Abstract
본 발명은 콜레라 독소를 민감하고 선택적으로 검출할 수 있는 생체 모방 전기화학 바이오센서에 관한 것으로, 본 발명의 바이오센서는 콜레라 독소가 특이적으로 결합하는 GM1이 풍부하게 존재하는 장내 상피세포막을 작동전극에 도포함으로써 콜레라 독소를 민감하고 선택적으로 검지할 수 있으며, 전기화학적 방식을 사용하여 전극의 임피던스 또는 전류값을 측정할 수 있는 기초장비만 있으면 어디에서나 간편하고 빠르게 콜레라를 검출할 수 있고, 바이오센서의 안정성이 높아 보관 및 배송이 용이해 다양한 지역에 공급하기 유리하므로, 급성으로 생명의 위협이 되며 광범위한 전염을 야기시킬 수 있는 콜레라를 조기에 검진하고 치료할 수 있는 효과가 있다.

인간이나 동물의 체내로 침입해 감염성 질병을 일으키는 생물체를 병원균 또는 병원체(pathogen)라고 명명하며, 병원균 중에서도 단세포 원핵생물인 세균(bacteria)의 경우 빠르게 증식하며 독소(toxin)를 분비해 세포나 조직을 손상시키고 물질대사에 관여해 독성을 일으킨다. 이 중, 고위험 병원체란 생물테러의 목적으로 이용되거나 사고 등에 의하여 외부에 유출될 경우 국민 건강에 심각한 위험을 초래할 수 있는 병원체로써 보건복지부령으로 정하는 것을 말한다. 고위험 병원체 종류로는, 페스트균(Yersinia pestis), 탄저균(Bacillus anthracis), 브루셀라균(Brucella melitensis, Brucella suis), 유비저균(Burkholderia pseudomallei), 비저균(Burkholderia mallei), 보툴리눔균(Clostridium botulinum), 이질균(Shigella dysenteriae Type 1), 클라미디아 시타키(Chlamydia psittaci), 큐열균(Coxiella burnetii), 야토균(Francisella tularensis), 발진티푸스균(Rickettsia prowazekii), 홍반열 리케치아균(Rickettsia rickettsii), 콕시디오이데스 이미티스균(Coccidioides immitis) 및 콜레라균(Vibrio cholerae) 등이 있다.
콜레라(Cholera)는 콜레라균(Vibrio cholerae)에 의해 감염되며 어패류 등의 해산물 식품매개로 전파되거나, 콜레라균에 감염된 사람의 분변처리가 잘 되지 않아 수로, 지하수 및 음용수 등에 오염되어 주변 사람들에게 전파된다. 비브리오 콜레라는 콜레라로 알려진 질병을 일으키는 주된 병원체로 알려져 있으며, 하나의 편모를 가지는 그람 음성균(gram negative)으로 분류되어 있다. 콜레라균은 외독소(exotoxin)인 콜레라 독소(cholera toxin, CT)을 통해 사람의 장 세포 통해 감염되어 설사(diarrhoea), 탈수(dehydration), 대사성 산성증(metabolic acidosis) 등을 일으키게 되며 치사율이 매우 높은 것으로 알려져 있어, 병원성 미생물인 비브리오 콜레라의 검출과 조기진단은 매우 중요한 부분으로 인식되고 있다. 콜레라는 특히 물을 통해 감염되기 때문에 전염력이 강한 특성을 지니고 있다. 현대에 와서 상수도정비 등의 수질개선을 통해 근본적인 문제를 해결하였고 콜레라를 치료할 수 있는 방법이 다양해지고 있지만, 수질개선이 어려운 갠지스 강 유역이나 개발 도상국 등의 지역에서는 풍토병화(endemic)되어 있는 상황이다. 콜레라균이 분비하는 콜레라 독소는 A 서브유닛(subunit)과 B 서브유닛으로 구성되어 있는데, 이 중 A 서브유닛만이 내부에서 독성을 띄는 기전을 가지며 B 서브유닛이 장 상피세포에 결합하는 역할을 한다. 장 상피세포에는 이 콜레라 독소와 특이적으로 결합하는 GM1(monosialotetrahexosylganglioside)이라는 수용체가 존재하는데, 콜레라 독소가 GM1과 결합하여 복합체를 형성하면 세포내 이입(endocytosis) 과정을 통해 장 세포 내로 들어간다. 장 세포막에 존재하는 G 단백질(protein)에서 GDP(guanosine diphosphate)가 GTP(guanosine triphosphate)로 인산화되면서 α subunit-GTP 복합체가 떨어져 나오고, 해당 복합체는 장 세포 내에서 아데닐산 고리화효소(adenylate cyclase)에 결합해 활성을 일으키며, 신호전달 물질인 cAMP(cyclic adenosine monophosphate)를 합성한다. 이렇게 합성된 cAMP는 protein kinase A를 활성화하고, 활성화된 protein kinase A는 장 세포막의 CFTR(Cystic fibrosis transmembrane conductance regulator, Cl 수송 막단백질)을 활성화시킨다. 활성화된 CFTR은 장 세포 내의 Cl 분자를 장내로 확산시키게 된다. 일반적으로 이 반응은 짧게 일어나며 GTP 가수분해효소인 GTPase가 아데닐산 고리화효소에 결합해있는 α subunit-GTP 복합체의 GTP를 GDP로 가수분해하여 cAMP 합성을 멈추고 G 단백질은 본래 구조로 돌아간다. 그러나, 상기 과정에서 장 세포 내로 들어간 콜레라 독소-GM1 복합체에서 A 서브유닛이 떨어져 나와 G 단백질의 α 서브유닛과 결합하게 되며, G 단백질에 결합된 A 서브유닛은 GTPase의 활성을 막아 활성화된 G 단백질의 GTP 분자가 GDP로 돌아가는 기작을 막아 cAMP 합성이 계속 지속되게 한다. 이에 따라 CFTR이 계속 열려 있게 되어 Cl 분자가 장내로 계속해서 빠져나가게 되고, 결과적으로 삼투현상이 일어나 장 세포와 혈관의 물 분자와 이온들이 장내로 빠져나가 설사와 탈수를 유발하게 되어, 심할 경우 사망에 이르게 할 수 있다.
콜레라 또는 콜레라 독소를 검출하는 방식은 이전부터 많이 제시되어 왔다. 측정 방식의 종류로는 크게 ELISA(enzyme-linked immunosorbent assay) 방식, SERS(surface-enhanced Raman spectroscopy) 방식, PCR(polymerase chain reaction)을 이용한 방식, 전기화학적(electrochemistry) 방식 등이 존재한다. 이중, ELISA 방식은 많은 문헌들에서 다뤄지고 있는 검증된 방식이며 항체만 있으면 어렵지 않게 항원을 특정하여 비색적으로 나타낼 수 있으나, 항체의 가격이 희소성에 따라서 매우 올라가거나 해당 항원에 대한 항체 자체가 존재하지 않을 수도 있으며, 또한 실험을 진행하는데 반복된 노동이 필요하며 측정 결과값을 얻는데까지 오랜 시간이 소요된다는 단점을 가지고 있다. SERS 방식은 라만산란광을 통해 항원의 기판부착을 검지해내는 원리로 감도 높은 선택성을 제공하지만, 마찬가지로 항체를 사용한다는 점과 함께 SERS 기판의 제작이 어려워 재현성이 떨어진다는 단점이 존재하고, 금나노입자를 기반으로 사용하는 방식이기 때문에 금나노입자의 제법이 표준화되어야 최적화된 신호값을 구할 수 있으며, 발색단을 포함하지 않는 단백질들은 SERS 신호가 약하다는 단점이 있다. PCR 방식은 재현성과 민감성, 선택성이 모두 월등하지만, 실험 실행을 위한 전문 장비와 인력이 갖춰져 있으며 비용이 크다는 단점을 가지고 있다. 상기의 방식들 모두 독소 검지 센서로써의 장점들을 어느정도 지니고 있지만, 콜레라 센서를 사용하는 곳이 주로 감염이 잦게 일어나는 빈곤국이라는 사실을 감안할 때, 어렵고 복잡하며 고급 실험설비 없이 측정이 불가능하다는 단점이 크게 작용한다. 또한, 전기화학적 방식을 이용한 콜레라 센서들도 여럿 존재하지만, 항원을 잡는 매개체로 주로 항체, 압타머(aptamer)를 사용한 방식들이 존재하며, 압타머 또한 항체와 마찬가지로 항원에 따라 고비용이고 그 자체로 전극 기판에 부착하려면 추가적인 처리가 필요한 문제가 있다.
본 발명의 목적은 콜레라 검출용 바이오센서를 제공하는 것이다.

본 발명의 전기화학적 방식의 바이오센서는 콜레라 독소가 특이적으로 결합하는 GM1이 풍부하게 존재하는 장내 상피세포막을 작동전극에 도포함으로써 콜레라 독소를 민감하고 선택적으로 검지할 수 있으며, 전기화학적 방식을 사용하여 전극의 임피던스 또는 전류값을 측정할 수 있는 기초장비만 있으면 어디에서나 간편하고 빠르게 콜레라를 검출할 수 있고, 바이오센서의 안정성이 높아 보관 및 배송이 용이해 다양한 지역에 공급하기 유리하므로, 급성으로 생명의 위협이 되며 광범위한 전염을 야기시킬 수 있는 콜레라를 조기에 검진하고 치료할 수 있는 효과가 있다.

본 발명에서 사용된 용어, "강글리오사이드 (ganglioside)"란 여러 개의 당 단위와 한 개 이상의 시알산을 포함하는 세라마이드(ceramide) 유도체로, 당사슬의 수와 시알산의 수에 의해서 일반적으로 GM1, GM2, GM3 등으로 불린다.
본 발명에서 사용된 용어, "콜레라 검출"이란 대상 시료 내에 콜레라 균주 또는 콜레라 독소가 존재하는지 여부를 알아내는 것 또는 시료 내 콜레라 균주 또는 콜레라 독소가 존재할 가능성이 높다고 판정하는 것을 의미하며, 상기와 같은 의미에서 검색, 검사, 검지, 판정 또는 분석 등으로 대체될 수 있다. 본 발명의 목적상, 상기 검출은 작동전극을 포함하는 바이오센서와 콜레라 독소의 직접적인 상호작용에 의한 검출 뿐만 아니라, 콜레라의 검출 표지(marker)와 상호작용을 하고 그 상호작용으로부터 발생한 신호에 의하여 콜레라의 존재 여부를 판정할 수 있는 경우를 포함한다. 바람직하게, 본 발명은 콜레라 독소와 검출 센서와의 상호작용을 전기적 신호로 확인함으로써 시료 내 콜레라 균주 또는 콜레라 독소의 존재를 검출할 수 있다. 따라서, 본 발명의 바이오센서는 콜레라 균주에서 생산되는 콜레라 독소를 검출함으로써 시료 내 비브리오 콜레라의 존재를 검출함을 특징으로 한다.
본 발명에 있어서, '바이오센서'는 생물이 가지고 있는 기능을 이용하여 물질의 성질 등을 조사하는 기계로서 피분석물의 검출을 위해 사용하는 분석장치를 지칭하는 것으로, 크게 감지성 생물적 요소(sensitivity biological element), 전도체 또는 검출 요소(transducer or the detector element) 및 바이오센서 해독 장치(biosensor reader device)를 가지는 것이 특징이며, 본 발명의 목적상 상기 바이오센서는 콜레라 독소 검출용 바이오센서로 해석될 수 있다.
본 발명에서 사용되는 작동전극은 작동전극 상에 세포막의 도포/코팅이 가능하고, 세포막에 전류를 흐르게 할 수 있다면 그 종류에 특별히 제한되지 않는다. 예를 들어, 상기 작동전극은 수은 전극, 아말감 전극, Pt, Au, Pd, Rh 전극과 같은 귀금속 전극, 열분해 흑연(pyrolytic graphite) 전극, 유리 탄소(glassy carbon) 전극, 탄소 페이스트 전극, 탄소 섬유 전극과 같은 탄소 전극, 인듐 산화 주석(Indium tin oxide, ITO) 전극 등일 수 있다.
일 측면에서, 본 발명은 본 발명의 콜레라 검출용 바이오센서를 포함하는 콜레라 검출용 키트에 관한 것이다.
본 발명에 따른 키트는 보다 효과적으로 콜레라 독소를 검출 또는 측정할 수 있도록 본 발명의 바이오센서 외에 채혈 또는 채변 등을 도와주는 도구 등을 추가로 포함할 수 있다. 상기 키트는 외부패키지를 포함할 수 있으며, 외부패키지는 구성요소들의 사용에 관한 사용설명서를 포함할 수 있다.

이하, 본 발명을 실시예를 통하여 더욱 상세히 설명하기로 한다. 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 콜레라 독소 검출용 바이오센서 제작
1-1. Caco-2 세포의 세포막(CCM) 분리
콜레라균으로부터 분비되는 콜레라 독소 중 B 서브유닛(CTxB)이 특이적으로 결합하는 수용체 GM1(monosialotetrahexosylganglioside)을 발현하는 장 상피 세포막을 추출하기 위해 Caco-2 세포를 10% FBS (Sigma-cat#F2442) 및 100units/ml의페니실린-스트렙토마이신 (Sigma-cat#P4333)을 포함하는 MEME (Sigma-cat#M4655 LOT#RNBJ9569) 배지에서 37℃의 온도 및 5%의 CO2 농도 조건의 인큐베이터에서 배양하고 3일에 한번씩 계대배양하였다 (도 1). 70 내지 90%의 밀도로 배양된 세포를 0.25% Trypsin-EDTA 용액 (Sigma - cat#T4049)를 이용하여 배양 접시에서 분리하고, 500×g로 10분 동안 원심분리하여 상등액을 제거함으로써 Caco-2 세포 펠릿을 얻은 뒤, 500×g로 10분씩 3회 세척한 뒤, 프로테이즈 억제제 칵테일을 포함하는 세포 파쇄 용액을 첨가하여 4℃에서 세포를 파쇄하였다. 이 후, 이의 펠릿을 균질기 (Daihan Scientific, Korea)를 이용하여 5000 rpm으로 20 pass 파쇄하였으며, 이의 현탁액을 800×g로 5분 동안, 10,000×g로 10분 동안 및 25,000×g로 20분 동안 순차적으로 원심분리함으로써 Caco-2 세포의 세포막 (미색의 펠릿)을 수득하였다.
1-2. 세포막이 도포된 전극 제작
상기 실시예 1-1에서 Caco-2 세포로부터 분리한 세포막(CCM)을 D.W.에 1000배 희석하여 0.1%의 농도를 가진 용액으로 만든 뒤, 3분 동안 초음파를 처리하는 최적화 조건으로 소낭을 자가형성시켰다. 이 후, 카본(Carbon)전극의 작동전극(Working electrode) 위에 상기 소낭화된 세포막 (0.1%)을 10uL 올리고 습한 조건에서 50℃로 열처리함으로써 세포막을 도포하였다. 이 때, 열처리 시간에 따른 전극의 저항값(EIS)을 확인한 결과, 전극의 저항값이 0~20분까지 지속적으로 증가하다가 30분에 포화되는 것을 확인하여 (도 2), 세포막 용액의 도포 시간을 30분으로 최적화하였다. 30분의 세포막 도포 (열처리) 후 전극을 증류수가 담긴 비커에 넣고 가볍게 흔들어 세척한 후, 먼지가 들어가지 않도록 흄 후드에서 건조시킴으로써 바이오센서의 센서부에 세포막이 코팅(도포)된 전극을 제작하고, 이를 포함하는 콜레라 독소 검출용 바이오센서를 제작하였다.
1-3. 바이오 센서 내 세포막 도포 확인
형광물질이 부착된 인지질인 PE-CF를 이용하여 상기 실시예 1-2에서 제작한 작동전극의 가변과 중앙부에 세포막을 도포하기 전과 세포막을 도포한 후의 형광이미지를 관찰한 결과, 세포막이 전극 위에 잘 도포된 것을 확인하였다 (도 3).
1-4. 전기화학적 측정
상기 실시예를 통해 제작한 콜레라 독소의 B 서브유닛(CTxB)이 특이적으로 결합하는 수용체 GM1이 발현된 장 상피 세포막으로 도포되어 콜레라 독소의 장내 감염 메커니즘 (도 4a)을 이용하여 콜레라 독서를 검출할 수 있는, 본 발명의 바이오센서 (도 4c), 세포막이 도포되지 않은 바이오센서 (도 4b), 및 본 발명의 바이오센서의 전극에 콜레라 독소의 B 서브유닛(CTxB)을 처리한 바이오센서에 각각 전해질 용액 70ul을 전극의 가운데를 중심으로 처리하고 EIS 프로그램을 이용하여 전기화학적 특성을 확인하고, 나이퀴스트 곡선 그래프를 확인하였다.
그 결과, 세포막을 도포하기 전의 바이오센서 (도 4e)에 비해 세포막을 도포한 후의 바이오센서에서 나이퀴스트 곡선 그래프의 반원부분이 커져 전하 전달 저항(Rct)이 커지는 것으로 나타났으며 (도 4f), 세포막을 도포한 본 발명의 바이오센서에 CTxB를 처리한 경우 아닌 경우에 비해 나이퀴스트 곡선 그래프의 반원부분이 커져 전하 전달 저항(Rct)이 커지는 것으로 나타났다 (도 4g).
이를 통해, 본 발명의 장 상피 세포막으로 도포된 작동전극을 포함하는 바이오센서는 시료 내 콜레라 독서의 존재 여부를 상기와 같은 특징적 저항 변화를 통하여 확인할 수 있음을 확인하였다.
실시예 2. 콜레라 독소 검출용 바이오센서와 콜레라 독소의 특이적 결합 확인
상기 실시예 1에서 제작한 콜레라 독소 검출용 바이오센서의 전극 위에 도포된 세포막에 존재하는 GM1과 CTxB가 특이적으로 결합하는지 확인하기 위해, 바이오센서에 Alexa 647이 결합된 CTxB를 처리하고 형광 현미경으로 작동전극을 관찰한 결과, 전극 위에 도포된 세포막의 GM1과 CTxB가 특이적으로 결합한 것을 확인할 수 있었다 (도 5a). 또한, 이를 교차 검증하기 위해, 전극의 표면 형상을 AFM(atomic force microscopy)로 측정하였다. 그 결과, 세포막을 도포한 바이오센서의 표면은 일정한 높이를 가져 단차가 나타나지 않는 반면 (도 5b 및 d), 여기에 CTxB를 처리한 바이오센서의 표면은 CTxB의 존재로 인해 큰 단차가 나타나는 것을 확인할 수 있었다 (도 5c 및 e).
실시예 3. 콜레라 독소 검출용 바이오센서의 검출 민감성 확인
전기화학 임피던스 분광법(Electrochemical Impedance Spectroscopy, EIS) 및 순환 전압 전류법(CV)을 이용하여 상기 실시예 1에서 제작한 콜레라 독소 검출용 바이오센서의 콜레라 독소 검출 성능을 확인하였다. 구체적으로, 세포막이 도포되지 않은 대조군 전극 (Bare electrode)을 포함하는 바이오센서, 본 발명의 바이오센서 (CCB), 및 CCB에 CTxB를 처리한 후의 바이오센서 (CTxB@CCB)를 각각 EIS와 CV로 측정하였으며, 이 때, CCB에 CTxB를 농도별로 처리하여 센서의 민감도를 확인하였다.
그 결과, 본 발명의 바이오센서의 전극 위에 세포막이 잘 도포되었으며, 도포된 세포막에 존재하는 GM1이 CTxB의 수용체로서 작용하는 것을 확인할 수 있었다 (도 6a 및 b). 또한, 콜레라 독소의 각 '농도'별 '임피던스 차이값'의 그래프의 기울기 및 결정계수는 각각 y=71.26 log(X)+300 및 R2=0.9984의 값을 가지는 것으로 나타났으며, 검출최저농도(LOD)는 11.46 nM로 나타났고, CCB에 CTxB를 농도별로 처리한 후 EIS 값을 측정한 결과가 경향성을 보여, 바이오센서로서 활용할 수 있음을 확인하였다 (도 6c 및 d).
실시예 4. 콜레라 독소 검출용 바이오센서의 선택성 및 안정성 확인
본 발명의 콜레라 독소 검출용 바이오센서의 선택성을 확인하기 위해, CTxB와 사람의 장 내에 존재하는 물질들 (AA: acetic acid; UA: uric acid; ASC: ascorbic acid; CA: citric acid; Glu: glucose; Chl: cholesterol; 및 CE: cellulose)을 각각 0.01 mg/mL로 CCB에 처리하고 EIS 값을 측정하였다. 또한, 바이오센서의 안정성을 확인하기 위해, 제작한 CCB를 0 내지 21일 동안 보관한 후 CTxB 0.01 mg/mL를 처리하여 보관 전 후의 EIS 값을 측정하여 비교하였다.
선택성을 확인한 결과, 본 발명의 콜레라 독소 검출용 바이오센서가 다른 물질들에 비해 CTxB에만 매우 높은 선택성을 나타내, 콜레라 독소 검출 센서로서 높은 선택성을 가지는 바이오센서임을 확인하였다 (도 6e). 또한, 안정성을 확인한 결과, 21일의 보관 후에도 같은 농도의 CTxB에 균일한 EIS 값을 나타내, 안정성이 우수한 것을 확인하였다 (도 6f).
Claims (17)
- 콜레라 독소와 특이적으로 결합하는 수용체를 포함하는 세포막이 도포된 작동전극(working electrode)을 포함하는, 콜레라 검출용 바이오센서.
- 제 1항에 있어서, 콜레라 독소와 특이적으로 결합하는 수용체는 강글리오사이드(ganglioside)를 포함하는, 콜레라 검출용 바이오센서.
- 제 2항에 있어서, 강글리오사이드는 GM1(monosialotetrahexosylganglioside), GM2, GM3, asialo GM1, GD1A, GD1B, GT1B, 락토실 세라마이드(lactosyl ceramide) 또는 글로보트리오실 세라마이드(globotriosyl ceramide)를 포함하는, 콜레라 검출용 바이오센서.
- 제 1항에 있어서, 장 상피 세포로부터 분리한 세포막인, 콜레라 검출용 바이오센서.
- 제 1항에 있어서, 세포막은 소낭 형태로 작동전극에 도포된, 콜레라 검출용 바이오센서.
- 제 1항에 있어서, 콜레라 독소가 세포막에 특이적으로 결합하여 전기적 특성이 변화되는, 콜레라 검출용 바이오센서.
- 제 6항에 있어서, 콜레라 독소는 콜레라 독소의 B 서브유닛(CTxB)인, 콜레라 검출용 바이오센서.
- 제 1항에 있어서, 전극에서 인식된 콜레라 독소에 의한 신호를 해독하는 해독(read)부를 추가로 포함하는, 콜레라 검출용 바이오센서.

- 제 13항에 있어서, 열처리는 20 내지 40분 동안 수행되는, 콜레라 검출용 바이오센서의 제조 방법.
