Abstract
본 발명은 베시클-단백질 상호작용 (vesicle-protein interaction, VPI)의 형광 이미징 및 정량화를 위한 신규한 프로토콜에 관한 것이다. 본 발명에 따른 염-교체 방법은 종래 알려진 베시클-표지 방법과 비교하여 적은 양의 염료만으로도 간단하고, 신속하게, 그리고 효과적으로 베시클을 표지할 수 있다. 특히, 상기 염-교체 방법은 샘플 내 잔류 염료를 완전히 제거할 수 있어 단일-베시클 수준에서 보다 정확한 베시클의 관찰이 가능하다. 또한, 다양한 세포-유래 베시클이 상기 방법을 통해 효과적으로 표지되는 것이 확인된 바, 본 발명은 프로테오리포좀 등의 인공 베시클부터 천연 베시클에 이르기까지 다양한 종류의 베시클에 범용적으로 적용될 수 있다. 뿐만 아니라, 이와 같이 표지된 베시클은 TIRF (total internal reflection fluorescence) 현미경을 이용하여 단일 베시클 수준에서 관찰할 수 있으므로, 베시클 및 단백질 상호작용의 보다 정확한 검출 및 정량화가 가능하다. 따라서, 본 발명은 베시클 및 타겟 단백질 간 상호작용의 정확한 분석을 위한 신규한 플랫폼을 제공하며, 베시클의 작용 매커니즘을 분석하기 위한 신규한 수단으로 다양한 분야에서 활용될 것으로 기대된다.
Description
본 발명은 세포 유래 베시클 (cell-derived vesicles) 등과 같은 베시클을 표지하는 방법 및 표지된 베시클을 이용하여 베시클-타겟 단백질의 상호작용을 검출하는 방법 등에 관한 것이다.
본 발명은 2022년 6월 8일에 출원된 한국특허출원 제10-2022-0069221호에 기초한 우선권을 주장하며, 상기 출원들의 명세서 및 도면에 개시된 모든 내용은 본 출원에 원용된다.
세포외 소포체 (세포외 베시클; extracellular vesicles, EV)은 원거리에 위치하는 세포 사이의 중요한 커뮤니케이션 수단이다. 이들 나노-규모의 막 (membrane) 결합 패킷 (packets)은 핵산, 단백질과 같은 세포 물질을 세포 사이에서 전달하여 세포간 커뮤니케이션을 매개한다. 이러한 EV의 특성을 활용하여, 최근에는 치료 효과가 있는 EV에 대한 연구가 활발히 이루어지고 있으며, EV를 약물 전달체로 활용하려는 시도들도 이루어지고 있다. 생리학적 또는 약학적 물질의 전달에 있어서, 베시클의 세포로의 흡수 (cellular uptake)를 결정하는 것은 베시클의 표면 단백질과 세포 막의 표면 단백질 사이의 상호작용이다. 관여하는 분자 사이의 집단적 친화도는 베시클-단백질 상호작용 (vesicle-protein interaction, VPI) 수준에 반영되며, 궁극적으로 베시클의 세포 내 혼입 (internalization) 효율을 결정한다.
VPI는 주로, 강하게 상호작용하는 단백질들에 의해 조절되지만, 베시클의 근본적인 가변성이 영향을 미칠 수 있다. 예를 들어, 여러 세포 부착 (cell adhesion) 분자들은 나노-규모의 클러스터로 구성된 올리고머를 형성하여, 베시클이 도킹할 수 있는 플랫폼 역할을 하는 것으로 알려져 있다. 또한, 상호작용하는 주요 단백질들에 인접하게 위치한 기타 조절 단백질들이 VPI의 안정성에 영향을 미쳐 결과적으로 베시클의 세포로의 흡수 효율에 변화를 일으킬 수 있다. EV는 나노-크기이고 무작위로 소량의 단백질들을 포함하고 있기 때문에, 본질적으로 생물물리학적 및 생화학적 특성이 각 단일 베시클마다 서로 이질적이다. 따라서, 베시클과 타겟 단백질의 상호작용을 정확하게 분석하기 위해서는, 단일 베시클 수준에서 VPI를 정확하게 측정할 수 있는 방법이 필요하다.
베시클 샘플의 표준 특성 평가 방법으로서 몇가지 접근법이 제안되어왔다. 특히, 자유롭게 확산하는 베시클에 대한 고-해상도 이미징 혹은 트래킹은 베시클의 크기 분포를 산출하는데 사용되며, 형광 표지된 베시클은 살아 있는 세포에서의 베시클 흡수를 모니터링하는데 사용된다. 그러나, 이의 중간 단계, 즉, VPI를 직접 확인할 수 있는 방법은 아직까지 개발되지 않았다. 특히, 개별 베시클과 단백질 사이의 VPI를 정량적으로 확인할 수 있는 방법이 요구되며, 살아있는 세포를 이용하는 방법에 비해 비용적, 시간적으로 효율적이면서도, 이와 유사한 환경에서 단백질 및 베시클의 상호작용을 관찰할 수 있는 방법에 대한 요구가 있으나, 아직까지 이와 같은 방법은 알려져 있지 않다.
한편, 베시클의 효과적인 라벨링 (표지)는 베시클 및 표적 단백질의 VPI를 정량적으로 확인하기 위한 전제 조건이다. 베시클을 라벨링하기 위해 친유성 염료, 면역염색, 수용성 염료, 베시클 막 염색법 등이 사용되고 있다. 그러나, 단백질을 이용한 라벨링의 경우 이미 베시클 자체에 여러 종류의 단백질이 풍부하게 포함되어 있기 때문에, 정확한 VPI 측정이 어려운 문제가 있다. 수용성 염료의 경우, 정제된 베시클 등에 혼입 (internalization) 시키기가 까다로운 문제가 있다. 베시클 막 염색은 밝기 강도가 매우 높은 선명한 라벨링이 가능하긴 하나, 지질단백질에 대한 off-target 라벨링의 문제 및 베시클과 유사한 크기의 잔류 염료입자(free-dye particles)와 같은 교란요인들이 있다. 친유성 염료를 사용한 기존의 막 염색 프로토콜 역시, 단일 베시클 이미징에서는 효과적이지 않다.
따라서, 베시클을 보다 효율적이고 효과적으로 염색하여, 단일 베시클 및 타겟 단백질 사이의 상호작용을 정밀하게 관찰할 수 있는 방법의 개발이 필요하다.
한편, 세포 유래 베시클 (cell-derived vesicles, CDVs)은 수집 및 농축에 상당한 노력이 요구되는 EV와 달리, 매우 간단한 과정으로 대량으로 수득할 수 있는 베시클이므로, 다양한 연구 및 산업 분야에서 엑소좀 및 엑토좀 등의 천연 EV의 유사체로 활용되고 있다. CDV는 세포를 점차 작아지는 규모의 마이크론-크기 공극을 통해 연속적으로 압출시키거나, 세포를 depth filter에 반복적으로 통과시키면서 Crude-CDV를 준비할 수 있다. 이후, Crude-CDV에서 불순물을 분리·정제하여 손쉽게 대량의 CDV를 수득할 수 있다. 또한, CDV의 막단백질 및 지질은, CDV의 모세포의 원형질막 (plasma membranes) 또는 세포소기관 (organelle mebranes)으로부터 유래한 것이다. 따라서, CDV는 다양한 분야에서의 EV의 적용 가능성을 테스트하는데 활용될 수 있다. 또한, CDV는 치료제로 조작될 수 있으며, 이 경우, VPI의 강도는 CDV의 표적에 대한 타겟팅 효능과 상관관계가 있다. 따라서, CDV를 이용한 다양한 약물 전달 기법, 치료제 개발 등에 있어서도 베시클-타겟 단백질의 상호작용 분석은 매우 중요한 이슈이다.
본 발명은 상기와 같은 문제점을 해결하기 위해 안출된 것으로서, 염 교체 (salt-change) 방법으로 베시클을 라벨링 (표지)하는 경우 베시클에 염료를 높은 효율로 도입할 수 있으며, 상기 라벨링을 이용하여 베시클 및 단백질의 상호작용을 정확하게 측정할 수 있음을 확인하여 완성된 것이다.
따라서, 본 발명의 목적은 베시클 (vesicle)의 표지 방법을 제공하는 것이다.
본 발명의 다른 목적은 상기 방법으로 표지된 베시클 및 단백질 사이의 상호작용을 이미징 하는 방법 내지 스크리닝하는 방법을 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명은 (S1) 제1 완충용액에서 베시클 (vesicles) 및 염료를 배양하는 단계;
(S2) 제2 완충용액에서 상기 베시클 및 염료를 더 배양하여 자유 염료 (free dye)의 응집체 형성을 유도하는 단계; 및
(S3) 상기 단계 (S2)의 배양액을 여과하여 상기 응집체를 제거하는 단계를 포함하고, 상기 제2 완충용액의 염 농도는 상기 제1 완충용액의 염 농도의 5 내지 20배인 것을 특징으로 하는, 베시클의 표지 방법을 제공한다.
본 발명의 일 구현예에서, 상기 베시클은 세포 유래 베시클 (cell-derived vesicles), 프로테오리포좀 (proteoliposomes), 엑소좀 (exosomes), 및 엑토좀 (ectosomes)으로 이루어진 군에서 선택된 하나 이상일 수 있고, 가장 바람직하게는 세포 유래 베시클일 수 있으나, 이에 한정되지 않는다.
본 발명의 다른 구현예에서, 상기 세포는 줄기세포, 중간엽 줄기세포, 유도만능줄기세포, 배아줄기세포, 침샘 줄기세포, 암세포, 선포세포, 근상피세포, 적혈구, 면역세포, 단핵구, 수지상 세포, 자연살해 세포, T 세포, B 세포, 대식세포, 내피세포, 표피세포, 신경세포, 신경아교세포, 성상세포, 근육세포, 및 혈소판으로 이루어진 군에서 선택된 하나 이상일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 염은 NaCl, KCl, CaCl2, NaHCO3, NH4Cl, 및 CuCl2로 이루어진 군에서 선택된 하나 이상일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 미세공극은 셀룰로오스 아세테이트 (Cellulose acetate)로 제조된 것일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 단계 (S2)의 제2 완충용액은 상기 단계 (S1)에서 배양을 마친 후 제1 완충용액에 염 농축액을 첨가하여 제조될 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 제1 완충용액의 염 농도는 전체 완충용액 대비 1 내지 50 mM 일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 제2 완충용액의 염 농도는 전체 완충용액 대비 100 내지 300 mM 일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 단계 (S3)의 여과는 미세공극을 통해 이루어지고, 상기 미세공극의 직경은 0.2 μm 이하일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 염료는 친유성 염료일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 염료는 형광 염료일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 방법은 상기 (S3) 단계 후 베시클을 정제하는 단계를 더 포함할 수 있으나, 이에 한정되지 않는다.
또한, 본 발명은 (S1) 형광 단백질-태깅된 타겟 단백질이 테더링된 글라스를 제조하는 단계;
(S2) 상기 글라스에 상기 표지 방법에 따라 표지된 베시클을 첨가하여 배양하는 단계로서, 상기 베시클은 형광 염료로 표지된 것인, 단계; 및
(S3) 상기 타겟 단백질 및 상기 표지된 베시클의 결합을 관찰하는 단계를 포함하는, 베시클-단백질 상호작용 (vesicle-protein interaction)의 스크리닝 방법을 제공한다.
본 발명의 일 구현예에서, 상기 글라스는 표면이 상기 형광 단백질에 특이적인 항체로 코팅된 것일 수 있으나, 이에 한정되지 않는다.
본 발명의 다른 구현예에서, 상기 단계 (S3)의 관찰은 전반사현미경 (TIRF)을 이용하여 수행되는 것일 수 있으나, 이에 한정되지 않는다.
또한, 본 발명은 (S1) 형광 단백질-태깅된 타겟 단백질이 담지된 지질이중층이 코팅된 글라스를 제조하는 단계;
(S2) 상기 글라스에 상기 표지 방법에 따라 표지된 베시클을 첨가하여 배양하는 단계로서, 상기 베시클은 형광 염료로 표지된 것인, 단계; 및
(S3) 상기 타겟 단백질 및 상기 표지된 베시클의 결합을 관찰하는 단계를 포함하는, 베시클-단백질 상호작용의 스크리닝 방법을 제공한다.
본 발명의 일 구현예에서, 상기 단계 (S1)의 지질이중층은 글라스 위에 제조될 수 있으며, 하기 단계에 의해 제조될 수 있으나, 이에 한정되지 않는다:
(i) 형광 단백질-태깅된 타겟 단백질을 발현하는 재조합 벡터로 형질전환된 세포를 준비하는 단계;
(ii) 상기 형질전환된 세포를 균질화하여 세포 유래 베시클을 수득하는 단계; 및
(iii) 상기 세포 유래 베시클을 리포좀을 혼합하여 융합시킨 후 글라스 위에 로딩하는 단계.
본 발명의 다른 구현예에서, 상기 리포좀은 페길화된 POPC (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 리포좀일 수 있으나, 이에 한정되지 않는다.
본 발명의 또 다른 구현예에서, 상기 단계 (S3)의 관찰은 전반사현미경 (TIRF)을 이용하여 수행되는 것일 수 있으나, 이에 한정되지 않는다.
또한, 본 발명은 상기 표지 방법으로 표지된 베시클, 글라스, 및 지시서를 포함하는 베시클-단백질 상호작용의 스크리닝용 키트로서, 상기 지시서는 상기 스크리닝 방법이 기재되어 있는 것을 특징으로 하는, 키트를 제공한다.
본 발명은 베시클-단백질 상호작용 (vesicle-protein interaction, VPI)의 형광 이미징 및 정량화를 위한 신규한 프로토콜에 관한 것이다. 본 발명에 따른 염-교체 방법은 종래 알려진 베시클-표지 방법과 비교하여 적은 양의 염료만으로도 간단하고, 신속하게, 그리고 효과적으로 베시클을 표지할 수 있다. 특히, 상기 염-교체 방법은 샘플 내 잔류 염료를 완전히 제거할 수 있어 단일-베시클 수준에서 보다 정확한 베시클의 관찰이 가능하다. 또한, 다양한 세포-유래 베시클이 상기 방법을 통해 효과적으로 표지되는 것이 확인된 바, 본 발명은 프로테오리포좀 등의 인공 베시클부터 천연 베시클에 이르기까지 다양한 종류의 베시클에 범용적으로 적용될 수 있다. 뿐만 아니라, 이와 같이 표지된 베시클은 TIRF (total internal reflection fluorescence) 현미경을 이용하여 단일 베시클 수준에서 관찰할 수 있으므로, 베시클 및 단백질 상호작용의 보다 정확한 검출 및 정량화가 가능하다. 따라서, 본 발명은 베시클 및 타겟 단백질 간 상호작용의 정확한 분석을 위한 신규한 플랫폼을 제공하며, 베시클의 작용 매커니즘을 분석하기 위한 신규한 수단으로 다양한 분야에서 활용될 것으로 기대된다.

본 발명은 단일 베시클의 형광-이미징을 위한 베시클의 표지 방법 및 이를 이용한 베시클-단백질 상호작용의 시각화 방법 내지 스크리닝 (또는 검출) 방법에 관한 것이다.
본 발명자들은 구체적인 실시예를 통해 본 발명에 따른 염-교체 방법이 종래의 베시클 표지 방법과 비교하여 더욱 신속하고 간편하게 베시클을 라벨링 ("표지", "염색" 등으로도 지칭됨) 할 수 있음을 확인하였으며, 특히 베시클을 염색하지 않은 잔류 염료를 효과적으로 제거할 수 있어 베시클 관찰의 정확도를 높일 수 있음을 확인하였다.
또 다른 실시예에서는 상기 방법으로 표지된 세포 유래 베시클 (cell-derived vesicles, CDVs)을 이용하여 CDV와 ICAM-1 사이의 베시클-단백질 상호작용 (vesicle-protein interaction, VPI)을 시각화하였다. 실제로, 본 발명자들은 글라스 표면에 테더링되거나 지질이중층에 내장된 (embedded) 타겟 단백질 (ICAM-1)과 CDV 사이의 상호작용을 전반사 현미경 (total internal reflection fluorescence, TIRF)으로 이미징하였으며, 단일-베시클 해상도 (single-vesicle resolution)로 ICAM-1 및 CDV의 VPI를 관찰하고, 정량화할 수 있음을 확인했다.
따라서, 본 발명의 목적은 하기 단계를 포함하는, 베시클 (vesicle)의 표지 방법을 제공하는 것이다:
(S1) 제1 완충용액에서 베시클 (vesicles) 및 염료를 배양하는 단계;
(S2) 제2 완충용액에서 상기 베시클 및 염료를 더 배양하여 자유 염료 (free dye)의 응집체 형성을 유도하는 단계; 및
(S3) 상기 단계 (S2)의 배양액을 여과하여 상기 응집체를 제거하는 단계.
이 때, 상기 제2 완충용액의 염 농도는 상기 제1 완충용액의 염 농도보다 높은 것을 특징으로 한다. 상기 표지 방법은 "염-교체 방법 (salt-exchange method)" 등으로도 지칭될 수 있다.
즉, 본 발명은 종래 사용되던 고농도의 염 (예를 들어, 100 내지 200 mM)을 함유한 완충용액에서 이루어지던 베시클 표지 방법 (직접 염색 방법)에 있어서, 이의 전처리 단계로서 저농도의 염을 함유한 완충용액에서 베시클 및 염료를 배양하는 단계를 추가로 포함하는 것을 특징으로 한다.
바람직하게는, 상기 베시클은 세포 유래 베시클 (cell-derived vesicles), 프로테오리포좀 (proteoliposomes), 엑소좀 (exosomes), 및 엑토좀 (ectosomes)으로 이루어진 군에서 선택된 하나 이상인 것을 특징으로 하며, 가장 바람직하게는, 세포 유래 베시클인 것을 특징으로 한다.
본 발명에 있어서, "세포 유래 베시클 (Cell derived vesicles, CDV)"이란 유핵세포에서 인위적으로 제조된 베시클을 지칭한다. CDV는 거의 모든 종류의 세포에서 세포막으로부터 유리되어 생성될 수 있고, 세포막의 구조인 이중 인지질 (phospholipid) 막 형태를 가지고 있는 것을 특징으로 한다. 본 발명에 따른 세포 유래 베시클은 마이크로미터 수준의 크기를 가진다. 예를 들어 CDV의 직경은 0.2 ㎛ 미만일 수 있다. 더욱 구체적으로, 상기 CDV의 직경은 0.03 내지 1 ㎛, 0.03 내지 0.8 ㎛, 0.03 내지 0.5 ㎛, 0.03 내지 0.3 ㎛, 0.03 내지 0.2 ㎛, 0.03 내지 0.18 ㎛, 0.1 내지 0.2 ㎛, 0.1 내지 1.5 ㎛, 0.1 내지 0.5 ㎛, 0.05 내지 0.3 ㎛, 0.05 내지 0.2 ㎛, 0.01 내지 0.2 ㎛, 또는 0.01 내지 0.3 ㎛ 일 수 있으나, 이에 한정되는 것은 아니다. 본 발명의 세포 유래 베시클은 자연적으로 분비되는 베시클, 예를 들어 엑소좀 (exosome)과는 구별된다. 본 명세서 전반에 있어서, 용어 "베시클 (vesicles)"은 유래한 세포의 세포막 성분으로 이루어진 지질 이중막에 의해 내부와 외부가 구분되며, 세포의 세포막 지질과 세포막 단백질, 핵산 및 세포 성분 등을 가지고 있으며, 원래 세포보다 크기가 작은 것을 의미하지만, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 "세포"는 세포 유래 베시클의 분리가 가능한 세포라면 제한 없이 사용될 수 있으며, 자연계 생물 개체로부터 분리된 세포를 모두 포함한다. 또한, 상기 세포는 인간 및 비인간 포유류를 포함하는 임의 유형의 동물, 식물 유래일 수 있다. 따라서, 본 발명에 따른 세포 유래 베시클을 수득할 수 있는 세포의 종류에는 특별히 제한이 없으나, 예를 들면, 줄기세포, 암세포, 선포세포, 근상피세포, 적혈구, 면역세포, 단핵구, 수지상 세포, 자연살해 세포, T 세포, B 세포, 대식세포, 내피세포, 표피세포, 신경세포, 신경아교세포, 성상세포, 근육세포, 및 혈소판 등으로부터 상기 세포 유래 베시클을 수득할 수 있다. 다양한 종류의 면역세포, 종양세포, 줄기세포, 선포세포, 근상피세포 또는 혈소판일 수 있으며, 바람직하게는, 상기 줄기세포는 중간엽 줄기세포, 유도만능줄기세포, 배아줄기세포 및 침샘 줄기세포로 이루어진 군에서 선택된 어느 하나 이상일 수 있으며, 바람직하게는 지방세포 유래의 중간엽 줄기세포 또는 제대 유래의 중간엽 줄기세포일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 있어서, 상기 베시클은 유핵세포를 포함하는 현탁액을 압출, 초음파 분해, 세포 용해, 균질화, 냉동-해동, 전기천공, 화학 물질 처리, 기계적 분해 및 외부적으로 세포에 힘을 가한 물리적 자극의 처리로 이루어진 군으로부터 선택된 방법을 사용하여 제조할 수 있으나, 이에 제한되는 것은 아니다. 본원발명에서는 예시적으로 유핵세포를 포함하는 현탁액을 압출기를 사용하여 압출하는 방법으로 베시클을 제조하였다. 본 발명의 유핵세포 유래 베시클을 제조하기 위하기 위하여 가하는 압출기의 압출력은 5 내지 200 psi, 10 내지 150 psi, 10 내지 100 psi, 10 내지 50 psi, 10 내지 40 psi, 또는 50 내지 100 psi일 수 있다.
또한, 본 발명에 따른 베시클은 세포를 포함하는 시료를 미세공극에 압출하는 단계를 포함하는 제조방법을 통해 제조될 수 있으며, 바람직하게는 미세공극의 크기가 큰 것으로부터 미세공극의 크기가 작은 것으로 순차적으로 압출하는 단계에 의해 제조될 수 있다. 상기 미세공극의 직경은 0.01 내지 100 μm, 0.01 내지 80 μm, 0.01 내지 60 μm, 0.01 내지 40 μm, 0.01 내지 20 μm, 0.01 내지 10 μm, 0.01 내지 5 μm, 0.01 내지 3 μm, 0.01 내지 1 μm, 0.01 내지 0.5 μm, 0.01 내지 0.1 μm, 5 내지 50 μm, 10 내지 50 μm, 1 내지 10 μm, 또는 1 내지 5 μm 일 수 있으나, 이에 한정되지 않는다. 예를 들어, 상기 순차적인 압출은 공극 직경이 2 내지 10 μm인 필터, 0.5 내지 2 μm인 필터, 및 0.1 내지 1 μm인 필터를 순차적으로 이용하여 이루어질 수 있다. 상기 미세공극의 크기는 사용하는 세포의 종류에 따라 적절히 조절할 수 있다. 상기 과정을 통해 수득한 CDV는 추가 정제 과정을 거칠 수 있다.
한편, 본 발명에 따른 베시클은 세포를 포함하는 시료를 깊이필터 (depth filter)에 통과시켜 압출하는 단계를 포함하는 제조방법을 통해 제조될 수 있다. 상기 깊이필터의 압출력은 5 내지 200 psi, 10 내지 150 psi, 10 내지 100 psi, 10 내지 50 psi, 10 내지 40 psi, 또는 50 내지 100 psi 일 수 있으나, 이에 한정되지 않는다. 상기 과정을 통해 수득한 CDV는 추가 정제 과정을 거칠 수 있다.
본 발명에 있어서, 상기 "세포를 포함하는 시료"는 유핵세포 또는 이의 형질전환된 세포를 포함하는 시료일 수 있으며, 베시클의 제조가 가능한 세포라면 제한 없이 포함하는 개념이다.
본 발명에 있어서, 상기 압출은 미세공극을 포함하는 것으로서 샘플의 여과가 가능한 형태의 멤브레인 (membrane) 구조의 필터라면 제한 없이 사용할 수 있으나, 바람직하게는 폴리카보네이트 멤브레인 필터일 수 있다.
이 밖에도, 본 발명에 따른 세포 유래 베시클은 지지체, 중간 필터, 및 멤브레인 필터 등을 포함하는 세포 압출기를 이용하여 제조될 수 있다. 상기 압출기에 대한 구체적인 설명은 대한민국 등록특허공보 제10-2134906호에서 확인 가능하다.
본 발명에 따른 베시클의 표지 방법은, 제2 완충용액의 염 농도가 상기 제1 완충용액의 염 농도보다 높은 것을 특징으로 한다. 즉, 제2 완충용액의 염 농도는 제1 완충용액의 염 농도에 비해 증가된 것을 특징으로 한다.
즉, 본 발명은 저염 완충용액 (제1 완충용액)에서 베시클 및 염료를 배양하여 베시클의 표지 (라벨링)를 유도한 후, 저염 완충용액을 고염 완충용액 (제2 완충용액)으로 교체함으로써 자유 염료 (free dye)의 응집을 유도하고, 이어서 미세공극을 이용한 여과 과정을 통해 상기 염료 응집체를 제거하는 것을 특징으로 한다. 따라서, 상기 방법에 의하면, 최종적으로는 샘플 내 잔류 염료가 모두 제거된 상태로 표지된 베시클만을 수득할 수 있다.
본 발명에 있어서, "증가"는 대조군과 비교하여 적어도 1%, 2%, 3%, 4%, 5%, 10% 또는 그 이상, 예를 들어, 5%, 10%, 20%, 30%, 40%, 또는 50%, 60%, 70%, 80%, 90% 또는 그 이상 높은, 및/또는 0.5배, 1.1배, 1.2배, 1.4배, 1.6배, 1.8배, 2배, 3배, 4배, 5배, 6배 또는 그 이상 높은 수준을 나타낸다.
따라서, 본 발명에 있어서, 상기 제2 완충용액의 염 농도는 상기 제1 완충용액의 염 농도의 5 내지 20배, 5 내지 15배, 5 내지 10배, 5 내지 8배, 5 내지 6배, 2 내지 10배, 3 내지 10배, 4 내지 10배, 4 내지 8배, 또는 4 내지 7배일 수 있으나, 이에 한정되는 것은 아니다.
또한, 본 발명에 있어서, 상기 제1 완충용액의 염 농도는 전체 완충용액 대비 1 내지 50 mM, 1 내지 40 mM, 1 내지 30 mM, 1 내지 25 mM, 1 내지 20 mM, 1 내지 10 mM, 5 내지 50 mM, 10 내지 50 mM, 10 내지 40 mM, 10 내지 30 mM, 10 내지 25 mM, 15 내지 30 mM, 15 내지 25 mM, 또는 18 내지 22 mM 일 수 있으나, 이에 한정되는 것은 아니다.
또한, 본 발명에 있어서, 상기 제2 완충용액의 염 농도는 전체 완충용액 대비 100 내지 300 mM, 100 내지 250 mM, 100 내지 200 mM, 100 내지 180 mM, 100 내지 160 mM, 110 내지 160 mM, 120 내지 160 mM, 130 내지 160 mM, 140 내지 160 mM, 130 내지 200 mM, 140 내지 200 mM, 또는 150 내지 200 mM 일 수 있으나, 이에 한정되는 것은 아니다.
본 명세서에서 사용된 용어, "완충용액 (buffer solution, 또는 buffer)"은 pH 완충용액 또는 수소이온 완충용액을 의미하는 것으로서, 약산 및 짝염기 혹은 이의 반대의 혼합물로 이루어진 수성 용액을 지칭한다. 상기 완충용액은 일반적으로 산이나 염기를 가해도 공통이온 효과에 의해 수소 이온 농도 (pH)가 일정하게 유지된다. 또한, 본 발명의 완충용액은 삼투압을 일정하게 유지하는 등장액의 역할도 하여 세포 유래 베시클이 온전한 상태를 유지할 수 있도록 한다.
상기 완충용액은 구체적인 종류에 제한되는 것은 아니나, 바람직하게는 PBS (Phosphate-buffered saline), 포스페이트 버퍼 (phosphate buffer), 아세테이트 버퍼 (acetate buffer), 생리식염수, 및 해패스 (HEPES) 버퍼등으로부터 선택될 수 있다. 바람직하게는, 상기 완충용액은 PBS 또는 포스페이트 버퍼이다. 제1 완충용액 및 제2 완충용액은 서로 동일한 종류일 수 있고, 혹은 상이한 종류의 버퍼일 수 있으나, 바람직하게는 동일한 종류인 것을 특징으로 한다.
또한, 상기 염은 NaCl, KCl, HEPES, CaCl2, NaHCO3, NH4Cl, 및 CuCl2로 이루어진 군에서 선택된 하나 이상일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 따른 베시클의 표지 방법에 있어서, 상기 단계 (S2)의 배양은 단계 (S1)에서 배양을 마친 후 제1 완충용액을 제2 완충용액으로 버퍼를 교체함으로써 이루어진다. 상기 버퍼-교체는 구체적인 방법에 제한되지 않으나, 바람직하게는 상기 단계 (S1)에서 배양을 마친 후의 제1 완충용액에 염 농축액을 첨가하여 이루어질 수 있다. 즉, 상기 단계 (S2)의 제2 완충용액은 상기 단계 (S1)에서 배양을 마친 이후의 제1 완충용액에 염 농축액을 첨가하여 제조될 수 있다. 예를 들어, 제1 완충용액으로 PBS를 사용한 경우, 염료 및 베시클의 배양을 마친 PBS에 고농도의 NaCl을 포함하는 PBS 농축액을 첨가하여 전체 용액의 NaCl 농도를 증가시킴으로써, 단계 (S2)를 수행하기 위한 제2 완충용액을 제조할 수 있다.
본 발명에 있어서, 상기 단계 (S3)의 여과는 미세공극을 통해 이루어지는 것을 특징으로 한다. 예컨대, 상기 여과는 미세공극을 포함하는 멤브레인 구조의 필터, 예컨대 시린지 필터 등을 사용하여 수행될 수 있다.
바람직하게는, 상기 단계 (S3)의 미세공극은 셀룰로오스 아세테이트 (Cellulose acetate)로 제조된 것일 수 있다. 즉, 상기 미세공극을 포함하는 멤브레인은 셀룰로오스 아세테이트로 이뤄진 것일 수 있다. 셀룰로오스 아세테이트로 제조된 미세공극이 아닌 경우, 베시클을 여과했을 때 베시클의 손실율이 클 수 있으므로, 셀룰로오스 아세테이트 멤브레인을 사용하는 것이 바람직하다.
상기 단계 (S3)의 미세공극의 직경은 바람직하게는 베시클의 직경보다는 크고, 염료 응집체의 직경보다는 작은 것을 특징으로 한다. 즉, 상기 미세공극은 단계 (S2)에서 생성된 염료 응집체는 통과시키지 않고, 염색된 베시클만을 통과시키므로, 베시클을 제외한 염료 응집체만 선택적으로 제거할 수 있다. 따라서, 본 발명은 잔류 염료를 제거하기 위한 복잡한 정제과정 (예를 들어, 밀도 구배 혹은 원심분리)를 필요로 하지 않는다.
상기 미세공극의 직경은 0.3 μm 미만, 0.25 μm 미만, 0.2 μm 이하, 0.2 μm 미만, 또는 0.1 μm 미만일 수 있으나, 이에 한정되는 것은 아니며, 당업자는 베시클 및 염료의 종류에 따라 적절한 미세공극을 선택할 수 있다. 보다 구체적으로는, 상기 미세공극의 직경은 0.01 내지 0.3 μm, 0.01 내지 0.25 μm, 0.01 내지 0.2 μm, 0.1 내지 0.2 μm, 0.1 내지 2.5 μm, 0.1 내지 0.3 μm, 0.15 내지 0.25 μm, 또는 0.15 내지 0.2 μm 일 수 있으나, 이에 한정되지 않는다.
본 발명에 있어서, 상기 염료는 베시클을 표지할 수 있는 것이라면 제한 없이 사용될 수 있으나, 바람직하게는 친유성 염료인 것을 특징으로 한다. 구체적인 실시예에서, 본 발명자들은 본 발명에 따른 표지 방법이 다양한 친유성 염료에 적용될 수 있는 것을 확인하였다. 따라서, 본 발명에 따른 표지 방법은 친유성 염료에 범용적으로 사용될 수 있는 것으로서, 염료는 친유성을 갖는 것이면 충분하고, 구체적인 종류에 제한되는 것은 아니나, DiI, DiD, DiO, DiA, 및 DiR 등을 예로 들 수 있다.
또한, 본 발명에 있어서, 상기 염료는 형광 염료인 것을 특징으로 한다. 따라서, 본 발명에 따른 표지 방법에 의해 형광 염료로 표지된 베시클은 형광현미경 등으로 관찰이 가능하다.
본 발명에 따른 표지 방법으로 표지된 베시클의 경우, 하나의 베시클당 1 내지 5개, 1 내지 4개, 1 내지 3개, 또는 1 내지 2개의 염료 분자를 함유할 수 있다. 즉, 본 발명에 따른 방법에 의해 특정 염료로 표지된 베시클 샘플에서, 상기 염료 분자는 단일-베시클의 이미징을 가능하게 하는 밀도로 분포하는 것을 특징으로 한다.
본 발명에 따른 표지 방법은 상기 (S3) 단계 후 베시클을 정제하는 단계를 추가로 포함할 수 있으나, 이는 선택적인 것에 불과하다. 상기 정제는 당업계에 알려진 베시클 정제 방법을 제한 없이 사용할 수 있다.
또한, 본 발명은 염료, 완충용액, 염, 미세공극을 포함하는 필터, 및/또는 지시서 등을 포함하는, 베시클 표지용 키트를 제공한다. 상기 지시서에는 본 발명에 따른 베시클 표지 방법이 기재되어 있을 수 있다.
본 발명에 있어서, "키트 (kit)"는 구체적인 형태나 종류에 제한이 없으며, 당업계에서 통상적으로 사용되는 형태의 키트를 사용할 수 있다. 예를 들어, 본 발명에 따른 키트는 상기 구성 성분들 외에도 보관 용기, 초순수 (ultrapure water), 시린지 등을 추가로 포함할 수 있다.
또한, 본 발명은 하기 단계를 포함하는, 베시클-단백질 상호작용 (vesicle-protein interaction, VPI)의 스크리닝 방법 (검출 방법, 정량화 방법)을 제공한다:
(S1) 형광 단백질-태깅된 타겟 단백질이 테더링된 글라스를 제조하는 단계;
(S2) 상기 글라스에 상기 표지 방법에 따라 표지된 베시클을 첨가하여 배양하는 단계로서, 상기 베시클은 형광 염료로 표지된 것인, 단계; 및
(S3) 상기 타겟 단백질 및 상기 표지된 베시클의 결합을 관찰하는 단계.
즉, 상기 스크리닝 방법은 글라스 표면에 테더링된 타겟 단백질과 베시클 간의 상호작용을 이미징 혹은 가시화하는 것을 목적으로 한다. 이 때, 상기 형광 단백질-태깅된 타겟 단백질은 2 종류 이상일 수 있으며, 이 때 각 타겟 단백질을 태깅하는 형광 단백질은 서로 다른 파장의 형광을 생성하는 것이 바람직하다.
타겟 단백질은 베시클과의 상호작용 여부를 확인하고자 하는 임의의 관심 단백질 (proteins of interest)을 의미한다.
상기 스크리닝 방법은 형광 (fluorescence) 검출에 기반한 것으로서, 타겟 단백질 및 베시클 모두 형광으로 표지되는 것을 특징으로 한다.
상기 타겟 단백질은 형광 단백질 (예를 들어, GFP, eGFP, RFP, mCherry, DsRed 등)로 태깅되어 형광-표지된 것을 특징으로 한다. 이 때, 상기 형광 단백질 태그는 바람직하게는 상기 타겟 단백질의 C-말단에 융합되어, 베시클 및 타겟 단백질 사이의 상호작용을 방해하지 않는 것을 특징으로 한다.
상기 글라스는 상기 형광 단백질에 특이적으로 결합하는 항체로 코팅된 것을 특징으로 한다. 따라서, 형광 단백질-태깅된 타겟 단백질을 글라스에 처리하면, 상기 형광 단백질이 글라스 상의 항체와 결합하므로 타겟 단백질이 글라스 표면에 부착될 수 있다. 즉, 상기 타겟 단백질은 형광 단백질-항체의 결합을 통해 글라스 표면에 테더링 될 수 있다.
일 구현예에서, 상기 글라스는 표면이 polyethylene glycols (PEG), Biotin, 및/또는 Avidin 단백질로 코팅될 수 있다.
또한, 상기 스크리닝 방법은 상기 단계 (S2) 이후에 시료를 완충액으로 세척하여 타겟 단백질에 결합하지 않은 베시클을 제거하는 단계를 추가로 포함할 수 있다.
본 발명에 있어서, 상기 단계 (S3)의 관찰은 전반사현미경 (Total Internal Reflection Fluorescence, TIRF)을 통해 수행될 수 있다. 상기 전반사현미경은 레이저를 광원으로 사용하며, 레이저를 전반사시켜 만들어진 evanescent wave를 통해 커버글라스로부터 약 200 nm까지의 거리에서 일어나는 현상을 관찰할 수 있는 현미경이다. TIRF를 이용하면, 일반 형광현미경이나 공초점현미경으로는 관찰이 불가능한 베시클등의 막 표면 상의 현상을 관찰할 수 있다. 특히, 본 발명에 따른 표지 방법으로 표지된 베시클은 단일-베시클 관찰이 가능한 정도의 밀도의 염료로 표지되므로, TIRF 현미경을 이용한 형광 스팟의 관찰을 통해 단일 베시클을 이미징할 수 있으며, 베시클 및 타겟 단백질 각각의 형광 표지를 검출함으로써 베시클-단백질 상호작용을 단일 베시클 수준에서 검출할 수 있다.
또한, 본 발명은 하기 단계를 포함하는, 베시클-단백질 상호작용 (vesicle-protein interaction, VPI)의 스크리닝 방법 (검출 방법, 정량화 방법)을 제공한다:
(S1) 형광 단백질-태깅된 타겟 단백질이 담지된 지질이중층이 코팅된 글라스를 제조하는 단계;
(S2) 상기 글라스에 상기 표지 방법에 따라 표지된 베시클을 첨가하여 배양하는 단계로서, 상기 베시클은 형광 염료로 표지된 것인, 단계; 및
(S3) 상기 타겟 단백질 및 상기 표지된 베시클의 결합을 관찰하는 단계.
즉, 상기 스크리닝 방법은 실제 세포막 단백질과 같이, 막 (membrane)에 내장 (embedded)된 상태의 타겟 단백질과 베시클 간의 상호작용을 가시화하는 것을 목적으로 한다. 즉, 상기 스크리닝 방법은 원형질막과 유사한 지질이중층에 담지된 상태의 타겟 단백질과 베시클의 상호작용 여부를 관찰할 수 있으므로, 실제 세포와 유사한 환경에서의 VPI 확인을 가능하게 한다. 상기 지질이중층은 글라스 표면 위에 코팅된 것으로서, "지지된 지질이중층 (supported lipid bilayer, SLB)"로 지칭될 수 있다.
상기 단계 (S3)의 관찰은 위에서 서술한 바와 같이 전반사현미경 (TIRF)을 이용하여 수행될 수 있다.
형광 단백질-태깅된 타겟 단백질 및 상기 관찰에 대한 설명은 위에서 서술한 바와 같다.
본 발명에 있어서, 상기 단계 (S1)의 지질이중층은 글라스 위에 제조되는 것으로서, 하기 단계에 의해 제조될 수 있다:
(i) 형광 단백질-태깅된 타겟 단백질을 발현하는 재조합 벡터로 형질전환된 세포를 준비하는 단계;
(ii) 상기 형질전환된 세포를 균질화 (homogenization 또는 sonication)하여 세포 유래 베시클을 수득하는 단계; 및
(iii) 상기 세포 유래 베시클 및 리포좀을 혼합하여 함께 융합시킨 후 글라스 위에 로딩하는 단계.
상기 단계 (i)의 세포는 재조합 벡터로 형질전환되어 재조합 벡터가 코딩하는 단백질을 발현할 수 있는 것이라면 제한이 없으나, 바람직하게는 HEK 세포일 수 있다. 여기서, 상기 재조합 벡터는 형광 단백질-태깅된 타겟 단백질을 코딩하는 유전자를 하나 이상 포함할 수 있다. 즉, 상기 타겟 단백질은 2 종류 이상일 수 있으며, 이 때 각 타겟 단백질을 태깅하는 형광 단백질은 서로 다른 파장의 형광을 생성하는 것이 바람직하다.
상기 단계 (ii)에서 생성된 세포 유래 베시클은 상기 단계 (i)의 재조합 벡터로부터 발현된 단백질이 담지된 것을 특징으로 한다. 상기 단계 (ii)의 세포 유래 베시클을 수득하는 단계는 구체적으로 다음의 단계로 나뉠 수 있다:
(ii-1) 형질전환된 세포를 homogenization하여 조혼합물 (crude mixture)를 수득하는 단계;
(ii-2) 상기 조혼합물을 500 내지 3,000×g에서 원심분리하여 상층액을 수득하는 단계; 및
(ii-3) 상기 상층액을 50,000 내지 200,000×g에서 원심분리하여 베시클 펠렛을 수득하는 단계.
상기 (ii-2) 단계의 원심분리는 500 내지 3,000×g, 500 내지 2,000×g, 500 내지 1,500×g, 700 내지 1,500×g, 또는 800 내지 1,200×g에서 수행될 수 있으나, 이에 한정되지 않는다.
상기 (ii-3) 단계의 원심분리는 50,000 내지 200,000×g, 70,000 내지 200,000×g, 80,000 내지 200,000×g, 50,000 내지 150,000×g, 70,000 내지 120,000×g, 또는 80,000 내지 120,000×g에서 수행될 수 있으나, 이에 한정되지 않는다.
또한, 상기 형질전환된 세포의 세포 유래 베시클은 위에서 서술한, 압출 과정을 이용한 세포 유래 베시클 수득 방법을 통해 수득할 수도 있다.
상기 단계 (iii)의 리포좀은 세포 유래 베시클과 융합 후, 글라스 표면에 로딩시켰을 때 지지된 지질이중층 형성을 촉진하는 합성 리포좀인 것이 바람직하다. 구체적인 종류로 제한되는 것은 아니나, 상기 리포좀은 천연 리포좀 또는 합성 리포좀일 수 있다. 바람직하게는, 상기 리포좀은 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) 리포좀일 수 있고, 더욱 바람직하게는 페길화된 리포좀일 수 있다. 일 구현예에서, 상기 리포좀은 클로로포름에 용해된 POPC를 PEG5000-ceramide와 혼합하여 제조됐다.
상기 단계 (iii)에서 상기 세포 유래 베시클 펠렛은 리포좀 용액과 혼합한 후 sonication 등을 이용하여 함께 융합된다.
이 때, 상기 세포 유래 베시클 : 리포좀의 혼합 비율은 1 : 2 내지 30, 1 : 2 내지 25, 1 : 2 내지 20, 1 : 2 내지 15, 1 : 2 내지 10, 1 : 2 내지 5, 1 : 5 내지 30, 1 : 10 내지 30, 1 : 15 내지 30, 또는 1 : 10 내지 20 (v:v) 일 수 있으나, 이에 한정되지 않는다.
상기 세포 유래 베시클-리포좀이 융합된 하이브리드 베시클이 포함된 용액을 글라스 표면 위에 로딩하면, 글라스에 의해 지지되는, 타겟-단백질이 내장된 지질이중층이 완성된다.
또한, 본 발명은 본 발명에 따른 방법으로 형광-표지된 베시클, 글라스 및 지시서 등을 포함하는, 베시클-단백질 상호작용의 스크리닝용 키트를 제공한다. 키트에 대한 설명은 상술한 바와 같다.
상기 지시서에는 본 발명에 따른 VPI 스크리닝 방법이 기재되어 있을 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실험예]
[실험 재료 및 방법]
1. CDV (Cell-derived vesicles, CDV)의 제조
NK-CDV, U937-CDV, 및 UCMSC-CDV는 [Jang, S. C.; Kim, O. Y.; Yoon, C. M.; et al., ACS Nano 2013, 7 (9), 7698-7710 및 Zhang C. et al., Nanomedicine. 2021 Oct;37:102448]에 기술된 방법에 따라 제조됐다. 먼저 세포를 PBS에 각각 세포 종류에 따라 5×106개 세포/ml, 2.5×105개 세포/ml, 및 2.5×105개 세포/ml의 농도로 재현탁시키고, 폴리카보네이트 (polycarbonate) 트랙-에칭 친수성 멤브레인 필터 (Cytiva, Whatman)를 통과시켜 압출하였다. NK-CDV의 경우 pore 직경이 5, 1, 및 0.4 μm인 멤브레인을; U937-CDV의 경우 pore 직경이 10, 3, 및 0.4 μm인 멤브레인을; UCMSC-CDV의 경우 pore 직경이 10, 3, 및 0.4 μm 멤브레인을 연속적으로 사용하여 압출했다. 이어서, CDV를 함유한 용액에 DNase (Merck Millipore, Benzonase Nuclease HC)를 처리하여 CDV 외부의 DNA를 제거하였으며, 3000×g에서 10분 동안 원심분리하여 응집체 (aggregates)를 제거했다. 그리고, CDV를 tangential flow filtration system (Repligen, KrosFlo KR2i)로 정제하여 CDV 외부의 단백질, 핵산 등의 불순물을 제거하였으며, 0.45-μm 필터를 사용해 최종 여과했다. CDV의 농도 및 크기 분포는 나노입자 트래킹 분석 (Particle Metrix, Zetaview)을 사용해 측정했다. 정제된 CDV는 사용 전까지 ~1×1011개 입자/ml의 농도로 -80 ℃의 온도에서 보관했다.
2. CDV의 특성 분석
SDS-PAGE로 CDV 샘플의 단백질을 분석하기 위해, NK-CDV, U937-CDV, 및 UCMSC-CDV를 각각의 기원 세포의 펠렛과 함께 1% Triton X-100 (Sigma)로 용해시키고, 총 단백질 농도를 측정했다 (Bio-Rad, 5000112). 이어서, 동량의 20 μg 샘플을 단백질 분리를 위해 8% polyacrylamide gel 상에 로딩하고, Coomassie brilliant blue R-250로 염색했다. 웨스턴 블롯팅의 경우, 동일한 농도의 CDV 샘플 (2×109개 입자/ml)를 SDS (Sigma)로 용해시켰다. 샘플을 10% (CD11a, CD18 분석) 및 12% (CD63 분석) polyacrylamide gel 상에 로딩하여 단백질을 분리하고, PVDF 멤브래인에 트랜스퍼 했다. 트랜스퍼가 완료된 멤브레인은 TBS (tris-buffered saline)에 포함된 5% skim milk로 1시간 동안 실온에서 블로킹했다. 이어서, 멤브레인을 1차 항체인 CD11a (BioLegend, 301202), CD18 (Abcam, ab176540), 및 CD63 (BioLegend, 353017)와 4 ℃에서 밤새 인큐베이션했다. 이후, 멤브레인을 TBS 에 포함된 Tween-20 (TBST)로 세척하고, 2차 항체 (Abcam, ab205718 및 ab205719)로 실온에서 1시간 동안 인큐베이션하였으며, 그 후 TBST로 다시 세척했다. 단백질 밴드는 화학발광 반응액 (Cytivia, RPN2232)으로 가시화했다. 나노입자 트래킹 분석 (nanoparticle tracking analysis, NTA)의 경우, CDV 샘플을 필터링된 PBS로 ~109개 입자/ml의 농도로 희석하고, NTA 장비 (Nanosight, LM10-HS)에서 405-nm 레이저로 조광했다. 각 CDV 샘플을 분석하기 위해 30번의 측정이 수행됐다.
3. CDV의 형광 라벨링 (Fluorescent labeling)
PBS에 포함된 CDV 용액을 초순수 (ultrapure water)로 희석하여 NaCl 농도를 20 mM 미만으로 낮추고, 37 ℃에서 20분 동안 2 μg/ml DiI 또는 DiD와 반응시켰다. 이어서 소량의 PBS 농축액 (10Х)을 첨가하여 NaCl 농도를 ~150 mM으로 증가시켰다. 혼합물은 0.2-μm 시린지 필터 (Advantec, 13CP045AS)로 여과하여 염료 분자 응집체를 제거하였으며, 추가 실험을 수행했다.
4. 형광 라벨링 효율 측정
베시클 라벨링 효율을 측정하기 위해, DiI (1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine; DiIC18(3))-염색된 CDV 샘플을 CD63 항체 (BioLegend, 353017)가 표면-테더링된 PEG-코팅된 글라스 커버슬립 상에 로딩했다 (도 5a 참고). Home-built total internal reflection (TIRF) 현미경 상에서 샘플에 523-nm 레이저 (30 mW)를 조사했으며, 베시클이 광표백 (photobleached) 될 때까지 전 시간에 걸쳐 베시클의 형광 스팟을 촬영했다. 수득한 이미지로부터, 각 스팟의 형광 광도를 추적하여 광표백 동안 단계 크기를 결정했다.
5. 단백질 발현
형광 단백질과의 태깅 (tagging)을 위해, ICAM-1-코딩 ORF 및 CD9-코딩 ORF를 각각 pEGFP-C1 및 pmCherry-C1 벡터에 클로닝했다. 플라스미드의 일시적 발현을 위해, polyethyleneimine (Sigma)으로 플라스미드를 HEK 세포에 형질주입했다. 세포는 10% fetal bovine serum (Welgene) 및 1% penicillin-streptomycin (Welgene)로 보충된 Dulbecco's modified Eagle's medium (Sigma)에서 37 ℃및 5% CO2의 조건으로 24시간 동안 배양됐다. 배양을 마친 세포는 15,000×g에서 10분 동안 원심분리하여 수확했다. 수확된 세포는 사용 전까지 -80 ℃에서 보관했다. VPI 분석에 사용될 형광 단백질의 준비를 위해, 세포를 1% Triton X-100을 함유한 버퍼로 용해시켰다. 원심분리로 세포 잔해물 (cell debris)를 제거한 후, 형광 단백질의 농도를 flurometer로 측정했다.
6. 표면-테더링된 (tethered) 단백질을 이용한 VPI 이미징
TIRF 현미경 분석을 위한 이미징 챔버는 공지된 방법을 따라 제조했다. 간략히 서술하면, 샘플 쳄버는 양면 테이프를 사용해 글라스 커버슬립 및 글라스 슬라이드를 함께 접착시켜 제조했다. 글라스 표면은 polyethylene glycols (PEG) (Laysan Bio, mPEG-SVA, MW 5,000)로 코팅했으며, 이 코팅된 PEG 중 3%는 biotin (Laysan Bio, BIO-PEG-SVA, MW 5,000)으로 태깅되어, NeutrAvidin (Thermo Fisher, 31000)를 테더링하는데 사용되었다. ICAM-1-eGFP의 표면 테더링을 위해, NeutrAvidin, biotin-conjugated GFP antibody (Abcam, ab6658), 및 ICAM-1-eGFP 용액을 샘플 챔버에 순차적으로 주입하되, 각 단계별로 5 내지 10분 동안 인큐베이션한 후 PBS (Biosesang, pH 7.2)으로 세척했다. CD9 및 ICAM-1/CD9 co-pulldown 실험을 위한 표면 준비는 biotin-conjugated RFP antibody (Abcam, ab34771) 및 CD9-mCherry 용액을 추가로 사용하는 것 외에는 유사하게 수행됐다. 필요한 경우, VPI 측정 전에 표면-테더링된 표적 단백질을 이미징 및 정량화했다. 이어서, 형광으로 라벨링된 CDV를 단백질-코팅된 표면에 도입하고, 15분 동안 상호작용할 수 있도록 인큐베이션한 후, PBS로 세척하여 결합되지 않은 베시클을 제거했다. VPI 블로킹 실험은, 면역블롯팅 분석과 유사하게, CDV에 먼저 블로킹 항체 (5 μg/ml)를 처리한 후, VPI를 측정했다.
7. SLB (supported lipid bilayers)에서의 VPI 이미징
합성 페길화 리포좀 (Synthetic PEGylated liposomes) 및 HEK 세포 멤브레인을 각각 제조한 후 혼합하여 지지된 지질이중층 (supported lipid bilayer) [Pace, H. P.; Hannestad, J. K.; Armonious, A.; et al., Chem. 2018, 90 (21), 13065-13072.]의 형성을 촉진했다. 합성 베시클의 경우, 클로로포름에 용해된 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC; Avanti, 850457C)를 7.4 μg의 PEG5000-ceramide (Avanti, 880280P)와 혼합했다. 멤브레인의 형광 이미징이 필요한 경우, 상기 혼합물에 0.002 내지 2 nmol의 DiD를 선택적으로 첨가하였으나, 일반적인 VPI 측정시에는 첨가하지 않았다. 상기 지질 혼합물은 진공 하에서 완전히 건조시켰으며, 1 ml PBS에 재현탁하고, pore 직경이 100-nm인 필터 (Avanti, 610023)를 통해 압출하여 PEG-POPC 베시클 용액을 수득했다. HEK 세포 멤브레인의 경우, ICAM-1-eGFP 및/또는 CD9-mCherry를 발현하는 ~4×107 개 HEK 세포를 수확한 후, 균질화 (homogenization; DH.WHG02016) 방식으로 세포를 파괴하였다. 세포 파괴 후 수득한 조혼합물 (crude mixture)을 1,000×g에서 30분 동안 원심분리하여 큰 잔해 (debris)를 제거하고, 상층액을 100,000×g에서 30분 동안 2회 더 원심분리하여 멤브레인-유래 베시클(HEK-CDV)을 수집했다. 원심분리된 HEK-CDV 베시클 펠렛은 PBS에 재현탁하고, 상기 PEG-POPC 베시클 용액과 혼합하였다. 혼합 비율은 전반적으로 5:95 내지 20 : 80 (HEK-CDV : PEG-POPC 베시클) (v:v) 이었다. 이어서 15분간의 초음파 처리(sonication)를 통해 베시클을 융합하여 하이브리드 베시클을 형성했다. SLB의 자발적 형성을 유도하기 위해, 하이브리드 베시클을 piranha-세척된 글라스 커버슬립 위에 로딩했다. 최종적으로, VPI 측정을 위해 생성된 SLB에 형광 라벨링 된 CDV 샘플을 로딩했다.
8. 광표백 후 형광 회복 (Fluorescence recovery after photobleaching, FRAP) 분석
지지된 이질 지중층 (supported lipid bilayers, SLBs)의 유동성을 확인하기 위해, FRAP 실험을 수행했다. 반경이 ω인 균일한 원형의 관심 영역에서 시간 경과에 따른 형광 강도 (초기 형광으로 정규화됨)의 부분 회복 (fractional recovery)는 다음 식에 따라 모델링됐다:
실시예에서, ω은 FRAP이 이루어지는 이질 지중충의 직경인 9 μm가 사용 되었다.
9. 단일-베시클 면역블롯팅 분석
LFA-1의 검출을 위해, 라벨링되지 않은 CDV (1×1010개 입자/ml)를 anti-CD11a 항체 (BioLegend, 301202) 및 anti-CD18 항체 (Abcam, ab176540) (5 μg/ml)와 함께 1시간 동안 인큐베이션했다. 샘플을 18,000×g에서 1시간 동안 원심분리하여 CDV에 결합하지 않은 자유 항체를 함유한 상층액은 제거하고, 수집된 베시클은 PBS에 재현탁했다. 추가 정제를 위해, 원심분리 후 상층액을 제거하고 재현탁하는 과정을 반복했다. 상기 실험방법 6을 참고하여, 이미징 챔버에서의 면역블롯팅을 위해, 상기 anti-CD11a 항체 및 anti-CD18 항체가 결합된 CDV를 비오틴화 (biotinylated)된 2차 항체 (Jackson ImmunoResearch, 715-065-150) 및 NeutrAvidin를 통해 PEG-코팅된 표면에 테더링했다. 이어서 단일-베시클 염색을 위해 anti-CD11a 항체 및 anti-CD18 항체에 결합할 수 있는 Cy3-결합된 2차 항체 (Jackson ImmunoResearch, 715-165-150)를 첨가했다.
10. TIRF 현미경 및 이미지 분석
샘플 슬라이드는 조광 (illumination) 및 이미징을 위해 도립현미경 (Olympus IX73) 및 60Х오일-침지 렌즈를 사용하여 home-built TIRF 현미경으로 이미징했다. 조광 (Cobolt)에 사용된 레이저 파장은 다음과 같다: eGFP의 경우 488 nm, mCherry 및 DiI의 경우 532 nm, 및 DiD의 경우 633 nm. 이미지는 일반적으로 100-ms 해상도 및 ~100-μm의 시계 (field of view)를 가진 sCMOS 카메라 (Teledyne Photometrics, Prime BSI Express)로 촬영했다. 형광의 정량화는 영상의 초기 10개 프레임을 평균화하고, 사용자 지정 MATLAB 코드를 사용하여 형광 스팟의 수 또는 전체 영역에 대한 총 형광강도를 측정함으로써 이루어졌다.
[실험 결과]
1. CDV의 형광 라벨링
1-1. 제조된 CDV의 특성 확인
일반화할 수 있는 (generalizable) 방법으로 지질 베시클의 형광 라벨링 방법을 개발하기 위해, CDV를 세포외베시클 (EV)의 모델로 사용했다. 상술한 실험방법 1.과 같이, 본 실험에 사용된 CDV는 공개된 방법에 따라 인간 기원의 자연살해세포 (natural killer cells)로부터 제조하였다. (이하, "NK-CDV"로 지칭함). 나노입자 트래킹 분석 (NTA)을 통해 NK-CDV의 95% 이상이 300 nm 미만의 직경을 가지며, 평균적으로는 140 nm의 직경을 가지는 것을 확인했다. 즉, 본 실험에서 사용된 CDV는 세포에서 자연적으로 분비되는 엑소좀과 상당히 유사한 크기를 갖는 것을 확인했다 (도 1a). U937 또는 UCMSC 로부터 제조한 CDV도 크기 분포가 NK-CDV와 거의 동일했으며, 각 세포에서 유래한 CDV의 전반적인 단백질 조성은 대체로 모세포의 프로테옴 (proteomes)를 반영하는 것으로 나타났다 (도 1b). 상기 결과는 CDV의 생산이 물리적으로나 화학적으로 편향되지 않았음을 보여준다.
1-2. 염색 방법 차이에 따른 CDV 형광 라벨링 효율 확인
1-2-1. 직접 염색 방법에 따른 CDV 형광 라벨링 효율
이어서, VPI 이미징을 위해 친유성 형광 염료로 CDV를 라벨링하고, 형광 현미경에서 시각화했다 ("직접 염색 방법"; 도 2a의 A 부분). 용액의 얇은 층 내에 존재하는 형광 입자를 선택적으로 이미징하기 위해, 글라스 커버슬립의 표면으로부터 ~200 nm 깊이까지 조광하는 TIRF 현미경을 사용했다 (도 2a의 C 부분). 직접 염색 방법을 이용해 PBS에서 표지된 CDV를 TIRF 현미경으로 이미징한 결과, 소수의 희미한 형광 스팟만이 검출되었다 (도 2b). 즉, 이는 종래 베시클 표지 방법의 라벨링 효율이 매우 낮다는 것을 보여준다. 본 발명자들은 라벨링의 낮은 효율이 PBS에서의 친유성 염료의 낮은 가용화 (solubilization)에 기인한 것이라고 추측했다. 관찰된 표지 수준은 다량의 베시클 또는 세포막을 조사하기 위한 대량 분석에는 적합할 수 있으나, 단일 베시클 수준의 정량적 분석을 할 수 있을만큼 형광이 충분히 밝지는 않았다 (도 2b 내지 2d).
1-2-2. 염 농도에 따른 염료 입자의 특성 변화 확인
라벨링 효율을 제한하는 요소를 확인하기 위해, 다양한 농도의 NaCl를 함유한 수성 매질에서 DiI의 상태를 직접 확인했다. TIRF 현미경으로 이미징했을 때, ~150 mM NaCl이 포함된 인산염 완충액 중의 DiI 분자는 베시클 막에 삽입될 수 없을 정도로 큰 크기의 응집체를 형성하는 것이 확인됐다 (도 3a). 샘플을 0.2-μm 직경의 pore로 필터링하였을 때, 상기 응집체들이 완전히 제거되는 것이 확인된 바, 입자들이 대부분 마이크론 크기임을 알 수 있었다 (도 3b). 또한, NaCl의 농도가 ~20 mM로 낮아짐에 따라 희미한 입자들의 수가 증가하는 것이 확인된 바, 응집체들이 점차 분산되는 것을 알 수 있었다 (도 3a 내지 3c). 염에 의한 이러한 응집체의 변화는 DiI의 농도는 동일하게 유지되는 상태에서 발생했는데, 이는 (detergent에 의해 완전히 용해된 후) 샘플의 전체 (bulk) 형광 강도에 변화가 없는 것으로부터 알 수 있었다 (도 3d). 반면, 미세공극 (micropore) 필터링 이후 detergent-용해된 형광은 최소였는데, 이는 고염 버퍼에 존재하던 염료 분자의 대부분이 응집체에 포획되어 제거되었음을 시사한다.
1-2-3. 염-교체 방법에 따른 CDV 형광 라벨링 효율 확인
상기 결과는 저염 버퍼에서의 DiI의 분산이 DiI의 막 분할 (membrane partitioning)을 증진시킬 수 있으며, 과잉의 염료는 고염 버퍼에서 다시 염료의 응집을 유도함으로써 제거할 수 있음을 시사한다. 따라서, 본 발명자들은 가역적 DiI 응집을 이용하여 CDV의 라벨링 방법을 개선하고자 하였다. 먼저, CDV를 저염 버퍼 (20 mM 미만의 NaCl)에서 염색한 후, 버퍼를 고염 버퍼 (~150 mM NaCl)로 교체하여 자유 염료 분자의 응집을 유도하고, 이어서 미세공극 (pores)을 지닌 통상적인 시린지 필터를 사용하여 필터링하는 것이다 ("염-교체 방법"; 도 2a의 B 부분). 염-교체 방법으로 CDV를 염색한 후 TIRF 현미경으로 관찰한 결과, 형광 베시클의 수가 약 80배 증가한 것을 확인할 수 있었다 (도 2b 및 2c). 또한, 라벨링된 베시클들의 형광 밝기는 기존 PBS 방법으로 염색된 것에 비해 2배 이상 더 밝았다 (도 2d). 형광 베시클의 수 및 밝기가 모두 증가한 것은, DiI의 베시클로의 도입이 전반적으로 2배 이상 증가하였음을 의미한다 (도 2d의 곡선 아래 면적으로 계산됨). 특히, NTA로 측정했을 때, 염-교체 염색방법은 CDV 샘플의 크기 분포를 왜곡시키지 않는 것으로 나타났다 (도 4a). 반면, 자유 DiI는 필터링에 의해 거의 완전히 제거된 것으로 나타났다 (도 3d). 라벨링 및 필터링 이후에도 베시클의 60% 이상이 일관되게 회수되는 것으로 나타났다. (도 4b).
1-3. 표면-테더링된 단백질을 활용한 라벨링된 CDV의 형광 효율 확인
라벨링 밀도 (예를 들어, 베시클당 염료 분자의 수)를 더욱 면밀히 측정하기 위해, 실험방법 4을 통해 준비된 anti-CD63 항체가 표면에 테더링된 글라스에서 실험방법 3을 통해 라벨링된 CDV를 안정적으로 캡쳐하고 형광 강도를 측정했다. 이 데이터를 사용하여, 초기 형광에서 광표백 단계 크기에 이르기까지 각 베시클 내의 염료 분자의 수를 측정했다 (도 5a 내지 5e). 결과적인 단계 크기는 ~65 였다 (본 실험의 TIRF 설정에서 측정 단위) (도 5b). 이어서, 초기 프레임 (0 내지 4초)의 단일 베시클의 형광을 확인된 단계 크기와 비교했다. 분포는 다시 비슷한 간격 (~65)으로 몇 가지 균등한 간격의 Gaussian 분포를 나타냈으며, 이는 베시클에 소수의 DiI 분자가 포함되어 있음을 의미한다 (도 5c). 마지막으로, 모든 베시클로부터 초기 강도를 확인하여 분석한 결과 평균이 122임을 확인했다 (도 5d). 따라서, 라벨링 효율은 122/65 = 1.9로 계산됐다. 추가로, 단계 크기에 의해 이산화된 강도 분포는 평균이 1.9인 Poisson 분포에 잘 맞았으며, 베시클당 소수의 염료 분자의 무작위성과 일치하는 것으로 확인됐다 (도 5e). 종합하면, 단일 CDV에는 일반적으로 1 내지 3개의 DiI 분자가 함유되어 있으며, 이들 수는 Poisson 분포를 따른다는 것을 알 수 있었다 (도 5e). 상기 결과는 베시클의 작은 부분만이 염색되지 않은 채로 남아 있으며, 동시에 지질막에서의 염료의 몰분율 (mole fraction)은 0.01% 미만을 차지할 것임을 나타낸다. 즉, 이는 본 발명에 따른 염-교체 방법으로 달성한 베시클의 라벨링 밀도가 단일-베시클 이미징에 충분할 뿐만 아니라, 막의 고유 특성을 방해할 정도로 극단적이지도 않다는 것을 의미한다. DiD를 사용한 실험에서도, CDV 염색과 유사한 결과가 확인됐다 (도 6). 즉, 이는 염-교체 염색방법이 본 실험에 개시된 DiI 또는 DiD 염료 외에 기타 친유성 염료에도 일반적으로 적용 가능하다는 것을 보여준다.
2. 타겟 단백질의 글라스 표면 부착
실험방법 3을 통해 준비한 형광-라벨링된 CDV를 사용하여, 실험방법 6에서 서술한 내용과 같이 TIFR 현미경에서 VPI를 측정하는 방법을 설계했다 (도 7a). 이 방법에서, 타겟 단백질 분자는 항체를 사용하여 글라스 표면에 특이적으로 테더링시켰으며, 형광 CDV를 첨가하여 표면 결합 단백질과 상호작용하도록 했다. 이 때, TIRF 광학장치는 글라스 표면 근처의 얇은 용액 시트를 조광하기 때문에, 표면-테더링된 타겟 단백질과 상호작용하는 CDV의 형광 신호만이 선택적으로 검출되며, 자유롭게 떠다니는 CDV에 의한 백그라운드 형광은 감지되지 않는다. 또한, 안정한 VPI를 구체적으로 확인하기 위해 인큐베이션 이후에 CDV 용액을 제거할 수 있다.
2-1. eGFP-tagged ICAM-1의 특성 확인
원리-증명 (proof-of-principle) 실험을 위해, 본 발명자들은 CDV와 표면-테더링된 ICAM-1 사이의 상호작용을 측정했다. ICAM-1은 백혈구와 내피세포 사이의 부착을 컨트롤하여 염증반응을 조절하는 주요 세포부착 분자이다. 베시클과 관련하여, ICAM-1은 면역세포 유래 EV 또는 치료용 EV의 타겟으로 기능할 수 있다. ICAM-1-코팅된 표면을 준비하기 위해, eGFP로 태깅된 재조합 ICAM-1을 HEK293 (HEK) 세포에 과발현시키고, 상기 세포를 용해시켜 얻은 추출물로부터 GFP 항체를 사용해 eGFP-태깅된 재조합 ICAM-1을 풀다운 했다 (도 7a). eGFP를 사용하면 복잡한 단백질 정제 과정 없이 거의 모든 타겟 단백질을 간단하게 포획할 수 있을 뿐만 아니라, 풀다운된 단백질의 형광-기반 정량화도 가능하다. EGFP 태그는 ICAM-1의 C-말단에 융합되어 세포 내측 도메인에 위치해 있으므로, 막 국소화 (membrane localization)을 교란하거나 세포외도메인을 방해하지 않는다 (도 8a 및 8b).
광학적 CMOS 카메라가 장착된 TIRF 현미경은 개별 GFP 분자의 형광 신호를 안정적으로 감지할 수 있다 (도 7b). 동시에, 센서의 높은 동적 범위는 전체 밝기를 기반으로 높은 밀도의 ICAM-1 집단을 정확하게 측정할 수 있다 (도 7c). 예를 들어, 세포 용해물의 인풋 (input) 양을 증가시키면, 형광 스팟의 직접 카운팅은 ~1 molecule/μm2의 광학 해상도까지 신뢰할 수 있고, 총 형광 강도는 표면 점유에 의해 제한될 때까지 더 증가할 수 있다. 이러한 방식으로, 본 실시예에서 얻을 수 있는 ICAM-1-eGFP의 최대 밀도는 ~40 molecules/μm2로 계산됐다. 이 상한선은 분자간 간격으로 환산하면 평균적으로 ~160 nm에 해당하며, 이 간격 내로 인접한 ICAM-1 분자들은 단일 CDV를 끌어들이는데 협력할 수 있다. 또한, 일부 ICAM-1 분자는 세포 용해 동안 불완전하게 해리된 이량체 또는 더 큰 올리고머로 포획될 수 있다. 실제로, ICAM-1 용액이 고농도로 유지됐을 때, 풀다운된 형광 반점은 저농도의 대응물에 비해 평균적으로 최소 2배 더 밝았으며 (도 8c 및 8d), 이는 다량체 복합체 (multimeric complexes)의 잠재적 기여를 시사한다.
3. CDV 및 표면-테더링된 단백질 사이의 VPI
ICAM-1-코팅된 표면에서의 VPI 이미징을 위해, 형광 CDV 용액을 첨가했다. VPI 측정은 다양한 베시클 샘플의 타겟-결합 특성을 비교하는데 유용하다. 이를 위해, 위에서 사용된 NK-CDV 외에도, 2가지 유형의 CDV를 더 준비(실험방법 1)하여 VPI를 측정했다: 하나는 pro-monocytic U937 세포에서 유래한 것이고 (U937-CDV), 다른 것은 재대혈 중간엽 줄기세포 (umbilical cord mesenchymal stem cells)에서 유래한 것이다 (UCMSC-CDV). TIRF 조광이 일시적인 상호작용을 포함하여 ICAM-1 표면 근처의 모든 CDV를 가시화하기 때문에, 10분의 배양 후 소포 용액을 세척해 제거하고, 안정적인 CDV 및 타겟 단백질간 결합을 분석하는데 집중했다 (실험방법 6).
3-1. CDV와 표면-테더링된 ICAM-1의 상호작용 확인
그 결과, 라벨링된 베시클 및 ICAM-1-코팅된 표면 사이의 강력하고 특정적인 (specific) 상호작용을 관찰할 수 있었다 (도 7d). 베시클의 표면 결합은 ICAM-1을 반드시 필요로 하며, 베시클-단백질의 결합 수준은 ICAM-1의 양에 거의 비례하는 것으로 나타났다 (도 7e). 3가지 CDV 샘플 중, NK-CDV 및 U937-CDV는 ICAM-1과 지속적으로 강한 상호작용을 나타낸 반면, UCMSC-CDV는 실험에 사용된 농도 범위 어디에서도 결합이 관찰되지 않았다. 흥미롭게도, U937-CDV의 결합은 ~1 molecule/μm2 밀도의 ICAM-1에서도 관찰됐으나, NK-CDV의 결합은 ~10 molecules/μm2의 밀도에서부터 검출되며, ICAM-1 밀도에 대해 다소 초-선형 의존성 (super-linear dependence)을 갖는 것으로 나타났다. 이러한 결과는 인접한 ICAM-1 분자가 NK-CDV의 결합에 협력할 수 있는 반면, 분리된 ICAM-1 단량체는 U937-CDV와 함께 LFA-1 매개 VPI를 유도하기에 충분할 수 있음을 암시한다. 한편, ICAM-1 표면을 동일하게 준비하고, NK-CDV 및 U937-CDV의 인풋 농도를 증가시켰을 때, VPI 신호가 증가하는 것이 확인됐다 (도 7f). NK-CDV 및 U937-CDV의 VPI는 베시클 농도에 대한 2차 (quandratic) 의존성을 보였는데, 이는 타겟 결합에서 베시클 간의 약한 협력성이 관여한다는 것을 나타내는 것일 수 있다. 표면-결합된 CDV는 1일 후에도 안정적인 결합을 유지하였는데 (도 9), 이는 살아있는 세포의 베시클 흡수를 과정에 있어서 이러한 베시클-단백질 상호작용이 영향을 줄 수 있음을 시사한다.
3-2. ICAM-1과 상호작용하는 CDV의 단백질 확인
다음으로, CDV의 어떤 단백질이 ICAM-1 의존성 VPI를 매개하는지 확인했다. ICAM-1은 본래 모든 주요 유형의 백혈구에서 널리 발현되는 이종이량체 인테그린 (αLβ2 또는 CD11a/CD18)인 LFA-1의 강력한 리간드로 알려져 있다. 또한, EV 표면 상에서의 LFA-1-ICAM-1 상호작용은 면역학적 관점에서 연관되어 있다. 따라서, 본 발명자들은 U937-CDV 및 NK-CDV의 LFA-1가 표면-테더링된 ICAM-1을 인식하여 VPI를 유도하는 것이라고 추측했다. 이를 확인하기 위해, 본 발명자들은 3가지 다른 수준에서 LFA-1의 연관성을 확인했다. 첫째, 웨스턴 블롯팅 결과 U937-CDV 및 NK-CDV에서만 상당한 양의 LFA-1 서브유닛 (CD11a 및 CD18) 단백질이 존재하는 것이 확인됐다 (도 10a). 다음으로, 온전한 베시클에서 단백질 분자를 특이적으로 검출할 수 있는 단일-베시클 면역블롯팅 분석을 수행하였다(실험방법 9, 도 10b). 상기 방법은 위의 VPI 측정과 동일한 화학적, 광학적 설정을 사용하여 베시클을 확인하기 때문에, 기능적 상호작용자의 양에 대한 강력한 기준 역할을 한다. 단일-베시클 면역블롯팅 분석 결과, U937-CDV 및 NK-CDV에서 LFA-1 분자의 존재가 재확인됐다 (도 10c, 10d). 마지막으로, LFA-1의 ICAM-1-결합 영역을 차단하였을 때 VPI에 변화가 있는지 확인하기 위해, LFA-1 항체 (anti-CD11a 항체, anti-CD18 항체)를 이용했다. 그 결과, LFA-1의 세포외도메인을 타겟팅하는 항체를 첨가했을 때 베시클의 ICAM-1 결합이 두드러지게 감소하는 걸 확인할 수 있었다 (도 10e). 반면, 항-mCherry 항체를 사용한 음성 대조군 (mock)은 베시클 결합에 영향을 미치지 않았다. 종합하면, 상기 결과들은 LFA-1 및 ICAM-1의 상호작용이 베시클과 ICAM-1 간의 VPI를 매개하는 핵심 요소임을 분명하게 보여준다.
4. 관련 인자에 의한 VPI의 조절
상기 실험예들은 주어진 베시클 및 표적 단백질의 VPI를 정량적으로 측정하는 방법과 그 결과를 보여준다. 그러나, 생리학적 환경에서 베시클의 표적 단백질 결합은 금속 이온, 조절 단백질 등과 같은 주변 환경의 다른 요소들에 의한 영향을 받을 수 있다. 이와 같은 기타 변수들의 조절 효과를 확인하기 위해, 버퍼 조성 또는 표면 조성에 변화를 가해 분석을 진행했다. 구체적으로, LFA-1-ICAM-1 상호작용에 영향을 미치는 것으로 알려진 Mg2+ 및 테트라스파닌 (tetraspanin) 단백질 CD9에 의해 NK-CDV 와 ICAM-1 사이의 VPI가 어떻게 변화하는지 확인했다.
4-1. Mg
2+
의 변화에 따른 LFA-1-ICAM-1 상호작용 확인
LFA-1-ICAM-1 상호작용의 생리학적 활성화 및 후속적인 세포-세포 상호작용은 기타 표면 수용체 및 액틴 세포골격과도 연관이 있으나, Mg2+는 LFA-1에 대한 ICAM-1의 결합을 직접 조절할 수 있다 (Dransfield, I. et al, J. Cell Biol. 1992, 116 (1), 219-226). 따라서, 본 발명자들은 Mg2+가 LFA-1 의존성 VPI를 유사하게 촉진하는지 확인하고자 하였다. Ca2+가 없는 상태에서 1 내지 5 mM의 Mg2+를 첨가하면, NK-CDV 및 ICAM-1 사이의 상호작용이 Mg2+가 처리되지 않은 U937-CDV의 VPI 수준에 도달할 때까지 점진적으로 증가했다 (도 11a). 본 발명자들은 상기 VPI의 증가는 생리학적 반응과 유사한 LFA-1의 구조적 변화로 인한 것이라고 추측했다. 대조적으로, U937-CDV에서 관찰된 높은 수준의 VPI는 Mg2+를 추가해도 더 이상 증가하지 않았다. 최소 ~9×104개 베시클/mm2까지 카운팅할 수 있음을 고려하면 (도 7f), 도 11a에 나타난 U937-CDV의 VPI 검출 범위(약 2.5×104 개 베시클/mm2)에 의해 측정된 VPI 수준이 제한된 것은 아니다.
4-2. VPI에 대한 ICAM-1 및 CD9의 시너지 효과 확인
테트라스파닌 CD9는 대부분의 백혈구와 내피세포에서 광범위하게 발현되어 ICAM-1과 같은 세포부착 단백질의 활성을 조절한다. 따라서, 본 발명자들은 표면-테더링된 ICAM-1 근처에 있는 CD9 분자가, 세포부착에서의 CD9의 생리학적 기능과 마찬가지로, NK-CDV의 부착을 돕는지 확인하고자 하였다. mCherry로 태깅된 재조합 CD9 단백질은 ICAM-1-eGFP와 함께 HEK 세포에서 공동-발현되었으며, PEG-코팅된 글라스에 풀다운됐다 (실험방법 6). Co-immunoprecipitation 실험과 달리, ICAM-1 및 CD9는 서로 독립된 항체를 이용하여 풀다운됐다 (도 11b). 따라서, 이 두 단백질의 인접여부는 오로지 우연에 의해서만 결정된다. 다양한 인풋 농도로 공동-발현된 단백질을 풀다운하면, 비슷한 양의 ICAM-1 및 CD9를 함유하는 여러 표면이 제조된다 (도 11c, "ICAM-1/CD9"). ICAM-1 및 CD9 사이의 시너지 효과를 구체적으로 평가하기 위해, 각 표면에서 다양한 밀도 범위로 존재하는, 표면-테더링된 ICAM-1 및/또는 CD9을 포함하는 글라스를 준비했다 (도 11d). 그 다음, 3가지 케이스 모두에 대해 병렬적으로 VPI 측정을 수행했다. 실제로, 유사한 양의 개별 단백질들을 사용해 얻은 결과와 비교했을 때, VPI는 ICAM-1 및 CD9가 혼합되어 테더링된 표면에서 현저하게 높은 것으로 나타났다 (도 11e, 11f). 더욱 놀랍게도, 상기 ICAM-1 및 CD9가 테더링된 표면에서의 VPI 수준은 각각의 단백질로 테더링된 표면에서 산출된 결과의 합보다 훨씬 높은 것으로 드러났다 (도 11g). 상기 결과는 CD9 및 ICAM-1 사이의 시너지적인 협력에 기인한 것으로 추측된다.
5. 막에 내장된 (membrane-embedded) 단백질과의 VPI 측정
상기 실험예는 베시클과 타겟 단백질 사이의 상호작용을 명확하게 관찰할 수 있으나, 지질막이 없는 상태에서 관찰이 이루어진다는 문제가 있다. 이에, 개발된 방법을 보완하기 위한 다음 단계로서, 지질이중층 표면 상에서 동일한 VPI를 조사할 수 있는 방법을 고안했다 (실험방법 7, 도 12a).
5-1. 타겟 막 단백질을 포함하는 지지된 지질이중층(SLB)의 특성 확인
타겟 막 단백질을 포함하는, 지지된 지질이중층 (supported lipid bilayers, SLBs)를 제조하기 위해, ICAM-1-eGFP 및 CD9-mCherry를 포함하는 HEK 세포의 세포막을 균질화 (homogenization) 방식으로 파괴하여 HEK-CDV를 생성하고, 생성된 HEK-CDV를 SLB 형성을 촉진하는 합성 리포좀(PEG-POPC 리포좀)과 함께 융합시킨 뒤, 깨끗한 글라스 표면에 로딩시켰다 [Pace, H.; Simonsson Nystrom, L.; Gunnarsson, A. et al. Anal. Chem. 2015, 87 (18), 9194-9203]. 이전에 설명한 바와 같이, 친유성 염료로 염색된 SLB는 균질하며 유동적이어서, 염료 분자의 빠른 확산 (diffusion)이 가능하다 (분산 계수는 0.5 내지 1.0 μm2/s) (도 13a 내지 13d). 특히, ICAM-1-eGFP 및 CD9-mCherry는 모두 SLB로 성공적으로 분획화되었으며 (도 12b), 각각의 밀도는 HEK-CDV와 합성 베시클 사이의 혼합 비율을 조정함으로써 조절이 가능했다 (도 13e). 그러나 도달 가능한 밀도는 위에서 사용된 표면-테더링된 단백질을 활용하는 방법에 비해 상대적으로 낮았다 (SLB의 경우, 최대 0.2 molecules/μm2; 표면 테더링의 경우, 최대 약 40 molecules/μm2).
4-2. SLB에서의 ICAM-1 및 CD9의 공동 국소화(Colocalization) 정도 확인
SLB에서 ICAM-1 및 CD9의 공동 국소화 (colocalization) 정도가 현저하게 높은 것으로 나타났다 (도 12c). 이는 ICAM-1/CD9 복합체가 살아있는 세포 내에서는 조립되어 (assembled) 온전하게 유지되어 있을 것임을 시사한다. ICAM-1 및 CD9를 유사한 밀도로 준비했을 때, CD9 분자의 약 70%가 ICAM-1과 공동 국소화되었고, 30%는 ICAM-1와 공동 국소화되었다 (과량의 ICAM-1으로 인해 더 낮음). 흥미롭게도, 공동 국소화된 스팟은 표면-테더링된 단백질에 복합체에 비해 형광이 더 밝은 경향성을 가졌다 (도 12b). 이는 베시클을 포획하기 위해 SLB 상에서 다수의 단백질 분자가 협력할 수 있는 "핫 스팟 (hot spots)"을 생성하였음을 시사한다. 상기 클러스터링이 특히 CD9를 포함하는 복합체에서 두드러지게 나타났는데 (도 12d), 이는 나노-클러스터 형성에 CD9가 기여한다는 것을 의미한다.
4-3. SLB에서의 VPI 확인
마지막으로, ICAM-1 및 CD9를 포함하는 SLB에서 VPI를 측정했다 (도 12e). 이 때에는 U937-CDV를 분석하여 천연 막과 유사한 막 환경을 제공하는 것이 강력한 상호작용을 더욱 향상시키는지 확인하는데 초점을 맞췄다. 놀랍게도, 표면-테더링 실험과 비교하여 단백질 밀도의 엄청난 차이에도 불구하고, U937-CDV와 막-내장된 ICAM-1/CD9 복합체 사이에 매우 강력한 상호작용이 일어나는 것이 확인됐다 (도 12f). 예를 들어, 표면 테더링된 ICAM-1을 포함하는 글라스에서의 U937-CDV의 VPI는 단위시야 (104 μm2)에서 ~5,000 유닛의 테더링된 ICAM-1 분자로 식별할 수 있었는데, 테더링 실험의 eGFP 형광강도 대비 1/10의 eGFP 형광강도를 가진 막-내장 ICAM-1에서도 U937-CDV의 동등한 수준의 상호작용이 관찰된 바 (도 13f), 베시클-결합력이 ~10배 증가된 것으로 추측된다. 상호작용하는 베시클의 존재는 주로 ICAM-1 및 CD9가 모두 존재하는 스팟에서 확인됐는데, 이는 테더링된 단백질의 실험에서와 마찬가지로, CD9의 존재가 베시클과 ICAM-1 사이의 상호작용을 촉진한다는 것을 시사한다 (도 12g). 상기 결과는 동일한 영역에서 ICAM-1-단독 스팟 (즉, CD9과 공동 국소화 되지 않은 스팟)에 대해, 베시클과 결합한 스팟의 수와 전체 ICAM-1 단독 스팟의 수를 비교하면 더욱 두드러지는데 (도 12h), 이는 ICAM-1이 CD9과의 공동 국소화로 인해 베시클에 유효한 친화력이 한 차원 증가하였음을 시사한다. 종합하면, 상기 결과들은 막단백질의 막횡단 도메인이 지질이중층에 안정적으로 위치하여 기능적 다량체 복합체(functional multimeric complexes)의 형성을 가능하게 할 때 VPI가 현저하게 증가할 수 있음을 보여준다.
이상에서 살펴본 바와 같이, 본 발명자들은 단일 베시클 수준에서 베시클을 관찰 가능하게 하는 신규한 베시클 표지방법을 개발하였으며, 또한 상기와 같이 표지된 베시클을 이용하여 단일-베시클 수준에서 베시클-단백질 상호작용을 시각화할 수 있는 형광-기반 기술을 개발하였다. 대표예로서 세포 유래 베시클 (CDV)을 형광 염료로 표지하여 베시클 및 ICAM-1 사이의 상호작용을 관찰한 결과, 둘 사이의 VPI는 LFA-1에 의해 매개되고, Mg2+ 및 CD9에 의해 촉진되며, 타겟 단백질이 지질이중층에 내장되어 있을 때는 상호작용이 더욱 강화된다는 것을 확인하였다. 본 발명에 따른, 베시클의 표지 및 베시클-단백질 상호작용 이미징 플랫폼은 자연적인 세포외베시클 또는 인공 나노소포와 세포 사이의 상호작용에 관여하는 분자들을 밝히는데 유용히 활용될 것으로 기대된다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야 한다.
본 발명은 베시클-단백질 상호작용 (vesicle-protein interaction, VPI)의 형광 이미징 및 정량화를 위한 신규한 프로토콜에 관한 것이다. 본 발명에 따른 염-교체 방법은 종래 알려진 베시클-표지 방법과 비교하여 적은 양의 염료만으로도 간단하고, 신속하게, 그리고 효과적으로 베시클을 표지할 수 있다. 특히, 상기 염-교체 방법은 샘플 내 잔류 염료를 완전히 제거할 수 있어 단일-베시클 수준에서 보다 정확한 베시클의 관찰이 가능하다. 또한, 다양한 세포-유래 베시클이 상기 방법을 통해 효과적으로 표지되는 것이 확인된 바, 본 발명은 프로테오리포좀 등의 인공 베시클부터 천연 베시클에 이르기까지 다양한 종류의 베시클에 범용적으로 적용될 수 있다. 뿐만 아니라, 이와 같이 표지된 베시클은 TIRF (total internal reflection fluorescence) 현미경을 이용하여 단일 베시클 수준에서 관찰할 수 있으므로, 베시클 및 단백질 상호작용의 보다 정확한 검출 및 정량화가 가능하다. 따라서, 본 발명은 베시클 및 타겟 단백질 간 상호작용의 정확한 분석을 위한 신규한 플랫폼을 제공하며, 베시클의 작용 매커니즘을 분석하기 위한 신규한 수단으로 다양한 분야에서 활용될 것으로 기대된다.
Claims (20)
- (S1) 제1 완충용액에서 베시클 (vesicles) 및 염료를 배양하는 단계;(S2) 제2 완충용액에서 상기 베시클 및 염료를 더 배양하여 자유 염료 (free dye)의 응집체 형성을 유도하는 단계; 및(S3) 상기 단계 (S2)의 배양액을 여과하여 상기 응집체를 제거하는 단계를 포함하고,상기 제2 완충용액의 염 농도는 상기 제1 완충용액의 염 농도의 5 내지 20배인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 베시클은 세포 유래 베시클 (cell-derived vesicles), 프로테오리포좀 (proteoliposomes), 엑소좀 (exosomes), 및 엑토좀 (ectosomes)으로 이루어진 군에서 선택된 하나 이상인 것을 특징으로 하는, 베시클의 표지 방법.
- 제2항에 있어서,상기 세포는 줄기세포, 중간엽 줄기세포, 유도만능줄기세포, 배아줄기세포, 침샘 줄기세포, 암세포, 선포세포, 근상피세포, 적혈구, 면역세포, 단핵구, 수지상 세포, 자연살해 세포, T 세포, B 세포, 대식세포, 내피세포, 표피세포, 신경세포, 신경아교세포, 성상세포, 근육세포, 및 혈소판으로 이루어진 군에서 선택된 하나 이상인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 염은 NaCl, KCl, CaCl2, NaHCO3, NH4Cl, 및 CuCl2로 이루어진 군에서 선택된 하나 이상인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 단계 (S2)의 제2 완충용액은 상기 단계 (S1)에서 배양을 마친 후 제1 완충용액에 염 농축액을 첨가하여 제조되는 것인, 베시클의 표지 방법.
- 제1항에 있어서,상기 제1 완충용액의 염 농도는 전체 완충용액 대비 1 내지 50 mM인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 제2 완충용액의 염 농도는 전체 완충용액 대비 100 내지 300 mM인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 단계 (S3)의 여과는 미세공극을 통해 이루어지고, 상기 미세공극의 직경은 0.2 μm 이하인 것을 특징으로 하는, 베시클의 표지 방법.
- 제8항에 있어서,상기 미세공극은 셀룰로오스 아세테이트 (Cellulose acetate)로 제조된 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 염료는 친유성 염료인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 염료는 형광 염료인 것을 특징으로 하는, 베시클의 표지 방법.
- 제1항에 있어서,상기 방법은 상기 (S3) 단계 후 베시클을 정제하는 단계를 더 포함하는, 베시클의 표지 방법.
- (S1) 형광 단백질-태깅된 타겟 단백질이 테더링된 글라스를 제조하는 단계;(S2) 상기 글라스에 제1항의 방법에 따라 표지된 베시클을 첨가하여 배양하는 단계로서, 상기 베시클은 형광 염료로 표지된 것인, 단계; 및(S3) 상기 타겟 단백질 및 상기 표지된 베시클의 결합을 관찰하는 단계를 포함하는, 베시클-단백질 상호작용 (vesicle-protein interaction)의 스크리닝 방법.

- 제16항에 있어서,상기 단계 (S1)의 지질이중층은 글라스 위에 제조되는 것으로서, 하기 단계에 의해 제조되는 것인, 베시클-단백질 상호작용의 스크리닝 방법:(i) 형광 단백질-태깅된 타겟 단백질을 발현하는 재조합 벡터로 형질전환된 세포를 준비하는 단계;(ii) 상기 형질전환된 세포를 균질화하여 세포 유래 베시클을 수득하는 단계; 및(iii) 상기 세포 유래 베시클 및 리포좀을 혼합하여 융합시킨 후 글라스 위에 로딩하는 단계.
- 제17항에 있어서,상기 리포좀은 페길화된 POPC (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) 리포좀인 것을 특징으로 하는, 베시클-단백질 상호작용의 스크리닝 방법.
- 제16항에 있어서,상기 단계 (S3)의 관찰은 전반사현미경 (TIRF)을 이용하여 수행되는 것인, 베시클-단백질 상호작용의 스크리닝 방법.
