Abstract
飲料中の遊離型のカルボキシメチルリジン(CML)、遊離型のカルボキシエチルリジン(CEL)および遊離型のメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)の合計含有量が434ppb以上である、低糖質ビールテイストアルコール飲料が開示されている。この低糖質ビールテイストアルコール飲料では、コクが増強されている。
Description
本発明は、ビールテイストアルコール飲料およびその製法に関する。
近年、健康志向の高まりにより、糖質が低減されたビール風味飲料のニーズが拡大している。糖質が低減されたビールテイストアルコール飲料においては、糖質低減により香味バランス調節効果が低下するため、コクが低下したり、嫌な酸味が目立つなど、香味バランスを損ねてしまうという問題がある。
一方で、メイラード反応によって得られる産物のうち、1000~5000Daの分子量からなるペプチドが、コクの増強に寄与することが報告されている(非特許文献1)が、具体的な寄与成分は明らかにされておらず、また、ビール類における香味バランスに対する作用は知られていない。

本発明者らは、低糖質のビールテイストアルコール飲料において、加熱・熟成工程で生成するメイラード反応産物(MRP:Maillard reaction product)の一部が、特定の濃度範囲でビールテイストアルコール飲料のコクを増強し得ることを見出した。本発明はこの知見に基づくものである。
従って、本発明は、コクが増強された低糖質ビールテイストアルコール飲料およびその製法を提供する。
そして、本発明には、以下の発明が包含される。
(1)飲料中の遊離型のカルボキシメチルリジン(CML)、遊離型のカルボキシエチルリジン(CEL)および遊離型のメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)の合計含有量が434ppb以上である、低糖質ビールテイストアルコール飲料。
(2)飲料中の結合型CML、結合型CELおよび結合型MG-H1の合計含有量が65ppb以上であり、
前記結合型のCML、結合型のCELおよび結合型のMG-H1が、分子量400~3,000Daのペプチド画分に含まれるものである、前記(1)に記載の低糖質ビールテイストアルコール飲料。
(3)飲料中の結合型CML、結合型CELおよび結合型MG-H1の合計含有量が72ppb以上である、前記(2)に記載の低糖質ビールテイストアルコール飲料。
(4)飲料中の遊離型CML、遊離型CEL、遊離型MG-H1、結合型CML、結合型CELおよび結合型MG-H1の合計含有量が499ppb以上であり、
前記結合型のCML、結合型のCELおよび結合型のMG-H1が、分子量400~3,000Daのペプチド画分に含まれるものである、前記(1)に記載の低糖質ビールテイストアルコール飲料。
(5)飲料中の遊離型CML、遊離型CEL、遊離型MG-H1、結合型CML、結合型CELおよび結合型MG-H1の合計含有量が510ppb以上である、前記(4)に記載の低糖質ビールテイストアルコール飲料。

本発明によれば、低糖質ビールテイストアルコール飲料において、コクを増強することが可能となる。また、本発明によれば、低糖質ビールテイストアルコール飲料において、嫌な酸味を低減することも可能である。さらに、本発明によれば、低糖質ビールテイストアルコール飲料において、アルコール辛さを低減することも可能である。
本発明において有効成分とされる3物質:カルボキシメチルリジン(CML)、カルボキシエチルリジン(CEL)およびメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)は、以下の化学構造を有するものである。
遊離型の上記3物質は、飲料中において、上記の構造そのもの、あるいは一部イオン化した状態で存在するものである。結合型の上記3物質は、飲料中において、分子量400~3,000Daのペプチド画分に含まれ、リジンないしアルギニンの側鎖が糖化修飾された残基として存在するものである。本発明において示される上記物質の量および濃度は、遊離型および結合型の両方について、上記の構造を有する化合物としての質量および濃度である。本明細書では、遊離型の上記3物質を「遊離型有効3成分」といい、結合型の上記3物質を「結合型有効3成分」ということがある。
本発明において「ビールテイストアルコール飲料」とは、ビール(麦芽およびホップを原料として用い、ビール酵母による発酵によって得られるアルコール飲料)、またはビールと同様の風味を有するアルコール飲料を意味する。本発明において「アルコール飲料」とは、アルコール(エタノール)の濃度が1v/v%以上の飲料を意味する。本発明のビールテイストアルコール飲料は、好ましくはホップを原料として用いることによりホップの香気が付与された発酵飲料である。本発明のビールテイストアルコール飲料は、好ましくは発酵麦芽飲料、すなわち、原料として少なくとも麦芽を使用した飲料を意味する。このような発酵麦芽飲料としては、ビール、発泡酒などが挙げられる。本発明のビールテイストアルコール飲料は、好ましくは麦芽使用比率(麦芽比率ともいう)0質量%以上100質量%以下、より好ましくは25質量%以上100質量%以下、さらに好ましくは50質量%以上100質量%以下の発酵麦芽飲料である。「麦芽使用比率」とは、平成30年4月1日が施行日の酒税法および酒類行政関係法令等解釈通達に従って計算された値を意味する。
本発明のビールテイストアルコール飲料は、低糖質のビールテイストアルコール飲料、すなわち、糖質の含有量が通常よりも低減されたビールテイストアルコール飲料とされる。この「通常よりも低減された」とは、そのビールテイストアルコール飲料を製造する際に糖質の含有量を低下させるための工夫がなされていることを意味する。このような低糖質ビールテイストアルコール飲料における具体的な糖質濃度の数値は特に限定されるものではないが、例えば、1.5g/100mL以下、好ましくは1.5g/100mL未満、より好ましくは1.4g/100mL以下、さらに好ましくは1.3g/100mL以下、さらに好ましくは1.2g/100mL以下、さらに好ましくは1.1g/100mL以下、さらに好ましくは1.0g/100mL以下、さらに好ましくは1.0g/100mL未満、さらに好ましくは0.9g/100mL以下、さらに好ましくは0.8g/100mL以下、さらに好ましくは0.7g/100mL以下、さらに好ましくは0.6g/100mL以下、さらに好ましくは0.5g/100mL以下、さらに好ましくは0.5g/100mL未満とすることができる。一つの実施態様において、低糖質ビールテイストアルコール飲料中の糖質の濃度は0.4g/100mL以下である。
糖質濃度の測定は公知の方法によって行うことができ、当該試料の質量から、水分、たんぱく質、脂質、灰分および食物繊維量を除いて算出する方法(食品表示基準について(平成27年3月30日 消食表第139号)別添 栄養成分等の分析方法等 参照)に従って行うことができる。
本発明において「コク」とは、味の強さ、複雑さ、および持続性から成る総合評価により決定される風味をいう。
本発明において「嫌な酸味」とは、後味などに感じる不快な酸味をいう。
本発明において「アルコール辛さ」とは、アルコール飲料の飲用時に感じられるピリピリとした刺激を伴う辛さをいう。
本発明において、「ppm」という単位は「mg/L」と同義であり、「ppb」という単位は「μg/L」と同義である。
本発明のビールテイストアルコール飲料は、飲料中の遊離型有効3成分の合計含有量が所定の範囲にあるものである。また、本発明の好ましい実施態様によれば、本発明のビールテイストアルコール飲料は、結合型有効3成分をも含有し、その合計含有量が所定の範囲にあるものとされる。このようなビールテイストアルコール飲料は、その製造の際に、遊離型有効3成分の合計含有量や結合型有効3成分の合計含有量を調整することにより得ることができる。遊離型有効3成分および結合型有効3成分の濃度調整の具体的手段は特に限定されるものではなく、例えば、遊離型有効3成分または結合型有効3成分の添加、遊離型有効3成分または結合型有効3成分を含有する原料の使用量の増減、遊離型有効3成分または結合型有効3成分を最終製品内に生成する原料の使用量の増減、酵母による発酵によって遊離型有効3成分または結合型有効3成分に変換される物質の濃度調整等が挙げられる。
本発明のビールテイストアルコール飲料中の遊離型有効3成分の合計含有量は、例えば、434ppb以上とされ、好ましくは438ppb以上、より好ましくは700ppb以上とされる。遊離型有効3成分の合計含有量が多いビールテイストアルコール飲料では高いコク増強効果や嫌な酸味低減効果、アルコール辛さ低減効果が認められるため、その上限は特に制限されるものではないが、あえて設定するとすれば、例えば、3000ppb、好ましくは2000ppb、より好ましくは1800ppb、さらに好ましくは1300ppb、さらに好ましくは1100ppbとすることができる。遊離型有効3成分は、原料由来のものであってもよく、植物原料とは別に添加されたものであってもよく、さらに発酵により生成されたものであってもよい。遊離型有効3成分の濃度は、例えば、原料の組成、原料の加工条件、仕込条件、および発酵条件などをコントロールすることにより、制御することができる。
本発明の好ましい実施態様によれば、本発明のビールテイストアルコール飲料中の結合型有効3成分の合計含有量は、0ppb以上、より好ましくは65ppb以上、さらに好ましくは72ppb以上、さらに好ましくは100ppb以上、さらに好ましくは117ppb以上とされる。結合型有効3成分の合計含有量が多いビールテイストアルコール飲料では高いコク増強効果や嫌な酸味低減効果、アルコール辛さ低減効果が認められるため、その上限は特に制限されるものではないが、あえて設定するとすれば、例えば、2000ppb、好ましくは1600ppb、より好ましくは1000ppb、さらに好ましくは832ppbとすることができる。結合型有効3成分は、原料由来のものであってもよく、植物原料とは別に添加されたものであってもよく、さらに発酵により生成されたものであってもよい。結合型有効3成分の濃度は、例えば、原料の組成、原料の加工条件、仕込条件、および発酵条件などをコントロールすることにより、制御することができる。本発明の別の好ましい実施態様によれば、本発明のビールテイストアルコール飲料中の結合型有効3成分の合計含有量は、アルコール辛さ低減の観点から、65ppb以上が好ましい。
本発明の一つの実施態様によれば、本発明のビールテイストアルコール飲料中の遊離型有効3成分の合計含有量は、例えば434ppb以上とされ、好ましくは438ppb以上、さらに好ましくは700ppb以上とされ、かつ、結合型有効3成分の合計含有量は、0ppb以上、好ましくは65ppb以上、より好ましくは72ppb以上、さらに好ましくは100ppb以上、さらに好ましくは117ppb以上とされる。本発明の他の一つの実施態様によれば、本発明のビールテイストアルコール飲料中の遊離型有効3成分の合計含有量は1003ppb以上とされ、かつ、結合型有効3成分の合計含有量は、0ppb以上、好ましくは65ppb以上、より好ましくは72ppb以上、さらに好ましくは117ppb以上とされる。
本発明の他の一つの実施態様によれば、本発明のビールテイストアルコール飲料中の遊離型有効3成分の合計含有量は、434~1800ppbとされ、好ましくは438~1100ppbとされ、かつ、結合型有効3成分の合計含有量は、0~832ppb、好ましくは65~832ppb、より好ましくは72~832ppb、さらに好ましくは117~832ppbとされる。
本発明の好ましい実施態様によれば、本発明のビールテイストアルコール飲料中の遊離型有効3成分および結合型有効3成分の計6成分の合計含有量は、499ppb以上、より好ましくは510ppb以上、さらに好ましくは550ppb以上、さらに好ましくは672ppb以上、さらに好ましくは1042ppb以上、さらに好ましくは1203ppb以上とされる。これら6成分の合計含有量が多いビールテイストアルコール飲料では高いコク増強効果や嫌な酸味低減効果、アルコール辛さ低減効果が認められるため、その上限は特に制限されるものではないが、あえて設定するとすれば、例えば、5000ppb、好ましくは4000ppb、より好ましくは2500ppb、さらに好ましくは2350ppb、さらに好ましくは1932ppbとすることができる。この実施態様における遊離型有効3成分の合計含有量および結合型有効3成分の合計含有量のそれぞれの好ましい数値範囲は、上記の段落に記載されている通りである。
本発明の好ましい実施態様によれば、本発明のビールテイストアルコール飲料において、結合型有効3成分の合計含有量(B ppb)に対する遊離型有効3成分の合計含有量(F ppb)の比(F/B)は、0.53以上とされる。本発明のさらに好ましい実施態様によれば、F/Bは0.53~5.05とされ、より好ましくは0.53~3.62とされる。本発明の別のさらに好ましい実施態様によれば、F/Bは6.66以上とされ、より好ましくは8.38以上とされる。遊離型有効3成分の合計含有量が多いビールテイストアルコール飲料では高いコク増強効果や嫌な酸味低減効果が認められるため、F/Bの上限は特に制限されるものではないが、あえて設定するとすれば、例えば、50、好ましくは30、より好ましくは20、さらに好ましくは18、さらに好ましくは15.34とすることができる。この実施態様における遊離型有効3成分の合計含有量および結合型有効3成分の合計含有量のそれぞれの好ましい数値範囲は、上記の段落に記載されている通りである。
ビールテイストアルコール飲料中の遊離型有効3成分および結合型有効3成分の定量は、後述の実施例に記載するLC-MS/MS分析により行うことができる。さらに、より正確な濃度測定のためには、既知の濃度の標準品を添加した幾つかのサンプルの測定値に基づいて作成した検量線を用いることが望ましい。
本発明のビールテイストアルコール飲料中のプリン体濃度は特に限定されるものではないが、一般的な淡色ビールと比較して低減されていることが好ましい。具体的には、本発明のビールテイストアルコール飲料のプリン体濃度は、例えば、5.0mg/100mL以下が挙げられ、これよりも低い濃度(例えば、4.5mg/100mL以下、4.0mg/100mL以下、3.5mg/100mL以下、3.0mg/100mL以下、2.5mg/100mL以下、2.0mg/100mL以下、1.5mg/100mL以下、1.4mg/100mL以下、1.3mg/100mL以下、1.2mg/100mL以下、1.1mg/100mL以下、1.0mg/100mL以下、0.9mg/100mL以下、0.8mg/100mL以下、0.7mg/100mL以下、0.6mg/100mL以下、0.5mg/100mL以下、0.5mg/100mL未満)に設定することが好ましい。なお、プリン体濃度が0.5mg/100mL未満の飲料は「プリン体ゼロ飲料」に対応する。本発明のビールテイストアルコール飲料のプリン体濃度の下限は特に制限されるものではないが、あえて設定するとすれば、例えば、0.001mg/100mL以上、好ましくは0.01mg/100mL以上とすることができる。
プリン体の測定は公知の方法によって行うことができ、例えば、過塩素酸による加水分解後にLC-MS/MS(液体クロマトグラフィー質量分析法)を用いて検出する方法(食衛誌55(2):110-116(2014)参照)により測定することができる。なお、本明細書中、「プリン体濃度」とは、アデニン、キサンチン、グアニン、ヒポキサンチンのプリン体塩基4種の総量を指す。
本発明のビールテイストアルコール飲料におけるプリン体濃度の低減は公知の方法に従って行うことができる。ビールテイストアルコール飲料においてプリン体濃度を低減させる方法の非制限的な例としては、発酵前液や発酵液を活性炭やゼオライト等の吸着剤と接触させて飲料中のプリン体濃度を低減させる方法(特開2003-169658号公報、特開2004-290071号公報、特開2004-290072号公報、特開2015-112090号公報等参照)や、プリン体の持込みが少ない大麦麦芽以外の原料(例えば、小麦麦芽、大豆タンパク、コーングリッツ)を用いて飲料中のプリン体濃度を低減させる方法(特開2014-117204号公報、特開2014-117205号公報、特開2022-140798号公報等参照)が挙げられる。
本発明のビールテイストアルコール飲料中のアルコール濃度は、特に限定されるものではないが、好ましくは1体積%(v/v%)超とされ、より好ましくは2体積%(v/v%)以上とされ、さらに好ましくは3体積%(v/v%)以上とされ、さらに好ましくは3.5体積%以上とされ、さらに好ましくは4体積%以上とされる。ビールテイストアルコール飲料のアルコール濃度の上限は、本発明の効果が奏される限り特に限定されるものではないが、例えば20体積%、好ましくは10体積%、より好ましくは8体積%である。本発明の一つの実施態様によれば、本発明のビールテイストアルコール飲料中のアルコール濃度は、好ましくは2~10体積%以下とされ、より好ましくは3~10体積%以下、さらに好ましくは3~8体積%とされる。
本発明のビールテイストアルコール飲料は、炭酸飲料とすることができる。炭酸ガス圧は好みに応じて適宜調整することができ、例えば、0.05~0.4MPa(20℃におけるガス圧)の範囲で調整することができる。
本発明のビールテイストアルコール飲料は、pHを、例えば、2.0~5.0、好ましくは2.3~4.9、より好ましくは2.9~4.9に調整することができる。飲料のpHは市販のpHメーター(例えば、卓上型pHメーター、堀場製作所)を使用して容易に測定することができる。
本発明のビールテイストアルコール飲料は、好ましくは容器詰飲料として提供される。本発明のビールテイストアルコール飲料に使用される容器は、飲料の充填に通常使用される容器であればよく、例えば、金属缶、樽容器、プラスチック製ボトル(例えば、PETボトル、カップ)、紙容器、瓶、パウチ容器等が挙げられるが、好ましくは金属缶、樽容器、プラスチック製ボトル(例えば、PETボトル)、または瓶とされる。
本発明の一つの実施態様によれば、本発明のビールテイストアルコール飲料は、飲料中の遊離型有効3成分および結合型有効3成分の濃度調整以外は、通常の低糖質ビールテイストアルコール飲料の製造方法に従って製造することができる。通常の製法としては、例えば、少なくとも水および麦芽を含んでなる発酵前液を発酵させる方法、すなわち、麦芽等の醸造原料から調製された麦汁(発酵前液)に発酵用ビール酵母を添加して発酵を行い、所望により発酵液を低温にて貯蔵した後、ろ過工程により酵母を除去する方法が挙げられる。また、発酵工程完了前のいずれかのタイミング(例えば、仕込中、発酵中、あるいは仕込中/発酵中の両方)で、糖質分解酵素、繊維素分解酵素等による分解処理を行ってもよい。糖質分解酵素としては、麦芽等の醸造原料由来酵素、市販酵素剤(αアミラーゼ、βアミラーゼ、グルコアミラーゼ、αグルコシダーゼ、プルナラーゼ、イソアミラーゼ等)などが挙げられる。繊維素分解酵素としては、市販酵素剤(βグルカナーゼ、キシラナーゼ、ヘミセルラーゼ等)が挙げられる。
本発明のビールテイストアルコール飲料の製造過程では、いずれかの工程でホップ(ホップの加工品を含む)を添加することができる。ホップの添加量は、典型的には、発酵工程における発酵前液の容量に対して乾燥毬花換算で0.1~5g/Lとなるように調整することができ、好ましくは0.1~2g/L、より好ましくは0.2~1.5g/Lとすることができる。
本発明では、麦芽、ホップおよび水以外に、米、とうもろこし、こうりゃん、馬鈴薯、でんぷん、糖類(例えば、液糖)、果実、コリアンダー等の酒税法で定める副原料や、タンパク質分解物、酵母エキス等の窒素源、色素、起泡・泡持ち向上剤、水質調整剤、発酵助成剤等のその他の添加物を醸造原料として使用することができる。また、未発芽の麦類(例えば、未発芽大麦(エキス化したものを含む)、未発芽小麦(エキス化したものを含む))を醸造原料として使用してもよい。
本発明の別の態様によれば、低糖質ビールテイストアルコール飲料において、コクを増強する方法が提供され、該方法では、飲料中の遊離型のカルボキシメチルリジン(CML)、遊離型のカルボキシエチルリジン(CEL)および遊離型のメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)の合計含有量が、434ppb以上、好ましくは438ppb以上に調整される。
本発明の別の態様によれば、低糖質ビールテイストアルコール飲料において、嫌な酸味を低減する方法が提供され、該方法では、飲料中の遊離型のカルボキシメチルリジン(CML)、遊離型のカルボキシエチルリジン(CEL)および遊離型のメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)の合計含有量が、434ppb以上、好ましくは438ppb以上に調整される。
本発明の別の態様によれば、低糖質ビールテイストアルコール飲料において、アルコール辛さを低減する方法が提供され、該方法では、飲料中の遊離型のカルボキシメチルリジン(CML)、遊離型のカルボキシエチルリジン(CEL)および遊離型のメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)の合計含有量が、434ppb以上、好ましくは438ppb以上に調整される。
以下の実施例に基づいて本発明を具体的に説明するが、本発明はこれらの例に限定されるものではない。
以下の実施例において示される遊離型メイラード反応産物(F-MRP)の濃度は、遊離型CML、遊離型CELおよび遊離型MG-H1の濃度の合計である。結合型メイラード反応産物(B-MRP)の濃度は、結合型CML、結合型CELおよび結合型MG-H1の濃度の合計である。
例1:ビールテイスト飲料の試験醸造
以下の通り、低糖質(糖質ゼロを含む)・ビールテイスト飲料を試験醸造により調製(製造ともいう)した。粉砕した大麦麦芽を麦芽比率50質量%以上の配合比率となるよう用い、糖質分解酵素、繊維素分解酵素を用いて糖化分解を行った。50~60℃で保持された温水が入った仕込槽に投入した後、段階的に昇温して、温度、時間条件を調整して糖化液を得た。その後、濾過して麦芽粕を除去し、麦汁を得た。得られた麦汁に、ホップを添加して煮沸した後に固液分離処理し、冷却して清澄な麦汁を得た。酵母を添加し、発酵温度及び発酵時間を調整して得られた発酵液を濾過することで、試醸品A~K(サンプル番号1~3、9~11、16、19、23~24、27)、並びに試醸品L~O、P~R及びT~X(サンプル番号30~33、34、49、50、及び52~56)をそれぞれ調製した。試醸品S(サンプル番号51)については、大麦麦芽の一部を小麦麦芽に変更する以外は、上記試醸品A~R及びT~Xと同様に調製した。得られたサンプルはいずれも、アルコール濃度4.6%、pH3.8~4.1であった。また、試醸品A~K及びP~Vの糖質量は0.3~0.4g/100mL、試醸品L~O、W及びXの糖質量は0.6~0.9g/100mLであった。
以下の通り、低糖質(糖質ゼロを含む)・ビールテイスト飲料を試験醸造により調製(製造ともいう)した。粉砕した大麦麦芽を麦芽比率50質量%以上の配合比率となるよう用い、糖質分解酵素、繊維素分解酵素を用いて糖化分解を行った。50~60℃で保持された温水が入った仕込槽に投入した後、段階的に昇温して、温度、時間条件を調整して糖化液を得た。その後、濾過して麦芽粕を除去し、麦汁を得た。得られた麦汁に、ホップを添加して煮沸した後に固液分離処理し、冷却して清澄な麦汁を得た。酵母を添加し、発酵温度及び発酵時間を調整して得られた発酵液を濾過することで、試醸品A~K(サンプル番号1~3、9~11、16、19、23~24、27)、並びに試醸品L~O、P~R及びT~X(サンプル番号30~33、34、49、50、及び52~56)をそれぞれ調製した。試醸品S(サンプル番号51)については、大麦麦芽の一部を小麦麦芽に変更する以外は、上記試醸品A~R及びT~Xと同様に調製した。得られたサンプルはいずれも、アルコール濃度4.6%、pH3.8~4.1であった。また、試醸品A~K及びP~Vの糖質量は0.3~0.4g/100mL、試醸品L~O、W及びXの糖質量は0.6~0.9g/100mLであった。
例2:遊離型および結合型のメイラード反応産物(MRP)の定量
(1)遊離型メイラード反応産物(F-MRP)の定量
遊離型メイラード反応産物画分の定量はLC-MS/MSで行った。液体サンプルまたは凍結乾燥品に超純水を加えて再溶解した。そこに等量の6Nスルホサリチル酸を加えて撹拌し、13,000rpmで5分間遠心分離して得られた上清を採って1/3量の20%(v/v)メタノールと混合した。固相抽出カラム(Bond Elut C18,Agilent Technologies製)に分析サンプルをロードし、続いて等量の10%メタノールをロードすることで、得られた溶出液を合わせて撹拌した。その後、別途準備した標準液(CEL,CML:0,50,100,200ppb/MG-H1:0,100,200,400ppb)と1:1で混合し、以下の条件でLC-MS/MSに供した。得られた2イオンのピーク面積を使い、標準添加法でサンプル中のメイラード反応産物濃度を算出した。
(1)遊離型メイラード反応産物(F-MRP)の定量
遊離型メイラード反応産物画分の定量はLC-MS/MSで行った。液体サンプルまたは凍結乾燥品に超純水を加えて再溶解した。そこに等量の6Nスルホサリチル酸を加えて撹拌し、13,000rpmで5分間遠心分離して得られた上清を採って1/3量の20%(v/v)メタノールと混合した。固相抽出カラム(Bond Elut C18,Agilent Technologies製)に分析サンプルをロードし、続いて等量の10%メタノールをロードすることで、得られた溶出液を合わせて撹拌した。その後、別途準備した標準液(CEL,CML:0,50,100,200ppb/MG-H1:0,100,200,400ppb)と1:1で混合し、以下の条件でLC-MS/MSに供した。得られた2イオンのピーク面積を使い、標準添加法でサンプル中のメイラード反応産物濃度を算出した。
(2)ゲル濾過分画
サンプルを0.45μmフィルターで濾過し、濾過済み発酵液を計量して凍結乾燥した。乾燥物を100mM NaCl溶液で溶解して5倍濃縮液を調製した。得られた濃縮液を以下の条件でゲル濾過分画を行い、0.66CV(カラムボリューム)~0.86CVの画分を分取して分子量分析および結合型メイラード反応産物の定量に供した。
サンプルを0.45μmフィルターで濾過し、濾過済み発酵液を計量して凍結乾燥した。乾燥物を100mM NaCl溶液で溶解して5倍濃縮液を調製した。得られた濃縮液を以下の条件でゲル濾過分画を行い、0.66CV(カラムボリューム)~0.86CVの画分を分取して分子量分析および結合型メイラード反応産物の定量に供した。
(3)HPLCゲル濾過法による分子量分析
(2)で分取した画分は、50mMリン酸ナトリウム緩衝液(pH7.0 150mM NaCl含む)となるよう溶解した。これを分子量分析サンプルとして、以下の方法でHPLCゲル濾過を行った。

得られたクロマトグラムから分子量を算出するため、分子量既知のペプチドを0.1~5mg/mLで適宜超純水に溶解したものを50μL注入して同様の条件でHPLCゲル濾過分析を行い、保持時間を確認した(表5)。その保持時間、分子量から検量線を作成し、(2)で分取した画分が示すクロマトグラムにおける主要なピークの始点から終点の保持時間に基づいて分子量範囲を求めた。その結果、当該画分の分子量は400~3,000Daであることがわかった。
(4)結合型メイラード反応産物(B-MRP)の定量
結合型メイラード反応産物の定量は、サンプル中のペプチドやタンパク質を酵素分解し、酵素ブランクの遊離型メイラード反応産物をLC-MS/MSで分析し、酵素ブランクとの差分を結合型メイラード反応産物量とした。サンプル処理は以下の通り行った。

(2)で分取した画分を1kDaの透析膜で24時間透析し、内液を凍結乾燥した。その後、ペプシンを含む0.02M塩酸に溶解し、37℃で24時間反応した。続いて、プロナーゼEを含有するTris緩衝液(pH8.2)を加えて混合し、37℃で24時間反応した。さらに、アミノペプチダーゼMおよびプロリダーゼを加えて混合し、37℃で24時間反応することで、ペプチド結合を切断した。この酵素反応時に酵素を含まない緩衝液を添加するものをブランクとして、同様に37℃24時間の反応を3回実施した。サンプルを凍結乾燥し、超純水を加えて再溶解した。そこに等量の6Nスルホサリチル酸を加えて撹拌し、13,000rpmで5分間遠心分離して得られた上清を採って1/3量の20%(v/v)メタノールと混合した。固相抽出カラム(Bond Elut C18)に分析サンプルをロードし、続いて等量の10%メタノールをロードすることで、得られた溶出液を合わせて撹拌した。その後、別途準備した標準液(CEL,CML:0,50,100,200ppb/MG-H1:0,100,200,400ppb)と1:1で混合し、(1)と同様の条件でLC-MS/MSに供することで、標準添加法によりサンプル中の結合型メイラード反応産物濃度を算出した。
例3:精製MRP画分の添加試飲
(1)メイラード反応産物画分の精製
例1に記載の「コクの強度」官能評価スコアが4.2以上(下記の評価基準に従って評価したときのスコア)であったサンプルから、例2(2)の方法にて得た分取画分を、C18固相抽出カラム(Bond Elut C18)に吸着させた。純水で洗浄した後、得られた素通り画分および洗浄液をさらにダイヤイオンHP20(三菱ケミカル製)カラムに吸着させ、純水で洗浄処理を行った。C18固相抽出カラムの吸着物は、50%(v/v)エタノール水溶液にて溶出した。C18固相抽出カラムの溶出液は濃縮乾固を行い、これを純水にて復水することで、結合型メイラード反応産物の精製物とした。
(1)メイラード反応産物画分の精製
例1に記載の「コクの強度」官能評価スコアが4.2以上(下記の評価基準に従って評価したときのスコア)であったサンプルから、例2(2)の方法にて得た分取画分を、C18固相抽出カラム(Bond Elut C18)に吸着させた。純水で洗浄した後、得られた素通り画分および洗浄液をさらにダイヤイオンHP20(三菱ケミカル製)カラムに吸着させ、純水で洗浄処理を行った。C18固相抽出カラムの吸着物は、50%(v/v)エタノール水溶液にて溶出した。C18固相抽出カラムの溶出液は濃縮乾固を行い、これを純水にて復水することで、結合型メイラード反応産物の精製物とした。
また、上記のダイヤイオンHP20カラムの素通り画分は、凍結乾燥濃縮を行い、分子量100~500Daの透析膜にて約10時間、NaClによる電気伝導度が低下するまで透析を行った。さらに透析外液を交換して20時間透析を行った。NaClを除去した後の透析外液を凍結乾燥した後、これを純水にて復水し、濃縮液とした。これを遊離型メイラード反応産物の精製物とした。
各精製物に含まれる結合型メイラード反応産物(B-MRP)、および遊離型メイラード反応産物(F-MRP)は、それぞれ例2に記載の方法で定量し、添加試飲に用いた。
(2)精製MRP画分の添加試飲
例1で調製した試醸品B、および試醸品C~K(サンプル番号3、11、16、19、24、27)に対して、精製MRPを下記の表に示す濃度となるように添加して各サンプルを得た(サンプル番号4~8、12~15、17~18、20~22、25~26、28~29)。得られた試飲サンプルについて、試醸品A(サンプル番号1)を対照として、訓練された5名のパネルにより官能評価を行った。評価項目および評価基準は以下の通りとした。
例1で調製した試醸品B、および試醸品C~K(サンプル番号3、11、16、19、24、27)に対して、精製MRPを下記の表に示す濃度となるように添加して各サンプルを得た(サンプル番号4~8、12~15、17~18、20~22、25~26、28~29)。得られた試飲サンプルについて、試醸品A(サンプル番号1)を対照として、訓練された5名のパネルにより官能評価を行った。評価項目および評価基準は以下の通りとした。
評価項目1として、コクの強さ(味の強さ、複雑さ、持続性から成る総合評価)を1点(弱い)~9点(強い)の9段階で評価した。また、評価項目2として、「嫌な酸味」(後味などに感じる不快な酸味)を1点(弱い)~9点(強い)の9段階で評価した。官能評価に際しては、例1で調製した試醸品Aを対照(コクの強さ: 2.4、嫌な酸味: 5.0と定義)とした。
評価結果を以下の表に示す。
対照である試醸品A、および試醸品Bは、「コクの強さ」のスコアが3未満、「嫌な酸味」のスコアが5.0以上となり、コクがとても弱く、嫌な酸味が感じられた。「コクの強さ」のスコアが4.2以上、「嫌な酸味」のスコアが4.8以下のサンプルは、コクを感じ、嫌な酸味が弱く感じられた。さらに「コクの強さ」のスコアが5.4以上、「嫌な酸味」のスコアが4.0以下のサンプルは、コクを強く感じ、嫌な酸味が弱く感じられた。これらサンプルは、ビールテイスト飲料として良好な香味であると考えられた。従って、ビールテイスト飲料の香味として、「コクの強さ」のスコアが4.2以上であることが好ましく、5.4以上であることがより好ましいものと評価した。
MRP濃度と「コクの強さ」評価結果との関係を示すプロット図(縦軸:結合型MRP濃度、横軸:遊離型MRP濃度、バブルサイズ:コクの強さ)を図1に示す。この図から、ビールテイストアルコール飲料中の遊離型MRPの合計含有量は、438ppb以上が好ましいことがわかった。また、結合型MRPの合計含有量は限定する必要はないが、72ppb以上であることが好ましく、117ppb以上がさらに好ましいことがわかった。
次に、上記の評価結果を、F-MRPとB-MRPの合計量で並べ替えて表記した結果を以下の表に示す。
MRP濃度と「コクの強さ」評価結果との関係を示すプロット図(縦軸:結合型MRP濃度、横軸:遊離型MRP濃度、バブルサイズ:コクの強さ、斜め補助線:F-MRPとB-MRPの合計量の境界線)を図2に示す。この図から、F-MRP438ppb以上の範囲において、ビールテイストアルコール飲料中のF-MRPとB-MRPとの合計量は、510ppb以上のとき、「コクの強さ」のスコアが4.2以上、「嫌な酸味」のスコアが4.8以下となり、コクを感じ、嫌な酸味が弱く感じられ、ビールテイスト飲料として良好な香味であると考えられた。また、F-MRPとB-MRPとの合計量が550ppb以上のとき、「コクの強さ」のスコアが4.2以上、「嫌な酸味」のスコアが4.6以下となり、コクを感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。さらに、F-MRPとB-MRPとの合計量が672ppb以上のとき、「コクの強さ」のスコアが4.4以上、「嫌な酸味」のスコアが4.2以下となり、コクを感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。さらに、F-MRPとB-MRPとの合計量が1,042ppb以上のとき、「コクの強さ」のスコアが5.2以上、「嫌な酸味」のスコアが3.6以下となり、コクを強く感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。さらに、F-MRPとB-MRPとの合計量が1,203ppb以上のとき、「コクの強さ」のスコアが6.4以上、「嫌な酸味」のスコアが3.6以下となり、コクを強く感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。
次に、上記の評価結果を、F-MRPとB-MRPの比率(F-MRP/B-MRP)で並べ替えて表記した結果を以下の表に示す。
MRP濃度と「コクの強さ」評価結果との関係を示すプロット図(縦軸:結合型MRP濃度、横軸:遊離型MRP濃度、バブルサイズ:コクの強さ、斜め補助線:F-MRP/B-MRP比の境界線)を図3に示す。この図から、F-MRP438ppb以上の範囲において、ビールテイストアルコール飲料中のF-MRP/B-MRP比が5.06~6.65のとき、「コクの強さ」のスコアが4.2~4.8、「嫌な酸味」のスコアが4.8~4.0となり、コクを感じ、嫌な酸味が弱く感じられ、ビールテイスト飲料として良好な香味であると考えられた。さらに、F-MRP/B-MRP比が6.66~8.37のとき、「コクの強さ」のスコアが4.2~5.8、「嫌な酸味」のスコアが4.6~3.6となり、コクを感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。さらに、F-MRP/B-MRP比が3.63~5.05のとき、「コクの強さ」のスコアが6.0~6.4、「嫌な酸味」のスコアが3.6~3.2となり、コクを感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。さらに、F-MRP/B-MRP比が0.53~3.62のとき、「コクの強さ」のスコアが5.4~8.2、「嫌な酸味」のスコアが4.0~2.6となり、コクを強く感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。さらに、F-MRP/B-MRP比が8.38以上のとき、「コクの強さ」のスコアが5.0~6.4、「嫌な酸味」のスコアが3.8~3.0となり、コクを強く感じ、嫌な酸味が弱く感じられ、さらに好ましいと考えられた。
(3)糖質1g/100mL以下のビールテイスト飲料の官能評価
例1で調製した試醸品Aを対照として、糖質1g/100mL以下のビールテイスト飲料試醸品L~O(サンプル番号30~33)を、訓練された5名のパネルにより官能評価を行った。評価項目および評価基準は上述の通りとした。

上記(2)と同様の数値範囲で、上述した評価結果と同様のコクの増強、嫌な酸味の抑制が確認されたことから、糖質1g/100mL以下のビールテイスト飲料に対しても、同等の効果が期待できるものと考えられた。
(4)B-MRP濃度の和が等しい、個々の成分の組成が異なるサンプルの評価
F-MRPないしB-MRP濃度の和がほぼ等しいが、個々の成分の組成が異なるサンプルの組合せと官能評価の結果を以下の表に示した。

サンプル9は、サンプル10と比較して結合型のCELおよびMG-H1が少なく、CMLが多くなっているが、3成分の合計量とコクの強さおよび嫌な酸味は同程度であった。
以上の結果から、有効3成分は、個々の構成成分の組成によらず、合計量が同等であれば同等の香味上の効果が得られるものと考えられた。
(5)プリン体濃度の測定
例1で調製した試醸品E、Jのプリン体濃度の測定を行った。具体的には、食衛誌55(2):110-116(2014)を参考に、サンプルを過塩素酸で分解し、LC-MS/MSによりプリン体濃度(mg/100mL)を測定した。なお、LC-MS/MSは上記文献の測定方法IIを参考に行った。測定結果を以下の表に示す。
例1で調製した試醸品E、Jのプリン体濃度の測定を行った。具体的には、食衛誌55(2):110-116(2014)を参考に、サンプルを過塩素酸で分解し、LC-MS/MSによりプリン体濃度(mg/100mL)を測定した。なお、LC-MS/MSは上記文献の測定方法IIを参考に行った。測定結果を以下の表に示す。
サンプル10とサンプル24のプリン体濃度は同程度であった。
例4:精製MRP画分の添加試飲(2)
(1)メイラード反応産物画分の精製
結合型メイラード反応産物の精製物および遊離型メイラード反応産物の精製物は、例3(1)に記載の精製物を用いた。

結合型メイラード反応産物の精製物および遊離型メイラード反応産物の精製物は、例3(1)に記載の精製物を用いた。
(2)精製MRP画分の添加試飲
例1で調製した試醸品P(サンプル34)に対して、精製MRPを下記の表に示す濃度となるように添加して各サンプルを得た(サンプル35~48)。なお、サンプル39とサンプル40は、個々の成分の組成が異なる遊離型MRPを、遊離型MRP濃度として同濃度となるよう添加した。得られた試飲サンプルについて、試醸品P(サンプル34)を対照として、訓練された6名のパネルにより官能評価を行った。評価項目および評価基準は以下の通りとした。
例1で調製した試醸品P(サンプル34)に対して、精製MRPを下記の表に示す濃度となるように添加して各サンプルを得た(サンプル35~48)。なお、サンプル39とサンプル40は、個々の成分の組成が異なる遊離型MRPを、遊離型MRP濃度として同濃度となるよう添加した。得られた試飲サンプルについて、試醸品P(サンプル34)を対照として、訓練された6名のパネルにより官能評価を行った。評価項目および評価基準は以下の通りとした。
評価項目1として、例3(2)と同様、コクの強さ(味の強さ、複雑さ、持続性から成る総合評価)を1点(弱い)~9点(強い)の9段階で評価した。また、評価項目2として、例3(2)と同様、「嫌な酸味」(後味などに感じる不快な酸味)を1点(弱い)~9点(強い)の9段階で評価した。さらに、評価項目3として、「アルコール辛さ」(ピリピリとした刺激を伴う辛さ)を1点(アルコール辛さがほとんどない)~9点(アルコール辛さが強い)の9段階で評価した。官能評価に際しては、例1で調製した試醸品P(サンプル34)を対照(コクの強さ: 2.5、嫌な酸味: 5.2、アルコール辛さ: 5.2と定義)とした。
評価結果を以下の表に示す。
対照である試醸品P(サンプル34)は、「コクの強さ」のスコアが3未満、「嫌な酸味」のスコアが5.0以上、「アルコール辛さ」のスコアが5.0以上となり、コクがとても弱く、嫌な酸味が感じられ、アルコール辛さが感じられた。「コクの強さ」のスコアが4.0以上、「嫌な酸味」のスコアが4.8以下のサンプルは、コクを感じ、嫌な酸味が弱く感じられた。また、「アルコール辛さ」のスコアが4.0以下のサンプルは、アルコール辛さが弱く感じられた。これらサンプルは、ビールテイスト飲料として良好な香味であると考えられた。従って、ビールテイスト飲料の香味として、「コクの強さ」のスコアが4.0以上である場合も好ましいと評価できる。
(3)遊離型MRP(F-MRP)濃度が等しく、個々の成分の組成が異なるサンプルの評価
例4(2)におけるサンプル39とサンプル40はF-MRP濃度は等しいが、個々の成分の組成が異なるサンプルであり、その組成を以下の表に示した。

サンプル39は、サンプル40と比較して遊離型のCELおよびCMLが少なく、MG-H1が多くなっている。一方、表12に示されるように、3成分の合計量とコクの強さ、嫌な酸味およびアルコール辛さは同程度であった。
以上の結果から、有効3成分は、個々の構成成分の組成によらず、合計量が同等であれば同等の香味上の効果が得られるものと考えられた。
例5:MRP濃度の異なるビールテイスト飲料試醸品Qの試飲
(1)ビールテイスト飲料の試飲
例1で調製した試醸品Qを試飲サンプルとして、例4と同様の方法により官能評価を行った。評価項目および評価基準は例4と同様である。

上記例4(2)と同様の数値範囲で、上述した評価結果と同様のコクの増強、嫌な酸味の抑制、アルコール辛さの抑制が確認されたことから、MRP濃度の異なる試醸品の試飲サンプルに対しても、同等の効果が期待できるものと考えられた。
例6:プリン体濃度の異なるビールテイスト飲料試醸品R~Yの試飲
(1)プリン体濃度の測定
例1で調製した試醸品R~Xのプリン体濃度の測定を、例3(5)に記載のプリン体濃度の測定と同様に行った。測定結果を以下の表に示す。

例1で調製した試醸品P(サンプル34)を対照として、プリン体濃度1.5mg/100mL以下のビールテイスト飲料試醸品R~X(サンプル番号50~56)を試飲サンプルとして、例4と同様の方法により官能評価を行った。評価項目および評価基準は例4と同様である。評価結果を以下の表に示す。
例1で調製した試醸品P(サンプル34)を対照として、プリン体濃度1.5mg/100mL以下のビールテイスト飲料試醸品R~X(サンプル番号50~56)を試飲サンプルとして、例4と同様の方法により官能評価を行った。評価項目および評価基準は例4と同様である。評価結果を以下の表に示す。
例4(2)で記載のように、対照である試醸品P(サンプル34)は、「コクの強さ」のスコアが3未満、「嫌な酸味」のスコアが5.0以上、「アルコール辛さ」のスコアが5.0以上となり、コクがとても弱く、嫌な酸味が感じられ、アルコール辛さが感じられた。一方、サンプル50~56は、「コクの強さ」のスコアが5.3以上、「嫌な酸味」のスコアが4.0以下であり、コクを強く感じ、嫌な酸味が弱く感じられた。また、「アルコール辛さ」のスコアが4.0以下のサンプルは、アルコール辛さが弱く感じられた。したがって、ビールテイスト飲料として良好な香味であると考えられ、プリン体濃度1.5mg/100mL以下のビールテイスト飲料に対しても、同等の効果が期待できるものと考えられた。また、上記サンプル番号50~56は、ビールテイストアルコール飲料中のF-MRP438ppb以上であり、F-MRP/B-MRP比が1.95~2.07であり、「コクの強さ」のスコアが5.3~5.7、「嫌な酸味」のスコアが3.3~3.9となり、コクを強く感じ、嫌な酸味が弱く感じられ、例3(2)と同様、ビールテイスト飲料として、さらに好ましいと考えられた。従って、ビールテイスト飲料の香味として、「コクの強さ」のスコアが5.3以上である場合もさらに好ましいと評価できる。
Claims (14)
- 飲料中の遊離型のカルボキシメチルリジン(CML)、遊離型のカルボキシエチルリジン(CEL)および遊離型のメチルグリオキサール由来ヒドロイミダゾロン-1(MG-H1)の合計含有量が434ppb以上である、低糖質ビールテイストアルコール飲料。
- 飲料中の結合型CML、結合型CELおよび結合型MG-H1の合計含有量が65ppb以上であり、
前記結合型のCML、結合型のCELおよび結合型のMG-H1が、分子量400~3,000Daのペプチド画分に含まれるものである、請求項1に記載の低糖質ビールテイストアルコール飲料。 - 飲料中の結合型CML、結合型CELおよび結合型MG-H1の合計含有量が72ppb以上である、請求項2に記載の低糖質ビールテイストアルコール飲料。
- 飲料中の遊離型CML、遊離型CEL、遊離型MG-H1、結合型CML、結合型CELおよび結合型MG-H1の合計含有量が499ppb以上であり、
前記結合型のCML、結合型のCELおよび結合型のMG-H1が、分子量400~3,000Daのペプチド画分に含まれるものである、請求項1に記載の低糖質ビールテイストアルコール飲料。 - 飲料中の遊離型CML、遊離型CEL、遊離型MG-H1、結合型CML、結合型CELおよび結合型MG-H1の合計含有量が510ppb以上である、請求項4に記載の低糖質ビールテイストアルコール飲料。
- 飲料中の遊離型CML、遊離型CEL、遊離型MG-H1、結合型CML、結合型CELおよび結合型MG-H1の合計含有量が1042ppb以上である、請求項4に記載の低糖質ビールテイストアルコール飲料。
- 飲料中の結合型CML、結合型CELおよび結合型MG-H1の合計含有量(B ppb)に対する遊離型CML、遊離型CELおよび遊離型MG-H1の合計含有量(F ppb)の比(F/B)が0.53以上であり、
前記結合型のCML、結合型のCELおよび結合型のMG-H1が、分子量400~3,000Daのペプチド画分に含まれるものである、請求項1に記載の低糖質ビールテイストアルコール飲料。 - F/Bが0.53~3.63である、請求項7に記載の低糖質ビールテイストアルコール飲料。
