<1> 光合成細菌を含む、がんの治療薬又は診断薬。
<2> 光合成細菌が、バクテリオクロロフィルを有する光合成細菌である、<1>に記載のがんの治療薬又は診断薬。
<3> 光合成細菌が、紅色光合成細菌、又は緑色光合成細菌である、<1>又は<2>に記載のがんの治療薬又は診断薬。
<4> 光合成細菌が、ロドシュードモナス属(Rhodopseudomonas)細菌、ブラストクロリス属(Blastochloris)細菌、アフィフェラ属(Afifella)細菌、ロドバクター属(Rhodobacter)細菌、パラロドスピリラム属(Pararhodospirillum)細菌、ロドミクロビウム属(Rhodomicrobium)細菌、ロドブラム属(Rhodovulum)細菌、又はマリクロマチウム属(Marichromatium)細菌である、<1>から<3>の何れか一項に記載のがんの治療薬又は診断薬。
<5> 光照射と組み合わせて使用する、<1>から<4>の何れかに記載のがんの治療薬又は診断薬。
<6> 光照射が近赤外光の照射である、請求項5に記載のがんの治療薬又は診断薬。
低酸素、免疫回避、化学走性に由来する明確な機構に従い、腫傷に対する高い選択性の発現が可能である。
副作用の強い抗がん剤は利用せず、使用する細菌は低毒性であるので、副作用や毒性が低い。
使用する細菌は無限に自己複製することが可能であり、製造費用は安い。
酸素を使う光線力学モードのみならず、酸素を必要としない温熱モードも発現可能であり、さらに、光を使用せずに細菌を投与するだけでがんを縮退させる効果がある。
細菌は固形がんに普遍的な微小環境に対して高選択的に集積、生育、増殖が可能である。
細菌は腫瘍深部に集積・生育・増殖が可能である。
遺伝子組み換えの必要がなく、腫傷に対する高選択性を発現できる。
本発明で使用する光合成細菌としては、バクテリオクロロフィルを有する光合成細菌が好ましい。バクテリオクロロフィルとしては、バクテリオクロロフィルa、バクテリオクロロフィルb、バクテリオクロロフィルc、バクテリオクロロフィルd、バクテリオクロロフィルf、又はバクテリオクロロフィルgなどが挙げられ、これらのうちの一以上を有する光合成細菌を使用することができる。一例としては、バクテリオクロロフィルa又はバクテリオクロロフィルbを有する光合成細菌を挙げることができる。より具体的には、紅色光合成細菌、又は緑色光合成細菌を使用することができる。図1は、近赤外光で駆動する紅色光合成細菌の概念図を示す。図2は、バクテリアクロロフィル(BChl a)の化学構造を示す。
上記の中でも、ロドシュードモナス属(Rhodopseudomonas)細菌、ブラストクロリス属(Blastochloris)細菌、アフィフェラ属(Afifella)細菌、ロドバクター属(Rhodobacter)細菌、パラロドスピリラム属(Pararhodospirillum)細菌、ロドミクロビウム属(Rhodomicrobium)細菌、ロドブラム属(Rhodovulum)細菌、又はマリクロマチウム属(Marichromatium)細菌が好ましい。
緑色光合成細菌の具体例としては、Thermochromatium tepidum、及びChlorobaculum tepidumなどが挙げられる。
上記の中でも、Rhodopseudomonas Palustris、Blastochloris viridis、Pararhodospirillum oryzae、Pararhodospirillum sulfurexigens、Rhodomicrobium udaipurense、Rhodomicrobium vannielii、Rhodovulum sulfidophilum、Afifella marina、Rhodobacter sphaeroides、Marichromatium litoris、Rhodobacter capsulatus、又はBlastochloris sulfoviridisが特に好ましい。

上記以外にも、場合によっては国内の大学の研究室から分与を受けることが可能な細菌もある。
近赤外光の波長は特に限定されないが、好ましくは700nm~2000nmであり、700nm~1400nm、750nm~1200nm、750nm~1000nm、又は750nm~900nmでもよい。一例としては、808nmである。
光照射の時間は本発明の効果が得られる限り特に限定されないが、一般的には1分から30分、好ましくは1分から20分である。
光照射は1回だけ行ってもよいし、2回以上の複数回行ってもよい。
上記した投与量を、1回もしくは複数回にわたる分割用量(2、3、または4回の用量など)または単一の製剤で投与することができる。
<A1> 光合成細菌を対象に投与することを含む、がんの治療方法。
<A2> 光合成細菌を投与した対象に光を照射することをさらに含む、<A1>に記載のがんの治療方法。
<A3> 光合成細菌を対象に投与することを含む、がんの診断方法。
<B1> がんの治療又は診断において使用するための、光合成細菌。
<B2> 光照射と組み合わせて使用する、<B1>に記載の光合成細菌。
<C1> がんの治療薬又は診断薬の製造のための、光合成細菌の使用。
本研究で使用した細菌は、National Institute of Technology and Evaluation Biological Resource Center(NBRC)およびAmerican Type Culture Collection(ATCC)より取得した。
Rhodopseudomonas Palustris(NBRC16661)、Blastochloris viridis(NBRC 102659)、Pararhodospirillum oryzae(NBRC107573)、Pararhodospirillum sulfurexigens(NBRC104433)、Rhodomicrobium udaipurense(NBRC109057)、Rhodomicrobium vannielii(NBRC100050)、Rhodovulum sulfidophilum(ATCC35886)、Afifella marina(NBRC100434)、Rhodobacter sphaeroides(NBRC12203)、Marichromatium litoris(NBRC104939)、Rhodobacter capsulatus(NBRC16435)、Blastochloris sulfoviridis(NBRC103805)は、543 ATCC medium中で タングステンランプを照射しながら26-30℃の温度で嫌気培養した。Bifidobacterium bifidum(NBRC 100015)は、385NBRC medium中で 37℃の温度で嫌気培養した。
細菌分散液の吸光スペクトルは、室温でUV-Vis-NIR spectrophotometer(V-730 BIO;Jasco)を用いて測定した。細菌分散液の蛍光は、fluorescence spectrometers(FP-8600 NIR Spectrofluorometer; Jasco or Fluorolog-3; HORIBA Jobin Yvon)を用いて測定した。
R.Palustrisの蛍光スペクトル(励起波長:805nm)を図4に示す。また、励起波長と蛍光波長を変更した際の各細菌の蛍光スペクトルを図26及び図27に示す。
レーザー照射した細菌分散液の温度変化は以下の方法で調査した。細菌を分散させたPBS緩衝液(100μL)あるいは細菌を添加していないPBS緩衝液(100μL)に808nmの波長のファイバーカップル型の連続波レーザー(レーザースポット径, 約5mm;出力,1.2W,約61.1mW mm-2; CivilLaser)を照射した。レーザー照射時の溶液の温度変化は温度センサー(AD-5601A;A&D)を用いて測定した。
近赤外レーザー照射時[波長:808nm、出力:1.2W(ca.61.1mW/mm2)、照射時間:5min]の各細菌((a)Rhodopseudomonas Palustris,、(b)Blastochloris viridis、(c)Bifidobacterium bifidum)の分散液の温度変化を、図5に示す。
ROS分析は底辺が透明でメインボディが黒色の96-well plate(Thermo Fisher Scientific)と一重項酸素検出試薬singlet oxygen sensor green(SOSG)(Invitrogen)を用いた。R.Palustrisを分散させたPBS緩衝液(100μL,5×109CFUml-1)をSOSGを含むPBS緩衝液で希釈した。R.PalustrisとSOSGの当該系中の終濃度は、それぞれ1.3×109CFU ml-1、1μMであった。次いで、サンプルに808nmの波長の近赤外レーザーを出力1.2W(ca.61.1mWmm-2)、5分間照射した。R.Palustrisを含まないPBS緩衝液をコントロールとして用いた。ROS発生に伴う緑色蛍光はマイクロプレートリーダー(Infinite 200 PRO M Plex)(励起波長485nm、蛍光波長535 nm)を用いて測定した。
近赤外レーザー照射時[波長:808nm、出力:1.2Wca.61.1mW/mm2)、照射時間:5min]のR.Palustris分散液からの活性酸素種(ROS)一重項酸素の発生挙動を図6に示す。
マウス結腸がん細胞(Colon26)ならびにヒト正常二倍体線維芽細胞(MRC5)は、the Japanese Collection of Research Bioresources Cell Bankより入手した。ヒト肺胞基底上皮腺がん細胞(A549)、ヒト結腸腺がん(HT29)は、DS Pharma Biomedicalより購入した。マウスマクロファージ(RAW264.7)は、Riken Bio Resource Centerより得た。Colon26の培養は、10%fetal bovine serum、2-mM l-glutamine、1-mM sodium pyruvate、gentamycin、penicillin-streptomycin(100 IU ml-1)を含むRoswell Park Memorial Institute(RPMI)1640 Medium(Gibco)を使用した。他の細胞は、10%fetal bovine serum、2-mM l-glutamine、1-mM sodium pyruvate、gentamycin、penicillin-streptomycin(100 IU ml-1)、Hank’s balanced salt solution(Life Technologies)を含むDulbecco’s Modified Eagle’s Medium(DMEM)(Gibco)中で培養した。細胞は37℃、5%のCO2雰囲気下の加湿チャンバー中で培養した。
がん細胞生存性を評価するためにColon26、A549、HT29細胞(5×103 cells/well)を96-well plateに播種し一晩インキュベートした。細胞は各濃度(0.16、0.31、0.63、1.25×109CFU ml-1)のR.Palustrisを分散させた細胞培養培地(100μL)あるいはR.Palustrisを含まない細胞培養培地(100μL)で処理後、レーザー(波長808nm、出力1.2 W,~61.1mW mm-2)を5分間照射した。レーザー照射後、細胞を洗浄し、新鮮な培地中でインキュベートした。細胞生存性はCCK-8 kitを用いて、レーザー照射直後ならびにレーザー照射後24時間インキュベートしたサンプルを調査した。
RAW264.7細胞(2.5×105cells well-1)を24-well plateに播種し一晩インキュベートした。細胞はR.Palustrisを分散させた細胞培養培地(1×108 CFU)あるいはR.Palustrisを含まない細胞培養培地に曝露後、37℃、5%CO2の条件下で2時間培養した。細胞を洗浄し、新鮮な培地中で4時間インキュベートし、近赤外蛍光ミラーユニット(IRDYE800-33LP-A-U01;Semrock)と対物レンズ(×60 magnification, aperture 1.35;UPLSAPO60X,Olympus)を搭載する蛍光顕微鏡システム(IX73;Olympus)を用いて室温で観察した。
全ての動物実験は、北陸先端科学技術大学院大学の動物実験委員会の承認を得て実施した。4週齢のメスの野生型マウス(n=10;average weight=15g; BALB/cCrSlc)をJapan SLC,Incより購入し、一週間馴化させた。Colon26細胞分散液(1×106cells)(100μL)とculture medium/matrigel(Corning)から成る混合溶液(v/v,1:1)をマウス側面腹部皮下2ヵ所に投与し、結腸がんモデルマウスを作成した。約2週間後、固形がん(~400mm3)を形成させたマウスの尾静脈内にR.Palustris(1× 109CFU ml-1)あるいはPBS緩衝液を各200μL投与した。右側の固形がんにのみ2日に一度のペースで近赤外レーザー[波長808 nm、出力(713 mW, 36.3 mW mm-2)]を3分間照射した。IRサーモグラフィー(i7; FLIR, Nashua)を用いてレーザー照射時のマウスの腫瘍表面温度を測定した。

このとき、Vは固形がん体積、Lは固形がんの長さ、Wは固形がんの幅をそれぞれ示している。
Colon26を播種した結腸がんモデルマウス(female,8 weeks;n= 5;average weight=19g;average tumor size~ 400mm3;BALB/cCrSIc;Japan SLC)の尾静脈にR.Palustris(5×109CFU ml-1)、B.viridis(5×109CFU ml-1)、細菌培養培地をそれぞれ200μL投与した。1、24、48、144時間後に各組織と腫瘍を摘出し、切断後、重量を測定した。ペストルを使って4℃の温度下、PBS緩衝液中でホモジナイズ後、混合物を20分間、15℃、380rpm min-1の速度で振とうした。上澄をPBS緩衝液で10倍希釈し、各サンプル(100μL)を寒天培地上に播種し、7日間、嫌気培養した。形成した細菌コロニー数はマニュアルで測定した。
各処置後のマウスの写真(矢印はレーザーを照射した各腫瘍を示す)を図11に示す。
各種処置を施した34日後の摘出した腫瘍の写真を図12に示す。
各処置における34日間のマウス生存率を図13示す。
各種臓器ならびに腫瘍中のR.Palustris生菌数[R.Palustris(200μL、1×109CFU/mL)をColon26担がんモデルマウスに尾静脈投与した7日後に各種臓器ならびに腫瘍を取り出し、抽出液を寒天培地上、嫌気条件で培養]を図16に示す。
各種臓器ならびに腫瘍のR.Palustrisに由来する蛍光強度[R. Palustris(200μL、1×109 CFU/mL)をColon26担がんモデルマウスに尾静脈投与し、経時的に蛍光バイオイメージング解析し、蛍光強度を測定]を図17に示す。
各種臓器ならびに腫瘍のR.Palustrisの生菌数[R. Palustris(200μL、1×109 CFU/mL)をColon26担がんモデルマウスに尾静脈投与し、経時的に各種臓器ならびに腫瘍を取り出し、抽出液を寒天培地に塗布後、嫌気条件下で培養し、形成したコロニー数を測定]を図18に示す。
各種臓器ならびに腫瘍中のBlastochloris viridis生菌数[Blastochloris viridis(200μL、1×109CFU/mL)をColon26担がんモデルマウスに尾静脈投与した7日後に各種臓器ならびに腫瘍を取り出し、抽出液を寒天培地上、嫌気条件で培養]を図20に示す。
NIR-I蛍光を用いた細菌の生体内分布を測定するためにColon26から成るマウス結腸がんモデルマウス(female;8weeks;n=3;average weight=19g;average tumor size=430mm3;BALB/cCrSIc;Japan SLC)の尾静脈にR.Palustris(200μL, 1×109CFU)を含む細菌培養培地あるいは細菌を含まないPBS緩衝液を投与した。マウスならびに主要生体組織の近赤外蛍光像をVISQUETM InVivo Smart-LF(Vieworks)を用いて観察した。励起波長ならびに蛍光波長はそれぞれλex=740-790nm,λem=810-860nmである。また画像解析はCleVueTM software(Vieworks)を用いた。
Colon26担がんモデルマウスのIn vivo近赤外I領域(NIR-I)蛍光バイオイメージング(Ex:740nm-790nm、Em:810nm-860nm)[画像はR.Palustris(200μL、1×109CFU/mL)あるいはPBS(200μL)をマウスの尾静脈投与した5日後に撮影、矢印は腫瘍の位置を示す]を図14に示す。
PAイメージングは、multi spectral optoacoustic tomography (MSOT)inVision 256-TF system(SYS-MSOTiV256TF;iThera Medical)を用いてSummit Pharmaceuticals Internationalによって実施された。R. Palustris(20μL,1×108CFU)を分散させたPBS緩衝液をHT29から成るヒト結腸腺がんモデルマウス(female;10weeks;n=3;average weight=21g;average tumor size=100mm3;BALB/cSlc-nu/nu;Japan SLC)の固形がんに投与することでPA像を得た。
データ中の±は標準偏差、nは使用したサンプル数を示している。データの統計解析は、Student’s t-testを用いた。*、**、***は、それぞれ< 0.05、< 0.005、< 0.001のp値を示している。

- 光合成細菌が、紅色光合成細菌、又は緑色光合成細菌である、請求項1又は2に記載のがんの治療薬又は診断薬。
- 光合成細菌が、ロドシュードモナス属(Rhodopseudomonas)細菌、ブラストクロリス属(Blastochloris)細菌、アフィフェラ属(Afifella)細菌、ロドバクター属(Rhodobacter)細菌、パラロドスピリラム属(Pararhodospirillum)細菌、ロドミクロビウム属(Rhodomicrobium)細菌、ロドブラム属(Rhodovulum)細菌、又はマリクロマチウム属(Marichromatium)細菌である、請求項1から3の何れか一項に記載のがんの治療薬又は診断薬。
