Description
本発明は、化合物の生物における作用メカニズムを決定及び/又は推定する方法、装置及びプログラムに関する。また、本発明は、抗てんかん薬のスクリーニング方法、装置及びプログラムに関する。
神経活動を反映する現象として、カルシウム振動(calcium oscillation)が使用されている(非特許文献1)。同期したカルシウム振動は培養皮質ニューロンで観察され(非特許文献2及び3)、カルシウム振動はニューロン培養における活動電位のバーストによって駆動される(非特許文献4)。てんかんは神経障害の一種であり、カルシウム振動は、抗てんかん薬を含むいくつかの化合物を特徴づけるための表現型スクリーニング法である可能性が示唆されている(非特許文献5)。しかし、これらは定性的な指標であり、解析が困難である。

本発明は、化合物の生物における作用メカニズムを決定及び/又は推定する方法を提供することを目的とする。また、本発明は、抗てんかん薬のスクリーニング方法を提供することを目的とする。
本発明者らは、上記目的を達成すべく鋭意研究を重ねた結果、カルシウム振動で得られる各種データを主成分分析の手法を用いて解析することにより、公知化合物のメカニズムによって、その特徴が異なることを見出し、本発明を完成するに至った。
本発明は、例えば、以下の項に記載の主題を包含する。

化合物の生物における作用メカニズムを決定及び/又は推測する方法であって、
作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で神経細胞の活動を解析し、当該解析結果を比較することにより、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する、方法。

本発明により、化合物の生物における作用メカニズムを決定及び/又は推定することができるので、効率的に化合物のスクリーニングを行うことができる。また、本発明により、抗てんかん薬の候補となる化合物のスクリーニングを効率的に行うことができる。
以下、本発明に包含される各実施形態について、さらに詳細に説明する。
作用メカニズムを決定及び/又は推測する方法
本発明の化合物の生物における作用メカニズムを決定及び/又は推測する方法は、
作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で神経細胞の活動を解析し、当該解析結果を比較することにより、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する(以下、「本発明の決定/推測方法」と称することもある)。
本発明の化合物の生物における作用メカニズムを決定及び/又は推測する方法は、
作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で神経細胞の活動を解析し、当該解析結果を比較することにより、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する(以下、「本発明の決定/推測方法」と称することもある)。
本明細書中で用いる語句及び用語について、以下に詳述する。
本発明で使用することができる神経細胞としては、活動を解析し、作用メカニズムが未知の化合物の作用メカニズムを決定及び/又は推測することができる神経細胞であれば特に限定されず、例えば、生体の脳から抽出した神経細胞、胎児(胎仔)の脳から抽出した神経細胞、脳オルガノイドから抽出した神経細胞、多能性幹細胞から分化誘導して製造した神経細胞、又はこれらをさらに培養した神経細胞、神経細胞の細胞株などを挙げることができる。神経細胞の具体例としては、大脳皮質初代培養神経細胞が挙げられる。多能性幹細胞としては、胚性幹(ES)細胞、核移植により得られるクローン胚由来の胚性幹(ntES)細胞、精子幹細胞(GS細胞)、胚性生殖細胞(EG細胞)、人工多能性幹(iPS)細胞、培養線維芽細胞及び骨髄幹細胞由来の多能性幹細胞(Muse細胞)などが挙げられる。
本発明における神経細胞(例えば、大脳皮質初代培養神経細胞など)は、例えば、ヒト、サル、ラット、マウス、ウサギ、ウマ、ウシ、ヤギ、ヒツジ、イヌ、ネコなどの哺乳動物から得られるものであり、好ましくは、ラット、より好ましくは、ラット胎仔から得られるものである。
大脳皮質神経細胞の初代培養は、公知の方法に準じて行うことができる(例えば、Cell Rep. 2017 Jan 17;18(3):791-803参照)。具体的には、以下の手順で実施する。また、大脳皮質神経細胞の初代培養以外の神経細胞の培養についても、公知の方法に準じて行うことができる。
ラットは、数日間の馴化飼育を行ってから使用することができる。健康状態の良好な妊娠ラットの子宮から胎仔を切り離し、胎仔から全脳を摘出する。摘出した全脳は、氷冷した培地に浸漬する。浸漬した全脳から大脳皮質を分取する。その後、リン酸緩衝食塩水(PBS)で洗浄し、酵素反応により細胞を分散する。使用する酵素反応溶液としては、パパイン/DNase I溶液が挙げられ、反応条件としては、例えば30~40℃、好ましくは36~38℃で、例えば5~60分間、好ましくは20~30分間の反応を行う。次にNeurobasal mediumなどを添加し、ピペットで分散する。これをナイロンメッシュなどで組織片及び細胞凝集塊を除去し、細胞を回収する。回収した細胞は、1000rpmで5分間遠心分離を行いNeurobasal mediumなどで再分散させる。再分散させた細胞の生細胞密度を0.5%トリパンブルーなどを用いて算出し、必要な細胞数を懸濁して96ウェル又は384ウェルの培養プレートに播種する。この際、播種する細胞密度は、およそ1~4×105個/cm2が挙げられる。播種後は、5%二酸化炭素、36~38℃の条件で培養する。そして、週に2~3回程度培地の半量交換を行う。
作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物における化合物としては、特に制限なく使用することができ、例えば、低分子化合物、高分子化合物、生体高分子(ペプチド、タンパク質、核酸、多糖類等)などが挙げられ、このような化合物を含む種々の化合物ライブラリーも使用できる。本発明の決定/推測方法に使用する作用メカニズムが既知の化合物の種類は、例えば、1~238個、1~118個、1~78個、1~58個、1~28個、1~18個、1~13個程度でありうる。
作用メカニズムが既知の化合物としては、神経細胞の活動に作用するメカニズムを有する化合物であれば特に制限されず、例えば、神経細胞の活動に作用するメカニズムを有することが知られている各種公知の化合物(例えば、既に承認されている医薬品、開発段階にある化合物、開発等が中止された化合物、神経細胞の活動に作用するメカニズムを有することが特許、文献等で報告されている化合物、PerkinElmer、Selleck、MedChemExpressなどにより市販されている化合物)などを使用することができる。作用メカニズムが既知の化合物の具体例としては、抗てんかん薬、抗統合失調症薬(統合失調症としては、認知障害を伴う難治性(治療抵抗性、慢性)統合失調症、認知障害を伴わない難治性(治療抵抗性、慢性)統合失調症等を含む)、抗気分障害薬、抗依存症薬(依存症としては、アルコール依存症、薬物依存症等を含む)、抗高次脳機能障害薬などが挙げられる。上記抗てんかん薬などの「抗~薬」とは、上記疾患の予防薬及び/又は治療薬を意味する。
本発明の決定/推測方法は、生物における作用メカニズムを決定及び/又は推測する方法であり、ここでの生物としては、例えば、ヒト、サル、ラット、マウス、ウサギ、ウマ、ウシ、ヤギ、ヒツジ、イヌ、ネコなどの哺乳動物であり、好ましくは、ヒトである。
神経細胞の活動を解析する際には、作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で解析を行う。このような方法としては、例えば、細胞とこれらの化合物を含む溶液(例えば、Tyrode'sバッファー)とを混合する方法が挙げられる。
溶液中の化合物の濃度は、神経細胞の活動を解析できる濃度であれば、特に限定されず、例えば、0.001~5000μMであり得る。当該範囲の上限又は下限は、例えば0.001, 0.003, 0.005, 0.01, 0.03, 0.05, 0.1, 0.3, 0.5, 1, 3, 5, 10, 30, 50, 100, 300, 500, 1000, 3000, 5000μMである。化合物の濃度がこの範囲であると、化合物による変化をデフォルトによる変化とより区別することができるようになり、且つカルシウム振動をより確認することができるようになる。また、同じ化合物について、2以上の複数の種類の濃度を使用して測定してもよい。さらに、化合物の種類毎に異なる濃度を使用してもよい。
神経細胞の活動を解析する方法は、特に限定されず、各種公知の方法を使用することができる。そのような神経細胞の活動は、例えば、カルシウム振動によって捕捉することができる。
さらに、カルシウム振動は、カルシウム指示薬を使用して細胞内のカルシウムを測定することで検出することが可能である。このようなカルシウム指示薬としては、カルシウムイオンとの結合前後に蛍光強度が変化する蛍光カルシウム指示薬などが挙げられ、カルシウム指示薬として各種公知の化合物、例えば、Cal-520などのCalbryte、Fura-2 AM、Fluo-3 AM、Fulo-4 AM、BioTrackerなどの市販のカルシウム指示薬、GCaMPなどのGenetic encoded calcium indicatorを使用することが可能である。カルシウム指示薬を使用することでカルシウム振動により蛍光を発するので、蛍光分析法によりカルシウム振動を可視化することができる。すなわち、特定の励起波長の光を細胞に照射して、特定の発光波長の強度を経時的に測定することによりカルシウム振動を可視化することができる。
蛍光の測定は、プレートリーダーを用いて測定することができる。プレートリーダーとしては、マイクロプレートリーダーが好ましく、96ウェル/384ウェルの同時分注、同時測定が可能なマイクロプレートリーダーがさらに好ましい。例えば、FDSS/μCell(浜松ホトニクス株式会社製)を用いると、96ウェル/384ウェルの同時分注、同時測定が可能である。
蛍光の測定時間は、例えば、1~20分間程度であり得る。当該範囲の上限又は下限は、例えば2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19分であってもよい。
このように神経細胞の活動を解析することで得られた解析結果を比較することによって、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する。生物における作用メカニズムを決定及び/又は推測する方法としては、作用メカニズムを決定及び/又は推測することが可能である限り特に限定されない。本発明の決定/推測方法の具体的な実施態様としては、以下の少なくとも1つの工程を含むものが挙げられる。
(a)作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で神経細胞のカルシウム振動に伴う蛍光を測定する工程、
(b)工程(a)で測定した蛍光波形を解析する工程、
(c)工程(b)の解析結果を主成分分析する工程、
(d)工程(c)の主成分分析の結果に基づき、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する工程。

工程(a)は上で既に説明されているものである。工程(b)では、カルシウム振動を蛍光分析法で経時的に記録することで得られた波形を解析する。これらの波形の解析は目視で行ってもよいし、コンピュータを用いてもよい。コンピュータを使用する場合は、SpotfireのWave Finder(Data Visualization&Analytics Software-TIBCO Spotfire、http://spotfire.tibco.com/)を使用することにより波形の特徴を抽出することができるし、R又はPythonでも実施可能である。また、例えば、AI(人工知能)等を活用したアプリケーションを用いることにより、多面的に解析することも可能である。
解析項目としては、例えば、波形ピーク数(総ピーク数、単位時間当たりのピーク数)、ピーク間時間(平均、標準偏差、最大、最小)、ピーク高(平均、標準偏差、変動係数)、ピーク振幅(平均、標準偏差)、ピーク輝度値/ボトム輝度値レシオ(平均、標準偏差)、ボトム輝度値(平均、標準偏差)、立ち上がりスロープ(0~100%、10~90%、20~80%、30~70%)(平均、標準偏差)、立ち下がりスロープ(0~100%、10~90%、20~80%、30~70%)(平均、標準偏差)、ピーク幅(10%、20%、30%、40%、50%、60%、70%、80%、90%、100%)(平均、標準偏差)、ピーク間幅(平均、標準偏差)、ピーク総面積(平均、標準偏差)、ピーク高/ピーク幅(平均、標準偏差)、ピークの頂点からピークが50%の高さになるまでのピーク幅(平均、標準偏差)を挙げることができる。
工程(c)では工程(b)の蛍光波形を解析結果に基づき主成分分析を行う。主成分分析に用いるデータは解析項目から2種類以上、例えば、2種類、3種類、4種類、5種類、6種類、7種類、8種類、9種類を選択する。選択したデータは、R、JMP、SPSS、SAS、MATLAB(商標)などのソフトウェアを使用して主成分分析を行うことができる。散布図は、例えば、第一主成分、第二主成分を用いて作成することができる。
主成分分析を行う際に使用する化合物の濃度は、1濃度のみでもよいし、2種類以上の濃度を使用することもできる。化合物の濃度は、例えば、1~10種類程度であり得る。当該範囲の上限又は下限は、例えば2、3、4、5、6、7、8、9種類であってもよい。2種類以上の濃度を使用する場合は、2種類以上の異なる濃度を用いて、各濃度で主成分分析を行うことにより、濃度による変化具合から、単一の濃度では区別ができなかった化合物を区別することも可能となり得る。
工程(d)では、主成分分析の結果に基づいて、作用メカニズムが既知の化合物の主成分分析の分布と比較し、分布が合致した既知の化合物の作用メカニズムを、作用メカニズムが未知の化合物のメカニズムと決定及び/又は推測することができる。作用メカニズムが未知の化合物の分布が、作用メカニズムが既知の化合物の分布のいずれとも合致しないときは、新たな作用メカニズムを有する可能性が推察されるため、新たな作用メカニズムを有する化合物であると決定及び/又は推測することができる。
なお、本発明の決定/推測方法は、上記工程(a)~(c)を含む実施形態、上記工程(a)~(d)を含む実施形態、上記工程(b)~(d)を含む実施形態、も包含する。

本発明の決定/推測方法は、神経細胞の活動に作用するメカニズムであれば、メカニズムを決定及び/又は推測することができる。神経細胞の活動に作用するメカニズムを有する疾患としては、例えば、てんかん、統合失調症、気分障害、依存症、高次脳機能障害などを挙げることができ、これらの治療薬、予防薬、診断薬のメカニズムを決定及び/又は推測することができる。
スクリーニング方法
本発明の抗てんかん薬、抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬(好ましくは、抗てんかん薬)のスクリーニング方法は、
抗てんかん作用、抗統合失調症作用、抗気分障害作用、抗依存症作用又は抗高次脳機能障害作用(好ましくは、抗てんかん作用)を有することが既知の化合物及び被検物質の存在下で神経細胞の活動を解析し、当該解析結果を比較することにより、被検物質が前記化合物と同様の作用メカニズムを有している場合に、被検物質を抗てんかん薬、抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬(好ましくは、抗てんかん薬)の候補物質として選択する(以下、「本発明のスクリーニング方法」と称することもある)。
本発明の抗てんかん薬、抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬(好ましくは、抗てんかん薬)のスクリーニング方法は、
抗てんかん作用、抗統合失調症作用、抗気分障害作用、抗依存症作用又は抗高次脳機能障害作用(好ましくは、抗てんかん作用)を有することが既知の化合物及び被検物質の存在下で神経細胞の活動を解析し、当該解析結果を比較することにより、被検物質が前記化合物と同様の作用メカニズムを有している場合に、被検物質を抗てんかん薬、抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬(好ましくは、抗てんかん薬)の候補物質として選択する(以下、「本発明のスクリーニング方法」と称することもある)。
本発明の抗てんかん薬、抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬(好ましくは、抗てんかん薬)のスクリーニング方法は、前述する本発明の決定/推測方法の実施形態の1つであり、「抗てんかん作用、抗統合失調症作用、抗気分障害作用、抗依存症作用又は抗高次脳機能障害作用(好ましくは、抗てんかん作用)を有することが既知の化合物」が「作用メカニズムが既知の化合物」に、「被検物質」が「作用メカニズムが未知の化合物」に対応する。本発明の抗てんかん薬、抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬(好ましくは、抗てんかん薬)のスクリーニング方法は、特に明示しない限り、本発明の決定/推測方法と基本的には同様に実施することができる。
被検物質としては、特に制限なく使用することができ、例えば、低分子化合物、高分子化合物、生体高分子(ペプチド、タンパク質、核酸、多糖類等)などが挙げられ、このような化合物を含む種々の化合物ライブラリーも使用できる。
本発明のスクリーニング方法の具体的な実施態様としては、以下の少なくとも1つの工程を含むものが挙げられる。
(A)抗てんかん作用、抗統合失調症作用、抗気分障害作用、抗依存症作用又は抗高次脳機能障害作用(好ましくは、抗てんかん作用)を有することが既知の化合物及び被検物質の存在下で神経細胞のカルシウム振動に伴う蛍光を測定する工程、
(B)工程(A)で測定した蛍光波形を解析する工程、

決定/推測及びスクリーニングの処理手順
続いて、本発明の決定/推測方法及びスクリーニング方法の処理手順の一例について説明する。図1は、本発明の決定/推測方法及びスクリーニング方法の処理手順を示すフローチャートであり、図2は、該方法の工程S1~S6を実行するための装置1及び蛍光測定器2のブロック図である。
続いて、本発明の決定/推測方法及びスクリーニング方法の処理手順の一例について説明する。図1は、本発明の決定/推測方法及びスクリーニング方法の処理手順を示すフローチャートであり、図2は、該方法の工程S1~S6を実行するための装置1及び蛍光測定器2のブロック図である。
装置1は、汎用のコンピュータで構成することができ、ハードウェア構成として、CPU、GPUなどのプロセッサ、DRAM、SRAMなどの主記憶装置、及びHDD、SSDなどの補助記憶装置を備えている。また、装置1は、複数のコンピュータで構成してもよい。
装置1は、機能ブロックとして、解析部11と、主成分分析部12と、決定/推測部13と、候補物質選択部14と、を含む。これらの各部は、装置1のプロセッサが、本発明のプログラムを主記憶装置に読み出して実行することにより、ソフトウェア的に実現することができる。当該プログラムは、インターネット等の通信ネットワークを介して装置1にダウンロードしてもよいし、当該プログラムを記録したCD-R等のコンピュータ読み取り可能な非一時的な記録媒体を介して装置1にインストールしてもよい。
図1に示すフローチャートでは、工程S1において、蛍光測定器2が、作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物(被検物質)の存在下で神経細胞のカルシウム振動に伴う蛍光を測定する。測定された蛍光波形は、装置1に送信される。本実施形態では、前記作用メカニズムが抗てんかん作用であり、前記作用メカニズムが既知の化合物が抗てんかん薬である。他の実施形態としては、前記作用メカニズムが抗統合失調症作用、抗気分障害作用、抗依存症作用又は抗高次脳機能障害作用であり、前記化合物が抗統合失調症薬、抗気分障害薬、抗依存症薬又は抗高次脳機能障害薬であることが挙げられる。特許請求の範囲に記載の工程(a)及び工程(A)は、工程S1に対応する。
工程S2において、装置1の解析部11が、工程S1で測定した蛍光波形を解析する。特許請求の範囲に記載の工程(b)及び工程(B)は、工程S2に対応する。
工程S3において、装置1の主成分分析部12が、工程S3の解析結果を主成分分析する。特許請求の範囲に記載の工程(c)及び工程(C)は、工程S3に対応する。
工程S4において、装置1の決定/推測部13が、工程S3の主成分分析の結果に基づき、作用メカニズムが未知の化合物(被検物質)の生物における作用メカニズムを決定及び/又は推測する。特許請求の範囲に記載の工程(d)は、工程S4に対応する。
その結果、被検物質が既知の化合物と同様の作用メカニズム(抗てんかん作用)を有している場合(工程S5においてYES)、工程S6に移行し、装置1の候補物質選択部14が、被検物質を抗てんかん薬の候補物質として選択する。特許請求の範囲に記載の工程(D)は、工程S4~S6に対応する。
本明細書において引用されるすべての特許文献及び非特許文献の開示は、全体として本明細書に参照により組み込まれる。
なお、本明細書において「含む」とは、「本質的にからなる」と、「からなる」をも包含する(The term "comprising" includes "consisting essentially of” and "consisting of.")。また、本発明は、本明細書に説明した構成要件を任意の組み合わせを全て包含する。
また、上述した本発明の各実施形態について説明した各種特性(性質、構造、機能等)は、本発明に包含される主題を特定するにあたり、どのように組み合わせられてもよい。すなわち、本発明には、本明細書に記載される組み合わせ可能な各特性のあらゆる組み合わせからなる主題が全て包含される。
本発明は、更に以下の実施例によって詳しく説明されるが、これらは本発明を限定するものではなく、また本発明の範囲を逸脱しない範囲で変化させてもよい。
神経細胞の調製
妊娠18日目のラット(Crl:CD(SD))の胚をPapain Dissociation System(Worthington Biochemical Corp)を使用して解離し皮質ニューロンを調製した。
妊娠18日目のラット(Crl:CD(SD))の胚をPapain Dissociation System(Worthington Biochemical Corp)を使用して解離し皮質ニューロンを調製した。
具体的には、大脳皮質を氷冷HBSSに収集し、インキュベーター内で0.25%パパイン/Dnase I含有PBS中で30分間解離させた。Neurobasal培地を10 mL加え、10回ピペッティングした。上清を回収し、セルストレーナー(#352350、株式会社ファルコンバイオシステムズ)を使用してろ過し、遠心分離(1,000 rpm、5分)した。ペレットをOvomucoidプロテアーゼ阻害剤/HBSSに懸濁し、遠心分離(800 rpm、5分)し、B27サプリメント(#17504044、Gibco)、GlutaMAX(#35050061、Gibco)、ペニシリン-ストレプトマイシン溶液(#P4333、Sigma-Aldrich)を含むNeurobasal培地(#21103-049、Gibco)で再懸濁した。
解離した細胞を60ウェル(96ウェルプレート(#3842、Corning)の周辺ウェルを除く)に75,000細胞/ウェルの密度で播種した。細胞培養は37℃の5% CO2インキュベーター内で維持し、培地の半分は週に2回交換した。すべての実験において、培養物はDIV14を使用した。
カルシウムイメージングの準備
Cal-520(#21130、AAT-Bioquest)のバイアルを、0.01% Pluronic(商標) F-127(#P3000MP、Invitrogen)を含むTyrode'sバッファー(0.1 mM Mg2+)に2μMになるように再懸濁した。培養物をタイロードバッファーで3回洗浄し、1ウェルあたり80μlのCal-520で37℃ 1時間インキュベートした。培養プレートをアッセイ場所に設置し、測定前に10分以上安定化させた。

FDSS/μCell(浜松ホトニクス株式会社)を使用して、カルシウムシグナルを蛍光強度データとして記録した。インキュベーション後、FDSS/μCellの96ディスペンサーヘッドを使用して表1に示した試験化合物をタイロードバッファーに溶解させた溶液及び溶媒対照を20μL分注した。分注から5分以上後にカルシウムシグナルを以下の測定条件で5分間測定した。

蛍光強度データの数値化
蛍光強度データは、FDSS/μCellソフトウェアから抽出した。データ分析と可視化は、SpotfireのWave Finder(Data Visualization&Analytics Software-TIBCO Spotfire、http://spotfire.tibco.com/)を用いた。蛍光強度データから平均検出ピーク高、検出ピーク高の変動係数、平均ピーク高/ピーク幅、50%ピーク幅、曲線下総面積、総検出ピーク数の6項目を数値化した。
蛍光強度データは、FDSS/μCellソフトウェアから抽出した。データ分析と可視化は、SpotfireのWave Finder(Data Visualization&Analytics Software-TIBCO Spotfire、http://spotfire.tibco.com/)を用いた。蛍光強度データから平均検出ピーク高、検出ピーク高の変動係数、平均ピーク高/ピーク幅、50%ピーク幅、曲線下総面積、総検出ピーク数の6項目を数値化した。
主成分分析(PCA)
PCA分析用にRによって作成されたカスタムコードを使用して処理した。PCA分析の特徴は、特徴の相関と各波形の目視検査に基づいて選択した。各ウェルの統計分析を実行し、GraphPad Prism 7を使用して視覚化した。

結果
96ウェルプレートでラット初代皮質神経細胞を培養し、インビトロで14日目にTyrode'sバッファーに溶解した蛍光カルシウムインジケーターを使用してカルシウム振動を観察した。結果を図3に示す。てんかんにおける神経細胞の活動を模倣するために、低マグネシウム濃度(0.1 mM)のTyrode'sバッファーを使用した。FDSS/μCellを使用し同時に60ウェルの同期したカルシウム振動を観察することに成功した(図3)。各ウェルにおいて5分間で40以上のピークが観察された。それ故、次の薬理学的実験においてもこれらの条件でカルシウム振動を測定することに決めた。
96ウェルプレートでラット初代皮質神経細胞を培養し、インビトロで14日目にTyrode'sバッファーに溶解した蛍光カルシウムインジケーターを使用してカルシウム振動を観察した。結果を図3に示す。てんかんにおける神経細胞の活動を模倣するために、低マグネシウム濃度(0.1 mM)のTyrode'sバッファーを使用した。FDSS/μCellを使用し同時に60ウェルの同期したカルシウム振動を観察することに成功した(図3)。各ウェルにおいて5分間で40以上のピークが観察された。それ故、次の薬理学的実験においてもこれらの条件でカルシウム振動を測定することに決めた。
カルシウム振動の形状を評価するために、Wave Finder softwareを使用して波形の6つの特徴を抽出した(図4参照)。当該特徴は、各ウェルについて、平均検出ピーク高(Mean Peak Height)、検出ピーク高の変動係数(CV of Peak Height)、平均ピーク高/ピーク幅(Mean Peak Height /Peak Width)、平均50%ピーク幅(Mean peak width 50%:ピークトップの時間からシグナルがピークトップの半分まで落ちる時間までのピーク幅)、平均曲線下総面積(AUC)、総検出ピーク数(Peak Number)。試験化合物としては、GABA作動性阻害活性のポジティブアロステリックモジュレーター(PAM)であるジアゼパム(diazepam)(10μM)及びクロナゼパム(clonazepam)、電位依存性ナトリウムチャネルを介した興奮性入力の阻害剤であるカルバマゼピン(carbamazepine)(30μM)、ラモトリギン(lamotrigine)(10μM)及びラコサミド(lacosamide)(100μM)、抗てんかん薬として広く使用されているレベチラセタム(levetiracetam)(100μM)、ブリバラセタム(brivaracetam)(100μM)、ペランパネル(perampanel)(100μM)を使用した(表1)。各化合物は、FDSS/μCellのオートディスペンサーによってプレートの6ウェルに分注、5分間インキュベートし、5分間カルシウム振動のシグナルを記録した。結果を図5に示す。DMSO(0.1%)が分注されたウェルと比べて、試験化合物が分注されたウェルのカルシウム振動の特徴について変化しているものがあった。
次に、主成分分析(PCA)を使用して化合物によるカルシウム振動の特徴を比較した。結果を図6に示す。2つの成分(PC1、PC2)の固有値は1以上であった。2つの成分は、変動性の83.8%を説明した。
次に、PCAプロットにおいてDMSOのプロットから離れた位置にある化合物の濃度依存性を確認した。レベチラセタムとブリバラセタムはカルシウム振動の特徴にほとんど影響を与えなかったので、この実験から除外した。6つの化合物を3種類の濃度で、1条件当たり3つのウェルに分注した。前述するように全ての検出されたピークから特徴を抽出した。結果を図7に示す。また、主成分分析の結果を図8に示す。濃度が増加するほど、プロットはDMSOのプロットから分離されており、化合物は様々な軌道に沿って移動した。同じメカニズムであるジアゼパムとクロナゼパムは濃度が増加しても同じ方向に移動したのに対して、異なるメカニズムであるカルバマゼピンとペランパネルは単一の濃度とは異なり軌道によって分離していた。
本発明によれば、作用メカニズムが未知の化合物の作用メカニズムを容易に決定又は推測できるため、作用メカニズムが未知の化合物の作用メカニズムの決定又は推測が迅速に行うことができる。

Claims (17)
- 化合物の生物における作用メカニズムを決定及び/又は推測する方法であって、
作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で神経細胞の活動を解析し、当該解析結果を比較することにより、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する、方法。 - 前記神経細胞の活動の解析が、カルシウム振動法に基づいて実施される、請求項1に記載の方法。
- (a)作用メカニズムが既知の化合物及び作用メカニズムが未知の化合物の存在下で神経細胞のカルシウム振動に伴う蛍光を測定する工程、
(b)工程(a)で測定した蛍光波形を解析する工程、及び
(c)工程(b)の解析結果を主成分分析する工程
を含む、請求項2に記載の方法。 - (d)工程(c)の主成分分析の結果に基づき、作用メカニズムが未知の化合物の生物における作用メカニズムを決定及び/又は推測する工程
