Abstract

본 발명은 금 나노입자, 상기 금 나노입자의 표면에 부착된 폴리에틸렌 글리콜(Polyethylene glycol, PEG), 및 상기 폴리에틸렌 글리콜에 부착된 형광 물질을 포함하는 활성화 산소 센싱용 나노입자 및 이를 이용한 활성화 산소 정량 센싱 방법에 관한 것으로, 이를 이용하여 상기 금 나노입자에 의해 발생되는 활성화 산소만을 선택적으로 정량 측정할 수 있다.

Description

활성화 산소 센싱용 나노입자 및 이를 이용한 활성화 산소 센싱 방법
본 발명은 금 나노입자 표면에서 발생하는 활성화 산소만을 선택적으로 검출할 수 있는 활성화 산소 센싱용 나노입자 및 이를 이용한 활성화 산소 센싱 방법에 관한 것이다.
본 출원은 2018년 7월 27일자로 출원된 한국특허출원 제 10-2018-0087670호에 대한 우선권 주장출원으로서, 해당출원의 명세서 및 도면에 개시된 모든 내용은 인용에 의해 본 출원에 원용된다.
기존의 방사선 치료 시스템의 경우 고선량 방사선의 영향으로 정상세포에 대한 부작용을 유도하고 비효율적 전달 및 치료 거동을 보인다는 문제점이 있다.
이와 같은 단점을 보완하기 위하여 방사선을 효과적으로 흡수할 수 있는 큰 원자량을 갖는 물질들을 방사선 감응성 제재로 주입하려는 연구가 진행 되었다.
일례로, 종양 제어를 위한 방사선 치료 시스템에서, 상기 방사선 감응성 제재로 금속 나노입자를 사용할 수 있다. 상기 금속 나노입자와 고속 이온 간의 쿨롱 충돌(Coulomb scattering)에 의한 다량의 2차 전자가 상기 방사선 감응성 제재(금속 나노입자)의 표면에 생성되고, 이는 세포 내에서 활성화 산소의 생성을 증가시킨다. 생성된 활성화 산소는 저선량의 방사선 에너지에 의해 종양의 DNA 손상을 통한 사멸을 유도할 수 있다.
이를 바탕으로 최적의 방사선 치료를 위하여 상기 방사선 감응성 제재의 표면에 발생되는 활성화 산소 정량 필요성이 대두되었다.

이에 본 발명의 목적은 방사선 에너지와 큰 원자량을 갖는 금 나노입자간의 상호작용으로 인해 금 나노입자의 표면에 생성되는 활성화 산소를 정량할 수 있는 활성화 산소 센싱용 나노입자 및 이를 이용한 활성화 산소 센싱 방법을 제공하는 것이다.
본 발명은 금 나노입자, 상기 금 나노입자의 표면에 부착된 폴리에틸렌 글리콜(Polyethylene glycol, PEG), 및 상기 폴리에틸렌 글리콜에 부착된 형광 물질을 포함하는 활성화 산소 센싱용 나노입자를 제공한다.
상기 금 나노입자는 입경이 10 내지 100 nm인 것일 수 있다.
상기 형광물질은 로다민(Rhodamine), 로다민 유도체 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나인 것일 수 있다.
상기 폴리에틸렌 글리콜은 하기 화학식 1로 표시되는 것일 수 있다.

상기 금 나노입자에 대한 상기 폴리에틸렌 글리콜의 중량비(w/w)는 1:10 내지 1:20인 것일 수 있다.
상기 폴리에틸렌 글리콜은 한 쪽 말단에 티올기(-SH)를 포함하고 다른 쪽 말단에 카르복실기(-COOH)를 포함하는 것일 수 있다.
상기 금 나노입자와 상기 폴리에틸렌 글리콜 사이에 개재되며 상기 금 나노입자의 표면을 둘러싸고 있는 계면활성제를 더 포함하는 것일 수 있다.
본 발명은 또한 금 나노입자, 상기 금 나노입자의 표면에 부착된 폴리에틸렌 글리콜(Polyethylene glycol, PEG), 및 상기 폴리에틸렌 글리콜에 부착된 형광 물질을 포함하는 활성화 산소 센싱용 나노입자를 용매에 분산시키는 단계, 상기 활성화 산소 센싱용 나노입자를 포함하는 용액에 방사선을 조사하는 단계, 및 발광 상태의 형광 물질의 형광 광도를 측정하는 단계를 포함하는 활성화 산소 정량 센싱 방법을 제공한다.
상기 방사선을 조사하는 단계에서 조사되는 방사선량은 2 내지 10 Gy인 것일 수 있다.
본 발명에 따른 활성화 산소 센신용 나노입자 및 이를 이용한 활성화 산소 센싱 방법에 의하면, 방사선 조사로 인해 금 나노입자의 표면에 생성되는 활성화 산소를 정량할 수 있다.
또한, 본 발명에 따르면, 나노바이오 구조체와 생체간의 상호작용에 대한 기초 연구를 실행할 수 있다.

이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
이하, 도 1을 참조하면서 본 발명을 보다 상세히 설명한다.
도 1은 본 발명의 일 실시예에 따른 활성화 산소 센싱용 나노입자(10)의 구조를 개략적으로 나타낸 도면이다. 상기 활성화 산소 센싱용 나노입자(10)는 금 나노입자(1), 상기 금 나노입자(1)의 표면에 부착된 폴리에틸렌 글리콜(Polyethylene glycol, PEG)(2), 및 상기 폴리에틸렌 글리콜(2)에 부착된 형광 물질(4)을 포함한다.
상기 금 나노입자(1)는 196.97 g/mol의 비교적 큰 원자량 값을 가지는 물질로, 방사선 흡수율이 우수하다. 따라서 저선량의 방사선 에너지에도 반응성이 높아 방사선 감응성 제재로 적합하다.
상기 금 나노입자(1)를 용매에 분산시킨 후 방사선을 조사하면 방사선에 의해 이온화된 고속 이온과 금 나노입자(1)와의 쿨롱 충돌(Coulomb scattering)에 의하여 상기 금 나노입자(1)의 표면에 2차 전자가 생성되고 생성된 2차 전자가 주변 용매와 반응하여 활성화 산소(Reactive oxygen species, ROS)가 생성된다.
상기 금 나노입자(1)의 입경은 10 내지 100 nm인 것일 수 있다. 상기 금 나노입자(1)의 형태는 본 발명에서 제한이 없고, 일 예를 들면 원기둥, 사각기둥, 삼각기둥, 오각기둥, 육각기둥, 팔각기둥, 구, 표면의 높낮이가 일정하지 않은 구, 반구, 구의 일부분, 타원구, 반타원구, 타원구의 일부분, 사각뿔, 사각쌍뿔, 사각뿔대, 삼각뿔, 삼각쌍뿔, 삼각뿔대, 원뿔, 원뿔대, 링, 큐브(cube) 및 이들의 조합 형태로 이루어진 군에서 선택되는 어느 하나 등일 수 있다.
상기 금 나노입자(1)로부터 생성된 활성화 산소는 종양의 DNA 손상을 통한 사멸을 유도하거나, 강한 살균 작용으로 병원체로부터 세포를 보호하는 등 여러 응용이 가능하다.
그러나 용매에 분산된 금 나노입자(1)에 활성화 산소를 발생시키기 위하여 방사선을 조사하는 경우, 금 나노입자(1)의 표면에서 발생되는 활성화 산소와, 주변 용매의 분해에 의해 발생되는 활성화 산소의 구분이 어렵다는 문제점이 있다. 이를 제어하지 못하는 경우 생체내(invivo) 환경에 적용시 주변 정상 세포에도 부작용을 일으키게 된다. 따라서 적당한 방사선량을 조절하기 위하여서는 상기 금 나노입자의 표면에서 발생되는 활성화 산소만을 측정할 필요가 있다.
이에 본 발명에서는 활성화 산소를 측정하기 위한 구성으로 상기 형광 물질(4)을 조합 사용하였다.
상기 형광 물질(4)은 활성화 산소와 반응하여 형광을 유발할 수 있는 물질로, 로다민(Rhodamine) 및 이의 유도체(Rhodamine derivatives)인 것이 금 나노 입자 흡광 영역대와 겹치지 않는다는 면에서 바람직하다.
상기 로다민은 로다민 B, 로다민 123, 로다민 6G일 수 있고, 상기 로다민 유도체는 카복시테트라메틸 로다민(Carboxytetramethylrhodamine, TAMRA), 테트라메틸 로다민(tetramethylrhodamine, TMR), 및 이들의 이소티오시아네이트 유도체(isothiocyanate derivative, TRITC), 설포로다민 101(sulforhodamine 101) 및 이의 설포닐 염화물인 텍사스 레드(Texas Red sulfonyl chloride), 로다민 레드(Rhodamine Red) 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나인 것일 수 있다.
그러나 금 나노입자(1)는 로다민의 퀜처(quencher)로 작용하여 일정 거리 이내에 위치하는 로다민의 방사 형광 빛을 모두 흡수하거나 일정 거리 밖에 위치하는 로다민은 금 나노입자(1)에서 발생시키는 활성화 산소를 측정할 수 없다는 문제점이 있다.
본 발명에서는 상기 문제점을 해결하기 위한 것으로, 방사선 조사에 의한 금 나노입자(1) 표면 발생 활성화 산소를 측정하기 위하여 금 나노입자(1)와 형광 물질(4) 사이의 적당한 거리를 위한 스페이서로 폴리에틸렌 글리콜(2)을 더 포함한다.
상기 폴리에틸렌 글리콜(2)은 하기 화학식 1로 표시되는 것일 수 있다.

화학식 1에서 n은 45 내지 60의 정수일 수 있고, 50 내지 55인 것이 더욱 바람직하다. n이 45 미만인 경우 금 나노 입자가 퀜처(quencher)로 작용하는 문제가 있을 수 있고, 60을 초과하는 경우 금 나노 입자 표면에서 생성되는 활성화 산소를 인지하지 못하는 문제가 있을 수 있다.
상기 폴리에틸렌 글리콜(2)의 길이는 3 내지 10 nm인 것일 수 있다. 본 발명에 따른 활성화 산소 센싱용 나노입자(10)는 금 나노입자(1)와 형광 물질(4)의 거리가 중요하다. 상기 폴리에틸렌 글리콜(2)의 길이가 3 nm 미만인 경우 금 나노입자(1)가 형광 물질(4)의 퀜처(quencher)로 작용하여 형광 물질(4)의 방사 형광 빛을 모두 흡수하여 활성화 산소의 측정이 불가능하고, 반대로 10 nm를 초과하는 경우 금 나노입자(1)의 표면에서 발생되는 활성화 산소만을 측정할 수 없다는 문제점이 있다.
상기 폴리에틸렌 글리콜(2)은, 재료가 인근 조직의 흉터에 해롭지 않거나 자극을 주지 않거나, 이식되어야 하는 생체 조직의 특성에서 의도된 기능을 저하시키지 않는 생체적합성(Biocompatible)을 나타내므로 스페이서로 바람직하다.
상기 폴리에틸렌 글리콜(2)은 로다민과의 결합을 위하여 한 쪽 말단에 작용기(3)를 더 포함할 수 있고, 상기 작용기(3)는 카르복실기(-COOH)일 수 있다.
또한 상기 폴리에틸렌 글리콜(2)은 금 나노입자(1)와의 결합을 위하여 다른 쪽 말단에 티올기(-SH)(도시하지 않음)를 더 포함하는 것일 수 있다.
상기 금 나노입자(1) 표면에 결합되는 폴리에틸렌 글리콜(2)의 밀도가 높을수록, 형광 물질(4)의 접합(Conjugation) 효율을 높일 수 있다. 금 나노입자(1) 표면에 결합되는 폴리에틸렌 글리콜(2)의 밀도를 금 나노입자(1) 중량에 대한 폴리에틸렌 글리콜(2)의 중량으로 표현할 수 있고, 구체적으로 상기 금 나노입자(1)에 대한 상기 폴리에틸렌 글리콜(2)의 중량비(w/w)는 1:10 내지 1:20인 것일 수 있다. 상기 중량비는 금 나노입자(1)의 중량:폴리에틸렌 글리콜(2)의 중량 즉, 금 나노입자(1) 1 중량부에 대하여 폴리에틸렌 글리콜(2) 10 내지 20 중량부가 결합될 수 있음을 의미한다.
상기 금 나노입자(1)와 폴리에틸렌 글리콜(2)의 사이에 개재되며 상기 금 나노입자(1)의 표면을 둘러싸고 있는 계면활성제(5)를 더 포함할 수 있다.
상기 계면활성제(5)로는 카르복실메틸폴리비닐알콜, 폴리비닐알콜, 폴리아크릴아마이드, 폴리에틸렌이민, 폴리아미도이민, 폴리에틸렌글라이콜, 폴리라틱산, 폴리아크릴산, 폴리카프로락톤, 폴리스티렌설포네이트, 폴리수산화에틸 (메트)아크릴레이트, 키토산, 폴리솔베이트 80(TWEEN 80), 폴리비닐술폰산, 텍스트란, 소듐 콜레이트 하이드레이트, n-옥틸글루코시드, 옥틸티오글루코시드, N-옥타노일-N-메틸글루카민, N-노나노일-N-메틸글루카민, 퀼라야 껍질(quillaja bark) 유래 사포닌, 폴리옥시에틸렌 소르비탄 모노라우레이트, 도데실 황산 나트륨, 테트라메틸암모늄 하이드록사이드 용액, 헥사데실트리메틸암모늄 브로마이드(CTAB), 디도데실디메틸암모늄 브로마이드(DMAB), N,N-비스(3-D-글루콘아미도프로필)데옥시콜아미드(deoxy-BIGCHAP), N,N-비스(3-D-글루콘아미도프로필)콜아미드(BIGCHAP), 폴리에틸렌글리콜 도데실 에테르, 플루로닉 F-68, 트리톤 X-100, 트리톤 X-114, 트윈 40, 이게팔(Igepal) CA-630, 이게팔 CO-210, 이게팔 CO-520, 이게팔 CO-630, 이게팔 CO-720, 이게팔 CO-890, 이게팔 DM-970, 이게팔 CA-210, 이게팔 CA-520, 이게팔 CA-630, N-데카노일-N-메틸글루카민, 노닐페닐-폴리에틸렌 글리콜, 브리지 76(Brij 76), 브리지 58, 브리지 35P, 브리지 30, 사이클로헥실메틸-β-D-말토시드(Cymal-1), 2-사이클로헥실에틸-β-D-말토시드(Cymal-2), 5-사이클로헥실펜틸-β-D-말토시드(Cymal-5), 6-사이클로헥실헥실-β-D-말토시드(Cymal-6), 디기토닌, 데실-β-D-말토피라노시드, 라우릴-β-D-말토시드(DDM), n-헥사데실-β-D-말토시드, 운데실-β-D-말토시드, 데실-β-D-1-티오말코피라노시드, 데실-β-D-1-티오글루코피라노시드, 디메틸데실포스핀 옥사이드, 도데실디메틸포스핀 옥사이드 또는 이들의 혼합물이 예시될 수 있다. 이러한 계면활성제들은 필요에 따라 개질되어 사용될 수 있다.
이하, 상기 활성화 산소 센싱용 나노입자의 센싱 방법에 대하여 설명한다.
상기 활성화 산소 센싱용 나노입자(10)를 용매 내에 분산시키는 경우 상기 형광 물질(4)과 금 나노입자(1)의 퀜칭(quenching) 현상으로 인하여 비발광 상태를 나타낸다.
상기 용매는 벤젠, 노말부탄올, 부틸아세테이트, 사염화탄소, 클로로포름, 시클로헥산, 디클로로에탄, 디클로로메탄, 에틸아세테이트, 디에틸에스터, 헵탄, 헥산, tert-부틸메틸에스터, 메틸에틸케톤, 펜탄, 디이소프로필에스터, 테트라하이트로퓨란, 톨루엔, N-메틸-2-피롤리돈, 아세톤, 아세토니트릴, 사염화탄소, 클로로포름, 사이클로헥산, 1,2-디클로로에탄, 디클로로메탄, 디에틸에테르, 디메틸 포름아마이드, 디메틸 설폭사이드, 1,4-디옥산, 에탄올, 에틸 아세테이트, 메탄올, 메틸 3급 부틸 에테르(methyl-tert-buryl ether), 1-프로판올, 2-프로판올, 2,2,4-트리메틸펜탄, 물, PBS(phosphate buffered saline) 및 이들의 혼합물로 이루어진 군에서 선택되는 어느 하나인 것일 수 있으나 이에 제한되는 것은 아니다.
상기 용액에 방사선을 조사하여 활성화 산소를 유도할 수 있다. 이때 상기 조사되는 방사선량은 2 내지 10 Gy일 수 있고, 더욱 바람직하게 2 내지 6 Gy일 수 있다.
방사선에 의하여 금 나노입자(1)로부터 활성화 산소가 발생되고 발생된 활성화 산소가 상기 형광 물질(4)과 결합하여 발광 상태를 나타낸다.
상기 방사선량을 변화시키면서 방사선량에 따른 형광 광도를 측정하여 활성화 산소를 정량할 수 있다.
본 발명에 따른 일 실시예에서 조사한 방사선량이 2 내지 10 Gy일 때 형광 여기 파장(fluorescence excitation wavelengths, λex)은 500 nm이고, 방출 파장(emission wavelengths, λem)은 530 nm이다.
이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
[제조예 1: 활성화 산소 센싱용 나노입자 제조]
- 금 나노입자: 표면 안정제인 triton X-100 솔루션에 환원제인 질산은(AgNO3) 솔루션을 넣어준 후 금 이온 솔루션(HAuCl4)을 넣어준 다음, 환원제인 아스코르브산(Ascorbic acid)을 순차적으로 넣어준 후 마그네틱 바를 이용하여 30 초 동안 교반하고 5 분 방치하는 방법으로 반지름이 20 nm인 구형의 금 나노입자를 합성하였다.
- 폴리에틸렌 글리콜: 폴리에틸렌 글리콜은 한 쪽 말단에 티올기(-SH)를 포함하고 다른 쪽 말단에 카르복실기(-COOH)를 포함하는 HS-PEG-COOH를 사용하였다. 이때, 상기 폴리에틸렌 글리콜의 주쇄는 하기 화학식 1로 표시되는 것을 사용하였다.

- 형광 물질: 형광 물질로 디하이드로로다민 123(Dehydrorhodamine 123)을 이용하였다.
- 활성화 산소 센싱용 나노입자: 금 나노입자의 표면과 친화도(Affinity)가 높은 티올기를 이용하여 상기 금 나노입자의 표면에 상기 HS-PEG-COOH을 결합시키는 방법으로 상기 금 나노입자를 코팅하였다. 그 후 HS-PEG-COOH의 -COOH기와 디하이드로로다민 123의 -NH2기를 EDC/NHS 방법을 통해 결합하여 활성화 산소 센싱용 나노입자를 제조하였다.
디하이드로로다민 123을 접합시킨 후 세척하는 단계를 통하여 결합되지 않은 디하이드로로다민 123을 제거해줌으로써 금 나노입자 표면이 아닌 용액에 분산되어 있을 디하이드로로다민 123을 줄여주었다.
[실험예 1: 코팅 밀도에 따른 활성화 산소 센싱]
화학적 방법으로 활성화 산소를 생성시키는 팬톤반응(Fenton reaction)을 통하여 금 나노입자 표면에서의 활성화 산소가 아닌 용액 전체에 랜덤한 거리에 활성화 산소를 생성시키는 방법으로 금 나노입자 표면에서 발생하는 활성화 산소만을 측정할 수 있음을 검증하였다.

폴리에틸렌 글리콜의 코팅 밀도에 따른 형광 물질의 배치 변화를 도 2에 나타내었다.

실시예 1 및 2는 금 나노입자의 표면에 결합되는 폴리에틸렌 글리콜의 밀도에 따른 로다민의 효율을 알아보기 위하여, 상기 금 나노입자에 대한 폴리에틸렌 글리콜의 결합 중량비를 각각 1:15, 1:5 (w/w)로 설정하여 비교한 것이다. 용매 5 mL에 금 나노입자 10 mg이 분산되어 있는 용액에 폴리에틸렌 글라이콜을 각각 150 mg 과 50 mg 을 첨가하여 코팅시킨 후, 세척(Washing)하고 각각 형광물질(디하이드로로다민 123)을 접합(Conjugation)시킨 결과 형광물질의 접합 효율(Conjugation efficiency)이 폴리에틸렌 글리콜 150 mg일 때 더 우수한 것으로부터, 폴리에틸렌 글리콜의 코팅 밀도가 높을수록 로다민과의 접합(Conjugation) 효율을 높일 수 있음을 확인하였다.
비교예 1은 폴리에틸렌 글리콜을 포함하지 않는 경우 금 나노입자와 로다민과의 분산 형태를 나타낸 것이고, 비교예 2는 물에 분산된 로다민을 나타낸 것이다.
[실험예 2: 방사선량에 따른 활성화 산소 센싱]
<비교예 3>
대조군으로 금 나노입자 없이 폴리에틸렌 글리콜과 로다민만을 결합시킨 입자를 용매에 분산시킨 후 각각 2 Gy, 4 Gy, 6 Gy의 방사선을 조사하였다. 방사선을 조사한 후 생성되는 활성화 산소를 센싱하여 방사선량에 따른 상대적 형광 유닛(relative fluorescence units, RFU)을 도 3a 및 도 4a에 나타내었다.
<실시예 1>
상기 제조예 1의 실시예 1에서 제조한 활성화 산소 센싱용 나노입자를 PBS(phosphate buffered saline)에 분산시킨 후 각각 2 Gy, 4 Gy, 6 Gy, 8 Gy, 10 Gy의 방사선을 조사한 후 활성화 산소를 센싱하여 도 3b 및 도 4b에 나타내었다.
<실시예 2>
상기 제조예 1의 실시예 2에서 제조한 활성화 산소 센싱용 나노입자를 PBS(phosphate buffered saline)에 분산시킨 후 각각 2 Gy, 4 Gy, 6 Gy, 8 Gy, 10 Gy의 방사선을 조사한 후 활성화 산소를 센싱하여 도 3c에 나타내었다.
방사선을 조사하기 이전에는 모두 형광을 띠지 않음을 확인하였다. 도 3a 내지 도 3c 및 도 4a와 도 4b를 비교한 결과, 본 발명에 따른 활성화 산소 센싱용 나노입자는 방사선 조사량에 따라 비례하게 형광 강도가 증가함을 확인하였고 이로부터 방사선량이 증가할수록 생성되는 활성화 산소량이 증가하는 것을 알 수 있었다. 특히 도 3b 및 도 3c를 비교하면, 폴리에틸렌 글리콜의 함량이 150 mg인 경우의 형광 강도가 폴리에틸렌 글리콜의 함량이 50 mg인 경우의 형광 강도에 비하여 총 전달량에 따라 2 배 정도 높은 것을 확인할 수 있었다.
이상에서 본 발명의 바람직한 실시예에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고 다음의 청구범위에서 정의하고 있는 본 발명의 기본 개념을 이용한 당업자의 여러 변형 및 개량 형태 또한 본 발명의 권리범위에 속하는 것이다.

Claims (10)

  1. 금 나노입자,
    상기 금 나노입자의 표면에 부착된 폴리에틸렌 글리콜(Polyethylene glycol, PEG), 및
    상기 폴리에틸렌 글리콜에 부착된 형광 물질
    을 포함하는 활성화 산소 센싱용 나노입자.
  2. 제1항에 있어서,
    상기 금 나노입자는 입경이 10 내지 100 nm인 것을 특징으로 하는 활성화 산소 센싱용 나노입자.
  3. 제1항에 있어서,
    상기 형광 물질은 로다민(Rhodamine), 로다민 유도체 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나인 것을 특징으로 하는 활성화 산소 센싱용 나노입자.
  4. 제1항에 있어서,
    상기 폴리에틸렌 글리콜은 하기 화학식 1로 표시되는 것을 특징으로 하는 활성화 산소 센싱용 나노입자.

  5. 제1항에 있어서,
    상기 폴리에틸렌 글리콜의 길이는 3 내지 10 nm인 것을 특징으로 하는 활성화 산소 센싱용 나노입자.
  6. 제1항에 있어서,
    상기 금 나노입자에 대한 상기 폴리에틸렌 글리콜의 중량비(w/w)는 1:10 내지 1:20인 것을 특징으로 하는 활성화 산소 센싱용 나노입자.
  7. 제1항에 있어서,
    상기 폴리에틸렌 글리콜은 한 쪽 말단에 티올기(-SH)를 포함하고, 다른 쪽 말단에 카르복실기(-COOH)를 포함하는 것을 특징으로 하는 활성화 산소 센싱용 나노입자.
  8. 제1항에 있어서
    상기 금 나노입자와 상기 폴리에틸렌 글리콜 사이에 개재되며, 상기 금 나노입자의 표면을 둘러싸고 있는 계면활성제를 더 포함하는 것을 특징으로 하는 활성화 산소 센싱용 나노입자.
  9. 금 나노입자, 상기 금 나노입자의 표면에 부착된 폴리에틸렌 글리콜(Polyethylene glycol, PEG), 및 상기 폴리에틸렌 글리콜에 부착된 형광 물질을 포함하는 활성화 산소 센싱용 나노입자를 용매에 분산시키는 단계,
    상기 활성화 산소 센싱용 나노입자를 포함하는 용액에 방사선을 조사하는 단계, 및
    발광 상태의 형광 물질의 형광 광도를 측정하는 단계
    를 포함하는 활성화 산소 정량 센싱 방법.
  10. 제9항에 있어서,
    상기 방사선을 조사하는 단계에서 조사되는 방사선량은 2 내지 10 Gy인 것을 특징으로 하는 활성화 산소 정량 센싱 방법.
