Description

2抗原を認識するiPS細胞由来キメラ抗原受容体発現T細胞療法
 本発明は、新たなキメラ抗原受容体発現T細胞(CAR-T細胞)及びこれを用いる医薬に関する。
 キメラ抗原受容体発現T細胞(CAR-T細胞)は、キメラ抗原受容体を導入したT細胞であり、画期的ながん治療薬として注目されている。現在医薬品承認されているCAR-T細胞療法は、がん患者末梢血より誘導するT細胞にキメラ抗原受容体を導入してCAR-T細胞を作製し、患者に投与される自家CAR-T細胞療法であり、極めて高額であるという問題がある。一方で臨床試験の段階であるが、他家CAR-T細胞療法も開発されており、作製コストの軽減が利点であるが、免疫拒絶による抗腫瘍効果減弱の問題がある。
 一方、抗原特異的細胞傷害性T細胞(CTL)からT-iPS細胞を樹立し、抗原特異性を維持したまま再び、機能的に若返ったCTLに分化誘導する技術が開発されている(特許文献1、非特許文献1)。

 しかしながら、CTL-iPS細胞又はCAR-T細胞療法の抗がん効果は抗原によっては、あるいはがん種によっては未だ十分でなく、更に効率よく増殖してより優れた抗腫瘍効果を奏し、生体内に長期生存し得る免疫細胞療法が望まれている。
 そこで本発明者は、抗原特異的細胞傷害性T細胞(CTL)を原料とし、これをiPS細胞とし、次いでキメラ抗原受容体(CAR)を導入して分化させれば、特にiPS細胞由来CTLとCARによる2つの抗原を認識することで、抗腫瘍効果が相乗的に増強され、かつ生体内に長期間生存するiPS細胞が得られることを見出し、本発明を完成した。
 すなわち、本発明は、次の〔1〕~〔8〕を提供するものである。
〔1〕キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞。
〔2〕抗原特異的細胞傷害性T細胞の抗原が、ウイルス抗原又は腫瘍抗原である〔1〕記載のiPS細胞。
〔3〕キメラ抗原受容体が、腫瘍表面抗原を認識するCARである〔1〕又は〔2〕記載のiPS細胞。
〔4〕〔1〕~〔3〕から選ばれるいずれかに記載のiPS細胞を有効成分とする医薬。
〔5〕腫瘍抗原又はウイルス抗原特異的細胞傷害性T細胞(CTL)にOct3/4、Sox2、Klf4、c-Myc及びSV40 large T抗原の遺伝子を導入して抗原特異的細胞傷害性T細胞(CTL)由来iPS細胞を得(T-iPSC)、得られたT-iPSCにキメラ抗原受容体(CAR)を導入して分化誘導することを特徴とするキメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞の製造法。
〔6〕がん又はウイルス感染疾患を治療するための、キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞。
〔7〕がん又はウイルス感染疾患の治療薬製造のための、キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞の使用。
〔8〕キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞を、必要な患者に投与することを特徴とする、がん又はウイルス疾患の治療方法。
 本発明のCARが導入されたCTL由来iPS細胞は、CAR遺伝子の導入効率が約100%と効率よく、またCTLの抗原特異的細胞傷害とCARによる抗腫瘍効果の相乗効果により抗腫瘍効果が極めて高く、かつ生体内に長期間生存するためがん及びウイルス感染疾患の治療薬として有用である。

 本発明の細胞は、CARが導入されたウイルス特異的もしくは腫瘍抗原特異的CTL由来のiPS細胞である。当該iPS細胞は、CTLを人工的に初期化してTCRを保持したiPS細胞とし、次いでこれにCARを遺伝子導入し分化させることにより得られる。より具体的には、CTLクローンにOct3/4、Sox2、Klf4、c-Myc及びSV40 large T抗原の遺伝子を導入してCTL由来iPS細胞を得、得られたT-iPS細胞にCARを導入し、分化することにより得られる。
 原料として用いられるCTLは、抗原特異性を有することにより細胞傷害性を生じているT細胞である。特に多くの成人が既感染であるウイルスはCTLとして優れている。エプスタイン バール ウイルス(Epstein Barr Virus)のLMP1抗原、LMP2抗原、EBNA抗原など、アデノウイルス抗原のペントン抗原、ヘキソン抗原など、サイトメガロウイルスのCMVpp65抗原、IE1抗原、IE2抗原、水痘帯状疱疹ウイルス(VZV)のIE61抗原、IE62抗原、IE63抗原、ORF10抗原、単純ヘルペスウイルス(HSV)の抗原などがあげられる。また、ウイルス関連腫瘍に関するウイルスとして、ヒトパピローマウイルス(HPV)のE6抗原、E7抗原など、HTLV-1ウイルスのTax抗原などのウイルス特異的CTLも2抗原を標的とするiPSC由来CAR-rejT(rejuvenated CTL)を作製するのに優れている。しかしウイルス抗原のみならずWT1抗原、NY-ESO-1抗原、MAGE-1抗原、MART-1抗原などの腫瘍抗原など他の抗原特異的CTLからも作成可能である。
 本発明に用いられるCTLは、ヒトCTLが好ましい。このT細胞の起源であるヒトとしては、健常人であっても、ウイルス感染症を患っているヒトであってもよい。また、T細胞の起源として好ましいヒトは、本発明によって製造されたCAR搭載iPS細胞を投与されるべき患者とHLAの型が一致しているヒトであり、さらにCAR搭載iPS細胞が投与されるヒトと同一人が好ましい。他家細胞を用いる場合は、CTLエピトープのHLA拘束性が一致するHLAを持っていれば、HLAをゲノム編集したヒトiPS細胞より誘導されたCTLも使用することができる。ウイルスとしては、ほとんどの成人が感染しているEBウイルス、サイトメガロウイルスが好ましいが、それに限らない。
 このようなCTLは、例えばヒトの組織から公知の手法により単離することができる。ヒトの組織としては、前記T細胞を含む組織、例えば、末梢血、リンパ節、骨髄、胸腺、脾臓、臍帯血、病変部組織が挙げられる。これらの中では、ヒトに対する侵襲性が低く、調製が容易であるという観点から、末梢血が好ましい。ヒトT細胞を単離するための公知の手法としては、例えば、細胞分離用磁気ビーズなどを用いる磁気セレクション、CD4又はCD8等の細胞表面マーカーに対する抗体と、セルソーターとを用いたフローサイトメトリー、抗CD3抗体、抗CD28抗体を用いる活性化T細胞誘導法などが挙げられる。抗原特異性を有するT細胞を含むヒトの組織より単離する場合には、所望の抗原を結合させたMHC(主要組織適合遺伝子複合体)を多量体化させたもの(例えば、「MHCテトラマー」、「プロ5(登録商標)MHC クラスIペンタマー」)を用いて、ヒトの組織より所望の抗原特異性を有するT細胞を精製することができる。
 CTLを初期化するために用いられる遺伝子は、Oct3/4、Sox2、Klf4、c-Myc及びSV40 large T抗原である。
 本発明において、CTLに前記遺伝子を導入する方法としては特に制限はなく、公知の手法を適宜選択して用いることができる。例えば、前記遺伝子をコードする核酸の形態にて前記CTLに導入する場合においては、前記遺伝子をコードする核酸(例えば、cDNA、RNA)を、T細胞で機能するプロモーターを含む適当な発現ベクターに挿入し、該発現ベクターを感染、リポフェクション法、リポソーム法、エレクトロポレーション法、リン酸カルシウム共沈殿法、DEAEデキストラン法、マイクロインジェクション法、エレクトロポレーション法、CRISPR/Cas9などのゲノム編集にて細胞に導入することができる。
 このような発現ベクターのうち、前記遺伝子を含むステルス型RNA発現ベクターを用いるのが、がん化の危険性の低減、導入効率の点から、より好ましい。
 ステルス型RNA発現ベクターとは、ベクターが染色体に入ることを回避し、核内ではなく細胞質内で、持続的で安定して遺伝子が発現するように設計されたベクターである。1万3000塩基対以上の大きな遺伝子の導入や、10個の遺伝子の同時導入もでき、細胞に傷害を与えず、導入遺伝子が不要な時には除去が可能で、細胞がベクターを異物として認識できないステルス性を持っている。
 このようなステルス型RNA発現ベクターとしては、下記(1)~(8)のRNA配列を含むマイナス一本鎖RNA(A)と、一本鎖RNA結合タンパク質(B)、RNA依存性RNA合成酵素からなり、自然免疫構造を活性化させない複合体が挙げられる。

(3)前記RNA依存性RNA合成酵素が認識する転写開始シグナル配列、

(6)前記RNA依存性RNA合成酵素をコードするRNA配列、
(7)前記RNA依存性RNA合成酵素の活性を調節するタンパク質をコードするRNA配列、
(8)前記一本鎖RNA結合タンパク質をコードするRNA配列。
 また、T-iPS細胞を樹立する際には、前記CTLは、前記遺伝子の導入前に、インターロイキン-2(IL-2)、インターロイキン-7(IL-7)又はインターロイキン-15(IL-15)の存在下にて抗CD3抗体、抗CD28抗体によって刺激して活性化することが好ましく、フィトヘマグルチニン(PHA)、インターロイキン-2(IL-2)、同種抗原発現細胞、抗CD3抗体、抗CD28抗体、CD3アゴニスト及びCD28アゴニストからなる群から選択される少なくとも1の物質によって刺激して活性化してもよい。かかる刺激は、例えば、培地中に、PHA、IL-2、抗CD3抗体及び/又は抗CD28抗体等を添加して前記T細胞を一定期間培養することによって行うことができる。また、抗CD3抗体及び/又は抗CD28抗体は磁性ビーズ等が結合されているものであってもよく、さらにこれらの抗体を培地中に添加する代わりに、抗CD3抗体及び/又は抗CD28抗体を表面に結合させた培養ディッシュ上で前記CTLを一定期間培養することによって刺激を与えてもよい。さらにまた、前記CTLが認識する抗原ペプチドをフィーダー細胞とともに培地中に添加することによって刺激を与えても良い。
 かかる刺激を前記CTLに与えるために、培地中に添加するPHAの濃度としては特に制限はないが、1~100μg/mLであることが好ましい。また、培地中に添加するIL-2の濃度としては特に制限はないが、1~200ng/mLであることが好ましい。さらに、培地中に添加する抗CD3抗体及び/又は抗CD28抗体の濃度としては特に制限はないが、前記CTLの培養量の1~10倍量であることが好ましい。また、かかる刺激を前記CTLに与えるために、培養ディッシュの表面上に結合させた抗CD3抗体及び/又は抗CD28抗体の濃度としては特に制限はないが、コーティングの際の濃度は抗CD3抗体では0.1~100μg/mL、好ましくは1~100μg/mL、抗CD28抗体では0.1~10μg/mLであることが好ましい。
 また、かかる刺激を行うための培養期間は、前記CTLに対してかかる刺激を与えるのに十分な期間であって、前記遺伝子の導入に必要な細胞数までCTLを増殖しうる期間であれば特に制限はないが、通常2~7日間であるが、遺伝子導入効率の観点から、好ましくは3~5日間である。15mLチューブ内でT細胞とベクターを混合することにより感染させる、もしくは遺伝子導入効率を上げるという観点から、レトロネクチンがコートしてある培養ディッシュ上にて培養することが好ましい。
 前記CTLを培養し、PHA、IL-2、抗CD3抗体及び/又は抗CD28抗体等を添加する培地としては、例えば、前記CTLの培養に適した公知の培地(より具体的には、他のサイトカイン類、ヒト血清を含む、ロズウェルパーク記念研究所(RPMI)1640培地、AIM VTM medium、NS-A2を用いることができる。培地には、PHA、IL-2、抗CD3抗体及び/又は抗CD28抗体以外にも、培養に必要なアミノ酸(例えば、L-グルタミン)、抗生物質(例えば、ストレプトマイシン、ペニシリン)が添加してあっても良い。また、IL-2のかわりに培地にIL-7及びIL-15を添加することも好ましい。IL-7及びIL-15の添加濃度としては特に制限はないが、それぞれ1~100ng/mLであることが好ましい。
 また、前記CTLに前記遺伝子を導入する際、又はその後の条件としては特に制限はないが、前記遺伝子を導入した前記CTLは、フィーダーフリー条件下で培養するのが好ましい。例えば、ラミニン511E8断片であるiMatrix-511溶液又はビトロネクチンもしくはマトリジェルでコーティングされたウェルが挙げられる。フィーダー細胞条件下での培養でも樹立可能であり、フィーダー細胞としては例えば、放射線の照射や抗生物質処理により細胞分裂を停止させたマウス胎児繊維芽細胞(MEF)、STO細胞、SNL細胞が挙げられる。
 さらに、前記CTLからT-iPS細胞に誘導する過程において、翌日からiPS細胞の培地を添加しておくことが好ましい。その後は1日おきに半量ずつ培地交換を行い、徐々にCTL培地からiPS培地に置換するのが好ましい。
 また、前記CTLからiPS細胞への移行に合わせて、前記CTLの培養に適した公知の培地から、iPS細胞の培養に適した培地に徐々に置換していきながら培養することが好ましい。かかるiPS細胞の培養に適した培地としては、公知の培地を適宜選択して用いることができ、例えば、iMatrixコーティングした場合にはStemFit AK03Nもしくはビトロネクチンでコーティングした場合にはEssential 8 Medium、マトリジェルの場合はmTeSR、MEF細胞などのフィーダー細胞上ではノックアウト血清代替物、L-グルタミン、非必須アミノ酸、2-メルカプトエタノール、及びb-FGF等を含有する、ダルベッコ変法イーグル培地/F12培地(ヒトiPS細胞培地)が望ましい。
 このようにしてT-iPS細胞の選択は、公知の手法を適宜選択することによって行うことができる。かかる公知の手法としては、例えば、ES細胞/iPS細胞様コロニーの形態を顕微鏡下にて観察して選択する方法が挙げられる。一方でシングルセルであるCTLクローンから樹立したT-iPSの場合、性質が似ていることが多いため、T-iPS細胞の各コロニーを選択せず、樹立できたコロニーを全てそのまま継代する方法もある。
 このようにして選択された細胞がT-iPS細胞であるということの確認は、例えば、選択された細胞における未分化細胞特異的マーカー(SSEA-4、Tra-1-60、及びTra-1-81等)の発現をRT-PCR等によって検出する方法やALP染色で確認する方法、選択された細胞をマウスに移植して、そのテラトーマ形成を観察する方法により行うことができる。また、このようにして選択された細胞が前記CTL由来であることの確認は、TCR遺伝子再構成の状態をゲノムPCRによって検出することにより行うことができる。
 これらの細胞を選択して回収する時期は、コロニーの生育状態を観察しながら、またセンダイウイルス前記因子ベクターにGFPなどの標識が入っている場合は、GFPが消失したことを蛍光顕微鏡で確認できた頃に回収するのが好ましい。概ね、前記遺伝子を前記T細胞に導入してから10~40日、好ましくは14日~28日である。培養環境としては、上記にて特に断りのない限り、好ましくは、5%CO2、35~38℃、より好ましくは37℃の条件である。
 得られたCTL由来iPS細胞にCARを導入するには、レンチウイルスベクターなどウイルスベクターを用いる方法、CRISPR/Cas9などのゲノム編集、トランスポゾンを用いる方法などがある。
 CARは、腫瘍細胞の表面抗原への特異性とT細胞の活性化能を有するキメラ抗原受容体である。CAR-T療法とは、このキメラ抗原受容体をコードする核酸をT細胞に導入し、得られた遺伝子導入T細胞を体外で増殖させて輸注する療法である。
 本明細書において「CAR」とは、抗原に結合する細胞外ドメイン、前記細胞外ドメインとは異なるポリペプチドに由来する膜貫通ドメイン及び少なくとも1つの細胞内ドメインを含む融合タンパク質を示す。「キメラ抗原受容体(CAR)」は、「キメラ受容体」、「T-body」、「キメラ免疫受容体(CIR)」と呼ばれることがある。「抗原に結合する細胞外ドメイン」は、ある抗原に結合することができる任意のオリゴペプチド又はポリペプチドを示し、「細胞内ドメイン」は細胞内で生物学的プロセスの活性化又は阻害をもたらすシグナルを伝達するドメインとして機能することが知られている任意のオリゴペプチド又はポリペプチドを意味する。
 本明細書において「腫瘍抗原」とは、細胞のがん化に伴って新たに発現が認められるようになる、抗原性を有する生体分子を意味する。腫瘍抗原の検出、例えば免疫学的な検出は、がん化した細胞とその母細胞の区別に有用である。本発明における腫瘍抗原は、腫瘍特異抗原(腫瘍細胞のみに存在し、他の正常細胞には見られない抗原)、又は腫瘍関連抗原(他の臓器・組織又は異種異系の正常細胞にも存在する抗原、発生・分化の途上において発現する抗原)を含む。
 本明細書において「単鎖抗体(scFv)」とは、抗原との結合能力を保持した、抗体由来の一本鎖ポリペプチドを意味する。例えば組換えDNA技術により形成され、スペーサー配列を介して免疫グロブリン重鎖(H鎖)及び軽鎖(L鎖)フラグメントのFv領域を連結した抗体ポリペプチドが例示される。scFvの各種作製方法が公知であり、米国特許第4694778号公報、サイエンス(Science)、第242巻、第423-442頁(1988)、ネイチャー(Nature)、第334巻、第54454頁(1989)、サイエンス(Science)、第242巻、第1038-1041頁(1988)に記載されている方法が挙げられる。
 本明細書において「ドメイン」とは、ポリペプチド内の一領域であって、他の領域とは独立して特定の構造に折りたたまれる(フォールディングされる)領域を意味する。
 代表的なCARの構造は、腫瘍細胞の表面抗原を認識する単鎖抗体(single chain variable fragment:scFv)、膜貫通ドメインとT細胞を活性化させるTCR複合体CD3ζの細胞内ドメインから構成される。このような構成のCARは第一世代CARと呼ばれている。単鎖抗体部分の遺伝子は、例えば標的とする抗原を認識するモノクローナル抗体を産生するハイブリドーマから単離される。CARを発現するT細胞は、腫瘍細胞上の主要組織適合抗原クラスIの発現とは無関係に腫瘍細胞の表面抗原を直接認識し、同時にT細胞を活性化することで、効率よく腫瘍細胞を殺傷することが可能である。
 第一世代CARのT細胞活性化能を増強する目的で、T細胞の共刺激分子であるCD28もしくは腫瘍壊死因子(TNF)受容体スーパーファミリーであるCD137(4-1BB)の細胞内ドメインを連結した第二世代CARが開発されている。さらなる改良型として、CD28、4-1BB又はCD134(OX40)のうち2つの共刺激分子をタンデムに連結した第三世代CARの開発も進んでいる(Current Opinion in Immunology,Vol.21,p215-223(2009)。また、安全システムとして副作用発現時にCAR-T細胞に細胞死を誘導できるようにするために、自殺遺伝子inducible caspase-9を組み入れたCARも有用である。
 CARの導入手段は、具体的には、レンチウイルスなどのウイルスベクターにより遺伝子導入を行う、またはpiggy bacなどのトランスポゾンを用いる、またはCRISR/Cas9などを用いたゲノム編集の方法がある。
 CARが導入された細胞の確認は、scFvと結合するプロテインLを染色することによりCAR遺伝子導入効率をフローサイトメトリーで解析する。
 CARが導入された細胞は、分化誘導することにより、CARが導入されたCAR-rejT細胞にすることができる。分化誘導手段は、CAR遺伝子導入後のCAR-T-iPS細胞を細かく砕きフィーダー細胞上で2週間培養し、その後Notch Ligandが発現するフィーダー細胞上で更に4週間サイトカインと共に培養し、4週間後浮遊細胞を確認しT細胞受容体(TCR)刺激する。刺激後CAR遺伝子導入効率と抗原特異性を前述の方法で確認する。分化誘導されたCAR-rejTはCAR遺伝子が導入されており、更にもとの抗原特異性を保持している。
 最終細胞の採取は、抗原特異性のある細胞が少ない場合は抗原特異性のある細胞をテトラマーを用いてビーズセレクション、もしくはFACSソートを行う。CAR遺伝子導入効率が少ない場合はプロテインL染色を行い、CAR陽性細胞をソートする。その他CD8陽性細胞のセレクションなど必要な細胞を選別し、増幅させることができる。
 本発明の方法により製造されるCARが導入されたiPS細胞由来のCTL細胞は、2つの抗原を標的とするため、腫瘍細胞の逃避機構が働きにくく、効率よく殺細胞効果が発揮できる。各種の悪性腫瘍、ウイルス関連腫瘍の治療薬として有用である。
 悪性腫瘍治療薬の例としてはSurvivin,NY-ESO-1,WT-1,MAGE3などの腫瘍抗原特異的rejTにGD2-CAR,HER2-CAR,NY-ESO-1-CAR,MUC1-CAR,CD19-CARなどを組み合わせることができる。
 ウイルス関連腫瘍治療薬の例としては、EBウイルス関連腫瘍であるEBウイルス関連リンパ腫、上咽頭がんに対するLMP2特異的rejTにLMP1-CAR/LMP2-rejTもしくはLMP1特異的rejTにLMP2-CARを組み合わせたLMP2-CAR/LMP1-rejT,EBウイルス陽性B細胞リンパ腫に対するCD19-CAR/LMP1-rejT,CD19-CAR/LMP2-rejT,成人T細胞白血病(ATL)に対するTax特異的rejTにNY-ESO-1-CARを組み込んだNY-ESO-1/Tax-rejT,HBZ特異的rejTにNY-ESO-1-CARを組み込んだNY-ESO-1/HBZ-rejTなどが挙げられる。
 また、生体内での耐久性に優れているとされるウイルス特異的rejTとCARとの組み合わせも有用である。例えばLMP2特異的rejT、LMP1特異的rejT、CMV特異的rejT、アデノウイルス特異的rejT、VZV特異的rejT、HSV特異的rejTなどにGD2-CAR, HER2-CAR,NY-ESO-1-CAR,MUC1-CAR,CD19-CARなどのCARを組み合わせる方法も有用である。
 本発明の免疫細胞治療薬は、特に限定されないが、好ましくは、非経口投与、例えば、静脈内、腹腔内、皮下又は筋肉内投与することができ、より好ましくは、静脈内投与することができる。あるいは、患部に局所投与することもできる。
 本発明の医薬組成物は、本発明の方法によって製造したT-iPS細胞を、公知の製剤学的方法により製剤化することにより調製することができる。例えば、カプセル剤、液剤、フィルムコーティング剤、懸濁剤、乳剤、注射剤(静脈注射剤、点滴注射剤等)、などとして、主に非経口的に使用することができる。
 これら製剤化においては、薬理学上許容される担体又は媒体、具体的には、滅菌水や生理食塩水、植物油、溶剤、基剤、乳化剤、懸濁剤、界面活性剤、安定剤、ベヒクル、防腐剤、結合剤、希釈剤、等張化剤、無痛化剤、増量剤、崩壊剤、緩衝剤、コーティング剤、滑沢剤、着色剤、溶解補助剤あるいはその他の添加剤等と適宜組み合わせることができる。また、前記疾患の治療又は予防に用いられる公知の医薬組成物や免疫賦活剤等と併用してもよい。
 本発明の医薬組成物を投与する場合、その投与量は、対象の年齢、体重、症状、健康状態、組成物の種類(医薬品、飲食品等)等に応じて、適宜選択される。
 以下に実施例を挙げて本発明をさらに説明するが、本発明はこれら実施例に限定されるものではない。
実施例1 ヒトパピローマウイルス(HPV)特異的CTLクローンよりセンダイウイルスベクターを用いてT-iPS細胞の樹立。
1)健常人末梢血より末梢血単核球を分離後、抗原提示目的に樹状細胞を誘導した。7日後、誘導した樹状細胞にHPV抗原ペプチド(HPV16-E6,A2402)を添加し15分間CO2インキュベーターで培養後、末梢血単核球と共培養開始した。更に樹状細胞との共培養を繰り返し、約8~10日後にHPV特異的CTL検出のため、CTLをMHCテトラマーで染色後、フローサイトメトリーにてテトラマー陽性率を確認した。HPV特異的CTLを確認後、シングルセルソートもしくはテトラマー/PEビーズセレクション後限界希釈法を行い、50GyのX線照射後のPBMCとIL2、PHAを加えて刺激した。
2)約3~6週間後立ち上がったコロニーのテトラマー染色を行い、フローサイトメトリーでCTLクローン樹立の確認をした。樹立確認後CTLクローンをCD3/28刺激後、以下のA)のベクターを用い遺伝子導入した。iMatrixでコートした6wellプレートに遺伝子導入後のCTLを移動し、IL-2を加えたCTLメディウムを培地としてCO2インキュベーターで培養開始した。(図1)
 A)SeV4因子ベクター + SV40 large T抗原
3)SeV遺伝子導入翌日、iPSメディウム(StemFitAK03N)を等量加え、その後は1日おきに半量ずつStem FitAK03Nに置換した。
4)7日後にT-iPS細胞のコロニーが観察でき、その後コロニーピックアップを行った。
実施例2 エプスタイン・バールウイルス(EBV)特異的CTLクローンよりセンダイウイルスベクターを用いてT-iPS細胞の樹立。
1)健常人末梢血より末梢血単核球を分離後、抗原提示目的に樹状細胞を誘導した。7日後、誘導した樹状細胞にEBV LMP2抗原ペプチド(A2402)を添加し15分間CO2インキュベーターで培養後、末梢血単核球と共培養開始した。更に樹状細胞との共培養を繰り返し、約8~10日後にLMP2特異的CTL検出のため、CTLをMHCテトラマーで染色後、フローサイトメトリーにてテトラマー陽性率を確認した。LMP2特異的CTLを確認後、シングルセルソートもしくはテトラマー/PEビーズセレクション後限界希釈法を行い、50GyのX線照射後のPBMCとIL2、PHAを加えて刺激した。
2)約3~6週間後立ち上がったクローンのテトラマー染色を行い、フローサイトメトリーでLMP2CTLクローン樹立の確認をした。樹立確認後LMP2CTLクローンをCD3/28刺激後、センダイウイルスベクターを用い、MOI10で遺伝子導入した。iMatrixでコートした6wellプレートに遺伝子導入後のCTLを移動し、IL-2を加えたCTLメディウムを培地としてCO2インキュベーターで培養開始した。
3)約2週間後にLMP2CTL由来T-iPSコロニーが多数樹立でき、コロニーピックアップして拡大培養を行なった。その後レンチウイルス由来LMP1-CARベクターを感染させLMP1-CAR/LMP2-T-iPSを作成後LMP1-CAR/LMP2-rejTを分化誘導した。
4)分化誘導したLMP1-CAR/LMP2-rejTのLMP1-CAR発現率は100%と非常に高かった。
5)細胞傷害性試験、共培養試験を行うと効率よく腫瘍細胞を排除することができた。
 図2に末梢血から誘導したLMP2-CTLと樹立したCTLクローンのテトラマー染色結果を示す。

実施例3
(細胞傷害性試験)
1)EBウイルス関連リンパ腫細胞株に対するLMP1-CAR/LMP2-rejTの細胞傷害性と末梢血由来LMP1-CARTの細胞傷害性細胞傷害性を比較するために51クロム放出試験を行った。エフェクターとしてLMP1-CAR/LMP2-rejTもしくは末梢血由来LMP1-CART、クロムでラベルしたターゲットであるHLA一致EBウイルス関連リンパ腫細胞株(節外性NK/T細胞リンパ腫,鼻型;ENKL)、コントロールターゲットであるHLA不一致EBウイルス感染腫瘍細胞株(LCL)をエフェクター:ターゲット比を5:1と2.5:1で6時間共培養した。
2)共培養後培養上清を別のカウンター用のプレートに移し、乾燥後プレートリーダーで測定した。
3)LMP1-CAR/LMP2-rejTはHLA一致のEBウイルス関連リンパ腫細胞株に対して強い抗原特異的細胞傷害性を示したが(70-80%)、コントロールのHLA不一致腫瘍細胞株に対しては10%以下と細胞傷害性を示さなかった。末梢血由来LMP1-CARTはHLA一致のEBウイルス関連リンパ腫細胞株に対して30から50%程度の細胞傷害性を示した。LMP1-CAR/LMP2-rejTの細胞傷害性はLMP1-CARTより強力であった(図6)。
実施例4
(共培養試験)
1)EBウイルス関連リンパ腫細胞株に対するLMP1-CAR/LMP2-rejTの細胞傷害性と末梢血由来LMP1-CARTの細胞傷害性細胞傷害性を比較するために共培養試験を行った。エフェクターとしてLMP1-CAR/LMP2-rejT、コントロールエフェクターとしてEBウイルスではなく他の抗原特異性を持つrejTを用いた。ターゲットとしてHLA一致EBウイルス関連リンパ腫細胞株(ENKL)を用いた。エフェクター:ターゲット比を5:1で9日間共培養した。
2)共培養後腫瘍細胞とT細胞の比率をCD56,CD3染色後フローサイトメトリーで解析した。
3)LMP1-CAR/LMP2-rejTはHLA一致のEBウイルス関連リンパ腫細胞を9日目には排除したが、抗原特異性を持たないコントロールのrejTでは腫瘍細胞を排除することができず、腫瘍の増殖を認めた(図7)。
実施例5
(抗がん効果)
 マウスモデルによるLMP1-CAR/LMP2-rejTとLMP2-rejTの生存率比較試験
 EBウイルス関連リンパ腫細胞株(ENKL)を免疫不全マウスの腹腔内に移植後、LMP1-CAR/LMP2-rejT治療群、LMP2-rejT治療群、無治療コントロールの3群に分けたマウスの生存率を比較した。
 結果を図8に示す。
実施例6
(CID投与によるアポトーシス)
 LMP1-CAR/LMP2-rejTにダイマライザーである低分子化合薬CIDを投与した48時間後のAnnexin/7-AAD陽性率を解析し、アポトーシスの割合を調べた。
 結果を図9に示す。

Claims (8)

  1.  キメラ抗原受容体が導入された抗原特異的細胞傷害性T細胞由来のiPS細胞。
  2.  抗原特異的細胞障害性T細胞の抗原が、ウイルス抗原又は腫瘍抗原である請求項1記載のiPS細胞。
  3.  キメラ抗原受容体が、腫瘍表面抗原を認識するCARである請求項1又は2記載のiPS細胞。
  4.  請求項1~3のいずれか1項記載のiPS細胞を有効成分とする医薬。
  5.  腫瘍抗原又はウイルス抗原特異的細胞傷害性T細胞(CTL)にOct3/4、Sox2、Klf4、c-Myc及びSV40 large T抗原の遺伝子を導入して抗原特異的細胞傷害性T細胞(CTL)由来iPS細胞を得(T-iPSC)、得られたT-iPSCにキメラ抗原受容体(CAR)を導入して分化誘導することを特徴とするキメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞の製造法。
  6.  がん又はウイルス感染疾患を治療するための、キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞。
  7.  がん又はウイルス感染疾患の治療薬製造のための、キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞の使用。
  8.  キメラ抗原受容体(CAR)が導入された抗原特異的細胞傷害性T細胞(CTL)由来のiPS細胞を、必要な患者に投与することを特徴とする、がん又はウイルス疾患の治療方法。
