Description

최적화된 약물 전달 나노입자의 생산을 위한 연속 흐름기반 통합형 인공지능 미세유체 플랫폼
본 발명은 인공지능 기반 최적화된 약물 전달용 나노입자의 제조를 위해 인공지능 알고리즘 기반 통합형 미세유체 연속 흐름 플랫폼에 관한 것이다.
약물 전달 시스템(Drug delivery systems, DDS)은 의학 분야에서 중요한 도구로 등장하여 치료제의 효과적이고 표적화된 전달에 중추적인 역할을 하고 있다. 이러한 시스템은 약물 생체 이용률 향상, 부작용 감소 및 치료 효능 향상과 같은 기존 약물 투여 방법에 비해 많은 이점을 제공한다. 약물 전달 시스템은 특수 담체 또는 운반체 내에 약물을 캡슐화함으로써 치료제의 분해를 방지하고 순환 시간을 연장하며 원하는 작용 부위에 약물의 축적을 촉진할 수 있다.
약물 전달 시스템의 개발에서는 다양한 접근법이 합성을 위해 사용되었으며, 그 중에서도 미세유체 접근법이 상당한 주목을 받고 있다. 미세유체공학(Microfluidics)은 미세 규모에서 유체를 조작하고 제어하는 분야로, 약물의 합성 및 제작을 정밀하게 제어할 수 있다. 미세유체 접근법은 액적 크기와 균일성을 정밀하게 제어할 수 있어 다양한 담체 물질 내에 소수성 및 친수성 약물을 효율적으로 캡슐화할 수 있을 뿐만 아니라 기존 접근법에 비해 확장성과 재현성이 향상된다.
약물 전달 시스템의 잠재력을 최대한 활용하려면 약물이 담지(loading)된 운반체의 합성 및 특성화를 위한 효율적인 방법을 개발하는 것이 필수적이다. 이러한 나노입자는 약물을 캡슐화하고 신체의 특정 표적 부위에 전달하는 운반체 역할을 하여 방출을 제어하고 치료 결과를 개선한다.
한편, 화학, 재료과학 등 화학 실험 분야 역시 자율주행 기술의 등장으로 상당한 발전을 이루었다. 자율주행 연구는 자동화, 로봇공학, 인공지능(AI) 기술을 통합하여 최소한의 인간 개입으로 화학 실험을 수행하는 것에 목적이 있다. 기존 실험 과정에는 화학 물질의 수동 처리, 반복 작업, 주관적인 의사 결정이 포함되는 경우가 많으며, 이는 시간이 많이 걸리고 오류가 발생하기 쉬우며 리소스 집약적일 수 있다.
따라서, 약물이 담지된 나노입자의 합성 및 특성화를 위해 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조를 위한 통합형 미세유체 연속 흐름 플랫폼을 구현함으로써 이러한 한계를 극복할 수 있으며, 수많은 이점을 제공할 것이다.
본 발명의 목적은 약물 전달용 나노입자의 최적화 및 자율적인 제조를 위한 인공지능 기반 통합형 미세유체 연속 흐름 플랫폼을 제공하는 것이다.

본 발명의 일 실시예에 따른 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조를 위한 통합형 미세유체 연속 흐름 플랫폼에 있어서, 대상 약물이 담지된 나노입자를 합성하는 합성 모듈; 상기 합성 모듈로부터 유입되는 상기 나노입자와 상기 나노입자에 담지되지 않은 약물을 분리하는 정제 모듈; 상기 정제 모듈을 통해 분리된 상기 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양을 측정하는 실시간 분석 장치; 및 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 담지양에 따른 상기 합성 모듈에 대한 복수의 파라미터 값을 결정하는 실시간 분석용 센서, 모듈 및 장치를 포함할 수 있다.
상기 합성 모듈은, 총 유량, 대상 약물과 지질의 유량 비율, 음이온성 지질 농도, POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 지질과 콜레스테롤의 비율 및 대상 약물의 농도를 포함하는 상기 복수의 파라미터 값을 이용하여 상기 나노입자를 합성할 수 있다.
상기 실시간 분석 장치는, 상기 나노입자의 크기를 측정하는 제1실시간 분석 장치; 및 상기 나노입자에 담지된 대상 약물의 양을 측정하는 제2실시간 분석 장치를 포함할 수 있다.

상기 전자장치는, 초기 대상 약물의 양 대비 상기 나노입자에 담지된 대상 약물의 양을 나타내는 캡슐화 효율성, 및 초기 지질의 양 대비 상기 나노입자에 담지된 대상 약물의 양을 나타내는 예상 로딩 용량을 연산할 수 있다.
상기 전자장치는, 캡슐화 효율성과 예상 로딩 용량을 최대화하는 동시에 측정된 상기 나노입자의 크기와 타겟 크기 간의 차이를 최소화하도록 하는 상기 파라미터 값을 결정할 수 있다.
상기 전자장치는, 결정된 상기 파라미터 값에 기초하여 상기 나노입자를 합성하도록 상기 합성 모듈을 제어할 수 있다.
본 발명의 일 실시예에 따른 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조를 위한 전자장치에 있어서, 합성 모듈에 의해 합성된 대상 약물이 담지된 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신하고, 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 양에 따른 상기 합성 모듈에 대한 복수의 파라미터의 파라미터 값을 결정하는 프로세서;를 포함할 수 있다.
상기 나노입자는, 상기 합성 모듈로부터 유입되어 정제모듈을 통해 상기 나노입자에 담지되지 않은 약물과 분리된 것을 특징으로 한다.
상기 프로세서는, 실시간 분석 장치로부터 측정된 상기 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신할 수 있다.
상기 복수의 파라미터는, 총 유량, 대상 약물과 지질의 유량 비율, 음이온성 지질 농도, POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 지질과 콜레스테롤의 비율 및 대상 약물의 농도를 포함할 수 있다.
본 발명의 일 실시에에 따른 통합형 미세유체 연속 흐름 플랫폼을 이용한 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조 방법에 있어서, 복수의 파라미터에 기초하여 대상 약물이 담지된 나노입자를 합성하는 단계; 상기 합성 모듈로부터 유입되는 상기 나노입자와 상기 나노입자에 담지되지 않은 약물을 분리하는 단계; 분리된 상기 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양을 측정하는 단계; 및 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 양에 따른 상기 복수의 파라미터의 파라미터 값을 결정하는 단계를 포함할 수 있다.
본 발명의 일 실시예에 따르면, 향상된 치료 결과와 함께 고도로 최적화되고 효과적인 약물 담지 나노입자의 개발을 촉진함으로써 약물 전달 분야를 크게 발전시킬 수 있는 잠재력을 가지는 통합형 미세유체 연속 흐름 플랫폼을 제공할 수 있다.
본 발명의 일 실시예에 따르면, 개인 맞춤형 치료 솔루션에 대한 수요 증가에 맞춰 나노의학 합성의 패러다임 전환을 제시하고 암 전이와 관련된 맞춤형 의학부터 효율적인 다기관 전달에 이르기까지 다양한 의료 시나리오를 해결하기 위한 역동적인 접근 방식을 제공할 수 있다.
본 발명의 일 실시예에 따르면, 통합형 미세유체 연속 흐름 플랫폼에서 한번의 실험을 수행함에 있어 기존 시스템과 비교하여 아주 적은 시간이 소요되며, 실험당 적은 양의 샘플을 사용하기 때문에 효율적이다.

이하, 본 발명에 따른 바람직한 실시 형태를 첨부된 도면을 참조하여 상세하게 설명한다. 첨부된 도면과 함께 이하에 개시될 상세한 설명은 본 발명의 예시적인 실시형태를 설명하고자 하는 것이며, 본 발명이 실시될 수 있는 유일한 실시형태를 나타내고자 하는 것이 아니다. 도면에서 본 발명을 명확하게 설명하기 위해서 설명과 관계없는 부분은 생략할 수 있고, 명세서 전체를 통하여 동일 또는 유사한 구성 요소에 대해서는 동일한 참조 부호를 사용할 수 있다.
도 1은 본 발명의 일 실시예에 따른 약물 전달용 나노입자의 자동 및 자율 제조를 위한 통합형 미세유체 연속 흐름 플랫폼을 도시한 개략도이다.
본 발명의 일 실시예에 따르면, 통합형 미세유체 연속 흐름 플랫폼(1)(이하, 플랫폼(1)이라고도 한다.)은 나노입자 합성 모듈(10), 정제 모듈(20), 실시간 분석 장치(30) 및 전자장치(100)(분석용 전자장치(100)라고도 한다.)를 포함할 수 있다.
나노입자 합성 모듈(10)(이하, 합성 모듈(10)이라고도 한다.)은 대상 약물이 담지(loading)된 운반체(carrier), 즉 나노입자를 합성하는 장치로, 나노입자를 합성하는데 필요한 복수의 파라미터들을 조절함으로써 최적화된 나노입자를 합성할 수 있다. 합성 모듈(10)은 다양한 조합의 물질간 반응에 의한 자율화 나노입자 합성 모듈(Autonomous Nanoparticle Synthesizer, ANS)이라고도 한다.
이때, 나노입자는 대상 약물을 캡슐화하여 신체의 특정 표적 부위에 전달하는 운반체 역할을 하는 것으로, 다양한 유형으로 형성될 수 있다.
나노입자는, 예를 들어, 지질 이중층으로 구성된 소포 구조로 된 리포솜 형태로 형성될 수 있다. 리포솜은 친수성 및 소수성 약물을 캡슐화할 수 있는 수성 코어가 있는 속이 빈 구체를 형성한다. 리포솜은 고유한 생체 적합성, 약물 생체 이용률을 향상시키는 능력, 약물 방출 동역학을 제어하기 위해 정밀하게 조작할 수 있는 능력을 포함하여 약물 운반체로서 매우 바람직하게 만드는 몇 가지 주요 이점을 가지고 있다. 다만 이 외에도 나노입자는 고분자 나노입자(PEG-PLGA)일 수 있으며, 어느 하나에 제한되지 않는다.
정제 모듈(20)은 합성 모듈(10)로부터 유입되는 나노입자와 나노입자에 담지되지 않은 약물(자유 약물이라고도 한다.)을 분리하는 장치이다. 정제 모듈(20)은 합성 모듈(10)과 연결될 수 있으며, 나노입자에 담지된 대상 약물과 자유 약물을 분리하여 나노입자 및 나노입자에 담지된 대상 약물을 분석하기 용이하게 한다. 정제 모듈(20)은 유동 투석 모듈(flow dialysis module)이라고도 한다.
실시간 분석 장치(30)는 정제 모듈(20)을 통해 분리된 나노입자의 크기 및 나노입자에 담지된 대상 약물의 양을 측정하는 장치이다. 실시간 분석 장치(30)는 나노입자의 크기를 측정하는 제1실시간 분석 장치 및 나노입자에 담지된 대상 약물의 양을 측정하는 제2실시간 분석 장치를 포함할 수 있다. 제2실시간 분석 장치는, 나노입자를 파괴하는 용액을 이용하여 파괴된 나노입자로부터 담지된 대상 약물의 양을 측정할 수 있다.
전자장치(100)는 합성 모듈(10)이 나노입자를 합성하는데 필요한 복수의 파라미터의 파라미터 값을 결정하고, 제어하는 장치로, 컴퓨터, 서버, 스마트 폰, 태블릿 PC, 스마트 패드, 노트북 등으로 구현될 수 있다.
전자장치(100)는 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 나노입자의 크기 및 대상 약물의 양에 따른 합성 모듈(10)에 대한 복수의 파라미터의 파라미터 값을 결정할 수 있다.
본 발명에서는 약물 전달을 위한 약물이 담지된 나노입자의 합성 및 특성화를 위해 맞춤화된 완전 자동화된 플랫폼을 제안한다.
이때, AI 알고리즘을 통합하면 지능적인 의사결정과 적응형 실험이 가능해지고, 시약 분배, 용액 혼합, 반응 모니터링과 같은 일반적인 작업을 자동화하여 인적 오류를 줄이고 더 빠르고 정확한 실험을 가능하게 한다.
AI 기반 플랫폼(1)은 방대한 양의 데이터를 분석하고 패턴을 식별하며 실험 조건을 최적화하여 원하는 결과를 얻을 수 있다. 또한, AI 기반 플랫폼(1)은 담체 구성, 약물 대 담체 비율, 합성 조건 등 다양한 파라미터를 분석하여 나노입자의 형성을 최적화할 수 있다. 플랫폼(1)은 기계 학습 알고리즘을 활용하여 나노입자 특성을 반복적으로 최적화하여 원하는 물리적 특성과 향상된 약물 로딩을 달성할 수 있다. 이러한 반복적인 최적화 프로세스는 기존 접근 방식과 관련된 시행착오를 크게 줄여 효율적인 나노입자 기반 약물 전달 플랫폼(1)의 개발을 가속화할 수 있다.
이하, 도면들을 참조하여 본 발명의 일 실시예에 따른 플랫폼(1)의 구성 및 동작에 대해 구체적으로 설명한다.

본 발명의 일 실시예에 따른 전자장치(100)는 입력부(110), 통신부(120), 표시부(130), 저장부(140) 및 프로세서(150)를 포함할 수 있다.
입력부(110)는 전자장치(100)의 사용자 입력에 대응하여 입력데이터를 발생시킨다. 예를 들어, 사용자 입력은 전자장치(100)의 동작을 시작하게 하는 사용자 입력, 합성 모듈(10)의 초기 파라미터 값을 설정하는 사용자 입력, 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 설정하기 위한 사용자 입력, 타겟 나노입자 크기를 설정하는 사용자 입력 등일 수 있으며, 이 외에도 자동화된 통합형 미세유체 연속 흐름 플랫폼(1)을 구축하기 위해 필요한 사용자 입력인 경우 제한하지 않고 적용 가능하다.
입력부(110)는 적어도 하나의 입력수단을 포함한다. 입력부(110)는 키보드(key board), 키패드(key pad), 돔 스위치(dome switch), 터치패널(touch panel), 터치 키(touch key), 마우스(mouse), 메뉴 버튼(menu button) 등을 포함할 수 있다.
통신부(120)는 실시간 분석 장치(30)가 측정한 나노입자의 크기 및 나노입자에 담지된 대상 약물의 양, 새로 결정된 파라미터 값 등을 송수신하기 위해 합성 모듈(10), 실시간 분석 장치(30) 등 외부장치와의 통신을 수행할 수 있다.
이를 위해, 통신부(120)는 5G(5th generation communication), LTE-A(Long Term Evolution-Advanced), LTE(Long Term Evolution), Wi-Fi(Wireless Fidelity), Bluetooth 등의 무선 통신, 혹은 LAN(Local Area Network), WAN(Wide Area Network), 전력선 통신 등의 유선 통신을 수행할 수 있다.
표시부(130)는 전자장치(100)의 동작에 따른 표시 데이터를 표시한다. 표시부(130)는 합성 모듈(10), 정제 모듈(20) 및 실시간 분석 장치(30) 등의 동작 상태 등을 표시하는 화면, 다목적 베이지안 최적화 알고리즘에 따라 결정된 파라미터 값을 표시하는 화면, 사용자 입력을 수신하는 화면 등을 표시할 수 있다.
표시부(130)는 액정 디스플레이(Liquid Crystal Display, LCD), 발광 다이오드(Light Emitting Diode, LED) 디스플레이, 유기 발광 다이오드(Organic LED, OLED) 디스플레이, 마이크로 전자기계 시스템(Micro Electro Mechanical Systems. MEMS) 디스플레이 및 전자 종이(electronic paper) 디스플레이를 포함한다. 표시부(130)는 입력부(110)와 결합되어 터치 스크린(touch screen)으로 구현될 수 있다.
저장부(140)는 전자장치(100)의 동작 프로그램들을 저장한다. 저장부(140)는 전원의 제공 유무와 무관하게 데이터(정보)를 보존할 수 있는 비휘발성 속성의 스토리지(storage)와, 프로세서(150)에 의해 처리되기 위한 데이터가 로딩되며 전원이 제공되지 않으면 데이터를 보존할 수 없는 휘발성 속성의 메모리(memory)를 포함한다. 스토리지에는 플래시메모리(flash-memory), HDD(Hard-Disc Drive), SSD(Solid-State Drive), ROM(Read Only Memory) 등이 있으며, 메모리에는 버퍼(buffer), 램(Random Access Memory, RAM) 등이 있다.
저장부(140)는 실시간 분석 장치(30)가 측정한 나노입자의 크기 및 나노입자에 담지된 대상 약물의 양, 개별 측정 별로 설정된 파라미터 값, 새로 결정된 파라미터 값, 다목적 베이지안 최적화 알고리즘 등을 저장할 수 있으며, 측정 정보 수신, 캡슐화 효율성 및 예상 로딩 용량 연산, 파라미터 값 결정, 결정된 파라미터 값에 따른 제어 정보 전송 등을 수행하는 과정에서 필요한 연산 프로그램 등을 저장할 수 있다.
프로세서(150)는 프로그램 등 소프트웨어를 실행하여 전자장치(100)의 적어도 하나의 다른 구성요소(예: 하드웨어 또는 소프트웨어 구성요소)를 제어할 수 있고, 다양한 데이터 처리 또는 연산을 수행할 수 있다.
본 발명의 일 실시예에 따른 프로세서(150)는 합성 모듈에 의해 합성된 대상 약물이 담지된 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신하고, 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 양에 따른 상기 합성 모듈에 대한 복수의 파라미터의 파라미터 값을 결정할 수 있다.
이때, 프로세서(150)가 다목적 베이지안 최적화 알고리즘을 구축하거나, 기 구축된 다목적 베이지안 최적화 알고리즘을 외부로부터 수신 및 저장하여 이용할 수 있으며 어느 하나에 한정되는 것은 아니다.
한편, 프로세서(150)는 상기 동작들을 수행하기 위한 데이터 분석, 처리, 및 결과 정보 생성 중 적어도 일부를 규칙 기반 또는 인공지능(Artificial Intelligence) 알고리즘으로서 기계학습, 신경망 네트워크(neural network), 또는 딥러닝 알고리즘 중 적어도 하나를 이용하여 수행할 수 있다. 신경망 네트워크의 예로는, CNN (Convolutional Neural Network), DNN (Deep Neural Network), RNN (Recurrent Neural Network), 트랜스포머(transformer)와 같은 모델을 포함할 수 있다.
도 3은 본 발명의 일 실시예에 따른 전자장치의 동작 흐름도를 도시한 도면이다.
본 발명의 일 실시예에 따르면, 프로세서(150)는 합성 모듈(10)에 의해 합성된 대상 약물이 담지된 나노입자의 크기 및 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신할 수 있다(S10).
프로세서(150)는 실시간 분석 장치(30)로부터 대상 약물이 담지된 나노입자의 크기 및 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신할 수 있다.
본 발명의 일 실시예에 따르면, 프로세서(150)는 다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 나노입자의 크기 및 대상 약물의 양에 따른 합성 모듈(10)에 대한 복수의 파라미터의 파라미터 값을 결정할 수 있다(S20).
본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼(1)의 목적은 합성 모듈(10)이 타겟하는 크기의 나노입자를 합성하면서도, 최대한 많은 양의 약물을 담지하도록 자동적으로 최적화함에 있다.
합성 모듈(10)은 복수의 파라미터의 파라미터 값을 제어하여 나노입자를 합성할 수 있으며, 복수의 파라미터는 총 유량, 대상 약물과 지질의 유량 비율, 음이온성 지질 농도, POPC 지질과 콜레스테롤의 비율 및 대상 약물의 농도를 포함할 수 있다.
나노입자 합성에 사용되는 지질은, 음이온성 지질, POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 지질과 콜레스테롤로, 음이온성 지질의 양은 나노입자, 특히 리포솜의 안정성과 기능성에 기여하며 통합형 미세유체 연속 흐름 플랫폼(1)의 효과에 매우 중요하다. 또한, POPC 지질과 콜레스테롤의 비율은 리포솜의 크기와 캡슐화 효율에 대한 중추적인 영향을 줄 수 있다.
합성 모듈(10)의 합성 성능을 평가하기 위해, 프로세서(150)는 초기 대상 약물의 양 대비 나노입자에 담지된 대상 약물의 양을 나타내는 캡슐화 효율성(Encapsulation Efficiency, EE)을 연산할 수 있다. 캡슐화 효율성은 다음 수학식 1을 이용하여 연산할 수 있다.

또한, 프로세서(150)는 초기 지질의 양 대비 나노입자에 담지된 대상 약물의 양을 나타내는 예상 로딩 용량(Expected Loading Capacity, ELC)을 연산할 수 있다. 예상 로딩 용량은 다음 수학식 2를 이용하여 연산할 수 있다.

프로세서(150)는 베이지안 최적화 알고리즘을 이용하여 캡슐화 효율성(EE)과 예상 로딩 용량(ELC)을 최대화하는 동시에 측정된 나노입자의 크기와 타겟 크기 간의 차이를 최소화하도록 하는 파라미터 값을 결정할 수 있다. 이 과정을 사이즈 타겟팅(size targeting)이라고도 한다.
베이지안 최적화 알고리즘은, 복수의 파라미터의 파라미터 값들과 나노입자의 크기와 타겟 크기 간의 차이(절대치), 캡슐화 효율성(EE) 및 예상 로딩 용량(ELC) 3가지의 타겟값들 간의 관계를 정의한 목적 함수를 예측해가며, 3가지를 모두 만족하게 하는 파라미터 값들을 찾는 알고리즘이다.
구체적으로, 프로세서(150)는 나노입자의 크기와 타겟 크기 간의 차이는 작고, 캡슐화 효율성(EE) 및 예상 로딩 용량(ELC)은 크게 하는 파라미터 값을 결정할 수 있다. 파라미터 값들을 통해 최적화하고자 하는 대상이 복수인 바, 다목적 베이지안 최적화 알고리즘이라고도 한다.
본 발명의 일 실시예에 따르면, 프로세서(150)는 결정된 파라미터 값에 기초하여 나노입자를 합성하도록 합성 모듈(10)을 제어할 수 있다(S30).
이 과정을 주문형 나노입자 합성(On-demand synthesis of nanoparticles)이라고도 한다. 프로세서(150)가 결정된 파라미터 값에 따라 합성 모듈(10)이 나노입자를 합성하도록 제어하는 방법은 다양할 수 있으며, 예를 들어, 프로세서(150)는 합성 모듈(10)의 자동화된 펌핑 장치, 선택기 벨브 등에 결정된 파라미터 값에 관한 정보를 전송하는 등의 방법으로 제어할 수 있다. 구체적인 내용은 도 5a를 참조하여 설명한다.
본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼(1)은 원하는 특성, 나노입자의 크기와 타겟 크기간의 차이 최소화, 캡슐화 효율성(EE) 및 예상 로딩 용량(ELC)에 도달할 때까지 S10 내지 S30을 반복할 수 있다.
본 발명의 일 실시예에 따르면, 통합형 미세유체 연속 흐름 플랫폼(1)은 향상된 치료 결과와 함께 고도로 최적화되고 효과적인 약물 담지 나노입자의 개발을 촉진함으로써 약물 전달 분야를 크게 발전시킬 수 있는 잠재력을 가지고 있다.
도 4는 본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼의 동작 모습을 도시한 도면이다.
도 4에 도시된 것처럼 본 발명에서는 약물이 담지된 나노입자의 합성 및 실시간 특성화를 위한 완전히 통합되고 자동화된 AI 기반 통합형 미세유체 연속 흐름 플랫폼(1)을 제시한다.
본 발명의 일 실시예에 따른 플랫폼(1)에는 1) 자동화된 나노입자 합성 모듈(Nanoparticle Synthesizer), 2) 정제 모듈(Purification), 3) 베이지안 최적화를 통한 특성화 및 결정 정책(Characterization & Decision policy with Bayesian Optimization)이 포함되어 있다. 이러한 구성 요소들은 약물이 담지된 나노입자의 효율적이고 정확한 제조에 시너지 효과를 발휘할 수 있다.
플랫폼(1)의 첫 번째 단계는 약물이 담지된 나노입자를 합성하기 위한 복수의 파라미터와, 약물 및 운반체의 구성을 자동으로 제어하는 것이다. 이 자동화는 일관되고 재현 가능한 결과를 보장하여 인적 오류와 변동성을 최소화할 수 있다. 플랫폼(1)은 구성 요소의 유량과 비율을 정밀하게 제어함으로써 원하는 특성을 갖는 나노입자의 합성을 가능하게 한다. 먼저 리포솜과 고분자 나노입자(PCL)라는 두 가지 유형의 담체의 자동적인 합성을 위해 이 모듈의 성능을 평가하였고, 자율 나노입자 합성모듈(ANS)를 통해 인간의 개입 없이 시간당 30번의 실험 규모로 고처리량 방식으로 최적의 크기와 균일성을 갖는 나노입자를 합성할 수 있었다.
약물이 담지된 나노입자를 위한 완전히 통합된 플랫폼(1)을 구축하기 위해 합성 모듈(10)을 정제 모듈(20)과 통합할 수 있다.
구체적으로, 캡슐화되지 않은 자유 약물과 대상 약물이 담지된 나노입자를 분리하여 나노입자에 캡슐화된 약물의 양을 측정하는 것이다. 이를 위해 캡슐화되지 않은 자유 약물을 신속하게 제거하고 나노입자를 분리하기 위한 정제 모듈(20)을 도입하였다.
정제 모듈(20)에 의해 분리된 나노입자로부터 대상 약물을 방출하기 위해 용매, 예를 들어, 메탄올을 이용할 수 있다.
마지막 단계에서는 제1실시간 분석 장치, 예를 들어, 동적 광 산란(Dynamic Light Scattering, DLS) 기기를 사용하여 나노입자 크기를 지속적으로 실시간 모니터링할 수 있다. 또한, 제2실시간 분석 장치, 예를 들어, UV-Vis 분광 기구를 통해 분광법을 이용하여 나노입자 내부에 캡슐화된 약물의 양을 측정할 수 있다.
나노입자 합성부터 캡슐화된 입자와 캡슐화되지 않은 입자의 분리 및 실시간 특성화에 이르는 이 포괄적인 플랫폼(1)은 인간의 개입 없이 자율적으로 작동될 수 있다.
최적의 크기와 약물 로딩을 위한 전체 흐름을 최적화하기 위해 플랫폼 (1)에 다목적 베이지안 최적화 알고리즘을 도입하여 합성 및 특성화 단계를 더욱 개선하는 강력한 의사결정 정책을 확립할 수 있다.
일반 배치(Normal Batch) 구세대 시스템의 경우, 여러 실험에 몇 시간, 며칠이 소요되는데 반하여, 본 발명의 일 실시예에 따른 플랫폼(1)은 합성, 정제, 특성화 및 AI 의사결정을 수행하는 한 번의 실험에 20분 미만이 소요되는 점에서 이점이 있다. 또한 플랫폼(1)은 실험당 매우 적은 양의 샘플(~3mL)을 사용하는 바 효율적으로 나노입자를 제조할 수 있다.
도 5a는 본 발명의 일 실시예에 따른 합성 모듈의 동작 모습 및 도 5b 내지 도 5f는 실험 결과를 도시한 도면이다.
이 플랫폼(1)의 적용 가능성을 보여주기 위해 항암제인 독소루비신(Doxorubicin)을 대상 약물로 두고, 독소루비신의 캡슐화를 위해 두 가지 다른 나노입자인 리포솜과 PEG-PLGA 나노입자를 사용했다.
나노기술 및 약물 전달 영역에서 합성 모듈(10)의 개발은 기존의 비자동화 플랫폼이 제기하는 문제를 해결하는 중요한 솔루션으로 떠오르고 있다. 나노입자 형성에 대한 전통적인 접근 방식은 노동 집약적이고 시간 소모적이며 변형이 발생하기 쉬우므로 일관되고 효율적인 결과를 달성하는 데 심각한 장애물이 된다. 기존 방법은 다양한 단계에서 수동 개입이 필요한 경우가 많으므로 입자 크기, 구성 및 기타 중요한 매개변수에 대한 정밀한 제어를 유지하는 것이 어렵다. 더욱이, 자동화가 부족하면 확장성이 방해되어 다양한 응용 분야에 대한 나노입자의 생산 용량이 제한된다. 합성 모듈(10)은 나노입자 형성 프로세스를 간소화하는 완전 자동화된 플랫폼을 제공함으로써 이러한 환경에서 획기적인 발전을 보여준다.
본 발명의 일 실시예에 따른 합성 모듈(10)은 수동 합성의 힘들고 오류가 발생하기 쉬운 측면을 제거할 뿐만 아니라, 최첨단 나노기술 및 약물 전달 연구에 필수적인 재현성과 확장성을 보장한다.
또한, 합성 모듈(10)은 약물이 담지된 나노입자의 합성, 정제 및 특성화를 포괄하는 대규모 통합 플랫폼의 필수 구성 요소이다. 이러한 전체적인 접근 방식에서 합성 모듈(10)은 효율적이고 일관된 나노입자 생산을 위한 초석 역할을 하며 포괄적인 나노의학 연구 프로그램의 전반적인 성공과 타당성에 크게 기여한다.
합성 모듈(10)은 세 가지 주요 모듈로 구성될 수 있다. 먼저, 전구체 형성 모듈(Precursor Formulation Module)로 알려진 첫 번째 모듈은 전구체 물질을 준비하여 이 자동화 플랫폼에서 중추적인 역할을 한다. 이러한 정밀도는 최종 나노입자의 원하는 특성과 기능을 결정하는 기본 단계인 나노입자 빌딩 블록의 정확한 구성을 보장할 수 있다. 그 다음, 합성 모듈(10)의 핵심에는 나노입자가 시작 물질로부터 형태를 취하는 흐름 반응기 모듈(Flow Reactor Module)이 있다. 뛰어난 제어력과 정밀도로 작동하는 이 장치를 사용하면 다양한 응용 분야에 필요한 특정 특성을 가진 나노입자를 생성할 수 있다. 마찬가지로 중요한 것은 합성 과정에서 나노입자의 크기와 다분산 지수(Polydispersity Index, PDI)에 대한 실시간 통찰력을 제공하는 In-Situ 나노입자 크기 특성화 모듈(Nanoparticle Size Characterization Module)이다. 나노입자 크기와 PDI는 성능과 기능을 결정하는 데 중요한 파라미터이기 때문에 이러한 실시간 특성화 기능은 중요한 역할을 한다.
정확한 크기 제어는 나노입자가 효과적인 약물 전달을 위해 원하는 범위 내에 있도록 보장하는 반면, 낮은 PDI는 나노입자 간의 균일성을 나타낸다. 이러한 요소는 나노의학 응용 분야에서 일관되고 신뢰할 수 있는 결과를 달성하는 데 중요하다. 이 세 가지 모듈의 원활한 조정은 컴퓨터 제어 알고리즘에 의해 관리되므로 사람의 개입 필요성과 관련 오류 가능성이 크게 줄어든다. 이러한 자동화 수준은 나노입자 형성의 재현성과 효율성을 향상시킬 수 있다.
합성 모듈(10)은 인공 지능의 힘을 통합하여 다목적 베이지안 최적화 알고리즘을 활용한다. 전자장치(100)는 다목적 베이지안 최적화 알고리즘을 이용하여 결정한 파라미터를 결정하여 반복적으로 합성 모듈(10)을 통한 나노입자의 최적화를 수행할 수 있다. 전자장치(100)는 이전 실행의 데이터를 분석하고 다시 최적화함으로써, 지능적인 접근 방식을 통해 나노입자의 크기와 PDI가 엄격한 요구 사항을 충족하도록 보장한다.
도 5a 내지 도 5f를 살펴보면, 통합형 미세유체 연속 흐름 플랫폼(1)의 기능을 입증하기 위해 두 가지 종류의 나노입자의 자율 생산을 시도했다. 초기 연구에서는 리포솜 나노입자와 하나의 추가 고분자 나노입자가 고려되었다.
도 5a에 표시된 것처럼 연구의 초기 단계에서 리포솜 제조 공정 내에서 다양한 중요한 파라미터를 세심하게 규제하여 나노입자 크기 및 다분산 지수(PDI)에 대한 중대한 영향을 확인했다. 이 플랫폼(1)은 매우 짧은 기간(4시간 동안 120번의 실험) 내에 두 리포솜 나노입자의 신속한 합성 및 특성화를 촉진했다.
도 5b에서 볼 수 있듯이 나노입자의 크기는 약 40nm에서 약 300nm까지 다양하고 PDI는 약 0.05에서 0.5까지 다양하다.
정밀도와 반복성을 보장하기 위해 합성 모듈(10)의 자동화된 펌핑 장치를 사용하여 총 유량, 유량 비율, 1mg/mL~10mg/mL 범위의 리포솜 농도 등 연속 파라미터를 정밀하게 제어할 수 있다. - 이들 모두는 나노침전의 맥락에서 기본 변수로 인식된다.
POPC 지질과 콜레스테롤의 지질 용액 비율이 55:45로 설정된 지질 용액의 인라인 희석을 위한 용매로 에탄올을 사용하였고, 또한 10mol%의 음이온성 지질(POPG)도 0, 1, 5mol% 수준의 다양한 양의 PEG(polyethylene glycol)와 함께 사용되었다.
이러한 연속 파라미터 외에도 리포솜 합성에 중추적인 범주적 요인도 다루었다. 특히, 우리는 지질 조성에 통합된 PEG의 분자 백분율(mol%)을 특징으로 하는 지질 형성의 영향을 조사하기 위해 실험을 맞춤화했다. 더욱이, pH 수준의 변화에 따라 반용매를 선택하는 것이 중추적인 역할을 했으며, 이는 선택기 밸브를 사용하여 현명하게 제어되었다.
도 5a는 고려 중인 주요 합성 매개변수에 대한 간결한 요약을 제공한다. 이 경우 Tris 완충액, NaCl 용액, Citrate 완충액, PBS 완충액 등 4가지 유형의 항용매(완충액)가 선택되었으며, 자동화된 플랫폼(1)은 다목적 베이지안 최적화 알고리즘 제안에 따라 각각을 선택했다.
자동화된 플랫폼(1)의 진정한 잠재력을 평가하기 위해 다목적 베이지안 최적화 알고리즘을 사용하여 합성 조건을 동적으로 조정하여 크기와 PDI를 체계적으로 최적화했다. 이 조사에 활용된 결과 및 해당 실험 조건에 대한 포괄적인 개요는 도 5b에 묘사된 평행 좌표 차트에 표시된다. 이 도면에서 볼 수 있듯이, 실험 횟수가 증가한 후에는 AI 기반 다목적 베이지안 최적화 알고리즘에 의한 자동화된 변경에 의해 크기와 PDI가 최소한으로 크게 최적화되는 경향을 확인할 수 있다.
플랫폼(1)이 개발된 이후에는 재현성과 견고성을 철저히 평가하는 것이 필수적이다. 플랫폼(1)의 재현성과 견고성을 면밀히 조사하기 위해 동일한 합성 조건을 연속적으로 실행하여 합성 및 특성화 절차를 총 10회 수행했으며 결과는 도 5c에서 볼 수 있다. 여기서 목표는 나노입자 크기와 다분산지수에 초점을 맞춰 결과가 안정적으로 반복될 수 있는지 여부를 조사하는 것이었다.
또한 도 5d에 표시된 것처럼 견고성 평가를 위해 미리 정의된 실험 프레임워크 내에서 합성 조건이 반복적으로 변경될 때 플랫폼(1)이 얼마나 잘 작동하는지 확인하려고 했다. 이를 위해 세 가지 서로 다른 합성 조건을 선택하고 각 조건에서 연속적인 합성을 수행했으며, 다양한 조건에서 얻은 결과의 일관성과 신뢰성을 측정하기 위해 이 프로세스를 3번 반복했다. 다목적 베이지안 최적화 알고리즘의 뛰어난 최적화 기능을 보여주기 위해 SOBOL 샘플링이라는 또 다른 결정 정책과의 비교 분석했다.
도 5e와 같이 각 실험을 완료하는 데 필요한 평균 시간이 계산되었다. 이 시간에는 공식화 및 합성, 특성화, AI 알고리즘 계산, 다가오는 실험을 준비하기 위한 전체 플랫폼 정리 등 모든 일반적인 단계가 포함된다. 기존 시스템은 빠른 합성 및 특성화로 인해 높은 처리량 실험 작업에 적합한 선택이 아닌 것으로 생각되지만, 본 발명의 일 실시예에 따른 플랫폼(1)은 높은 처리량 스크리닝 및 최적화를 위한 이상적인 옵션이 될 수 있다.
도 5f에서 볼 수 있듯이 각 결정 정책에 대해 총 30회의 실험 라운드가 수행되었으며 결과를 기록했다. 각 정책에 의해 생성된 하이퍼볼륨을 비교함으로써 다목적 베이지안 최적화 전략의 탁월한 성능을 입증할 수 있었다.
이 두 가지 사례 연구의 결과는 통합형 미세유체 연속 흐름 플랫폼(1)에서 운반체로 사용될 나노입자의 제어되고 재현 가능하며 효율적인 제조에 도움이 될 수 있음을 시사한다.
도 6a는 본 발명의 일 실시예에 따른 정제 모듈의 동작 모습 및 도 6b 내지 도 6c는 실험 결과를 도시한 도면이다.
합성 모듈(10)을 통해 나노입자의 합성 이후에 중요한 단계는 나노캐리어 내의 캡슐화된 약물과 캡슐화되지 않은 약물을 분리하는 것이다.
이를 위해 정제 모듈(20)을 활용하며, 정제 모듈(20)은 멤브레인(RC 멤브레인 12000 KDE)으로 분할된 채널을 특징으로 하는 유동 투석 모듈로 구현될 수 있다. 정제 모듈(20)의 버퍼 섹션에는 확산을 활용하고 약물 캡슐화된 나노입자에서 자유 약물을 제거하기 위해 다중 버퍼가 도입되었다.
도 6a에 도시된 대로, 리포솜 샘플(합성 후 나노입자 및 자유 약물로 구성)은 정제 모듈(20) 내에서 10분 투석 과정을 거쳤다. 정제 모듈(20)에 의해 나노입자가 분리된 이후, 분리된 나노입자를 파괴용 용매에 도입하여 약물이 방출되도록 한다. 정제 모듈(20)의 출구에서는 캡슐화된 나노입자가 메탄올에 유입되어 리포솜 막을 파괴하고 캡슐화된 약물을 방출했다. 이때, 메탄올-물 혼합물의 휘발성으로 인해 발생하는 기포 문제를 해결하기 위해 가스 또는 기포 트랩을 사용하여 기포를 제거할 수 있다.
이후, 제2실시간 분석 장치를 이용하여 방출된 약물의 강도를 UV-vis 분광법을 사용하여 측정함으로써 캡슐화된 약물의 양을 측정할 수 있다. 전자장치(100)는 캡슐화된 약물의 양을 측정하여 캡슐화 효율성을 연산할 수 있다.
실제 샘플을 도입하기 전에 정제 모듈(20)의 성능을 검증하기 위해 자유 독소루비신의 제거 효율성을 평가하는 테스트를 수행했다. 95% 제거 임계값을 안전한 것으로 설정하고 0.1, 0.2, 0.3 mg/mL 농도의 독소루비신을 주입했다. 도 6b에 설명된 대로 배출구에서 관찰된 독소루비신 제거율은 지속적으로 95%를 초과하여 독소루비신의 안전한 제거를 보장하였다.
그러나 도 6c에서 알 수 있듯이 정제 모듈(20)의 멤브레인 오염에 따른 문제가 발생함을 발견하였다. 이 문제를 완화하기 위해 멤브레인 세척 메커니즘을 구현하여 도 6d에 도시된 바와 같이 샘플이 10분 동안 흐른 후 추가 10분 동안 더 높은 유속(2mL/분)으로 세척 용매(에탄올)를 주입했다. 이 접근 방식은 멤브레인 오염을 방지하고 사이클 전반에 걸쳐 재현성을 보장할 수 있다. 전반적인 안전 성능을 검증하기 위해 세척 과정을 10사이클 이상 반복했다. 특히, 이 실험 설정에서 DI 수는 투석 장치의 완충제로 사용되었으며, 샘플은 0.1몰 구연산염 완충액에 용해된 독소루비신 용액으로 구성되었으며, 에탄올은 세척 용매로 사용되었다.
캡슐화되지 않은 자유 약물과 약물로 캡슐화된 나노입자를 분리하기 위해, 본 발명의 일 실시예에 따른 정제 모듈(20)을 사용하면 기존 방법에 비해 몇 가지 뚜렷한 이점을 제공할 수 있다.
첫째, 정제 모듈(20)이 실시간으로 작동해 분리 과정을 지속적으로 모니터링할 수 있다. 이는 즉각적인 피드백을 제공하고 동적 조정을 가능하게 하여 전체 프로세스 동안 정확한 캡슐화 효율성 계산을 보장하므로 특히 유리하다.
둘째, 정제 모듈(20)은 연속 흐름 플랫폼(1)과 호환되므로 다른 연속 나노입자 합성 및 제제화 공정과의 통합에 매우 적합하다. 또한 플랫폼(1)에 원활하게 통합되어 높은 처리량과 효율적인 캡슐화 효율성 결정을 촉진할 수 있다.
마지막으로, 광범위한 시료 준비 및 후처리가 필요할 수 있는 다른 기존 방법과 달리 정제 모듈(20)은 복잡한 시료 처리 필요성을 줄여 작업 흐름을 단순화함으로써 시료 손실 또는 오염 위험을 최소화할 수 있다.

도 7은 약물이 담지된 나노입자의 엔드투엔드 자율 생산을 위한 완전히 통합된 플랫폼을 나타낸다.
앞서 도 3의 S20과 관련하여 서술한 바와 같이, 시료 내 음이온성 지질의 양과 POPC 지질과 콜레스테롤의 비율은 리포솜 형성에 중요한 영향을 미친다.
이때, 본 발명에서는 음이온성 지질을 POPG(phosphatidylglycerol)을 사용하였다.
도 7의 우측 하단에 도시된 바와 같이, 본 발명에서는 POPG(0, 10, 20 mol%)에 대해 세 가지 다른 수준과 POPC 대 콜레스테롤 비율(1:2, 1:1, 2:1 mol%)에 대해 세 가지 다른 수준을 고려하여 총 9가지 독특한 조합 결과를 얻었다.
이러한 체계적인 접근 방식을 통해 다양한 조합을 탐색하고 최적화하여 지질 형성 파라미터 간의 상호 작용과 독소루비신이 담지된 리포솜의 결과적인 특성을 철저히 이해하기 위한 토대를 마련할 수 있었다.
지질 형성 외에도 약물 독소루비신의 농도도 0.1 mg/mL ~ 0.3 mg/mL으로 고려되었다. 그리고 마지막으로 총유량, 약물 대 지질의 유속비 등의 유동 관련 파라미터들도 선택되었다.
지질 제형 및 독소루비신 용액의 선택은 자동화된 펌핑 장치와 연결된 선택기 밸브를 활용하여 진행되었다. 다목적 베이지안 최적화 알고리즘에 의해 제안된 파라미터 값을 기반으로 약물이 담지된 나노입자는 합성 모듈(10)과 정제 모듈(20)의 유속 불일치를 해결하려고 시도하는 샘플 수집기 구성 요소에 의해 합성 및 수집될 수 있다. 그런 다음 수집된 샘플을 10분의 유지 시간으로 정제 모듈(20)에 주입한 후 나노입자 파괴를 위해 메탄올을 주입하고 마지막으로 제1실시간 분석 장치(DLS) 및 제2실시간 분석 장치(UV-Vis 기기)를 각각 사용하여 나노입자의 크기 및 약물의 양을 측정했다.
도 8a 및 도 8b는 본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼의 효율성을 도시한 도면이다.
시간 및 시료 소비의 효율성은 특히 나노입자 기반 나노의약품의 합성, 정제, 특성화 및 최적화에서 우리가 개발한 통합 플랫폼을 기존의 수동 및 인간 제어 흐름 또는 배치 시스템과 차별화되는 중요한 측면이다. 이러한 요소의 중요성은 빠른 프로세스로 인해 더 빠른 발전과 향상된 환자 결과로 이어지는 약물 개발의 가속화된 속도에 있다.
시기적절한 생산이 핵심인 경우, 예를 들어, 암 치료의 경우 나노입자를 효율적으로 합성, 정제, 특성화 및 최적화하는 능력은 시기적절하고 효과적인 치료법을 제공하는 것이 가장 중요하다. 본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼(1)은 이 점에서 탁월하여 합성 시간을 크게 단축시킬 수 있다. 그림 8a에 표시된 것처럼 업스트림(upstream), 즉 나노입자 합성에 따른 시간 범위는 약 9~180초이며, 총 유량이 변화하는 경우에도 놀라운 효율성을 보여준다. 더욱이, 도 8a에 도시된 것처럼 전체 프로세스는 단지 16~20분에 불과하며, 다운스트림(downstream)인 정제 모듈(20)이 대부분의 시간(10~12분)을 구성한다. 이러한 효율성은 특히 긴급 의료 상황에서 환자에게 나노입자 기반 나노의약품을 신속하게 전달하는 데 매우 중요하다.
샘플 소비 측면에서 플랫폼(1)은 놀라운 효율성을 보여준다. 도 8b에서 볼 수 있듯이 유속 비율(1~20개의 독소루비신 대 지질 범위)을 기준으로 독소루비신과 지질의 누적 소비는 최소로 총 3.25mL이다. 또한 플랫폼(1)의 무결성을 유지하는 데 중요한 단계인 세척 과정에서는 5~8mL의 적당한 세척액을 사용한다. 플랫폼(1)의 전반적인 효율성은 전체 프로세스에서 인간의 개입을 제거할 뿐만 아니라 시료 소비를 크게 줄여 생물 의학 연구에서 지속 가능하고 자원 효율적인 관행에 대한 현대적인 강조와 일치한다. 요약하자면, 우리가 개발한 통합 플랫폼(1)은 신속하고 효과적인 합성을 보장할 뿐만 아니라 시간과 시료 활용 측면에서 비교할 수 없는 효율성으로 나노입자 기반 나노의학의 발전에 기여한다.
도 9a는 본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼의 동작 흐름도 및 도 9b 내지 도 9c는 실험 결과를 도시한 도면이다.
도 9a는 플랫폼(1)의 전반적인 효능을 입증하기 위해 리포솜 합성에 초점을 맞춘 프로세스의 자세한 흐름도를 도시한다. 처음에 다목적 베이지안 최적화 알고리즘은 실험을 위한 최적의 조건을 제안한다(AI-based Decision Making). 그 후, 플랫폼(1)은 5~8분 동안 지속되는 에탄올을 사용한 세척 단계를 거친다(Wash the system). 그런 다음 다목적 베이지안 최적화 알고리즘의 권장 사항에 따라 적절한 독소루비신 및 지질에 대한 밸브를 설정하고 펌프 장치를 채운다(Rotate the valve based on Recipe & Fill the pumps for synthesis).
그런 다음 플랫폼(1)은 0.25mL에 대해 1mL/분의 유속으로 설정된다(Warm-up system with 0.25mL sample). 후속 단계에서는 합성 모듈(10)에서 제안된 레시피에 따라 실험 샘플을 준비하고 수집 펌프에 의해 수집된다(Prepare the sample with recipe & Collect sample with sample collector).
수집된 샘플은 정제 모듈(20)에 주입되며, 이 과정에는 10분이 소요된다(Dialysis). 정제 모듈(20)의 출구에서는 메탄올을 투입하고(Methanol Injection), 방출된 약물을 UV-Vis를 이용하여 분석하여 캡슐화 효율성을 측정한다(Measure Drug-loading). 마지막으로 플랫폼(1)을 정지하고 DLS(동적 광산란) 장치를 사용하여 나노입자 크기를 측정한다(Measure size).
플랫폼(1)의 견고성을 평가하기 위해 도 9b에 설명된 것처럼 각각 6시간의 시간 간격을 두고 두 번의 실험에 대해 세 가지 다른 합성 조건에서 엔드투엔드 처리를 수행했다.
플랫폼(1)의 성능 평가를 위해 우리는 1) 나노입자 크기, 2) 캡슐화 효율성, 3) 예상 로딩 용량이라는 세 가지 목표를 정의했다. 중요한 지표인 캡슐화 효율성은 나노입자 내에 캡슐화된 약물의 비율을 반영하여 합성 과정의 효능에 대한 통찰력을 제공한다.
전반적으로 다목적 베이지안 최적화 알고리즘은 치료효과의 향상으로서 캡슐화 효율성을 극대화할 수 있는 조건을 제시하고자 하였으며, 예상 로딩 용량을 극대화하는 것은 지질의 소모를 줄이면서 독소루비신의 담지를 최대화하여 치료 효과를 높이는 동시에 지질에 의해 유발되는 세포 독성을 감소시킬 수 있다.
도 9b 크기에서 볼 수 있듯이 EE와 ELC는 6시간의 차이 과정에서 재현 가능한 결과를 보여주었으며, 이는 합성부터 정제, 최종 특성화까지 플랫폼(1)이 모든 수준에서 견고함을 나타낸다.
플랫폼(1)의 견고성과 재현성을 종합적으로 평가한 후, 캡슐화 효율성(EE)과 예상 로딩 용량(ELC)을 모두 포괄하여 약물 로딩을 최대화하는 데 중점을 두고 폐쇄 루프 자율 구동 프로세스를 위해 이 플랫폼(1)을 배포하는 것을 구상한다.
자체 최적화 프로세스를 시작하기 전에 사용자는 자신의 요구 사항에 맞는 특정 나노입자 크기를 선택할 수 있다. 그런 다음 플랫폼(1)은 빠른 시간 내에 최소한의 주기로 원하는 크기를 달성하기 위해 효율적으로 작동하며 동시에 다른 중요한 목표를 최적화한다. 이 새로운 접근법은 특히 맞춤형 의료의 경우 상당한 가능성을 갖고 있다. 사용자 정의 속성으로 특정 나노의학을 맞춤화해야 하는 시나리오의 경우 플랫폼은 개별화된 치료 요구 사항을 충족하도록 신속하게 적응할 수 있다. 이러한 맞춤형 맞춤화를 통해 나노의학은 원하는 치료 효과를 제공하도록 정밀하게 설계되어 개별 환자의 치료 결과를 최적화할 수 있다.
이 접근법은 다중 장기 전달이 필수적일 수 있는 암 전이의 맥락에서 또 적절하게 적용될 수 있다. 다양한 나노입자 크기가 특정 장기에 다양한 영향을 미칠 수 있다는 점을 인식한 이 플랫폼(1)은 나노의학의 신속한 준비, 정제 및 최적화에 탁월하다. 이를 통해 주문형 치료 효과가 가능해 나노의학이 지정된 크기 기준에 따라 특정 기관에 전달되도록 준비된다. 본질적으로, 본 발명의 일 실시예에 따른 플랫폼(1)은 맞춤형 치료 솔루션에 대한 수요 증가에 맞춰 나노의학 합성의 패러다임 전환을 제시하고 암 전이와 관련된 맞춤형 의학부터 효율적인 다기관 전달에 이르기까지 다양한 의료 시나리오를 해결하기 위한 역동적인 접근 방식을 제공할 수 있다.
도 9c에 도시된 바와 같이, 플랫폼(1)의 성능을 평가하기 위해 요구되는 크기 70, 170 및 350nm를 달성한다는 목표를 제시했다. 다목적 베이지안 최적화 알고리즘은 캡슐화 효율성(EE)과 예상 로딩 용량(ELC)을 최대화하는 동시에 측정된 크기와 대상으로 선택한 크기 간의 절대 차이를 최소화하는 작업을 수행했다.
도 9c를 살펴보면 베이지안 최적화 루프에서 단 한 번의 초기 무작위 실험으로 플랫폼(1)은 단 11번의 실험만으로 첫 번째 목표 크기에 성공적으로 도달했다. 이러한 초기 라운드에서 얻은 통찰력을 활용하여 플랫폼(1)은 170nm와 350nm의 다른 목표 크기에 도달하는 데 단 2개의 추가 단계만 필요했다.
이 프로세스 전반에 걸쳐 플랫폼(1)은 캡슐화 효율성을 60%의 일관된 수준으로 적절하게 유지하여 첫 번째 타겟팅 주기에서 10% 예상 로딩 용량을 달성하고 후속 원하는 단계에서 지속적으로 예상 로딩 용량을 3% 초과했다.
이러한 결과는 플랫폼(1)의 탁월한 적응성과 효율성을 나타내는 강력한 지표로 사용된다. 최소한의 실험으로 목표 크기에 대한 신속한 수렴은 특정 사용자 정의 목표를 충족하는 데 있어 자율주행 플랫폼의 효율성을 강조한다. 또한 다양한 과제 단계에서 높은 캡슐화 효율성과 상당한 예상 로딩 용량을 유지하는 플랫폼(1)의 능력은 견고성과 신뢰성을 강조한다. 이는 정밀 나노의학 합성을 위한 강력한 도구로서 플랫폼(1)의 잠재력을 강조하며, 맞춤형 치료 적용을 위한 귀중한 통찰력을 제공하고 약물 전달의 복잡한 과제를 효율적으로 해결할 수 있다.
도 10은 도 9c의 세부 결과를 나타낸 표이다.
도 10에 도시된 것처럼, 대상 1(70nm)에 대한 첫 11라운드에서 다목적 베이지안 최적화 알고리즘은 크기를 제어할 뿐만 아니라 더 높은 캡슐화 효율성 및 예상 로딩 용량에 대한 다양한 파라미터를 탐색하는 경향이 있다.
몇 번의 라운드 후에 다목적 베이지안 최적화 알고리즘은 다량의 POPG(20mol%)를 사용하고 P/C 비율을 2(2:1)로 고정하는 경향이 있다. 그 사이에 원하는 최소화 크기에 대한 적절한 흐름 조건을 찾으려고 한다. 첫 번째 목표의 마지막 사이클에서 다목적 베이지안 최적화 알고리즘은 P/C의 균형(1:1)을 사용하여 가능한 가장 높은 음이온성 지질을 사용하여 총 유량을 최고 및 중간 유량 비율로 설정했다.
이 라운드와 목표 변경 후에는 흐름 조건을 변경해야만 최적의 사이클이 발견되었으며, 이로 인해 캡슐화 효율성 및 예상 로딩 용량이 허용 가능한 범위로 유지되고 크기가 어느 정도 증가했다.
음이온성 지질이 약간 변형되어 예상 로딩 용량이 감소한 다음 타겟에서도 동일한 과정이 발생했지만 유속과 유속 비율을 거의 최소 수준으로 낮추어 원하는 크기를 달성했다.
도 11a 내지 도 11d는 본 발명의 일 실시예에 따른 통합형 미세유체 연속 흐름 플랫폼을 이용하여 PEG-PLGA 나노 입자를 합성하는 모습 및 실험결과를 도시한 도면이다.
구체적으로, 도 11a는 PEG-PLGA 나노입자 제조를 위한 합성 모듈의 개략도, 도 11b는 PEG-PLGA 나노입자 제조를 위한 폐쇄 루프 자율 플랫폼에서 획득한 크기 및 EE 결과를 나타내는 도면, 도 11c는 자율 플랫폼에서 얻은 크기 및 EE에 대한 최대 결과를 나타내는 도면, 도 11d는 폐쇄 루프 최적화 프로세스 전반에 걸쳐 실험 실행 및 제안 조건을 고려한 누적 결과를 나타내는 도면이다.
다양한 유형의 나노캐리어에 대해 개발된 플랫폼 활용의 다양성을 추가로 확인하기 위해, 도 11a와 같이 Dox가 탑재된 PEG-PLGA 나노입자의 자율 합성을 위한 시스템을 구축했다. 도 11a에 도시된 바와 같이, 리포솜을 이용한 플램폼과 비교하여 수정된 부분은 지질 제형 스톡은 5, 10, 15 및 20mg/mL로 설정된 농도를 갖는 아세토니트릴에 용해된 PEG-PLGA 중합체로 대체된 합성 모듈(ANS)이다.
또한, 리포솜(TFR, FRR)에 대한 이전 캠페인에서 고려된 일반적인 흐름 매개변수와 Dox의 농도가 최적화를 위한 매개변수로 선택되었다. 폐쇄 루프 최적화 실험은 20회(각각 3회 반복) 동안 수행되었으며 이에 따라 나노 입자의 크기와 EE가 측정되었다. 또한, 리포솜(TFR, FRR)에 대한 이전 캠페인에서 고려된 일반적인 흐름 매개변수와 Dox의 농도가 최적화를 위한 매개변수로 선택되었다. 폐쇄 루프 최적화 실험은 20회(각각 3회 반복) 동안 수행되었으며 이에 따라 나노 입자의 크기와 EE가 측정되었다.
도 11b를 참조하면, 이 실험의 전체 결과 및 파레토 경계(Pareto frontiers)로, 합성된 나노입자의 크기는 100nm보다 약간 작은 것부터 약 500nm까지, EE 값은 약 6%에서 약 88%까지 다양하게 나타남을 알 수 있다.
리포솜 나노입자에 대한 최적화 캠페인과 유사하게 qNEHVI 획득 기능을 갖춘 MOBO 알고리즘을 사용하여 크기를 최소화하고 EE를 최대화했다. 도 11c에 도시된 바와 같이, 여섯 번째 실험에서는 최소 크기 98nm가 관찰되었으며, 여덟 번째 실험에서는 EE의 최대값(88%)이 탐색되어 MOBO 알고리즘의 전반적인 성능이 우수함을 나타낸다. 도 11d에 표시된 대로 프로세스가 더 많은 수의 실험으로 진행됨에 따라 알고리즘은 일반적으로 더 높은 EE 값에 해당하는 더 높은 농도의 Dox(0.3mg/mL)와 더 낮은 농도의 PEG-PLGA를 갖는 조건을 제안하는 경향이 있다. 초기에 Dox의 낮은 농도가 선택된 초기 실험 실행과 대조적이다.
이 두 가지 실험 플랫폼은 나노입자 크기 타겟팅 또는 최적화를 위해 서로 다른 방향으로 다양한 유형의 약물이 탑재된 나노입자를 합성하고 불필요한 담체 독성을 최소화할 수 있는 약물 로딩 최적화를 위한 최첨단 플랫폼의 효율적인 활용을 승인함을 알 수 있다. 동시에 표적 전달 속도를 높이며, 핵산과 같은 다른 유형의 치료제의 캡슐화를 통해 개인화되고 체계적인 표적 전달을 달성할 수 있는 잠재력을 보유하고 있다.

Claims (17)

  1. 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조를 위한 통합형 미세유체 연속 흐름 플랫폼에 있어서,
    대상 약물이 담지된 나노입자를 합성하는 합성 모듈;
    상기 합성 모듈로부터 유입되는 상기 나노입자와 상기 나노입자에 담지되지 않은 약물을 분리하는 정제 모듈;
    상기 정제 모듈을 통해 분리된 상기 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양을 측정하는 실시간 분석 장치; 및
    다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 양에 따른 상기 합성 모듈에 대한 복수의 파라미터의 파라미터 값을 결정하는 전자장치;를 포함하는 플랫폼.
  2. 제1항에 있어서,
    상기 합성 모듈은,
    총 유량, 대상 약물과 지질의 유량 비율, 음이온성 지질 농도, POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 지질과 콜레스테롤의 비율 및 대상 약물의 농도를 포함하는 상기 복수의 파라미터의 파라미터 값을 이용하여 상기 나노입자를 합성하는 플랫폼.
  3. 제1항에 있어서,
    상기 실시간 분석 장치는,
    상기 나노입자의 크기를 측정하는 제1실시간 분석 장치; 및
    상기 나노입자에 담지된 대상 약물의 양을 측정하는 제2실시간 분석 장치를 포함하는 플랫폼.
  4. 제3항에 있어서,
    상기 제2실시간 분석 장치는,
    상기 나노입자를 파괴하는 용액을 이용하여 파괴된 나노입자로부터 담지된 대상 약물의 양을 측정하는 플랫폼.
  5. 제1항에 있어서,
    상기 전자장치는,
    초기 대상 약물의 양 대비 상기 나노입자에 담지된 대상 약물의 양을 나타내는 캡슐화 효율성, 및 초기 지질의 양 대비 상기 나노입자에 담지된 대상 약물의 양을 나타내는 예상 로딩 용량을 연산하는 플랫폼.
  6. 제5항에 있어서,
    상기 전자장치는,
    캡슐화 효율성과 예상 로딩 용량을 최대화하는 동시에 측정된 상기 나노입자의 크기와 타겟 크기 간의 차이를 최소화하도록 하는 상기 파라미터 값을 결정하는 플랫폼.
  7. 제1항에 있어서,
    상기 전자장치는,
    결정된 상기 파라미터 값에 기초하여 상기 나노입자를 합성하도록 상기 합성 모듈을 제어하는 플랫폼.
  8. 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조를 위한 전자장치에 있어서,
    합성 모듈에 의해 합성된 대상 약물이 담지된 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신하고,
    다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 양에 따른 상기 합성 모듈에 대한 복수의 파라미터의 파라미터 값을 결정하는 프로세서;를 포함하는 전자장치.
  9. 제8항에 있어서,
    상기 나노입자는,
    상기 합성 모듈로부터 유입되어 정제모듈을 통해 상기 나노입자에 담지되지 않은 약물과 분리된 것을 특징으로 하는 전자장치.
  10. 제9항에 있어서,
    상기 프로세서는,
    실시간 분석 장치로부터 측정된 상기 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양에 관한 정보를 수신하는 전자장치.
  11. 제10항에 있어서,
    상기 실시간 분석 장치는,
    상기 나노입자의 크기를 측정하는 제1실시간 분석 장치; 및
    상기 나노입자에 담지된 대상 약물의 양을 측정하는 제2실시간 분석 장치를 포함하는 것을 특징으로 하는 전자장치.
  12. 제11항에 있어서,
    상기 제2실시간 분석 장치는,
    상기 나노입자를 파괴하는 용액을 이용하여 파괴된 나노입자로부터 담지된 대상 약물의 양을 측정하는 것을 특징으로 하는 전자장치.
  13. 제8항에 있어서,
    상기 복수의 파라미터는,
    총 유량, 대상 약물과 지질의 유량 비율, 음이온성 지질 농도, POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 지질과 콜레스테롤의 비율 및 대상 약물의 농도를 포함하는 전자장치.

    상기 프로세서는,

    상기 프로세서는,
    캡슐화 효율성과 예상 로딩 용량을 최대화하는 동시에 측정된 상기 나노입자의 크기와 타겟 크기 간의 차이를 최소화하도록 하는 상기 파라미터 값을 결정하는 전자장치.
  16. 제8항에 있어서,
    상기 프로세서는,
    결정된 상기 파라미터 값에 기초하여 상기 나노입자를 합성하도록 상기 합성 모듈을 제어하는 전자장치.
  17. 통합형 미세유체 연속 흐름 플랫폼을 이용한 인공지능 기반 최적화된 약물 전달용 나노입자의 자동 제조 방법에 있어서,
    복수의 파라미터에 기초하여 대상 약물이 담지된 나노입자를 합성하는 단계;
    상기 합성 모듈로부터 유입되는 상기 나노입자와 상기 나노입자에 담지되지 않은 약물을 분리하는 단계;
    분리된 상기 나노입자의 크기 및 상기 나노입자에 담지된 대상 약물의 양을 측정하는 단계; 및
    다목적 베이지안 최적화(Multi-Objective Bayesian Optimization, MOBO) 알고리즘을 이용하여 상기 나노입자의 크기 및 대상 약물의 양에 따른 상기 복수의 파라미터의 파라미터 값을 결정하는 단계를 포함하는 방법.
