Abstract
Die Erfindung betrifft eine antibakterielle Beschichtung für ein medizinisches Implantat enthaltend legierte Nanopartikel aus Gold und Platin (AuPt-Nanopartikel) sowie ein Verfahren zur Herstellung einer entsprechenden Beschichtung.
Description
Antibakterielle Beschichtung für ein medizinisches Implantat sowie Herstellung
Die Erfindung betrifft eine antibakterielle Beschichtung enthaltend Gold (Au) und Platin (Pt) Nanopartikel, ihre Herstellung, sowie die Verwendung der Beschichtung zur Bereitstellung von antibakteriell beschichteten medizinischen Implantaten.
Zur Vermeidung von Implantat-assoziierten Infektionen ist es sinnvoll, medizinische Implantate vor ihrem Einsatz mit einer antibakteriellen Beschichtung zu versehen.
In Diefenbeck et al.„Gentamicin coating of plasma Chemical oxidized titanium alloy prevents im plant- related Osteomyelitis in rats“ Biomaterials 2016 Sep; 101 : 156-64 wird eine Antibiotika enthaltende Schicht für Titan-Implantate vorgeschlagen, welche in den ersten Tagen nach Einsatz des Implantats eine Infektion verhindern soll. Eine dauerhafte Freisetzung des
Antibiotikums ist nicht vorgesehen/angeraten, da die Ausbildung von Resistenzen vermieden werden soll.
Die DE 10 2012 109 272 schlägt vor, medizinische Geräte und Implantate mit einer galvanischen Kupferschicht und einer äußeren biokompatiblen Oberflächenbeschichtung, welche mittels der Gasphasenabscheidung aufgebracht wird, zu versehen. Die antibakterielle Wirkung der so beschichteten Implantate wird durch die Freisetzung des Kupfers, während der Lebensdauer des Implantates erzielt, kann kein Kupfer mehr freigesetzt werden, so erlischt die Wirkung.
Die antibakterielle Aktivität von AuPt-Nanopartikeln ist aus Zhao et al.„Tuning the Composition of AuPt Bimetallic Nanoparticles for Antibacterial Application, Angew. Chem. Int. Ed. 2014, 53, S. 8127 bekannt.
Aufgabe der Erfindung ist es eine antibakterielle Beschichtung für ein medizinisches Implantat enthaltend Gold (Au) und Platin (Pt) Nanopartikel bereitzustellen.
Desweiterten ist es Aufgabe der Erfindung eine antibakterielle Beschichtung enthaltend Gold (Au) und Platin (Pt) Nanopartikel, welche auf die Freisetzung von antibakteriellen Wirkstoffen verzichten kann und damit auch dauerhafte Wirkung entfaltet, bereitzustellen.
Ferner ist es Aufgabe der Erfindung ein Verfahren zur Herstellung der erfindungsgemäßen antibakteriellen Beschichtung bereitzustellen.
Darüber hinaus ist es Aufgabe der Erfindung ein mit der erfindungsgemäßen Beschichtung versehendes medizinisches Implantat bereitzustellen.
Die Aufgabe der Erfindung wird u.a. durch eine Beschichtung für ein medizinisches Implantat enthaltend Nanopartikel aus Gold und Platin (AuPt) gelöst.
Hierbei kann der mittlere Durchmesser der Nanopartikel aus Gold und Platin AuPt-Nanopartikel zwischen 2 und 200 Nanometern, bevorzugt zwischen 5 und 180 Nanometern, besonders bevorzugt zwischen 10 und 150 Nanometern liegen.
Die zu beschichtenden medizinischen Implantate müssen bis zu einer Temperatur von 300°C thermisch stabil sein.
Es handelt sich insbesondere um dentale Implantate, verschiedene Formen des
Gelenkersatzes sowie Teile davon und Materialien zur Behandlung von Knochenersatz.
Als Material für die zu beschichtenden medizinischen Implantate kommen Titan,
Titanlegierungen, insbesondere mit Nickel, und/oder Aluminium und/oder Vanadium, z. B. NiTi, TiAI6V4 und TiAI6Nb7, Edelstahl, Legierung aus Kobalt, Chrom und/oder Molybdän, z. B. CoCr- Legierungen und CoCrMo-Legierungen, keramischen Materialien, Formgedächtnislegierungen, und/oder Polymere in Frage.
Die Polymere können ausgewählt sein aus der Gruppe der Polyacrylate, Polymethacrylate, Fluorcarbone, Polyoxymethylene, Polysulfone, Polycarbonate, Polyaryletherketone, Polyamide, Polyethylene, Polypropylene und/oder Silicone.
Es kann sich auch um faserverstärkte, insbesondere carbonfaserverstärkte Composite von Methylmethacrylaten und/oder ultrahochmolekularem Polyethylen handeln.
Die Formgedächtnislegierungen können insbesondere ausgewählt sein aus der Gruppe der Nickel-Titan (Nitinol) (NiTi), Nickel-Titan-Kupfer (NiTiCu), Kupfer-Zink (CuZn), Kupfer-Zink- Aluminium (CuZnAI), Kupfer-Aluminium-Nickel (CuAINi), Eisen-Mangan-Silizium (FeMnSi), Eisen-Nickel-Aluminium (FeNiAl), Eisen-Nickel-Kobalt-Titan (FeNiCoTi) und/oder Zink-Gold- Kupfer (ZnAuCu)-Legierungen.
Die Oberflächen der Implantate können Nitride, Carbide, Carbonitride, Oxide und/oder Apatid aufweisen.
Die zu beschichtenden medizinischen Implantate sollten über eine Oberflächenrauheit Ra zwischen 100 nm und 0,5 nm verfügen.
Die benötigte Oberflächenrauheit lässt sich durch Anodisieren, Passivieren, Sintern, Beizen, Polieren, Schleifen, Läppen, Honen, Sandstrahlen, Bristle Blasting, Ätzen und/oder Bedampfen hersteilen.
Die erfindungsgemäße Beschichtung für ein medizinisches Implantat ist sehr thermostabil, säureresistent und beständig gegen thermische Spannungen, da die AuPt-Teilchen sehr gut auf den Oberflächen der Implantate haften.
Die erfindungsgemäßen antibakteriellen Beschichtungen können nach einem ebenfalls erfindungsgemäßen Verfahren hergestellt werden, wobei dieses die folgenden Schritte umfasst: i. Erwärmung des zu beschichtenden Implantats auf 300 bis 320°C,
ii. Eintauchen des heißen Implantats in eine Lösung A,
iii. Entnahme des Implantats aus der Lösung A,
iv. Abspülen / Waschen des beschichteten Implantats,
wobei die Lösung A überwiegend wässrig ist und in Wasser lösliche Precursoren zur
Abscheidung von Gold (Au) und Platin (Pt) Nanopartikeln enthält.
Als wasserlösliche Precursoren zu Abscheidung von Gold eignen sich Verbindungen des ein- und dreiwertigen Goldes, neben der Tetrachlorogoldsäure (HAuCU) sind hier noch weitere Salze, wie Nitrate z. B. ((N0)[Au(N03)4]) und Verbindungen wie z. B. Gold (III) Acetat
(AU(02CCH3)3), AU( AUCIPF3, K[AU(CN)2] und Na3[Au(S03)2] und diverse metallorganische Verbindungen (Au(CH3)3PF3), Au(CH3)2(acac) (mit acac=Acetylacetonat), CH3Au(P(C2H5)3) dem Fachmann geläufig. Die Precursoren zur Abscheidung des Platins (Pt) sollten ebenfalls wasserlöslich sein. Neben dem H2PtCI6 sind hier auch weitere Chloride wie PtCI2 und PtCI4, Acetylacetonat (acac)-Verbindungen (Pt(acac)2), Tetraamin-Verbindungen Pt(NH3)4(N03)2, Pt(NH3)4(OH)2, Pt(NH3)4(N02)2, Pt(NH3)4CI2 und Nitrate (z.B. (N0)2[Pt(N03)6]) und organo Platin- Komplexe z. B. mit den Liganden Methyl, Methylisonitril, Cyclooctadien, Cyclopentadien, Trimethylphophin bekannt.
Beim Eintauchen (Schritt ii.) sollte die Temperatur der Lösung A zwischen 10 und 30°C, bevorzugt zwischen 15 und 27°C, besonders bevorzugt zwischen 20 und 25°C liegen.
Der Vorgang des Eintauchens (Schritt ii.) sollte zwischen 0, 1 und 60 Sekunden, bevorzugt 0,5 bis 40 Sekunden, besonders bevorzugt 1 bis 20 Sekunden dauern.
Zum Abspülen der Lösungen (Schritt iv.) können dem Fachmann geläufige Lösungen, wie Wasser, bevorzugt demineralisiertes Wasser, besonders bevorzugt destilliertes Wasser und/oder Alkohole verwendet werden.
Bei Bedarf kann das Verfahren auch mehrfach hintereinander ausgeführt werden, da die erfindungsgemäße Beschichtung sehr temperaturstabil ist.
Bei dem erfindungsgemäßen Verfahren ist es von Wichtigkeit, dass beim schnellen Abkühlen von 300 bis 320°C während des Eintauchvorganges (Schritt ii.) die Leidenfrost-Temperatur (290°C) durchlaufen wird, dieses führt zu der Reduktion zu den legierten Metallen Au und Pt aus den Precursoren.
Die erfindungsgemäße Beschichtung für ein medizinisches Implantat kann legierte Nanopartikel aus Gold und Platin (AuPt-Nanopartikel) enthalten.
Die Beschichtung zeigt insbesondere bei der aus der Röntgendiffraktometrie bestimmbaren Gitterkonstanten der D1 i rEbene eine lineare Abhängigkeit vom Anteil der Komponenten in den AuPt-Nanopartikeln.
Ferner ist die Beschichtung bei einem Aufheizen auf bis zu 320°C stabil.
Die Beschichtung ist bei einem pH-Wert von kleiner/gleich 2 stabil.
Das erfindungsgemäße Verfahren zur Herstellung einer Beschichtung kann insbesondere die folgenden Schritte aufweisen:
i. Erwärmung des zu beschichtenden Implantats auf 300 bis 320°C,
ii. Eintauchen des heißen Implantats in eine Lösung A,
iii. Entnahme des Implantats aus der Lösung A,
iv. Waschen des beschichteten Implantats,
wobei die Lösung A überwiegend wässrig ist und in Wasser lösliche Precursoren zur Abscheidung der Metalle Au und Pt enthält.
Zur Umsetzung des Beschichtungsverfahrens können die wasserlöslichen Precursoren bevorzugt HAuCL und H2PtCI6 sein.
Abbildung 1 zeigt schematisch das Prinzip der Bildung der erfindungsgemäßen Beschichtung enthaltend legierte Nanopartikel aus Gold und Platin (AuPt).
Absolut überraschenderweise kommt es ohne Zusatz von Reduktionsmitteln, wie z. B. Citrat- Ionen allein aufgrund des Durchlaufens der Leidenfrost-Temperatur zu einer Reduktion und der Bildung der legierten PtAu-Nano-Teilchen, die im Größenbereich zwischen 2 und 200 nm liegen und bei einmaliger Durchführung des Schrittes (ii.) eine monomodale Größenverteilung aufweisen.
Bei Verwendung des Precursors HAuCL können folgende Reaktionsgleichung angenommen werden:
H20 -» OH* + H* (1)
3 H* + AuCL -» Au° + 3 HCl + CI (2)

Die zugehörigen Reduktionsgleichungen nebst Potential sind:
AuCl 4 + 3e — > Au + 4 CI 1 V
Tabelle 1 zeigt exemplarische Zusammensetzungen der Lösung A und die mittels energiedispersiver Röntgenspektroskopie (EDS) (aufgenommen mit einem INCAx-act, Oxford Instruments, UK) ermittelte resultierende Zusammensetzung der PtAu-Nano-Teilchen, sowie die ausschließlich den Pt oder Au enthaltenden Precursor als Vergleich.
Zur Erzeugung der erfindungsgemäßen Beschichtung für ein medizinisches Implantat sollte das molare Verhältnis von Au zu Pt in der Lösung A zwischen 1 :1 und 1 :10 liegen, bevorzugt sind die Verhältnisse 1 :1 , 1 :2, 1 :3, 1 :4 und 1 :5, besonders bevorzugt sind die Verhältnisse 1 :2, 1 :3 und 1 :4 ganz besonders bevorzugt ist das Verhältnis 1 :3.
Die erfindungsgemäß beschichteten Oberflächen wurden mittels Röntgendiffraktometrie (XRD) unter streifendem Einfall (Gl) (Gerät: X’Pert Pro diffractometer PANalytical, Holland; konstanter Einfallwinkel von 0 = 1°; monochromatische Cu Ka Strahlung mit l = 0,15418 nm) untersucht.
Die Ergebnisse sind in Abbildung 2 zu sehen. Es zeigt sich nur der zur 111-Ebene gehörende hkl-Reflex (dm), dieses zeigt die Bildung von legierten Partikeln. (Tabelle 1)

Abbildung 3 zeigt, dass die Vegard'sche Regel, welche die lineare Abhängigkeit der
Gitterkonstante vom prozentualen Anteil der Komponenten bei Legierungen formuliert, ideal erfüllt ist.
Dieses zeigt ebenfalls, dass es sich um legierte Teilchen handelt.
Abbildung 4 und Abbildung 5 zeigen die REM-Aufnahmen (Gerät Zeiss Ultra Plus) der erfindungsgemäßen Beschichtung. Die Pt39Au6rNanopartikel Lösung A ((2 mM H2PtCI6) 3:1 (2 mM HAuCL)) sind als helle Bereiche in den Abbildungen zu erkennen. Die
erfindungsgemäße Beschichtung ist auf einem rostfreien Edelstahl mit Titannitrit (TiN) Überzug aufgebracht.
Es sind deutlich die Dimensionen der AuPt-Nanopartikel zwischen 2 und 200 Nanometern, bevorzugt zwischen 5 und 180 Nanometern, besonders bevorzugt zwischen 10 und 150 Nanometern, sowie der bevorzugte mittlere Abstand zwischen den legierten AuPt- Nanopartikeln, der im Bereich zwischen 0,5 bis 10 mal des mittleren Durchmessers liegt, zu erkennen.
Ein besonderes Merkmal der Beschichtung ist dabei, dass sie sehr abriebfest und
freisetzungsfrei ist und eine sehr hohe Temperaturstabilität, Beständigkeit gegen thermische Spannungen und Säureresistenz aufweist. Das bedeutet, die Teilchen haften extrem fest auf der Oberfläche. Zur Demonstration dieser Eigenschaften wurde eine erfindungsgemäß erzeugte Beschichtung Pt39Au6 -Nanopartikel (Lösung A ((2 mM H2PtCI6) 3:1 (2 mM HAuCI4))) auf rostfreien Edelstahl mit Titannitrit (TiN) Überzug noch zwei weitere Male mit dem
erfindungsgemäßen Verfahren behandelt. Die REM-Aufnahmen (Gerät Zeiss Ultra Plus) der Proben sind in Abbildung 6, nämlich nach einmaliger erfindungsgemäßer Behandlung,
Abbildung 7, nämlich nach zweimaliger erfindungsgemäßer Behandlung, und Abbildung 8, nach dreimaliger erfindungsgemäßer Behandlung, zu sehen.
Die Anzahl und die mittlere Größe der Teilchen nehmen mit jedem Verfahrensschritt zu.
Dieses zeigt, dass die erfindungsgemäße Beschichtung das Erhitzen auf 300 bis 320°C
(Schritt i.) und das schnelle Abkühlen in wässrigen sauren Lösungen (pH zwischen 2 und 3) (Schritt ii.) unbeschadet überstanden hat.
Die Beschichtung ist:
- antibakteriell,
- temperaturstabil,
- beständig gegen thermische Spannungen,
- abriebfest,
- freisetzungsfrei und
- säureresistent.
In Abbildung 6 ist zu erkennen, dass die Größenverteilung der Teilchen nach einmaliger Durchführung des erfindungsgemäßen Verfahrens monomodal ist. In Abbildung 7 und 8 ist zu erkennen, dass die wiederholte Durchführung des erfindungsgemäßen Verfahrens auch zur Entstehung von größeren Teilchen, welche neben den kleineren vorliegen, führt.
Dieses zeigt deutlich an, dass die Teilchen gut auf der Oberfläche haften und somit auch das Anwachsen bei wiederholter Durchführung des erfindungsgemäßen Verfahrens erlauben.
Material und Methoden:
Die Allgemeinheit der Lehre nicht einschränken sollen hier die verwendeten Geräte und Methoden aufgelistet werden.
Zum Aufheizen der Proben wurde eine handelsübliche Heizplatte der Firma VWR verwendet. Die Temperaturkontrolle erfolgte mit einem Infrarot-Thermometer (Typ: Fluke 566 IR).
Die REM-Aufnahmen wurden mit einem Zeiss Ultra Plus gemacht.
Für die Röntgendiffraktometrie (XRD) unter streifendem Einfall (Gl) wurde das Gerät: X’Pert Pro diffractometer PANalytical, Holland; konstanter Einfallwinkel von 0 = 1 °; monochromatische Cu Ka Strahlung mit l = 0,15418 nm; verwendet.
Die energiedispersive Röntgenspektroskopie (EDS) erfolgte mit einem INCAx-act, Oxford Instruments, UK.

Claims
1. Beschichtung für ein medizinisches Implantat enthaltend legierte Nanopartikel aus Gold und Platin (AuPt-Nanopartikel).
2. Beschichtung nach Anspruch 1 ,
dadurch gekennzeichnet, dass
der mittlere Durchmesser der AuPt-Nanopartikel zwischen 2 und 200 Nanometern bevorzugt zwischen 5 und 180 Nanometern, besonders bevorzugt zwischen 10 und 150 Nanometern liegt.
3. Beschichtung nach Anspruch 1 oder 2,
dadurch gekennzeichnet, dass
die Gitterkonstante, bestimmbar aus der Röntgendiffraktometrie, der d Ebene eine lineare Abhängigkeit vom Anteil der Komponenten in den AuPt-Nanopartikeln zeigt.
4. Beschichtung nach einem der Ansprüche 1 , 2 oder 3,
dadurch gekennzeichnet, dass
die Beschichtung bei einem Aufheizen auf bis zu 320°C stabil ist.
5. Beschichtung nach einem der vorangehenden Ansprüche,
dadurch gekennzeichnet, dass
die Beschichtung bei einem pH-Wert von kleiner/gleich 2 stabil ist.
6. Verfahren zur Herstellung einer Beschichtung nach einem der vorangehenden Ansprüche, umfassend die folgenden Schritte:
i. Erwärmung des zu beschichtenden Implantats auf 300 bis 320°C,
ii. Eintauchen des heißen Implantats in eine Lösung A,
iii. Entnahme des Implantats aus der Lösung A,
iv. Waschen des beschichteten Implantats,
wobei die Lösung A überwiegend wässrig ist und in Wasser lösliche Precursoren zur Abscheidung der Metalle Au und Pt enthält.
7. Verfahren nach Anspruch 6,
dadurch gekennzeichnet, dass
die wasserlöslichen Precursoren bevorzugt HAuCL und H2PtCI6 sind.
8. Verfahren nach Anspruch 6 oder 7,
dadurch gekennzeichnet, dass
das molare Verhältnis von Au zu Pt in der Lösung A zwischen 1:1 und 1:10 liegt, bevorzugt bei 1:1, 1:2, 1:3, 1:4 oder 1 :5, besonders bevorzugt bei 1:2, 1:3 und 1 :4 ganz besonders bevorzugt bei 1:3.
