前記アンピシリン感受性である細菌は、さらにフシジン酸耐性菌でもあることが好ましい。また、前記アンピシリン感受性である細菌は、Staphylococcus属の細菌であることが好ましい。ただし、Staphylococcus属の細菌としては、S. aureusではないことが好ましい。すなわち、前記アンピシリン感受性である細菌としては、アンピシリン感受性かつフシジン酸耐性であるStaphylococcus属の細菌(S. aureusを除く)が特に好ましい様態として挙げられる。なお、本明細書においては、「医薬組成物」以外の発明についても記載しているが、前記「医薬組成物」以外の発明においても、アンピシリン感受性である細菌が記載されている場合には、アンピシリン感受性である細菌としては前記の細菌が好ましい。また、前記「医薬組成物」以外の発明としては、さらに医薬部外品や化粧品等も含まれるが、当該医薬部外品や化粧品の成分として、前記アンピシリン感受性である細菌、好ましくは前記アンピシリン感受性かつフシジン酸耐性であるStaphylococcus属の細菌(S. aureusを除く)が含まれ得る。
<アンピシリン感受性である細菌>
前記アンピシリン感受性である細菌は、S. epidermidis、 S. capitis、 S.lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、およびS. siinulansの一以上の種から選択されることが好ましく、S. cohniiまたはS.lentusから選択されることがより好ましく、S. cohniiが選択されることが最も好ましい。
また前記細菌は、16S rDNA遺伝子配列が、配列番号1~7のいずれか一つにて示される配列であるか、または配列番号1~7のいずれか一つにて示される配列に対して90、91、92、93、94、95、96、97、98または99%以上の配列同一性を有する配列からなることが好ましく、16S rDNA遺伝子配列が、配列番号1~7のいずれか一つにて示される配列であることが特に好ましい。
本発明の医薬組成物においては、前記細菌が、マウス由来の細菌またはヒト由来の細菌であることが好ましく、具体的には、マウス由来の細菌である、受託番号NITE BP-02848(配列番号1)、NITE BP-02850(配列番号2)、NITE BP-02851(配列番号3)、NITE BP-02852(配列番号4)、NITE BP-02845(配列番号6)、およびNITE BP-02847(配列番号7)、ならびにヒト由来の細菌である受託番号NITE BP-02853(配列番号5)から選択される一以上の株であることが好ましい。上記細菌はそれぞれ2018年12月25日(原寄託日)に、独立行政法人製品評価技術基盤機構特許微生物寄託センター(千葉県木更津市かずさ鎌足2-5-8 122号室)において寄託されている。
<S. cohnii>
前記S. cohniiは、16S rDNA遺伝子配列が、配列番号1~5のいずれか一つにて示される配列であるか、または配列番号1~5のいずれか一つにて示される配列に対して90、91、92、93、94、95、96、97、98または99以上%の配列同一性を有する配列からなることが好ましく、また16S rDNA遺伝子配列が、配列番号1~5のいずれか一つにて示されるものであることが特に好ましい。
<医薬組成物>
本発明の医薬組成物は上述したように、アンピシリン感受性である細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を有効成分とする、医薬組成物である。
前記皮膚疾患は皮膚炎であることが好ましく、アトピー性皮膚炎、IFN-α産生促進剤により惹起される皮膚炎、イミダゾキノリン誘導体を含む製剤により惹起される皮膚炎、またはイミキモドにより惹起される皮膚炎であることが特に好ましい。
<医薬組成物の製造方法>
本発明の一様態としては、前記医薬組成物の製造方法が挙げられる。
<活性化剤>
本発明の他の一様態としては、アンピシリン感受性細菌である細菌が含まれる活性化剤が挙げられる。当該細菌はフシジン酸耐性菌でもあることが好ましく、前記アンピシリン感受性である細菌がフシジン酸耐性菌であるStaphylococcus属の細菌(S. aureusを除く)であることがより好ましい。さらに前記細菌はS. epidermidis、 S. capitis、 S.lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択されることが特に好ましく、S. cohniiであることが最も好ましい。
なお、細菌の増殖が起きているかどうかは、例えば皮膚擦過サンプル中の細菌数(CFU/g)を一般的な手法を用いて測定してそれが平常時(健常者の測定値)よりも統計学的に有意に高いか否かを判定するなど、公知の手法によって決定することができる。

本発明の他の一様態としては、S. aureusの抑制剤が挙げられる。当該抑制剤の有効成分はS. aureusの増殖を抑制する作用を持つものであれば特に限定されるものではないが、S. aureusに対して物理的または化学的に作用して当該細菌を分解または溶解させることで該細菌を死滅させる物質を当該抑制剤として用いることは、当該細菌の菌体の分解により宿主に炎症作用を惹起させて皮膚炎をより悪化させる可能性を含んでおり、望ましくない。よって、有効成分としては、生体の免疫機構によってS. aureusを攻撃し死滅させる、抗S. aureus抗体を含むものであることが好ましく、またアンピシリン感受性細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を含むものが好ましい。さらに、前記抗S. aureus抗体ならびにアンピシリン感受性細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物のいずれか一種のみを有効成分として含むものでもよいし、抗S. aureus抗体またはアンピシリン感受性細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を共に有効成分として含むものでもよい。また、前記アンピシリン感受性である細菌はフシジン酸耐性菌であることがさらに好ましく、フシジン酸耐性菌であるStaphylococcus属の細菌(S. aureusを除く)であることがさらに好ましい。例えば、具体的には前記アンピシリン感受性である細菌は、S. epidermidis、 S. capitis、 S.lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択されることが好ましく、特にS.cohniiであることがさらに好ましい。また、前記有効成分は抗S. aureus抗体であることも好ましい。前記S. aureusの抑制剤におけるS. aureusの増殖を抑制する作用が奏する具体的な効果としては、皮膚疾患の治療、改善又は予防的効果を活性化する効果や当該細菌が増殖する効果、皮膚の状態の改善効果等が挙げられるが特に限定されるものではない。当該効果によって前記S. aureusの抑制剤は、例えば、前記医薬組成物、医薬部外品、または化粧品の成分として使用することができる。
<皮膚疾患治療剤>
本発明の他の一様態としては、アンピシリン感受性である細菌の菌体、ならびに該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を含む皮膚疾患治療剤が挙げられる。前記アンピシリン感受性である細菌はフシジン酸耐性であることが好ましく、フシジン酸耐性であるStaphylococcus属の細菌(S. aureusを除く)であることがさらに好ましい。そのような細菌としては、具体的には、例えば、S. epidermidis、 S. capitis、 S.lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択されることが特に好ましく、なかでも S. cohniiを選択することがさらに好ましい。
<S. aureusの増殖抑制方法>
本発明の他の一様態としては、抗S. aureus抗体、ならびにアンピシリン感受性である細菌の菌体、ならびに該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を含む物質を用いることによる、S. aureusの増殖抑制方法が挙げられる。前記アンピシリン感受性である細菌はフシジン酸耐性菌でもあることが好ましく、アンピシリン感受性かつフシジン酸耐性菌であるStaphylococcus属の細菌(S. aureusを除く)であることがより好ましい。具体的には、例えば、S. epidermidis、 S. capitis、 S.lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択されることが特に好ましく、S. cohniiであることが最も好ましい。より好ましい様態としては、例えば、S.cohniiの細菌の菌体、ならびに該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を患部に塗布する、S. aureusの増殖抑制方法が挙げられる。
<グルココルチコイド応答遺伝子の転写亢進方法>
本発明の他の一様態としては、アンピシリン感受性の細菌の菌体、ならびに該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、および代謝物から選択される一以上を用いる、グルココルチコイド応答遺伝子の転写亢進方法が挙げられる。前記アンピシリン感受性菌はフシジン酸耐性菌であることが好ましく、Staphylococcus属の細菌(S. aureusを除く)であることがより好ましい。具体的には、例えば、、S. epidermidis、 S. capitis、 S. lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択されることが好ましく、S. cohniiおよびS. lentusの一以上であることがより好ましい。また、前記グルココルチコイド応答遺伝子はFkbp5、Zbtb16、およびTsc22d3の一以上であることが好ましい。また、前記グルココルチコイド応答遺伝子はステロイド生合成関連タンパク質をコードする遺伝子であることも好ましく、またこの場合当該ステロイド生合成関連タンパク質はスクアレン合成酵素であることが好ましい。
<菌叢の分析方法>
本発明のさらに他の一様態としては、皮膚疾患を有する生物の、皮膚に存在する菌叢の分析方法が挙げられる。当該方法としては通常皮膚の菌叢を分析する方法として用いられる方法であるなら特に限定はされず、例えば綿棒等で対象となる生物の皮膚を擦過したものを適当な培地で培養する等の手法によって行われる。また対象となる生物種としては特に限定されず、ヒトまたはヒト以外の動物から選択することができる。
<治療方法の選択を補助するための方法>
本発明の菌叢の分析方法を用いる一様態として、皮膚疾患を有する生物のための治療方法の選択を補助するための方法であって、(1)前記生物の疾患部位から採取した皮膚検体中のS.aureusの皮膚常在細菌叢に占める比率を算出する工程、(2)前記比率が基準値より高い場合において、下記(a)~(d)から選択される一以上を有効成分とする、医薬組成物を選択する工程を含む方法が挙げられる。
(a)アンピシリン感受性である細菌の菌体または該細菌の構成成分
(b)アンピシリン感受性である細菌の培養上清または該培養上清からの精製物
(c)アンピシリン感受性である細菌の抽出物
(d)アンピシリン感受性である細菌の代謝物
前記基準値としては、(i)前記皮膚疾患を有する生物と同種の生物であって、皮膚疾患を有さない個体(前記皮膚疾患を有する生物がヒトの場合は健常人)の皮膚検体中のS.aureusの皮膚常在細菌叢に占める比率の平均値または(ii)皮膚疾患を有する生物の疾患部以外から採取した皮膚検体中におけるS. aureusの皮膚常在細菌叢に占める比率などを用いることができる。
<補助情報を取得する方法>
本発明の菌叢の分析方法を用いる一様態として、皮膚疾患を有する生物における皮膚疾患の治療についての補助情報を取得する方法が挙げられる。前記方法は、(1)皮膚疾患を有する生物であって皮膚疾患治療前の生物から採取した皮膚検体中の皮膚常在細菌叢における、S.aureusの比率を算出する工程、(2)皮膚疾患治療後の前記生物から採取した皮膚検体中の皮膚常在細菌叢における、S.aureusの比率を算出する工程、(3)工程(1)で測定した前記比率と、工程(2)で測定した前記比率と比較する工程を含む。この方法によれば、例えば、医師以外の人間が治療前後の皮膚常在細菌叢におけるS.aureusの比率を算出・比較した結果を治療の補助情報として医師に提供することができ、医師は当該補助情報を用いて、行った皮膚疾患治療の経過、例えば病状の改善・増悪などを判断することができる。
<抗S. aureus抗体およびステロイド生合成関連タンパク質>
本発明のさらに他の一様態としては、皮膚疾患の治療における使用のための抗S. aureus抗体および/またはステロイド生合成関連タンパク質、皮膚疾患の治療用医薬を製造するための抗S. aureus抗体およびステロイド生合成関連タンパク質から選択される1以上の使用、さらに抗S. aureus抗体およびステロイド生合成関連タンパク質から選択される一以上を患者に投与することにより皮膚疾患を治療する方法が含まれる。また、皮膚疾患の治療における使用のためのアンピシリン感受性である細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、または代謝物も本発明のさらなる一様態として挙げられる。この場合前記アンピシリン感受性である細菌がフシジン酸耐性菌であることが好ましく、前記アンピシリン感受性である細菌がフシジン酸耐性であるStaphylococcus属の細菌(S. aureusを除く)であることがさらに好ましく、具体的には例えば、前記アンピシリン感受性である細菌が、S. epidermidis、 S. capitis、 S. lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択されることが特に好ましい。
<治療における使用、治療用医薬の製造のための使用、および治療する方法>
本発明のさらに他の一様態としては、皮膚疾患の治療における使用のためのアンピシリン感受性である細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、または代謝物が挙げられ、前記アンピシリン感受性である細菌はフジシン酸耐性菌であることが好ましく、さらにフシジン酸耐性であるStaphylococcus属の細菌(S. aureusを除く)であることが好ましい。具体的には、S. epidermidis、 S. capitis、 S. lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択される細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、または代謝物であることが好ましく、S. cohniiの細菌の菌体、該細菌の構成成分、培養上清、該培養上清からの精製物、抽出物、または代謝物であることが特に好ましい。
<細菌株>
本発明のさらに他の一様態としては、受託番号がNITE BP-02848、NITE BP-02850、NITE BP-02851、NITE BP-02852、またはNITE BP-02853のいずれかで特定される、S. cohniiに属する細菌株が挙げられる。
<細菌>
前記アンピシリン感受性である細菌とは別の態様である、本発明の細菌としては、Methylobacterium sp. RD4.1、Methylobacterium sp. SKJH-1、Mesorhizobium alhagi、Sphingomonas sp. MK355、Sphingomonas sp. AC83、Pantoea sp. lan5、Weissella cibaria、Bradyrhizobium sp. STM 3843、Lactococcus lactis、Afipia genosp.1、Methylobacterium populi、Sediminibacterium sp. IV-37、Bradyrhizobium sp. Aust13C、Pelomonas saccharophila、Bradyrhizobium sp. Shinshu-th2、Methylobacterium sp. BF15、およびBradyrhizobium sp. BTAi1の一以上の種から選択される細菌が挙げられる。前記細菌や、S. cohniiは後述の実験例3で示したように、皮膚炎増悪(臨床スコア高)と負の相関を示していることから、これらの細菌もS. cohniiと同様に皮膚炎症に対して抑制的な効果を示すことが推測される。このため、前記<アンピシリン感受性である細菌>で挙げた細菌と同様の各種用途に使用することができる。
次に本発明について実験例を示してさらに詳細に説明するが、本発明はこれらによって限定されるものではない。
なお、本実施例で使用したクローン名、由来、菌種、受託番号、配列番号を下記表1に示す。各細菌はそれぞれ2018年12月25日(原寄託日)に、独立行政法人製品評価技術基盤機構特許微生物寄託センター(千葉県木更津市かずさ鎌足2-5-8 122号室)において寄託されている。
<Tmem79KOマウスの作製>
UC DAVISリポジトリのKOMP (knock out mouse project) (https://www.komp.org/)から、Project ID:CSD74985、Allele:Tmem79tm1Wtsi DeletionのマウスES細胞を取得した。FIMRe機関の凍結マウスリソースの個体化サービス(RIKEN BRC)(http://mus.brc.riken.jp/ja/mailnews/20080605#mn/20080604#2)にて、該変異細胞をキメラマウスに個体化した。得られた変異個体と、C57/BL6Nマウスの交配を繰り返し、ホモ接合体のTmem79KOマウスを得た。Tmem79KOマウスの同定は、PCRによる変異Tmem79遺伝子の確認、および目視による皮膚状態の差異比較に基づいて行った。
(Tmem79KOマウスにおける皮膚炎臨床スコア)
作製例1で作製したTmem79KOマウスを作成し、SPF(Specific pathogen free)環境下で飼育した(SPF-Tmem79KOマウス)。
(SPF-Tmem79KOマウスの病変部における菌叢変化)
上述したようにSPF-Tmem79KOマウスにおいては二峰性の皮膚炎を引き起こすが、特に病期の後半においては頸部を中心に皮膚炎が広がっている様子が観察された。
(臨床スコアと菌叢分布)
上記と同様の手法で取得したSPF-Tmem79KOマウスの皮膚サンプルの菌叢解析(メタ16s解析)を行い(図3A)、その結果および当該マウスの臨床スコアを用いてスピアマンの相関解析を行った。結果を図3Bに示す。
[実験例4]
(SPF-Tmem79KOマウス遺伝子解析)
1~15週齢のSPF-Tmem79KOマウスとSPF-WTマウスの皮膚から実験例2と同様の手法で皮膚サンプルを取得し、RNA-seq遺伝子発現によるPathway解析を行った。結果を図4に示す。SPF-Tmem79KOマウスは上記臨床スコアの増加(皮膚炎の悪化)に伴って、様々な免疫応答に関する遺伝子発現が亢進しており、S. cohniiの割合が高く皮膚炎が一時軽減する5~6週齢の時期では、SPF-Tmem79KOマウスのサイトカイン受容体シグナルやT細胞受容体シグナルなどの免疫応答に関与する遺伝子発現が抑制されていた。
(獲得免疫不全Tmem79KOマウスの解析)
Tmem79KOマウスとRag2KOマウス(The Jackson Laboratory)とを交配して獲得免疫不全Tmem79KOマウス(Rag2-/-Tmem79-/-マウス)を作製した。
臨床スコアからも病理組織検査の結果からも、獲得免疫不全Tmem79KOマウスはTmem79KOマウスで観察される後半の皮膚炎の発症が抑制されていることがわかり、Tmem79KOマウスの二峰性皮膚炎の後半の増悪には獲得免疫が重要な役割を果たすことが示唆された。
(ノトバイオート(gnotobiote)マウスの作成)
1mLのPBS(Phosphate buffered saline) またはBHI(Brain heart infusion )に浸漬した綿棒でSPFマウスの皮膚を擦過して採取した皮膚組織を、複数の培地(マンニット食塩卵黄付加寒天培地やトリプチックソイ寒天培地など)を用いて好気下で培養し、取得した単一コロニーの16S rDNA配列を解読し菌種を同定することで、S. aureus、S. epidermidis、およびS. lentusを単離した。なお、ここで単離したS. aureusは株名:BP-02846であり、S. epidermidisは株名BP-02845であり、およびS. lentusは株名BP-02847である。
(ノトバイオートTmem79KOマウスにおける皮膚炎の誘導)
作製例2で作製したS. aureus、S. epidermidisまたはS. lentusにより作製したノトバイオートTmem79KOマウスを用いて、皮膚常在菌によって当該皮膚炎は誘導され得るか、あるいは当該皮膚炎を誘導するのは特定の菌のみなのかについて、検討を行った。
Conventionalizedマウスと菌量が2.0×106CFUのS. aureusを塗布したノトバイオートTmem79KOマウスは皮膚炎を発症する一方、菌量が2.0×106CFUのS. epidermidisまたはS. lentusを塗布したノトバイオートSPF-Tmem79KOマウスではいずれも明らかな皮膚炎の誘導はみられなかった。結果を図8Bに示す。
[実験例6]
(S. cohniiの炎症抑制効果-1)
続いてSPF-Tmem79KOマウスにおける他の細菌による影響を検証した。
さらに両抗生物質によるS. aureusおよびS. cohniiの抗生物質に対する感受性の検証を行った。S. aureusは、アンピシリンおよびフシジン酸のいずれによっても増殖が抑制される(図9CおよびD)。一方、S. cohniiはアンピシリンにより増殖が有意に抑制されるが、フシジン酸によっては増殖の抑制は引き起こされないことがわかった(図9CおよびE)。
(S. cohniiの炎症抑制効果-2)
S. aureus、C.mastitidis、または S. cohniiの培養液について菌量を2.0×106CFUに調整し、GF-Tmem79KOマウスまたはGF-WTマウスに接種した。いずれの細菌もGF-WTマウスにおいては皮膚炎を起こさなかったが(図10C)、GF-Tmem79KOマウスにおいてはS. aureusまたはC. mastitidisは皮膚炎を惹起する一方で、S. cohniiは皮膚炎を起こさなかった(図10B)。
(皮膚炎に対するブドウ球菌の予防効果)
GF-Tmem79KOマウスにS. cohnii、S. epidermidisの培養液について、菌量を2.0×106CFUに調整したものをそれぞれ接種し、当該接種の14日後にS. aureusを接種した。
S. aureus接種21日後における頸部リンパ節腫脹も、臨床スコアと同様に、S. cohniiまたはS. epidermidisの予防投与によって抑制された(図11C)。
また、各マウスの皮膚を用いて、HE染色を行うことで病理組織検査を行った。染色像からはS. cohniiまたはS. epidermidisの予防投与によって明らかに皮膚炎が抑制されることが明らかとなった(図11E)。
(皮膚炎に対するブドウ球菌の治療効果)
さらにブドウ球菌による、S. aureusによる皮膚炎への治療的効果を検証した。
(S. cohniiによる遺伝子発現解析)
上記実験例におけるマウスにおけるS. cohnii等の影響を検討するため、ノトバイオートTmem79KOマウスの皮膚から抽出したtotal RNAを用いてHiSeq(登録商標;イルミナ株式会社)によりRNA-sequencing解析を施行した。
(グルココルチコイド遺伝子の発現亢進)
S. aureus、S. cohnii、S. epidermidis、およびS. lentusを用いたノトバイオートTmem79KOマウスの皮膚におけるグルココルチコイド応答遺伝子として知られる3つの遺伝子Fkbp5、Zbtb16、およびTsc22d3の発現量について解析を行った。
グルココルチコイド受容体のレポーターアッセイとして、GAL4 Reporter, Luciferase, HEK293 Cell Line (グルココルチコイドレセプター情報伝達経路測定用細胞株)に濁度 OD(600)で0.6に菌量を調整したS.cohniiまたはS.aureusを各ウェルに加えて24時間培養し、ONE-Step luciferase assay system (BPS Bioscience #60690-1)を用いて発光量を測定することで、GAL4レポーターアッセイを行った。細胞にS. cohniiを加えて培養することで、グルココルチコイド情報伝達経路が活性化していることがわかる。結果を図18に示す。
[実験例12]
(S. cohniiの新規単離株の炎症抑制効果)
上述の実験例においてはS. cohniiは、作製例2と同様の手法で単離した株名:BP-02848を用いている。他株のS. cohniiについてもBP-02848と同様の効果があることを確認するため、マウス皮膚から新規に単離したS. cohnii株である、BP-02850、BP-02851、BP-02852を用いて以下の実験を行った。
GF-WTマウスの耳にイミキモド(製剤名;ベセルナクリーム、持田製薬株式会社)を50mg/匹、1日1回、5連日間塗布した。最初の2日間についてはS. cohniiまたはS. epidermidisを2.0×107CFU/匹で1日1回、綿棒で同時に接種した。
[実験例13]
(S. cohniiの新規単離株のS. aureus惹起性皮膚炎の予防効果)
S. cohniiの株、BP-02848およびBP-02851を接種したGF-Tmem79KOマウスへ、当該接種の14日後に菌量を2.0×106CFUに調整したS. aureusの培養液を接種した。臨床スコアを鑑みると、BP-02848およびBP-02851株の両者とも皮膚炎の改善効果を有していることが示されており、また、各マウスの頸部リンパ節腫脹においても両株により腫脹は抑制されていた(図20BおよびC)。なお、臨床スコアおよび頸部リンパ節の腫脹の程度はいずれも各株によって異なっていた。
[実験例14]
(ステロイド生合成関連遺伝子の発現)
実験例10において、S. cohniiで作製したノトバイオートマウスにおいて、グルココルチコイド受容体(GCR)が最も強く遺伝子発現していることが示された。
(スクアレンによる炎症抑制効果)
実験例14の結果を裏付けるため、スクアレン投与がS. cohniiと同様に炎症を抑制するか、また、ミフェプリストン投与によりその炎症抑制効果が阻害されるか、について検討した。
最初の3日間についてはS. cohniiを2.0×107CFU/匹で1日1回、綿棒で接種するか、スクアレン(株式会社Sigma、S3626-10ML)を200μl/匹で、マウスの耳に5連日間(1日1回)塗布した。さらにマウスの他の一群においては、実験期間においてはミフェプリストン(Sigma-Aldrich社、M8046-1G)を210μg/mLで自由飲水投与した。
(ヒト由来S. cohniiによる炎症抑制効果)
本発明者らの解析により上記で用いたマウス由来のS. cohnii株 BP-02848がヒトに対しても同様の抗炎症効果を有する事が認められた。
健常者ボランティアの皮膚からのS. cohniiの単離は、理化学研究所「ヒト皮膚細菌叢の網羅的解析」番号H27-1にて承認されたプロトコールに基づいて行った。具体的にはPBSを浸漬した綿棒で、健常者ボランティアの額部皮膚を擦り、付着物を1mLのPBSに再分散させて菌液とし、マンニット食塩培地(日水製薬株式会社、05236)に菌液を塗布して培養し、形成されたコロニーから得た単離菌株に対する16SrDNA解析に基づいて、ヒト由来のS. cohnii 株BP-02853を同定した。
SPF-WTマウスの耳にイミキモド(ベセルナクリーム 持田製薬)を50mg/匹、1日1回、5連日間塗布した。
S. cohnii株BP-02853の接種で、BP-02848と同程度に炎症の程度やリンパ節腫脹が抑えられた(図23BおよびC)。
(ヒト由来S. cohniiおよびS. epidermidisの併用によるS. aureus依存性炎症抑制効果)
S. aureusを2.0×107CFU/匹でGF-Tmem79KOマウスに接種した。接種日から5日、6日、7日、12日、13日、および14日後に、2)S. cohniiのみ、3)S. epidermidisのみ、3)S. cohniiおよびS. epidermidisの接種をそれぞれ2.0×107CFU/匹で行った(S. cohniiおよびS. epidermidisはそれぞれ2×106CFU/匹ずつで計4×106CFU/匹で接種した)(図24A)。なお、1)はポジティブコントロールでありS. aureusのみの接種しかしておらず、5)無接種のものはネガティブコントロールであり、いずれの菌も接種していない。
[実験例18]
(ヒト由来S. cohniiおよびS. epidermidisの併用によるS. aureus依存性炎症予防効果)
GF-Tmem79KOマウスにS. cohniiのみ、S. epidermidisのみ、ならびにS. cohniiおよびS. epidermidisをそれぞれ2×106CFU/匹ずつ皮膚に接種した(S. cohniiおよびS. epidermidisはそれぞれ2×106CFU/匹ずつで計4×106CFU/匹で接種した)。接種の2週間後に、S. aureusを2×106CFU/匹ずつ皮膚に接種した。経時的に観測した結果から得られた臨床スコアを図25Aに、S. aureusの接種から21日後の頸部リンパ節の重量を図25Bに示す。
(ヒト由来S. cohniiおよびS. epidermidisの併用による菌量変化)
GF-Tmem79KOマウスに、S. aureusのみ、S. aureusおよびS. cohnii、S. aureusおよびS. epidermidis、ならびにS. aureusとS. cohniiおよびS. epidermidisの接種を2.0×107CFU/匹でそれぞれ行い、21日後に菌量の測定を行った。S. aureusおよびS. cohniiならびにS. aureusおよびS. epidermidisを接種したマウスにおいてはS. aureusのみを接種したマウスに比べてS. aureusの菌量について有意な差は見られなかったが、S. aureusとともにS. cohniiおよびS. epidermidisを接種したマウスにおいてはS. aureusの菌量が有意に減少しれいることがわかった(図26A)。なお、いずれのマウスにおいてもS. aureus、S. cohnii、およびS. epidermidisを併せたtotalの菌量については差が見られなかった(図26B)。さらに解析を行うと、S. aureusとともにS. cohniiおよびS. epidermidisを接種したマウスにおいては他のブドウ球菌属に対してS. aureusの割合が有意に低いことが判った(図26C)。なお、S. aureusとともにS. cohniiまたはS. epidermidisを単独接種したマウスにおいてはS. aureusの菌量はS. aureusのみを接種した場合と比較してそれほど差はみられなかった。
(皮膚FACS解析)
S. cohniiの局所的ステロイドメタボリズムの効果を解析するため、無菌B6N WT8週齢のマウスに、2×106CFU/匹のマウス由来S. cohnii(2A1)、マウス由来S. cohnii(7C1)、ヒト由来S. cohnii(10B1)、マウス由来S. aureus(1D1)をそれぞれ背部に接種した。投与から4週間後に皮膚を採取してT細胞についてFACS解析を行った。図27は各マウスにおける菌量およびFACS解析の結果である。
[実験例21]
(糖質コルチコイドの検出-1)
更にコルチコイドについて詳細な解析を行うために無菌B6N WT8週齢のマウスに、それぞれ2×106CFU/匹のマウス由来S. cohnii(2A1)、マウス由来S. cohnii(7C1)、ヒト由来S. cohnii(10B1)、マウス由来S. aureus(1D1)を接種して28日後の背部皮膚および頸部皮膚を用いてGC-MS脂肪酸解析を行った。コルチコステロンに関してはS. cohnii、特にマウス由来S. cohnii(2A1)を接種したマウスについては背部および頸部の両方において無処理のマウスより有意に多く検出された。一方コルチコステロンの前駆体であるプロゲステロンに関してはいずれのマウスの背部、頸部ともに有意な差は見られなかった(図28)。
(糖質コルチコイドの検出-2)
Tmem79KOマウスの頸部に2×106CFU/匹のマウス由来S. cohnii(2A1)、マウス由来S. cohnii(7C1)を接種して28日後の頸部皮膚について、実験例21と同様にGC-MS脂肪酸解析を行った。実験例21と同様にコルチコステロンおよびプロゲステロンの解析結果を図29に示す。コルチコステロンに関してはマウス由来S. cohnii(2A1)を接種したマウスについては頸部において無処理のマウスより有意に多く検出された。一方コルチコステロンの前駆体であるプロゲステロンに関してはいずれのマウスにおいても有意な差は見られなかった(図29)。
(考察)
上述した実験例等の結果から、本研究では、皮膚常在菌の一種であるS. cohniiやS. epidermidis等が皮膚炎に対して予防的、治療的効果を有することを明らかにした。さらにこれらの菌は宿主に作用し、宿主の皮膚におけるグルココルチコイド応答遺伝子の活性を亢進すること、特にスクアレン合成酵素が皮膚炎抑制効果を有する菌に特異的に発現していることを見出した。

- 下記(a)~(d)から選択される一以上を有効成分とする、医薬組成物。
(a)アンピシリン感受性である細菌の菌体または該細菌の構成成分
(b)アンピシリン感受性である細菌の培養上清または該培養上清からの精製物
(c)アンピシリン感受性である細菌の抽出物
(d)アンピシリン感受性である細菌の代謝物 - 前記アンピシリン感受性である細菌が、フシジン酸耐性菌である、請求項1に記載の医薬組成物。
- 前記アンピシリン感受性である細菌が、フシジン酸耐性であるStaphylococcus属の細菌(S. aureusを除く)である、請求項1または2に記載の医薬組成物。
- 前記アンピシリン感受性である細菌が、S. epidermidis、 S. capitis、 S. lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択される、請求項1~3のいずれか一項に記載の医薬組成物。
- 前記アンピシリン感受性である細菌の16S rDNA遺伝子配列が、
配列番号1~7のいずれか一つにて示される配列であるか、
または配列番号1~7のいずれか一つにて示される配列に対して少なくとも90%以上の配列同一性を有する配列からなる、請求項1~4のいずれか一項に記載の医薬組成物。 - 前記アンピシリン感受性である細菌が、受託番号NITE BP-02848、NITE BP-02850、NITE BP-02851、NITE BP-02852、NITE BP-02853、NITE BP-02845、およびNITE BP-02847の一以上から選択される、請求項1~5のいずれか一項に記載の医薬組成物。
- 前記アンピシリン感受性である細菌が、S. cohniiである、請求項1~6のいずれか一項に記載の医薬組成物。
- 前記S. cohniiの16S rDNA遺伝子配列が、
配列番号1~5のいずれか一つにて示される配列であるか、
または配列番号1~5のいずれか一つにて示される配列に対して少なくとも90%以上の配列同一性を有する配列からなる、請求項7に記載の医薬組成物。 - 前記S. cohniiが、受託番号NITE BP-02848、NITE BP-02850、NITE BP-02851、NITE BP-02852、およびNITE BP-02853の一以上の種から選択される、請求項7または8に記載の医薬組成物。
- 前記アンピシリン感受性である細菌が、ヒトに由来する細菌である、請求項1~9のいずれか一項に記載の医薬組成物。
- 前記アンピシリン感受性である細菌が、生菌、死菌またはそれらの混合物である、請求項1~10のいずれか一項に記載の医薬組成物。
- 皮膚疾患治療、改善または予防用である、請求項1~11のいずれか一項に記載の医薬組成物。
- 前記皮膚疾患が、皮膚炎である、請求項12に記載の医薬組成物。
- 前記皮膚疾患が、アトピー性皮膚炎である、請求項12に記載の医薬組成物。
- 前記皮膚疾患が、IFN-α産生促進剤により惹起される皮膚炎である、請求項12に記載の医薬組成物。

- 前記アンピシリン感受性である細菌が、フシジン酸耐性菌である、請求項21に記載の活性化剤。
- 前記アンピシリン感受性である細菌が、フシジン酸耐性菌であるStaphylococcus属の細菌(S. aureusを除く)である、請求項21または22に記載の活性化剤。
- 前記アンピシリン感受性である細菌が、S. epidermidis、 S. capitis、 S.lentus、 S. caprae、 S. saccharolyticus、 S. warneri、 S. pasteuri、 S. haemolyticus、 S. homonis、 S. lugdunesis、 S. auricularis、 S. saprophyticus、 S. cohnii、 S. xylosus、 およびS. siinulansの一以上の種から選択される、請求項21~23のいずれか一項に記載の活性化剤。

- 前記皮膚疾患を有する生物が、SPF(specific pathogen free)動物である、請求項53または54に記載の菌叢の分析方法。
- 皮膚疾患を有する生物のための治療方法の選択を補助するための方法であって、
(1)前記生物の疾患部位から採取した皮膚検体中のS.aureusの皮膚常在細菌叢に占める比率を算出する工程、
(2)前記比率が基準値より高い場合において下記(a)~(d)から選択される一以上を有効成分とする医薬組成物を選択する工程、
を含む治療方法の選択を補助するための方法。
(a)アンピシリン感受性である細菌の菌体または該細菌の構成成分
(b)アンピシリン感受性である細菌の培養上清または該培養上清からの精製物

(2)皮膚疾患治療後の前記生物から採取した皮膚検体中の皮膚常在細菌叢における、S.aureusの比率を算出する工程、
(3)工程(1)で測定した前記比率と、工程(2)で測定した前記比率とを比較する工程、
を含む、補助情報を取得する方法。 - 皮膚疾患の治療における使用のための抗S. aureus抗体および/またはステロイド生合成関連タンパク質。
- 皮膚疾患の治療用医薬を製造するための、抗S. aureus抗体およびステロイド生合成関連タンパク質から選択される一以上の使用。
