Definitions

  • the present invention refers to the area of therapeutical agents and relates to a me- dicament for treating inflammatory conditions of human skin associated with an increased expression of IL-6.
  • Interleukin-6 (short: IL-6, synonymous older names: Interferon- ⁇ 2 (IFNB2), B-cell stim- ulating factor, B-cell differentiation factor, liver cell stimulating factor) belongs to the inter- leukins (or more comprehensively to the cytokines), which regulate the inflammatory reaction of the organism.
  • IL-6 by the nature of its complex regulation and functions in the orchestra of other cytokines and cells, plays a key role in the transition from mechanisms of innate im- munity to mechanisms of acquired immunity within the inflammatory process.
  • IL-6 is encoded on chromosome 7 gene locus p21.
  • the gene segment contains 5 exons.
  • An inactive precursor protein consisting of 212 amino acids is transcribed. Transcription is induced by the transcription factors NF-kB, NF-AT, HSF1 and HSF2, among others.
  • the 184 amino acids long active interleukin-6 is cleaved from the precursor protein. Interleukin-6 can bind to two receptor types as an activating ligand:
  • IL-6R membrane-bound IL-6 receptor
  • IL-6 receptor a soluble IL-6 receptor
  • sIL-6R soluble IL-6 receptor
  • the resulting IL-6/sIL-6R complex binds to and acti- vates the glycoprotein gpl30, which is the only glycoprotein found in the membranes of many cell types.
  • This second process is called "IL-6-trans-signaling". It is important for the numerous cells that express gpl30 (which occurs ubiquitously) but not IL-6R on their cell surface. IL-6-trans-signaling is inhibited by naturally occurring soluble sgpl30, which inactivates the IL-6/sIL-6R complex, but not IL-6 alone and its effect on the membrane-bound IL-6 receptor.
  • glycoprotein Gpl30 is activated by an IL-6/sIL-6R complex or when an IL- 6R (to which gpl30 is associated) is activated by IL-6, a Janus-activated kinase at its domain inside the cell is phosphorylated at a tyrosine, thereby activating the JAK-STAT signalling pathway and the MAP kinase pathway, which then leads to the transcription of specific target genes in the cell nucleus.
  • interleukin-6 is usually considered an activator of acute phase proteins and a lymphocyte stimulating factor.
  • neutrophil granulocytes are recruited, infiltrate the site of inflammation, do their job, die rapidly and are replaced by a longer lasting popula- tion of more specific inflammatory cells such as lymphocytes and mononuclear cells.
  • interleukin-6 efficacy plays an important role: With the degree of infiltration by neutrophil granulocytes, the local concentration of SIL-6R increases, which triggers IL-6-trans-signaling in the surrounding tissue. This in turn leads to a limitation of the accumulation of neutrophilic granulocytes in the inflamed tissue via different pathways.
  • IL-6-trans- signaling attracts CD3+-T lymphocytes, which mark the transition of the innate immune re- sponse to a learned immune response.
  • IL-6 is involved in the regulation of leu- kocyte apoptosis with pro-apoptotic and anti-apoptotic agents (of which it is not yet known how they are balanced).
  • IL-6 has an anti-apoptotic effect on resting and activated T lympho- cytes, and regulates their differentiation, proliferation, polarization and immunoglobulin G secretion of B lymphocytes.
  • soluble SIL-6R is necessary to mediate these effects, because activated T lymphocytes usually do not have membrane- bound IL-6 receptors. Monocytes are more differentiated by IL-6 towards macrophages.
  • US 2007 036788 AA discloses peptides with IL-6 inhibitor activity obtained from an HL-cell line treated with cycloheximine.
  • US 2012 071549 AA refers to an anti-inflammatory use of macrolactin obtained from bacillus strains by inhibition of release of cytokines such IL-6.
  • US 2005 209201 A provides a method and composition which ameliorate inflammation, and suppress its onset, by allowing ATP receptor antagonists to act on ATP receptors of cells to block them and thereby inhibit the release of inflammatory cytokines, particularly interleukin-6 (IL-6) and/or interleukin-8 (IL-8), by the cells.
  • IL-6 interleukin-6
  • IL-8 interleukin-8
  • US 6,291,487 BA is related certain thiazole derivatives which are capable of inhibiting cytokine production.
  • JP 2009 203167 A deals with the provision of a cytokine production inhibi- tor that inhibits inflammation while retaining beneficial actions of IL-la or TNFa for cells by selectively controlling IL-6 and IL-8 with respect to inhibition or alleviation of skin inflamma- tion caused by external stimulation including irradiation with ultraviolet rays, and a cosmetic composition containing the same.
  • the cytokine production inhibitor comprises sericin and inhibits formation of interleukin-6 and/or interleukin-8.
  • interleukin-6 represents a chemokine that intervenes in many ways in biochemical processes of the body and is particularly associated with inflammatory processes.
  • IL-6 a number of substances are known from the state of the art that reduce or suppress the expression of IL-6, these are exclusively substances that are complex and expensive to produce.
  • the task of the present invention has therefore been to provide further drugs which reduce the expression of IL-6 while at the same time being more readily availa- ble. Particular consideration should be given to substances which are already known for cos- metic applications.
  • a first object of the present invention refers to agonists of olfactory receptors, par- ticularly OR2AT4, showing
  • Olfactory receptors also known as odorant receptors
  • ORs are expressed in the cell membranes of olfactory receptor neurons and are responsible for the detection of air-borne odour molecules that enter the nasal cavity.
  • Activated olfactory receptors trigger nerve im- pulses which transmit information about odour to the brain.
  • These receptors are members of the class A rhodopsin-like family of G protein-coupled receptors (GPCRs).
  • GPCRs G protein-coupled receptors
  • the olfactory re- ceptors form a multigene family consisting of around 800 genes in humans. Particularly, the human nose has roughly 400 types of scent receptors that can detect at least 1 trillion different odors.
  • olfactory receptors display affinity for a range of odour molecules, and conversely a single odorant molecule may bind to a number of ol- factory receptors with varying affinities, which depend on physio-chemical properties of mol- ecules like their molecular volumes. Once the odorant has bound to the odour receptor, the receptor undergoes structural changes and it binds and activates the olfactory-type G pro- tein on the inside of the olfactory receptor neuron.
  • the G protein (Golf and/or G s ) in turn acti- vates the lyase - adenylate cyclase - which converts ATP into cyclic AMP (cAMP).
  • the cAMP opens cyclic nucleotide-gated ion channels which allow calcium and sodium ions to enter into the cell, depolarizing the olfactory receptor neuron and beginning an action poten- tial which carries the information to the brain.
  • OR is the root name (Olfactory Receptor superfamily)
  • n - an integer representing a family (e.g., 1-56) whose members have greater than 40% sequence identity
  • OR1A1 is the first isoform of subfamily A of olfactory receptor family.
  • An overview on olfactory receptors can be found in the following paper submitted by Gaillard I, Rouquier S, Giorgi: D “Olfactory receptors ". Cellular and Molecular Life Sciences. 61 (4): 456-69 (2004).
  • the evaporation rate is the rate at which a material will vaporize (evaporate, change from liquid to vapor) compared to the rate of vaporization of butyl acetate, which is set as 1. This quantity is a ratio, therefore it is unitless.

Description

Medicament for fighting inflammatory conditions of human skin (II)
AREA OF INVENTION
[0001] The present invention refers to the area of therapeutical agents and relates to a me- dicament for treating inflammatory conditions of human skin associated with an increased expression of IL-6.
BACKGROUND OF THE INVENTION
[0002] Interleukin-6 (short: IL-6, synonymous older names: Interferon-β2 (IFNB2), B-cell stim- ulating factor, B-cell differentiation factor, liver cell stimulating factor) belongs to the inter- leukins (or more comprehensively to the cytokines), which regulate the inflammatory reaction of the organism. IL-6, by the nature of its complex regulation and functions in the orchestra of other cytokines and cells, plays a key role in the transition from mechanisms of innate im- munity to mechanisms of acquired immunity within the inflammatory process.
[0003] In humans, IL-6 is encoded on chromosome 7 gene locus p21. The gene segment contains 5 exons. An inactive precursor protein consisting of 212 amino acids is transcribed. Transcription is induced by the transcription factors NF-kB, NF-AT, HSF1 and HSF2, among others. The 184 amino acids long active interleukin-6 is cleaved from the precursor protein. Interleukin-6 can bind to two receptor types as an activating ligand:
(i) to a membrane-bound IL-6 receptor (IL-6R), which only occurs on liver cells and leuko- cytes and transmits its signals with the help of the receptor-associated, signal- transmitting glycoprotein gpl30,
(ii) to a soluble IL-6 receptor (sIL-6R). The resulting IL-6/sIL-6R complex binds to and acti- vates the glycoprotein gpl30, which is the only glycoprotein found in the membranes of many cell types. This second process is called "IL-6-trans-signaling".
[0004] When the glycoprotein Gpl30 is activated by an IL-6/sIL-6R complex or when an IL- 6R (to which gpl30 is associated) is activated by IL-6, a Janus-activated kinase at its domain inside the cell is phosphorylated at a tyrosine, thereby activating the JAK-STAT signalling pathway and the MAP kinase pathway, which then leads to the transcription of specific target genes in the cell nucleus. However, interleukin-6 is usually considered an activator of acute phase proteins and a lymphocyte stimulating factor.
[0005] During an acute inflammatory episode, neutrophil granulocytes are recruited, infiltrate the site of inflammation, do their job, die rapidly and are replaced by a longer lasting popula- tion of more specific inflammatory cells such as lymphocytes and mononuclear cells. Here, interleukin-6 efficacy plays an important role: With the degree of infiltration by neutrophil granulocytes, the local concentration of SIL-6R increases, which triggers IL-6-trans-signaling in the surrounding tissue. This in turn leads to a limitation of the accumulation of neutrophilic granulocytes in the inflamed tissue via different pathways. At the same time, IL-6-trans- signaling attracts CD3+-T lymphocytes, which mark the transition of the innate immune re- sponse to a learned immune response. Furthermore, IL-6 is involved in the regulation of leu- kocyte apoptosis with pro-apoptotic and anti-apoptotic agents (of which it is not yet known how they are balanced). IL-6 has an anti-apoptotic effect on resting and activated T lympho- cytes, and regulates their differentiation, proliferation, polarization and immunoglobulin G secretion of B lymphocytes. Especially in activated T lymphocytes, soluble SIL-6R is necessary to mediate these effects, because activated T lymphocytes usually do not have membrane- bound IL-6 receptors. Monocytes are more differentiated by IL-6 towards macrophages.

[0011] JP 2009 203167 A (KRACIE) deals with the provision of a cytokine production inhibi- tor that inhibits inflammation while retaining beneficial actions of IL-la or TNFa for cells by selectively controlling IL-6 and IL-8 with respect to inhibition or alleviation of skin inflamma- tion caused by external stimulation including irradiation with ultraviolet rays, and a cosmetic composition containing the same. The cytokine production inhibitor comprises sericin and inhibits formation of interleukin-6 and/or interleukin-8. OBJECT OF THE INVENTION
[0012] As explained above, interleukin-6 represents a chemokine that intervenes in many ways in biochemical processes of the body and is particularly associated with inflammatory processes. Although a number of substances are known from the state of the art that reduce or suppress the expression of IL-6, these are exclusively substances that are complex and expensive to produce. The task of the present invention has therefore been to provide further drugs which reduce the expression of IL-6 while at the same time being more readily availa- ble. Particular consideration should be given to substances which are already known for cos- metic applications.
BRIEF DESCRIPTION OF THE INVENTION
[0013] A first object of the present invention refers to agonists of olfactory receptors, par- ticularly OR2AT4, showing
(i) a molecular weight ranging from about 50 to about 300 Dalton and
(ii) an evaporation rate versus butyl acetate of about 3 to about 15 for use as a medicament for fighting inflammatory conditions of human skin associated with increased expression of interleukin-6.
[0014] Surprisi ngly it has been found that small and volatile molecules capable of activating olfactory receptors, in particular OR2AT4, which is found in human skin, are showing a signifi- cant reduction in IL-lbeta-induced IL-6 release in primary human fibroblasts. This proves that the substances are capable of fighting inflammatory conditions particularly when applied topical to human skin and a patient in need of.
Olfactory receptors
[0015] Olfactory receptors (ORs), also known as odorant receptors, are expressed in the cell membranes of olfactory receptor neurons and are responsible for the detection of air-borne odour molecules that enter the nasal cavity. Activated olfactory receptors trigger nerve im- pulses which transmit information about odour to the brain. These receptors are members of the class A rhodopsin-like family of G protein-coupled receptors (GPCRs). The olfactory re- ceptors form a multigene family consisting of around 800 genes in humans. Particularly, the human nose has roughly 400 types of scent receptors that can detect at least 1 trillion different odors.
[0016] Rather than binding specific ligands, olfactory receptors display affinity for a range of odour molecules, and conversely a single odorant molecule may bind to a number of ol- factory receptors with varying affinities, which depend on physio-chemical properties of mol- ecules like their molecular volumes. Once the odorant has bound to the odour receptor, the receptor undergoes structural changes and it binds and activates the olfactory-type G pro- tein on the inside of the olfactory receptor neuron. The G protein (Golf and/or Gs) in turn acti- vates the lyase - adenylate cyclase - which converts ATP into cyclic AMP (cAMP). The cAMP opens cyclic nucleotide-gated ion channels which allow calcium and sodium ions to enter into the cell, depolarizing the olfactory receptor neuron and beginning an action poten- tial which carries the information to the brain.
[0017] A nomenclature system has been devised for the olfactory receptor family and is the basis for the official Human Genome Project (HUGO) symbols for the genes that encode these receptors. The names of individual olfactory receptor family members are in the format "ORnXm" where:
• OR is the root name (Olfactory Receptor superfamily)
• n - an integer representing a family (e.g., 1-56) whose members have greater than 40% sequence identity,
• X = a single letter (A, B, C, ...) denoting a subfamily (>60% sequence identity), and
• m = an integer representing an individual family member (isoform).
[0018] For example, OR1A1 is the first isoform of subfamily A of olfactory receptor family. An overview on olfactory receptors can be found in the following paper submitted by Gaillard I, Rouquier S, Giorgi: D "Olfactory receptors ". Cellular and Molecular Life Sciences. 61 (4): 456-69 (2004).
[0019] The evaporation rate is the rate at which a material will vaporize (evaporate, change from liquid to vapor) compared to the rate of vaporization of butyl acetate, which is set as 1. This quantity is a ratio, therefore it is unitless.

[0042] Tetraalkyl ammonium salts. Cationically active surfactants comprise the hydropho- bic high molecular group required for the surface activity in the cation by dissociation in aqueous solution. A group of important representatives of the cationic surfactants are the tetraalkyl ammonium salts of the general formula: (R1R2R3R4N+) X-. Here R1 stands for C1-C8 alk(en)yl, R2, R3 and R4, independently of each other, for alk(en)yl radicals having 1 to 22 car- bon atoms. X is a counter ion, preferably selected from the group of the halides, alkyl sulfates and alkyl carbonates. Cationic surfactants, in which the nitrogen group is substituted with two long acyl groups and two short alk(en)yl groups, are particularly preferred.
[0043] Esterquats. A further class of cationic surfactants particularly useful as co-surfactants for the present invention is represented by the so-called esterquats. Esterquats are generally understood to be quaternised fatty acid triethanolamine ester salts. These are known com- pounds which can be obtained by the relevant methods of preparative organic chemistry. Reference is made in this connection to International patent application WO 91/01295 Al, according to which triethanolamine is partly esterified with fatty acids in the presence of hy- pophosphorous acid, air is passed through the reaction mixture and the whole is then quaternised with dimethyl sulphate or ethylene oxide. In addition, German patent DE 4308794 Cl describes a process for the production of solid esterquats in which the quaterni- sation of triethanolamine esters is carried out in the presence of suitable dispersants, prefer- ably fatty alcohols.
[0044] Typical examples of esterquats suitable for use in accordance with the invention are products of which the acyl component derives from monocarboxylic acids corresponding to formula RCOOH in which RCO is an acyl group containing 6 to 10 carbon atoms, and the amine component is triethanolamine (TEA). Examples of such monocarboxylic acids are ca- proic acid, caprylic acid, capric acid and technical mixtures thereof such as, for example, so- called head-fractionated fatty acid. Esterquats of which the acyl component derives from monocarboxylic acids containing 8 to 10 carbon atoms, are preferably used. Other esterquats are those of which the acyl component derives from dicarboxylic acids like malonic acid, suc- cinic acid, maleic acid, fumaric acid, glutaric acid, sorbic acid, pimelic acid, azelaic acid, sebac- ic acid and/or dodecanedioic acid, but preferably adipic acid. Overall, esterquats of which the acyl component derives from mixtures of monocarboxylic acids containing 6 to 22 carbon atoms, and adipic acid are preferably used. The molar ratio of mono and dicarboxylic acids in the final esterquat may be in the range from 1:99 to 99:1 and is preferably in the range from 50:50 to 90:10 and more particularly in the range from 70:30 to 80:20. Besides the quater- nised fatty acid triethanolamine ester salts, other suitable esterquats are quaternized ester salts of mono-/dicarboxylic acid mixtures with diethanolalkyamines or 1,2-dihydroxypropyl dialkylamines. The esterquats may be obtained both from fatty acids and from the corre- sponding triglycerides in admixture with the corresponding dicarboxylic acids. One such pro- cess, which is intended to be representative of the relevant prior art, is proposed in European patent EP 0750606 Bl. To produce the quaternised esters, the mixtures of mono- and dicar- boxylic acids and the triethanolamine - based on the available carboxyl functions - may be used in a molar ratio of 1.1:1 to 3:1. With the performance properties of the esterquats in mind, a ratio of 1.2:1 to 2.2:1 and preferably 1.5:1 to 1.9:1 has proved to be particularly ad- vantageous. The preferred esterquats are technical mixtures of mono-, di- and triesters with an average degree of esterification of 1.5 to 1.9. Superfatting agents and consistency factors
[0045] Superfatting agents may be selected from such substances as, for example, lanolin and lecithin and also polyethoxylated or acylated lanolin and lecithin derivatives, polyol fatty acid esters, monoglycerides and fatty acid alkanolamides, the fatty acid alkanolamides also serving as foam stabilizers.
[0046] The consistency factors mainly used are fatty alcohols or hydroxyfatty alcohols con- taining 12 to 22 and preferably 16 to 18 carbon atoms and also partial glycerides, fatty acids or hydroxyfatty acids. A combination of these substances with alkyl oligoglucosides and/or fatty acid N-methyl glucamides of the same chain length and/or polyglycerol poly-12- hydroxystearates is preferably used.

A detailed overview of suitable volatile silicones can be found in Todd et al. in Cosm. Toil. 91, 27 (1976).
Waxes and stabilizers
[0053] Besides natural oils used, waxes may also be present in the preparations, more espe- cially natural waxes such as, for example, candelilla wax, carnauba wax, Japan wax, espar- tograss wax, cork wax, guaruma wax, rice oil wax, sugar cane wax, ouricury wax, montan wax, beeswax, shellac wax, spermaceti, lanolin (wool wax), uropygial fat, ceresine, ozocerite (earth wax), petrolatum, paraffin waxes and microwaxes; chemically modified waxes (hard waxes) such as, for example, montan ester waxes, sasol waxes, hydrogenated jojoba waxes and syn- thetic waxes such as, for example, polyalkylene waxes and polyethylene glycol waxes.
[0054] Metal salts of fatty acids such as, for example, magnesium, aluminium and/or zinc stearate or ricinoleate may be used as stabilizers.
Primary sun protection filters
[0055] Primary sun protection factors in the context of the invention are, for example, organ- ic substances (light filters) which are liquid or crystalline at room temperature and which are capable of absorbing ultraviolet radiation and of releasing the energy absorbed in the form of longer-wave radiation, for example heat.
[0056] The formulations according to the invention advantageously contain at least one UV- A filter and/or at least one UV-B filter and/or a broadband filter and/or at least one inorganic pigment. Formulations according to the invention preferably contain at least one UV-B filter or a broadband filter, more particularly preferably at least one UV-A filter and at least one UV-B filter.
[0057] Preferred cosmetic compositions, preferably topical formulations according to the present invention comprise one, two, three or more sun protection factors selected from the group consistiung of 4-aminobenzoic acid and derivatives, salicylic acid derivatives, benzo- phenone derivatives, dibenzoylmethane derivatives, diphenyl acrylates, 3-imidazol-4-yl acryl- ic acid and esters thereof, benzofuran derivatives, benzylidene malonate derivatives, poly- meric UV absorbers containing one or more organosilicon radicals, cinnamic acid derivatives, camphor derivatives, trianilino-s-triazine derivatives, 2-hydroxyphenylbenzotriazole deriva- tives, phenylbenzimidazole sulfonic acid derivatives and salts thereof, anthranilic acid menthyl esters, benzotriazole derivativesand indole derivatives.
[0058] In addition, it is advantageous to combine compounds of formula (I) with active in- gredients which penetrate into the skin and protect the skin cells from inside against sun- light-induced damage and reduce the level of cutaneous matrix metalloproteases. Preferred respective ingredients, so called arylhydrocarbon receptor antagonists, are described in WO 2007/128723, incorporated herein by reference. Preferred is 2-benzylidene-5,6-dimethoxy- 3,3-dimethylindan-l-one.
