沉香木萃取物用于抗菌的用途及其组合物 技术领域
本发明是关于一种沉香木的用途,尤其指一种沉香木萃取物用于抗菌的用途及其组合物。
背景技术
沉香树为一种沉香属植物,当其木头因受外力而破损时,会分泌出树脂而与木质部分产生结香现象,因而生成所谓的“沉香”,其除了是一种珍贵的药材而可作为药物用途,也因其具有特殊的香气而被制成祭拜用的立香。
技术问题
沉香树除可生成沉香而具上述用途之外,其本身更有其实用价值,例如如中国台湾公告第I389701号发明专利,即公开一种沉香属果壳的萃取物及其于治疗癌症的用途;再如中国台湾公告第I687405号发明专利案,也公开一种沉香树籽萃取物、制备方法及其应用于抗过敏的用途。然而,现有的沉香树如所述果壳或树籽的萃取物,皆未以沉香木萃取物用于抗菌为用途。
技术解决方案
为解决上述课题,本发明提供一种沉香木萃取物用于抗菌的用途及其组合物,所述沉香木萃取物是萃取自沉香木而来。
本发明的一项实施例提供沉香木萃取物用于抗菌的用途,所述沉香木萃取物是取沉香木与一溶剂混合而在一萃取条件下预制一沉香木萃取液,再由沉香木萃取液通过过滤及干燥所获得,用于杀死或抑制微生物。
较佳地,萃取条件是所述沉香木与超纯水以1:10的比例混合,且在100℃的加热温度下萃取1小时。
较佳地,萃取条件是所述沉香木与酒以1:10的比例混合,且在20℃至50℃的加热温度下萃取2小时至8小时。
较佳地,所述过滤是经粗过滤后收集上清液,再对所收集的上清液进行细过滤。
较佳地,所述粗过滤是以10000rpm的转速对所述沉香木萃取液进行离心式过滤,过滤时间为5分钟。
较佳地,所述细过滤是将上清液先以定性滤纸过滤,再经玻璃纤维滤膜过滤。
较佳地,所述玻璃纤维滤膜的孔径为1.2μm。
较佳地,所述干燥是经所述过滤的沉香木萃取液于一低温环境放至结冰状态,再经冷冻干燥成粉状后储存。

本发明的一项实施例提供一种用于灰指甲抗菌的组合物,含有如前述的沉香木萃取物的溶液、水、甘油、尿素、尿囊素、苯氧乙醇和乙基己基甘油混合物,以及对羟基苯甲酸甲酯的组合物,所占重量百分比分别为沉香木萃取物的溶液40%~55%、水35%~50%、甘油1%~5%、尿素0.5%~1.5%、尿囊素0.05%~0.3%、苯氧乙醇和乙基己基甘油混合物0.05%~0.3%,以及对羟基苯甲酸甲酯0.01%~0.2%。
本发明的另一项实施例提供一种用于灰指甲抗菌的组合物,含有前述的沉香木萃取物的溶液、蒸馏水、1,3-丁二醇、黄原胶、苯氧乙醇和乙基己基甘油混合物,以及对羟基苯甲酸甲酯的组合物,其中所占重量百分比分别为沉香木萃取物的溶液40%~55%、蒸馏水35%~50%、1,3-丁二醇1%~8%、黄原胶0.1%~0.8%、苯氧乙醇和乙基己基甘油混合物0.1%~0.8%,以及对羟基苯甲酸甲酯0.01%~0.2%。
借此,本发明以沉香木进行萃取而获得沉香木萃取物,其经试验证明,将沉香木萃取物用于抗菌的用途,确实可有效杀死或抑制微生物而达到抗菌效果,以此作为沉香木应用的新用途。

为便于说明本发明于上述发明内容一栏中所表示的中心思想,现以具体实施例表达。实施例中各种不同对象是按适于说明的比例、尺寸、变形量或位移量而描绘,而非按实际组件的比例予以绘制,合先叙明。
本发明提供一种沉香木萃取物用于抗菌的用途及其组合物,本发明所述抗菌,是指杀死或抑制微生物的能力。所述沉香木萃取物,其萃取流程如图1所示,是取沉香木与一溶剂混合(如步骤S1),而在一萃取条件下预制一沉香木萃取液(如步骤S2),再由沉香木萃取液通过过滤及干燥(如步骤S3、S4)所获得,所述沉香木萃取物用于杀死或抑制微生物,以作为抗菌的用途。于本实施例中所述步骤S1,是对沉香木进行木屑的刨取,再以获得的木屑与所述溶剂混合,但不以此为限,例如将沉香木取块后进行搅碎成粉状也可。
较佳地,所述萃取条件是所述沉香木与一溶剂以1:10的比例混合,且在100℃的加热温度下萃取1小时,而所述溶剂于此是以超纯水为较佳实施例,但不以此为限,所述溶剂也可为酒,例如具有高酒精浓度的酒液(如酒精浓度在50度以上的),作为溶剂以浸泡沉香木。如溶剂为酒,是沉香木与酒同样以1:10的比例混合,而在20℃(常温)至50℃的温度下萃取2小时至8小时为所述萃取条件。
较佳地,所述过滤是经粗过滤后收集上清液,再对所收集的上清液进行细过滤。所述粗过滤系以10000rpm的转速对所述沉香木萃取液进行离心式过滤,且过滤时间为5分钟;所述细过滤是将上清液先以定性滤纸(使用Whatman NO.4滤纸)过滤,再经玻璃纤维滤膜过滤,所述玻璃纤维滤膜的孔径为1.2μm。
较佳地,所述干燥,是经所述过滤的沉香木萃取液于一低温环境放至结冰状态,再经冷冻干燥成粉状后储存(例如放置于药品柜),所述低温环境的温度以零下20℃为较佳。
为说明本发明的沉香木萃取物用于抗菌的用途,于本实施例中是以须毛癣菌(Trichophyton mentagrophytes , ATCC 28185)为试验菌株进行抗菌试验,此抗菌试验是参考JIS Z 2801(2012)抗菌测试方法。
于本次抗菌试验前,先进行试验分组,包括实验组以及对照组,其中实验组是以上述沉香木萃取物为检体,而对照组为0.85%的无菌生理食盐水。本次抗菌试验中,须毛癣菌期望的菌数,是经调配浓度至约107CFU/mL的菌量。
本次抗菌试验的进行,是取实验组以及对照组各10mL,且自调配须毛癣菌的检体各取0.1mL的菌种悬浮液分别接种于实验组以及对照组,接种菌量为1.6 x 105 CFU/mL,接种后在25±2℃的环境下作用3小时。接着,以9 mL的无菌生理食盐水分别将实验组以及对照组进行10倍连续(序列)稀释(Decimally Serial Dilution),再分别置于一培养基在25±2℃的环境下培养后(如图2所示),分别记录须毛癣菌在实验组以及对照组中的残留菌量(菌落数)。如图3所示,经记录发现须毛癣菌在实验组中的残留菌量为85 CFU/mL,须毛癣菌在对照组中的残留菌量为1.9 x 105 CFU/mL,如欲知其抗菌能力,则须进行抗菌率的计算,其计算公式如下:
抗菌率(%) = (对照组残留菌量-实验组残留菌量)/对照组残留菌量 x 100%。
经上述抗菌试验的结果发现,依须毛癣菌在实验组中的残留菌量(85 CFU/mL),以及须毛癣菌在对照组中的残留菌量(1.9 x 105CFU/mL)进行抗菌率的计算可知,含有本发明的沉香木萃取物的实验组,相对于对照组的抗菌率为99.95%,确实具有极佳的抗菌效果,以作为抗菌的用途。
本发明进一步提供一种用于灰指甲抗菌的组合物,是含有前述沉香木萃取物的溶液、水、甘油(Glycerin)、尿素(Urea)、尿囊素(Allantoin)、苯氧乙醇和乙基己基甘油混合物(Phenoxyethanol & Ethylhexylglycerin),以及对羟基苯甲酸甲酯(Methyl paraben)的组合物,所占重量百分比分别为沉香木萃取物的溶液40%~55%、水35%~50%、甘油1%~5%、尿素0.5%~1.5%、尿囊素0.05%~0.3%、苯氧乙醇和乙基己基甘油混合物0.05%~0.3%,以及对羟基苯甲酸甲酯0.01%~0.2%,以组成一种灰指甲抗菌水。
本发明并提供另一种用于灰指甲抗菌的组合物,是含有前述沉香木萃取物的溶液、蒸馏水、1,3-丁二醇(1.3-Butylene Glycol)、黄原胶(Xanthan gum)、苯氧乙醇和乙基己基甘油混合物,以及对羟基苯甲酸甲酯的组合物,其中所占重量百分比分别为沉香木萃取物的溶液40%~55%、蒸馏水35%~50%、1,3-丁二醇1%~8%、黄原胶0.1%~0.8%、苯氧乙醇和乙基己基甘油混合物0.1%~0.8%,以及对羟基苯甲酸甲酯0.01%~0.2%,以组成一种灰指甲抗菌凝露。
以上所举实施例仅用于说明本发明而已,非用于限制本发明的保护范围。举凡不违本发明所从事的种种修改或变化,皆属于本发明的保护范围。

  1. 一种沉香木萃取物用于抗菌的用途,其特征在于,所述沉香木萃取物是取沉香木与一溶剂混合而在一萃取条件下预制一沉香木萃取液,再由该沉香木萃取液通过过滤及干燥所获得,用于杀死或抑制微生物。
  2. 如权利要求1所述的沉香木萃取物用于抗菌的用途,其特征在于,该萃取条件是所述沉香木与超纯水以1:10的比例混合,且在100℃的加热温度下萃取1小时。
  3. 如权利要求1所述的沉香木萃取物用于抗菌的用途,其特征在于,该萃取条件是所述沉香木与酒以1:10的比例混合,且在20℃至50℃的加热温度下萃取2小时至8小时。

  12. 一种用于抗菌的组合物,其特征在于,所述组合物包括如权利要求1所述的沉香木萃取物的溶液、蒸馏水、1,3-丁二醇、黄原胶、苯氧乙醇和乙基己基甘油混合物,以及对羟基苯甲酸甲酯的组合物,其中所占重量百分比分别为沉香木萃取物的溶液40%~55%、蒸馏水35%~50%、1,3-丁二醇1%~8%、黄原胶0.1%~0.8%、苯氧乙醇和乙基己基甘油混合物0.1%~0.8%,以及对羟基苯甲酸甲酯0.01%~0.2%。
