有DDSポリマー)のSTARD7に対する解離定数(KD)は、例えば1.0×10-6M以下、好ましくは5.0×10-7M以下、より好ましくは3.0×10-7M以下、さらに好ましくは2.0×10-7M以下、さらにより好ましくは5.0×10-8M以下であり、例えば1.0×10-9M以上または1.0×10-8M以上であってよい。
PC基含有DDSポリマー中、PC基は、任意の位置に導入されてよく、好ましくはポリマー鎖の末端に導入される。ポリマー鎖の末端に導入されたPC基は、薬物-ポリマー複合体の表面に露出しやすいことから、STARD7に対する親和性が高い薬物-ポリマー複合体が好適に得られ得る。分岐型のPC基含有DDSポリマーにおいては、少なくとも1つのポリマー鎖にPC基が結合されており、2以上のポリマー鎖にPC基が結合されていることが好ましく、2以上のポリマー鎖の末端にPC基が結合されていることがより好ましい。
PC基含有DDSポリマーの分子量(直鎖型、分岐型を問わず、ポリマー全体としてのMw)は、例えば5000~100000、好ましくは10000~50000、より好ましくは12000~50000、さらに好ましくは15000~50000、さらにより好ましくは20000~40000であり、20000~35000または20000~30000であってよい。PC基含有DDSポリマーの分子量分散度(PDI)は、例えば1.1以下であり得る。また、PC基含有DDSポリマーが分岐型である場合、各ポリマー鎖の分子量(Mw)はそれぞれ、代表的には200以上、好ましくは1000以上であり、例えば1500以上または2000以上であってよく、代表的には50000以下、好ましくは5000以下であり、例えば4000以下または3000以下であってよい。各ポリマー鎖の分子量は、互いに同じであってもよく、異なっていてもよい。各ポリマー鎖の分子量が同程度であると、その末端基の露出度も同程度となり得る。分子量(Mw)は、例えば、ゲル濾過浸透クロマトグラフィー(GPC)法で測定することができる。PC基含有DDSポリマーの分子量および/または各ポリマー鎖の分子量が上記範囲内であると、in vivoにおけるミトコンドリアターゲティングがより好適に行われ得る。
PC基含有DDSポリマーのPC基当量(PC基含有DDSポリマーの分子量/PC基の数)は、例えば200~10000、好ましくは1000~5000、より好ましくは1000~4000であり、1500以上または2000以上であってよく、3000以下または2500以下であってよい。PC基当量が上記範囲であると、薬物-ポリマー複合体の表面におけるPC基の露出度が高くなり、上記STARD7に対する解離定数を満たす薬物-ポリマー複合体を好適に得ることができる。
分岐型のPC基含有DDSポリマーは、1つのポリマー鎖の末端にsulfo-cy5(以下、「スルホ-cy5」とも表記され得る)が結合され、その他のポリマー鎖の末端に非荷電性基(例えば、PC基、アセチル基)を有するポリマーとして測定された場合に、好ましくは0以下、より好ましくは-3以下のゼータ電位、例えば-15~-3、-12~-4、または-11~-5のゼータ電位を示すポリマーであり得る。1つのポリマー鎖が負の荷電性末端基(例えば、sulfo-cy5)を有し、他のポリマー鎖が非荷電性末端基を有する場合、荷電性末端基の露出度が高いと上記ゼータ電位は小さくなり、露出度が低いと上記ゼータ電位は大きくなると考えられる。よって、上記ゼータ電位は、分岐型のPC基含有DDSポリマーにおける各ポリマー鎖末端基の露出度に対応し得る。上記ゼータ電位を示すポリマー骨格において、ポリマー鎖末端にPC基が結合された構成を有するPC基含有DDSポリマーは、STARD7に対して十分に小さい解離定数を示し得る。
上記薬物としては、目的に応じて任意の適切な物質を用いることができる。例えば、細胞増殖抑制活性、抗腫瘍活性、免疫調節活性、抗ウイルス活性、抗菌活性、抗炎症活性等の生理活性を有する物質(生理活性物質)が好ましく用いられ得る。薬物の具体例としては、タンパク質またはペプチド(例えば、抗体またはその機能性断片、ホルモン、酵素)、核酸(例えば、プラスミドDNA、mRNA等の高分子核酸、siRNA、miRNA、アンチセンス核酸、アプタマー等の低分子核酸)、その他の生理活性物質(例えば、抗腫瘍剤、シグナル伝達阻害剤、代謝拮抗剤、鎮痛剤、抗炎症剤、抗菌剤)が挙げられる。また、薬物は、蛍光色素、造影剤等の検出試薬であってもよい。薬物は、1種のみを用いてもよく、2種以上を用いてもよい。2つ以上または2種以上の薬物を複合体の状態で用いてもよい。薬物は、PC基含有DDSポリマーの任意の適切な部位に結合され得、例えば、ポリマー鎖の末端またはポリマー鎖の側鎖に結合され得る。
薬物の質量(分子量)は特に制限されない。具体的には、薬物の質量(分子量)は、500未満あるいは500以上であってよい。薬物は、例えば、500未満の質量(分子量)を有する低分子薬物、500以上2000未満の質量(分子量)を有する中分子薬物、または2000以上の質量(分子量)を有する高分子薬物であり得る。1つの実施形態において、上記その他の生理活性物質は、低分子薬物または中分子薬物であり得る。タンパク質、核酸等の質量は、例えば1000Da~150000Da、また例えば3000Da~150000Daであり得る。
PC基含有DDSポリマーに結合される薬物の数(薬物-ポリマー複合体1つあたりの薬物の数)は、1以上であり、例えば2以上または3以上であってよい。上記薬物の数の上限は、上記STARD7に対する解離定数が維持される限りにおいて制限されず、例えば10000以下または100以下であり得る。異なる種類の薬物を組み合わせて用いてもよい。例えば、抗体と蛍光色素とが結合された薬物-ポリマー複合体は、蛍光色素の蛍光を検出することで抗体の送達効率等を調べることができる。
上記薬物-ポリマー複合体の平均粒子径は、例えば5.2nm超であり、好ましくは6nm超である。上記平均粒子径が例えば5nm以下、また例えば5.2nm以下または6nm以下であると、腎臓から尿中に排泄されやすい傾向がある。上記平均粒子径は、上記STARD7に対する解離定数を満たす限りにおいて制限されない。上記平均粒子径は、代表的には100nm以下であり、好ましくは30nm以下であり、例えば20nm以下または10nm以下であってよい。平均粒子径は、動的光散乱(DLS)測定装置を用いて測定することができる。
上記薬物-ポリマー複合体は、任意の適切な方法で製造することができる。例えば、薬物-ポリマー複合体は、末端に官能基Aが導入されたポリマー鎖を有する分岐型ポリマーと、官能基Aと反応可能な官能基およびPC基を有するPC基含有化合物と、を反応させて、PC基含有DDSポリマーを得ること、および、PC基含有DDSポリマーの未反応の官能基Aと、薬物が有する官能基と、を反応させること、を含む方法;末端に官能基Aが導入されたポリマー鎖を有する分岐型ポリマーと、官能基Aと反応可能な官能基を有する薬物と、を反応させて薬物結合ポリマーを得ること、薬物結合ポリマーの未反応の官能基Aと、官能基Aと反応可能な官能基およびPC基を有するPC基含有化合物と、を反応させること、を含む方法;末端に官能基Aが導入されたポリマー鎖と官能基Bが導入されたポリマー鎖とを有する分岐型ポリマーと、官能基Aと反応可能な官能基を有する薬物および官能基Bと反応可能な官能基およびPC基を有するPC基含有化合物と、を反応させること、を含む方法等によって得ることができる。PC基含有ポリマーとの反応に用いられる薬物の官能基は、薬物に固有の官能基であってもよく、クロスリンカー等を用いて薬物に導入された官能基であってもよい。
官能基の組み合わせの具体例としては、アジド基とアルキン、チオール基と(メタ)アクリロイル基、チオール基とマレイミド基、チオール基とチオール基、チオール基とカルボキシル基、(メタ)アクリロイル基とヒドロキシル基、(メタ)アクリロイル基とアミノ基、カルボキシル基とアミノ基、カルボキシル基とヒドロキシル基、アミノ基とヒドロキシル基、アミノ基とNHSエステル基、アミノ基とアルデヒド基等が挙げられる。また、種々の官能基(例えば、アミノ基、マレイミド、NHS、チオール、アクリロイルオキシ、カルボキシル)が導入された分岐型PEGおよびクロスリンカーが市販されている。また、PC基含有化合物の具体例としては、6-(O-ホスホリルコリン)ヒドロキシヘキサン酸、2-(メタ)アクリロイルオキシエチルホスホリルコリン、3-(メタ)アクリロイルオキシプロピルホスホリルコリン、4-(メタ)アクリロイルオキシブチルホスホリルコリン、6-(メタ)アクリロイルオキシヘキシルホスホリルコリン、10-(メタ)アクリロイルオキシデシルホスホリルコリン、ω-(メタ)アクリロイル(ポリ)オキシエチレンホスホリルコリン、2-(メタ)アクリルアミドエチルホスホリルコリン、3-(メタ)アクリルアミドプロピルホスホリルコリン、4-(メタ)アクリルアミドブチルホスホリルコリン、6-(メタ)アクリルアミドヘキシルホスホリルコリン、10-(メタ)アクリルアミドデシルホスホリルコリン、ω-(メタ)アクリルアミド(ポリ)オキシエチレンホスホリルコリン等が挙げられる。
A-2.第2の実施形態
第2の実施形態における薬物-ポリマー複合体は、PC基含有DDSポリマーを含む2つ以上のDDSポリマーと1つ以上の薬物とを含む。第2の実施形態における薬物-ポリマー複合体としては、上記2つ以上のDDSポリマーが薬物または担体に共有結合されている構成(構成1)および上記2つ以上のDDSポリマーが非共有結合性の相互作用によって会合している構成(構成2)が挙げられる。構成1において、DDSポリマーと薬物または担体との結合は、直接またはリンカーを介して行われ得る。
2つ以上のDDSポリマーを含む薬物-ポリマー複合体においても、上記STARD7に対する解離定数を満たすものであれば、ミトコンドリア内への薬物送達、なかでも、in vivoでのミトコンドリア内への薬物送達が好適に行われ得る。
2つ以上のDDSポリマーは、PC基含有DDSポリマーに加えて、PC基を有さないDDSポリマー(PC基非含有DDSポリマー)を含み得る。第2の実施形態における薬物-ポリマー複合体において、DDSポリマーは、薬物の可溶化、徐放化、血中滞留性の向上、酵素分解の回避等の効果も発揮し得る。
構成1の薬物-ポリマー複合体としては、例えば、1つの薬物にPC基含有DDSポリマーを含む2つ以上のDDSポリマーが結合した構成(構成1a)と、1つの担体または薬物にPC基含有DDSポリマーおよび薬物含有ポリマーが結合した構成(構成1b)と、が挙げられる。
第2の実施形態における薬物-ポリマー複合体は、PC基含有DDSポリマーを含む2つ以上のDDSポリマーと1つ以上の薬物とを含む。第2の実施形態における薬物-ポリマー複合体としては、上記2つ以上のDDSポリマーが薬物または担体に共有結合されている構成(構成1)および上記2つ以上のDDSポリマーが非共有結合性の相互作用によって会合している構成(構成2)が挙げられる。構成1において、DDSポリマーと薬物または担体との結合は、直接またはリンカーを介して行われ得る。
2つ以上のDDSポリマーを含む薬物-ポリマー複合体においても、上記STARD7に対する解離定数を満たすものであれば、ミトコンドリア内への薬物送達、なかでも、in vivoでのミトコンドリア内への薬物送達が好適に行われ得る。
2つ以上のDDSポリマーは、PC基含有DDSポリマーに加えて、PC基を有さないDDSポリマー(PC基非含有DDSポリマー)を含み得る。第2の実施形態における薬物-ポリマー複合体において、DDSポリマーは、薬物の可溶化、徐放化、血中滞留性の向上、酵素分解の回避等の効果も発揮し得る。
構成1の薬物-ポリマー複合体としては、例えば、1つの薬物にPC基含有DDSポリマーを含む2つ以上のDDSポリマーが結合した構成(構成1a)と、1つの担体または薬物にPC基含有DDSポリマーおよび薬物含有ポリマーが結合した構成(構成1b)と、が挙げられる。
A-2-1.構成1a
構成1aの薬物-ポリマー複合体において、PC基含有DDSポリマーを構成するポリマーとしては、A-1項で記載した親水性ポリマーに加えて、ポリ(プロピレングリコール)、ポリ(エチレングリコール)-ポリ(プロピレングリコール)、アルブミン等の可溶性タンパク質、多糖等が好ましく例示される。なかでも、ポリ(エチレングリコール)が好ましい。
構成1aの薬物-ポリマー複合体において、PC基含有DDSポリマーを構成するポリマーとしては、A-1項で記載した親水性ポリマーに加えて、ポリ(プロピレングリコール)、ポリ(エチレングリコール)-ポリ(プロピレングリコール)、アルブミン等の可溶性タンパク質、多糖等が好ましく例示される。なかでも、ポリ(エチレングリコール)が好ましい。
PC基含有DDSポリマーは、直鎖状であってもよく、分岐鎖状であってもよい。PC基含有DDSポリマーの分子量(分岐鎖状である場合は、各鎖のMw)は、例えば200以上、好ましくは1000以上であり、例えば1500以上または2000以上であってよく、代表的には50000以下、好ましくは5000以下であり、例えば4000以下または3000以下であってよい。PC基含有DDSポリマーは、1種のみ用いてもよく、2種以上を併用してもよい。2種以上を併用する場合、各PC基含有DDSポリマーの分子量は、互いに同じであってもよく、異なっていてもよい。また、PC基含有DDSポリマーが分岐鎖状の場合、各ポリマー鎖の分子量は、互いに同じであってもよく、異なっていてもよい。PC基含有DDSポリマーの分子量が上記範囲内であると、PC基の露出と薬物の被覆とが好適に両立され得る。
PC基含有DDSポリマーにおいて、PC基は、上記ポリマーの任意の適切な部位に結合され得、好ましくは上記ポリマーの末端に結合される。PC基が導入された末端と官能基が導入された末端とを有するPC基含有DDSポリマーを、当該官能基を用いて薬物と結合させることで、薬物の表面を上記ポリマーで層状に覆うことができ、さらに、上記ポリマー層の表面にPC基を露出させることができる。PC基含有DDSポリマーが有するPC基の数は、1以上であり、例えば1~8であり得る。
上記PC基非含有DDSポリマーについては、PC基を有さないこと以外はPC基含有DDSポリマーと同様の説明を適用することができる。PC基含有DDSポリマーとPC基非含有DDSポリマーとは、PC基の有無を除いて実質的に同じポリマーであってもよく、異なるポリマー(例えば、分子量、構成単位の種類または比率等が異なるポリマー)であってもよい。
1つの薬物に結合するPC基含有DDSポリマーの数は、薬物-ポリマー複合体が上記STARD7に対する解離定数を示す限りにおいて制限されない。1つの実施形態において、PC基含有DDSポリマーは、薬物-ポリマー複合体に含まれるPC基の数が、例えば2以上、好ましくは3以上、より好ましくは5以上となるように、薬物に結合され得る。1つの薬物に結合するPC基含有DDSポリマーの数は、例えば2以上、また例えば7以上であってよく、例えば5×107以下、5×104以下、または5×102以下であり得る。
1つの薬物に結合する各ポリマー数の比(PC基含有DDSポリマー:PC基非含有DDSポリマー)は、例えば100:0~1:99、好ましくは10:90~50:50、より好ましくは20:80~30:70であってよい。
薬物としては、上述と同様のものを用いることができ、タンパク質、核酸、多糖等の比較的サイズが大きいものが好ましく用いられる。薬物が、タンパク質、核酸、多糖等の生体高分子である場合、その質量は、例えば1000Da~150000Da、また例えば3000Da~150000Daであり得る。
上記薬物-ポリマー複合体の平均粒子径は、例えば5.2nm超であり、好ましくは6nm超である。上記平均粒子径が例えば5nm以下、また例えば5.2nm以下または6nm以下であると、腎臓から尿中に排泄されやすい傾向がある。上記平均粒子径は、上記STARD7に対する解離定数を満たす限りにおいて制限されない。上記平均粒子径は、好ましくは200nm以下であり、例えば150nm以下または100nm以下であってよい。平均粒子径は、動的光散乱(DLS)測定装置を用いて測定することができる。
上記薬物-ポリマー複合体は、任意の適切な方法で製造することができる。例えば、薬物-ポリマー複合体は、一方の端部に官能基Aが導入され、他方の端部に官能基Bが導入されたポリマーと、官能基Aと反応可能な官能基およびPC基を有するPC基含有化合物と、を反応させて、PC基含有DDSポリマーを得ること、および、PC基含有DDSポリマーの官能基Bと、官能基Bと反応可能な官能基を有する薬物と、を反応させること、を含む方法によって得ることができる。PC基含有DDSポリマーおよびPC基非含有DDSポリマーを薬物と反応させることもできる。
A-2-2.構成1b
構成1bの薬物-ポリマー複合体において、PC基含有DDSポリマーとしては、A-2-1項に記載のものと同様のものを用いることができる。薬物含有DDSポリマーとしては、PC基含有DDSポリマーにおいて、PC基の代わりに薬物を有するポリマーを例示することができる。また、PC基含有DDSポリマーまたは薬物含有DDSポリマーの代わりに、あるいは、これらに加えて、PC基と薬物とを有するPC基-薬物含有DDSポリマーを用いてもよい。PC基-薬物含有DDSポリマーは、例えば、側鎖に薬物が導入され、一方の末端にPC基、他方の末端に官能基を有し、当該官能基による反応を介して担体または薬物と結合され得る。構成1bの薬物-ポリマー複合体においては、目的に応じて、上記DDSポリマーに加えて、PC基も薬物も有さないPC基-薬物非含有DDSポリマーを担体または薬物と結合させてもよい。

DDSポリマーを結合させる担体または薬物としては、金等の金属ナノ粒子、酸化ガドリニウム等の金属酸化物ナノ粒子等が好ましく例示できる。担体の長径は、例えば5nm~40nm、また例えば5nm~15nmであり得る。これらのナノ粒子は、球状であってもよく、ロッド状であってもよい。また、クエン酸緩衝液等の緩衝液に分散されていてもよい。金属ナノ粒子は、造影剤としても機能し得ることから、当該用途においては、金属ナノ粒子は薬物であり得る。
1つの担体または薬物に結合するPC基含有DDSポリマーの数は、薬物-ポリマー複合体が上記STARD7に対する解離定数を示す限りにおいて制限されない。1つの担体または薬物に結合するPC基含有DDSポリマーの数は、例えば2以上、また例えば7以上であってよく、例えば5×107以下、5×104以下、または5×102以下であり得る。
1つの担体または薬物に結合するPC基含有DDSポリマーの数とPC基-薬物非含有DDSポリマーの数との比(前者:後者)は、例えば100:0~1:99、好ましくは10:90~50:50、より好ましくは20:80~30:70であってよい。
1つの担体または薬物に結合する薬物含有ポリマーの数は、薬物含有ポリマーに結合している薬物の数、種類等に応じて適宜設定され得る。1つの担体または薬物に薬物含有ポリマーを介して担持される薬物の数は、例えば1~100、また例えば5~30であり得る。
薬物含有ポリマーが有する薬物としては、A-1項に記載される薬物と同様のものを用いることができる。好ましくは比較的サイズが小さいものが好ましく用いられ、その質量は、例えば1000Da以下であり得る。
上記薬物-ポリマー複合体の平均粒子径は、例えば5.2nm超であり、好ましくは6nm超である。上記平均粒子径が例えば5nm以下、また例えば5.2nm以下または6nm以下であると、腎臓から尿中に排泄されやすい傾向がある。上記平均粒子径は、上記STARD7に対する解離定数を満たす限りにおいて制限されない。上記平均粒子径は、好ましくは200nm以下であり、例えば150nm以下または100nm以下であってよい。
上記薬物-ポリマー複合体は、任意の適切な方法で製造することができる。例えば、上記薬物-ポリマー複合体は、一方の末端に官能基を有する各DDSポリマーを準備し、当該官能基を担体または薬物が有する官能基と反応させること、当該官能基を用いて各DDSポリマーを担体または薬物表面に化学吸着させること等によって得られ得る。
A-2-3.構成2
構成2の薬物-ポリマー複合体は、非共有結合性の相互作用によって会合した2分子以上のDDSポリマー(好ましくはPC基含有DDSポリマー)と薬物とを含む会合体である。上記会合体としては、例えば、ミセル、ベシクル等のナノ粒子またはポリイオンコンプレックス(PIC)が挙げられる。薬物は、DDSポリマーに結合されていてもよく、結合されることなく、会合体(例えば、ナノ粒子)に内包されていてもよい。
構成2の薬物-ポリマー複合体は、非共有結合性の相互作用によって会合した2分子以上のDDSポリマー(好ましくはPC基含有DDSポリマー)と薬物とを含む会合体である。上記会合体としては、例えば、ミセル、ベシクル等のナノ粒子またはポリイオンコンプレックス(PIC)が挙げられる。薬物は、DDSポリマーに結合されていてもよく、結合されることなく、会合体(例えば、ナノ粒子)に内包されていてもよい。
上記薬物-ポリマー複合体の平均粒子径は、例えば400nm以下、好ましくは200nm以下、より好ましくは150nm以下、さらに好ましくは100nm以下、さらにより好ましくは80nm以下であり、例えば20nm以上または30nm以上である。平均粒子径は、動的光散乱(DLS)測定装置を用いて測定することができる。
上記薬物-ポリマー複合体としては、WO2020/116640の[0074]~[0080]に記載される会合体が例示できる。具体的には、電荷を有する薬物とPC基および上記薬物と反対の電荷を有する荷電性ポリマーとが静電相互作用によって会合して形成されるPIC;末端にPC基を有する親水性ポリマーセグメントと疎水性ポリマーセグメントとを含むPC基含有ブロックコポリマーによって形成され、薬物を内包するポリマーミセル;ポリグリコール酸(PGA)、ポリ乳酸(PLA)、およびそれらの共重合体(PLGA)、ポリε-カプロラクトン、キトサン等のPC基含有生体適合性ポリマーによって形成され、薬物を担持する微粒子;等が例示できる。これらの薬物-ポリマー複合体においても、上記STARD7に対する解離定数を満たすことにより、in vivoでのミトコンドリア内への薬物送達が好適に行われ得る。例えば、会合体を構成するDDSポリマーにおけるPC基含有DDSポリマーの割合を増加させることにより、会合体のSTARD7との親和性を向上させ得る。
B.医薬組成物
本実施形態による医薬組成物は、A項に記載の薬物-ポリマー複合体を含む。医薬組成物は、目的に応じて、薬物-ポリマー複合体中の薬物以外の薬物をさらに含んでいてもよい。また、必要に応じて、任意の適切な添加剤を含むことができる。添加剤は、薬学的に許容され得るものであればよく、例えば賦形剤、希釈剤、溶解補助剤、懸濁化剤、等張化剤、pH調整剤、緩衝剤、安定化剤等を例示できる。
本実施形態による医薬組成物は、A項に記載の薬物-ポリマー複合体を含む。医薬組成物は、目的に応じて、薬物-ポリマー複合体中の薬物以外の薬物をさらに含んでいてもよい。また、必要に応じて、任意の適切な添加剤を含むことができる。添加剤は、薬学的に許容され得るものであればよく、例えば賦形剤、希釈剤、溶解補助剤、懸濁化剤、等張化剤、pH調整剤、緩衝剤、安定化剤等を例示できる。
医薬組成物は、任意の適切な剤形に調製される。医薬組成物は、代表的には、非経口剤であり、皮下注射剤、静脈内注射剤、筋肉内注射剤、腹腔内注射剤等の注射剤、輸液剤、点眼剤、点鼻剤等であり得る。
C.ミトコンドリアに薬物を送達する方法
本発明の別の局面によれば、A項に記載の薬物-ポリマー複合体またはB項に記載の医薬組成物を処置対象の個体に投与することを含む、in vivoでミトコンドリアに薬物を送達する方法が提供される。

A項に記載の薬物-ポリマー複合体またはB項に記載の医薬組成物によって処置され得る疾患としては、ミトコンドリアが関与する疾患が挙げられる。ミトコンドリアが関与する疾患としては、ミトコンドリア病、がん、糖尿病、アルツハイマー病、パーキンソン病、神経変性疾患、心血管疾患、代謝性疾患等が挙げられる。1つの実施形態において、上記薬物-ポリマー複合体および医薬組成物は、がん細胞のミトコンドリアへの薬物の送達に用いられ得る。
処置対象の個体は、代表的には、ヒトまたはヒト以外の哺乳類である。ヒト以外の哺乳類としては、マウス、ラット、ハムスター、ウサギ、ヤギ、イヌ、ネコ、サル、チンパンジー、ウシ、ウマ、ブタ等が挙げられる。
個体への投与は、全身投与であってもよく、局所投与であってもよい。全身投与としては、静脈内注射、皮下注射、筋肉内注射等の注射または点滴が挙げられる。局所投与としては、皮膚、粘膜、鼻、眼等への投与が挙げられる。
投与量は、処置対象となる疾患、症状、処置対象の個体、投与方法等に応じて任意の適切な量に設定され得る。
以下、実施例を挙げて本発明を具体的に説明するが、本発明はこれらの実施例に限定されるものではない。以下の実施例において、統計解析は、2グループ間の比較に対して、two-tailed t testで行い、3グループ以上の比較に対して、analysis of variance (ANOVA) with the Tukey post-hoc testで行った。
[実験例1:薬物-ポリマー複合体の作製]
(1)PC結合薬物-ポリマー複合体
下記に示す構造を有し、3つの異なる分子量(Mw=10k、20k、または40k)を有する8分岐型ポリエチレングリコール(以下、「8-PEG10-NH2」、「8-PEG20-NH2」、「8-PEG40-NH2」)をCreative PEGWorksから購入した。これらのPDIは、1.02~1.06であった。8-PEG10-NH2、8-PEG20-NH2、および8-PEG40-NH2における各PEG鎖の分子量(Mw)はそれぞれ、約1250、約2500、および約5000であり、各PEG鎖の末端にはアミノ基が導入されている。
Sulfo-cyanine 5 succinimidyl ester(以下、「スルホ-Cy5-NHS」)をLumiprobeから購入した。
8-PEG10-NH2、8-PEG20-NH2、または8-PEG40-NH2をN,N-ジメチルホルムアミド(DMF)中のスルホ-Cy5-NHS(アミンに対して1当量)と混合し、室温で24時間撹拌した。得られたスルホ-Cy5標識ポリマーをメタノールおよび純水に対して交互に各3回透析することによって精製した(MWCO:3,500)。生成物を凍結乾燥により回収した。ポリマーに対するスルホ-Cy5の結合を蛍光検出および1H-NMR(アセトニトリル-d3;25℃)によって確認した。スルホ-Cy5標識ポリマーを水に溶解し(10mg/ml)、6-(O-phosphorylcholine)hydroxyhexanoic acid(富士フイルム和光純薬社)(アミンに対して10当量)および4-(4,6-Dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride n-hydrate(以下、「DMT-MM」、富士フイルム和光純薬社)(アミンに対して10当量)と、水中、室温で24時間混合した。追加のDMT-MM(アミンに対して2当量)を反応の間6時間毎に投入した。生成物を、Sephadex G-25を含む使い捨てPD-10脱塩カラム(Cytiva)を通過させることで精製し、凍結乾燥により回収した。これにより、薬物としてスルホ-Cy5が結合されたPC結合薬物-ポリマー複合体(PC-8-PEG10、PC-8-PEG20、PC-8-PEG40)を得た。
ポリマーへのPCの結合を、D2O(0.05%V/V TMS)中の1H-NMRによって調べたところ、PCのポリマーへの結合率は、72%~99%であった。

(1)PC結合薬物-ポリマー複合体

8-PEG10-NH2、8-PEG20-NH2、または8-PEG40-NH2をN,N-ジメチルホルムアミド(DMF)中のスルホ-Cy5-NHS(アミンに対して1当量)と混合し、室温で24時間撹拌した。得られたスルホ-Cy5標識ポリマーをメタノールおよび純水に対して交互に各3回透析することによって精製した(MWCO:3,500)。生成物を凍結乾燥により回収した。ポリマーに対するスルホ-Cy5の結合を蛍光検出および1H-NMR(アセトニトリル-d3;25℃)によって確認した。スルホ-Cy5標識ポリマーを水に溶解し(10mg/ml)、6-(O-phosphorylcholine)hydroxyhexanoic acid(富士フイルム和光純薬社)(アミンに対して10当量)および4-(4,6-Dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride n-hydrate(以下、「DMT-MM」、富士フイルム和光純薬社)(アミンに対して10当量)と、水中、室温で24時間混合した。追加のDMT-MM(アミンに対して2当量)を反応の間6時間毎に投入した。生成物を、Sephadex G-25を含む使い捨てPD-10脱塩カラム(Cytiva)を通過させることで精製し、凍結乾燥により回収した。これにより、薬物としてスルホ-Cy5が結合されたPC結合薬物-ポリマー複合体(PC-8-PEG10、PC-8-PEG20、PC-8-PEG40)を得た。
ポリマーへのPCの結合を、D2O(0.05%V/V TMS)中の1H-NMRによって調べたところ、PCのポリマーへの結合率は、72%~99%であった。

(2)アセチル化薬物-ポリマー複合体
上記と同様にして、スルホ-Cy5で標識した8-PEG10-NH2、8-PEG20-NH2、および8-PEG40-NH2を得た。これらのポリマーを超脱水アセトニトリルに溶解し、アルゴン雰囲気下でトリエチルアミン(以下、「TEA」)(アミンに対して100当量)を加えた。室温で30分撹拌後、酢酸無水物を超脱水アセトニトリルに溶解し、ポリマー溶液に滴下した。混合物を、アルゴン雰囲気下、室温で24時間撹拌した。アセトニトリルを蒸発により除去した。生成物(アセチル化ポリマー(Ac-8-PEG))を純水に再溶解し、PC結合薬物-ポリマー複合体と同様の方法で精製した。これにより、ネガティブコントロールとしてのアセチル化薬物-ポリマー複合体(Ac-8-PEG10、Ac-8-PEG20、Ac-8-PEG40)を得た。
アセチル化の程度を、アセトニトリル-d3(0.05%V/V TMS)中の1H-NMRによって調べたところ、アミンの72%~85%のアセチル化が確認された。
1H-NMR(C2D3N)δ=1.80-1.85ppm(3H:-CO-CH3)
上記と同様にして、スルホ-Cy5で標識した8-PEG10-NH2、8-PEG20-NH2、および8-PEG40-NH2を得た。これらのポリマーを超脱水アセトニトリルに溶解し、アルゴン雰囲気下でトリエチルアミン(以下、「TEA」)(アミンに対して100当量)を加えた。室温で30分撹拌後、酢酸無水物を超脱水アセトニトリルに溶解し、ポリマー溶液に滴下した。混合物を、アルゴン雰囲気下、室温で24時間撹拌した。アセトニトリルを蒸発により除去した。生成物(アセチル化ポリマー(Ac-8-PEG))を純水に再溶解し、PC結合薬物-ポリマー複合体と同様の方法で精製した。これにより、ネガティブコントロールとしてのアセチル化薬物-ポリマー複合体(Ac-8-PEG10、Ac-8-PEG20、Ac-8-PEG40)を得た。
アセチル化の程度を、アセトニトリル-d3(0.05%V/V TMS)中の1H-NMRによって調べたところ、アミンの72%~85%のアセチル化が確認された。

(3)TPP結合薬物-ポリマー複合体
上記と同様にして、スルホ-Cy5で標識した8-PEG10-NH2、8-PEG20-NH2、および8-PEG40-NH2を得た。これらのポリマーを水に溶解し(10mg/ml)、(3-Carboxybutyl)triphenylphosphonium Bromide(富士フイルム和光純薬社)(アミンに対して10当量)およびDMT-MM(アミンに対して50当量)と、水中、室温で24時間混合した。追加のDMT-MM(アミンに対して2当量)を反応の間6時間毎に添加した。生成物を、Sephadex G-25を含む使い捨てPD-10脱塩カラム(Cytiva)を通過させることで精製し、凍結乾燥により回収した。これにより、ポジティブコントロールとしてのTPP結合薬物-ポリマー複合体(TPP-8-PEG10、TPP-8-PEG20、TPP-8-PEG40)を得た。
ポリマーへのTPPの結合を、アセトニトリル-d3(0.05%V/V TMS)中の1H-NMRによって調べたところ、TPPのポリマーへの結合率は、86%~87%であった。
1H-NMR(アセトニトリル-d3)δ=6.0-9.0ppm(15H:(C6H5)3-P-)
上記と同様にして、スルホ-Cy5で標識した8-PEG10-NH2、8-PEG20-NH2、および8-PEG40-NH2を得た。これらのポリマーを水に溶解し(10mg/ml)、(3-Carboxybutyl)triphenylphosphonium Bromide(富士フイルム和光純薬社)(アミンに対して10当量)およびDMT-MM(アミンに対して50当量)と、水中、室温で24時間混合した。追加のDMT-MM(アミンに対して2当量)を反応の間6時間毎に添加した。生成物を、Sephadex G-25を含む使い捨てPD-10脱塩カラム(Cytiva)を通過させることで精製し、凍結乾燥により回収した。これにより、ポジティブコントロールとしてのTPP結合薬物-ポリマー複合体(TPP-8-PEG10、TPP-8-PEG20、TPP-8-PEG40)を得た。
ポリマーへのTPPの結合を、アセトニトリル-d3(0.05%V/V TMS)中の1H-NMRによって調べたところ、TPPのポリマーへの結合率は、86%~87%であった。
1H-NMR(アセトニトリル-d3)δ=6.0-9.0ppm(15H:(C6H5)3-P-)
上記(1)~(3)で得られた薬物-ポリマー複合体のイメージ図を図1に示す。
<粒子径およびゼータ電位の測定>
上記(1)~(3)で得られた薬物-ポリマー複合体の流体力学的直径および多分散度指数(PDI)を動的光散乱法(DLS)で測定した。また、薬物-ポリマー複合体の表面ゼータ電位を電気泳動光散乱法(ELS)で測定した。具体的には、各薬物-ポリマー複合体を脱イオン水に溶解したものを測定サンプル(4mg/ml)として、室温で、ダイオードレーザー(λ=532nm)を搭載したZatasizer Nano ZS90(Malvern Instruments Ltd.)を用いて、検出角度173°で測定を行った。結果を、各薬物-ポリマー複合体におけるリガンド(PC基、TPP基、またはAc基)の結合数およびスルホ-Cy5の結合数とともに表1に示す。なお、リガンドおよびスルホ-Cy5の結合数は、NMRデータから算出した推定結合数である。ポリマーの長さ、リガンドの種類等によってポリマーと溶媒との親和性が変化することから、当該推定結合数は実際の値から多少ずれる場合があるが、リガンドの推定結合数とスルホ-Cy5の推定結合数との合計が7以上である場合は、末端アミノ基の全てが反応したと推測できる。
上記(1)~(3)で得られた薬物-ポリマー複合体の流体力学的直径および多分散度指数(PDI)を動的光散乱法(DLS)で測定した。また、薬物-ポリマー複合体の表面ゼータ電位を電気泳動光散乱法(ELS)で測定した。具体的には、各薬物-ポリマー複合体を脱イオン水に溶解したものを測定サンプル(4mg/ml)として、室温で、ダイオードレーザー(λ=532nm)を搭載したZatasizer Nano ZS90(Malvern Instruments Ltd.)を用いて、検出角度173°で測定を行った。結果を、各薬物-ポリマー複合体におけるリガンド(PC基、TPP基、またはAc基)の結合数およびスルホ-Cy5の結合数とともに表1に示す。なお、リガンドおよびスルホ-Cy5の結合数は、NMRデータから算出した推定結合数である。ポリマーの長さ、リガンドの種類等によってポリマーと溶媒との親和性が変化することから、当該推定結合数は実際の値から多少ずれる場合があるが、リガンドの推定結合数とスルホ-Cy5の推定結合数との合計が7以上である場合は、末端アミノ基の全てが反応したと推測できる。
<STARD7に対する解離定数(KD)測定>
表面プラズモン共鳴(SPR)測定は、BIACORE T200(Cytiva)を用いて行った。
まず、EDCカップリング法を用いて、抗GST抗体をCM5センサーチップ表面のカルボキシル基に結合させた。具体的には、以下のとおりである。新たに調製した脱イオン水中の0.2M N-ethyl-N’-(3-demethylaminopropyl carbodiimide)と0.05M N-hydroxysuccineimideとの混合液を注入し、ゲル上の表面のカルボキシル基を化学的に活性化した。続いて、10mM酢酸ナトリウム(pH5.0)中の30μg/mL抗GST抗体溶液98μLを注入し、PBS(-)を移動相として流動させ、デキストランゲルの活性化表面に420秒間固定化した。最後に、128μLの1Mエタノールアミン/HCl(pH8.5)の注入により、デキストランゲル表面の未反応カルボキシル基を確実に失活させた。
次に、STARD7タンパク質を、GSTタグを媒介して抗GST抗体に固定化した。具体的には、以下のとおりである。センサーチップへの抗GST抗体の固定化後、移動相を150mM NaClおよび5mM CaCl2を含む50mM HEPES(RB;ランニングバッファー)に交換した。次に、RB中の2μg/mLのGSTタグ付きSTARD7(LifeSpan BioSciences社、「Human STARD7 Protein (Recombinant GST) (aa61-307)-LS-G22208」、Lot# LS-G22208/198058)溶液を、応答ユニットが200RUに達するまで注入した。STARD7溶液の代わりにRBを注入したものを参照チャネルとして抗体結合を行った。
次に、RB中の薬物-ポリマー複合体溶液を、一連の異なる濃度(48、24、12、6、および3μM)で、接触時間120秒、流速30μL/分で注入してSPR測定を行った。得られた応答曲線を、Biacore評価ソフトウェア(Cytiva)を用いて処理して、解離定数を決定した。結果を表2に示す。
なお、上記測定で用いたSTARD7のアミノ酸配列は、LifeSpan BioSciences社のHP(https://www.lsbio.com/proteins/human-stard7-protein-recombinant-gst-aa61-307-ls-g22208/22208)に開示されており、配列番号1として示されるとおりである。

次に、STARD7タンパク質を、GSTタグを媒介して抗GST抗体に固定化した。具体的には、以下のとおりである。センサーチップへの抗GST抗体の固定化後、移動相を150mM NaClおよび5mM CaCl2を含む50mM HEPES(RB;ランニングバッファー)に交換した。次に、RB中の2μg/mLのGSTタグ付きSTARD7(LifeSpan BioSciences社、「Human STARD7 Protein (Recombinant GST) (aa61-307)-LS-G22208」、Lot# LS-G22208/198058)溶液を、応答ユニットが200RUに達するまで注入した。STARD7溶液の代わりにRBを注入したものを参照チャネルとして抗体結合を行った。
次に、RB中の薬物-ポリマー複合体溶液を、一連の異なる濃度(48、24、12、6、および3μM)で、接触時間120秒、流速30μL/分で注入してSPR測定を行った。得られた応答曲線を、Biacore評価ソフトウェア(Cytiva)を用いて処理して、解離定数を決定した。結果を表2に示す。
なお、上記測定で用いたSTARD7のアミノ酸配列は、LifeSpan BioSciences社のHP(https://www.lsbio.com/proteins/human-stard7-protein-recombinant-gst-aa61-307-ls-g22208/22208)に開示されており、配列番号1として示されるとおりである。
以下の実験例において、特段の記載が無い場合における薬物-ポリマー複合体は、上記(1)~(3)で調製した薬物-ポリマー複合体である(表1に記載のもの)。
[実験例2:in vitroでのミトコンドリアへの集積]
BxPC3細胞を、10%V/V FBSおよび1%V/V ペニシリン-ストレプトマイシンを含むRPMI中、1,000,000細胞/ウェルの濃度で12ウェルプレートに播き、一晩インキュベートした。薬物-ポリマー複合体を、25μMの濃度でBxPC3細胞を含む上記ウェルに添加し、24時間インキュベートした。余分のポリマーを取り除くために細胞を新鮮な培地で十分かつ注意深く洗浄した後、0.1mg/mL Hoechst 33342と15分間インキュベーションして細胞の核を染色した。細胞を新鮮な培地で十分かつ注意深く洗浄した後、細胞を室温で15分間4%PFA(4%パラホルムアルデヒド含有PBS溶液)で固定した。試料を共焦点レーザー走査型顕微鏡(CLSM)(LSM 780;カールツァイス)で観察した。各蛍光を以下のように異なる励起(ex.)および発光(em.)フィルターによって観察した。Hoechst 33342:ex.405nm/em.410-485nm、GFP:ex.488nm/em.489-551nm、スルホ-Cy5:ex.633nm/em.638-746nm。製造業者のプロトコルに従ってミトコンドリア分離キット(Abcam)を用いて細胞からミトコンドリアを分離した。ミトコンドリア中の薬物-ポリマー複合体の含有量は、SPARK(R)マルチモードマイクロプレートリーダー(TECAN)を用いてミトコンドリア中のスルホ-Cy5の蛍光強度で評価した。結果を図2に示す。図中、(a)は顕微鏡観察画像であり、(b)はミトコンドリアにおける蛍光強度を示すグラフである。

図2に示されるとおり、PC結合薬物-ポリマー複合体は、すべての分子量において、Ac化薬物-ポリマー複合体およびTPP結合薬物-ポリマー複合体よりも良好なミトコンドリア集積性を示した。
[実験例3:金ナノ粒子のミトコンドリアへの送達]
図3に示すスキームでリガンドが結合された金ナノ粒子を調製した。
2-メタクリロイルオキシエチルホスホリルコリン(MPC)を、マイケル付加によってチオールおよび第一級アミン末端PEGと結合させて、PC-PEG-NH2を得た。TPP-COOHを、片側遊離で、もう片側はtert-ブトキシカルボニル(Boc)で保護されたアミン末端PEGと結合させて、結合後、Bocを脱保護することで、TPP-PEG-NH2を得た。リガンドが導入された第一級アミン末端PEGまたはメトキシおよび第一級アミン末端PEGを、DMF中の1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミドカップリングによってリポ酸と結合させた。生成物のジチオラン部分をジヒドロリポ酸に還元してチオール含有ポリマー(MeO-PEG-(SH)2、PC-PEG-(SH)2、TPP-PEG-(SH)2)を生成させ、これらを表3に示す所定の割合で混合して、水性条件下で金ナノ粒子にキレート化した。なお、用いたPEGは直鎖であり、その分子量(Mw)はいずれも、2000であった。
図3に示すスキームでリガンドが結合された金ナノ粒子を調製した。

ポリマーにキレート化された金ナノ粒子のサイズおよび表面電位をそれぞれ、DLSおよびELSを用いて評価するとともに、当該金ナノ粒子の形態をTEMにより観察した。次に、10%FBS含有RPMI中で、2.5×1012particles/mLの濃度の金ナノ粒子とともにBxPC3細胞を24時間インキュベートし、金ナノ粒子の細胞内微小分布を、TEM観察した。TEMのサンプルは、既報(Hirabayashi,Y.et al. Science 358,623-630(2017))に従って調製した。結果を表3および図4に示す。図4中、(a)および(b)はそれぞれ、MeO-PEG-S-AuNPとインキュベートした細胞のTEM画像およびそのミトコンドリア部分の拡大画像であり、(c)および(d)はそれぞれ、25%PC-PEG-S-AuNPとインキュベートした細胞のTEM画像およびそのミトコンドリア部分の拡大画像であり、(e)は(d)のミトコンドリアおよび金ナノ粒子を示す断層画像である((a)および(c)のスケールバー:500nm、(b)および(d)のスケールバー:200nm)。
図4に示されるとおり、PC基が導入された金ナノ粒子は、ミトコンドリアおよびエンドソームの内部に移行したことが確認されたが、細胞質および核への移行は確認されなかった。一方、TPP基が導入された金ナノ粒子は細胞質への移行が確認され、リガンドが導入されていない金ナノ粒子は主にエンドソーム内に分布していた。この結果は、PC基のミトコンドリアターゲティングの選択性がTPP基の選択性よりも高いことを裏付けるものである。
[実験例4:STARD7KD細胞におけるミトコンドリアへの集積]
下記のようにして、Lipofectamine(R)を用いたトランスフェクションよりBxPC3細胞のSTARD7を不活化(ノックアウト)した。STARD7は、ゴルジ体、滑面小胞体、細胞質等の周囲の細胞小器官からミトコンドリアにホスファチジルコリンを輸送するタンパク質である。
BxPC3細胞を、10%V/V FBSおよび1%V/V ペニシリン-ストレプトマイシンを含むRPMI中、5%のミトコンドリア-GFP(BacMam2.0)(Mito-GFP)と共にインキュベートし、成熟細胞を新鮮な培地で3回洗浄した。STARD7-ノックアウトsiRNAまたは陰性対照としてのスクランブルRNAを、RNAiMAXを用いて細胞にトランスフェクトした。RNAiMAXを含む培地を用いて細胞を24時間インキュベートした。新しい培地で細胞を3回洗浄した後、培地に溶解した25μM PC結合薬物-ポリマー複合体またはTPP結合薬物-ポリマー複合体を添加し、細胞を24時間インキュベートした。STARD7不活化細胞と本来の細胞(native cell)とにおけるミトコンドリアおよび薬物-ポリマー複合体の共局在の違いを、CLSM(LSM 780)で取得した共局在画像およびミトコンドリア単離からの定量データによって調べた。具体的には、薬物-ポリマー複合体とインキュベートしたSTARD7ノックダウンBxPC3細胞におけるAlexa 488標識ミトコンドリアをLSM 780を用いてイメージングすることにより、STARD7のノックダウンによる薬物-ポリマー複合体のミトコンドリア集積への影響を確認した。結果を図5に示す。また、薬物-ポリマー複合体のミトコンドリア集積を、ミトコンドリア単離アッセイで定量した。結果を図6に示す。

図5および6に示されるとおり、PC結合薬物-ポリマー複合体のミトコンドリアへの集積はSTARD7のノックダウンによって効果的に抑制された一方で、TPP結合薬物-ポリマー複合体のミトコンドリアへの集積はその影響を受けなかった。これらの結果は、PC結合薬物-ポリマー複合体はSTARD7に関連する経路によって輸送されるのに対し、TPP結合薬物-ポリマー複合体はこの経路で輸送されなかったことを示す。このことから、STARD7がPC結合薬物-ポリマー複合体をがん細胞内のミトコンドリアに輸送することが示唆される。
なお、Lipofectamineトランスフェクションを用いた処理前後の薬物-ポリマー複合体の細胞取り込みの差を比較したところ、siRNA処理BxPC3細胞とスクランブルRNA処理BxPC3細胞との間で細胞取り込みに差はなかった。
[実験例5:in vivoでの薬物-ポリマー複合体の生体内分布およびがん細胞への集積]
5週齢の雌BALB/cヌードマウスをチャールズリバーラボラトリーズジャパンから購入した。すべての動物実験は、東京大学が定める実験動物の飼養および使用に関するガイドラインに準拠して実施した。

BALB/cヌードマウスを腫瘍モデルに用いた。50μLのRPMI1640に懸濁したBxPC3細胞(5×106cells ml‐1)を雌BALB/cヌードマウス(5週齢)の右背部に皮下接種し、腫瘍モデルを作製した。腫瘍の体積が≧100mm3に達した時点で、生体内分布およびミトコンドリア集積評価に供した。
(生体内分布)
一連の薬物-ポリマー複合体(0.2mM、100μL)を、BxPC3腫瘍を有する8週齢のBALB/cヌードマウスに尾静脈投与した。24時間の循環後、マウスを屠殺し、腫瘍、肝臓、腎臓、脾臓、心臓、および肺を摘出した。臓器表面の蛍光はIVISイメージングシステムで評価した。結果を図7に示す。その後、臓器および腫瘍を秤量し、ダウンス型組織ホモジナイザーでホモジナイズした。ホモジナイズした各臓器および腫瘍の赤色蛍光強度(スルホ-Cy5)をプレートリーダーで測定した。結果を図8に示す。
一連の薬物-ポリマー複合体(0.2mM、100μL)を、BxPC3腫瘍を有する8週齢のBALB/cヌードマウスに尾静脈投与した。24時間の循環後、マウスを屠殺し、腫瘍、肝臓、腎臓、脾臓、心臓、および肺を摘出した。臓器表面の蛍光はIVISイメージングシステムで評価した。結果を図7に示す。その後、臓器および腫瘍を秤量し、ダウンス型組織ホモジナイザーでホモジナイズした。ホモジナイズした各臓器および腫瘍の赤色蛍光強度(スルホ-Cy5)をプレートリーダーで測定した。結果を図8に示す。
図7および図8から、PC-8-PEG20は、投与後24時間の時点で、腫瘍に選択的に集積することが分かる。これに対し、PEGの分子量が10Kである薬物-ポリマー複合体(PC-8-PEG10、TPP-8-PEG10、およびAc-8-PEG10)はいずれも、腫瘍および各臓器への集積性が低かった。PEGの分子量が10Kである薬物-ポリマー複合体は、サイズが5nm以下であることから、腎臓でろ過され、体外へ排出された可能性がある。また、PEGの分子量が40Kである薬物-ポリマー複合体(PC-8-PEG40、TPP-8-PEG40、およびAc-8-PEG40)およびAc-8-PEG20はいずれも、腫瘍および各臓器に非特異的に集積した。また、TPPリガンドは肝臓および腎臓への集積が多い傾向があった。上記結果から、以下のように考察される。分子量が40Kの薬物-ポリマー複合体は、PEGの分子量が大きいためにリガンドの露出度が低くなった結果、リガンド特異性が低くなった。一方、PC-8-PEG20は、良好な血中滞留性およびリガンド露出度を有し、リガンド依存性を有する細胞取り込み(例えば、リン脂質輸送タンパク質(PLTP)を介した細胞取り込み)およびその後のSTARD7を介したミトコンドリア輸送の点で有利であり得ることから、in vivoにおいてミトコンドリア(例えば、がん細胞のミトコンドリア)をターゲットするのに適切である。
(ミトコンドリア集積)
一連の薬物-ポリマー複合体(0.2mM、100μL)を、BxPC3腫瘍を有する8週齢のBALB/cヌードマウスに尾静脈投与した。24時間の循環後、Hoechst 33342(16mM、100μL)を尾静脈投与し、その30分後に屠殺し、腫瘍を摘出した。採取した腫瘍の一部をアセトン/ドライアイス混合物中で直ちに凍結し、クライオスタットで厚さ10μmに切片化した。切片を固定し、Rhodamineでミトコンドリアを染色した。残りの腫瘍の断面および固定化した切片をLSM 780を用いて観察した。腫瘍および切片の観察画像をそれぞれ図9および10に示す。
また、採収した腫瘍の一部を、組織用ミトコンドリア分離キットを用いてミトコンドリアを単離し、単離ミトコンドリアにおける蛍光強度を測定することで、腫瘍のミトコンドリアへの薬物-ポリマー複合体の集積を定量評価した。結果を図11に示す。

図9に示されるとおり、PEGの分子量が40Kの複合体はすべて腫瘍への集積が確認された。また、PEGの分子量が20Kまたは10Kの場合、PC結合薬物-ポリマー複合体が顕著に腫瘍全体へ集積していた。一方、TPP結合薬物-ポリマー複合体は、最も集積が少なかった。また、図10に示されるとおり、in vivoにおいてはPC結合薬物-ポリマー複合体に関してのみ、ミトコンドリアとの共局在が確認され、集積の定量評価においても、PC結合薬物-ポリマー複合体が効率的にミトコンドリアに集積することが確認された(図11)。
[実験例6:ミトコンドリアへのタンパク質デリバリー]
(リガンド結合DNASE1-ポリマー複合体の調製)
末端がアジド(-N3)およびアミン(-NH2)であり、20kの分子量(Mw)を有する8分岐型ポリエチレングリコール「8-PEG20-(NH2)7―(N3)」をCreative PEGWorksから購入した。8-PEG20-(NH2)7―(N3)における各PEG鎖の分子量(Mw)は約2500であり、7つのPEG鎖の各末端にはアミノ基が導入されており、残りの1つのPEG鎖の末端にはアジド基が導入されている。8-PEG20-(NH2)7―(N3)に下記3種のリガンドをPBS(-)の中でDMT-MM(アミンに対して10当量)とTEA(アミンに対して100当量)と混ぜて、室温で24時間混合した。追加のDMT-MM(アミンに対して2当量)を反応の間、6時間毎に投入した。6-(O-phosphorylcholine)hydroxyhexanoic acid(富士フイルム和光純薬社)(アミンに対して10当量)、(3-Carboxybutyl)triphenylphosphonium Bromide(富士フイルム和光純薬社)(アミンに対して10当量)、またはAcetic Acid(東京化成工業社)(アミンに対して10当量)を各リガンドとして反応させ、3種のリガンド結合ポリマー(PC-8-PEG20-(N3)、TPP-8-PEG20-(N3)、およびAc-8-PEG20-(N3))を合成した。
DBCO-PEG4-NHS Ester(CAS RN:1427004-19-0)を東京化成工業社から購入した。上記で合成した3種のリガンド結合ポリマーを各DMF中に20μg/mLで溶解したDBCO-PEG4-NHS Esterを10mMリン酸緩衝液中で1:1.1(リガンド:DBCO(モル比))で混合して、-30℃で凍結、4℃で溶解を3回繰り返し、-N3とDBCOでのクリック反応を行い、PC-8-PEG20-NHS、TPP-8-PEG20-NHS、およびAc-8-PEG20-NHSを合成した。得られた3種のポリマーを、超遠心フィルターをかけることによって精製した(MWCO:10000、3500g、8分)。
精製した3種のポリマーをリン酸緩衝液(pH8.0)の中でDNASE1と2:1のモル比で反応させた(4℃、オーバーナイト)。これにより、約7つのPEG鎖の末端にリガンド(PC、TPP、またはAc)が結合され、1つのPEG鎖の末端にDNASE1が結合されたリガンド結合DNASE1-ポリマー複合体を得た。異なるリガンドを有する3種のリガンド結合DNASE1-ポリマー複合体を、超遠心フィルターをかけることによって精製した(MWCO:35000、3500g、8分)。精製された複合体に搭載されたDNASE1の濃度はBCAタンパク質アッセイ(ThermoFisher社)を用いて定量した。
(リガンド結合DNASE1-ポリマー複合体の調製)
末端がアジド(-N3)およびアミン(-NH2)であり、20kの分子量(Mw)を有する8分岐型ポリエチレングリコール「8-PEG20-(NH2)7―(N3)」をCreative PEGWorksから購入した。8-PEG20-(NH2)7―(N3)における各PEG鎖の分子量(Mw)は約2500であり、7つのPEG鎖の各末端にはアミノ基が導入されており、残りの1つのPEG鎖の末端にはアジド基が導入されている。8-PEG20-(NH2)7―(N3)に下記3種のリガンドをPBS(-)の中でDMT-MM(アミンに対して10当量)とTEA(アミンに対して100当量)と混ぜて、室温で24時間混合した。追加のDMT-MM(アミンに対して2当量)を反応の間、6時間毎に投入した。6-(O-phosphorylcholine)hydroxyhexanoic acid(富士フイルム和光純薬社)(アミンに対して10当量)、(3-Carboxybutyl)triphenylphosphonium Bromide(富士フイルム和光純薬社)(アミンに対して10当量)、またはAcetic Acid(東京化成工業社)(アミンに対して10当量)を各リガンドとして反応させ、3種のリガンド結合ポリマー(PC-8-PEG20-(N3)、TPP-8-PEG20-(N3)、およびAc-8-PEG20-(N3))を合成した。
DBCO-PEG4-NHS Ester(CAS RN:1427004-19-0)を東京化成工業社から購入した。上記で合成した3種のリガンド結合ポリマーを各DMF中に20μg/mLで溶解したDBCO-PEG4-NHS Esterを10mMリン酸緩衝液中で1:1.1(リガンド:DBCO(モル比))で混合して、-30℃で凍結、4℃で溶解を3回繰り返し、-N3とDBCOでのクリック反応を行い、PC-8-PEG20-NHS、TPP-8-PEG20-NHS、およびAc-8-PEG20-NHSを合成した。得られた3種のポリマーを、超遠心フィルターをかけることによって精製した(MWCO:10000、3500g、8分)。
精製した3種のポリマーをリン酸緩衝液(pH8.0)の中でDNASE1と2:1のモル比で反応させた(4℃、オーバーナイト)。これにより、約7つのPEG鎖の末端にリガンド(PC、TPP、またはAc)が結合され、1つのPEG鎖の末端にDNASE1が結合されたリガンド結合DNASE1-ポリマー複合体を得た。異なるリガンドを有する3種のリガンド結合DNASE1-ポリマー複合体を、超遠心フィルターをかけることによって精製した(MWCO:35000、3500g、8分)。精製された複合体に搭載されたDNASE1の濃度はBCAタンパク質アッセイ(ThermoFisher社)を用いて定量した。
(リガンド結合DNASE1-ポリマー複合体のin vitroでのミトコンドリアへの集積)
BxPC3細胞を、10%V/V FBSおよび1%V/V ペニシリン-ストレプトマイシンを含むRPMI中、1,000,000細胞/ウェルの濃度で12ウェルプレートに播き、一晩インキュベートした。リガンド結合DNASE1-ポリマー複合体を、100μg/mLのDNASE1濃度でBxPC3細胞に添加し、3時間インキュベートした。余分のポリマーを取り除くために細胞を新鮮な培地で十分かつ注意深く洗浄した後、21時間培地でポストインキュベーションを行った。次に、製造業者のプロトコルに従ってミトコンドリア分離キット(Abcam)を用いて細胞からミトコンドリアを分離した。分離したミトコンドリアの中に残っているDNAの量を、PCRを用いて定量した。結果を図12に示す。

図12に示されるように、PC結合DNASE1-ポリマー複合体とインキュベーションしたサンプルにおいてのみミトコンドリア中のDNA量が減少した。これにより、PC結合DNASE1-ポリマー複合体がミトコンドリアまで送達されたことが示唆される。
[実験例7:ミトコンドリアへの核酸デリバリー]
(リガンド結合ASO-ポリマー複合体の調製)
ミトコンドリアmRNAであるCOX II(COX2)のmRNAを標的とし、DBCO修飾されたASOを北海道システムサイエンス株式会社から購入した。
・COX2標的ASO
5’-CUUGCGCUGCAUGUGCCAU-spacer-DBCO-3’(配列番号2)
末端がアジド(-N3)およびアミン(-NH2)であり、20kの分子量(Mw)を有する8分岐型ポリエチレングリコール「8-PEG20-(NH2)7―(N3)」をCreative PEGWorksから購入した。8-PEG20-(NH2)7―(N3)における各PEG鎖の分子量(Mw)は約2500であり、7つのPEG鎖の各末端にはアミノ基が導入されており、残りの1つのPEG鎖の末端にはアジド基が導入されている。
8-PEG20-(NH2)7―(N3)をPBSに10mg/mlの濃度で溶解した。上記溶液にPC-COOH(アミン10当量)またはTPP-COOH(アミン10当量)とDMT-MM(アミン10当量)とを添加して、室温で24時間反応させた。追加のDMT-MM(アミンに対して2当量)を反応の間、6時間毎に投入した。生成物を、Sephadex G-25(Cytiva)を含む使い捨てPD-10脱塩カラムを用いて精製した。凍結乾燥によって2種のリガンド結合ポリマー(PC-8-PEG20-(N3)、TPP-8-PEG20-(N3))を回収した。
8-PEG20-(NH2)7―(N3)を超脱水アセトニトリルに溶解した。上記溶液にアルゴン雰囲気下でTEA(アミン100当量)を加えた。室温で30分間混合した後、酢酸無水物を反応系に滴下で添加した。アルゴン雰囲気中で混合物を室温で24時間撹拌した。蒸発によってアセトニトリルを除去し、得られたアセチル化ポリマー(Ac-8-PEG20-(N3))を脱イオン水に溶解した。上記と同じ方法でAc-8-PEG20-(N3)の精製および回収を行った。
DBCOとアジド基とのクリックケミストリー反応を利用して上記ポリマーとASOとを結合した。具体的には、(PC、TPP、Ac)-8-PEG20-(N3)を脱イオン水に溶解し、N3に対して1当量のASOを混合し、凍結融解法(凍結:-30°C、融解:R.T.)を3回行った。
以上のようにして、3種類のリガンド結合ASO-ポリマー複合体(Ac-8-PEG20-COX2、TPP-8-PEG20-COX2、PC-8-PEG20-COX2)を得た。
(リガンド結合ASO-ポリマー複合体の調製)

DBCOとアジド基とのクリックケミストリー反応を利用して上記ポリマーとASOとを結合した。具体的には、(PC、TPP、Ac)-8-PEG20-(N3)を脱イオン水に溶解し、N3に対して1当量のASOを混合し、凍結融解法(凍結:-30°C、融解:R.T.)を3回行った。
以上のようにして、3種類のリガンド結合ASO-ポリマー複合体(Ac-8-PEG20-COX2、TPP-8-PEG20-COX2、PC-8-PEG20-COX2)を得た。
(PCRによる標的ミトコンドリアmRNAレベルの定量)
1ウェル当たり1mLの無血清RPMI培地を含む6ウェルプレート上で、37℃、5%CO2/空気雰囲気下、上記3種類のリガンド結合ASO-ポリマー複合体をBxPC3細胞(1×106細胞/ウェル)と3時間インキュベートした。細胞をPBSで3回洗浄し、完全培地で21時間ポストインキュベートした。RNeasy Mini Kit(Qiagen)を製造者のプロトコルに従って用いて、上記細胞から全RNAを精製した。High Capacity RNA-to-cDNAキット(Thermo fishier Scientific)を製造者のプロトコルに従って用いて、各サンプルから抽出されたRNA懸濁液から逆転写反応を行った。FastStart Universal SYBR Green Master(ROX)(Thermo fishier Scientific)を用いて、転写されたcDNAから定量的PCR解析を行った。
RT-PCRに使用したプライマーを以下に示す。

結果を図13に示す。図中、コントロールは、リガンド結合ASO-ポリマー複合体で処理されていない細胞サンプルである。
図13(a)は、コントロールmRNAの発現(転写)量に対する標的mRNAの発現量の割合を、コントロールにおける割合を1とした場合の相対値として示し、図13(b)は、β-アクチンmRNAの発現量を、コントロールにおける発現量を1とした場合の相対値として示す。図13(a)に示されるとおり、PC-8-PEG20-COX2で処理された細胞におけるND1に対するCOX2の発現レベルは大きく低下していた。ND1はミトコンドリアmRNAであるが、上記ASOの標的ではないこと、および、細胞mRNAであるβ-Actinの発現量が変化しなかったことを考慮すると、PC-8-PEG20-COX2は、細胞の他の状態に影響を与えることなく、ASOをミトコンドリア内に効果的に送達したと考えられる。
[実験例8:ミトコンドリアへの低分子薬物デリバリー]
(リガンド結合Sim-ポリマー複合体の調製)
8-PEG20-NH2(末端がすべてアミン(-NH2)であり、20kの分子量(Mw)を有する8分岐型PEG、Creative PEGWorks)を用いて、図14に示すスキームで、シンバスタチン(Sim)を薬物として含むリガンド結合Sim-ポリマー複合体を合成した。シンバスタチンは、コレステロールを下げるために使用されるHMG-CoAレダクターゼ阻害剤である。シンバスタチンは、また、抗がん作用を示す。具体的には、シンバスタチンは、ミトコンドリアの膜電位を減少させ、Smac/DIABLOおよびシトクロムCを細胞質に放出して、がん細胞のアポトーシスを引き起こす。このようなシンバスタチンによるミトコンドリア損傷は、ミトコンドリアへのシンバスタチン送達を増加することによって増大され得る。
まず、エステル結合を作るために、シンバスタチンの二級アルコールを塩化サクシニルと反応させた。ベンゼンに溶解したシンバスタチン(500mg、1当量)を凍結乾燥し、10mLの超脱水ジクロロメタン(DCM)に溶解した。塩化スクシニル(393μL、3当量)およびTEA(75μL、1.5当量)をシンバスタチンに添加し、室温で3時間撹拌した。その後、脱イオン水を加え、15分間撹拌して未反応のカルボン酸塩化物を不活性化した。合成したシンバスタチン-COOHをDCMと脱イオン水との相分離抽出により精製した。DCM相を採取し、追加の水で5回洗浄した。精製後、DCMを30℃で蒸発させ、サンプルをベンゼンに溶解して凍結乾燥することで、残留水分を可能な限り除去した。これにより、深紅色の粘性の液体が得られた。反応は、1H-NMR(メタノール-d4;25℃)で確認した。
上記シンバスタチン-COOHを、DMF中のEDC/NHSカップリングにより、8-PEG20-NH2の一級アミンの1つに結合した。超脱水DMF中で、8-PEG20-NH2(500mg、1当量)をシンバスタチン-COOH(39mg、3当量)と混合した。混合液を4℃で6時間撹拌した。EDC/HCl(7.2mg、1.5当量)およびTEA(5.2μL、1.5当量)を触媒として2時間ごとに添加した。ポリマーを純水およびメタノールに対して、交互に各3回透析して精製した(透析バッグのMWCO:3,500;スペクトラムラボラトリーズ)。シンバスタチンの結合は、1H-NMR(メタノール-d4;25℃)で確認した。以上のようにして、1つのアミノ末端にシンバスタチンが結合されたシンバスタチン-8-PEG20-(NH2)7を得た。
上記シンバスタチン-8-PEG20-(NH2)7に対して、上記と同様の方法でアミノ基を各リガンド(すなわち、PC、TPP、またはAc)でキャップして、リガンド結合Sim-ポリマー複合体(PC-8-PEG20-Sim、TPP-8-PEG20-Sim、およびAc-8-PEG20-Sim)を得た。各リガンド結合Sim-ポリマー複合体におけるリガンドおよびシンバスタチンの導入割合を、1H-NMR(メタノール-d4;25℃)で評価した。
また、室温でZatasizer Nano ZS90を用いて電気泳動光散乱(ELS)測定を行うことにより、各リガンド結合Sim-ポリマー複合体の表面ゼータ電位を測定した。
下記表に表示される結果から、8arm PEGに7つのリガンドと1つのSimとが結合されたことが示唆される。
(リガンド結合Sim-ポリマー複合体の調製)

上記シンバスタチン-8-PEG20-(NH2)7に対して、上記と同様の方法でアミノ基を各リガンド(すなわち、PC、TPP、またはAc)でキャップして、リガンド結合Sim-ポリマー複合体(PC-8-PEG20-Sim、TPP-8-PEG20-Sim、およびAc-8-PEG20-Sim)を得た。各リガンド結合Sim-ポリマー複合体におけるリガンドおよびシンバスタチンの導入割合を、1H-NMR(メタノール-d4;25℃)で評価した。
また、室温でZatasizer Nano ZS90を用いて電気泳動光散乱(ELS)測定を行うことにより、各リガンド結合Sim-ポリマー複合体の表面ゼータ電位を測定した。
下記表に表示される結果から、8arm PEGに7つのリガンドと1つのSimとが結合されたことが示唆される。
(リガンド結合Sim-ポリマー複合体の細胞毒性評価:MTTアッセイ)
培地(10%V/V FBSおよび1%V/V ペニシリン-ストレプトマイシンを含むRPMI1640)に懸濁したBxPC3細胞を、96ウェル細胞培養プレート(10,000細胞/ウェル)(IWAKI)上で一晩インキュベートした。培地を、種々のSim濃度(0~200μM)のリガンド結合Sim-ポリマー複合体または遊離シンバスタチンを含む培地と交換し、細胞を6時間インキュベートした。処理後の細胞を培地で洗浄し、さらに48時間ポストインキュベートした。その後、MTTアッセイキット(Dojindo Laboratories)を製造者のプロトコルを少し変更して用いて、細胞毒性を測定した。簡単に説明すると、細胞を培地で洗浄し、100μLのPBS(-)で置換し、20μLのMTTを添加して3時間インキュベートした。上清を慎重に除去し、100μLのDMSOを添加し、15分間混合した。530nmでの吸光度を、SPARK(R)マルチモードマイクロプレートリーダー(TECAN)を用いて測定した。結果を図15に示す。

(リガンド結合Sim-ポリマー複合体の細胞毒性評価:CCK-8アッセイ)
96ウェル細胞培養プレート(IWAKI)に、BxPC3細胞を5,000細胞/ウェルの濃度で分注した。細胞を上記MTTアッセイと同様にリガンド結合Sim-ポリマー複合体または遊離シンバスタチンで処理した。CCK-8アッセイキット(Dojindo Laboratories)を製造者のプロトコルに従って用いて、処理後のサンプルの細胞生存率を評価した。結果を図16に示す。
96ウェル細胞培養プレート(IWAKI)に、BxPC3細胞を5,000細胞/ウェルの濃度で分注した。細胞を上記MTTアッセイと同様にリガンド結合Sim-ポリマー複合体または遊離シンバスタチンで処理した。CCK-8アッセイキット(Dojindo Laboratories)を製造者のプロトコルに従って用いて、処理後のサンプルの細胞生存率を評価した。結果を図16に示す。
図15および図16に示されるとおり、TPP-8-PEG20-SimおよびPC-8-PEG20-Simは、37.5μMを超えるシンバスタチン濃度で、遊離シンバスタチンよりもBxPC3の細胞生存率を効果的に低下させた。一方、Ac-8-PEG20-Simは、遊離シンバスタチンと比較して細胞生存率の低下を示さなかった。この結果は、PCおよびTPPリガンドがシンバスタチンの細胞毒性を増大したことを示す。
(リガンド結合Sim-ポリマー複合体のミトコンドリア毒性評価)
5,000細胞/ウェルのBxPC3細胞を上記MTTアッセイと同様にリガンド結合Sim-ポリマー複合体で処理した。処理後の細胞に関して、MitoSOX(R) Redキット(Thermo Fisher Scientific)を用いてミトコンドリアから発生する活性酸素種(ROS)を測定すること、および、JC-1ミトコンドリア膜電位アッセイキット(Thermo Fisher Scientific)を用いてミトコンドリアの膜電位変化を測定することによって、BxPC3細胞のミトコンドリアに対するシンバスタチンの毒性を評価した。ミトコンドリアのROSおよび膜電位の測定はそれぞれ、製造者のプロトコルに従って行った。ROSの測定結果およびJC-1アッセイの結果をそれぞれ、図17ならびに図18Aおよび18Bに示す。
5,000細胞/ウェルのBxPC3細胞を上記MTTアッセイと同様にリガンド結合Sim-ポリマー複合体で処理した。処理後の細胞に関して、MitoSOX(R) Redキット(Thermo Fisher Scientific)を用いてミトコンドリアから発生する活性酸素種(ROS)を測定すること、および、JC-1ミトコンドリア膜電位アッセイキット(Thermo Fisher Scientific)を用いてミトコンドリアの膜電位変化を測定することによって、BxPC3細胞のミトコンドリアに対するシンバスタチンの毒性を評価した。ミトコンドリアのROSおよび膜電位の測定はそれぞれ、製造者のプロトコルに従って行った。ROSの測定結果およびJC-1アッセイの結果をそれぞれ、図17ならびに図18Aおよび18Bに示す。
ミトコンドリアの活性酸素種(ROS)の測定によって、ミトコンドリア活性を評価した。図17に示されるとおり、PC-8-PEG20-SimおよびTPP-8-PEG20-Simは、遊離SimまたはAc-8-PEG20-Simと比較して、75μMを超えるシンバスタチン濃度で増大したミトコンドリア毒性を示した。このことから、PCリガンドおよびTPPリガンドがSim-ポリマー複合体のミトコンドリア送達に寄与することが示唆される。
図18Aは正規化した緑色蛍光強度を示す。緑色蛍光の強度が大きいことは、ミトコンドリア膜の負電荷が弱いことを示す。図18Bは正規化した赤色蛍光強度を示す。赤色蛍光の強度が大きいことは、ミトコンドリア膜が強く負に帯電していることを示す。
図18Bに示されるとおり、赤色蛍光強度の減少は、PC-8-PEG20-Simにおいて最大であり、TPP-8-PEG20-Simよりも大きい。一方、シンバスタチンを有さないPCまたはTPP結合ポリマー(PC-8-PEG20、TPP-8-PEG20)で細胞を処理した場合、TPP-8-PEG20のみがミトコンドリアの表面電荷を大きく変化させた(データは示さず)。この結果は以下のことを示唆する。PC結合ポリマーはミトコンドリアの膜電荷に影響せずに、搭載薬物であるシンバスタチンをミトコンドリアに送達し、シンバスタチンの影響でミトコンドリアの膜電荷の変化が生じた。一方で、TPP結合ポリマーはそれ自体がミトコンドリアの膜電荷に影響し、さらにシンバスタチンを送達することでミトコンドリアの膜電荷を変化させた。また、本実験でPCおよびTPP結合ポリマーにおけるシンバスタチンの量が同等であることから、上記結果は、シンバスタチン送達システムにおいて、PCリガンドがTPPリガンドよりも効果的にミトコンドリアを標的としたことを示唆する。
一方、緑色蛍光強度の減少は、ミトコンドリア膜電位がより強く負に帯電したこと、または、細胞が死滅し、JC-1を捕捉した細胞の数が減少したことを示す。図18Aから、シンバスタチン濃度が高い場合、PC-8-PEG20-Simの細胞毒性のために細胞が大幅に減少したと考えられる。一方、TPP-8-PEG20-Simは、PC-8-PEG20-Simほど緑色蛍光強度を減少させず、シンバスタチンの濃度が低い場合にはむしろ、緑色蛍光強度を増大させた。これは、TPP-8-PEG20-Simがシンバスタチンの有効濃度未満でミトコンドリアの膜電位を減少させた可能性があり、TPPリガンド自体が細胞に悪影響を及ぼし得ることが示唆される。
図18Bに示されるとおり、赤色蛍光強度の減少は、PC-8-PEG20-Simにおいて最大であり、TPP-8-PEG20-Simよりも大きい。一方、シンバスタチンを有さないPCまたはTPP結合ポリマー(PC-8-PEG20、TPP-8-PEG20)で細胞を処理した場合、TPP-8-PEG20のみがミトコンドリアの表面電荷を大きく変化させた(データは示さず)。この結果は以下のことを示唆する。PC結合ポリマーはミトコンドリアの膜電荷に影響せずに、搭載薬物であるシンバスタチンをミトコンドリアに送達し、シンバスタチンの影響でミトコンドリアの膜電荷の変化が生じた。一方で、TPP結合ポリマーはそれ自体がミトコンドリアの膜電荷に影響し、さらにシンバスタチンを送達することでミトコンドリアの膜電荷を変化させた。また、本実験でPCおよびTPP結合ポリマーにおけるシンバスタチンの量が同等であることから、上記結果は、シンバスタチン送達システムにおいて、PCリガンドがTPPリガンドよりも効果的にミトコンドリアを標的としたことを示唆する。

[実験例9:血中滞留性評価]
8週齢のBALB/cマウスをイソフルラン吸入で麻酔した。マウスの右の耳たぶを生体内共焦点レーザー走査型顕微鏡(IVCLSM)上に固定した。実施例1で調製した一連のリガンド結合ポリマー(1とのスルホ-Cy5と7つのリガンドとが結合された10kDa、20kDa、または40kDaの8分岐PEG)をマウスに静脈内注射した。一連のリガンド結合ポリマーに結合されたスルホ-Cy5の蛍光強度を、IVCLSMを用いた耳たぶの連続イメージングによって評価した。結果を図19に示す。図19は、血管内の蛍光標識の強度を正規化し、定量化したグラフである。
8週齢のBALB/cマウスをイソフルラン吸入で麻酔した。マウスの右の耳たぶを生体内共焦点レーザー走査型顕微鏡(IVCLSM)上に固定した。実施例1で調製した一連のリガンド結合ポリマー(1とのスルホ-Cy5と7つのリガンドとが結合された10kDa、20kDa、または40kDaの8分岐PEG)をマウスに静脈内注射した。一連のリガンド結合ポリマーに結合されたスルホ-Cy5の蛍光強度を、IVCLSMを用いた耳たぶの連続イメージングによって評価した。結果を図19に示す。図19は、血管内の蛍光標識の強度を正規化し、定量化したグラフである。
図19に示されるとおり、10kDaのリガンド結合ポリマー、特にTPP-8-PEG10は、急速に循環から除去された。20kDaおよび40kDaのリガンド結合ポリマーは、10kDaのリガンド結合ポリマーよりも長い血液循環を示した。特に、PC-8-PEG20は、TPP-8-PEG20およびAc-8-PEG20よりも長い血液循環を示した。
[実験例10:血管外移行性評価]
BxPC3腫瘍を有する8週齢のBALB/cヌードマウスをイソフルラン吸入で麻酔した。Hoechst 33342を、麻酔の30分前に静脈内注射した。栄養血管を傷つけることなく、皮下腫瘍の周囲に皮弁を形成した。皮弁を元に戻して伸ばし、これにより、腫瘍をIVCLSMに装着および固定した。0.2mMの濃度の一連のリガンド結合ポリマー(PC-8-PEG20、TPP-8-PEG20、およびAc-8-PEG20)100μLを、蛍光強度が1000a.u.になるように濃度を調整した10k FITC標識デキストラン(TCI, Tokyo, Japan)水溶液に溶解し、マウスに静脈内注射した。腫瘍周囲の血管外漏出を、IVCLSMを用いた連続撮像によって可視化した。撮像された画像を図20に示す。また、図20中の四角形の枠内の関心領域(ROI)における各標識の蛍光強度を定量化した。上記ROIは、腫瘍のネスト部(nest site)に位置している。結果を図21に示す。図21中、Ac dextran 10K、TPP dextran 10K、およびPC dextran 10Kはそれぞれ、Ac-8-PEG20、TPP-8-PEG20、およびPC-8-PEG20を投与したマウスにおけるデキストラン由来のFITC蛍光強度を示す。
BxPC3腫瘍を有する8週齢のBALB/cヌードマウスをイソフルラン吸入で麻酔した。Hoechst 33342を、麻酔の30分前に静脈内注射した。栄養血管を傷つけることなく、皮下腫瘍の周囲に皮弁を形成した。皮弁を元に戻して伸ばし、これにより、腫瘍をIVCLSMに装着および固定した。0.2mMの濃度の一連のリガンド結合ポリマー(PC-8-PEG20、TPP-8-PEG20、およびAc-8-PEG20)100μLを、蛍光強度が1000a.u.になるように濃度を調整した10k FITC標識デキストラン(TCI, Tokyo, Japan)水溶液に溶解し、マウスに静脈内注射した。腫瘍周囲の血管外漏出を、IVCLSMを用いた連続撮像によって可視化した。撮像された画像を図20に示す。また、図20中の四角形の枠内の関心領域(ROI)における各標識の蛍光強度を定量化した。上記ROIは、腫瘍のネスト部(nest site)に位置している。結果を図21に示す。図21中、Ac dextran 10K、TPP dextran 10K、およびPC dextran 10Kはそれぞれ、Ac-8-PEG20、TPP-8-PEG20、およびPC-8-PEG20を投与したマウスにおけるデキストラン由来のFITC蛍光強度を示す。
図20および21から、PC-8-PEG20は、他の20kDaポリマー(すなわち、TPP-8-PEG20およびAc-8-PEG20)と比較して血管外漏出が促進されることがわかる。ポリマーの血管外漏出が促進されると、腫瘍へのポリマーの集積が増大し得る。
[実験例11:ミトコンドリアとリソソームの接触時間評価]
細胞内への薬物送達においてエンドソームエスケープは有効な手段である。その一方で、8分岐PEGを用いた上記リガンド結合薬物-ポリマー複合体はエンドソームエスケープを促進する構造を有さない。よって、エンドソームエスケープを伴わないミトコンドリアターゲティング経路の確認のために、以下、リソソームとミトコンドリアとの接触を評価した。
10%V/V FBSおよび1%V/V ペニシリン-ストレプトマイシンを含むRPMI中のBxPC3細胞を15,000細胞/ウェルの濃度で8ウェルカバーガラスチャンバーに分注し、8時間置いた。培地を5体積%の「CellLightTMリソソームGFP、BacMam 2.0」および「CellLightTMミトコンドリアRFP、BacMam 2.0」を含む培地と交換し、36時間インキュベートした。新鮮な培地で十分に洗浄した後、25μMの一連のリガンド結合ポリマーで8時間処理した。上記リガンド結合ポリマーを含む培地を新鮮な培地と交換した。その後、0.1mg/mLのHoechst 33342と15分間インキュベートすることにより、細胞の核を青色蛍光で染色した。培地を新鮮な培地と交換した。ConfoCor 3モジュールおよび63X油浸対物レンズを装備した共焦点レーザー走査型顕微鏡(CLSM)(LSM 780;Carl Zeiss)を用いて、細胞およびオルガネラを観察した。リソソームおよびミトコンドリアの運動の動画を、ConfoCor 3モジュールと63X油浸対物レンズを装備した共焦点レーザー走査型顕微鏡(CLSM)(LSM 880;Carl Zeiss)を用いた4秒ごとの連続イメージングによって作成した。
各蛍光は、以下の種々の励起(ex.)および発光(em.)フィルターによってイメージングされた:Hoechst 33342:ex.405nm/em.410~485nm、GFP:ex.488nm/em.489~551nm、RFP:ex.555nm/em.570~620nm、スルホ-Cy5:ex.633nm/em.638~746nm。LSM 880のZenソフトウェアを用いて共局在率を得た。PC-8-PEG20で処理した細胞に関して、観察開始後の複数の時点における観察画像および共局在率を図22に示す。また、リソソームとミトコンドリアとの接触時間と、リガンドの種類との関係を図23に示す。図23中、コントロールは、リガンド結合ポリマーを添加しなかったサンプルを示す。
細胞内への薬物送達においてエンドソームエスケープは有効な手段である。その一方で、8分岐PEGを用いた上記リガンド結合薬物-ポリマー複合体はエンドソームエスケープを促進する構造を有さない。よって、エンドソームエスケープを伴わないミトコンドリアターゲティング経路の確認のために、以下、リソソームとミトコンドリアとの接触を評価した。

図22に示されるとおり、リソソームとミトコンドリアとの接触部位において、リソソームからミトコンドリアへのPC-8-PEG20の輸送が観察された。また、経時的に、PC-8-PEG20のリソソームとの共局在が減少し、ミトコンドリアとの共局在が増加した。これは、PC-8-PEG20がリソソームからミトコンドリアに輸送されたことを示す。この結果から、PC-8-PEG20がリソソームからミトコンドリアに直接転換されたことが示唆される。また、図23に示されるとおり、リソソームとミトコンドリアとの接触時間は、PC-8-PEG20で処理した細胞において最も長く、TPP-8-PEG20で処理した細胞においてその次に長くなった。このことから、リガンド結合ポリマーがリソソームとミトコンドリアとの接触の助けになったことが示唆される。
本発明は、例えば、DDS分野において好適に用いられ得る。
Claims (16)
- 薬物と、ホスホコリン基を有する薬物送達用ポリマーと、を含み、
STARD7に対する解離定数(KD)が1.0×10-6M以下である、薬物-ポリマー複合体。 - ミトコンドリアに前記薬物を送達するための、請求項1に記載の薬物-ポリマー複合体。
- 平均粒子径が、5.2nmを超える、請求項1に記載の薬物-ポリマー複合体。
- 前記薬物送達用ポリマーが、2つ以上のポリマー鎖を有する分岐型ポリマーであって、
前記2つ以上のポリマー鎖の少なくとも1つに前記ホスホコリン基が結合されている、請求項1に記載の薬物-ポリマー複合体。 - 前記薬物送達用ポリマーが、前記2つ以上のポリマー鎖の1つの末端にsulfo-cy5が導入され、その他の末端に非荷電性基を有するポリマーとして測定された場合に、0以下のゼータ電位を示すポリマーである、請求項4に記載の薬物-ポリマー複合体。
- 前記薬物送達用ポリマーの分子量(Mw)が、5000~100000である、請求項4に記載の薬物-ポリマー複合体。
- 前記薬物送達用ポリマーの前記2つ以上のポリマー鎖の分子量(Mw)がそれぞれ、200~50000である、請求項4に記載の薬物-ポリマー複合体。
- 前記2つ以上のポリマー鎖の少なくとも1つに前記薬物が結合されている、請求項4に記載の薬物-ポリマー複合体。
- 前記薬物送達用ポリマーが、ポリ(エチレングリコール)、ポリ(サッカライド)、ポリ(ビニルピロリドン)、ポリ(ビニルアルコール)、ポリ(アクリルアミド)、ポリ(アクリル酸)、ポリ(メタクリルアミド)、ポリ(メタクリル酸)、ポリ(メタクリル酸エステル)、ポリ(アクリル酸エステル)、ポリ(アミノ酸)、ポリ(リンゴ酸)、またはこれらの誘導体から構成される親水性ポリマーを含む、請求項1に記載の薬物-ポリマー複合体。
- 前記薬物が、生理活性物質、蛍光試薬、および造影剤から選択される少なくとも1種を含む、請求項1に記載の薬物-ポリマー複合体。
- 請求項1から10のいずれかに記載の薬物-ポリマー複合体を含む、医薬組成物。
- 2つ以上のポリマー鎖を有し、
前記2つ以上のポリマー鎖の少なくとも1つにホスホコリン基が結合されており、
