一种用于活细胞成像的荧光染色方法,通过第一荧光生物标记物与第二荧光生物标记物对细胞进行双重染色,用荧光显微镜检测显现清晰的荧光细胞图像,所获取的图像可以在观察到细胞形态的同时能观察到细胞核形态。

本发明涉及细胞染色方法,具体涉及一种利用荧光生物标记物进行双重染色的方法,属于生物技术领域。
荧光染色方法与荧光显微技术的结合,使荧光显像在生物活性物质检测和细胞成像方面得到广泛应用,其中细胞成像技术是生命科学技术领域中重要的研究手段。相比于其他技术,荧光染色具有灵敏度高、选择性高、操作简单、反应灵敏等优点,是目前广泛应用于活细胞分析的一种高灵敏的可视化分析技术。
荧光是一种“光致发光”的冷发光现象。当某种常温物质经特定波长的入射光照射,吸收光能后进入激发态,并且立即发出比入射光的波长长的出射光,具有这种性质的出射光即被称为荧光。
荧光光谱包括激发光谱和发射光谱两种。激发光谱指荧光物质在不同波长光的激发下,该物质的某一发光谱线与谱带的强度或发光效率与激发光波长的关系;发射光谱指荧光物质在某一激发光的激发下,其不同波长的发光强度的强弱变化。每种荧光物质都有激发光谱和发射光谱,以及它最适宜的激发波段和发射波段。目前对于荧光的研究主要是研究荧光物质的激发光谱和发射光谱,以找到它们最适宜的激发波段和发射波段,例如临床上广泛使用的5-氨基乙酰丙酸(5-ALA)。
基于5-ALA的荧光检测恶性病变目前临床应用于脑外科、泌尿外科、胃肠外科,其中5-ALA诱导的原卟啉IX(PP IX)荧光检测 最近成为一种很有前途的术中检测恶性病变的方法。为了提高5-ALA荧光在强自发荧光条件下的检测精度,已有几种光谱分析方法(Valdes,P.A.et al.(2011)Neurosurg.115,11–17;Xu,H.&Rice,B.W.(2009)Journal of biomedical optics 14,064011;Harada,K.et al.(2013)International Journal of Molecular Sciences 14,23140–23152;Koizumi,N.et al.(2013)Ann.Surg.Oncol.20,3541–3548;Kondo,Y.et al.(2014)Int.J.Oncol.45,41–46)。虽然一些研究报道了5-ALA荧光检测方法在临床应用中的有效性,但由于发色团自身荧光背景强,经常出现检测错误。
一些荧光生物标记物已被研究,除了上述5-ALA,目前使用比较广泛的还有荧光素钠(Fluorescein,FS)和吲哚菁绿(ICG)。
荧光素钠(Fluorescein)是一种活体荧光染料,其水溶液可以对细胞胞体轮廓进行标记,用于改善肿瘤组织的可视化,其对肿瘤细胞没有特异性。这种染料在被波长在460–500nm范围内的光激发时,会发出波长在540–690nm的荧光辐射,在医学上有着广泛的应用,特别是在脑肿瘤手术中应用更为广泛(Copeman SM,Coke F,Gouldesbrough C.,BrMed J.(1929)2:233–42;Hamamcioglu MK.et al.,Clin Neurol Neurosurg(2016)143:39-45;O’goshi K,Serup J.,Ski Res Technol.(2006)12:155–61;Koc K.et al.,Br J Neurosurg(2008)22:99-103;Hara T.et al.,Am J Ophthalmol.(1998)126:560–4;Kuroiwa T.et al.,SurgNeurol(1998)50:41-8)。
吲哚菁绿(ICG)是一种两亲性小分子(<800道尔顿),一种近红外光谱(NIR)荧光团(峰激发=805nm,峰发射=835nm),当静脉注射时,通常留在血管内,主要与白蛋白和其他血浆蛋白结合,进行近红外成像以描绘血管系统。近年来又发现ICG在大鼠和人类患者肿瘤组织中的研究,发现ICG在肿瘤中积累,肿瘤和背景之间有显著的对比,ICG已被证明在标记肿瘤组织方面具有实用价值(Cho SS.et al.,(2019)Front.Surg.6:11;Hansen DA et al.,Surg Neurol.(1993) 40:451–6;Haglund MM.et al.,Neurosurgery.(1994)35:930-40;Haglund MM.,et al.,Neurosurgery.(1996)38:308–17;Madajewski B.,et al.,Clin Cancer Res.(2012)18:5741–51;30.Jiang JX.,et al.,Am J Nucl Med Mol Imaging.(2015)5:390–400;Zeh R.et al.,PLoS ONE.(2017)12:e0182034)。
美兰(Methylene Blue,MB),又称亚甲基蓝,是美国食品和药物管理局批准的用于治疗高铁血红蛋白血症的染料,是一种无毒的替代染料。美兰是目前除了ICG,另一个在人体临床试验可用的近红外荧光团,其在700nm处有峰值发射,与荧光素钠和ICG在不同的近红外波段发出荧光。
尽管目前对荧光染料的研究和应用非常广泛,但是这些荧光染料染色效果仍有不足之处,举例来说,荧光素钠和ICG没有特异性;5ALA特异性要高于荧光素钠,但是其敏感性不高,与周围正常组织对比微弱(OkudaT.et al.,J ClinNeurosci.(2012)19:1719–22;Acerbi F.et al.,Neurosurg.Focus.(2014)36:E5;Okuda T.et al.,J Clin Neurosci.(2010)17:118–121;35.Chen B.et al.,Int J Med Sci.(2012)9:708–714;Francaviglia N.et al.,Surg Neurol Int.(2017)8:145;Bowden SG.et al.,Neurosurgery.(2018)82:719–27);一些研究尝试了5-ALA和荧光素的双重注射,通过增加摄取5-ALA的肿瘤组织与摄取荧光素的瘤周区域之间的对比来增强对肿瘤组织的检测(Suero Molina E.et al.,J Neurosurg.(2018)128:399–405),效果有所改善,但仍然不足以满足临床需求。
上述为荧光染色剂在临床上的广泛应用,通过细胞团块对于染色剂的吸收与转化,结合荧光“光致发光”的冷发光现象,形成荧光成像,方便进行整体的组织的观察的案例。而在荧光显微观察活细胞时,由于目前这些染色剂在被单个细胞吸收时,仅能被细胞体吸收或被细胞核吸收。在活细胞荧光染色并进行荧光显微观察时,吖啶黄染色成像可以观察到细胞核形态,但无法观察细胞形态,荧光素钠染色 成像可以观察查到细胞形态,但无法观察细胞核形态。在进行显微观察时,仅能观察细胞核形态或细胞形态,不满足细胞观察时同时需要观察细胞核与细胞形态的需求。
综上所述,目前常用于活细胞成像分析的荧光染色方法在实际应用中存在需将细胞固定,使用过程繁琐,染色过程时间长、无特异性、对比度不足、无法同时清楚观察到细胞形态和细胞核形态等缺点。因此,提高可视化分析的选择性和精度是目前迫切需要解决的问题。
发明内容
本发明的主要目的在于提供一种新的染色方法,利用荧光生物标记物对活细胞进行双重染色以克服现有技术中的不足。
为实现前述目的,本发明提供如下技术方案:
本发明提供一种活细胞染色的方法,该方法包括以下步骤:同时或分别(i)使用第一荧光生物标记物对目标细胞进行染色;(ii)使用第二荧光生物标记物对前述目标细胞进行染色;(iii)用荧光显微镜获取前述目标细胞的荧光图像,所述方法在观察到细胞形态的同时能观察到细胞核形态。
优选地,上述第一荧光生物标记物具有460–800nm的激发波长。
优选地,上述第二荧光生物标记物具有350–670nm的激发波长。
优选地,上述第二荧光生物标记物的发射光谱最大值与第一荧光生物标记物的发射光谱最大值相差至少50nm。
优选地,当使用荧光显微镜获取所述目标细胞的荧光图像时,上述第一荧光生物标记物发光,同时第二荧光生物标记物吸收光。

本发明的组合物还使用一种或多种生理上可接受的载剂调制,所述载剂包含赋形剂和助剂,其有助于将活性剂加工成可药用的制剂。适当的调配物取决于所选择的施用途径。
前述稳定剂可选自山梨醇酯、聚氧乙烯氢化蓖麻油、聚乙烯醇中的一种或多种。
前述抗氧化剂可选自亚硫酸钠、亚硫酸钾、硫酸钠、硫酸钾、柠檬酸、二丁基羟基甲苯、叔丁基对苯二酚、枸橼酸中的一种或多种。
前述保护剂可选自羟丙基甲基纤维素、医用透明质酸钠、聚丙烯酰胺、卡波姆、木糖醇、葡萄糖、烷基糖苷的一种或多种。
前述pH调节剂可选自磷酸二氢钠、磷酸氢二钠、磷酸二氢钾、磷酸氢二钾、硼酸、硼砂、醋酸、醋酸钠、柠檬酸、柠檬酸钠、酒石 酸、酒石酸钠、碳酸钠、碳酸钾、碳酸氢钠、碳酸氢钾、氢氧化钠、氢氧化钾、三乙醇胺、盐酸和磷酸中的一种或多种。
前述防腐剂可选自苯扎氯铵,苯扎溴铵,三氯叔丁醇,山梨醇中的一种或多种。
本发明前述组合物在实际销售时可以有其他替代名称,例如试剂盒、套件、套组或系统等。前述组合物中两种物质可以混合在一起包装,也可独立分开包装。
本发明发现了荧光素钠和美兰双染色法进行成像可以显着提升成像效果,荧光素钠提高了与背景的对比度,同时美兰又可以很好的标记细胞核,如此一来就可以在观察到细胞形态的同时又清晰的观察到细胞核,特别是肿瘤细胞的核质比变化等重要信息,因此可以清晰的判断出正常组织和肿瘤组织,达到肿瘤和非肿瘤组织边界的可视化,以满足临床需求。并且目前这两种染料都已被批准在临床使用,安全性得到了保证。
在本发明的优选实施方案中,举例说明使用两种不同荧光波长的染剂。应当理解,可以结合使用三种或以上的不同染剂进行组织染色,只要其识别的标的彼此不同,并且具有不同荧光波长即可。
进一步地,荧光生物标记物还可选自以下染剂:异硫氰酸荧光素(FITC)、藻红蛋白(phycoerythrin)、藻蓝蛋白(phycocyanin)、别藻蓝蛋白(allophycocyanin)、邻苯二甲醛(ophthaldehyde)、若丹明(rhodamine)以及AlexaFluor系列染料、DAPI、Hoechst 33342噻唑橙、吖啶橙等。
在实际应用时,本发明的染色方法不仅可以应用于病理组织切片,而且可以应用于活体组织以及离体培养的细胞染色,可将待成像组织的细胞、微载体上的细胞或涂片上的细胞与本发明的染剂直接接触染色,进一步应用于外科手术、诊断、药物递送等临床用途。
与现有技术相比,本发明的有益效果在于:
本发明提供一种用于活细胞成像的荧光生物标记物双重染色法,其具有快捷、高效、安全等优势,在观察到细胞形态的同时能观察到细胞核形态,除此之外可以清晰的辨明正常细胞和肿瘤细胞,,对组织染色、细胞形态研究、图像引导外科手术等领域发展有着至关重要的意义。

小鼠
小鼠为C57BL6品系,购于南大动物中心。
荧光图像分析
荧光图像是由MCI显微镜(DiveScope)获得,将样本染色后置于470nm波长下观察,随机选取感兴趣的区域成像。
染色液配制
0.1%荧光素钠(Flourescein):称取0.01g的荧光素钠粉末,装进避光试管里,加入10mL的生理盐水,摇匀后用锡箔纸包裹配好的溶液,避光保存。
0.25%荧光素钠(Flourescein):称取0.025g的荧光素钠粉末,装进避光试管里,加入10mL的盐水清洁液,摇匀后用锡箔纸包裹配好的溶液,避光保存。
0.5%荧光素钠(Flourescein):称取0.05g的荧光素钠粉末,装进避光试管里,加入10mL的生理盐水,摇匀后用锡箔纸包裹配好的溶液,避光保存。
1%荧光素钠(Flourescein):称取0.1g的荧光素钠粉末,装进避光试管里,加入10mL的生理盐水,摇匀后用锡箔纸包裹配好的溶液,避光保存。
0.5%美兰(亚甲基蓝,Methylene blue):称取0.05g的亚甲基蓝粉末装进避光试管里,加入10mL的5%的碳酸氢钠溶液,摇匀后用锡箔纸包裹配好的溶液,避光保存。
1%美兰(亚甲基蓝,Methylene blue):称取0.1g的亚甲基蓝粉末装进避光试管里,加入10mL的5%的碳酸氢钠溶液,摇匀后用锡箔纸包裹配好的溶液,避光保存。
2%美兰(亚甲基蓝,Methylene blue):称取0.2g的亚甲基蓝粉末装进避光试管里,加入10mL的5%的碳酸氢钠溶液,摇匀后用锡箔纸包裹配好的溶液,避光保存。
3%美兰(亚甲基蓝,Methylene blue):称取0.3g的亚甲基蓝粉末装进避光试管里,加入10mL的5%的碳酸氢钠溶液,摇匀后用锡箔纸包裹配好的溶液,避光保存。
0.05%5-ALA(5-aminolevulinic acid):称取0.005g的5-ALA粉末,装进避光试管里,加入10mL的5%葡萄糖溶液,摇匀后用锡箔纸包裹配好的溶液,避光保存。
1%吖啶黄(acriflavine):称取0.1g的吖啶黄粉末,装进避光试管里,加入10mL的生理盐水,摇匀后用锡箔纸包裹配好的溶液,避光保存。
0.05%ICG(indocyanine-green):称取0.005g的ICG粉末,装进避光试管里,加入10mL的生理盐水,摇匀后用锡箔纸包裹配好 的溶液,避光保存。
0.05%结晶紫(crystal violet):称取0.005g的结晶紫粉末,装进避光试管里,加入10mL的生理盐水,摇匀后用锡箔纸包裹配好的溶液,避光保存。
实施例1:荧光素钠和美兰双染色法进行组织染色
1.1小鼠肾脏染色
使用小鼠进行组织染色,染色流程如下:
1.小鼠使用1%戊巴比妥钠进行腹腔注射麻醉,麻醉剂量为8–9mL/g。
2.去除小鼠背侧部体毛,剪开表皮,暴露出肾脏,将肾脏完整切除取出后,用刀片固定肾脏表面,于显微镜下用剪刀和镊子去除肾脏表面被膜。
3.用棉签止血,以0.25%荧光素钠对肾脏表面敷染2分钟后,用生理盐水清洗三次;接着以1%美兰染色液表面敷染2分钟,时间到后用生理盐水清洗三次,然后置于显微镜470nm波长下观察。
图1的结果显示小鼠肾脏在单独使用美兰染色、单独使用荧光素钠染色和美兰-荧光素钠双染色法成像效果比对。其中图1A为单独使用荧光素钠的染色在470nm波长下成像结果;图1B为单独使用美兰的染色在470nm波长下成像结果,可以看到在470nm波长下,美兰无法成像;图1C为单独使用美兰的染色在660nm波长下成像结果;图1D为荧光素钠和美兰双染色在在470nm波长下成像结果,可以清楚的看到经荧光素钠和美兰双染色的小鼠肾小管细胞轮廓以及细胞核的形态;图1E为HE染色对照。
1.2小鼠肝脏染色
使用小鼠进行组织染色,染色流程如下:
1.小鼠使用1%戊巴比妥钠进行腹腔注射麻醉,麻醉剂量为8–9mL/g。
2.去除小鼠腹部体毛,剪开表皮,暴露出肝脏,将肝脏完整切除取出后,用刀片固定肝脏表面。
3.以0.25%荧光素钠对肝脏表面敷染2分钟后,用生理盐水清洗三次;接着以1%美兰染色液表面敷染2分钟,时间到后用生理盐水清洗三次,然后置于显微镜470nm波长下观察。
图2为小鼠肝脏在单独使用美兰染色与单独使用荧光素钠和美兰双染色法成像效果比对。其中图2A为单独使用荧光素钠的染色在470nm波长下成像结果;图2B为单独使用美兰的染色在470nm波长下成像结果,可以看到在470nm波长下,美兰无法成像;图2C为荧光素钠和美兰双染色在在470nm波长下成像结果,可以清楚的看到经荧光素钠和美兰双染色的小鼠肝脏细胞轮廓以及细胞核的形态;图2D为HE染色对照。
由图1和2可以发现,使用荧光素钠和美兰双染色法比单独使用荧光素钠或者单独使用美兰的成像效果都有很大的提升。荧光素钠和美兰双染色法提高了与背景的对比度,可以清晰的观察到组织和细胞形态,同时又可以观察到清晰的细胞核,甚至是细胞核仁。这些信息都将帮助医生对于肿瘤与非肿瘤组织的判定,为医疗带来更大的价值。
实施例2:不同浓度荧光素钠和美兰染色双染色的效果
使用猪肾脏进行组织染色,染色流程如下:
1.采购新鲜猪肾脏若干,于4℃冰箱冷藏备用。
2.将新鲜猪肾脏用清水清洗干净,于显微镜下用剪刀和镊子去除肾脏表面被膜。
3.分别以0.1%、0.25%、0.5%、1%荧光素钠对肾脏表面敷染一分钟,然后用生理盐水清洗三次;接着分别以0.5%、1%、2%、3%美兰染色液表面敷染一分钟,时间到后用生理盐水清洗三次,然后置于显微镜470nm波长下观察。
图3为猪肾脏使用不同浓度荧光素钠和美兰双组合染色成像效果比对。其中图3A为0.1%荧光素钠+0.5%美兰组合染色在470nm波长下成像结果;图3B为0.25%荧光素钠+1%美兰组合染色在470nm波长下成像结果;图3C为0.5%荧光素钠+2%美兰组合染色在在470nm波长下成像结果;图3D为1%荧光素钠+3%美兰组合染色在470nm波长下成像结果。
由图3可以发现,不同浓度的荧光素钠和美兰组合,其中荧光素钠浓度为0.1%–1%之间,美兰浓度为0.5%–3%之间,在此浓度间两者组合染色效果相近,在此浓度范围内的组合,都能够清晰的分辨出细胞形态及细胞核结构。
实施例3:5-ALA和吖啶黄双染色法进行组织染色
使用猪肝脏进行组织染色,染色流程如下:
1.采购新鲜猪肝脏若干,于4℃冰箱冷藏备用。
2.将新鲜猪肝脏用清水清洗干净,于显微镜下用剪刀和镊子去除肝脏表面被膜。
3.以0.05%5-ALA对肝脏表面敷染3分钟后,用生理盐水清洗三次;接着以1%吖啶黄染色液表面敷染2分钟,时间到后用生理盐水清洗三次,然后置于显微镜635nm波长下观察。
图4为猪肝脏使用5-ALA和吖啶黄双染色的成像结果,可以清楚的看到经染色的肝脏细胞轮廓以及细胞核的形态。
实施例4:美兰和ICG双染色法进行组织染色
使用猪肝脏进行组织染色,染色流程如下:
1.采购新鲜猪肝脏若干,于4℃冰箱冷藏备用。
2.将新鲜猪肝脏用清水清洗干净,于显微镜下用剪刀和镊子去除肝脏表面被膜。
3.以1%美兰对肝脏表面敷染2分钟后,用生理盐水清洗三次;接着以0.05%ICG染色液表面敷染2分钟,时间到后用生理盐水清洗三次,然后置于显微镜835nm波长下观察。
图5为猪肝脏使用美兰和ICG双染色的成像结果,可以清楚的看到经染色的肝脏细胞轮廓以及细胞核的形态。
实施例5:荧光素钠和结晶紫双染色法进行组织染色
使用猪肾脏进行组织染色,染色流程如下:
1.采购新鲜猪肾脏若干,于4℃冰箱冷藏备用。
2.将新鲜猪肾脏用清水清洗干净,于显微镜下用剪刀和镊子去除肾脏表面被膜。
3.以0.25%荧光素钠对肾脏表面敷染2分钟后,用生理盐水清洗三次;接着以0.05%结晶紫染色液表面敷染3分钟,时间到后用生理盐水清洗三次,然后置于显微镜525nm波长下观察。
图6为猪肾脏使用荧光素钠和结晶紫双染色的成像结果,可以 清楚的看到经染色的肝脏细胞轮廓以及细胞核的形态。

Claims (15)
- 一种用于活细胞成像的染色方法,其特征在于,所述方法包括以下步骤:同时或分别(i)使用第一荧光生物标记物对目标细胞进行染色;(ii)使用第二荧光生物标记物对所述目标细胞进行染色;(iii)用荧光显微镜获取所述目标细胞的荧光图像,所述方法在观察到细胞形态的同时能观察到细胞核形态。
- 根据权利要求1所述的方法,其特征在于,所述第一荧光生物标记物具有460–800nm的激发波长,所述第二荧光生物标记物具有350–670nm的激发波长。
- 根据权利要求1所述的方法,其特征在于,所述第二荧光生物标记物的发射光谱最大值与所述第一荧光生物标记物的发射光谱最大值相差至少50nm。
- 根据权利要求1所述的方法,其特征在于,用荧光显微镜获取所述目标细胞的荧光图像时,所述第一荧光生物标记物发光,所述第二荧光生物标记物吸收光。
- 根据权利要求4所述的方法,其特征在于,所述第一荧光生物标记物选自荧光素钠、5-氨基乙酰丙酸和吲哚菁绿;所述第二荧光生物标记物选自美兰、吖啶黄和结晶紫。
- 根据权利要求5所述的方法,其特征在于,所述第一荧光生物标记物为荧光素钠,所述第二荧光生物标记物为美兰。
- 根据权利要求5所述的方法,其特征在于,所述第一荧光生物标 记物为5-氨基乙酰丙酸,所述第二荧光生物标记物为吖啶黄。
- 根据权利要求5所述的方法,其特征在于,所述第一荧光生物标记物为吲哚菁绿,所述第二荧光生物标记物为结晶紫。
- 根据权利要求1所述的方法,其特征在于,所述方法用于活体组织染色。
- 根据权利要求1所述的方法,其特征在于,所述活细胞包含癌细胞。
- 一种用于活细胞染色的组合物,其特征在于,所述组合物包括权利要求1所述的第一荧光生物标记物和第二荧光生物标记物。
- 根据权利要求11所述的组合物,其特征在于,所述第一荧光生物标记物为荧光素钠,所述第二荧光生物标记物为美兰。
- 根据权利要求11所述的组合物,其特征在于,所述第一荧光生物标记物为5-氨基乙酰丙酸,所述第二荧光生物标记物为吖啶黄。
- 根据权利要求11所述的组合物,其特征在于,所述第一荧光生物标记物为吲哚菁绿,所述第二荧光生物标记物为结晶紫。
- 根据权利要求11所述的组合物,其特征在于,所述组合物还包括稳定剂、抗氧化剂、保护剂、防腐剂、pH调节剂。
