Abstract
코난토킨-G의 분비를 위한 재조합 발현 벡터 및 이로 형질전환된 약독화 살모넬라 균주에 관한 것으로서, flgM 유전자; 및 코난토킨-G 유전자를 포함하는 재조합 발현 벡터로 형질전환된, 약독화 살모넬라 균주, 상기 약독화 살모넬라 균주를 유효성분으로 포함하는 항암용 약학적 조성물, 및 코난토킨-G의 분비능을 향상시킬 수 있는 재조합 발현 벡터를 제공한다.
Description
코난토킨-G의 분비를 위한 재조합 발현 벡터 및 이로 형질전환된 약독화 살모넬라 균주에 관한 것이다. 본 특허출원은 2022년 07년 21일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2022-0090612호에 대하여 우선권을 주장하며, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.

질병의 치료에 가장 효과적인 대응 체계는 면역력을 강화하는 것이나, 암세포는 외부 침입자가 아니기 때문에 인체의 면역반응을 유발하지 않고 회피한다. 특정 바이러스 또는 박테리아는 정상세포보다 암세포에 더 많이 감염되고, 감염된 박테리아가 면역세포의 공격대상이 될 수 있다. 이에 일부러 특정 바이러스 또는 박테리아를 감염시켜 인체 면역반응을 자극함으로써 암세포에 대항할 수 있도록 하는 항암치료 방법들이 제시되었다. 시겔라(Shigella), 콜레라균(Vibrioholera), 병원성 대장균(pathogenic E. coli)과 같은 감염성 세균들은 장관의 세포에 침투할 뿐, 면역반응을 일으키는 중요한 기관인 간과 비장에는 도달하지 못한다. 이와 대조적으로 살모넬라균은 림프절을 통하여 비장과 간에 침투하여 전신면역반응을 자극시킬 수 있다. 구체적으로, Leschner 등(J. Mol. Med. 2010, 88,763-773)은 CT26 종양을 갖는 마우스를 대상으로 형광을 나타내는 살모넬라균을 정맥주사로 감염시킨 후, 시간에 따른 감염 경로를 추적하였다. 그 결과 감염 직후에는 마우스의 혈액을 통해 전신이 감염된 것을 보여주었으며, 감염 20분 후에는 비장과 간에 살모넬라균이 축적되었으며, 24시간 후에는 종양 조직에만 살모넬라균이 집중적으로 축적되어 있음을 관측하였다.
하지만 살모넬라균은 식중독을 유발하는 대표적인 균으로 감염에 의해 패혈증을 유발하여 생명을 위협할 수 있으므로 직접적으로 암 치료에 사용하기에는 병원성이 너무 강하다. 미국 예일대학 연구팀은 살모넬라를 유전자 조작하면 종양을 공격하는 특성은 유지하면서 독성만 제거하여 약독화할 수 있으며, 상기 약독화된 살모넬라의 주입을 통해 면역 자극을 유도하여 종양을 억제할 수 있다고 발표하였다. 그러나 약독화된 살모넬라는 생존성이 낮고 면역유도능 역시 저하될 뿐 아니라 돌연변이로 인하여 약독화 균주가 야생형 살모넬라로 전환되어 패혈증을 일으킬 우려가 있다. 이에 더하여 연구개발자금의 부족 문제로 인하여 약독화된 살모넬라를 이용한 항암치료는 임상과정의 1단계에서 대부분 중단되거나 보류된 상태이다.
살모넬라와 같은 박테리아를 이용한 암 치료에 동반되어 발생할 수 있는 패혈증의 유발은 극복해야 할 매우 중요한 문제이며, 이에 더하여 면역반응의 자극 역시 활발하게 이루어질 수 있어야 한다. 이에 약독화 및 면역 활성화와 함께 항암치료에 도움이 되는 물질들을 추가로 발현 또는 억제시킬 수 있는 균주들을 개발하고, 이를 항암치료에 이용하고자 하는 연구들이 주를 이루고 있다. 등록특허 제10-0852687호는 약독화된 살모넬라 균주 내에 종양괴사인자 알파단백질 벡터를 형질도입한 종양괴사인자 알파를 발현하는 살모넬라 균주 및 이를 함유하는 항암 치료용 조성물에 대해 게시하였으며, 공개특허 제10-2021-0123556호는 필라멘트 성장 조절로 면역반응을 유도하는 항암 살모넬라에 대해 게시하였다.
“코난토킨-G”는 17개의 아미노산으로 구성된 펩타이드로서, 청자고둥(Cone snail)속에 속한 바다 달팽이가 분비하는 독이다. 이것은 NMDAR(N-methyl-D-aspartate receptor)의 길항제(antagonist)로 알려져 있으며 슬리퍼(sleeper) 펩타이드로 알려져 있다. 허혈성 및 흥분독성 및 통증 등에서 신경보호제의 가능성을 보여주며 약물중독 및 알츠하이머의 연구 도구로 잠재력이 있음이 보고되어 있다. 또한 흥분 독성 세포내 칼슘이온의 강도를 감소시키고, 신경 손상을 차단하는 것으로 알려져 있다. 특히 암의 성장에 NMDAR이 연관되어 있어, 종양 치료에 타겟으로 떠오르고 있다.
이에 본 발명자들은 flgM 유전자 및 코난토킨-G 단백질을 암호화하는 유전자를 연결시킨 유전자 구조물을 제조하고, 유전자 구조물을 약독화 살모넬라 균주에 도입함에 따라 flgM와 함께 코난토킨-G의 분비를 가능하게 함으로써, 살모넬라 고유의 암 세포 표적화 및 항암 활성에 더하여, 항암 활성을 보유하는 다량의 코난토킨-G의 작용에 의해 항암 효과를 보다 극대화시킬 수 있음을 확인하고, 이에 기초하여 본 발명을 완성하였다.
본 배경기술 부분에 기재된 상기 정보는 오직 본 발명의 배경에 대한 이해를 향상시키기 위한 것이며, 이에 본 발명이 속하는 기술분야에서 통상의 지식을 가지는 자에게 있어 이미 알려진 선행기술을 형성하는 정보를 포함하지 않을 수 있다.
본 발명의 목적은 우수한 항암효과를 보이는 코난토킨-G 단백질을 효과적으로 분비하기 위한 재조합 발현 벡터 및 이로 형질전환된, 약독화 살모넬라 균주를 제공하는데 있다.
본 발명의 다른 목적은 상기 약독화 살모넬라 균주를 유효성분으로 포함하는 항암용 약학적 조성물을 제공하는데 있다.

본 명세서에서 "작동 가능하게 연결된"은 하나의 기능이 다른 것에 의하여 영향을 받도록 단일 핵산 단편 상에서 뉴클레오티드 서열들이 연결된 것을 나타낼 수 있다. 일 구체예에서, flgM- conantokin-G 단백질을 발현시키기 위해 pBAD18 asd 플라스미드를 벡터로 사용하여 ara 프로모터에 작동 가능하게 유전자가 연결된 flgM-(linker)-conantokin-G-pBAD18의 재조합 발현벡터를 확립하였다. 상기 asd 유전자는 서열번호 10의 염기서열로 이루어질 수 있다.
일 구체예에서, 상기 코난토킨-G의 발현 효율을 향상시키기 위해 본 발명의 유전적 구조물 전단에 샤인-달가노(Shine-dalgarno) 서열이 삽입될 수 있다. 또한, 본 발명의 유전적 구조물을 벡터에 도입할 수 있도록 본 발명의 유전적 구조물 전단과 후단에 적절한 제한효소를 배치할 수 있다.
일 구체예에서, 상기 flgM 유전자는 서열번호 1의 염기서열로 이루어진 것 일 수 있고, 및/또는 상기 코난토킨-G 유전자는 서열번호 2의 염기서열로 이루어질 수 있다. 또한, 상기 재조합 발현 벡터는 flhDC 유전자를 추가로 포함할 수 있으며, 상기 flhDC 유전자는 서열번호 3의 염기서열로 이루어질 수 있다.
본 명세서에서, "변이체 (variants)"는 참조 엔터티(entity)(예컨대, 야생형 서열)와 상당한 구조적 동일성을 나타내지만 하나 이상에서 참조 엔터티와 구조적으로 상이한 엔터티를 지칭한다. 특정 엔터티가 참조 엔터티의 "변이체"로 적절하게 여겨지는지 여부는 참조 엔티티와의 구조적 또는 기능적 동일성의 정도를 기초로 한다. 임의의 생물학적 또는 화학적 참조 엔터티는 특정한 고유의 구조적 요소를 갖으며, 상기 변이체는, 정의상, 하나 이상의 그러한 고유의 구조적 요소를 공유하는 별개의 화학적 엔터티를 지칭한다. 예를 들어, 폴리뉴클레오티드는 선형 또는 3차원 공간에서 서로에 대해 지정된 위치를 갖는 복수의 뉴클레오티드 잔기를 포함하는 고유의 서열 요소를 가질 수 있다. 예를 들어, 폴리뉴클레오티드는 뉴클레오티드 서열의 하나 이상의 차이 및/또는 폴리뉴클레오티드 백본에 공유 결합된 화학적 모이어티(예를 들어, 탄수화물, 지질 등)의 하나 이상의 차이로 인해 참조 폴리뉴클레오티드와 상이할 수 있다. 일부 실시형태에서, 상기 변이체는 적어도 60%, 65%, 70%, 75%, 80%, 85%, 86%, 87%, 88%, 89%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98% 또는 99%의, 참조 폴리뉴클레오티드와의 전체 서열 동일성을 나타낸다.
일 구체예에서, 상기 flgM 유전자; 코난토킨-G 유전자; 및 flhDC 유전자는 동일한 유도성 프로모터에 의해 발현이 조절되는 것일 수 있다. 상기 flgM 유전자; 코난토킨-G 유전자; 및 flhDC 유전자는 동일한 유도성 프로모터에 의해 발현이 조절됨으로써, 상기 flgM 및 코난토킨-G 단백질의 균주 외 분비능이 향상된 것일 수 있으며, 상기 균주를 포함하는 조성물의 항암효과가 향상된 것일 수 있다.
본 발명에 따른 약독화 살모넬라 균주는 flgM 유전자-링커-코난토킨-G 유전자 구조물, 및 flhDC 유전자를 포함하는 재조합 발현 벡터에 의해 형질전환되었으며, 이를 통해, 높은 수준의 코난토킨-G의 발현 및 분비를 유도할 수 있다. 따라서, 일 양상에 따른 재조합 발현 벡터, 이로 형질전환된 약독화 살모넬라 균주 및 이를 포함하는 항암용 약학적 조성물은 암을 치료하는데 활용될 수 있다.

이하 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: 코난토킨-G 분비를 위한 플라스미드의 제작
1-1. flgM 유전자 및 conantokin-G 유전자가 연결된 유전적 구조물의 제작
야생형 살모넬라의 flgM DNA 서열(flgM anti-sigma-28 factor FlgM [Salmonella enterica subsp. enterica serovar Typhimurium str. LT2] Gene ID: 1252690, Locus tagSTM1172 NC_003197.2 (1257036..1257341, complement))과, conantokin-G에 대한 코돈 최적화를 통해 수득한 DNA 서열을 연결시켜, 일 실시예에 따른 유전적 구조물 제작하였다. 구체적으로, 상기 flgM DNA 서열 및 conantokin-G DNA 서열은 링커를 통하여 연결시켰고, flgM 유전자의 발현을 높이기 위해, flgM 유전자 앞에 14개의 샤인-달가노(shine dalgarno) 서열 (AGGAGGTTTGACCT)을 삽입하였다, 또한, 클로닝을 위해 맨 앞에는 NheⅠ site를 삽입하고, conantokin-G의 종결 코돈 뒤에는 SacⅠ site를 삽입하였다. 한편, 본 실시예에서 주요한 구조물의 유전 정보는 하기 표 1에 나타낸 바와 같다.

야생형 살모넬라 LT2의 genomic DNA를 주형으로 하기 표 2의 flhD-sac1-F 및 flhC-Sal1-r 프라이머를 이용하여 PCR에 의해 flhDC 유전자가 포함된 PCR 산물을 얻었으며, 구체적인 PCR 조건은 다음과 같다; 10x buffer 5μl, 5xQ solution 10 μl, 2 mM dNTPs 7.5 μl, 20 μM flhD-sac1-F 프라이머 2.5μl, 20μM flhC-Sal1-r 프라이머 2.5 μl, Taq polymerase 1 μl, 10 ng/μl genomic DNA2 μl 및 물 19.5 μl을 혼합하여 Qiagen system을 사용하여, 95℃ 5분과, 30회의 94℃ 30초, 49℃ 30초, 72℃ 1분 반복과 그 후에 72℃ 10분의 조건에서 PCR을 진행하였다. PCR purification kit로 정제한 PCR fragment 1μg과 sacI과 SalI을 각각 10U를 37℃에서 2시간 반응시키고, 상기 반응 산물을 PCR purification kit로 정제하여 서열번호 3의 flhDC insert DNA를 제작하였다.

37℃에서 2시간 반응시킨 후, 상기 반응 산물을 PCR purification kit로 정제하여 각각의 벡터 DNA를 얻었다. 상기 플라스미드 내에 삽입된 asd 유전자는 서열번호 10의 염기서열로 이루어질 수 있다. 이후, 각 벡터 DNA를 실시예 1-1의 flgM-conantokin-G insert DNA와 25℃에서 30분간 ligation시키고, DH5a 컴피턴트 세포에 형질전환시켰다. 이후, 상기 형질전환된 세포를 LB amp 고체 배지에 도말하고 37℃에서 배양하여 항생제 내성을 갖는 콜로니를 선별하였다. 선별된 콜로니의 후보군 6개를 선별하여 NheⅠ 및 SacⅠ 각각 10 U와 37℃에서 1시간 반응시킨 후 전기영동을 통해 400bp 밴드의 생성을 확인하였다. 이후, pBAD18을 사용하여 제작된 플라스미드에 대해 해당 후보군의 염기서열 분석을 실시하여, 상기 flgM, linker1, linker2, 및 conantokin-G 유전자의 삽입을 확인하고, 일 실시예에 따른 flgM-linker1-conantokin-G-pBAD18 플라스미드 및 flgM-linker2-conantokin-G-pBAD18 플라스미드를 확립하였다.
이후, 상기 제작한 flgM-linker1-conantokin-G-pBAD18 플라스미드 또는 flgM-linker2-conantokin-G-pBAD18 플라스미드 1μg을 sacⅠ 및 SalⅠ 10U와 37℃에서 2시간 반응시킨 후 반응산물을 PCR purification kit로 정제하여 벡터 DNA를 완성하였다. 이후, 각 벡터 DNA를 상기 실시예 1-2의 flhDC insert DNA와 25℃에서 30분 ligation 시킨 후 DH5a 컴피턴트 세포를 형질전환 시켰다. 형질전환된 세포를 LB amp 고체 배지에 도말하고 37℃에서 배양하여 항생제 내성을 갖는 콜로니를 선별하였다. 선별된 콜로니의 후보군 6개를 선별하여 sacⅠ 및 SalⅠ 10U와 37℃에서 1시간 반응시킨 후 전기영동을 통해 band의 생성을 확인하였다. 해당 후보군의 염기서열 분석을 실시하여, flhDC 유전자의 삽입을 확인하고, 이를 통하여, 일 실시예에 따른 flgM-linker1-conantokin-G-flhDC pBAD18 플라스미드 및 flgM-linker2-conantokin-G-flhDC pBAD18 플라스미드를 확립하였다.
실시예 2: flgM-conantokin-G을 분비하는 약독화 살모넬라 균주 제작
상기 실시예 1에서 제조된 플라스미드를 사용하여 약독화 살모넬라 균주를 형질전환시켰다. 약독화 살모넬라 균주로는 aro A aro D asd- 균주를 사용하였으며, 이를 통해 약독화 살모넬라 균주가 아라비노스에 의해 conantokin-G의 발현을 조절하고 이를 세포 밖으로 분비하도록 조절하였다. 상기 형질전환된 각 균주의 단일 콜로니를 LB amp 액체배지에 접종한 후 37℃에서 12~16시간 진탕배양하였다. 이후 신선한 LB amp 액체배지에 OD600이 0.05가 되도록 희석한 후 37℃에서 2시간 진탕배양하였다. 진탕배양에 의해 OD600이 0.4~0.6이 되면 아라비노스(arabinose)를 0.2 w%의 농도가 되도록 처리하여, 37℃에서 5시간 추가로 진탕배양시켰다. 이후, 배양액을 3000 rpm으로 원심분리하여 상층액을 분리하고, 0.2μm 필터로 여과하여 균을 완전히 제거하였다. 상기 균이 제거된 상층액 50μl를 SDS sample buffer와 혼합하여 100℃ 히팅 블록(heating block)에 5분간 변성시킨 후, 5분간 13000rpm 4℃에서 원심분리 하고 10% 폴리아크릴아마이드 겔에 전기영동을 하였다. 1시간 30분 동안 100V로 전기영동 후 Coomassie Brilliant Blue G 250로 염색 후 증류수를 이용하여 탈색하여 웨스턴블랏을 수행하였다. 이를 통해 배양액 중 flgM-conantokin-G의 양을 확인하였으며 나타내었다 (도 4). 상기 도 4에서, 1 및 2 레인은 flgM-linker1-conantokin-G-flhDC pBAD18 플라스미드로 형질전환된 군 (flgM-linker1-conantokin-G-flhDC pBAD18/aroA aro D asd-), 3 및 4 레인은 flgM-linker2-conantokin-G-flhDC pBAD18 플라스미드로 형질전환된 군 (flgM-linker2-conantokin-G-flhDC pBAD18/aroA aro D asd-)이며, 2 및 4 레인은 1 및 3 레인과 달리, 아라비노스를 처리하여 발현을 유도한 군이다.
그 결과, 도 4에 나타낸 바와 같이, 아라비노스가 처리된 2 및 4 레인은 아라비노스가 처리되지 않은 1 및 3 레인보다 동일 농도에서 다량의 양의 flgM-conantokin-G이 분비되었고, 이 중에서도 특정 링커가 적용된 4 레인에서의 분비량은 2 레인에 비해 매우 현저하였다. 상기 결과로부터, 일 실시예 따른 flgM-linker-conantokin-G-flhDC-pBAD18 플라스미드로 형질전환된 균주는 우수한 conantokin-G 분비능을 나타내며, 특히, 14mer 링커가 적용된 flgM-linker2-conantokin-G-flhDC pBAD18/aroA aro D asd- 균주의 분비능은 매우 우수함을 알 수 있었다. 이에, 본 발명자들은 일 실시예에 따른 flgM-linker2-conantokin-G-flhDC pBAD18/aroA aro D asd- 균주를 한국생명공학연구원 생물자원센터에 2023년 6월 26일자로 기탁하고, 수탁번호 KCTC15483BP를 부여받았다 (S-flgM-conantokin-pBAD18-ASD+/BRD509 asd-).
실시예 3: in vitro 에서의 conantokin-G의 항종양 효과 테스트
3-1. 분비된 flgM-conantokin-G의 활성 평가
실시예 2에서 얻은 형질전환된 flgM-linker2-conantokin-G-flhDC pBAD18/aroA aro D asd- 균주의 단일 콜로니를 LB amp 액체배지에 접종한 후 37℃에서 12~16시간 진탕배양하였다. 이후 신선한 LB amp 액체배지에 OD600이 0.05가 되도록 희석한 후 37℃에서 2시간 진탕배양하였다. 진탕배양에 의해 OD600이 0.4~0.6이 되면 아라비노스(arabinose)를 0.2 w%의 농도가 되도록 처리하여, 37℃에서 5시간 추가로 진탕배양시켰다. 비교를 위해서 아라비노스를 처리하지 않은 비교군을 동일 조건에서 진탕배양시켰다. 배양액을 3000 rpm으로 원심분리하여 상층액을 분리하고, 0.2μm 필터로 여과하여 균을 완전히 제거하였다. 이를 통해 flgM-conantokin-G 융합 단백질을 획득하였다.
한편, murine CT26 마우스 결장암 세포주, HT29 및 SW480 인체 대장암 세포주를 구입하여 각각의 세포들을 10% 소태아혈청(FBS) 및 1% 페니실린-스트렙토마이신을 포함하는 DMEM(Dulbecco's modified Eagle's medium)에서 세포를 성장시키고 세포 수가 2 X 105 개 일 때 6-well에 세포를 시딩(seeding)하였다. 24시간 후, 상기 과정을 통해 상층액으로부터 얻은 융합 단백질 200μl를 처리하고, 이를 48시간 배양한 후 관찰하였다.
그 결과, 도 5에 나타낸 바와 같이, 아라비노스를 처리하지 않은 비교군 (-)은 PBS를 처리한 대조군에 비해 약 5% 정도 세포 수준이 감소된 반면, 아라비노스를 처리하여, flgM-conantokin-G의 발현 또는 분비를 유도한 실험군(+)에서는 50% 이상 세포 수준이 감소됨을 확인하였다.
3-2. 암 세포 사멸 유도 효과 평가
암 세포 사멸 유도에 대한 효과를 확인하기 위하여 상기 실시예 3-2의 배양액 및 세포를 사용하여 CCK assay 및 LDH assay를 수행하였다. CCK assay를 위해 상기 배양한 세포주 상층액 100ul를 96well 플레이트에 옮기고 CCK 용액 100ul를 추가한 후, 450nm microreader로 측정하였다. 본 실험은 신뢰성을 높이기 위해 triplicate로 진행하였다. 또한, LDH assay를 위해 상기 배양한 세포주 상층액을 1000rpm에서 3분동안 원심분리하여 세포잔해를 제거하고 상층액 10ul과 LDH 용액 100ul를 혼합하여 실온에서 30분간 인큐베이션하고 450nm microreader로 측정하였다. CCK assay 과 마찬가지로 실험은 triplicate로 진행하였다.
그 결과, 도 6에 나타낸 바와 같이, flgM-conantokin-G의 발현 또는 분비를 유도한 실험군(+)은 비처리 또는 아라비노스를 처리하지 않은 비교군(-)과 비교하여, 검출된 세포 활성 수준이 가장 낮고, 세포가 터져 사멸한 세포의 수준이 가장 높음을 확인하였다. 상기의 실험 결과는, 일 실시예에 따른 형질전환된 약독화 살모넬라 균주는 conantokin-G 고유의 세포 사멸능을 지닌 채로, 제3형 분비계인 flgM과의 연결된 특유의 구조를 통해 그 분비능이 강화된 것임을 실험적으로 입증한 것이다.
실시예 4: 마우스 대장암 모델을 이용한 항종양 효과 평가
본 실시예에서 실험 동물로는 20g 내외의 6 주령 BALB/c 암컷 마우스를 구입하여 사용하였다. 승인된 프로토콜에 따라 모든 동물에 대한 관리 및 실험, 안락사가 수행되었다. 미국 세포주 은행(ATCC)으로부터 murine CT26 결장암종 세포를 구입하여 10% FBS 및 1% 페니실린-스트렙토마이신을 포함하는 DMEM 배지에서 배양하였다. 시험관 내 배양된 CT26 종양세포를 수거한 후, 20μl PBS에 현탁시킨 다음 1×106개의 세포를 BALB/c 마우스의 우측 등에 피하 주사하여 종양세포를 이종 이식하였다. 종양세포의 이식 약 2주 후, 종양의 크기가 100mm3에 도달하면 실시예 2에서 제작한 flgM-linker2-conantokin-G-flhDC pBAD18/aroA aro D asd- 균주를 PBS로 희석하여 약 5×107 CFU/mice용량으로 꼬리 정맥을 통해 주사하였다 (0day). 이후, 상기 마우스를 대상으로 flgM-conantokin-G의 발현 또는 분비 유도를 위하여 40% L-아라비노스 100 μl/mice를 매일 복강내 주사하였고, 매일 일정한 시간을 정하여 마우스의 체중과 종양의 크기를 측정하여 기록하였다. 종양의 부피는 (종양 길이×종양 폭×종양 높이)/2의 식에 의해 계산하였으며, 동물실험 윤리위원회의 지침에 따라 종양 부피가 1,500 mm3이상이되면 안락사시켰다. 한편, 대조군으로는 PBS 완충액만을 주사한 군 (PBS), 비교군으로 약독화 살모넬라 균주인 aroA aroD asd- 균주를 주사한 군 (Salmonella), flgM asd로 형질전환된 aroA aroD asd- 균주를 주사한 군 (Salmonella+flgM), 및 일 실시예에 따른 균주를 처리하되, 아라비노스를 처리하지 않은 군 (Co-)을 이용하였다.
도 7 내지 도 9에 나타낸 바와 같이, 일 실시예에 따른 형질전환된 약독화 살모넬라 균주를 주사에 의해 flgM-conantokin-G의 발현 또는 분비가 유도된 군 (Co+)은 종양의 성장을 현저하게 억제시킴을 확인하였다. 특히, 이러한 항종양 효과는 살모넬라 균주를 주사한 군들 (Salmonella, Salmonella+flgM, Co-)에 비해서도 매우 현저한 것이었다. 이로부터, 일 실시예에 따른 flgM-linker2-conantokin-G-flhDC pBAD18/aroA aro D asd- 균주는 암의 치료를 위한 유효 성분으로 활용할 수 있음을 알 수 있었다.
