半導体SWCNTが凝集せず、生体に投与した際に、特定の部位に集積することなく、肺等の血管を閉塞させることがない、毒性の低い生体イメージング用SWCNT分散液を提供することを目的とする。 上記課題を解決するため、本発明の生体イメージング用半導体SWCNT分散液は、平均粒子径が10nmより小さい半導体単層カーボンナノチューブと、前記半導体単層カーボンナノチューブの表面をコーティングする両親媒性物質からなる分散剤とを含むことを特徴とする。

Description

生体イメージング用半導体SWCNT分散液及びその製造方法
 本発明は、生体イメージング用の半導体単層カーボンナノチューブ(SWCNT)分散液及びその製造方法に関する。
 カーボンナノチューブ(以下、CNTともいう)は、炭素原子が平面的に六角形状に配置されて構成された炭素シート(いわゆる、グラファイトからなるシート)が円筒状に閉じた構造を有する炭素構造体である。このCNTには、多層のもの及び単層のものがあるが、単層CNT(以下、SWCNTともいう)の電子物性は、その巻き方(直径や螺旋度)に依存して、金属的性質又は半導体的性質を示すことが知られている。
 半導体SWCNTは、生体透過性の良い近赤外領域(800~2000nm)で光吸収及び発光することから、細胞や生体の機能を検出する蛍光プローブとして有用なものであると期待されている。特に、1200~1400nmの波長領域は、最も生体透過性が良い領域である。
 その半導体SWCNTに酸素原子や官能基を導入することによって、発光波長を変化させることができる。例えば、SWCNTを界面活性剤で分散した水溶液にオゾンを添加した水を混合し、光を照射しながら化学反応させることによって、ナノチューブ壁中の炭素を一部酸素原子に置き換える技術が知られている(非特許文献1及び2)。このようにして酸素原子を導入した場合、ほとんどの酸素原子はSWCNTの壁にエーテル結合し、SWCNTの発光エネルギーは元の発光エネルギーよりも約150meV小さくなる。このような化学修飾には、SWCNTの発光量子収率を増加するという利点もある。
 しかし、非特許文献1及び2で報告されている発光波長の長波長化では、現在最も研究されているSWCNTの一つであるカイラル指数(6,5)を有するSWCNTに対して、その発光波長は、近赤外蛍光プローブとして最も好ましいとされている約1300nm~1400nmより短い、約1140nm(約1.088eV)にピークを有するものが主な生成物であった。
 これに対し、特許文献1には、大気中で半導体単層カーボンナノチューブに直接紫外線を照射することにより、オゾンを発生させて半導体単層カーボンナノチューブを酸化処理することを特徴とする近赤外発光する半導体単層カーボンナノチューブの製造方法が開示されている。
 上記特許文献1によれば、グラム量のSWCNTに対し短時間で簡便に酸素原子を導入することができ、また、その発光波長のピークを980nm(1.265eV)から1280±13nm(0.9686±0.01eV)に変化させることができる。

 しかし、上記特許文献1に記載の方法によって製造された半導体SWCNTは、生体イメージング用分散液(生体用プローブ)として生体(マウス)に投与すると、短時間で特定の部位(特に、肝臓)に集積してしまい、蛍光分布を検出する際、ハレーションを起こすと共に、その他の領域での蛍光分布を検出することができない。また、半導体SWCNTは、凝集性が高いため、マウス等の生体内に投与すると凝集して肺等に詰まり死に至るリスクがあるという問題があった。さらに、半導体SWCNTが凝集することで、半導体SWCNTの発光量子効率が低下するという問題もあった。
 そこで本発明は、上記従来の状況に鑑み、半導体SWCNTが凝集せず、生体に投与した際に、特定の部位に集積することなく、肺等の血管を閉塞させることがない、毒性の低い生体イメージング用SWCNT分散液及びその製造方法を提供することを目的とする。
 本発明者らが鋭意検討を行った結果、特定の半導体SWCNTを、両親媒性物質によってコーティングすることによって半導体SWCNTの分散性が向上することを見い出した。さらに、半導体SWCNT分散液を製造する際に、まず、半導体SWCNTを界面活性剤の溶液中に分散させ、その後に、透析によって界面活性剤を両親媒性物質からなる分散剤で置換することで、分散性に優れた半導体SWCNT分散液が得られることを見い出し、発明を完成した。すなわち、本発明の要旨は以下のとおりである。
(1)平均粒子径が10nmより小さい半導体単層カーボンナノチューブと、前記半導体単層カーボンナノチューブの表面をコーティングする両親媒性物質からなる分散剤とを含む生体イメージング用半導体SWCNT分散液。
(2)前記半導体単層カーボンナノチューブに、酸素原子がエポキシドとして導入されている上記(1)に記載の生体イメージング用半導体SWCNT分散液。
(3)前記半導体単層カーボンナノチューブが、1200nm以上1400nm以下の波長領域に発光波長のピークを有する上記(1)又は(2)に記載の生体イメージング用半導体SWCNT分散液。
(4)前記半導体単層カーボンナノチューブが、大気中で直接紫外線を照射することにより酸化処理された半導体単層カーボンナノチューブである上記(1)に記載の生体イメージング用半導体SWCNT分散液。
(5)前記分散剤が、ポリエチレングリコール脂質誘導体である上記(1)~(4)のいずれか一つに記載の生体イメージング用半導体SWCNT分散液。
(6)上記(1)に記載の生体イメージング用半導体SWCNT分散液の製造方法であって、
 平均粒子径が10nmより小さい半導体単層カーボンナノチューブを、界面活性剤の溶液中に分散させる工程と、
 得られた分散液に、両親媒性物質からなる分散剤を溶解させる工程と、
 得られた溶液から、透析によって界面活性剤を除去する工程と、
を含む生体イメージング用半導体SWCNT分散液の製造方法。
(7)前記半導体単層カーボンナノチューブに、酸素原子がエポキシドとして導入されている上記(6)に記載の生体イメージング用半導体SWCNT分散液の製造方法。
(8)前記半導体単層カーボンナノチューブが、1200nm以上1400nm以下の波長領域に発光波長のピークを有する上記(6)又は(7)に記載の生体イメージング用半導体SWCNT分散液の製造方法。
(9)前記半導体単層カーボンナノチューブが、大気中で直接紫外線を照射することにより酸化処理された半導体単層カーボンナノチューブである上記(6)に記載の生体イメージング用半導体SWCNT分散液の製造方法。
(10)前記分散剤が、ポリエチレングリコール脂質誘導体である上記(6)~(9)のいずれか一つに記載の生体イメージング用半導体SWCNT分散液の製造方法。
(11)前記界面活性剤が、ラウリルベンゼン硫酸ナトリウムである上記(6)~(10)のいずれか一つに記載の生体イメージング用半導体SWCNT分散液の製造方法。
 本発明の生体イメージング用半導体SWCNT分散液は、半導体SWCNTの粒子径が微小である上に、分散剤によって分散状態が維持されるため、特定の臓器(主に、肝臓)に集積することなく、蛍光分布を検出する際のハレーションを低減でき、発光する蛍光の光量低下を防ぐことができる。また、肺等の血管を閉塞させることがなく、毒性が極めて小さい。具体的には、細胞と細胞の隙間に半導体SWCNTが点在して保持されることがあるが、毒性を発現するには至らない。
 また、生体イメージング用半導体SWCNT分散液を製造する際に、透析法によって界面活性剤を除去し、界面活性剤を分散剤によって置換することにより、毒性が著しく低減され、半導体SWCNTの分散性が向上する。

 以下、本発明を詳細に説明する。
 本発明の生体イメージング用半導体SWCNT分散液は、平均粒子径が10nmより小さい半導体SWCNTと、その半導体SWCNTの表面をコーティングする両親媒性物質からなる分散剤とを含むことを特徴とする。
 また、上記の半導体SWCNTは、大気中で直接紫外線を照射することにより酸化処理されたものであることが好ましい。大気中で直接紫外線を照射することにより、オゾンを発生させ、半導体SWCNTに酸素原子を導入する。大気中で直接紫外線を照射して得られる半導体SWCNTは、発光エネルギーを296±10meV低エネルギー側にシフトすることができ、特に、カイラル指数(6,5)を持つSWCNTに適用すると、その発光波長のピークは約980nmから1200nm以上1400nm以下の波長領域へと変化する。具体的には、例えば1280±13nmへと変化し、近赤外蛍光プローブとして好ましい、生体透過性を有する波長領域に発光波長のピークを有するものとなる。
 紫外線を照射することによる酸化処理について、上述の非特許文献1及び2等の従来の湿式による方法(SWCNTを水溶液中で反応させる方法)では、ほとんどの酸素はSWCNTとエーテル結合するために290meVを超える低エネルギーシフトは困難であった。これに対し、大気中で直接紫外線を照射した場合には、導入された酸素原子はほとんどがエポキシドとしてSWCNTに導入され、これによってSWCNTの発光エネルギーを296±10meV低エネルギー側にシフトさせることが可能となる。
 半導体SWCNTの合成方法は特に限定されず、公知の化学気相成長法、アーク放電法、レーザー蒸発法等の方法を用いて適宜合成することができる。特に、触媒の存在下、化学気相成長法によって合成することが好ましい。
 半導体SWCNTの平均粒子径は、10nmより小さく、好ましくは、6nm以上10nm未満の範囲内である。平均粒子径が10nmより小さい微小な半導体SWCNTは、肺の血管等を閉塞することがなく、毒性が低い。ここで、半導体SWCNTの平均粒子径とは、遠心沈降法によって測定される、重量基準の粒度分布における平均径をいう。
 大気中で直接紫外線を照射してオゾンを発生させる際には、密閉された空間内で行うことが好ましく、例えばUVオゾンクリーナ等の、紫外線を大気に照射することによってオゾンを発生させる装置が好ましく用いられる。また、紫外線の照射条件は、用いる装置によって異なるが、照射により半導体SWCNTが破壊されない条件下で行うことが好ましい。
 また、大気中で半導体SWCNTに直接紫外線を照射するために、予め基材上に半導体SWCNTを膜状に形成しておくことが好ましく、特に、酸素原子が導入される半導体SWCNTに均一に化学反応が起こるために、半導体SWCNTを厚さ1μm程度の薄膜状にした状態で紫外線を照射することが好ましい。
 半導体SWCNTの表面をコーティングする両親媒性物質からなる分散剤としては、特に限定されるものではなく、生体に対して毒性が小さく、半導体SWCNTとの親和性に優れるものであれば適宜用いることができる。具体的には、疎水性の脂質部位に親水性のPEGが結合してなるポリエチレングリコール脂質誘導体、核酸、ウシ血清アルブミン等を挙げることができる。特に、ジステアロイルホスファチジルエタノールアミン-PEG2000(DSPE-PEG2000)等のポリエチレングリコール脂質誘導体が好ましく用いられる。
 DSPE-PEG2000等の分散剤によって半導体SWCNTの表面をコーティングすることにより、半導体SWCNTの分散状態が維持され、半導体SWCNTの粒子径が微小であることと相まって、半導体SWCNTが特定の臓器に集積することなく、また、肺の血管等を閉塞することがない。
 酸化処理された半導体SWCNTと、両親媒性物質からなる分散剤との重量割合は、半導体SWCNTの表面が十分にコーティングされ、分散状態が維持できればよく特に限定されるものではないが、酸化処理された半導体SWCNT:分散剤の重量比が、1:2~1:20の範囲内であることが好ましい。
 以上のような生体イメージング用半導体SWCNT分散液を製造するに際しては、まず、上述のように好ましくは大気中で直接紫外線を照射することにより酸化処理された、平均粒子径が10nmより小さい半導体SWCNTを、界面活性剤の溶液中に分散させる。
 ここで、界面活性剤としては、半導体SWCNTを分散できるものであれば良く、陰イオン界面活性剤、陽イオン界面活性剤、両性イオン界面活性剤、非イオン性界面活性剤等の公知の各種界面活性剤から適宜選択して用いることができる。
 陰イオン界面活性剤としては、例えば、アルキルベンゼンスルホン酸塩、アルキルナフタレンスルホン酸塩、アルキルスルホン酸塩、ジアルキルスルホコハク酸塩、アルキル硫酸塩、ポリオキシエチレンアルキルエーテル硫酸塩、アルキルリン酸塩、ポリオキシエチレンアルキルエーテルリン酸塩、コール酸塩、デオキシコール酸塩、グリココール酸塩、タウロコール酸塩、タウロデオキシコール酸塩等を挙げることができる。
 また、陽イオン界面活性剤としては、例えば、テトラアルキルアンモニウム塩、トリアルキルベンジルアンモニウム塩、アルキルピリジウム塩等を挙げることができる。
 両性イオン界面活性剤としては、例えば、2-メタクロイルオキシホスホリルコリンのポリマーやポリペプチド等の両性イオン高分子、3-(N,N-ジメチルステアリルアンモニオ)-プロパンスルホネート、3-(N,N-ジメチルステアリルアンモニオ)プロパンスルホネート、3-(N,N-ジメチルミリスチルアンモニオ)プロパンスルホネート、3-[(3-コラミドプロピル)ジメチルアンモニオ]プロパンスルフォネート(CHAPS)、3-[(3-コラミドプロピル)ジメチルアンモニオ]-2-ヒドロキシプロパンスルホネート(CHAPSO)、n-ドデシル-N,N’-ジメチル-3-アンモニオ-1-プロパンスルホネート、n-ヘキサデシル-N,N’-ジメチル-3-アンモニオ-1-プロパンスルホネート、n-オクチルホスホコリン、n-ドデシルホスホコリン、n-テトラデシルホスホコリン、n-ヘキサデシルホスホコリン、ジメチルアルキルベタイン、パーフルオロアルキルベタイン及びN,N-ビス(3-D-グルコナミドプロピル)-コラミド、レシチン等を挙げることができる。
 さらに、非イオン性界面活性剤としては、例えば、ポリオキシエチレンアルキルエーテル、ポリオキシエチレンアルキルフェニルエーテル、ポリオキシエチレン多価アルコール脂肪酸部分エステル、ポリグリセリン脂肪酸エステル等を挙げることができる。
 特に、半導体SWCNTの分散性に優れることから、ラウリルベンゼン硫酸ナトリウム(SDBS)等のアルキルベンゼンスルホン酸塩が好ましく用いられる。
 半導体SWCNTを、界面活性剤の溶液中に分散させる方法としては、各種ホモジナイザー等を用いて行うことができる。
 得られた分散液は、必要に応じて、遠心分離を行い、上澄みを回収することにより、半導体SWCNTの孤立分散性を高めることができる。孤立分散の半導体単層SWCNTは、生体に投与した場合に、蛍光量子効率の向上、ステルス性の向上、クリアランスの向上という利点があるため好ましい。
 続いて、半導体SWCNTを界面活性剤の溶液中に分散させた分散液に、上述のポリエチレングリコール脂質誘導体等の両親媒性物質からなる分散剤を溶解させ、その後、得られた溶液から透析によって界面活性剤を除去する。これにより、半導体SWCNTの周囲に存在していた界面活性剤がポリエチレングリコール脂質誘導体等の分散剤によって置換され、半導体SWCNTの表面が分散剤によってコーティングされる。
 表面が分散剤でコーティングされた半導体SWCNTは、生体イメージング用半導体SWCNT分散液として、生体に投与した場合に、分散状態が維持されるため、半導体SWCNT自体の粒子径が微小であることと相まって、特定の臓器(主に、肝臓)に集積せず、ハレーションを低減することができる。また、肺の血管等を閉塞することがなく、SDBS等の界面活性剤が透析により除去されているため毒性が極めて低い。また、良好に分散することで、凝集性が低下し、発光する蛍光の光量低下を防ぐことができる。
 以下、実施例及び比較例に基づき本発明をさらに詳細に説明するが、本発明はこれらの実施例に限定されるものではない。
(実施例1)
 カーボンナノチューブ(CoMoCAT SG65i、平均直径0.8nm、以下「半導体SWCNT」という)1mgをエタノール10mlに加え、バスソニケーションに5分ほどかけて半導体SWCNTをエタノールに分散させた。続いて、減圧濾過器にオムニポアメンブレン(φ47mm、5μmポアをセットし、半導体SWCNT/エタノール分散液を入れてろ過し、フィルター上に半導体SWCNTを均一に載せた。次に、半導体SWCNTをフィルターに載せたまま薬包紙で挟み、フィルターが丸まらないよう軽く重石をしながら60℃で30分乾燥させた。そして、フィルターに載せた半導体SWCNTをフィルターごと60~70秒オゾン処理した(光源は水銀ランプ、半導体SWCNT上での紫外線強度は約19mW/cm)。
 オゾン処理を行った後、半導体SWCNTをフィルターごと10mlの1%SDBS-HOに入れ、チップ型ホモジナイザーで氷冷しながら20分間(ON:OFF=1秒:1秒)超音波破砕し、半導体SWCNTをSDBS溶液中に分散させた。続いて、フィルターを取り除いた半導体SWCNT分散溶液を超遠心分離機(104,000g、3時間)にかけ、上澄みを回収し、半導体SWCNT孤立分散液を得た。
 半導体SWCNT孤立分散液に、ジステアロイルホスファチジルエタノールアミン-PEG2000(DSPE-PEG2000)を3mg/mlになるよう添加し、バスソニケーションに5分ほどかけて、DSPE-PEG2000の粉末を溶解させた。そして、透析膜(Spectrum、G235070)にこの溶液を入れ、2リットルの水で透析を行った。この過程で、SDBSがDSPE-PEG2000に置換される。
 2時間後、透析外液を分析用に5ml採取した後に捨て、水と交換した。同様に、1晩後、2日後、3日後に水を交換し、それぞれ分析用にとっておいた透析外液の吸収スペクトルを測定して透析率を計算し、95%以上のSDBSの溶出が確認できたところで、透析を終了した。
 最後に、半導体SWCNT/DSPE-PEG2000分散液の吸収スペクトル、PLスペクトルを測定し、CNTの凝集や消光がないことを確認し、目的の生体イメージング用半導体SWCNT分散液を製造した。なお、生体イメージング用半導体SWCNT分散液について、遠心沈降法による粒子径測定を行ったところ、分散液における半導体SWCNTの平均粒子径は6.5nmであった。
(比較例1)
 カーボンナノチューブ(CoMoCAT SG65i、平均直径0.8nm、以下「半導体SWCNT」という)1mgをエタノール10mlに加え、バスソニケーションに5分ほどかけて半導体SWCNTをエタノールに分散させた。続いて、減圧濾過器にオムニポアメンブレン(φ47mm、5μmポアをセットし、半導体SWCNT/エタノール分散液を入れてろ過し、フィルター上に半導体SWCNTを均一に載せた。次に、半導体SWCNTをフィルターに載せたまま薬包紙で挟み、フィルターが丸まらないよう軽く重石をしながら60℃で30分乾燥させた。そして、フィルターに載せた半導体SWCNTをフィルターごと60~70秒オゾン処理した(光源は水銀ランプ、半導体SWCNT上での紫外線強度は約19mW/cm)。
 オゾン処理を行った後、半導体SWCNTをフィルターごと10mlの0.3%ジステアロイルホスファチジルエタノールアミン-PEG2000(DSPE-PEG2000)水溶液に入れ、チップ型ホモジナイザーで氷冷しながら20分間(ON:OFF=1秒:1秒)超音波破砕し、半導体SWCNTをDSPE-PEG2000水溶液中に分散させた。続いて、フィルターを取り除いた半導体SWCNT分散液を超遠心分離機(104,000g、3時間)にかけ、上澄みを回収し、最後に、半導体SWCNT/DSPE-PEG2000分散液の吸収スペクトル、PLスペクトルを測定し、CNTの凝集や消光がないことを確認し、比較例の生体イメージング用半導体SWCNT分散液を製造した。なお、生体イメージング用半導体SWCNT分散液について、遠心沈降法による粒子径測定を行ったところ、分散液における半導体SWCNTの平均粒子径は8nmであった。
(in vivoイメージング)
 実施例1及び比較例1で得られた生体イメージング用半導体SWCNT分散液を0.1mlマウスに投与し、0~6時間後の蛍光を島津製作所SAI-1000装置を用いて観察した。その結果を図1(実施例1)及び図2(比較例1)に示す。
 図1及び図2に示すように、実施例1の半導体SWCNT分散液は、比較例1に比べて投与後の時間が経過しても特定の臓器に集積しづらく、ハレーションを低減できることが明らかとなった。これは、比較例1では、半導体SWCNTを界面活性剤(SDBS)の溶液中に分散させる工程を経ないため、孤立分散性が低く、DSPE-PEG2000によるコーティングが不十分であり、半導体SWCNTが良好に分散せず一部凝集したためと考えられる。

Claims (11)

  1.  平均粒子径が10nmより小さい半導体単層カーボンナノチューブと、前記半導体単層カーボンナノチューブの表面をコーティングする両親媒性物質からなる分散剤とを含む生体イメージング用半導体SWCNT分散液。
  2.  前記半導体単層カーボンナノチューブに、酸素原子がエポキシドとして導入されている請求項1に記載の生体イメージング用半導体SWCNT分散液。
  3.  前記半導体単層カーボンナノチューブが、1200nm以上1400nm以下の波長領域に発光波長のピークを有する請求項1又は2に記載の生体イメージング用半導体SWCNT分散液。
  4.  前記半導体単層カーボンナノチューブが、大気中で直接紫外線を照射することにより酸化処理された半導体単層カーボンナノチューブである請求項1に記載の生体イメージング用半導体SWCNT分散液。
  5.  前記分散剤が、ポリエチレングリコール脂質誘導体である請求項1~4のいずれか一項に記載の生体イメージング用半導体SWCNT分散液。
  6.  請求項1に記載の生体イメージング用半導体SWCNT分散液の製造方法であって、
     平均粒子径が10nmより小さい半導体単層カーボンナノチューブを、界面活性剤の溶液中に分散させる工程と、
     得られた分散液に、両親媒性物質からなる分散剤を溶解させる工程と、
     得られた溶液から、透析によって界面活性剤を除去する工程と、
    を含む生体イメージング用半導体SWCNT分散液の製造方法。
  7.  前記半導体単層カーボンナノチューブに、酸素原子がエポキシドとして導入されている請求項6に記載の生体イメージング用半導体SWCNT分散液の製造方法。
  8.  前記半導体単層カーボンナノチューブが、1200nm以上1400nm以下の波長領域に発光波長のピークを有する請求項6又は7に記載の生体イメージング用半導体SWCNT分散液の製造方法。
  9.  前記半導体単層カーボンナノチューブが、大気中で直接紫外線を照射することにより酸化処理された半導体単層カーボンナノチューブである請求項6に記載の生体イメージング用半導体SWCNT分散液の製造方法。
