Abstract

希釈条件や生体内環境でもディスク状の構造体を維持することができ、調製が容易で、生体適合性及び皮膚透過力に優れるバイセルを提供する。 pH6~8の領域において分子全体が負に帯電しているリン脂質を50質量%以上含む、バイセル、並びに該バイセルを含む化粧料、キャリア材料、フォトニック材料及びNMR配向剤。

[0001] 本発明はバイセル及び/《イセルを含む各種材料に関する。
背景技術
[0002] バイセルは、 リン脂質二分子膜のディスク状分子集合体である。 バイセル の模式図を図 1 に示す。 従来、 ( 1) ディスク状の平面部分 1 を構成する疎 水性基が長い長鎖リン脂質及び (2) ディスク状の屈曲部分 2を構成する疎 水性基が短い短鎖リン脂質又は界面活性剤、 の少なくとも 2成分から構成さ れるバイセルが知られている。
このようなバイセルは最小の脂質二分子膜モデルとして生体膜の研究に利 用されている。 バイセルは、 磁場配向する性質から NMRを用いた膜タンパク質 の解析に利用されたり、 薬物送達システムのキャリア材料、 化粧料及び皮膚 外用剤等に利用されている (特許文献 1 _2、 非特許文献 1 _7) 。

発明が解決しようとする課題
[0005] しかしながら、 従来のバイセルは、 (1) 長鎖リン脂質及び (2) 短鎖リ ン脂質又は界面活性剤の種類に応じて、 構成成分の配合量や配合割合などデ ィスク状構造を形成する条件の検討が必要であった。
また、 従来の少なくとも 2成分で構成されるバイセルの分散液は、 希釈す ると、 その構成成分である (2) 短鎖リン脂質又は界面活性剤が乖離してし まい、 ディスク状の構造体を維持することができないという問題があった。 同様に、 生体内の夾雑物が多い環境などでも (2) 短鎖リン脂質又は界面活 性剤が乖離してしまい、 ディスク状の構造体を維持することができず、 また 乖離した界面活性剤は生体に有害なため、 使用条件や使用環境に制限があっ た。
[0006] 従って、 本発明の目的は、 希釈条件や生体内環境でもディスク状の構造体 を維持することができ、 調製が容易で、 生体適合性及び皮膚透過力に優れる バイセルを提供することにある。
課題を解決するための手段
[0007] 本発明者らは、 上記課題を解決するため鋭意研究した結果、 下記のバイセ ルが上述した課題を解決できることを見出し、 本発明を完成した。
即ち、 本発明は、 次の発明を提供するものである。

発明を実施するための形態
[0010] 以下本発明のバイセルについて詳細に説明するが、 本発明はこれらに限定 されるものではない。 また、 本発明において 「室温」 とは周囲環境温度をい う。
[バイセル]
本発明のバイセルは、 卩昍〜 8の領域で分子全体が負に帯電しているリン脂 質を 5 0質量%以上含む。 一実施形態においては、 バイセルの平面部分及び バイセルの屈曲部分 (周縁部) のいずれも当該リン脂質を含んでいる。
前記リン脂質のバイセル中の含有量は 5 0質量%以上であり、 7 0〜 1 0 0質量%であることが好ましく、 9 0〜 1 0 0質量%であることがより好ま しく、 9 5〜 1 0 0質量%であることが更に好ましく、 1 0 0質量%である ことが特に好ましい。
バイセル中に含まれるリン脂質は 1種類であつてもよいし複数種類であつ てもよい。
本発明のバイセルは、 前記リン脂質を主成分とするものであり、 調製が容 易であり、 希釈した場合等でもディスク状の構造体を維持することができる 〇 また、 本発明のバイセルは、 N 3 +、 、

[0017] 一般式 (1) で表される化合物には、 上記例示した化合物のナトリウム塩 、 カリウム塩などの金属塩、 及びアンモニウム塩などの有機塩も含まれる。 一般式 (1) で表される化合物は、 1種でも単独で用いても、 異なる 2種 以上を組み合わせて用いてもよい。
[0018] 本発明のバイセルは、 前記リン脂質以外の化合物を含んでもよい。 前記リ ン脂質以外の化合物としては、 前記リン脂質以外のリン脂質、 コレステロー ルといった疎水性分子等が挙げられるが、 これらのバイセル中の含有量は 0 〜 5 0質量%であり、 好ましくは〇〜 3 0質量%であり、 より好ましくは 0 〇 2020/175281 8 卩(:170? 2020 /006556

また、 本発明のバイセルに機能性を付与するため、 前記リン脂質以外の化 合物には、 蛍光ラベル化剤、 リガンド、 薬剤分子等を結合したものを用いて もよい。
[0019] 本発明において、 バイセルの大きさとは、 ディスク状の平面部分の長径を いい、 透過型電子顕微鏡観察で測定される値をいう。 本発明のバイセルの大 きさは調製条件によっても異なるが、 通常 10 〜10 111程である。 また、 動的 光散乱法によって測定した粒径 (体積基準の頻度分布において累積頻度が 5 0 %となる粒径) から、 バイセルの大きさを測定することもできる。 例えば 、 バイセル分散液に、 超音波処理を施した場合、 超音波処理時間が長いほど バイセルの大きさが小さくなることが動的光散乱法によって測定した粒径か ら把握される。
また、 本発明において、 バイセルの厚さは通常 2〜 6 程である。
[0020] [バイセルの製造方法]
本発明のバイセルは、 以下の工程 1及び工程 2を有する方法により製造す ることができる。

工程 1では、 卩昍〜 8の領域において分子全体が負に帯電しているリン脂質 を、 2質量%以上の濃度で、 前記リン脂質の相転移温度未満の温度で、 水系 分散液に分散する。
[0022] 前記リン脂質の、 工程 1の水系分散液における濃度は、 2〜 1 0質量%で あり、 好ましくは 3〜 7質量%、 より好ましくは 5質量%である。 本発明の バイセルが前記リン脂質以外の化合物を含む場合も、 工程 1 における水系分 散液における混合物の濃度は、 前記の範囲とすることが好ましい。
[0023] 工程 1 において水分散媒を調製する際の温度は、 前記リン脂質の相転移温 度未満の温度であり、 好ましくは、 室温である。

工程 2では、 工程 1で調製した水系分散液を、 前記リン脂質の相転移温度 以上の温度に加熱する。 前記リン脂質を 2種以上用いる場合は、 加熱温度を 最も相転移温度が高い化合物の相転移温度以上とする。 加熱する温度は、 前 記リン脂質の相転移温度より 2 °〇以上 3 0 °〇以下の温度であることが好まし い。 加熱する方法は、 分散液全体が所望とする温度以上になる方法であれば 特に限定されず、 例えば、 容器に入れた分散液を湯浴に浸潰して温める方法 、 電子レンジでマイクロ波を照射する方法などが挙げられる。 加熱する時間 は、 加熱する方法にもよるが、 1 5分程度であることが好ましい。
[0026] 工程 2の後、 必要に応じて以下の工程 3を行なってもよい。

工程 3では、 工程 2で加熱した水系分散液を、 前記リン脂質の相転移温度 未満の温度に冷却する。 冷却する温度は、 前記リン脂質の相転移温度より好 ましくは 2 °〇以下の温度、 より好ましくは 1 5 °〇以下の温度である。 冷却す る方法は、 分散液全体が所望とする温度以下になる方法であれば特に限定さ れず、 例えば、 容器に入れた分散液を室温で静置する、 氷浴に浸潰する方法 が挙げられる。 冷却する時間は、 冷却する方法にもよるが、 1時間程度であ ることが好ましい。
[0027] 必要に応じて、 工程 3の後得られるバイセルの分散液を分散媒で希釈して もよい。 本発明のバイセルは、 その分散液中での前記リン脂質の濃度が 1 〇 〜 0 . 2質量%であり、 この濃度に応じて分散液は構造色を示す。 特に、 前記 リン脂質の濃度が 0. 5〜 1 . 5質量%の濃度範囲において、 11 領域 (350 ) か ら可視光領域 (650n.ii) まで、 幅広い波長の構造色を調整することができる。
[0028] また、 本発明のバイセルの分散液は、 分散液の温度変化に応じて可逆的に 異なる色を呈する。 この温度変化により生じる構造色の変化は、 バイセルの 第一の構造と第二の構造とに起因するものである。 第一の構造は前記リン脂 〇 2020/175281 10 卩(:170? 2020 /006556
質が平面的な集合構造 (ゲル相) であり、 第二の構造は前記リン脂質が波状 の集合構造 (リップル相) であり、 バイセルがいずれの構造となるかは、 前 記リン脂質の相転移温度によって決まる。

本発明のバイセルは、 生体に対する有害性が懸念される界面活性剤を必要 とせず、 生体適合性に優れるリン脂質を主成分とするものであり、 生体適合 性に優れる。 また、 本発明のバイセルは、 希釈条件や生体内環境でもデイス ク状の構造体を維持することができ、 皮膚透過力にも優れる。 したがって、 本発明のバイセルはスキンケア材料をはじめとする各種化粧料や薬物送達シ ステム用のキャリア材料に有用である。

が式 (2 _ 2) である) 場合、 該化合物は極性基 に親水性に優れるヒドロキシル基を 2つ有する。 このようなジオール構造を 有する化合物 (例えば、 セラミ ドなど) は、 保湿剤として利用されており、 本発明のバイセルはそれ自身が高い保湿力を有し、 かつ皮膚透過力が高い化 粧料として期待される。 化粧料は、 本発明のバイセル及び目的に応じて添加 される各種成分を含む組成物として使用できる。 目的に応じて添加される各 種成分として、 酸化防止剤、 !~1調整剤、 粘度調整剤、 香料、 顔料、 消毒剤 、 抗菌活性剤、 医薬活性剤等が挙げられる。
[0031 ] また、 本発明のバイセルは、 その構造中に疎水性物質を内包できる。 した がって、 例えば、 調製時に疎水性物質を内包させることで、 キャリア材料と して利用できる。 本発明のバイセルは、 皮膚透過力に優れ、 皮膚表面 (表皮 ) のみならず、 真皮及び皮下組織にまで透過するため、 経皮吸収用の薬物等 のキャリアに有用である。 また、 目的部位に結合するリガンドを配合したバ イセルを作成することで標的指向性を付与させることが可能になる。

ヘアレスラッ トから摘出した皮膚をフランツセルにセッ トし、 皮下投与実 験を行なった。 フランツセルに対する初期の投与は 2(« のバイセル分散液を 使用した。 皮下投与実験の概要を図 9 A及び巳に、 皮膚組織からの透過量を 図 9 Cにそれぞれ示す。 サイズが小さい 30nmのバイセルの場合だけでなく、 サイズが大きい 200 nmのバイセルの場合でも 24時間後に皮膚組織を透過したバ イセルからの蛍光が観察できた。
また、 過去に報告されている界面活性剤とリン脂質の二成分から構成され るバイセルを用いた例においては 20wt%のバイセルを用いて皮下投与を行って も皮膚を透過するバイセルは観察されていない (非特許文献 5) 。
[0047] [試験例 9 ] 蛍光ラベル DPPGバイセルの皮膚組織の集積量測定
試験例 8のバイセルのうち、 DLS測定において 200nmの粒径のバイセルを皮 膚組織へ投与して 24時間後の皮下における各々の組織中への集積量を図 1 0 に示す。 DPPGバイセルは、 最表層から 20 Mm以内の角質だけでなく、 それより も深部である表皮や真皮、 さらには皮膚組織を透過するバイセルを確認でき た。

試験例 8における DPPGバイセルの皮下浸透試験において、 投与して 24時間 後の皮膚組織からの透過物を SAXS解析した結果を図 1 1 に示す。 ディスク状 の構造体が形成されている場合に特徴的な 「-2」 の傾きを示しており、 皮膚 組織透過後も DPPGバイセルはディスク状の形状を維持していることがわかっ た。

実施例 1の DPPGバイセル (リン脂質濃度 0. 01 -1 wt%) をがん細胞 (Hep3B細 胞) に投与して 24時間後の細胞生存率を Ce U Count i ng K i tにより評価した。 比較として、 常法により調製したジミリストイルホスファチジルグリセロ —ル(DMPG)ミセル (リン脂質濃度 0. 01 -1 wt%) も同様にがん細胞 (Hep3B細胞 ) に投与して 24時間後の細胞生存率を Ce U Count i ng K i tにより評価した。 結 果を図 1 2に示す。
