  • Compound 4 (34 mg, 56.5103 ⁇ mol) was separated by Prep-HPLC-Gilson with the following conditions: Column, CHIRAL ART Cellulose-SC column (2 cm ⁇ 25 cm, 5 ⁇ m) ; mobile phase, Hex (0.1%DEA) /EtOH (60: 40) ; Flowing rate: 20 mL/min to give Compound 4A (7 mg, the first eluting isomer, Retention Time 6.478 min) and Compound 4B (6 mg, the second eluting isomer, Retention Time 7.675 min) respectively.
  • Compound 30-1 was synthesized based on the same method as Compound 1-3 in Example 1, except that “azepane” in Example 1 is replaced with “4, 4-difluoroazepane hydrochloride” .
  • Compound 33-1 was synthesized based on the same method as Compound 1-1 in Example 1, except that “azepane” in Example 1 is replaced with “3-oxa-6-azabicyclo [3.1.1] heptane 4-methylbenzenesulfonate” .
  • Compound 34-1 was synthesized based on the same method as Compound 1-2 in Example 1, except that “azepane” in Example 1 is replaced with “ (1R, 4R) -2-oxa-5-azabicyclo [2.2.1] heptane” .
  • Compound 36-4 was synthesized based on the same method as Compound 1-4 in Example 1, except that “azepane” in Example 1 is replaced with “ (1R, 4R) -2-oxa-5-azabicyclo [2.2.1] heptane” .
  • Compound 61-4 was synthesized based on the same method as Compound 1-4 in Example 1, except that “azepane” in Example 1 is replaced with “8-oxa-3-azabicyclo [3.2.1] octane hydrochloride” .
  • Compound 62-4 was synthesized based on the same method as Compound 1-4 in Example 1, except that “azepane” in Example 1 is replaced with “3-oxa-6-azabicyclo [3.1.1] heptane 4-methylbenzenesulfonate” .
  • K-Ras G12D K-Ras G12V, K-Ras G12C, K-Ras G13D, K-Ras G12A, K-Ras G12R, K-Ras Q61H and K-Ras WT proteins were used in this assay.
  • K-Ras His tag, aa 1-169 pre-loaded with GDP was pre-incubated with each of compounds in the presence of 10 nM GDP in a 384-well plate (Greiner) for 15-60 mins, then purified SOS1 ExD (Flag tag, aa 564-1049) , BODIPY TM FL GTP (Invitrogen) and monoclonal antibody anti 6HIS-Tb cryptate Gold (Cisbio) were added to the assay wells and incubated for 4 hours at 25 °C (Specially, we did not add SOS1 in the K-Ras G13D assay) .
  • GppNp is an analog of GTP.
  • K-Ras G12D, K-Ras G12V, K-Ras G12C, K-Ras G13D, K-Ras G12A, K-Ras G12R, K-Ras Q61H and K-Ras WT proteins were used in this assay.
  • K-Ras His tag, aa 1-169 pre-loaded with GppNp was pre-incubated with each of compounds in the presence of 200 ⁇ M GTP in a 384-well plate (Greiner) for 15-60 mins, then cRAF RBD (GST tag, aa 50-132, CreativeBioMart) , monoclonal antibody anti GST-d2 (Cisbio) and monoclonal antibody anti 6HIS-Tb cryptate Gold (Cisbio) were added to the assay wells and incubated for 2 hours at 25 °C. Wells containing same percent of DMSO served as vehicle control, and wells without K-Ras served as low control.
  • MKN-1 with K-Ras WT amplification is also a K-Ras dependent cell line.
