Abstract

The present disclosure discloses a composition comprising ajwain oil; at least one surfactant; and at least one base. The composition is free from alcohol content. The composition of the present disclosure is in form of a disinfectant and also hand sanitizer. Also, the process for preparing the composition has been disclosed in the present disclosure.

Description

COMPOSITION COMPRISING AJWAIN OIL, AND USES THEREOF
FIELD OF INVENTION
[001] The present disclosure relates to the field of consumer and institutional use products in general and herbal sanitising and disinfecting composition in particular. There is provided a composition comprising ajwain oil and uses thereof.
BACKGROUND OF THE INVENTION
[002] Maintaining proper sanitization and following effective hygiene practice ensures healthy lifestyle and reduced risk towards manifestation of undesirable infections. Public places such as hospitals, religious/public/office buildings, restaurants form a virtual abode of disease causing microorganisms. Apart from the public places, areas in home such as kitchen and bathroom also house many such organisms.
[003] Cleaning compositions such as disinfectants or surface cleansers are used for sanitizing non-living surfaces such as floors, furniture and worktops. Such composition mainly comprises one or more antimicrobial agents. Most of the disinfectants available in the market contain chemical actives and synthetic agents such as halogenated compounds, plasticizers and alcohols. The above mentioned antimicrobial agents though effective, are toxic in nature. Disposal of such compositions is hazardous as they contaminate the environment and particularly the water bodies. In addition to being hazardous to the environment such compositions are also corrosive in nature and often damage furniture and worktops in long run.
[004] Sanitizer compositions on the other hand mainly comprise plasticisers dispersed in an alcoholic base. Several sanitizer compositions that are available commonly suffer from the shortcoming of inflammability and high volatility. Also, alcohols such as isopropyl alcohol and ethanol that are commonly employed in such compositions are harsh on skin. The alcohols commonly cause excessive dryness of skin and have also been reported to cause eczema and rash. [005] EP0003625 describes liquid cleaning compositions for non-living surfaces containing hypochlorite and paraffin sulfonates. The composition is an alkaline aqueous solution and produces hypochlorite ions in aqueous solution. Hypochlorite is known to be corrosive and also potentially toxic to living beings. Hence, the usage of such compositions requires special care and protective gear such as gloves and mask.
[006] WO2014158662 discloses cleaning compositions for hard surfaces comprising alkoxylate compounds derived from propylene oxide. Though alkoxylate compounds represent a milder form of surfactant, these compositions are still environmentally toxic.
[007] There are range of sanitisers and cleansers that are currently being marketed. However, as stated above, these products comprise multiple hazardous and synthetic chemical actives that in turn pose risk towards the environment and individuals in general. Therefore, constant efforts are being made for developing cleaning compositions that are based on natural ingredients and at the same time provide with maximum efficiency.
SUMMARY OF INVENTION
[008] In an aspect of the present disclosure, there is provided a composition comprising: (a) ajwain oil; (b) at least one surfactant; and (c) at least one base.

The common disadvantage associated with most of them is their composition that majorly comprise toxic and hazardous chemical agents. Natural ingredients such as essential oils are one of the alternatives that are currently being explored for the replacement of such chemicals. Apart from being safe towards individuals and environment, the bio-actives present in these oils are effective against a range of microorganisms.
[0022] Ajwain oil is an essential oil that is obtained from the fruits of the plant Trachyspermum ammi, also known as Ajowan caraway. The oil is an important source of thymol and carvacrol, which in turn have antiseptic and anti-microbial properties.
[0023] The present disclosure reveals a cleansing composition comprising ajwain oil for use as disinfectants and hand sanitiser. Other components used in the composition are mineralizing agents, antioxidants, surfactants, pH adjusting agents, fragrance and skin conditioners. [0024] The present disclosure is not to be limited in scope by the specific embodiments described herein, which are intended for the purposes of exemplification only. Functionally-equivalent products, compositions, and methods are clearly within the scope of the disclosure, as described herein.

EXAMPLES
[0089] The disclosure will now be illustrated with working examples, which is intended to illustrate the working of disclosure and not intended to take restrictively to imply any limitations on the scope of the present disclosure. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood to one of ordinary skill in the art to which this disclosure belongs. Although methods and materials similar or equivalent to those described herein can be used in the practice of the disclosed methods and compositions, the exemplary methods, devices and materials are described herein. It is to be understood that this disclosure is not limited to particular methods, and experimental conditions described, as such methods and conditions may vary.
[0090] The examples below describe the present invention in detail. The examples illustrate formulations using components as described in subsequent paragraphs. Cleansing composition for disinfectant and hand sanitizer comprising ajwain oil have been exemplified. Working weight percentage for each of the components have also been determined for preparation of both the cleansing compositions. Materials and methods
[0091] Rideal-Walker coefficient (RWC): In order to confirm the efficiency of the disinfectant and the hand sanitizer RW coefficient of the formulations were determined. It is the ratio of the dilution of the disinfectant required to kill microorganism to the dilution of phenol required to kill the organism in the same time under identical conditions. The procedure followed was same as specified in the Indian Drug and Cosmetics Act. The ajwain oil was procured from AG industries (22% to 60% thymol).
[0092] The final trials that passed the RW coefficient test, were sent to Haffkin Institute (Govt, of India recognised) in Mumbai for efficacy testing against hard to kill pathogenic bacteria and fungi as well as viruses and satisfactory results were obtained. Testing was also done at Intertek (internationally acclaimed testing organisation) as per European guidelines EN 1240 and EN 1275 for disinfection efficacy with successful results.
Example 1- Preparation of disinfectant and hand sanitizer (Composition 1):

[0094] Following steps were followed for the preparation Composition 1 : . Ajwain oil along with the surfactant was added to the mixing container and stirred at 100-3000 RPM with the help of a stirrer for 3 to 15 minutes. Other surfactants, if desired can also be added (Any lumps of surfactant and oil is to be avoided at this step.).
2. Demineralized water was added to the above mixture till desired volume was achieved. This mixture of oil, surfactant and water was stirred at 100-3000RPM till completely dissolved. For use of the composition as disinfectant for non-living surfaces, the pH was adjusted to 8-11 using alkaline solution, NaOH. For use as hand sanitiser, pH was adjusted to 5-6 using mild pH adjusting agents such as citric acid and sodium citrate.
Precautions: (a) In all of the above steps, temperature of any of the solutions or components must not be less than 6 °C or more than 40 °C.
(b) Excessive frothing should be avoided while stirring.
Example 2- Preparation of disinfectant and hand sanitizer (Composition 2):

Ajwain oil was mixed with the surfactant and stirred at 100-3000 RPM with the help of a manual/mechanical stirrer for 3 to 15 minutes. Other surfactants, if desired can also be added at this stage (Any lumps of surfactant and oil is to be avoided at this step.).
2. Demineralized water was added to the above mixture to make the volume to 60% of the desired volume. The mixture was stirred at 100-3000 RPM for proper mixing. 3. Next, isopropyl alcohol equivalent to 30% of the total volume of the total desired volume was added. The desired volume was attained by adding demineralized water. This mixture of oil, surfactant, alcohol and water was stirred at 100-3000RPM till completely dissolved.
Precautions: (a) In the above steps, temperature of any of the solutions or components must not be less than 6 °C or more than 40 °C.
(b) Excessive frothing should be avoided while stirring.
Example 3- Preparation of disinfectant
[0097] Different components used in preparation of disinfectant are illustrated below- 1. Surfactant: Sodium lauryl sulphate (SLS)
. Mineralizing agent: Magnesium sulphate and sodium chloride (MgS04+NaCl)
3. Antioxidants: Butylated Hydroxy Anisole (BHA) and Butylated Hydroxy Toluene (BHT)
. Fragrance: citrus fragrance
5. pH adjusting agent: Sodium hydroxide (NaOH) and citric acid
6. Base: Demineralized water
[0098] A two-step procedure was followed for preparation of the disinfectant: 1. Preparation of solution A: Antioxidants BHA and BHT were added to Ajwain oil along with citrus fragrance in a mixing container. The mixture was stirred at 100-3000 RPM with the help of a stirrer for 10 seconds to 1 minute. Surfactant, in this case, SLS (Sodium Lauryl Sulphate) was added to the above mixture of oils and stirred continuously for 3 to 15 minutes. Other surfactants, if desired can also be added (Any lumps of surfactant and oil is to be avoided at this step). 2. Preparation of solution B: demineralized water, approximately 80% of total volume was added in a separate container. Mineralizing agents, magnesium sulphate and sodium chloride was added. pH was adjusted to 8-11 using alkaline solution, NaOH and Citric acid and stirring was done at 100-3000 RPM.
Solution B was added to solution A while stirring continuously at 100 - 3000RPM. Desired volume was made up with demineralized water. pH if required can further be adjusted to 8-1 lusing NaOH and Citric acid.
Precautions: (a) In all of the above steps, temperature of any of the solutions or components must not be less than 6 °C or more than 40 °C.
(b) Excessive frothing should be avoided while stirring.

8. Base or solvent: Demineralized water.
[00101] A five-step procedure was followed for preparation of the disinfectant:
1. Preparation of solution A: Ajwain oil alone or in combination with Citrus fragrance, were added to a mixing container and stirred at 100-3000 RPM with the help of a stirrer for 10 seconds to 1 minute. Surfactant, in this case, Hydrogenated Castor Oil RH 40 was added to the above mixture oil and fragrance and stirred continuously for 3 to 15 minutes. Other surfactants, if desired can also be added. Next, the skin conditioners, propylene glycol and glycerine were added while continuously stirring the solution for minimum 30 seconds to maximum 5 minutes (any lumps of surfactant and oil is to be avoided at this step.).
2. Preparation of solution B (optional step): For adding additional skin conditioners like aloe vera gel, addition was done in a separate container and diluted with demineralised water in a ratio of 1:20 of the gel to water. The mixture was stirred in order to form a uniform mixture. There should be no lumps in this mixture of gel and water.
3. Solution C: solution B was mixed to solution A and stirred continuously until uniform solution was achieved.
4. Solution D: demineralized water, 5-10% of total volume was added in a separate container and heated to boiling point. Antioxidant agents, BHA and BHT was added to the above water and stirred at 100-3000RPM till completely dissolved. The mixture of water and antioxidant agents was cooled to approximately 40 °C, post which mineralizing agents, magnesium sulphate and sodium chloride was added. pH was adjusted to a skin friendly range of 5-7 using citric acid and sodium citrate solution. 5. Solution D was then added to solution C while stirring continuously at 100 - 3000RPM. Desired volume was made up with demineralized water. pH if required can further be adjusted using citric acid and sodium citrate solution to 5-7.
Precautions: (a) In all of the above steps, temperature of any of the solutions or components must not be less than 6°C or more than 40°C, unless specifically mentioned.
(b) Excessive frothing should be avoided while stirring.
Various formulations were prepared for hand sanitizer composition by varying the weight percentage of different components. Results for the hand sanitizer formulations have been depicted in Table 8, 9, 10, 11 and 12. All the values in the table represent weight percentages of the different components. [00102] Table 8, 9, 10, 11 and 12 depicts the composition for different formulations of hand sanitizer and their observations with respect to clarity of the solution, feel of the formulation, colour during storage and pH of the final formulation.
[00103] Table 8: Weight percentage of ajwain oil 0.100%, was kept constant in all the formulations. Column El displays the formulation which satisfies all the observation criteria. However, it was observed that with 0.100% of ajwain oil the RW Coefficient obtained was not satisfactory.

Example 5 - Efficacy of the formulations
Antimicrobial Activity
[00108] Sanitizer formulation
[00109] The antimicrobial activity of sanitizer formulation of the present disclosure was performed by EN 1040 method of testing. It was found to display 99.99% of antimicrobial activity against Salmonella genus.
[00110] Hand sanitizer formulation
[00111] The antimicrobial activity of hand sanitizer formulation of the present disclosure was performed by EN 1040 method of testing. It was found to display 99.99% of antimicrobial activity against each of E. coli, S. aureus, and P. aeruginosa.
[00112] Further, the antimicrobial activity was also tested by a method EN 1275. It was found to display 99.99% of antimicrobial activity against each of Aspergillus niger and Candida albicans.
[00113] Evaluation of anti-bacterial and anti-fungal ability by Percentage killing method:
[00114] It was observed that the sanitizer sample of the present disclosure in an undiluted form at 30 seconds brings about 99.99% killing/inactivation of test species E. coli (ATCC25922), S. typhi (NCTC 786), P. aeruginosa (Fisher's Immuno Type IV), and Methicllin resistant Staphylococcus aureus (MRSA-ATCC 25923). Further, the sanitizer sample undiluted at 60 seconds brings about 99.99% killing/inactivation of both the test fungal species Candida albicans (ATCC 10231) and Aspergillus niger (ATCC 16404).
Viricidal efficacy of the formulation of the present disclosure
[00115] The disinfectant formulation of the present disclosure was tested for its virucidal efficacy against viruses Influenza A HlNl, Rotavirus, and Herpes Simplex 2 virus. The method for testing was as disclosed in Gretorex JS. 2010. Plos One., e8987. Doi: 10.1371; and Springothorpe VS. 1986. J. Hyg., Camb, 97, 139-161.
[00116] It was observed that the sanitizer sample of the present disclosure show toxicity with a change in pH of the medium. This was inferred by the lysis of cell sheath of the susceptible cells. Both these phenomena further implicate the killing of the virus. Thus, a strong cytotoxic activity at the undiluted phase can be considered empirically to be offering potent enough activity to kill all the three viruses under in-vitro conditions. Advantages of the present disclosure:
Overall, the present disclosure provides composition comprising ajwain oil. The disclosure also provides use of the composition in preparation of cleansing compositions such as, disinfectant and hand sanitizer. Other components included in the composition are surfactants, mineralizing agents, antioxidants, and fragrance. The cleansing compositions of the instant disclosure comprise natural active ingredient and therefore pose minimum risk to an individual and environment.

Claims

I/We Claim
1. A composition comprising:
(a) ajwain oil;
(b) at least one surfactant; and
(c) at least one base.
2. The composition as claimed in claim 1, wherein the at least one surfactant is selected from the group consisting of anionic surfactant, amphoteric surfactant, cationic surfactant, non-ionic surfactant, and combinations thereof.
3. The composition as claimed in claim 1, wherein the at least one base is selected from the group consisting of water, isopropyl alcohol, ethyl alcohol, and combinations thereof.
4. The composition as claimed in claim 1, wherein the composition is alcohol free.
5. The composition as claimed in claim 1, wherein the composition is substantially alcohol free.
6. The composition as claimed in claim 1, comprising:
(a) ajwain oil having weight by volume percentage in the range of 0.04%-9% with respect to the composition;
(b) at least one surfactant having weight by volume percentage in the range of 0.25- 40% with respect to the composition; and
(c) at least one base having weight percentage as much as is required to make the composition 100%.
7. The composition as claimed in claim 1, optionally comprising at least one mineralizing agent, at least one antioxidant, at least one pH adjusting agent, at least one fragrance, at least one skin conditioner, at least one colorant, and combinations thereof.
8. The composition as claimed in claim 7, wherein the at least one mineralizing agent is selected from the group consisting of sodium chloride, magnesium sulphate, calcium chloride, potassium bicarbonate, magnesium chloride, calcium carbonate, magnesium carbonate, calcium sulphate, potassium sulphate, manganese sulphate, calcium sulphate, ferrous sulphate, and combinations thereof, and the at least one mineralizing agent has a weight by volume percentage in the range of 0.01-0.5% with respect to the composition.

The composition as claimed in claim 7,wherein the at least one anti-oxidant is selected from the group consisting of butylated hydroxy anisole, butylated hydroxy toluene, ascorbic acid, ascorbyl palmitate, beta carotene, tocopherol, tocopheryl acetate, isoflavones, polyphenols, propyl gallate, dodecyl gallate, kojic acid, manganese dioxide, acetyl cysteine, tetrahydrodiferuloylmethane, quercetin, tertiary butylhydroquinone, ascorbyl palmitate, potassium metabisulfite, sodium metabisulfite, and combinations thereof, and the at least one anti-oxidant has a weight by volume percentage in the range of 0.01 - 5% with respect to the composition.

The composition as claimed in claim 7, wherein the at least one pH adjusting agent is selected from the group consisting of sodium hydroxide, sodium bicarbonate, citric acid, sodium citrate, acetic acid, and combinations thereof, and the at least one pH adjusting agent has a weight by volume percentage in the range of 0.01% - 0.5% with respect to the composition.

The composition as claimed in claim 7, wherein the at least one fragrance is selected from the group consisting of synthetic fragrances, natural fragrances and combinations thereof, and the at least one fragrance has a weight by volume percentage in the range of 0.1% -5% with respect to the composition.

The composition as claimed in claim 7, wherein the at least one skin conditioner is selected from the group consisting of dimethicone copolyols, glycerin, propylene glycol, aloevera gel, tocopherol, D-Panthenol, polyqauternium polymers, shea butter, cocoa butter, petrolatum and combinations thereof, and the at least one skin conditioner has a weight by volume percentage in the range of 0.02-4% with respect to the composition.
13. A disinfectant composition comprising: (a) ajwain oil;
(b) at least one surfactant; and
(c) at least one base.
14. A disinfectant composition comprising:
(a) ajwain oil;
(b) at least one surfactant;
(c) at least one mineralizing agent;
(d) at least one anti-oxidant;
(e) at least one pH adjusting agent; and
(f) at least one base.
15. A hand sanitizer composition comprising:
(a) ajwain oil;
(b) at least one surfactant; and
(c) at least one base.
16. A hand sanitizer composition comprising:
(a) ajwain oil;
(b) at least one surfactant;
(c) at least one mineralizing agent;
(d) at least one anti-oxidant;
(e) at least one fragrance;
(f) at least one pH adjusting agent;
(g) at least one skin conditioner; and
(h) at least one base.
17. A process for preparing the composition as claimed in any one of the claims 1-4, comprising the steps of: (a) obtaining ajwain oil; (b) obtaining at least one surfactant; (c) obtaining at least one base; and (d) contacting ajwain oil, the at least one surfactant, and the at least one base to obtain the composition.
18. A process for preparing the composition as claimed in any one of the claims 1-12, comprising the steps of: (a) obtaining ajwain oil; (b) obtaining at least one surfactant; (c) obtaining at least one mineralizing agent, at least one anti-oxidant, at least one pH adjusting agent, at least one fragrance, one at least one colorant; (d) obtaining at least one base; and (e) contacting ajwain oil, the at least one surfactant, the at least one mineralizing agent, the at least one anti-oxidant, the at least one pH adjusting agent, the at least one fragrance, the at least one colorant, the at least one skin conditioner and the at least one base to obtain the composition.
19. The composition as claimed any one of the claims 1-12, wherein the composition is used as any substance selected from a group consisting of disinfectant, hand sanitizer, wet wipes, concentrate, hand wash, shower gel, liquid soap, bar, and dish-wash liquid.
