Abstract
結晶スポンジ法に利用可能な新規な細孔性三次元ネットワーク構造体およびその製造方法、ならびに、細孔性三次元ネットワーク構造体を用いた構造解析対象化合物の新規な構造解析方法の提供を目的とする。本発明によれば、金属塩およびα-ヒドロキシカルボン酸を含む溶液と、二座配位子(前記二座配位子中の配位部位間の距離は8.0Å以上である)を含む溶液とを混合し、静置することにより形成される、新規な細孔性三次元ネットワーク構造体およびその製造方法が提供される。本発明によればまた、細孔性三次元ネットワーク構造体からなる新規な結晶構造解析用試料およびその製造方法ならびに細孔性三次元ネットワーク構造体を用いた新規な構造解析方法が提供される。
Description
本願は、先行する日本国出願である特願2021-164174(出願日:2021年10月5日)の優先権の利益を享受するものであり、その開示内容全体は引用することにより本明細書の一部とされる。
本発明は、細孔性三次元ネットワーク構造体およびその製造方法に関する。本発明はまた、結晶構造解析用試料およびその製造方法ならびに構造解析対象化合物の構造解析方法に関する。
創薬、食品、化粧品、材料科学等の分野において、化合物の立体構造は薬効や毒性、味や香り、材料の機能発現等の多方面に大きな影響を及ぼす。このため、化合物の立体構造を正確に決定する必要があるが、一般的に、立体構造の解析は難易度が高い。
物質の分子構造を正確に決定する手法として単結晶X線構造解析法が知られているが、解析対象物質の結晶化が困難な場合等にはその利用が制限されていた。近年、X線結晶解析により構造決定する革新的な手法として「結晶スポンジ法」が注目されている。結晶スポンジ法は、解析対象物質を含む溶液に結晶スポンジと呼ばれる多孔性単結晶を浸漬させ、三次元的に規則正しく整列した空隙を有する細孔内に解析対象物質を安定的に配置することでX線結晶解析により構造決定することができる解析手法である(特許文献1)。この手法は結晶化のプロセスを経ることなく解析対象物質の構造を決定できる点で極めて有用性が高い。
しかしながら、解析対象物質の特性や分子量の大きさによっては結晶スポンジの細孔内に取り込ませることが困難なものが存在し、結晶スポンジ法で解析可能な物質が限定されているという問題があった。
本発明は、結晶スポンジ法に利用可能な新規な細孔性三次元ネットワーク構造体およびその製造方法を提供することを目的とする。本発明はまた、細孔性三次元ネットワーク構造体を用いた新規な構造解析方法を提供することを目的とする。
本発明者らは、金属塩およびα-ヒドロキシカルボン酸を含む溶液と、二座配位子(前記二座配位子中の配位部位間の距離は8.0Å以上である)を含む溶液とを混合し、静置することにより、新規な細孔性三次元ネットワーク構造体が形成されること、該細孔性三次元ネットワーク構造体に解析対象化合物を取り込むことで、該化合物の立体構造を決定できることを見出した。本発明はこれらの知見に基づくものである。

本発明の細孔性三次元ネットワーク構造体によれば、これまで結晶スポンジ法による構造解析が困難であった分子量500以上の化合物の立体構造解析を行うことができる。分子量が500以上のいわゆる中分子化合物は新しい創薬モダリティとして注目されており、本発明によればこのような中分子化合物の立体構造解析を容易かつ迅速に行うことができる点で有利である。

本発明において、基の全部または一部としての「脂肪族炭化水素基」は、直鎖または分岐鎖の炭化水素鎖を意味する。炭素数1~6の脂肪族炭化水素基としては、例えば、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基が挙げられる。本発明において「脂肪族炭化水素基」は、1個または複数個の二重結合を有していてもよい。1個または複数個の二重結合を有する炭素数1~4の脂肪族炭化水素基としては、エテニル基、プロペニル基、ブテニル基が挙げられる。本発明において「脂肪族炭化水素基」は、1個または複数個の三重結合を有していてもよい。1個または複数個の三重結合を有する炭素数1~4の脂肪族炭化水素基としては、エチニル基、プロピニル基、ブチニル基が挙げられる。

本発明において、基の全部または一部としての「脂環式炭化水素基」は、炭素原子が環状に結合した構造を有する炭化水素鎖であり、後記の「芳香族炭化水素基」に属さないものを意味する。炭素数3~6の脂環式炭化水素基としては、例えば、シクロプロピル基、シクロブチル基、シクロペンチル基、シクロヘキシル基が挙げられる。本発明において、基の全部または一部としての「複素環基」は、酸素原子、窒素原子および硫黄原子からなる群から選択される1種または2種以上の異種原子を環員原子として含む、5~18員の複素環であり、後記の「芳香族複素環基」に属さないものを意味する。複素環基は、単環式の複素環であっても、2または3環式以上(好ましくは、2、3または4環式)の複素環であってもよい。異種原子の個数は、1個または複数個とすることができ、例えば、1~4個、1~3個、1~2個または1個である。本発明において、「脂環式炭化水素基」および「複素環基」は置換基を有していてもよく、例えば、オキソ基(=O)が挙げられる。

本発明において、「ハロゲン原子」としては、塩素(Cl)、臭素(Br)、フッ素(F)、ヨウ素(I)が挙げられる。
本発明において、「置換されていてもよい」あるいは「置換基を有してもよい」とは、置換される基または原子上の1または複数個の水素原子が別の基に置換されることを意味する。
本発明において、「配位子」は、配位部位を2つ以上有する配位子を意味し、このうち配位部位を2つ有する配位子を二座配位子という。ここで、「配位部位」とは、配位結合が可能な非共有電子対(孤立電子対)を有する、配位子中の原子または基(原子団)をいう。
本発明において、リガンドとは配位部位を有する配位子であり、リガンド1(L1)およびリガンド2(L2)は配位子1および配位子2とそれぞれいうことがある。
<<細孔性三次元ネットワーク構造体>>
本発明の細孔性三次元ネットワーク構造体は、前記式(I)の組成式を満たす金属錯体構造を基本構造とするものである。すなわち、本発明の構造体においては、前記式(I)で示される金属錯体構造が規則的に組み合わさることにより細孔性の三次元ネットワーク構造が形成されている。本発明の構造体は金属イオンおよび有機化合物の錯体を基本構造とするため、細孔性金属有機構造体と同義である。
本発明の細孔性三次元ネットワーク構造体は、前記式(I)の組成式を満たす金属錯体構造を基本構造とするものである。すなわち、本発明の構造体においては、前記式(I)で示される金属錯体構造が規則的に組み合わさることにより細孔性の三次元ネットワーク構造が形成されている。本発明の構造体は金属イオンおよび有機化合物の錯体を基本構造とするため、細孔性金属有機構造体と同義である。

本発明の構造体において、L2は二座配位子であり、金属イオンMと配位結合を形成する配位部位を2つ有する。金属イオンMと配位結合を形成する配位部位としては、例えば、窒素原子、酸素原子、硫黄原子またはリン原子等の異種原子;ニトロ基、アミノ基、シアノ基、カルボキシ基または水酸基等の基が挙げられ、好ましくは窒素原子または窒素原子を含む基である。二座配位子(L2)は、好ましくは、二座配位子中の2つの配位部位が、二座配位子の中心位置に対して直線状またはほぼ直線状に存在するものを選択することができる。ここで、本発明において、「中心位置」とは二座配位子を構成する分子の化学構造における中心位置をいい、例えば、重心を中心位置とすることができる。本発明において、「直線状」とは二座配位子の2つの配位部位のそれぞれの中心位置を直線で結んだ場合に該直線が該二座配位子の中心位置を通過する位置関係にあることをいい、「ほぼ直線状」とは二座配位子の中心位置と2つの配位部位のそれぞれの中心位置を直線で結んだ場合に該2本の直線が形成する角度が、120°以上180°未満(好ましくは150°以上180°未満であり、より好ましくは170°以上180°未満であり、特に好ましくは175°以上180°未満である)であることをいう。
本発明の構造体において、二座配位子(L2)中の配位部位間の距離は8.0Å以上である。ここで、配位部位間の距離は、二座配位子の各配位部位間の直線距離を意味する。前記配位部位間の距離の下限値(以上または超える)は、分子量500以上の中分子化合物が細孔内に進入できる程度の細孔を形成する観点から、8.0Å、8.5Å、9.0Å、9.5Å、10.0Å、10.5Å、11.0Åまたは11.5Åとすることができる。一方、前記配位部位間の距離の上限値(以下または未満)は、細孔性三次元ネットワーク構造体の安定性の観点から、50.0Å、40.0Åまたは30.0Åとすることができる。これらの前記配位部位間の距離の上限値および下限値はそれぞれ任意に組み合わせることができる。本発明の構造体における前記配位部位間の距離の範囲は、例えば、8.0~50.0Åまたは9.0~40.0Åとすることができ、分子量500以上の中分子化合物が細孔内に進入できる程度の細孔を形成し、かつ、細孔性三次元ネットワーク構造体の安定性の観点から、好ましくは11.0~30.0Åである。なお、分子量は化学構造式から算出される理論値をいう。

式(IV)においてA1およびA2は、例えば、それぞれ独立して1価の脂肪族炭化水素基、1価の脂環式炭化水素基、1価の芳香族炭化水素基、1価の芳香族複素環基から選択することができるが、特に限定されることなく、さらに置換基を有してもよい。A1およびA2は金属イオンMと配位結合を形成できる原子または基(例えば、窒素原子、酸素原子、硫黄原子およびリン原子等の異種原子;ニトロ基、アミノ基、シアノ基、カルボキシ基および水酸基等の基)を配位部位に有する。配位部位としては、好ましくは窒素原子または窒素原子を含む基である。前記置換基としては、配位結合を阻害する等の影響を与えない限り限定されるものではないが、例えば、ハロゲン基、ヒドロキシ基、アミノ基、アルキル基、フルオロアルキル基、スルホ基、メルカプト基、アルコキシ基、ニトロ基、アシル基およびニトリロ基等が挙げられる。
式(IV)において好ましくは、A1およびA2はそれぞれ独立して非置換の芳香族複素環基(特に1個の窒素を含有する6員芳香族複素環基)を表し、より好ましくはA1およびA2は同じ非置換の芳香族複素環基(特に1個の窒素を含有する6員芳香族複素環基)を表す。
式(IV)において好ましくは、A1およびA2はそれぞれ独立して、窒素原子、酸素原子、硫黄原子またはリン原子等の異種原子;ニトロ基、アミノ基、シアノ基、カルボキシ基または水酸基等の基により置換された芳香族炭化水素基(特に1個のニトロ基、アミノ基、シアノ基またはカルボキシ基により置換された6員芳香族炭化水素基)を表し、より好ましくはA1およびA2はニトロ基、アミノ基、シアノ基、カルボキシ基または水酸基により置換された、同じ芳香族炭化水素基(特に1個のニトロ基、アミノ基、シアノ基、カルボキシ基または水酸基により置換された6員芳香族炭化水素基)を表す。
式(IV)においてA1またはA2が6員の環式基を表す場合、配位結合を形成できる原子または基の位置は、特に限定されるものではないが、A1またはA2とXとの結合に対してパラ位であることが好ましい。
式(IV)においてXが表すリンカーは、二座配位子中の配位部位間の距離が8.0Å以上となるものであれば限定されないが、例えば、(i)-Z1-(式中、Z1は、イミノ基(>C=N-または-N=C<)、アミド基(-C(=O)-NH-または-NH-C(=O)-)、ジスルフィド基(-S-S-)、2価の炭素数2~4の脂肪族炭化水素基(1または2個の芳香族炭化水素基または芳香族複素環基により置換されていてもよく、また、二重結合または三重結合を少なくとも1つ有していてもよい。以下同様)、2価の脂環式炭化水素基、2価の複素環基、2価の芳香族炭化水素基または2価の芳香族複素環基を表す。);

上記式(iii)においてZ4、Z5およびZ6は好ましくはそれぞれ独立して、イミノ基、アミド基、ジスルフィド基、2価の炭素数2または3の炭化水素基(1または2個の芳香族炭化水素基により置換されていてもよく、また、二重結合または三重結合を1つ有していてもよい)、2価の複素環基、2価の単環式芳香族炭化水素基または2、3もしくは4環式の芳香族炭化水素基(より好ましくはフェニレン基またはナフチレン基)、あるいは2価の単環式芳香族複素環基または2、3もしくは4環式の芳香族複素環基を表し、より好ましくは、Z4およびZ6がそれぞれ独立してイミノ基、アミド基、ジスルフィド基、または2価の炭素数2または3の炭化水素基(1または2個の芳香族炭化水素基により置換されていてもよく、また、二重結合または三重結合を1つ有していてもよい)を表し、Z5が2価の単環式または2環式芳香族炭化水素基(特にフェニレン基、ナフチレン基)あるいは2価の単環式または2環式芳香族複素環基を表し、より好ましくはZ4およびZ6が同じイミノ基、アミド基、ジスルフィド基、または2価の炭素数2または3の炭化水素基(1または2個の芳香族炭化水素基により置換されていてもよく、また、二重結合または三重結合を1つ有していてもよい)を表し、Z5が2価の単環式または2環式芳香族炭化水素基(特にフェニレン基、ナフチレン基)あるいは2価の単環式または2環式芳香族複素環基を表す。前記複素環基はオキソ基(=O)により置換されていてもよい。
式(i)、(ii)および(iii)においてZ1、Z2、Z3、Z4、Z5およびZ6が2価の6員環式基を表す場合、これらの基がそれぞれ有する2つの結合のうち一方の結合に対する他方の結合の位置はパラ位であることが好ましい。例えば、式(i)、(ii)および(iii)においてZ1、Z2、Z3、Z4、Z5およびZ6がフェニレン基を表す場合、1,4-フェニレン基が好ましい。また式(i)、(ii)および(iii)においてZ1、Z2、Z3、Z4、Z5およびZ6がナフチレン基を表す場合、それぞれ独立に1,4-ナフチレン基、1,5-ナフチレン基および2,6-ナフチレン基からなる群から選択されるナフチレン基が好ましい。

本発明の構造体は、結晶スポンジとして結晶構造解析に用いることができる。すなわち本発明の構造体は結晶構造解析用試料として提供することができる。ここで、結晶スポンジとは、規則正しい細孔構造を有する単結晶であり、例えば、金属有機構造体(MOF)、共有結合性有機構造体(COF)、細孔性有機分子結晶(POMC)、ゼオライトのような無機化合物等から得ることができものであり、細孔内に化合物をゲストとして取り込む性質を有する単結晶をいう。
本発明の構造体は、金属塩およびα-ヒドロキシカルボン酸を含む溶液(溶液3)と、二座配位子(前記二座配位子中の配位部位間の距離は8.0Å以上である)を含む溶液(溶液1)とを接触させ、静置することにより調製することができる。ここで、金属塩は金属イオンとカウンターアニオンで組成されるものであってもよく、このような組成の金属塩でない場合は全体の電荷が0となるようにカウンターアニオンにより調整することができる。
本発明の構造体において、溶液1および溶液3ならびに後記の溶液2を調製する際に使用する溶媒は、構造体の作製に支障を与えるものでない限り、特に限定されるものではないが、例えば、脂肪族炭化水素類(例えば、n-ヘキサン、ペンタン、ヘプタン、オクタン、ノナン、デカン、シクロペンタン、シクロヘキサン等)、芳香族炭化水素類(例えば、ベンゼン、トルエン、キシレン等)、ハロゲン化炭化水素類(例えば、クロロホルム、ジクロロメタン、クロロメタン、ジクロロエタン、クロロエタン等)、エーテル類(例えば、ジエチルエーテル、ジメチルエーテル、メチルtert-ブチルエーテル、テトラヒドロフラン等)、エステル類(例えば、酢酸メチル、酢酸エチル、ギ酸メチル、ギ酸エチル等)、ケトン類(例えば、アセトン、メチルエチルケトン、ペンタノン等)、アルコール類(例えば、メタノール、エタノール、プロパノール、ブタノール、ペンタノール、ヘキサノール等)、ニトリル類(例えば、アセトニトリル等)、アミド類(例えば、ジメチルホルムアミド、ジメチルアセトアミド等)、ジメチルスルホキシドまたは水および上記溶媒を任意の割合で混合したもの等が挙げられる。また、本発明の構造体の各原料化合物の性質や原料化合物の相性を考慮して、使用する溶媒を選択することが望ましい。
本発明の構造体の製造では、溶液1と溶液3との接触は、各成分が時間をかけて混じり合うように行うことが好ましい。例えば、溶液1を容器に入れ、有機溶媒(例えば溶液1と溶液3を1:1で混ぜた溶液)からなる溶液2を溶液1の上に重層し、さらに溶液3を溶液2の上に重層して3層溶液を調製し、0~80℃、1~14日間静置することで溶液1の成分が溶液3に移行して溶液3の成分と混じり合い、本発明の細孔性三次元ネットワーク構造体を結晶として析出させることができる。
<<結晶構造解析用試料およびその製造方法>>
本発明の結晶構造解析用試料は、本発明の構造体および構造解析対象化合物からなるものである。

本発明の解析用試料は、本発明の構造体に解析対象化合物を取り込ませて作製することができる。本発明の構造体に解析対象化合物を取り込ませる方法は、解析対象化合物がゲストとして本発明の構造体の細孔内に取り込まれる限り、特に限定されるものではない。例えば、IUCrJ 2016, 3, 139-151、Chem. Eur. J. 2017, 23, 15035-15040、CrystEngComm, 2017, 19, 4528-4534、Org. Lett. 2018, 20, 3536-3540、Science 2016, 353, 808-811、Chem. Commun. 2015, 51, 11252-11255等に記載の方法によって解析対象化合物をゲストとして本発明の構造体の細孔内に取り込ませることができる。
本発明の解析用試料において、解析対象化合物は、本発明の構造体の細孔内に取り込まれる限り、特に限定されるものではないが、本発明の構造体は分子量500以上の化合物を細孔内に取り込むことができるため、解析対象化合物は分子量500以上の化合物とすることができる。解析対象化合物の分子量の範囲は、創薬モダリティとして注目を集める中分子領域の化合物がターゲットとなり得るとの観点から、500~4000、好ましくは500~2000とすることができる。
本発明の解析用試料においてまた、解析対象化合物は、周期表中第3周期までの元素で構成されるものとすることができる。

<<構造解析対象化合物の構造解析方法>>
本発明の構造解析対象化合物の構造解析方法は、(A)本発明の細孔性三次元ネットワーク構造体に構造解析対象化合物を取り込ませて結晶構造解析用試料を調製する工程と、(B)前記結晶構造解析用試料の結晶構造解析を行う工程とを含んでなるものである。
本発明の構造解析対象化合物の構造解析方法は、(A)本発明の細孔性三次元ネットワーク構造体に構造解析対象化合物を取り込ませて結晶構造解析用試料を調製する工程と、(B)前記結晶構造解析用試料の結晶構造解析を行う工程とを含んでなるものである。
工程(A)では、本発明の結晶構造解析用試料の記載に従って、本発明の細孔性三次元ネットワーク構造体に構造解析対象化合物を取り込ませて結晶構造解析用試料を調製することができる。
工程(B)では、工程(A)で作製した結晶解析用試料を結晶構造解析することにより、対象物質の分子構造を決定することができる。結晶構造解析には、X線回折、中性子線回折および電子線回折のいずれの方法も使用することができる。また、測定データの解析、すなわち、解析対象化合物の構造解析は公知の方法に従って実施することができる。例えば、国際公開第2014/038220号やIUCrJ 2016, 3, 139-151の記載に従って実施することができる。
本発明の構造解析方法において、構造解析対象化合物は、本発明の結晶解析用試料についての記載と同様に、本発明の構造体の細孔内に取り込まれる限り、特に限定されるものではないが、本発明の構造体は分子量500以上の化合物を細孔内に取り込むことができるため、分子量500以上の化合物とすることができる。解析対象化合物の分子量の範囲は、創薬モダリティとして注目を集める中分子領域の化合物がターゲットとなり得るとの観点から、500~4000、好ましくは500~2000とすることができる。本発明の構造解析方法によれば、分子量500以上の化合物の立体構造を決定できる点で有利である。
以下の例に基づき本発明をより具体的に説明するが、本発明はこれらの例に限定されるものではない。
測定機器等
本実施例において、X線回折測定は単結晶X線回折装置(Synergy-R、リガク)を使用して行った。得られた回折データの解析は、ソフトウェア(CrysAlisPro、リガク)を用いて積分および吸収補正を行った後、SHELX T(Acta Crystallogr. Sect. A 2015, 71, 3-8)により初期構造を取得し、SHELX L(Acta Crystallogr. Sect. C 2015, 71, 3-8)を用いて精密化した。
本実施例において、X線回折測定は単結晶X線回折装置(Synergy-R、リガク)を使用して行った。得られた回折データの解析は、ソフトウェア(CrysAlisPro、リガク)を用いて積分および吸収補正を行った後、SHELX T(Acta Crystallogr. Sect. A 2015, 71, 3-8)により初期構造を取得し、SHELX L(Acta Crystallogr. Sect. C 2015, 71, 3-8)を用いて精密化した。
例1:結晶スポンジの作製(1)
例1では、[Zn2(R-2-Cl-man)2(dpb)3](TfO)2からなる結晶スポンジ(単結晶)を作製した。

1,4-ジ(4-ピリジル)ベンゼン(dpb)(東京化成)11.6mgをo-キシレン(1,2-ジメチルベンゼン)(東京化成)5mLに溶解した溶液を「溶液1」とした。o-キシレンとメタノールを体積比1:1で混合した溶媒5mLを「溶液2」とした。(R)-2-クロロマンデル酸(R-2-Cl-man)(東京化成)74.6mgおよびトリフルオロメタンスルホン酸亜鉛(東京化成)145.4mgをメタノール5mLに溶解した溶液を「溶液3」とした。溶液1を試験管に移し、溶液2を溶液1の上層に静かに添加した。さらに溶液3を溶液2の上層に静かに添加した。得られた3層溶液を室温で7日間静置して析出した結晶を回収し、o-キシレン中で保存した。そして、この結晶について、X線回折測定および回折データの解析を行った。

例3:結晶スポンジ法による対象化合物の構造解析(1)
例3では、例2で作製した結晶スポンジを用いて対象化合物の構造解析を行った。

例2で作成した結晶スポンジとして[Zn2(S-2-Cl-man)2(dpb)3](TfO)2を保存溶媒であるo-キシレンとともに1粒をV底型1.2mLのガラスバイアルに移し、o-キシレンを除去した後、イソプロパノール1mLで洗浄し溶媒を置換した。洗浄液であるイソプロパノールを除去した後、対象化合物としてベクロメタゾンプロピオン酸エステル(東京化成、CAS番号:5534-09-8、分子式:C28H37ClO7、分子量:521.042)をイソプロパノールに5mg/mLに溶解したサンプルを60μL添加し、結晶スポンジを対象化合物の溶液に浸漬した(対象化合物300μg、イソプロパノール60μL)。バイアルに蓋をして密封し、60℃で6日間静置して対象化合物を細孔内に取り込ませた。その後、結晶を取り出して、X線回折測定
および回折データの解析を行った。

例5では、例3および4とは異なる結晶スポンジを使用して、対象化合物の構造解析が可能かどうかを検討した。具体的には、分子量500未満の化合物に対して多数の構造解析実績を有し、三座配位子である2,4,6-トリス(4-ピリジル)-1,3,5-トリアジン(tpt)とハロゲン化亜鉛から成る結晶スポンジを用いた。
例5では、例3および4とは異なる結晶スポンジを使用して、対象化合物の構造解析が可能かどうかを検討した。具体的には、分子量500未満の化合物に対して多数の構造解析実績を有し、三座配位子である2,4,6-トリス(4-ピリジル)-1,3,5-トリアジン(tpt)とハロゲン化亜鉛から成る結晶スポンジを用いた。
2,4,6-トリス(4-ピリジル)-1,3,5-トリアジン(tpt)(東京化成)および塩化亜鉛(富士フイルム和光純薬)を用いて、Chem.Eur.J.2017,23,15035-15040記載の方法に従って[(ZnCl2)3(tpt)2]nの結晶スポンジ(単結晶)を調製した。[(ZnCl2)3(tpt)2]nはn-ヘキサン中で保存した。[(ZnCl2)3(tpt)2]n単結晶1粒をV底型1.2mLガラスバイアルに移し、n-ヘキサンを除去した後、イソプロパノール1mLで洗浄して溶媒の置換を試みた。しかし、この洗浄工程でイソプロパノールを添加した後、[(ZnCl2)3(tpt)2]nの結晶が黒ずんでしまい結晶性が低下したことが示唆された。その後、[(ZnCl2)3(tpt)2]nをガラスバイアル中で300μgのベクロメタゾンプロピオン酸エステル(対象化合物)を含む60μLのイソプロパノール溶液に浸った状態にし、60℃で静置したが、3日後にはバイアル中に存在した結晶が溶解し、消失してしまっており、結晶スポンジ法が適用できなかった。
例6:結晶スポンジ法による対象化合物の構造解析(4)
例6では、結晶スポンジを洗浄する溶媒および対象化合物の溶解に使用する溶媒を例5とは異なるものを用いて、対象化合物の構造解析が可能かどうかを検討した。

例5の結果と同様に、6日後にはバイアル中に存在した結晶が溶解または消失により、結晶スポンジ法が適用できなかった。
例7:結晶スポンジ法による対象化合物の構造解析(5)
例7では、結晶スポンジを洗浄する溶媒および対象化合物の溶解に使用する溶媒を例5とは異なるものを用いて、対象化合物の構造解析が可能かどうか検討した。

結晶スポンジである[(ZnCl2)3(tpt)2]nを洗浄する溶媒および対象化合物であるベクロメタゾンプロピオン酸エステルを溶解する溶媒を、n-ヘキサンとジメトキシエタンを体積比9:1で混合して調製した溶媒に変更したこと以外は例5の記載と同様にして対象化合物の構造解析が可能かどうかを検討した。
その結果、結晶スポンジへの浸漬期間の6日間経過後に[(ZnCl2)3(tpt)2]nの結晶がバイアル中にそのまま残っており、X線回折測定が可能であった。そこで、例1(1)の記載と同様にしてX線回折測定および回折データの解析を行ったところ、結晶スポンジの細孔内にはn-ヘキサンおよびジメトキシエタン(溶媒)が観測され、構造解析ターゲットであるベクロメタゾンプロピオン酸エステルは観測されなかった(図11)。すなわち、本例では細孔内にベクロメタゾンプロピオン酸エステルが取り込まれなかったことが示唆された。
例8:結晶スポンジ法による対象化合物の構造解析(6)

結晶スポンジとして[Zn2(S-2-Cl-man)2(dpb)3](TfO)2の代わりに[Zn2(S-2-Cl-man)2(bp)3](TfO)2(bpは4,4’-ビピリジンを表す)を用いること以外は例3(1)の記載と同様にしてX線回折測定および回折データの解析を行った。

結果は図12に示す通りであった。例1と同様に、用いたリガンド(bp)の配位部位間の距離を算出すると7.1Åであった。細孔内には溶媒であるイソプロパノールのみが観測され、構造解析対象化合物であるベクロメタゾンプロピオン酸エステルは観測されなかった。すなわち細孔内にベクロメタゾンプロピオン酸エステルが取り込まれなかったことが示唆された。
結果は図12に示す通りであった。例1と同様に、用いたリガンド(bp)の配位部位間の距離を算出すると7.1Åであった。細孔内には溶媒であるイソプロパノールのみが観測され、構造解析対象化合物であるベクロメタゾンプロピオン酸エステルは観測されなかった。すなわち細孔内にベクロメタゾンプロピオン酸エステルが取り込まれなかったことが示唆された。

例10:結晶スポンジ法による対象化合物の構造解析(8)
例10では、例2で作製した結晶スポンジを用いて対象化合物の構造解析を行った。

例2で作成した結晶スポンジとして[Zn2(S-2-Cl-man)2(dpb)3](TfO)2を保存溶媒であるo-キシレンとともに1粒をV底型1.2mLのガラスバイアルに移し、o-キシレンを除去した後、イソプロパノール1mLで洗浄し溶媒を置換した。洗浄液であるイソプロパノールを除去した後、対象化合物としてシクレソニド(東京化成、CAS番号:126544-47-6、分子式:C32H44O7、分子量:540.69)をイソプロパノールに5mg/mLに溶解したサンプルを60μL添加し、結晶スポンジを対象化合物の溶液に浸漬した(対象化合物300μg、イソプロパノール60μL)。バイアルに蓋をして密封し、50℃で6日間静置して対象化合物を細孔内に取り込ませた。その後、結晶を取り出して、X線回折測定および回折データの解析を行った。
(2)結果
結果は図14、図15および表5に示す通りであった。図14は、非対称単位構造を示す。図14の点線枠に示すように非対称単位あたり2分子のシクレソニド(ゲストA、ゲストB)が観測された。ゲストAおよびゲストBともに二回らせん軸周辺に位置しており、対称操作で生じるゲスト自身とディスオーダーしていた。また、ゲストBはゲストA、およびゲストAの2回らせん対称操作で生じるゲストとディスオーダーしており、これらを踏まえ最小二乗法でゲストAとゲストBの占有率を計算したところ、ゲストAが32.6(3)%、ゲストBが34.7(5)%となった。表5に結晶データを示す。また、図15は、単結晶の細孔内で観測されたシクレソニドのORTEP図(ゲストA、30%確率(probability))および構造式を示す。これらの結果から、[Zn2(S-2-Cl-man)2(dpb)3](TfO)2の細孔中にシクレソニドが取り込まれ、結晶スポンジ法によりその立体構造を明確に観測し、決定することができることが確認され、C、H、Oで構成される分子量500以上の化合物の結晶構造解析が可能であることが示された。
結果は図14、図15および表5に示す通りであった。図14は、非対称単位構造を示す。図14の点線枠に示すように非対称単位あたり2分子のシクレソニド(ゲストA、ゲストB)が観測された。ゲストAおよびゲストBともに二回らせん軸周辺に位置しており、対称操作で生じるゲスト自身とディスオーダーしていた。また、ゲストBはゲストA、およびゲストAの2回らせん対称操作で生じるゲストとディスオーダーしており、これらを踏まえ最小二乗法でゲストAとゲストBの占有率を計算したところ、ゲストAが32.6(3)%、ゲストBが34.7(5)%となった。表5に結晶データを示す。また、図15は、単結晶の細孔内で観測されたシクレソニドのORTEP図(ゲストA、30%確率(probability))および構造式を示す。これらの結果から、[Zn2(S-2-Cl-man)2(dpb)3](TfO)2の細孔中にシクレソニドが取り込まれ、結晶スポンジ法によりその立体構造を明確に観測し、決定することができることが確認され、C、H、Oで構成される分子量500以上の化合物の結晶構造解析が可能であることが示された。
例11:結晶スポンジの作製(3)
例11では、[Zn2(R-4-OH-man)2(dpb)3](TfO)2からなる結晶スポンジ(単結晶)を作製した。

(1)方法
1,4-ジ(4-ピリジル)ベンゼン(dpb)(東京化成)11.6mgをo-キシレン(1,2-ジメチルベンゼン)(東京化成)5mLに溶解した溶液を「溶液1」とした。o-キシレンとメタノールを体積比1:1で混合した溶媒5mLを「溶液2」とした。(R)-2-ヒドロキシ-2-(4-ヒドロキシフェニル)酢酸((R)-4-ヒドロキシマンデル酸(R-4-OH-man))(BLD Pharmatech)67.3mgおよびトリフルオロメタンスルホン酸亜鉛(東京化成)145.4mgをメタノール5mLに溶解した溶液を「溶液3」とした。溶液1を試験管に移し、溶液2を溶液1の上層に静かに添加した。さらに溶液3を溶液2の上層に静かに添加した。得られた3層溶液を室温で7日間静置して析出した結晶を回収し、o-キシレン中で保存した。そして、この結晶について、X線回折測定および回折データの解析を行った。

結果は図16、図17および表6に示す通りであった。図16および図17は、非対称単位構造およびパッキング構造をそれぞれ示す。表6は結晶データを示す。これらの結果から、1.57nmの細孔径を有する新規な単結晶(結晶スポンジ)を取得したことが確認された。また、用いたdpbの配位部位間の距離は11.4ÅであることがX線回折データから確認できた。
結果は図16、図17および表6に示す通りであった。図16および図17は、非対称単位構造およびパッキング構造をそれぞれ示す。表6は結晶データを示す。これらの結果から、1.57nmの細孔径を有する新規な単結晶(結晶スポンジ)を取得したことが確認された。また、用いたdpbの配位部位間の距離は11.4ÅであることがX線回折データから確認できた。

L2は二座配位子を表し、但し、前記二座配位子中の配位部位間の距離は8.0Å以上であり、

a、b、c、dおよびeはそれぞれ0を超える任意の整数を表す。)
の組成式で示される金属錯体を単位構造とする細孔性三次元ネットワーク構造体。 - 二座配位子が、式(IV):A1-X-A2
(上記式中、A1およびA2は、それぞれ独立に、金属イオンMと配位結合を形成する配位部位を有する基であり、Xは、リンカーを表す。)
で示される、請求項1または2に記載の構造体。 - 二座配位子の2つの配位部位が、二座配位子の中心位置に対して直線状またはほぼ直線状に存在するものである、請求項1または2に記載の構造体。
- 二座配位子の2つの配位部位が、窒素原子、酸素原子、硫黄原子、リン原子、ニトロ基、アミノ基、シアノ基、カルボキシ基および水酸基からなる群からそれぞれ独立に選択される、請求項1または2に記載の構造体。
- 請求項1または2に記載の細孔性三次元ネットワーク構造体の製造方法であって、金属塩およびα-ヒドロキシカルボン酸を含む溶液と、二座配位子(前記二座配位子中の配位部位間の距離は8.0Å以上である)を含む溶液とを接触させる工程を含む、製造方法。
- 請求項1または2に記載の細孔性三次元ネットワーク構造体および構造解析対象化合物からなる、結晶構造解析用試料。
- 構造解析対象化合物の分子量が500以上である、請求項7に記載の解析用試料。
- 結晶構造解析用試料の製造方法であって、請求項1または2に記載の細孔性三次元ネットワーク構造体に構造解析対象化合物を取り込ませる工程を含む、製造方法。
- (A)請求項1または2に記載の細孔性三次元ネットワーク構造体に構造解析対象化合物を取り込ませる工程と、(B)前記結晶構造解析用試料の結晶構造解析を行う工程とを含む、構造解析対象化合物の構造解析方法。
- 構造解析対象化合物の分子量が500以上である、請求項10に記載の構造解析方法。
