Abstract
머신 러닝 기반 디지털 홀로그래피 장치 및 이의 동작 방법이 개시된다. 일 실시예에 따른 디지털 홀로그래피 방법은, 시료에 대한 2차원 정보를 포함하는 광학 영상을 획득하는 단계와, 상기 광학 영상에 기초하여 상기 시료에 대한 3차원 정보를 포함하는 홀로그램 영상을 생성하는 단계를 포함한다.
Description
실시예들은 머신 러닝 기반 디지털 홀로그래피 장치 및 이의 동작 방법에 관한 것이다.
백색광을 사용하는 광학 현미경(Bright-field microscopy)은 산업, 생물학, 의학 분야를 포함한 다양한 영역에서 널리 사용되는 영상 기술이다. 광학 영상(bright-field image)은 일반적으로 오염물질, 미생물, 생체 시료와 같은 미세 물체를 관찰하는데 사용된다. 그러나 광학 영상은 수 μm의 얇은 초점 심도(depth of focus) 내 시료의 2차원 빛의 세기 정보만 제공하기 때문에, 검출 및 진단 정확도가 보통 90% 이하로 높지 않고, 3차원 공간 형태로 존재하는 시료를 검출하기 어렵다.
결맞음성(coherent) 빛을 시료에 조사하면, 시료에 의해 회절된 물질파와 참조파의 간섭으로 홀로그램이 생성되며, 생성된 홀로그램에는 시료의 3D 영상정보가 2D 간섭무늬 형태로 담겨있다. 즉, 디지털 홀로그래픽 현미경(digital holography microscope(DHM)) 기법은 한 장의 홀로그램 영상으로부터 3D 공간 내에 존재하는 시료들의 3D 위치 및 형상 정보와 함께 광학 정보를 획득할 수 있는 영상기술이다. 이러한 DHM 기법은 별도의 라벨링 및 기계적 스캐닝 과정 없이 홀로그램으로부터 시료의 3D 정보를 비침습적으로 획득할 수 있기 때문에 생체 시료, 오염물질의 모니터링, 입자 및 세포의 3D 동적거동 분석 등 다양한 분야에서 응용되어 왔다.
홀로그램을 얻기 위해서 일반적으로 고가의 결맞음성이 좋은 레이저 광원, 고품질의 공간 필터, 거울, 렌즈 등 같은 값비싼 광학 부품들이 요구된다. 최근에는 DHM 기법에 딥러닝을 적용하여 홀로그램의 공간 해상도를 향상시키고, 수치적 재생 과정과 위상 정보 복원 알고리즘을 개선하여 생체 시료의 분류 및 모니터링 분야에 응용되고 있다. 그러나 근본적인 DHM 기법의 기술적 한계인 고가의 레이저 및 광학 부품을 대체하는 기술은 아직까지 제시된 적이 없다.
아래 실시예들은 머신 러닝을 이용하여 백색광 기반의 광학 영상을 홀로그램 영상으로 변환시켜 시료의 3차원 정보를 획득하는 기술을 제공할 수 있다.
다만, 기술적 과제는 상술한 기술적 과제들로 한정되는 것은 아니며, 또 다른 기술적 과제들이 존재할 수 있다.
일 실시예에 따른 디지털 홀로그래피 방법은, 시료에 대한 2차원 정보를 포함하는 광학 영상을 획득하는 단계와, 상기 광학 영상에 기초하여 상기 시료에 대한 3차원 정보를 포함하는 홀로그램 영상을 생성하는 단계를 포함한다.
상기 광학 영상은, 상기 시료에 백색광을 비추어 촬영한 영상일 수 있다.
상기 생성하는 단계는, 상기 광학 영상을 뉴럴 네트워크에 입력하여 상기 홀로그램 영상을 생성하는 단계를 포함할 수 있다.
상기 뉴럴 네트워크는, 동일한 시료에 백색광 및 단색광을 동시에 비추어 획득한 광학 영상 및 홀로그램 영상에 기초하여 학습된 뉴럴 네트워크일 수 있다.
상기 디지털 홀로그래피 방법은, 상기 홀로그램 영상을 깊이 방향으로 수치적으로 재생(numerical reconstruction)한 영상을 한 평면에 투영시켜 상기 시료의 2차원 위치 정보를 획득하는 단계를 포함할 수 있다.
상기 디지털 홀로그래피 방법은, 상기 홀로그램 영상을 깊이 방향으로 수치적으로 재생(numerical reconstruction)한 영상을 상기 시료의 중심을 기준으로 분할하는 단계와, 분할한 영상의 선명도에 기초하여 상기 시료의 초점 평면 영상 및 상기 시료의 깊이 정보를 획득하는 단계를 포함할 수 있다.
상기 디지털 홀로그래피 방법은, 깊이 방향으로 상기 홀로그램 영상의 빛 강도 분포를 축적하여 빛의 산란 패턴을 획득하는 단계와, 상기 빛의 산란 패턴에 기초하여 상기 시료의 실초점 거리 및 허초점 거리 중 적어도 하나를 추출하는 단계를 포함할 수 있다.
일 실시예에 따른 디지털 홀로그래피 장치는, 인스트럭션들을 포함하는 메모리와, 상기 인스트럭션들을 실행하기 위한 프로세서를 포함하고, 상기 프로세서에 의해 상기 인스트럭션들이 실행될 때, 상기 프로세서는, 시료에 대한 2차원 정보를 포함하는 광학 영상에 기초하여 상기 시료에 대한 3차원 정보를 포함하는 홀로그램 영상을 생성한다.
상기 광학 영상은, 백색광을 상기 시료에 비추어 촬영한 광학 영상일 수 있다.
상기 프로세서는, 동일한 시료에 백색광 및 단색광을 동시에 비추어 획득한 광학 영상 및 홀로그램 영상에 기초하여 학습할 수 있다.
상기 프로세서는, 상기 홀로그램 영상을 깊이 방향으로 수치적으로 재생(numerical reconstruction)한 영상을 한 평면에 투영시켜 상기 시료의 2차원 위치 정보를 획득할 수 있다.

일 실시예에 따른 디지털 홀로그래피 장치의 학습 방법은, 동일한 시료에 백색광 및 단색광을 동시에 비추어 획득한 광학 영상 및 제1 홀로그램 영상을 획득하는 단계와, 제1 뉴럴 네트워크를 통해 상기 광학 영상에 기초하여 제2 홀로그램 영상을 생성하는 단계와, 제2 뉴럴 네트워크를 통해 상기 제1 홀로그램 영상 및 상기 제2 홀로그램 영상의 진위를 평가하는 단계와, 상기 제1 홀로그램 영상, 상기 제2 홀로그램 영상 및 진위 평가 결과에 기초한 손실 함수를 최적화하는 단계를 포함한다.
상기 손실 함수는, 상기 제1 뉴럴 네트워크에 대한 제1 손실 함수 및 상기 제2 뉴럴 네트워크에 대한 제2 손실 함수를 포함하고, 상기 최적화하는 단계는, 상기 제1 손실 함수 및 제2 손실 함수가 각각 최소 값을 가지도록 상기 제1 뉴럴 네트워크 및 제2 뉴럴 네트워크의 가중치를 조절하는 단계를 포함할 수 있다.

디지털 홀로그래피 장치(100)는 시료에 대한 불충분한 정보를 제공하는 종래의 광학 현미경과 정교한 평행광이 필요한 디지털 홀로그래피 현미경(digital holography microscopy(DHM))의 기술적 한계를 극복하기 위하여 머신 러닝을 이용할 수 있다.
디지털 홀로그래피 장치(100)는 백색광을 이용하여 시료의 3차원 정보를 획득하고, 3차원 정보에 기초하여 시료를 검출할 수 있다. 예를 들어, 디지털 홀로그래피 장치(100)는 시료에 백색광을 비추어 촬영한 광학 영상(bright-light(BF) image)에 기초하여 시료의 3차원 정보를 획득할 수 있다. 이때 백색광은 할로겐 램프(halogen lamps), LED 램프, 스마트폰 플래시 등을 포함할 수 있다.

디지털 홀로그래피 장치(100)는 생성한 홀로그램 영상으로부터 시료의 3차원 정보를 추출할 수 있다. 예를 들어, 디지털 홀로그래피 장치(100)는 빛의 확산 공식을 적용하여 여러 깊이에서 홀로그램 영상을 수치적으로 재생(numerical reconstruction)하여 시료의 3차원 정보를 획득할 수 있다.
디지털 홀로그래피 장치(100)는 뉴럴 네트워크(neural network)을 이용하여 특정 깊이에 위치한 시료의 초점이 맞지 않는(defocused) 광학 영상을 이와 동등한 홀로그램 영상으로 변환시켜, 값비싼 광학 부품을 사용하지 않고 백색광을 이용하여 시료의 3D 정보를 획득하고, 이를 시료 검출에 활용할 수 있다.
뉴럴 네트워크는 심층뉴럴 네트워크(Deep Neural Network)를 포함할 수 있다. 뉴럴 네트워크는 CNN(Convolutional Neural Network), RNN(Recurrent Neural Network), 퍼셉트론(perceptron), FF(Feed Forward), RBF(Radial Basis Network), DFF(Deep Feed Forward), LSTM(Long Short Term Memory), GRU(Gated Recurrent Unit), AE(Auto Encoder), VAE(Variational Auto Encoder), DAE(Denoising Auto Encoder), SAE(Sparse Auto Encoder), MC(Markov Chain), HN(Hopfield Network), BM(Boltzmann Machine), RBM(Restricted Boltzmann Machine), DBN(Depp Belief Network), DCN(Deep Convolutional Network), DN(Deconvolutional Network), DCIGN(Deep Convolutional Inverse Graphics Network), GAN(Generative Adversarial Network), LSM(Liquid State Machine), ELM(Extreme Learning Machine), ESN(Echo State Network), DRN(Deep Residual Network), DNC(Differentiable Neural Computer), NTM(Neural Turning Machine), CN(Capsule Network), KN(Kohonen Network) 및 AN(Attention Network)를 포함할 수 있다.

프로세서(150)는 메모리(110)에 저장된 데이터를 처리할 수 있다. 프로세서(150)는 메모리(110)에 저장된 컴퓨터로 읽을 수 있는 코드(예를 들어, 소프트웨어) 및 프로세서(150)에 의해 유발된 인스트럭션(instruction)들을 실행할 수 있다.
프로세서(150)는 목적하는 동작들(desired operations)을 실행시키기 위한 물리적인 구조를 갖는 회로를 가지는 하드웨어로 구현된 데이터 처리 장치일 수 있다. 예를 들어, 목적하는 동작들은 프로그램에 포함된 코드(code) 또는 인스트럭션들(instructions)을 포함할 수 있다.
예를 들어, 하드웨어로 구현된 데이터 처리 장치는 마이크로프로세서(microprocessor), 중앙 처리 장치(central processing unit), 프로세서 코어(processor core), 멀티-코어 프로세서(multi-core processor), 멀티프로세서(multiprocessor), ASIC(Application-Specific Integrated Circuit), FPGA(Field Programmable Gate Array)를 포함할 수 있다.
도 2는 도 1에 도시된 디지털 홀로그래피 장치를 학습시키기 위한 학습 데이터를 생성하는 하이브리드 영상 장치를 나타내는 도면이고, 도 3a 및 도 3b는 도 2에 도시된 하이브리드 영상 장치가 생성한 학습 데이터를 나타내는 도면이다.
하이브리드 영상 장치(200)는 디지털 홀로그래피 장치(100)의 학습에 이용되는 학습 데이터를 생성할 수 있다. 예를 들어, 하이브리드 영상 장치(200)는 시료의 광학 영상 및 광학 영상에 대응되는 홀로그램 영상을 획득할 수 있다.
하이브리드 영상 장치(200)는 제1 카메라(210), 제2 카메라(220), 이색성 거울(dichroic mirror)(230), 대물 렌즈(240) 및 미세 채널(250)을 포함할 수 있다.
하이브리드 영상 장치(200)는 미세 채널(250)을 통과하는 시료에 백색광 및 단색광을 동시에 비출 수 있다. 이때, 단색광은 공간 필터(spatial filter)를 거친 레이저 평행광일 수 있다.
미세 채널(250)은 주입 및 분리 부분을 가지는 복수의 미세 채널로 구성될 수 있다. 미세 채널(250)을 통해 시료가 흘러 지나갈 수 있으며, 백색광 및 단색광이 미세 채널(250)을 흘러지나는 시료를 비출 수 있다.
대물 렌즈(240)는 미세 채널(250)에 백색광 및 단색광을 비추어 획득한 광학 영상 및 홀로그램 영상을 확대할 수 있다. 대물 렌즈(240)는 튜브 렌즈(tube lens)를 포함할 수 있다.
이색성 거울(230)은 특정 파장대의 빛은 반사하고 나머지 빛들은 통과시킬 수 있다. 즉, 이색성 거울(230)은 빛의 파장에 따라 빛을 투과 또는 반사시킬 수 있다. 이색성 거울(230)은 단색광을 제2 카메라(220) 방향으로 반사시키고, 백색광을 제1 카메라(210) 방향으로 통과시킬 수 있다.
제1 카메라(210)는 백색광에 의한 광학 영상을 획득할 수 있다. 예를 들어, 제1 카메라(210)는 도 3a에 도시된 것과 같은 시료의 광학 영상을 획득할 수 있다.
제2 카메라(220)는 단색광에 의한 홀로그램 영상을 획득할 수 있다. 예를 들어, 제2 카메라(220)는 도 3b에 도시된 것과 같은 시료의 홀로그램 영상을 획득할 수 있다.
제1 카메라(210) 및 제2 카메라(220)는 CCD(charge coupled device) 카메라 또는 CMOS(complementary metal oxide semiconductor) 카메라일 수 있다.
하이브리드 영상 장치(200)가 획득한 시료의 광학 영상 및 홀로그램 영상은 디지털 홀로그래피 장치(100)의 학습 데이터로 이용될 수 있다. 디지털 홀로그래피 장치(100)는 동일한 시료에 대한 광학 영상 및 홀로그램 영상을 학습할 수 있다.
도 4는 도 1에 도시된 디지털 홀로그래피 장치가 포함하는 네트워크 구조를 나타내는 도면이고, 도 5는 도 1에 도시된 디지털 홀로그래피 장치가 생성한 홀로그램 영상을 나타내는 도면이다.
디지털 홀로그래피 장치(100)는 생성적 적대 신경망(generative adversarial network(GAN))을 포함할 수 있다. 예를 들어, 디지털 홀로그래피 장치(100)는 GAN(400)을 이용하여 광학 영상을 홀로그램 영상으로 변환할 수 있다.
GAN(400)은 하이브리드 영상 장치(200)에 의해 생성된 학습 데이터에 기초하여 학습할 수 있다. 백색광을 이용하여 획득한 광학 영상의 경우, 대물 렌즈의 배율에 따른 초점 심도로 인해 시료의 깊이 좌표가 증가할수록 신호의 세기가 약해지고 흐릿해질 수 있다. 결맞음성 빛을 비추어 시료에 의해 회절된 물질파와 시료를 통과하지 않은 참조파의 간섭현상으로 형성된 홀로그램 영상의 경우, 시료의 깊이 좌표가 증가할수록 간섭 무늬 간격이 증가한다. GAN(400)은 깊이에 따른 시료의 광학 영상 및 광학 영상에 대응하는 홀로그램 영상 사이의 관계를 통계적으로 학습할 수 있다.
GAN(400)은 제1 뉴럴 네트워크(410) 및 제2 뉴럴 네트워크(430)를 포함할 수 있다. 예를 들어, 제1 뉴럴 네트워크(410)는 생성 모델(generator)(410)일 수 있고, 제2 뉴럴 네트워크(430)는 분류 모델(discriminator)(430)일 수 있다.
생성 모델(410)은 입력 영상에 기초하여 새로운 영상을 생성할 수 있다. 예를 들어, 생성 모델(410)은 학습 영상 중 광학 영상에 기초하여 광학 영상에 대응되는 홀로그램 영상을 생성할 수 있다. 생성 모델(410)은 광학 영상 내에 존재하는 간섭 무늬와 같은 다층 구조를 학습하여, 홀로그램 영상을 생성할 수 있는 CNN(convolutional neural network)를 변형한 네트워크 구조를 가질 수 있다.
도 4에서 특징 맵(feature map) 또는 뉴런(neuron)의 수는 생성 모델(410) 및 판별 모델(430)의 왼쪽에 표시된 값일 수 있다. 특징 맵의 크기는 생성 모델(410) 및 판별 모델(430)의 오른쪽에 표시된 값일 수 있다. 서로 다른 색상의 화살표는 각각의 유형의 연결을 나타낼 수 있다. 색상이 다른 두 블록의 병합은 특징 맵의 연결을 나타낼 수 있다.
분류 모델(430)은 생성 모델(410)이 생성한 홀로그램 영상 및/또는 학습 영상을 획득한 홀로그램 영상의 진위 여부를 평가할 수 있다. 예를 들어, 분류 모델(430)은 실제 홀로그램 영상과 생성된 홀로그램 영상의 진위 여부를 판단하여, 실제 원본인 홀로그램 영상일 확률이 높다고 판단할 경우 1에 가까운 값을 출력하고, 생성된 홀로그램 영상일 확률이 높다고 판단하는 경우 0에 가까운 값을 출력하는 CNN일 수 있다.
GAN(400)은 학습 과정에서 생성 모델(410) 및 분류 모델(430)의 서로 반대되는 손실 함수(loss function)의 최적화를 시도할 수 있다. 예를 들어, 생성 모델(410)은 분류 모델(430)이 실제 홀로그램 영상과 생성된 홀로그램 영상을 분류할 수 없도록 학습하고, 분류 모델(430)은 분류에 성공할 확률을 높일 수 있도록 서로 적대적으로 학습할 수 있다.

디지털 홀로그래피 장치(100)는 여러 깊이에서의 빛 강도 분포를 깊이 방향으로 축적하여 빛의 산란 패턴을 획득할 수 있다. 빛 산란 패턴(light scattering)은 빛이 시료를 지나면서 어떻게 발산하고 수렴하는지를 나타내는데, 빛의 산란 패턴은 시료의 형상과 광학적인 특성에 따라 달라질 수 있다.

- 제1항에 있어서,상기 생성하는 단계는,상기 광학 영상을 동일한 시료에 백색광 및 단색광을 동시에 비추어 획득한 광학 영상 및 홀로그램 영상에 기초하여 학습된 뉴럴 네트워크에 입력하여 상기 홀로그램 영상을 생성하는 단계를 포함하는, 디지털 홀로그래피 방법.
- 제1항에 있어서,상기 홀로그램 영상을 깊이 방향으로 수치적으로 재생(numerical reconstruction)하여 상기 시료에 대한 3차원 정보를 획득하는 단계를 더 포함하는, 디지털 홀로그래피 방법.
- 제1항에 있어서,깊이 방향으로 상기 홀로그램 영상의 빛 강도 분포를 축적하여 빛의 산란 패턴을 획득하는 단계; 및상기 빛의 산란 패턴에 기초하여 상기 시료의 실초점 거리 및 허초점 거리 중 적어도 하나를 추출하는 단계를 더 포함하는, 디지털 홀로그래피 방법.
- 인스트럭션들을 포함하는 메모리; 및상기 인스트럭션들을 실행하기 위한 프로세서를 포함하고,상기 프로세서에 의해 상기 인스트럭션들이 실행될 때, 상기 프로세서는,시료에 대한 2차원 정보를 포함하는 광학 영상에 기초하여 상기 시료에 대한 3차원 정보를 포함하는 홀로그램 영상을 생성하는, 디지털 홀로그래피 장치.
- 제6항에 있어서,상기 광학 영상은,백색광을 상기 시료에 비추어 촬영한 광학 영상인, 디지털 홀로그래피 장치.
- 제6항에 있어서,상기 프로세서는,동일한 시료에 백색광 및 단색광을 동시에 비추어 획득한 광학 영상 및 홀로그램 영상에 기초하여 학습하는, 디지털 홀로그래피 장치.
- 제6항에 있어서,상기 프로세서는,상기 홀로그램 영상을 깊이 방향으로 수치적으로 재생(numerical reconstruction)하여 상기 시료에 대한 3차원 정보를 획득하는, 디지털 홀로그래피 장치.
- 제6항에 있어서,상기 프로세서는,깊이 방향으로 상기 홀로그램 영상의 빛 강도 분포를 축적하여 빛의 산란 패턴을 획득하고, 상기 빛의 산란 패턴에 기초하여 상기 시료의 실초점 거리 및 허초점 거리 중 적어도 하나를 추출하는, 디지털 홀로그래피 장치.
